Abstract Dermanura gnoma (Handley, 1987) is a phyllostomid bat that occurs in South America. In Brazil, most of the records are concentrated in the Amazon Forest, with sparse data from Cerrado and Atlantic Forest. This study presents the first record of D. gnoma for Alagoas state, and for the Pernambuco Endemism Center (PEC), the most threatened region of the Brazilian Atlantic Forest, characterized by its high rate of endemism, habitat loss, and fragmentation. The species was surveyed at Murici Ecological Station and its identification was supported by morphological and molecular data. We provide the first COI and CYTB gene sequences for this species in the entire Atlantic Forest. Phylogenetic analyses reveal that the generated sequences were recovered in the same clade as the published sequences of D. gnoma from the Amazonian Forest, corroborating the disjunct distribution pattern between Amazonia and PEC, as already reported for other vertebrate taxa. Also, this record increases the number of bat species to 71 for the PEC and extends the geographic distribution of D. gnoma by 758 kilometers to the northeast of its range.

Abstract The progressive fragmentation and loss of habitats represent the main threats for endangered species, causing genetic consequences that may have potential implications for a population’s long-term persistence. Large mammals are the most affected species among vertebrates. The giant armadillo Priodontes maximus is a large South American mammal threatened species, showing nocturnal, solitary and fossorial behavior, occurring at low population densities, and its population dynamics are still poorly known. In this study, we carried out the first assessment of genetic variability and population genetic structure of the species, using a panel of 15 polymorphic microsatellites developed by high-throughput genome sequencing. The spatial Bayesian clustering, Fst and Dest results indicated the presence of two genetic clusters (K = 2) in the study area. These results suggest a reduction in gene flow between individuals inhabiting the Brazilian savanna (Cerrado) and the Pantanal wetlands, with the increased human-driven habitat modifications possibly contributing for this scenario. A bottleneck signal was detected in both populations, and a subpopulation structuring in the Cerrado may also be reflecting consequences of the extensive habitat modifications. Findings from this study provide important and useful information for the future maintenance of genetic diversity and long-term conservation of this flagship species.

Forest corridor has been considered the main strategy for maintaining gene flow between isolated populations, yet their effectivity is poorly tested. Assessing signatures of genetic variation loss, gene flow reduction and inbreeding may be helpful for conservation of the biodiversity that needs large continuous areas. Here we evaluated the genetic structure and diversity of the largest neotropical mammal, the lowland tapir (Tapirus terrestris), living in the largest Atlantic forest corridor in Brazil. We used fecal-derived DNA, genotyped nine polymorphic microsatellite loci of 75 tapirs, and quantified genetic differentiation, genetic diversity, and landscape resistance to gene flow. We found genetic differentiation between the inland and coastal populations, which may be explained by elevation. Expected heterozygosity ranged between 0.64 (inland population) and 0.78 (coastal population), and a small Ne was observed in both populations. We demonstrated that even large continuous rainforests are not totally permeable to the gene flow of large organisms. Our study also changes our perception about the pristine of continuous corridors and their role for long-term survival of large mammals, suggesting that tapir conservation efforts should be taken even for populations in the large protected areas.

Abstract Introduced in South America at the end of the 19th century, the European hare population has expanded dramatically and now represents a risk to native Brazilian forest rabbits. Monitoring the invasive Lepus europaeus and its coexistence with native Sylvilagus brasiliensis is a challenge that can be efficiently addressed by the use of molecular tools. This work describes a set of primers useful for amplifying three mini‐barcodes for the molecular identification of both invasive and native leporid species using degraded fecal DNA. In addition, tests in silico indicate that these mini‐barcodes can successfully amplify the DNA sequences of a number of leporids. These mini‐barcodes constitute a powerful tool for the monitoring and management of the invasive L. europaeus and the conservation of native rabbits.

Abstract Schizodon encompasses approximately 15 species of Neotropical headstanding fishes. Integrative taxonomy, combining molecular and morphometric analyses with traditional taxonomic methods, was used to investigate Schizodon vittatus and its potential new sister species. Molecular differences between the two species in the barcode are greater than intra-specific variation recovered in species of Schizodon, and the two species represent distinct lineages for approximately one million years. The two species are morphologically very similar, and the meristic data showed great overlap. Morphometric analyses also showed overlap among the putative species but indicated differences in caudal-peduncle depth, orbital diameter, and length of anal-fin rays. Color pattern seems to provide a clear diagnostic feature for the two species. Schizodon vittatus usually has four dark brown transversal bars on body, and its sister species has three conspicuous bars, with the fourth, if present, inconspicuous and dorsal to the lateral line. Schizodon vittatus is redescribed based on the type and recently collected specimens, its type locality is revisited, and its known distribution restricted to the Araguaia and Tocantins drainages. The new species, sister to S. vittatus, distributed in the Xingu and Tapajós drainages, is described. A key for the identification of the Amazon clade species of Schizodon is provided.

The Epinephelidae form a group of species of high biological and economical interests. It´s phylogeographic patterns are not well known especially the distributed populations in the western region of the Atlantic Ocean. Among the representatives is a small species called Cephalopholis fulva, Coney, which presents a wide geographical distribution, polychromia, hermaphroditism and is quickly becoming a large target for the exploration of commercial fishing. The genetic and historical demography were obtained through the partial sequence analysis of Control Region from six locations on the coastline of Brazil from the northeast coast to the southwest coast, including the oceanic islands of Rocas Atoll and Fernando de Noronha Archipelago. The analyzed samples revealed a high genetic variability and a strong gene flow among the sampled locations. Additionally, the genetic data revealed that population expansions probably occurred due to the changes in the sea levels that occurred during the Pleistocene. The large population connectivity found in Coney constitutes relevant conditions for their biological conservation.

Prochilodus is one of the most important fish resources of South America, in addition to the important role it plays in nutrient cycling of Neotropical rivers. In the present study, we describe the isolation and characterization of nine novel microsatellite loci in Prochilodus argenteus. The number of alleles per polymorphic locus varied from 5 (Par76) to 21 (Par85), revealing a total of 116 alleles. The values of observed and expected heterozygosities ranged from 0.629 (Par69) to 0.926 (Par85 and Par86) and from 0.643 (Par66) to 0.931 (Par80), respectively. Furthermore, the ability of these and other previously described microsatellite markers to amplify orthologous loci was tested in two related species, Prochilodus costatus and Prochilodus lineatus. These loci will be useful for studies of population genetic structure in this group of fishes, and in aiding future genetic mapping studies of P. argenteus.

Curimbatá-pióa (Prochilodus costatus Valenciennes, 1850) is an endemic species from the São Francisco River Basin with migratory habits, important ecological role in the ecosystem and relevant fishing importance. The present study aimed to assess the genetic variation in P. costatus in order to investigate its population genetic structure. Genetic variation was studied at three sites downstream the Três Marias dam (Upper-middle São Francisco River, State of Minas Gerais) through six specific microsatellite loci. Fish from the three sites had quite similar genetic diversity levels and no genetic differentiation was detected, suggesting that P. costatus might represent a single reproductive unit in the studied area. Alternatively, the present study was not able to detect putative coexistent and comigrating populations along the main channel river. Our findings could be very helpful for the management and conservation of this fish.

Prochilodus lineatus, an abundant species in the Mogi-Guaçu river basin, represents a large part of the region’s fishing potential. Karyotypic analyses based on classic cytogenetic techniques have revealed the presence of 54 meta-submetacentric type chromosomes, together with the occurrence of small supernumerary chromosomes with intra and interindividual variations. This paper describes the genomic organization of two families of satellite DNA in the P. lineatus genome. The chromosomal localization these two repetitive DNA families through fluorescence in situ hybridization (FISH) demonstrated that the SATH1 satellite DNA family, composed of approximately 900 bp, was located in the pericentromeric region of a group of chromosomes of the standard complement, as well as on all the B chromosomes. The SATH2 satellite family has a monomeric unit of 441 bp and was located in the pericentromeric regions of some chromosomes of the standard complement, but was absent in the B chromosomes. Double FISH analyses showed that these two families participate jointly in the pericentromeric organization of several chromosomes of this species. The data obtained in this study support the hypothesis that the B chromosomes derive from chromosomes of the standard complement, which are carriers of the SATH1 satellite DNA. Objective. Here we aimed to identify the main points of animal death by roadkill in the view of helping mitigation plans and reducing the impact over the local fauna of a protected area. Materials and methods. We surveyed the roads around a protected area of Cerrado (São Paulo, Brazil) from May 2012 to August 2013. We recorded the local of roadkills, biometric and morphologic data of the animals, and collected samples of tissue for molecular species confirmation. Results. Thirty-one roadkilled animals were registered, including threatened species: Leopardus pardalis; Cuniculus paca and Chrysocyon brachyurus. Most roadkills were represented by mammals (54.8%) and reptiles (38.7%), and the mortality rate was 1.46 animals/km/year. Three roadkill hotspots were detected, suggesting that they were important points of animal crossing, probably because of the existence of natural remnant vegetation and intersection of roads by riparian vegetation. Conclusions. This work provided strong evidence of the most critical points where mitigation strategies should be immediately implemented and highlighted the importance of detecting roadkill hotspots and the species or taxonomic groups more affected, helping to elaborate effective actions that can improve fauna conservation.
