Abstract Background Identifying children and adolescents with cardiometabolic risk at an early stage is crucial for effective treatment and prevention. From a practical perspective, this could be accomplished by assessing the presence of abdominal obesity, which serves as a surrogate indicator of increased cardiometabolic risk and is easy to measure. However, the assessment of abdominal obesity via waist circumference has not yet become a standard procedure in pediatric healthcare. The present study aimed to analyze the secular trends in increased cardiometabolic risk, as indicated by waist circumference among Spanish children and adolescents. Methods This study included 4861 children and adolescents aged 8 to 16 years from two nationwide representative cross-sectional surveys, the EnKid study and the PASOS study, conducted in 1998–2000 and 2019–2020, respectively. Anthropometric variables were measured in both surveys by trained personnel. Three different waist-to-height (WHtR) cutoffs were used to define abdominal obesity as criteria for cardiometabolic risk. Abstract Despite the increase in research of hamstring stiffness through the use of ultrasound-based shear wave elastography, the active stiffness of biceps femoris long head (BFlh) and semitendinosus (ST) muscles under fatigue conditions at various contraction intensities has not been sufficiently explored. This study aimed to compare the effects of knee flexor’s isometric contraction until exhaustion performed at 20% vs. 40% of maximal voluntary isometric contraction (MVIC), on the active stiffness responses of BFlh and ST. Eighteen recreationally active males performed two experimental sessions. The knee flexors’ MVIC was assessed before the fatiguing task, which involved a submaximal isometric contraction until failure at 20% or 40% of MVIC. Active muscle stiffness of the BFlh and ST was assessed using shear wave elastography. BFlh active stiffness remained relatively unaltered at 20% of MVIC, while ST active stiffness decreased from ≅ 91% contraction time (55.79 to 44.52 kPa; p 0.05) or ST (63.62 to 53.54 kPa; p > 0.05) at 40% of MVIC session. Intermuscular active stiffness at 20% of MVIC differed until 64% contraction time (p 0.05), but a steeper increase in BFlh/ST ratio was found for 20% (0.004 ± 0.003 ratio/%) compared to 40% (0.001 ± 0.003 ratio/%) of MVIC (p = 0.003). These results suggest that contraction duration could play a major role in inducing changes in hamstrings’ mechanical properties during fatigue tasks compared to contraction intensity.

Abstract Soil-transmitted helminth (STH) infections account for a significant global health burden, necessitating mass drug administration with benzimidazole-class anthelmintics, such as albendazole (ALB), for morbidity control. However, ALB efficacy shows substantial variability, presenting challenges for achieving consistent treatment outcomes. We have explored the potential impact of the baseline gut microbiota on ALB efficacy in hookworm-infected individuals through microbiota profiling and machine learning (ML) techniques. Our investigation included 89 stool samples collected from hookworm-infected individuals that were analyzed by microscopy and quantitative PCR (qPCR). Of these, 44 were negative by microscopy for STH infection using the Kato-Katz method and qPCR 21 days after treatment, which entails a cure rate of 49.4%. Microbiota characterization was based on amplicon sequencing of the V3–V4 16S ribosomal RNA gene region. Alpha and beta diversity analyses revealed no significant differences between participants who were cured and those who were not cured, suggesting that baseline microbiota diversity does not influence ALB treatment outcomes. Furthermore, differential abundance analysis at the phylum, family and genus levels yielded no statistically significant associations between bacterial communities and ALB efficacy. Utilizing supervised ML models failed to predict treatment response accurately. Our investigation did not provide conclusive insights into the relationship between gut microbiota and ALB efficacy. However, the results highlight the need for future research to incorporate longitudinal studies that monitor changes in the gut microbiota related to the infection and the cure with ALB, as well as functional metagenomics to better understand the interaction of the microbiome with the drug, and its role, if there is any, in modulating anthelmintic treatment outcomes in STH infections. Interdisciplinary approaches integrating microbiology, pharmacology, genetics and data science will be pivotal in advancing our understanding of STH infections and optimizing treatment strategies globally. Abstract Background Patients with active cancer and aortic stenosis may be under-referred for valve interventions due to concerns over a prohibitive risk. However, whether active cancer impacts outcomes after transcatheter aortic valve replacement (TAVR) remains unknown. Methods We searched PubMed, Embase, and Cochrane Library in December 2023 for studies comparing the post-TAVR outcomes of patients with versus without active cancer. We pooled odds ratios (OR) and adjusted hazard ratios (aHR) with 95% confidence intervals (CI) applying a random-effects model. Statistical analyses were performed in R version 4.3.2. Results We included nine observational studies analyzing 133,906 patients, of whom 9,792 (7.3%) had active cancer. Abstract Background The number of people with palliative care needs is projected to increase globally. Chile has recently introduced legislation for universal access to palliative care services for patients with severe and terminal illnesses, including non-cancer conditions. We aimed to estimate the number of people affected by serious health-related suffering and need for palliative care in Chile to 2050. Methods We used data on all deaths registered in Chile between 1997-2019 and population estimates for 1997–2050. We used Poisson regression to model past trends in causes of death adjusted by age, sex and population estimates, to project the number of deaths for each cause from 2021 to 2050. We applied the Lancet Commission on Palliative Care and Pain Relief weights to these projections to identify decedents and non-decedents with palliative care needs. Results Population palliative care needs in Chile are projected to increase from 117 (95% CI 114 to 120) thousand people in 2021 to 209 (95% CI 198 to 223) thousand people in 2050, a 79% increase (IRR 1.79; 95% CI 1.78–1.80). This increase will be driven by non-cancer conditions, particularly dementia (IRR 2.9, 95% CI 2.85–2.95) and cardiovascular conditions (IRR 1.86, 95% CI 1.83–1.89). By 2050, 50% of those estimated to need palliative care will be non-decedents (not expected to die within a year). Conclusions Chile will experience a large increase in palliative care needs, particularly for people with dementia and other non-cancer conditions. Improved availability of high-quality services, expanded clinician training and new sustainable models of care are urgently required to ensure universal access to palliative care.

BackgroundMalaria is a devasting parasitic disease that causes over half a million deaths every year. The necessity for prompt and thorough antimalarial drug discovery and development is accelerated by the rise in multidrug resistance and the lack of an effective vaccine. The Plasmodium spp. proteasome represents a prospective target for antimalarial treatment since several chemotherapy types have been shown to potently and selectively limit the growth of parasites. Combined with first-line artemisinin medicines, it creates synergy, even in the artemisinin-resistant parasites.MethodsPRISMA guidelines were used in the development of this systematic review. A literature search was performed in March 2024 in PubMed, Science Direct, and Scopus databases, with the following keywords: ((antimalarial resistance) AND (plasmodium OR malaria) AND (proteasome)) NOT (cancer [Title/Abstract]). Only articles with the susceptibility assessment were included.ResultsHerein, 35 articles were included in the systematic review, which was divided into two subcategories: those that studied the UPS inhibitors, which accounted for 25 articles, and those that studied genetic modifications, including knockouts, knockdowns, and mutations, in the UPS toward antimalarial resistance, accounting for 16 articles. 6 articles included both subcategories. In total, 16 categories of inhibitors were analyzed, together with two knockdowns, one knockout, and 35 mutations.ConclusionIn this study, we reviewed the literature for available inhibitors and their respective susceptibility and ability to develop resistance toward Plasmodium spp. 26 s proteasome. The proteasome was highlighted as a potential antimalarial target and as an artemisinin partner drug. However, host toxicity and susceptibility to resistance appear as the main obstacle in the development of highly potent drugs, indicating a need for additional scrutiny during any further drug development efforts.

IntroductionDomestic wildlife markets have important nutritional, medicinal, cultural, and financial significance for local communities, but the scale and diversity of wildlife trade that passes through them is also associated with negative impacts on biodiversity, poor animal welfare, and potential human health risk. To design, and monitor the effectiveness of, interventions to ameliorate such impacts, an understanding of the species sold at the markets and their purpose is required, together with a robust (and potentially flexible) baseline. Here we focus on Belén (the largest open wildlife market in the Peruvian Amazon) and Modelo market, in Iquitos, Peru.MethodsWe surveyed wildlife products for sale at both markets approximately weekly over a year, using two different survey methods (open and discreet). To provide a baseline to support future conservation monitoring, we estimated a number of different market metrics (including indices of product availability, volume (observed per survey), and price), for the most frequently observed species ‘groups’, and compared indices of trade volume with daily river water levels. To provide a complete understanding of the range of species involved, we also described all threatened species recorded at the markets, the products sold, and their uses, including those that were only observed occasionally.ResultsBoth markets sold predominantly wild meat, and some pets; at Belén Market >30% observations were of decorative, spiritual, or medicinal products. At least 71 unique species (including mammals, reptiles, birds, and invertebrates) were observed in total. The most frequently observed species ‘groups’ were: lowland paca, peccaries, caiman, river turtles, boas, yellow footed tortoise, parrots, and brocket deer. 27.7% of species were threatened or Near Threatened globally or nationally but there was no evidence that discreet surveys increased their detection. Daily river water levels were positively correlated with indices of trade volume for lowland paca, caiman, and yellow-footed tortoise, and negatively correlated with indices of trade volume for parrots and river turtle eggs.DiscussionBeyond providing a comparative dataset, and insights regarding the apparent availability and value of a diversity of products (including food items, live pets, and other decorative, spiritual, and medicinal items), we suggest that simulations using these data could be used to optimize future monitoring efforts. Finally, our observations of correlations of per survey trade volumes of some species with daily river water levels in Iquitos may inform optimal time of year for species- specific surveys.

Abstract Given the ongoing environmental degradation from local to global scales, it is fundamental to develop more efficient means of gathering data on species and ecosystems. Local ecological knowledge, in which local communities can consistently provide information on the status of animal species over time, has been shown to be effective. Several studies demonstrate that data gathered using local ecological knowledge (LEK)‐based methods are comparable with data obtained from conventional methods (such as line transects and camera traps). Here, we employ a consensus analysis to validate and evaluate the accuracy of interview data on LEK. Additionally, we investigate the influence of social and bioecological variables on enhancing data quality. We interviewed 323 persons in 19 villages in the Western and Central Amazon to determine the level of consensus on the abundance of hunted and non‐hunted forest species. These villages varied in size, socio‐economic characteristics and in the experience with wildlife of their dwellers. Interviewees estimated the relative abundance of 101 species with a broad spectrum of bioecological characteristics using a four‐point Likert scale. High consensus was found for species population abundance in all sampled villages and for 79.6% of interviewees. The village consensus of all species abundance pooled was negatively correlated with village population size. The consensus level was high regardless of the interviewees' hunting experience. Species that are more frequently hunted or are more apparent had greater consensus values; only two species presented a low consensus level, which are rare and solitary species. We show in our study in the Amazon that information gathered by local peoples, Indigenous as well as non‐Indigenous, can be useful in understanding the status of animal species found within their environment. The high level of cultural consensus we describe likely arises from knowledge sharing and the strong connection between the persons interviewed and the forest. We suggest that consensus analysis can be used to validate LEK‐generated data instead of comparing these types of data with information obtained by conventional methods. Abstract Background Nursery pigs undergo stressors in the post-weaning period that result in production and welfare challenges. These challenges disproportionately impact the offspring of primiparous sows compared to those of multiparous counterparts. Little is known regarding potential interactions between parity and feed additives in the post-weaning period and their effects on nursery pig microbiomes. Therefore, the objective of this study was to investigate the effects of maternal parity on sow and offspring microbiomes and the influence of sow parity on pig fecal microbiome and performance in response to a prebiotic post-weaning. At weaning, piglets were allotted into three treatment groups: a standard nursery diet including pharmacological doses of Zn and Cu (Con), a group fed a commercial prebiotic only (Preb) based on an Aspergillus oryzae fermentation extract, and a group fed the same prebiotic plus Zn and Cu (Preb + ZnCu). Results Although there were no differences in vaginal microbiome composition between primiparous and multiparous sows, fecal microbiome composition was different (R 2 = 0.02, P = 0.03). Background: Visual exploration is abnormal in schizophrenia; however, few studies have investigated the physiological responses during selecting objectives in more ecological scenarios. This study aimed to demonstrate that people with schizophrenia have difficulties observing the prominent elements of an image due to a deficit mechanism of sensory modulation (active sensing) during natural vision. Methods: An electroencephalogram recording with eye tracking data was collected on 18 healthy individuals and 18 people affected by schizophrenia while looking at natural images. These had a prominent color element and blinking produced by changes in image luminance. Results: We found fewer fixations when all images were scanned, late focus on prominent image areas, decreased amplitude in the eye-fixation-related potential, and decreased intertrial coherence in the SCZ group. Conclusions: The decrease in the visual attention response evoked by the prominence of visual stimuli in patients affected by schizophrenia is generated by a reduction in endogenous attention mechanisms to initiate and maintain visual exploration. Further work is required to explain the relationship of this decrease with clinical indicators.

AbstractThe present study examines the relationship between state anxiety, trait anxiety, and social anxiety among students enrolled in a private university in South Lima. The research design employed was correlational, utilizing a nonexperimental cross-sectional approach. The sample consisted of 449 male and female university students, aged 18 to 30, enrolled in psychology and law programs from the third to eleventh cycle. The participants completed an online survey utilizing two validated instruments: the State-Trait Anxiety Inventory (STAI) and the Social Anxiety Questionnaire for Adults (CASO-A30). Descriptive and inferential statistics were employed for the purpose of data analysis. The findings indicated a stronger correlation between state anxiety and social anxiety than between trait anxiety and social anxiety. The mean scores for state anxiety and trait anxiety were both moderate, with trait anxiety exhibiting a slightly higher mean score. Approximately 29.84% of students reported high levels of state anxiety, while 25.39% reported high levels of trait anxiety. Significant differences were observed in anxiety levels based on age, career, and study cycle, but not on gender. In conclusion, this study demonstrated a positive and highly significant correlation between state anxiety, trait anxiety, and social anxiety among university students. State anxiety exhibited a stronger association with social anxiety than trait anxiety.

ABSTRACTBackground Due to the increase in global temperature, it is necessary to investigate solutions so that athletes competing in hot conditions can perform in optimal conditions avoiding loss of performance and health problems. Therefore, this study aims to evaluate the effect of pre-exercise glycerol supplementation during a rectangular test at ambient temperature mid (28.2ºC) on dehydration variables in international race walkers.Methods Eight international male race walkers (age: 28.0 years (4.4); weight: 65.6 kg (6.6); height: 180.0 cm (5.0); fat mass: 6.72% (0.66); muscle mass: 33.3 kg (3.3); VO2MAX: 66.5 ml · kg−1·min−1 (1.9)) completed this randomized crossover design clinical trial. Subjects underwent two interventions: they consumed placebo (n = 8) and glycerol (n = 8) acutely, before a rectangular test where dehydration, RPE, metabolic, kinematic, and thermographic variables were analyzed before, during and after the test.Results After the intervention, significant differences were found between groups in body mass in favor of the placebo (Placebo: −2.23 kg vs Glycerol: −2.48 kg; p = 0.033). For other variables, no significant differences were found.Conclusion Therefore, pre-exercise glycerol supplementation was not able to improve any dehydration, metabolic, kinematic, or thermographic variables during a rectangular test at temperature mid in international race walkers. Possibly, a higher environmental temperature could have generated a higher metabolic and thermoregulatory stress, generating differences between groups like other previous scientific evidence.

Activity simulation protocols offer useful applications in research and practice; however, the specificity of such protocols to basketball game-play is currently lacking. Consequently, this study aimed to develop a game-specific basketball activity simulation protocol representative of typical playing durations and assess its reliability and discriminant validity. The simulation protocol was modified from an original version (i.e., Basketball Exercise Simulation Test) to incorporate regular breaks indicative of time-outs, free-throws, and substitutions. Twelve competitive male and female adult basketball players competing in the fourth or fifth Spanish basketball division underwent repeated trials of the simulation protocol (min. 4 to max. 14 days apart) for reliability analyses. In turn, 13 competitive male (fifth division), 9 competitive female (fourth division), and 13 recreational male adult basketball players completed the simulation protocol to assess discriminant validity via comparisons between sexes (competitive players) and playing levels (males). A range of physical, technical, and perceptual-physiological variables were collected during and following the simulation protocol. Several physical and heart rate variables displayed the strongest reliability (intraclass correlation coefficient [ICC] = 0.72–0.96; coefficient of variation [CV] = 1.78–6.75%), with physical decrement, technical, blood lactate concentration, and rating of perceived exertion (RPE) variables having the weakest (ICC = 0.52–0.75; CV = 10.34–30.85%). IntroductionThere are no microbiological regulatory limits for viruses in animal feed and feed ingredients.MethodsA performance objective (PO) was proposed in this study to manufacture a spray-dried porcine plasma (SDPP) batch absent of any infectious viral particles. The PO levels of −7.0, −7.2, and −7.3 log TCID50/g in SDPP were estimated for three batch sizes (10, 15, and 20 tons).Results and discussionA baseline survey on the presence of porcine epidemic diarrhea virus (PEDV) in raw porcine plasma revealed a concentration of −1.0 ± 0.6 log TCID50/mL as calculated using a TCID50-qPCR derived standard curve. The mean African swine fever virus (ASFV) concentration in raw plasma was estimated to be 0.6 log HAD50/mL (0.1–1.4, 95% CI) during a pre-clinical scenario (collected from asymptomatic and undetected viremic pigs). Different processing scenarios (baseline: spray-drying + extended storage) and baseline + ultraviolet (UV) radiation were evaluated to meet the PO levels proposed in this study. The baseline and baseline + UV processing scenarios were >95 and 100% effective in achieving the PO for PEDV by using different batch sizes. For the ASFV in SDPP during a pre-clinical scenario, the PO compliance was 100% for all processing scenarios evaluated. Further research is needed to determine the underlying mechanisms of virus inactivation in feed storage to further advance the implementation of feed safety risk management efforts globally.

Abstract Background The particle structure of Emiliania huxleyi virus (EhV), an algal infecting member of nucleocytoplasmic large DNA viruses (NCLDVs), contains an outer lipid membrane envelope similar to that found in animal viruses such as African swine fever virus (ASFV). Despite both being enveloped NCLDVs, EhV and ASFV are known for their stability outside their host environment. Method Here we report for the first time, the application of a viability qPCR (V-qPCR) method to describe the unprecedented and similar virion thermal stability of both EhV and ASFV. This result contradicts the cell culture-based assay method that suggests that virus “infectivity” is lost in a matter of seconds (for EhV) and minutes (for ASFV) at temperature greater than 50 °C. Confocal microscopy and analytical flow cytometry methods was used to validate the V-qPCR data for EhV. Results We observed that both EhV and ASFV particles has unprecedented thermal tolerances. These two NCLDVs are exceptions to the rule that having an enveloped virion anatomy is a predicted weakness, as is often observed in enveloped RNA viruses (i.e., the viruses causing Porcine Reproductive and Respiratory Syndrome (PRRS), COVID-19, Ebola, or seasonal influenza). Using the V-qPCR method, we confirm that no PRRSV particles were detectable after 20 min of exposure to temperatures up to 100 °C. We also show that the EhV particles that remain after 50 °C 20 min exposure was in fact still infectious only after the three blind passages in bioassay experiments. Conclusions This study raises the possibility that ASFV is not always eliminated or contained after applying time and temperature inactivation treatments in current decontamination or biosecurity protocols. This observation has practical implications for industries involved in animal health and food security. Finally, we propose that EhV could be used as a surrogate for ASFV under certain circumstances.

AbstractBackground The COVID-19 pandemic, caused by SARS-CoV-2 infection, has become the most devastating zoonotic event in recent times, with negative impacts on both human and animal welfare as well as on the global economy. Although SARS-CoV-2 is considered a human virus, it likely emerged from animals, and it can infect both domestic and wild animals. This constitutes a risk for human and animal health including wildlife with evidence of SARS-CoV-2 horizontal transmission back and forth between humans and wild animals.Aim Molecular surveillance in different wildlife rehabilitation centers and wildlife associated institutions in Chile, which are critical points of animal-human interaction and wildlife conservation, especially since the aim of wildlife rehabilitation centers is to reintroduce animals to their original habitat.Materials and Methods The survey was conducted in six WRCs and three wildlife associated institutions. A total of 185 samples were obtained from 83 individuals belonging to 15 different species, including vulnerable and endangered species. Each specimen was sampled with two different swabs: one oropharyngeal or nasopharyngeal according to the nostril diameter, and/or a second rectal sample. RNA was extracted from the samples and two different molecular assays were performed: first, a conventional RT-PCR with pan-coronavirus primers and a second SARS-CoV-2 qPCR targeting the N and S genes.Results All 185 samples were negative for SARS-CoV-2.Clinical relevance This study constitutes the first report on the surveillance of SARS-CoV-2 from wildlife treated in rehabilitation centers in Chile, and supports the biosafety procedures adopted in those centers.

Introduction: Due to the possible impact of the thermoregulatory process on sports performance, it is necessary to explore the existing relationships between kinetic, mechanical, and physiological variables. The objective of this study was to evaluate metabolic stress using thermography in the lower limb after the Spanish Championship 2023 walk. Method: A descriptive study was carried out on national and international race walkers before and after the 2023 Spanish Championships. The participants performed different tests within the same circuit. Five walkers completed the long-distance race of 35 km, four walkers completed the middle-distance race of 20 km and finally, two walkers completed the short-distance race of 10 km. Result: Statistically significant changes were observed in the lower limbs of the walkers after completing the test. We observed a decrease in skin temperature in all the anatomical regions analyzed, except for the back of the leg. More specifically, the decrease was in the hip (−1.92 °C: p = 0.004), quadriceps, hamstrings (−1.23 °C: p = 0.002), and tibia (−1.23 °C: p = 0.030). However, in the posterior region of the leg, a significant increase in temperature was observed (+0.50 °C: p = 0.011) following the competition. Discussion and Conclusions: The findings in this descriptive investigation support the notion that thermography may serve as a useful tool in the acute analysis of muscle functional activation and metabolic response in professional marching athletes. Moreover, the results confirmed that the change in skin temperature is the result of a variation in acute metabolic and functional activation in the lower extremities of race walkers during competition, with infrared thermography representing an instrument capable of detecting such a change in a rapid and non-invasive manner.

Este escrito se propone pensar el papel de las imágenes y de la «invención» en el proyecto de la Unidad Popular. Si la inventiva, como forma de entender el trabajo colectivo, surge en distintas iniciativas de la UP que buscan mejorar las condiciones de vida de los trabajadores, a la vez que intentando legitimar el proyecto socialista al consolidar sus logros y su capacidad de funcionar como alternativa a los modelos de producción capitalista, la invención en tanto «combinación original de rayos imitativos» (Tarde) opera en paralelo como una forma de asociatividad inédita promovida por el proyecto popular y, en esa medida, las trayectorias y relaciones que suscita son menos lineales que lo que las imágenes, entendidas como mecanismo de captura y fijación representacional, permiten entender. Es por esto que el artículo se propone pensar otros modos de narrar la Unidad Popular en los que las imágenes funcionen como máquinas o «agenciamientos maquínicos» (Guattari), esto es, como registros de lo múltiple que emergen dentro de una formación social compleja y en constante mutación. Para ello, el texto conecta dos hitos de la UP que inscriben la invención como un eje articulador de la redefinición de lo social: la colección «Nosotros los chilenos», de Quimantú (1971-1973), y el edificio UNCTAD III (1971-1972).

Trail runners (TRs) must carry an extra load of equipment, food (bars and gels) and liquids, to delay the anticipation of fatigue and dehydration during their competitions. Therefore, we aimed to evaluate how an extra load can influence the metabolic level. Thirteen well-trained trail runners performed a randomized crossover study (total n = 39), completing three treadmill running sessions with a weighted vest of 0%, 5% and 10% of their body mass during a combined test (rectangular test + ramp test). In addition, biomarkers of oxygen metabolism, acid–base and electrolyte status pre-, during and post-test, as well as the rectangular from capillary blood of the finger and time to exhaustion, were analyzed. Repeated-measures ANOVA showed no significant difference between conditions for any of the analyzed biomarkers of blood gas. However, one-way ANOVA showed a significant difference in trial duration between conditions (p ≤ 0.001). Tukey’s post hoc analysis observed a significant decrease in time to exhaustion in the weighted vest of 10% compared to 0% (p ≤ 0.001) and 5% (p ≤ 0.01) and 5% compared to 0% (p = 0.030). In addition, repeated-measures ANOVA detected a significant difference in pH in the group x time interaction (p = 0.035). Our results show that increasing the weighted vest (5% and 10%) anticipates fatigue in runners trained in TR. In addition, increasing the load decreased pH by a smaller magnitude at 10% compared to 0% and 5% at the end of the exercise protocol.

During some trail running races, athletes have to carry hydration support, food, and technical and safety equipment, which generates an additional load that must be mobilized during the race. The aim of the present study was to determine the physiological responses to overload running and the effect they may have on metabolic zones. Seventeen well-trained male trail runners (n = 17) completed three maximal treadmill tests with weighted vests at 0%, 5%, and 10% of their body mass (L0, L5, and L10). Their gas exchange was monitored to assess their ventilatory thresholds 1 (VT1) and 2 (VT2), maximal fat oxidation zone (FatMax), and peak oxygen consumption (VO2peak). Their heart rate (HR), power, and velocity (V) were tracked to compare their behavior. One-way ANOVA showed significant differences in the V (p < 0.001; ηp2 = 0.4620) as a limitation for reaching the peak velocity (Vpeak), with a significant decrease in the Vpeak with the L10 compared to the L0 (p = 0.002) and L5 (p = 0.004). In addition, one-way ANOVA showed significant differences in the peak absolute power (p < 0.001; ηp2 = 0.468) among the groups, detecting higher power production between the L10 and L0 (p < 0.001) and between the L10 and L5 (p = 0.015). Loads higher than L5 could generated important physiological and mechanical modifications, while a load of L5 managed to maintain the working conditions without overloading. These insights shed light on nuanced strategies for optimizing performance and endurance, offering valuable considerations for athletes seeking to enhance their training regimens during overload conditions.

To evaluate the antioxidant activity and oxidative damage by relaxing, wounding, and seeding of a saibo of different origin on Pteria sterna hosts, five oyster treatments were included: (1) relaxed (REL) but neither wounded nor seeded; (2) relaxed and wounded (WOU) but not seeded; (3) relaxed, wounded, and seeded with an allograft (ALL); (4) relaxed, wounded, and seeded with an autograft (AUT); and (5) unrelaxed, unwounded, and unseeded as control (CTR). Superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), and thiobarbituric acid (TBARS) activity were quantified between 3 and 24 h post-seeding. Compared to the CTR oysters, which did not suffer oxidative stress, SOD activity significantly decreased in the gonad and digestive gland in all treatments and decreased in mantle tissue in AUT oysters; this indicates that the entire process of preparing oysters for pearl culture (relaxing, wounding, and seeding) generates oxidative stress in the host. CAT was not a sensitive enzyme for measuring the short-term response of oysters to the wounding–seeding processes but rather a more prolonged or chronic stress. Similar to SOD, the lowest GPx and TBARS activity in seeded oysters evidenced their susceptibility to oxidative stress and damage, particularly in the WOU treatment. Evidence from this study indicates that SOD is a more sensitive enzyme for measuring the short-term response of the host oyster to the wounding and seeding of a saibo. It is also clear that the host undergoes stress at all stages of the pearl culture process, mostly during gonad wounding and regardless of the origin of saibo.

Grid-connected power inverters are indispensable in modern electrical systems, playing a pivotal role in enhancing the integration of renewable energies into power grids. Their significance, primarily when functioning as grid-forming inverters, extends to maintaining the grid’s inertia and strength—a distinct advancement over traditional grid-following operations. As grid-forming inverters, these devices emulate the characteristics of synchronous generators and can act as robust voltage sources, providing essential ancillary services. This behavior is particularly relevant when integrating energy storage systems on the converters’ direct current side. Among the various inverter topologies, the current source inverter (CSI) has emerged as a promising yet underexplored alternative for grid-forming applications. CSIs, when paired with their AC output filters, can effectively operate as voltage sources, utilizing control strategies that facilitate the integration of renewable energies into the electrical system. Their design inherently manages output current fluctuations, reducing the need for restrictive current limitations or additional protective measures. This paper examines the operational region of CSIs, obtained through detailed modeling, to explore their advantages, challenges, and potential for enhancing grid-connected systems. Analyzing the operating region from the converter model verifies the limits of where the converter can operate in a plane of active and reactive powers. For a small prototype model operating with 7 amperes in DC and 120 V in AC, it is possible to supply or absorb active power exceeding 1000 W and manage maximum reactive power values around 500 VAr, as determined by its operating region. Simulations also confirm that small changes in the control reference, as little as 5%, towards the region’s right limits cause significant oscillations in the dynamic control responses. This research aims to deepen our understanding of CSIs’ operational capabilities and highlight their unique benefits in advancing grid-connected systems and promoting the integration of renewable energy using this technology.

This scoping review aimed to characterize and quantify the external load demands of professional female team sports, in terms of total distance [TD], moderate-speed [MSR] and high-speed running [HSR], sprint, accelerations [ACC], and decelerations [DEC]. A search was conducted in PubMed, Scopus, and Web of Science until 15/04/2023. The Risk of Bias Assessment Tool for Nonrandomized Studies (RoBANS) was used. Eighty-six articles were eligible for inclusion in this review, with 40 in soccer, 23 in rugby (6 rugby union, 3 rugby league, and 14 rugby sevens), 8 in field hockey, 8 in basketball, 6 in handball, and 1 in futsal. Soccer is the most investigated sport, and players perform ~9500 m TD, of which ~580 m is performed in HSR, and with a great number of ACC, DEC, and sprints. Rugby league and union players cover a greater distance (~5450 m) when compared to rugby sevens (~1550 m); however, rugby sevens is more demanding in terms of high-intensity actions. Field hockey players perform ~5400 m TD with high-intensity and sprint actions. Women’s indoor sports are less studied, and basketball players cover ~5300 m TD, of which 7% is performed in MSR. Handball players perform ~3500 m TD and cover ~423 m in MSR and ~141 m in HSR, and futsal players perform ~5 m×min −1 in HSR and they do a great number of high-intensity activities (HSR, ACC, and DEC). Considering the high physical demands experienced by female athletes, professionals could use the present results for training and return to competition schedules.

Abstract The emergence and reemergence of mosquito-borne diseases in Brazil such as yellow fever, zika, chikungunya, and dengue have had serious impacts on public health. Concerns have been raised due to the rapid dissemination of the chikungunya virus across the country since its first detection in 2014 in Northeast Brazil. In this work, we carried out on-site training activities in genomic surveillance in partnership with the National Network of Public Health Laboratories that have led to the generation of 422 chikungunya virus genomes from 12 Brazilian states over the past two years (2021–2022), a period that has seen more than 312 thousand chikungunya fever cases reported in the country. These genomes increased the amount of available data and allowed a more comprehensive characterization of the dispersal dynamics of the chikungunya virus East-Central-South-African lineage in Brazil. Tree branching patterns revealed the emergence and expansion of two distinct subclades. Phylogeographic analysis indicated that the northeast region has been the leading hub of virus spread towards other regions. Increased frequency of C > T transitions among the new genomes suggested that host restriction factors from the immune system such as ADAR and AID/APOBEC deaminases might be driving the genetic diversity of the chikungunya virus in Brazil.

Abstract Background A processing methodology of raw starch extraction from avocado seeds (ASs) and a sequential hydrolysis and fermentation bioprocess in just a few steps was successfully obtained for the bioethanol production by a single yeast Saccharomyces cerevisiae strain and this research was also to investigate the optimum conditions for the pretreatment of biomass and technical procedures for the production of bioethanol. It successfully resulted in high yields and productivity of all the experiments from the laboratory scale and the pilot plant. Ethanol yields from pretreated starch are comparable with those in commercial industries that use molasses and hydrolyzed starch as raw materials. Results Before the pilot-scale bioethanol production, studies of starch extraction and dilute sulfuric acid-based pretreatment was carefully conducted. The amount of starch extracted from dry and fresh avocado seed was 16.85 g ± 0.34 g and 29.79 ± 3.18 g of dry starch, representing a yield of ∼17% and 30%, respectively. After a dilute sulfuric acid pretreatment of starch, the released reducing sugars (RRS) were obtained and the hydrolysate slurries containing glucose (109.79 ± 1.14 g/L), xylose (0.99 ± 0.06 g/L), and arabinose (0.38 ± 0.01 g/L). The efficiency of total sugar conversion was 73.40%, with a productivity of 9.26 g/L/h. The ethanol fermentation in a 125 mL flask fermenter showed that Saccharomyces cerevisiae (Fali, active dry yeast) produced the maximum ethanol concentration, p max at 49.05 g/L (6.22% v/v) with a yield coefficient, Y p/s of 0.44 gEthanol/gGlucose, a productivity or production rate, r p at 2.01 g/L/h and an efficiency, Ef of 85.37%. The pilot scale experiments of the ethanol fermentation using the 40-L fermenter were also successfully achieved with essentially good results. The values of p max, Y p/s , r p , and Ef of the 40-L scale were at 50.94 g/L (6.46% v/v), 0.45 gEthanol/gGlucose, 2.11 g/L/h, and 88.74%, respectively. Because of using raw starch, major by-products, i.e., acetic acid in the two scales were very low, in ranges of 0.88–2.45 g/L, and lactic acid was not produced, which are less than those values in the industries. Conclusions The sequential hydrolysis and fermentation process of two scales for ethanol production using the combination of hydrolysis by utilizing dilute sulfuric acid-based pretreatment and fermentation by a single yeast Saccharomyces cerevisiae strain is practicable and feasible for realistic and effective scale-up strategies of bioethanol production from the starch of avocado seeds.

Abalone pearl farming is an aquaculture activity with great potential for future growth and diversification of the global pearl sector, which has been dominated for decades by pearls of marine pearl oysters of the genus Pinctada. Despite this potential, there is a significant lack of knowledge regarding methods for nucleus (bead) seeding and the process of pearl formation in abalone, compared to the well-studied pearl oyster model. To address this problem, this review first compares the main anatomical differences and similarities between both groups of mollusks, particularly those associated with the structure and role of mantle tissue on the synthesis of CaCO3 compounds that integrate the inner nacreous layer. The iridescence (orient) and color properties of nacre in a cultured pearl is particularly related to the microstructure of aragonite crystals during biomineralization in each mollusk group. We then analyze the advantages and disadvantages of the adoption in abalone of methods originally developed for pearl oysters to produce bead-seeded pearls and half-pearls (or mabé pearls). For the red abalone Haliotis rufescens in particular, updated information on commercial production of both bead-seeded and mabé pearls is provided as a case of success for Latin America (Chile). Finally, the current status and future prospects for abalone pearl farming are discussed, assessing technology gaps and challenges to be addressed for the pearl sector to achieve its full potential, particularly for Latin America.

IntroductionThe aims of this study were to compare several countermovement jump (CMJ) kinetic variables between professional (PRO) and semi-professional (SEMI-PRO) futsal players and examine the differences amongst playing positions.MethodsCMJ performance from 56 male futsal players (25.2 ± 4.8 years; weight: 74.4 ± 6.4 kg) was analysed. Players were separated into PRO (n = 29; 27.0 ± 4.4 years; 75.4 ± 6.0 kg) and SEMI-PRO (n = 27; 22.7 ± 4.3 years; 73.1 ± 6.8 kg), and according to playing position: defenders (n = 16; 25.4 ± 3.7 years; 75.2 ± 6.0 kg), wingers (n = 26; 23.5 ± 4.5 years; 72.0 ± 6.9 kg), and pivots (n = 14; 28.0 ± 5.6 years; 77.8 ± 4.3 kg). Linear mixed models and effect sizes were used for the analyses based on the mean of two jumps for each variable.ResultsPRO players presented a deeper center of mass (COM) displacement (p = 0.002, ES = 0.83), greater eccentric (Ecc) absolute (p = 0.019, ES = 0.61) and relative peak power (p = 0.046, ES = 0.52), and achieved greater Ecc peak velocities (p = 0.004, ES = 0.76) when compared to SEMI-PRO. Non-significant and trivial-to-small differences were observed in all the other CMJ variables according to the competitive level and playing position.DiscussionEcc capabilities (i.e., deeper COM displacement, greater Ecc absolute and relative peak power, and peak velocity) during vertical jump seem to differentiate PRO and SEMI-PRO players. However, CMJ variables do not discriminate amongst playing positions in futsal players.

IntroductionMalaria parasites increasingly develop resistance to all drugs available in the market, hampering the goal of reducing malaria burden.MethodsHerein, we evaluated the impact of a single-nucleotide variant, E738K, present in the 26S proteasome regulatory subunit rpn2 gene, identified in Plasmodium chabaudi resistant parasites. Plasmids carrying a functional rpn2 interspecies chimeric gene with 5’ recombination region from P. falciparum and 3’ from P. chabaudi were constructed and transfected into Dd2 P. falciparum parasites.Results and discussionThe 738K variant parasite line presented increased parasite survival when subjected to dihydroartemisinin (DHA), as well as increased chymotrypsin-like activity and decreased accumulation of polyubiquitinated proteins. We thus conclude that the ubiquitin-proteasome pathway, including the 738K variant, play an important role in parasite response to DHA, being the first report of a mutation in a potential DHA drug target enhancing parasite survival and contributing to a significant advance in the understanding the biology of artemisinin resistance.

Context: The majority of people with serious health-related suffering in low- and middle-income countries lack access to palliative care (PC). Increased access to PC education is greatly needed. Objectives: This paper describes the process to adapt an advanced PC training course for a Chilean context. Methods: A joint team of intercultural PC educators from the US and Chile conducted a series of key informant interviews and a target audience survey to iteratively design a PC training course in Chile. Results: Eight key informant interviews identified a strong need for formal PC education pathways, confirmed the five central learning domains, and helped to identify potential course sub-topics. A target audience survey of 59 PC providers from across Chile confirmed a strong desire to participate in such a course. Conclusion: Our team of intercultural PC educators adapted an advanced PC course to the unique context of Chilean providers.

ABSTRACT Lactoferrin is a protein from breast milk involved in early-life microbiome assembly. It is believed to promote the growth of lactic acid-producing bacteria and inhibit pathogens by limiting iron availability and enabling an acidic environment in the gut. Previous reports suggest that oral administration of lactoferrin in neonates promotes the increase of gut microbiota diversity and the enrichment of beneficial microbes. However, its effect on infant gut microbiota over time has yet to be thoroughly studied to fully understand its potential as a prebiotic. Using 16s rRNA gene amplicon sequencing, we analyzed the gut microbiota composition of 60 toddlers at 12–18 months of age from Lima, Peru, who received daily oral administration of lactoferrin or placebo for 6 months since enrollment. Samples were analyzed at 0, 2, and 4 months after beginning treatment. Our results show that lactoferrin treatment does not increase gut microbiota diversity over time nor affect its bacterial composition compared to the placebo group. IMPORTANCE Previous studies have suggested that oral lactoferrin enhances diversity in the gut microbiota in infants while inhibiting the growth of opportunistic pathogens. However, the effect of lactoferrin on infant gut microbiota over time has yet to be thoroughly studied. Our study suggests that lactoferrin oral treatment in infants aged 12–18 months does not affect gut microbiome diversity and composition over time. To our knowledge, this is the first study to report the effect of lactoferrin on infant gut microbiome composition over time and helps elucidate its impact on infant health and its therapeutic potential.

A rapidly emerging area of interest in high-pressure environments is that of pupillometry, where handheld quantitative infrared pupillometers (HQIPs) are able to track psycho-physiological fatigue in a fast, objective, valid, reliable, and non-invasive manner. However, the application of HQIPs in the context of athlete monitoring is yet to be determined. Therefore, the main aim of this pilot study was to examine the potential usefulness of a HQIP to monitor game-induced fatigue inside a professional female basketball setting by determining its (1) test-retest repeatability, (2) relationship with other biomarkers of game-induced fatigue, and (3) time-course from rested to fatigued states. A non-ophthalmologic practitioner performed a standardized Pupil Light Reflex (PLR) test using a medically graded HQIP among 9 professional female basketball players (2020–2021 Euro Cup) at baseline, 24-h pre-game (GD-1), 24-h post-game (GD+1) and 48-h post-game (GD+2). This was repeated over four subsequent games, equalling a total of 351 observations per eye. Two out of seven pupillometrics displayed good ICCs (0.95–0.99) (MinD and MaxD). Strong significant relationships were found between MaxD, MinD, and all registered biomarkers of game-induced fatigue (r = 0.69–0.82, p < 0.05), as well as between CV, MCV, and cognitive, lower-extremity muscle, and physiological fatigue markers (r = 0.74–0.76, p < 0.05). Three pupillometrics were able to detect a significant difference between rested and fatigued states. In particular, PC (right) (F = 5.173, η 2 = 0.115 p = 0.028) and MCV (right) (F = 3.976, η 2 = 0.090 p = 0.049) significantly decreased from baseline to GD+2, and LAT (left) (F = 4.023, η 2 = 0.109 p = 0.009) significantly increased from GD-1 to GD+2. HQIPs have opened a new window of opportunity for monitoring game-induced fatigue in professional female basketball players. However, future research initiatives across larger and heterogenous samples, and longer investigation periods, are required to expand upon these preliminary findings.

Se evaluó el uso de las reservas energéticas de reproductores juveniles de Pteria sterna (50 mm de altura; 9 meses de edad) durante su primera madurez sexual en condiciones de laboratorio. Se empleó una dieta microalgal base (Dieta M) complementada con fécula de maíz rica en carbohidratos (dietas F1 y F2) y con microcápsulas ricas en lípidos (dietas L1 y L2), equivalente a una ración diaria del 1 y 2 % de la biomasa seca de tejidos blandos. Se hicieron muestreos a los días 0 (d0), 15 (d15) y 30 (d30) para recolectar tejidos y determinar las variaciones temporales en el índice de tejido gonadal (ITG, %), diámetro teórico de ovocitos (DT, µm) y composición bioquímica de carbohidratos, proteínas y lípidos en la gónada, glándula digestiva, músculo y manto (mg/g). Debido a la talla de los organismos y carácter protándrico de la especie, los machos dominaron sobre las hembras en todos muestreos. Al d30, los organismos maduraron en diferente proporción dependiendo del sexo y dieta, siendo el ITG significativamente mayor en machos y hembras que se alimentaron con la dieta L1, y el DT de ovocitos significativamente mayor en las hembras de la dieta L2. La concentración de carbohidratos del músculo aductor y gónada disminuyó significativamente en ambos muestreos (d15 y d30) y en casi todas las dietas (excepto F2); esto sugiere un proceso de lipogénesis a partir de este combustible en ambos tejidos, particularmente en el músculo, lo que a su vez explica el incremento en los niveles de lípidos en la gónada al d30 en las ostras de las dietas L1 y L2. El contenido de lípidos de la glándula digestiva disminuyó significativamente al d30 con casi todas las dietas (a excepción de L2), indicando que este tejido moviliza a la gónada lípidos previamente almacenados para asegurar la maduración de gametos. La dieta L2 parece la mejor alternativa, incluso combinada con la dieta F2, para la maduración sexual de P. sterna, con resultados que podrían estar asociados a su actividad gametogénica inicial protándrica.

About 6.5 million people are infected with Chagas disease (CD) globally, and WHO estimates that $ > million people worldwide suffer from ChHD. Sudden cardiac death (SCD) represents one of the leading causes of death worldwide and affects approximately 65% of ChHD patients at a rate of 24 per 1000 patient-years, much greater than the SCD rate in the general population. Its occurrence in the specific context of ChHD needs to be better exploited. This paper provides the first evidence supporting the use of machine learning (ML) methods within non-invasive tests: patients' clinical data and cardiac restitution metrics (CRM) features extracted from ECG-Holter recordings as an adjunct in the SCD risk assessment in ChHD. The feature selection (FS) flows evaluated 5 different groups of attributes formed from patients' clinical and physiological data to identify relevant attributes among 57 features reported by 315 patients at HUCFF-UFRJ. The FS flow with FS techniques (variance, ANOVA, and recursive feature elimination) and Naive Bayes (NB) model achieved the best classification performance with 90.63% recall (sensitivity) and 80.55% AUC. The initial feature set is reduced to a subset of 13 features (4 Classification; 1 Treatment; 1 CRM; and 7 Heart Tests). The proposed method represents an intelligent diagnostic support system that predicts the high risk of SCD in ChHD patients and highlights the clinical and CRM data that most strongly impact the final outcome.

Hamstring strain injuries (HSIs) are one of the most common injuries in sprint-based sports. In soccer, the ability to sprint is key, not only because of its relation to performance but also due to its possible protective effect against HSIs. Although many authors have focused on the “how”, “when”, and “what” training load should be implemented, there is a lack of practical proposals for sprint training in a high-level professional environment. The objective of this narrative review is, after a deep review of the scientific literature, to present a practical approach for sprint training, trying to answer some of the questions that most strength and conditioning coaches ask themselves when including it in soccer. Once the literature published on this topic was reviewed and combined with the practical experience of the authors, it was concluded that sprint training in soccer, although it presents an obvious need, is not something about which there is methodological unanimity. However, following the practical recommendations from this narrative review, strength and conditioning coaches can have a reference model that serves as a starting point for optimal management of the internal and external training load when they wish to introduce sprint training in the competitive microcycle in professional soccer players, with the aim of reducing HSIs.

The aims of this study were to analyze the effects of a congested period (three games in four days) on countermovement (CMJ) jump-landing metrics, heart rate variability (HRV), and total recovery quality (TQR) score in under-19 male futsal players, and to detect the differences between those who played for more minutes (HIGHMIN) and less minutes (LOWMIN). Fourteen youth futsal players (age: 17.5 ± 0.5 years; body mass: 70.2 ± 8.5 kg; height: 1.80 ± 0.1 m) participated. HRV, TQR questionnaire, and CMJ metrics (i.e., CMJ height, relative peak power (PPREL), eccentric and concentric impulse, braking time, and time to peak force) were registered. A linear mixed model and effect sizes (ESs) were used to assess the differences between groups and days. Considering the total sample, a significant decrease was found in the PPREL and TQR score (p = 0.001–0.013 and ES = 0.28–0.99) on Days 2, 3, and 4 when compared to Day 1. HIGHMIN group presented a significant decrease in PPREL on Day 3 (p = 0.004; ES: 0.62; 95% CI: 0.39–2.65) when compared to Day 1, and in the TRQ score on Day 3 (p = 0.002; ES: 1.98; 95% CI: 0.18–2.46) and 4 (p = 0.003; ES: 2.25; 95% CI: 0.52–3.38) when compared to Day 1. Non-significant differences were found for the rest of the metrics and in the group LOWMIN. In summary, neuromuscular performance (i.e., CMJ PPREL) and subjective recovery were impaired in players with higher playing minutes during a match-congested period when compared to those with less on-court time.

When playing basketball, players are required to have high explosive power, which requires the ability to move in efficient, specific, and game-specific movement patterns that combine both horizontal and vertical abilities. Differences have been seen between young male and female basketball players in this measure. The aim of this study was to examine differences in players’ unique movements by gender, age, and playing positions using a novel test for basketball players. This study included 232 young basketball players, male and female, from a range of Israeli leagues, who were divided into three categories: under-14, under-16, and under-18. Our findings showed that males presented better results than females in all age categories. Moreover, females in the under-18 category presented better results than those in the under-14 category, but not more than those in the under-16 category. Differences in playing positions were only examined between males and females in the under-18 category, where players begin to specialize in playing positions, and here, guards showed better results than forwards and centers. Our conclusions highlight the importance of including unique, sport-specific tests in talent identification and selection processes, as these tests can provide valuable information about a player’s skill set and potential for success. The findings are presented in an achievement table of the expected physical fitness results by age and gender for the benefit of basketball coaches and strength and conditioning coaches when assessing their players.

On behalf of the Strength and Conditioning Society (SCS) and the Hellenic Society of Biochemistry and Exercise Physiology (EEVFA), we are pleased to present the abstracts of the SCS 6th Annual Meeting and EEVFA—11th International Congress of Biochemistry and Physiology of Exercise. The event was held at the Hellenic Olympic Committee headquarters in Athens, Greece, on 19–22 October 2023, and comprised several invited sessions from international and national speakers on a variety of topics related to biochemistry and exercise physiology, strength and conditioning practices and their application to health, injury prevention and sports performance. These included strength training in high-performance sports, sport science and training–competition load management in elite environments, biochemistry and exercise physiology and prescription, nutrition and biomechanics, among others. The conference also included different practical workshops conducted by renowned academics and practitioners on eccentric training, change of direction ability and strength and power training in professional team-sports, and ergospirometry and exercise prescription in specific populations. Finally, the event disseminated up-to-date strength and conditioning research by providing practitioners and researchers with the opportunity to present their most recent findings. In this regard, all abstracts of the communications presented at the SCS 6th Annual Meeting—EEVFA—11th International Congress of Biochemistry and Physiology of Exercise can be found in this Conference Report.

Sprinting is a decisive action in soccer that is considerably taxing from a neuromuscular and energetic perspective. This study compared different calculation methods for the metabolic power (MP) and energy cost (EC) of sprinting using global positioning system (GPS) metrics and electromyography (EMG), with the aim of identifying potential differences in performance markers. Sixteen elite U17 male soccer players (age: 16.4 ± 0.5 years; body mass: 64.6 ± 4.4 kg; and height: 177.4 ± 4.3 cm) participated in the study and completed four different submaximal constant running efforts followed by sprinting actions while using portable GPS-IMU units and surface EMG. GPS-derived MP was determined based on GPS velocity, and the EMG-MP and EC were calculated based on individual profiles plotting the MP of the GPS and all EMG signals acquired. The goodness of fit of the linear regressions was assessed by the coefficient of determination (R2), and a repeated measures ANOVA was used to detect changes. A linear trend was found in EMG activity during submaximal speed runs (R2 = 1), but when the sprint effort was considered, the trend became exponential (R2 = 0.89). The EMG/force ratio displayed two different trends: linear up to a 30 m sprint (R2 = 0.99) and polynomial up to a 50 m sprint (R2 = 0.96). Statistically significant differences between the GPS and EMG were observed for MP splits at 0–5 m, 5–10 m, 25–30 m, 30–35 m, and 35–40 m and for EC splits at 5–10 m, 25–30 m, 30–35 m, and 35–40 m (p ≤ 0.05). Therefore, the determination of the MP and EC based on GPS technology underestimated the neuromuscular and metabolic engagement during the sprinting efforts. Thus, the EMG-derived method seems to be more accurate for calculating the MP and EC in this type of action.

The supplementation of live yeast in pig diets is common in the post-weaning phase due to its prebiotic and probiotic effects, but little is known regarding the potential of feeding live yeast to gestating or lactating sows for transferring such benefits to their offspring through maternal programming. The objective of this study was to investigate the effects of live yeast supplementation in sow diets during late gestation and lactation on their reproductive performance and its impact on offspring performance and gut microbiomes in the post-weaning period. Three dietary treatments were imposed on 92 mixed-parity sows during late gestation and lactation based upon the inclusion level of live yeast in corn/soybean meal-based diets: Control (0% yeast), Low (0.1% yeast), and High (0.5% yeast). Nursery pigs in the Low group displayed the highest feed intake in the post-weaning period and greater total gain and average daily gain in comparison to pigs in the High group. The gut microbiomes of nursery pigs differed in composition according to maternal dietary treatment groups at days 4 and 28 post weaning, highlighting higher abundances of bacterial genera typically associated with fermentation roles in the gut microbiomes of offspring of yeast-fed sows. These results indicate that the supplementation of live yeast in sow diets, depending on the inclusion level, may result in beneficial performance and specific microbiome traits for their offspring in the post-weaning period.

Hamstring injuries in soccer continue to be a challenge for professionals who work with soccer players daily. Although its origin is multifactorial, the proper management of neuromuscular fatigue during the training microcycle is a very important factor to consider. There are no clear guidelines regarding the weekly distribution of certain exercises that demand the hamstrings. The main objective of this study was to describe the usual training practices of professional European soccer teams. An international observational survey design was applied to some of the strength and conditioning coaches of professional soccer teams. The survey included different neuromuscular demanding exercises for the hamstrings. For each exercise, the strength and conditioning coaches had to respond in relation to their frequency of use and timepoint depending on the day of the weekly microcycle. Although there is no strong consensus in this regard, there does seem to be a trend when applying certain exercises, especially on the days matchday-4 and matchday-3.

In hemophilia, the unmet needs regarding adherence to prophylaxis and lack of effective long-term prophylaxis regimens, especially in patients with inhibitors, led to the production of emicizumab, the first non-factor medicine for subcutaneous administration in patients with severe and moderate hemophilia A with or without factor VIII inhibitors. This review describes the research steps behind the development of this game-changing medication as well as its success in the prophylaxis of bleeding episodes, as witnessed by the results of pivotal clinical trials but also by real-life use in the frame of a still expanding global market. We also discuss potential and actual adverse events and the nuances related to clinical use, such as laboratory monitoring, development of neutralizing antidrug antibodies, risk of thrombosis/hypercoagulability and role in the management of surgical operations. The potential of emicizumab to prevent bleeding in other congenital and acquired coagulation disorders is also outlined.

While external stimulation can reliably trigger neuronal activity, cerebral processes can operate independently from the environment. In this study, we conceptualize autogenous cerebral processes (ACPs) as intrinsic operations of the brain that exist on multiple scales and can influence or shape stimulus responses, behavior, homeostasis, and the physiological state of an organism. We further propose that the field should consider exploring to what extent perception, arousal, behavior, or movement, as well as other cognitive functions previously investigated mainly regarding their stimulus–response dynamics, are ACP-driven.

Abstract Background Phenylketonuria (PKU) is an autosomal recessive disease resulting from a deficiency of the enzyme phenylalanine hydroxylase (PAH). Hyperphenylalaninemias (HPA) due to PAH deficiency are accompanied by a wide variety of clinical, biochemical, and molecular features. To identify and characterize pathogenic variants in the PAH gene and establish a correlation between genotype and biochemical phenotype in patients with PKU from state of Pará in the North Region of Brazil. Methods All 13 exons of the PAH gene from 32 patients (21 PKU and 11 non‐PKU HPA) were amplified by PCR and submitted to DNA sequencing (Sanger). Biochemical data were obtained from the patients' medical records. Results Molecular analysis identified 17 pathogenic variants and 3 nonpathogenic variants. The most frequent pathogenic variants were IVS10‐11G>A (7.9%), p. Arg261Gln (7.9%), p. Val388Met (6.3%) and p. Ile65Thr (4.7%). Was observed correlations and inconsistencies between genotype and biochemical phenotype. Conclusion In PKU patients from state of Pará, North Region of Brazil, a heterogeneous mutation spectrum was revealed, in which the most frequent mutations are variants commonly observed in other Brazilian studies and in the region of the Iberian Peninsula.

IntroductionThis study aimed to investigate the physical and individual technical-tactical performance of elite futsal players.MethodsThirteen National Team futsal players (27.4 ± 4.7 years, 70.3 ± 7.6 kg, 176.3 ± 5.5 cm) competed in 15 official matches in three distinct competitions. Session rate of perceived exertion (sRPE) and player load (PL) were measured for assessing physical internal and external load, respectively. Individual tactical actions were analyzed using Instat Scout®, considering variables such as fouls, shots, shots on target, goals, successful dribbles, passes, successful passes, ball losses, ball recoveries, and challenges won.ResultsThe cluster analysis classified the players into two groups: “high-involvement” (HIGH) and “low-involvement” (LOW), based on their average effective playing time, sRPE, and PL. A linear mixed model was used to examine the differences in external and internal load and technical-tactical variables between the two groups, accounting for individual repeated measures. Considering absolute values, HIGH players presented higher sRPE, RPE, total PL, shots, goals, passes, successful passes, ball losses and recoveries, and challenges won (p = 0.001–0.039; ES = 0.43–2.48), when compared to their LOW counterparts. However, when analyzed relative to effective time, LOW players reached higher total PL and shots per minute (p = 0.001–0.012; ES = 0.39–0.93) when compared to HIGH players. Non-significant differences were found for the rest of the variables.DiscussionIn summary, these results emphasize the importance of normalizing internal and external load variables and technical-tactical performance variables per effective playing time in futsal. The current findings indicate that players with lower involvement can present the same or even higher physical and technical-tactical performance compared to teammates with more playing time.

Background: The development of adipose tissue during adolescence may provide valuable insights into obesity-associated diseases. We propose an automated convolutional neural network (CNN) approach using Dixon-based magnetic resonance imaging (MRI) to quantity abdominal subcutaneous adipose tissue (SAT) and visceral adipose tissue (VAT) in children and adolescents. Methods: 474 abdominal Dixon MRI scans of 136 young healthy volunteers (aged 8–18) were included in this study. For each scan, an axial fat-only Dixon image located at the L2–L3 disc space and another image at the L4–L5 disc space were selected for quantification. For each image, an outer and an inner region around the abdomen wall, as well as SAT and VAT pixel masks, were generated by expert readers as reference standards. A standard U-Net CNN architecture was then used to train two models: one for region segmentation and one for fat pixel classification. The performance was evaluated using the dice similarity coefficient (DSC) with fivefold cross-validation, and by Pearson correlation and the Student’s t-test against the reference standards. Results: For the DSC results, means and standard deviations of the outer region, inner region, SAT, and VAT comparisons were 0.974 ± 0.026, 0.997 ± 0.003, 0.981 ± 0.025, and 0.932 ± 0.047, respectively. Pearson coefficients were 1.000 for both outer and inner regions, and 1.000 and 0.982 for SAT and VAT comparisons, respectively (all p = NS). Conclusion: These results show that our method not only provides excellent agreement with the reference SAT and VAT measurements, but also accurate abdominal wall region segmentation. The proposed combined region- and pixel-based CNN approach provides automated abdominal wall segmentation as well as SAT and VAT quantification with Dixon MRI and enables objective longitudinal assessment of adipose tissues in children during adolescence.

Cetacean morbillivirus (CeMV) causes illness and death in cetaceans worldwide; the CeMV strains circulating in the Southern Hemisphere are poorly known. We detected a pilot whale CeMV strain in 3 short-finned pilot whales (Globicephala macrorhynchus) stranded in Brazil during July–October 2020. Our results confirm this virus circulates in this species.

Abstract The accumulation of α-synuclein (α-syn) in intracellular formations known as Lewy bodies (LBs) is associated with several neurodegenerative diseases including Parkinson’s disease and Lewy Body Dementia. There is still limited understanding of how α-syn and LB formation is associated with cellular dysfunction and degeneration in these diseases. To examine the clearance and production dynamics of α-syn we transduced organotypic murine brain slice cultures (BSCs) with recombinant adeno-associated viruses (rAAVs) to express Dendra2-tagged human wild-type (WT) and mutant A53T α-syn, with and without the addition of exogenous α-syn fibrillar seeds and tracked them over several weeks in culture using optical pulse labeling. We found that neurons expressing WT or mutant A53T human α-syn show similar rates of α-syn turnover even when insoluble, phosphorylated Ser129 α-syn has accumulated. Taken together, this data reveals α-syn aggregation and overexpression, pSer129 α-syn, nor the A53T mutation affect α-syn dynamics in this system. Prion-type seeding with exogenous α-syn fibrils significantly slows α-syn turnover, in the absence of toxicity but is associated with the accumulation of anti-p62 immunoreactivity and Thiazin Red positivity. Prion-type induction of α-syn aggregation points towards a potential protein clearance deficit in the presence of fibrillar seeds and the ease of this system to explore precise mechanisms underlying these processes. This system facilitates the exploration of α-syn protein dynamics over long-term culture periods. This platform can further be exploited to provide mechanistic insight on what drives this slowing of α-syn turnover and how therapeutics, other genes or different α-syn mutations may affect α-syn protein dynamics.
