Strong risk factors for developing asthma are genetic predisposition and environmental exposure to inhaled substances that may provoke allergic reactions. Asthma guidelines recommend identifying causal or trigger allergens with specific IgE (sIgE) testing after a diagnosis of asthma has been made. Allergy testing with sIgE targets subpopulations of patients considered at high risk, such as those with frequent exacerbations, emergency visits or hospitalizations, or uncontrolled symptoms. Specific recommendations apply to preschool children, school-age children, patients with persistent or difficult-to-control asthma, patients needing oral corticosteroids or high-dose inhaled steroids, patients seeking understanding and guidance about their disease, and candidates for advanced therapies (biologics, allergen immunotherapy). Allergen skin testing is common in specialized settings but less available in primary care. Blood tests for total and sIgE are accessible and yield quantifiable results for tested allergens, useful for detecting sensitization. Results are interpreted in the context of the patient’s clinical presentation, age, and relevant allergen exposures. Incorporating sIgE testing into asthma management adds objective information to identify specific allergies and can guide personalized treatment plans, which reinforce patient-doctor communication. Test results can also be used to predict exacerbations and response to therapies. Additional diagnostic information can be gleaned from (i) eosinophil count ≥ 300 μL, which significantly increases the odds of having exacerbations, and emerging eosinophil biomarkers (eg, eosinophil-derived neurotoxin), which can be measured in plasma or serum samples, and (ii) fractional exhaled nitric oxide (FeNO), with values ≥ 25 ppb regarded as the cutoff for diagnosis, evaluating inhaled corticosteroid response, and of probable response to anti-IgE, anti-IL4 and anti-IL5 receptor biologics. 5. Host age influence on the intensity of experimental Trichuris suis infection in pigs

Abstract

The impact of age-related resistance on the regulation of population dynamics of adult Trichuris suis was investigated in an experimental pig model. Helminth-naive pigs varying in age from five weeks to four years were infected with T. suis to determine susceptibility to infection. Sows had a significantly lower establishment of adult T. suis worms compared with weaner pigs. Adult worm populations were highly overdispersed in both sows and grower pigs contrasted by a more even distribution among weaner pigs. Sows had significantly lower worm fecundities compared to weaner and grower pigs; T. suis from grower pigs, in turn, had reduced fecundity compared to worms in weaner pigs. In conclusion, we provide the first controlled experimental evidence that age-related resistance to T. suis occurs in pigs.

6. Experimental infection of pigs with three dose levels of Trichuris suis

Abstract

The objective of the study was to follow the course of Trichuris suisinfection in pigs given infective eggs at low (400 eggs), medium (4,000 eggs) and high inoculation dose (40,000 eggs), respectively. Interestingly, despite a 100-fold difference in dose level no significant difference was found in either blood parameters, total faecal egg excretion, fecundity or worm burdens at necropsy 12 weeks post inoculation. The highest and lowest median faecal egg output was found in the medium and high dose group, respectively. With increasing dose level, worm size, establishment and prevalence of T. suis positive pigs decreased while worms were dislocated aborally. In addition there was a highly significant correlation between female worm burden and faecal egg excretion.

Abstract

This tutorial aims at giving an introductory treatment of the circuit analysis of superconducting qubits, i.e., two-level systems in superconducting circuits. It also touches upon couplings between such qubits and how microwave driving and these couplings can be used for single- and two-qubit gates, as well as how to include noise when calculating the dynamics of the system. We also discuss higher-dimensional superconducting qudits. The tutorial is intended for new researchers with limited or no experience with the field but should be accessible to anyone with a bachelor's degree in physics. The tutorial introduces the basic methods used in quantum circuit analysis, starting from a circuit diagram and ending with a quantized Hamiltonian, that may be truncated to the lowest levels. We provide examples of all the basic techniques throughout the discussion, while in the last part of the tutorial we discuss several of the most commonly used circuits for quantum-information applications. This includes both worked examples of single qubits and examples of how to analyze the coupling methods that allow multiqubit operations. Abstract

Physical habitat is important in determining the carrying capacity of juvenile brown trout, and within freshwater management. Summer daytime physical habitat selection for the parr lifestage (7–20 cm) juvenile brown trout (Salmo trutta) was assessed in 6 small lowland streams. Habitat preference was determined for the four variables; water velocity, water depth, substrate and cover, and the preferences for physical habitat selection were expressed in terms of habitat suitability indices (HSI’s). The statistical confidence of HSI’s was evaluated using power analysis. It was found that a minimum of 22 fish observations was needed to have statistical confidence in the HSIs for water depth, and a minimum of 92 fish observations for water velocity during daytime summer conditions. Generally parr were utilising the deeper habitats, indicating preference for deeper water. Cover was also being selected for at all sites, but selection was inconsistent among sites for the variables substrate and velocity. The results indicate that during daytime summer conditions water depth is a significant variable for parr habitat selection in these small lowland streams, with cover also being important. Therefore, daytime refugia may be a critical limiting factor for parr in small lowland streams, and important for stream management actions under the Water Framework Directive.

Abstract

The diel partitioning of juvenile brown trout Salmo trutta foraging behaviour is controlled by a number of factors including predation risk, competition, temperature and food availability. The present study uses PIT-tagging and visual observation to asses the use of shelter and foraging behaviour of Danish wild juvenile brown trout (13.5–15.6 cm). The experiment was conducted in a fluvarium and the fish were fed to satiation. It was hypothesised that food satiation would promote nocturnal foraging and increase daytime shelter use. Our results showed a significant difference in diel shelter use between day and night with a significant increase in shelter use during daytime conditions. Visual observations showed a significant preference for nocturnal feeding. Together with the significantly reduced shelter use during the night, our results support the hypothesis that young stream living trout only feed during the day to the extent needed to sustain growth.

Abstract

LOFT (Large Observatory For x-ray Timing) is one of the ESA M3 missions selected within the Cosmic Vision program in 2011 to carry out an assessment phase study and compete for a launch opportunity in 2022-2024. The phase-A studies of all M3 missions were completed at the end of 2013. LOFT is designed to carry on-board two instruments with sensitivity in the 2-50 keV range: a 10 m 2 class Large Area Detector (LAD) with a <1{\deg} collimated FoV and a wide field monitor (WFM) making use of coded masks and providing an instantaneous coverage of more than 1/3 of the sky. The prime goal of the WFM will be to detect transient sources to be observed by the LAD. However, thanks to its unique combination of a wide field of view (FoV) and energy resolution (better than 500 eV), the WFM will be also an excellent monitoring instrument to study the long term variability of many classes of X-ray sources. The WFM consists of 10 independent and identical coded mask cameras arranged in 5 pairs to provide the desired sky coverage. We provide here an overview of the instrument design, configuration, and capabilities of the LOFT WFM. 31. Spin-orbit interaction and the 'metal-insulator' transition observed in two-dimensional hole systems

Abstract

We present calculations of the spin and phase relaxation rates in GaAs/AlGaAs $p$-type quantum wells. These rates are used to derive the temperature dependence of the weak-localization correction to the conductivity. In $p$-type quantum wells both weak localization and weak anti-localization are present due to the strong spin-orbit interaction. When determining the total conductivity correction one also have to include the term due to hole-hole interaction. The magnitude of the latter depends on the ratio between the thermal energy and the Fermi energy, $k_{\rm B}T/E_{\rm F}$ and whether the system can be considered as ballistic $(k_{\rm B}T \tau_{\rm tr} / \hbar>1)$ or diffusive ($k_{\rm B}T \tau_{\rm tr}/\hbar<1$). We argue that due to the relatively low Fermi energy and the moderate mobilities, in the $p$-type systems in question, the conductivity correction arising from hole-hole interactions is negligible at the highest temperatures accessible in the experiments. Hence the 'metal-insulator' transition observed at these relatively high temperatures could be caused by interference effects. We compare our calculations of the weak anti-localization correction with the experimental results from different independent groups with special emphasis on the experiments by Simmons et al. Abstract

We have experimentally investigated the magnetisation of a mesoscopic aluminum loop at temperatures well below the superconducting transition temperature $T_{c}$. The flux quantisation of the superconducting loop was investigated with a $\mu$-Hall magnetometer in magnetic field intensities between $\pm 100 {Gauss}$. The magnetic field intensity periodicity observed in the magnetization measurements is expected to take integer values of the superconducting flux quanta $\Phi_{0}=h/2e$. A closer inspection of the periodicity, however, reveal a sub flux quantum shift. This fine structure we interpret as a consequence of a so called giant vortex state nucleating towards either the inner or the outer side of the loop. Abstract

We study electron decoherence by measuring the temperature dependence of Aharonov-Bohm (AB) oscillations in quasi-1D rings, etched in a high-mobility GaAs/GaAlAs heterostructure. The oscillation amplitude is influenced both by phase-breaking and by thermal averaging. Thermal averaging is important when the temperature approaches the energy scale, on which the AB oscillations shift their phase. For the phase-breaking, it is demonstrated that the damping of the oscillation amplitude is proportional to the length of the interfering paths. For temperatures $T$ from 0.3 to 4 $\rm K$ we find the phase coherence length $\it L_{\phi}$ $\propto$ $T^{-1}$, close to what has been reported for open quantum dots. This might indicate that the $T^{-1}$ decoherence rate is a general property of open and ballistic mesoscopic systems., Comment: 5 pages, 4 figures; to appear in Phys. Rev. B

Abstract

We have for the first time experimentally investigated the weak localisation magnetoresistance in a AlGaAs/GaAs p-type quantum well. The peculiarity of such systems is that spin-orbit interaction is strong. On the theoretical side it is not possible to treat the spin-orbit interaction as a perturbation. This is in contrast to all prior investigations of weak localisation. In this letter we compare the experimental results with a newly developed diffusion theory, which explicitly describes the weak localisation regime when the spin-orbit coupling is strong. The spin relaxation rates calculated from the fitting parameters was found to agree with theoretical expectations. Abstract

We present new results of the ``0.7'' 2(e^2)/h structure or quasi plateau in some of the most strongly confined point contacts so far reported. This strong confinement is obtained by a combination of shallow etching and metal gate deposition on modulation doped GaAs/GaAlAs heterostructures. The resulting subband separations are up to 20 meV, and as a consequence the quantized conductance can be followed at temperatures up to 30 K, an order of magnitude higher than in conventional split gate devices. We observe pronounced quasi plateaus at several of the lowest conductance steps all the way from their formation around 1 K to 30 K, where the entire conductance quantization is smeared out thermally. We study the deviation of the conductance from ideal integer quantization as a function of temperature, and we find an activated behavior, exp(-T_a/T), with a density dependent activation temperature T_a of the order of 2 K. We analyze our results in terms of a simple theoretical model involving scattering against plasmons in the constriction., Comment: RevTex (4 pages) including 2 postscript figures. Abstract

We investigate experimentally and theoretically the spin-orbit effects on the weak localization in a (110) GaAs 2-dimensional electron gas (2DEG). We analyze the role of two different terms in the spin splitting of the conduction band: the Dresselhaus terms, which arise due to the lack of inversion center in the bulk GaAs, and the Rashba terms, which are caused by the asymmetry of the quantum well. It is shown that in A3B5 quantum wells the magnetoresistance due to the weak localization depends qualitatively on the orientation of the well. In particular, it is demonstrated that the (110) geometry has a distinctive feature that in the absence of the Rashba terms the ``antilocalization'' effect, i.e. the positive magnetoresistance, does not exist. Abstract

Climate change, including both increasing temperatures and changing snow regimes, is progressing rapidly in the Arctic, leading to changes in plant phenology and in the seasonal patterns of plant properties, such as tissue nitrogen (N) content and community aboveground biomass. However, significant knowledge gaps remain over how these seasonal patterns vary among Arctic plant functional groups (i.e., shrubs, grasses, and forbs) and across large geographical areas. We used three years of in situ field vegetation sampling from an 80,000-km² area in Arctic Alaska, remotely sensed vegetation data (daily normalized difference vegetation index [NDVI]), and modeled output of snow-free date to determine and model the seasonal trends and primary controls on leaf percent nitrogen and biomass (in grams per square meter) among Arctic vegetation functional groups. We determined relative vegetation phenology stage at a 500-m spatial scale resolution, defined as the number of days between the date of the seasonal maximum NDVI and the vegetation field sampling date, and relative snow phenology stage (90-m spatial scale) was determined as the number of days between the date of snow-free ground and the sampling date. Models including relative phenology stage were particularly important for explaining seasonal variability of %N in shrubs, graminoids, and forbs. Similarly, vegetation and snow phenology stages were also important for modeling seasonal biomass of shrubs and graminoids; however, for all functional groups, the models explained only a small amount of seasonal variability in biomass. Relative phenology stage was a stronger predictor of %N and biomass than geographic position, indicating that localized controls on phenology, acting at spatial scales of 500 m and smaller, are critical to understanding %N and biomass.

Abstract

Agricultural robots present an opportunity to strengthen agrifood systems by addressing labour shortages and reducing CO2 emissions. This study proposes a method to investigate the potential use of an agricultural robot (AGR) based on a case study on a typical high-tech Dutch farm that implements both an AGR and a conventional tractor (TRC) on a single farm of 200 ha in the region of Oldambt. Five crops were chosen. Price of labor and fuel as well as time needed for remote supervision of AGR were found to be critical factors determining the benefit and use of the AGR.

Abstract

Due to increasing challenges of labour scarcity and cost, autonomous field machines have recently gained attention and some application in agriculture. Field crop robots are considered to address economic, environmental and social sustainability. Based on a survey of farmers from four case areas in Europe, this study identifies major drivers of demand for robotic solutions from farmers’ point of view. The main drivers are found to be: to save labour cost, intention to reduce soil compaction and improve efficiency in input use. Expectations about likely impact of robots (e.g. to reduce amount of chemical input use including fuel and pesticide, increase worker safety, profit, etc.) could also be important drivers of interest in robotic applications. Therefore, labour-saving, lighter, and affordable robots need to be made available and accessible to farmers. Further study on the relative importance of the identified drivers, possibly disaggregated by operation type, crop and other geographical factors is needed.

49. Revealing population heterogeneity in vesicle-based nanomedicines using automated, single particle Raman analysis

Abstract

The intrinsic heterogeneity of many nanoformulations is currently challenging to characterise on both the single particle and population level. Therefore there is great opportunity to develop new techniques to describe and understand nanomedicine heterogeneity, which will aid translation to the clinic by informing manufacturing quality control, characterisation for regulatory bodies, and connecting nanoformulation properties to clinical outcomes to enable rational design. Here, we present an analytical technique to provide such information, whilst measuring the nanocarrier and cargo simultaneously with label-free, non-destructive single particle automated Raman trapping analysis (SPARTA). We first synthesised a library of model compounds covering a range of hydrophilicities and providing distinct Raman signals. These compounds were then loaded into model nanovesicles (polymersomes) that can load both hydrophobic and hydrophilic cargo into the membrane or core regions respectively. Using our analytical framework, we characterised the heterogeneity of the population by correlating the per particle membrane and cargo signals. We found that core and membrane loading can be distinguished, and we detected sub-populations of highly loaded particlesin certain cases. We then confirmed suitability of our technique in liposomes, another Peer reviewed version of the manuscript published in final form in ACS Nano (2023) 2 nanovesicle class, including the commercial formulation Doxil. Our label-free analytical technique precisely determines cargo location alongside loading and release heterogeneity in nanomedicines, which could be instrumental for future quality control, regulatory body protocols and development of structure-function relationships, to bring more nanomedicines to the clinic
