We discuss recent advances towards matter-wave interference experiments with free beams of metallic and dielectric nanoparticles. They require a brilliant source, an efficient detection scheme and a coherent method to divide the de Broglie waves associated with these clusters: We describe an approach based on a magnetron sputtering source which ejects an intense cluster beam with a wide mass dispersion but a small velocity spread of 10%. The source is universal as it can be used with all conducting and many semiconducting or even insulating materials. Here we focus on metals and dielectrics with a low work function of the bulk and thus a low cluster ionization energy. This allows us to realize photoionization gratings as coherent matter-wave beam splitters and also to realize an efficient ionization detection scheme. These new methods are now combined in an upgraded Talbot-Lau interferometer with three 266 nm depletion gratings. We here describe the experimental boundary conditions and how to realize them in the lab. This next generation of near-field interferometers shall allow us to soon push the limits of matter-wave interference to masses up to 10 megadaltons., Comment: 10 pages, 5 figures

3. Assessment of the FRET-based Teen sensor to monitor ERK activation changes preceding morphological defects in a RASopathy zebrafish model and phenotypic rescue by MEK inhibitor

Abstract Background RASopathies are genetic syndromes affecting development and having variable cancer predisposition. These disorders are clinically related and are caused by germline mutations affecting key players and regulators of the RAS-MAPK signaling pathway generally leading to an upregulated ERK activity. Gain-of-function (GOF) mutations in PTPN11, encoding SHP2, a cytosolic protein tyrosine phosphatase positively controlling RAS function, underlie approximately 50% of Noonan syndromes (NS), the most common RASopathy. A different class of these activating mutations occurs as somatic events in childhood leukemias. Method Here, we evaluated the application of a FRET-based zebrafish ERK reporter, Teen, and used quantitative FRET protocols to monitor non-physiological RASopathy-associated changes in ERK activation. In a multi-level experimental workflow, we tested the suitability of the Teen reporter to detect pan-embryo ERK activity correlates of morphometric alterations driven by the NS-causing Shp2D61G allele. Results Spectral unmixing- and acceptor photobleaching (AB)-FRET analyses captured pathological ERK activity preceding the manifestation of quantifiable body axes defects, a morphological pillar used to test the strength of SHP2 GoF mutations. Last, the work shows that by multi-modal FRET analysis, we can quantitatively trace back the modulation of ERK phosphorylation obtained by low-dose MEK inhibitor treatment to early development, before the onset of morphological defects. Conclusion This work proves the usefulness of FRET imaging protocols on both live and fixed Teen ERK reporter fish to readily monitor and quantify pharmacologically- and genetically-induced ERK activity modulations in early embryos, representing a useful tool in pre-clinical applications targeting RAS-MAPK signaling.

We explore a wide range of fundamental magnetic phenomena by measuring the dephasing of matter-wave interference fringes upon application of a variable magnetic gradient. The versatility of our interferometric Stern-Gerlach technique enables us to study alkali atoms, organic radicals and fullerenes in the same device, with magnetic moments ranging from a Bohr magneton to less than a nuclear magneton. We find evidence for magnetization of a supersonic beam of organic radicals and, most notably, observe a strong magnetic response of a thermal C$_{60}$ beam consistent with high-temperature atom-like deflection of rotational magnetic moments.

Abstract Current genetic modification and phenotyping methods in teleost fish allow detailed investigation of vertebrate mechanisms of development, modeling of specific aspects of human diseases and efficient testing of drugs at an organ/organismal level in an unparalleled fast and large-scale mode. Fish-based experimental approaches have boosted the in vivo verification and implementation of scientific advances, offering the quality guaranteed by animal models that ultimately benefit human health, and are not yet fully replaceable by even the most sophisticated in vitro alternatives. Thanks to highly efficient and constantly advancing genetic engineering as well as non-invasive phenotyping methods, the small zebrafish is quickly becoming a popular alternative to large animals’ experimentation. This approach is commonly associated to invasive procedures and increased burden. Here, we present a rapid and minimally invasive method to obtain sufficient genomic material from single zebrafish embryos by simple and precise tail fin scratching that can be robustly used for at least two rounds of genotyping already from embryos within 48 h of development. The described protocol betters currently available methods (such as fin clipping), by minimizing the relative animal distress associated with biopsy at later or adult stages. It allows early selection of embryos with desired genotypes for strategizing culturing or genotype–phenotype correlation experiments, resulting in a net reduction of “surplus” animals used for mutant line generation.

7. Distinct role of AtCuAOβ- and RBOHD-driven H2O2 production in wound-induced local and systemic leaf-to-leaf and root-to-leaf stomatal closure

Polyamines (PAs) are ubiquitous low-molecular-weight aliphatic compounds present in all living organisms and essential for cell growth and differentiation. The developmentally regulated and stress-induced copper amine oxidases (CuAOs) oxidize PAs to aminoaldehydes producing hydrogen peroxide (H2O2) and ammonia. The Arabidopsis thaliana CuAOβ (AtCuAOβ) was previously reported to be involved in stomatal closure and early root protoxylem differentiation induced by the wound-signal MeJA via apoplastic H2O2 production, suggesting a role of this enzyme in water balance, by modulating xylem-dependent water supply and stomata-dependent water loss under stress conditions. Furthermore, AtCuAOβ has been shown to mediate early differentiation of root protoxylem induced by leaf wounding, which suggests a whole-plant systemic coordination of water supply and loss through stress-induced stomatal responses and root protoxylem phenotypic plasticity. Among apoplastic ROS generators, the D isoform of the respiratory burst oxidase homolog (RBOH) has been shown to be involved in stress-mediated modulation of stomatal closure as well. In the present study, the specific role of AtCuAOβ and RBOHD in local and systemic perception of leaf and root wounding that triggers stomatal closure was investigated at both injury and distal sites exploiting Atcuaoβ and rbohd insertional mutants. Data evidenced that AtCuAOβ-driven H2O2 production mediates both local and systemic leaf-to-leaf and root-to-leaf responses in relation to stomatal movement, Atcuaoβ mutants being completely unresponsive to leaf or root wounding. Instead, RBOHD-driven ROS production contributes only to systemic leaf-to-leaf and root-to-leaf stomatal closure, with rbohd mutants showing partial unresponsiveness in distal, but not local, responses. Overall, data herein reported allow us to hypothesize that RBOHD may act downstream of and cooperate with AtCuAOβ in inducing the oxidative burst that leads to systemic wound-triggered stomatal closure.

Browning reactions caused by oxidative stress occurring during postharvest procedures significantly compromise the quality of ready‐to‐eat (RTE) vegetables, negatively affecting their market value. Polyphenol oxidase (PPO) and peroxidase (POD) are two major enzymes involved in this phenomenon, as they oxidize phenolic compounds to quinones, which in turn polymerize to brown pigments. Recently, there has been an increasing interest in developing antibrowning treatments using food by‐products. Herein, the efficiency of a kiwi peel extract in reducing enzymatic browning of minimally processed lettuce (Lactuca sativa) has been investigated. PPO and POD activities showed an opposite spatial distribution within the leaf, with a higher POD activity in the midvein (MV) and in the inner lamina tissues, and a prevalence of PPO activity in the mesophyll. Considering that MV lignified tissues are those mainly affected by browning, the temporal trend of POD activity over a 20‐day storage period at 4°C was investigated. Data showed that treatment with a kiwi peel extract hinders both the increasing trend of POD activity and browning development compared to control leaves. These results could be potentially useful for the industry as they confirm that natural extracts, such as kiwi peel extract, can be valuable for extending the shelf‐life of RTE products. This guide, provided by a partisan education policy public interest group, seeks to answer the most frequently asked questions about teaching, designing, and evaluating an English immersion classroom and the research underpinnings in favor of English immersion. This guide aims to help teachers and policymakers better understand what English immersion is all about and the specific difficulties that affect English language learners (ELLs) at different grade levels, especially older students. After an introduction by Linda Chavez, the guide is divided into three parts and nine chapters. Part 1, "Why English Immersion?" has two chapters: "Is One Year Enough? Can English Learners Attain Proficiency in One-Year Immersion Programs" by Christine H. Rossell; and "What Works? What the Research Says" by Russell Gersten. Part 2, "Nuts and Bolts" has five chapters: "English Immersion for All Grades: The Bethlehem, PA English Acquisition Program" by Ann Goldberg; "Teaching Juan and Maria to Read: Reading Instruction Techniques for English Learners in Primary Grades" by Janet Siano; "Teaching English to High School Students" by Richard K. - Abstract
The history of bilingual education in Massachusetts since its first bilingual education law in 1971 is examined and compared to the experience of California, which is offered as an example of how bilingual education policy and practice may evolve in other states. It is suggested that Massachusetts has paralleled the California experience in failing to collect student performance data over the last two decades, to demonstrate good or bad results of transitional bilingual education programs. The reading test administered to third-graders across Massachusetts in 1997 and 1998 is proposed as an example of this resistance to accountability. Increased flexibility in local program choice and participation of English language learners in the assessment process is recommended for Massachusetts. (MSE)

The spring issue focuses on the evolution in the education of language minority students. Rosalie Pedalino Porter's article, "On the State of Bilingual Education 1990-1995: 'Forked Tongue' Continued," highlights the advances made by local school districts where students are being introduced to academic content learning in English at a younger age and where principles of effective instruction are being incorporated into models of bilingual education. Shereen Arraf's article, "The Bilingual and Compensatory Education Program of the Dearborn Schools, Michigan: A Model for Systematic Change and Integration of Services," depicts the grass roots movement in Michigan to demand and implement substantial reforms for more flexible and creative math, science, reading, and bilingual programs by pooling Title 1 and bilingual education resources. The fall issue's two articles feature two studies of central importance in the consideration of language education policy in the United States. Polyamines (PAs) are ubiquitous low-molecular-weight aliphatic compounds present in all living organisms and essential for cell growth and differentiation. The developmentally regulated and stress-induced copper amine oxidases (CuAOs) oxidize PAs to aminoaldehydes producing hydrogen peroxide (H2O2) and ammonia. The Arabidopsis thaliana CuAOβ (AtCuAOβ) was previously reported to be involved in stomatal closure and early root protoxylem differentiation induced by the wound-signal MeJA via apoplastic H2O2 production, suggesting a role of this enzyme in water balance, by modulating xylem-dependent water supply and stomata-dependent water loss under stress conditions. Furthermore, AtCuAOβ has been shown to mediate early differentiation of root protoxylem induced by leaf wounding, which suggests a whole-plant systemic coordination of water supply and loss through stress-induced stomatal responses and root protoxylem phenotypic plasticity. Among apoplastic ROS generators, the D isoform of the respiratory burst oxidase homolog (RBOH) has been shown to be involved in stress-mediated modulation of stomatal closure as well. In the present study, the specific role of AtCuAOβ and RBOHD in local and systemic perception of leaf and root wounding that triggers stomatal closure was investigated at both injury and distal sites exploiting Atcuaoβ and rbohd insertional mutants. Data evidenced that AtCuAOβ-driven H2O2 production mediates both local and systemic leaf-to-leaf and root-to-leaf responses in relation to stomatal movement, Atcuaoβ mutants being completely unresponsive to leaf or root wounding. Instead, RBOHD-driven ROS production contributes only to systemic leaf-to-leaf and root-to-leaf stomatal closure, with rbohd mutants showing partial unresponsiveness in distal, but not local, responses. Overall, data herein reported allow us to hypothesize that RBOHD may act downstream of and cooperate with AtCuAOβ in inducing the oxidative burst that leads to systemic wound-triggered stomatal closure.

Plant defence responses to adverse environmental conditions include different stress signalling, allowing plant acclimation and survival. Among these responses one of the most common, immediate, and effective is the modulation of the stomatal aperture, which integrates different transduction pathways involving hydrogen peroxide (H2O2), calcium (Ca2+), nitric oxide (NO), phytohormones and other signalling components. The Arabidopsis thaliana copper amine oxidases β (AtCuAOβ) encodes an apoplastic CuAO expressed in guard cells and root protoxylem tissues which oxidizes polyamines to aminoaldehydes with the production of H2O2 and ammonia. Here, its role in stomatal closure, signalled by the wound-associated phytohormone methyl-jasmonate (MeJA) was explored by pharmacological and genetic approaches. Obtained data show that AtCuAOβ tissue-specific expression is induced by MeJA, especially in stomata guard cells. Interestingly, two Atcuaoβ T-DNA insertional mutants are unresponsive to this hormone, showing a compromised MeJA-mediated stomatal closure compared to the wild-type (WT) plants. Coherently, Atcuaoβ mutants also show compromised H2O2-production in guard cells upon MeJA treatment. Furthermore, the H2O2 scavenger N,N1-dimethylthiourea (DMTU) and the CuAO-specific inhibitor 2-bromoethylamine (2-BrEtA) both reversed the MeJA-induced stomatal closure and the H2O2 production in WT plants. Our data suggest that AtCuAOβ is involved in the H2O2 production implicated in MeJA-induced stomatal closure.

“Un encuentro entre testeros” és un projecte que busca superar l'homogeneïtat dels polígons residencials dels anys 60 introduint diversitat programàtica i residencial en el barri de Ciutat Meridiana i centrant la discussió en la manca d'espais de trobada i punts d'intensitat urbana a la ciutat existent, agreujada pel fort pendent sobre la qual es construeix el barri. Per a això, es reflexiona sobre la tipologia del bloc lineal aïllat, amb l'objectiu de generar noves agrupacions de blocs que puguin passar a considerar-se com les unitats tipològiques mínimes del barri. Aquestes unitats, en vincular-se a l'espai públic existent, busquen constituir punts d'intersecció i confluència en una trama actualment estèril. La proposta busca intervenir tant en l'espai construït dins d'aquestes agrupacions, requalificant els habitatges, com en l'espai lliure entre els blocs, recuperant les àrees intersticials per al seu ús comunitari en realitzar-se una topografia artificial i generant una infraestructura d'enllaç entre cotes centralitzada entre les testeres dels blocs., “Un encuentro entre testeros” es un proyecto que busca superar la homogeneidad de los polígonos residenciales de los años 60 introduciendo diversidad programática y residencial en el barrio de Ciudad Meridiana y centrando la discusión en la falta de espacios de encuentro y puntos de intensidad urbana en la ciudad existente, agravada por la fuerte pendiente sobre la que se construye el barrio. Para ello, se reflexiona sobre la tipología del bloque lineal aislado, con el objetivo de generar nuevas agrupaciones de bloques que puedan pasar a considerarse como las unidades tipológicas mínimas del barrio. Estas nuevas unidades, al vincularse al espacio público existente, pretenden constituir puntos de intersección y confluencia en una trama actualmente estéril. La propuesta busca intervenir tanto en el espacio construido dentro de estas agrupaciones, recualificando las viviendas, como en el espacio libre entre los bloques, recuperando las áreas intersticiales para su uso comunitario al realizarse una topografía artificial y generando una infraestructura de enlace entre cotas centralizada entre los testeros de los bloques., "Un encuentro entre testeros" is a project whose aim is to overcome the homogeneity of the residential polygons of the 1960s by introducing programmatic and residential diversity in the neighborhood of Ciudad Meridiana and focusing the discussion on the lack of meeting spaces and points of urban intensity in the existing city, aggravated by the steep slope where the neighborhood is built. To this end, there is a reflection on the typology of the isolated linear block, with the intention to generate new groupings of blocks that may be considered as the minimum typological units of the neighborhood. These aggregations, connecting to the existing public space, seek to provide points of intersection and confluence along a currently sterile grid. It is presented the outcome of a clinical study conducted on children subjected to violent conditions treated with homeopathy. The approach employed was unicist, according to the parameters of the criteria supplied by Hahnemann, Kent and Elizalde. They were also through the clinical evolution, as well as the Questionnaire of Conners and the SNAP-IV, for Attention Deficit Disorder with Hyperactivity and Impulsivity (ADDHI), and the diagnostic criteria for Behavior Disorder and Oppositional Defiant Disorder (DSM-IV 312.8; 313;81)

We discuss recent advances towards matter-wave interference experiments with free beams of metallic and dielectric nanoparticles. They require a brilliant source, an efficient detection scheme and a coherent method to divide the de Broglie waves associated with these clusters: We describe an approach based on a magnetron sputtering source which ejects an intense cluster beam with a wide mass dispersion but a small velocity spread of 10%. The source is universal as it can be used with all conducting and many semiconducting or even insulating materials. Here we focus on metals and dielectrics with a low work function of the bulk and thus a low cluster ionization energy. This allows us to realize photoionization gratings as coherent matter-wave beam splitters and also to realize an efficient ionization detection scheme. These new methods are now combined in an upgraded Talbot-Lau interferometer with three 266 nm depletion gratings. We here describe the experimental boundary conditions and how to realize them in the lab. This next generation of near-field interferometers shall allow us to soon push the limits of matter-wave interference to masses up to 10 megadaltons., Comment: 10 pages, 5 figures

The authors discuss violence as a current epidemics and violent behavior in children and adolescents. They present a repertory study including the characteristic and peculiar symptoms of 12 patients, with clinical diagnoses of Attention Deficit Hyperactivity Disorder (ADHD), Conduct Disorder and Oppositional Defiant Disorder, and the symptoms corresponding to the diagnostic criteria in the DSM-IV transcribed in repertory language. The result is a materia medica for patterns of violent behavior. Sweet basil (Ocimum basilicum L.) is one of the most common aromatic herbs cultivated all over the world. It is widely used for its aromatic and culinary purposes. Consumer demand for fresh sweet basil has been on the rise and many farmers of leafy vegetables and aromatic herbs have oriented their production schemes to include basil. The genetic material is a key preharvest factor and the major determinant of variation in yield and quality of leafy vegetables. Another important preharvest factor that may modulate yield and nutritional quality of sweet basil could be the growing medium. The aim of this research was to assess in a multi-factorial approach the effects of two sweet basil cultivars (Napoletano and Genovese) and two substrates (rockwool and an innovative inert substrate, SISAL fiber) on plant growth, marketable yield, nitrogen content and nutritional quality. The plant growth parameters, yield and especially the bioactive compounds were mostly affected by the growing media and to a lesser extent by the genetic material. When averaged over growing media, the highest leaf number was observed in 'Genovese' while an opposite trend was recorded for the leaf area. Irrespective of the sweet basil cultivar, plants grown on rockwool exhibited a higher leaf area, leaf fresh weight and dry biomass compared to those grown on SISAL fiber substrate. Contrarily to crop productivity, sweet basil grown on SISAL was characterized by high levels of lipophilic antioxidant activity, total chlorophyll and carotenoids, whereas no significant effect among treatments were recorded on total phenols. Although the yield reduction with SISAL was quite significant, the basil plants grown on this innovative substrate produced leaves with high concentrations of bioactive compounds. Additional researches should focus on elucidating the Genotype × Substrate interaction in order to select the best combinations.

Since 1980, Field Epidemiology Training Programs (FETPs) have trained highly qualified field epidemiologists to work for ministries of health (MOH) around the world. However, the 2013–2015 Ebola epidemic in West Africa, which primarily affected Guinea, Liberia, and Sierra Leone, demonstrated a lack of field epidemiologists at the local levels. Trained epidemiologists at these levels could have detected the Ebola outbreak earlier. In 2015, the US Centers for Disease Control and Prevention (CDC) launched FETP-Frontline, a 3-month field training program targeting local MOH staff in 24 countries to augment local public health capacity. As of December 2016, FETP-Frontline has trained 1,354 graduates in 24 countries. FETP-Frontline enhances global health security by training local public health staff to improve surveillance quality in their jurisdictions, which can be a valuable strategy to strengthen the capacity of countries to more rapidly detect, respond to, and contain public health emergencies at the source.

The 2014–2015 epidemic of Ebola virus disease in West Africa primarily affected Guinea, Liberia, and Sierra Leone. Several countries, including Mali, Nigeria, and Senegal, experienced Ebola importations. Realizing the importance of a trained field epidemiology workforce in neighboring countries to respond to Ebola importations, the Centers for Disease Control and Prevention Field Epidemiology Training Program unit implemented the Surveillance Training for Ebola Preparedness (STEP) initiative. STEP was a mentored, competency-based initiative to rapidly build up surveillance capacity along the borders of the at-risk neighboring countries Côte d’Ivoire, Mali, Senegal, and Guinea-Bissau. The target audience was district surveillance officers. STEP was delivered to 185 participants from 72 health units (districts or regions). Timeliness of reporting and the quality of surveillance analyses improved 3 months after training. STEP demonstrated that mentored, competency-based training, where learners attain competencies while delivering essential public health services, can be successfully implemented in an emergency response setting.

49. Nanoscale Magnetism Probed in a Matter-Wave Interferometer

We explore a wide range of fundamental magnetic phenomena by measuring the dephasing of matter-wave interference fringes upon application of a variable magnetic gradient. The versatility of our interferometric Stern-Gerlach technique enables us to study the magnetic properties of alkali atoms, organic radicals, and fullerenes in the same device, with magnetic moments ranging from a Bohr magneton to less than a nuclear magneton. We find evidence for magnetization of a supersonic beam of organic radicals and, most notably, observe a strong magnetic response of a thermal C_{60} beam consistent with high-temperature atomlike deflection of rotational magnetic moments.
