4. Genetic Diversity of Mexican Germplasm Collection of Jatropha Curcas L

Abstract

The aim of this study was to determine the genetic diversity in accessions of Jatropha curcas located in the Mexican germplasm collection. Mexican collection has 400 germplasm accessions and is located in the municipality of Tuxtla Chico, Chiapas to 400 meters above sea level under humid tropical conditions. In this collection was conducted agronomic characterization, morphological and molecular. The results showed differences among accessions. The variation in yield of fruit was 3 grams to 2.6 kilograms per tree. The average male to female flower ratio was 12:1, however it was detected trees with flowers completely pistillate. Morphological characterization was carried out in 90 accessions using 68 descriptors of leaf, flower, fruit and seed. The results showed that variation is reflected by the shape of leaf, fruit and seed and the weight of fruit and seed. Molecular characterization was based on Mexican germplasm comprising 88 accessions using 6 primer of AFLP (Amplified Fragment Length Polymorphism) and 16 primer SSR (Simple Sequence Repeat) markers. This study revealed high genetic diversity over 70%. We conclude that there is considerable genetic diversity in germplasm Jatropha Mexican which extends the possibility of obtaining new varieties with desirable agronomic and industrial characteristic., Proceedings of the 18th European Biomass Conference and Exhibition, 3-7 May 2010, Lyon, France, pp. 542-543

6. Diallel Analysis of Resistance to Macrophomina phaseolinain Sorghum

Abstract

Charcoal rot of sorghum, caused by the fungus Macrophomina phaseolina, is a severe disease in regions where sorghum encounters post-anthesis drought stress. Previous reports disagree with respect to the narure of the genetic factors that condition resistance to charcoal rot. A diallel cross was made using nine genetically diverse sorghum lines. Parental lines and Fl progenies were evaluated in the field in 1997 and 1998 at two locations. Lines were significantly more resistant than Fls, indicating that heterosis was not affecting resistance. General combining ability (GCA) and specific combining ability (SCA) effects were significant for both locations over both years. However, the magnitudes of SCA effects were much smaller than GCA effects. These observations indicate that resistance to charcoal rot is conditioned by both additive and nonadditive effects.

Abstract

La germinación in vitro del polen de sorgo es útil en estudios de viabilidad, fisiología y transformación genética. Sin embargo, los medios reportados son poco eficientes. El objetivo del presente trabajo fue formular un medio artificial y determinar las condiciones óptimas para germinar el polen de sorgo in vitro. Para ello se usó un arreglo factorial de concentraciones de sacarosa, ácido bórico y nitrato de calcio; se evaluó el efecto del pH, la humedad relativa, el estado físico del medio y la etapa de desarrollo floral sobre la germinación. El medio óptimo se conformó por 0.7 M de sacarosa, 3.81 mM de nitrato de calcio, 1.21 mM de ácido bórico y 1% de agar con pH 6. La mayor germinación se obtuvo con polen colectado justo después de la antesis en una humedad relativa superior a 70%. Los granos de polen sin germinar y la humedad relativa se correlacionaron negativamente (-0.47, p0.01). Con las condiciones descritas en este trabajo se obtuvo hasta un 51% de germinación in vitro del polen de 14 materiales de sorgo. Estas evidencias sugieren que para incrementar la germinación in vitro se requiere una formulación óptima del medio, controlar la humedad relativa (superior a 70%) y seleccionar la etapa fenológica más adecuada para la colecta del polen (justo después de la antesis).

Abstract

Background: Low protein digestibility and lysine content of white sorghum grain limit its use as a foodstuff. The increase in γ-kafirin cross-linking, has an important role in the reduction of protein digestibility. The objective of this study was to characterize the γ-kafirin gene in 12 Mexican tannin-free white sorghum genotypes and its relationship with protein digestibility and lysine content., Results: Two alleles of γ-kafirin gene were identified: alleles 1 and 7. The predicted amino acid sequence of allele 7 showed seven point mutations; six were silent, and one missense (C235G), causing the substitution P79A in the deduced amino acid sequence. In silico analysis showed that γ-kafirin codified by allele 1 has five α-helixes without disulfide bonds, while γ-kafirin coding by allele 7 has four α-helixes and three disulfide bonds. Genotypes with allele 7 had higher lysine content than those with allele 1, showing no differences in the kafirin electrophoretic profile, neither a correlation with the protein content nor the in vitro pepsin digestibility., Conclusions: Mexican tannin-free white sorghum genotypes showed two γ-kafirin alleles, 1 and 7. Allele 7 was associated with higher lysine content; in silico analysis showed that the substitution of P79A in this allele could modify γ-kafirin secondary structure. © 2015 Society of Chemical Industry., (© 2015 Society of Chemical Industry.)
