Genomic prediction (GP) has been evaluated in durum wheat breeding programs for several years, but prediction accuracy (PA) remains insufficient for some traits. Recently, multivariate (MV) analysis has gained much attention due to its potential to significantly improve PA. In this study, PA was evaluated for several agronomic traits using a univariate (UV) model in durum wheat, subsequently, different multivariate genomic prediction models were performed to attempt to increase PA. The panel was phenotyped for 10 agronomic traits over two consecutive crop seasons and under two different field conditions: high nitrogen and well-watered (HNW), and low nitrogen and rainfed (LNR). Multivariate GP was implemented using two cross-validation (CV) schemes: MV-CV1, testing the model for each target trait using only the markers, and MV-CV2, testing the model for each target trait using additional phenotypic information. These two MV-CVs were applied in two different analyses: modelling the same trait under both HNW and LNR conditions, and modelling grain yield together with the five most genetically correlated traits. PA for all traits in HNW was higher than LNR for the same trait, except for the trait yellow index. Among all traits, PA ranged from 0.34 (NDVI in LNR) to 0.74 (test weight in HNW). In modelling the same traits in both HNW and LNR, MV-CV1 produced improvements in PA up to 12.45% (NDVI in LNR) compared to the univariate model. By contrast, MV-CV2 increased PA up to 56.72% (thousand kernel weight in LNR). The MV-CV1 scheme did not improve PA for grain yield when it was modelled with the five most genetically correlated traits, whereas MV-CV2 significantly improved PA by up to ~18%. This study demonstrated that increases in prediction accuracy for agronomic traits can be achieved by modelling the same traits in two different field conditions using MV-CV2. Silybum marianum (L.) Gaertn. is a multipurpose crop native to the Mediterranean and middle east regions and mainly known for the hepatoprotective properties of fruit-derived silymarin. Despite growing interest in milk thistle as a versatile crop with medicinal value, its potential in agroindustry is hindered by incomplete domestication and limited genomic knowledge, impeding the development of competitive breeding programs. The present study aimed to evaluate genetic diversity in a panel of S. marianum accessions (n = 31), previously characterized for morphological and phytochemical traits, using 5,178 polymorphic DArTseq SNP markers. The genetic structure investigated using both parametric and non-parametric approaches (e.g. PCA, AWclust, Admixture), revealed three distinctive groups reflecting geographical origins. Indeed, Pop1 grouped accessions from Central Europe and UK, Pop3 consisted mainly of accessions of Italian origin, and Pop2 included accessions from different geographical areas. Interestingly, Italian genotypes showed a divergent phenotypic distribution, particularly in fruit oleic and linoleic acid content, compared to the other two groups. Genetic differentiation among the three groups, investigated by computing pairwise fixation index (FST), confirmed a greater differentiation of Pop3 compared to other subpopulations, also based on other diversity indices (e.g. private alleles, heterozygosity). Finally, 22 markers were declared as putatively under natural selection, of which seven significantly affected some important phenotypic traits such as oleic, arachidonic, behenic and linoleic acid content. These findings suggest that these markers, and overall, the seven SNP markers identified within Pop3, could be exploited in specific breeding programs, potentially aimed at diversifying the use of milk thistle. Indeed, incorporating genetic material from Pop3 haplotypes carrying the selected loci into milk thistle breeding populations might be the basis for developing milk thistle lines with higher levels of oleic, arachidonic, and behenic acids, and lower levels of linoleic acid, paving new avenues for enhancing the nutritional and agronomic characteristics of milk thistle. Climate change poses a major threat to global food security, significantly reducing crop yields as cause of abiotic stresses, and for boosting the spread of new and old pathogens and pests. Sustainable crop management as a route to mitigation poses the challenge of recruiting an array of solutions and tools for the new aims. Among these, the deployment of positive interactions between the micro-biotic components of agroecosystems and plants can play a highly significant role, as part of the agro-ecological revolution. Endophytic microorganisms have emerged as a promising solution to tackle this challenge. Among these, Arbuscular Mycorrhizal Fungi (AMF) and endophytic bacteria and fungi have demonstrated their potential to alleviate abiotic stresses such as drought and heat stress, as well as the impacts of biotic stresses. They can enhance crop yields in a sustainable way also by other mechanisms, such as improving the nutrient uptake, or by direct effects on plant physiology. In this review we summarize and update on the main types of endophytes, we highlight several studies that demonstrate their efficacy in improving sustainable yields and explore possible avenues for implementing crop-microbiota interactions. The mechanisms underlying these interactions are highly complex and require a comprehensive understanding. For this reason, omic technologies such as genomics, transcriptomics, proteomics, and metabolomics have been employed to unravel, by a higher level of information, the complex network of interactions between plants and microorganisms. Industrial hemp (Cannabis sativa L.) is receiving increasing attention for its multiple end-uses; therefore, an improvement in its production is needed to meet the increased demand. In the present study, the effect of seed inoculation with two Trichoderma afroharzianum strains, T-AA and T-22, on plant growth and root morphology of hemp plants at sixth-leaf (S6) and tenth-leaf (S10) stages was assessed for two consecutive years (2020 and 2021). In addition, the ability of the two strains to enhance the accumulation of phenolic compounds in hemp leaves was also evaluated. The results obtained revealed the ability of T-22 to improve the growth and root morphology of hemp plants both in 2020 and 2021, although with different impact, probably ascribable to the different weather conditions in the two years. In 2020, the positive effects of T-22 were detected at S10 stage with significant increases in the shoot and root length (38% and 17%, respectively) and dried biomass (35% and 30%, respectively) compared to untreated plants. The total root surface area and the number of tips, forks, and crossings also increased significantly (24–36%) at this stage. In 2021, significant increases in the shoot length and dried biomass (40% and 30%, respectively) were observed at S6 stage, whereas root length and dried biomass increased significantly at S6 (55% and 47%, respectively) and S10 stage (121% and 40%, respectively). Significant increases in the total surface area and volume, as well as in the number of tips, forks, and crossings were also observed at both S6 and S10 stage (50–63% and 105–187%, respectively). Interestingly, in both years and at both stages, the two strains induced significant increases in the leaf accumulation of phenolic compounds and the antioxidant activity, which were greater in T-22- compared to T-AA-treated plants (18–102% and 13–34%, respectively). The results are discussed in light of the potential practical applications of T-22 as a biostimulant of hemp plant growth under favorable and unfavorable environmental conditions, and of both strains as promising tools for the improvement of the leaves' economic value as a source of health-promoting compounds. Vernalization, the requirement of a period of low temperature to induce transition from the vegetative to reproductive state, is an evolutionarily and economically important trait in the Triticeae. The genetic basis of vernalization in cultivated barley (Hordeum vulgare subsp. vulgare) can be defined using the two-locus VRN-H1/VRN-H2 model. We analyzed the allelic characteristics of HvBM5A, the candidate gene for VRN-H1, from ten cultivated barley accessions and one wild progenitor accession (subsp. spontaneum), representing the three barley growth habits - winter, facultative, and spring. We present multiple lines of evidence, including sequence, linkage map location, and expression, that support HvBM5A being VRN-H1. While the predicted polypeptides from different growth habits are identical, spring accessions contain a deletion in the first intron of HvBM5A that may be important for regulation. While spring HvBM5A alleles are typified by the intron-localized deletion, in some cases, the promoter may also determine the allele type. The presence/absence of the tightly linked ZCCT-H gene family members on chromosome 4H perfectly correlates with growth habit and we conclude that one of the three ZCCT-H genes is VRN-H2. The VRN-H2 locus is present in winter genotypes and deleted from the facultative and spring genotypes analyzed in this study, suggesting the facultative growth habit (cold tolerant, vernalization unresponsive) is a result of deletion of the VRN-H2 locus and presence of a winter HvBM5A allele. All reported barley vernalization QTLs can be explained by the two-locus VRN-H1/VRN-H2 model based on the presence/absence of VRN-H2 and a winter vs. spring HvBM5A allele.

In the present study, the phytochemical content and the antioxidant activity in the inflorescences of the monoecious hemp cultivar Codimono grown in southern Italy were assessed, and their elicitation was induced by foliar spray application of 50 mg/L and 250 mg/L of chitosan (CHT) at three different molecular weights (low, CHT L; medium, CHT M; high CHT H). The analysis of the phytochemical profile confirmed that cannabinoids were the most abundant class (54.2%), followed by flavonoids (40.3%), tocopherols (2.2%), phenolic acids (1.9%), and carotenoids (1.4%). Cannabinoids were represented almost exclusively by cannabidiol, whereas cannabigerol and Δ9-tetrahydrocannabinol were detected at very low levels (the latter was below the legal limit of 0.3%). The most abundant flavonoids were orientin and vitexin, whereas tocopherols were mainly represented by α-tocopherol. The antioxidant activity was found to be positively correlated with flavonoids and tocopherols. Statistical analysis revealed that the CHT treatments significantly affected the phytochemical content and the antioxidant activity of hemp inflorescences. Notably, a significant increase in the total phenolic content (from +36% to +69%), the α-tocopherol (from +45% to +75%) and β+γ-tocopherol (from +35% to +82%) contents, and the ABTS radical scavenging activity (from +12% to +28%) was induced by all the CHT treatments. In addition, treatments with CHT 50 solutions induced an increase in the total flavonoid content (from +12% to +27%), as well as in the vitexin (from +17% to +20%) and orientin (from +20% to +30%) contents. Treatment with CHT 50 L almost always resulted in the greatest increases. Overall, our findings indicated that CHT could be used as a low-cost and environmentally safe elicitor to improve the health benefits and the economic value of hemp inflorescences, thus promoting their employment in the food, pharmaceutical, nutraceutical, and cosmetic supply chains. Vascular inflammation (VI) represents a pathological condition that progressively affects the integrity and functionality of the vascular wall, thus leading to endothelial dysfunction and the onset of several cardiovascular diseases. Therefore, the research of novel compounds able to prevent VI represents a compelling need. In this study, we tested erucin, the natural isothiocyanate H2S-donor derived from Eruca sativa Mill. (Brassicaceae), in an in vivo mouse model of lipopolysaccharide (LPS)-induced peritonitis, where it significantly reduced the amount of emigrated CD11b positive neutrophils. We then evaluated the anti-inflammatory effects of erucin in LPS-challenged human umbilical vein endothelial cells (HUVECs). The pre-incubation of erucin, before LPS treatment (1, 6, 24 h), significantly preserved cell viability and prevented the increase of reactive oxygen species (ROS) and tumor necrosis factor alpha (TNF-α) levels. Moreover, erucin downregulated endothelial hyperpermeability and reduced the loss of vascular endothelial (VE)-Cadherin levels. In addition, erucin decreased vascular cell adhesion molecule 1 (VCAM-1), cyclooxygenase-2 (COX-2) and microsomal prostaglandin E-synthase 1 (mPGES-1) expression. Of note, erucin induced eNOS phosphorylation and counteracted LPS-mediated NF-κB nuclear translocation, an effect that was partially abolished in the presence of the eNOS inhibitor L-NAME. The potential of the model grass Brachypodium distachyon L. (Brachypodium) for studying grass-pathogen interactions is still underexploited. We aimed to identify genomic regions in Brachypodium associated with quantitative resistance to the false brome rust fungus Puccinia brachypodii. The inbred lines Bd3-1 and Bd1-1, differing in their level of resistance to P. brachypodii, were crossed to develop an [F.sub.2] population. This was evaluated for reaction to a virulent isolate of P brachypodii at both the seedling and advanced growth stages. To validate the results obtained on the [F.sub.2], resistance was quantified in [F.sub.2]-derived [F.sub.3] families in two experiments. Disease evaluations showed quantitative and transgressive segregation for resistance. A new AFLP-based Brachypodium linkage map consisting of 203 loci and spanning 812 cM was developed and anchored to the genome sequence with SSR and SNP markers. Three false brome rust resistance QTLs were identified on chromosomes 2, 3, and 4, and they were detected across experiments. This study is the first quantitative trait analysis in Brachypodium. Resistance to P. brachypodii was governed by a few QTLs: two acting at the seedling stage and one acting at both seedling and advanced growth stages. The results obtained offer perspectives to elucidate the molecular basis of quantitative resistance to rust fungi. (Brachypodium) pour l'etude des interactions plante-pathogene est encore sous-utilise. Les auteurs ont voulu identifier des regions genomiques du Brachypodium qui sont associees a la resistance quantitative a l'agent fongique de la rouille brachypode, Puccinia brachypodii. Les lignees fixees Bd3-1 et Bd1-1, lesquelles different en ce qui a trait a leur resistance au P. brachypodii, ont ete croisees pour produire une population [F.sub.2]. Celle-ci a ete evaluee pour sa reaction a un isolat virulent du P. brachypodii a la fois au stade plantule et aux stades plus avances. Pour valider les resultats obtenus chez la [F.sub.2], la resistance a ete quantifiee chez des familles [F.sub.3] derivees d'individus [F.sub.2] au cours de deux essais. Les evaluations de la maladie ont montre une segregation quantitative et transgressive. Une nouvelle carte genetique de marqueurs AFLP comprenant 203 locus et s'etendant sur 812 cM a ete developpee et ancree a la sequence genomique grace a des marqueurs SSR et SNP. Trois QTL de resistance a la rouille brachypode ont ete identifies sur les chromosomes 2, 3 et 4, et ils ont ete detectes dans tous les essais. Ce travail constitue la premiere analyse d'un caractere quantitatif chez Brachypodium.
