Abstract

Background Increasing awareness of milder presentations of cystic fibrosis (CF) and greater interest in non-CF bronchiectasis are likely to lead to more CF screening by respiratory clinicians. As a result, adults who may not strictly fulfil CF diagnostic criteria yet display evidence of abnormal CF transmembrane conductance regulator (CFTR) function are being identified. The degree of agreement on diagnosis and care needs in these cases between CF clinicians remains unknown, and has implications for patient care, including access to CFTR modulator therapies. Methods We surveyed adult CF physicians in Canada, the USA, the UK and Ireland, and presented them with anonymised vignettes of adult patients referred for assessment of possible CF. Diagnostic inter-rater agreement over diagnosis, ease of classifying cases and appropriate follow-up was assessed using Krippendorff's reliability coefficient (α). Abstract

Abstract Rationale The University of California, San Diego Shortness of Breath Questionnaire (UCSDSOBQ) is a frequently used domain-specific dyspnea questionnaire; however, there is little information available regarding its use and minimum important difference (MID) in fibrotic interstitial lung disease (ILD). We aimed to describe the key performance characteristics of the UCSDSOBQ in this population. Methods UCSDSOBQ scores and selected anchors were measured in 1933 patients from the prospective multi-center Canadian Registry for Pulmonary Fibrosis. Anchors included the St. George’s Respiratory Questionnaire (SGRQ), European Quality of Life 5 Dimensions 5 Levels questionnaire (EQ-5D-5L) and EQ visual analogue scale (EQ-VAS), percent-predicted forced vital capacity (FVC%), diffusing capacity of the lung for carbon monoxide (DLCO%), and 6-min walk distance (6MWD). Concurrent validity, internal consistency, ceiling and floor effects, and responsiveness were assessed, followed by estimation of the MID by anchor-based (linear regression) and distribution-based methods (standard error of measurement). Results The UCSDSOBQ had a high level of internal consistency (Cronbach’s alpha = 0.97), no obvious floor or ceiling effect, strong correlations with SGRQ, EQ-5D-5L, and EQ-VAS (|r| > 0.5), and moderate correlations with FVC%, DLCO%, and 6MWD (0.3

Abstract

Abstract Background Comorbidities are frequent and have been associated with poor quality of life, increased hospitalizations, and mortality in patients with interstitial lung disease (ILD). However, it is unclear how comorbidities lead to these negative outcomes and whether they could influence ILD disease progression. The goal of this study was to identify clusters of patients based on similar comorbidity profiles and to determine whether these clusters were associated with rate of lung function decline and/or mortality. Methods Patients with a major fibrotic ILD (idiopathic pulmonary fibrosis (IPF), fibrotic hypersensitivity pneumonitis, connective tissue disease-associated ILD, and unclassifiable ILD) from the CAnadian REgistry for Pulmonary Fibrosis (CARE-PF) were included. Hierarchical agglomerative clustering of comorbidities, age, sex, and smoking pack-years was conducted for each ILD subtype to identify combinations of these features that frequently occurred together in patients. The association between clusters and change in lung function over time was determined using linear mixed effects modeling, with adjustment for age, sex, and smoking pack-years. Kaplan Meier curves were used to assess differences in survival between the clusters. Results Discrete clusters were identified within each fibrotic ILD. In IPF, males with obstructive sleep apnea (OSA) had more rapid decline in FVC %-predicted (− 11.9% per year [95% CI − 15.3, − 8.5]) compared to females without any comorbidities (− 8.1% per year [95% CI − 13.6, − 2.7]; p = 0.03). Females without comorbidities also had significantly longer survival compared to all other IPF clusters. There were no significant differences in rate of lung function decline or survival between clusters in the other fibrotic ILD subtypes. Conclusions The combination of male sex and OSA may portend worse outcomes in IPF. Further research is required to elucidate the interplay between sex and comorbidities in ILD, as well as the role of OSA in ILD disease progression.

Abstract

Abstract Rationale Lumacaftor/ivacaftor (LUM/IVA) modestly improves lung function following 1 month of treatment but it is unknown if this translates into improvements in exercise endurance and exertional symptoms. Methods Adult CF participants completed a symptom-limited constant load cycling test with simultaneous assessments of dyspnea and leg discomfort ratings pre- and 1 month post-initiation of LUM/IVA. Results Endurance time, exertional dyspnea and leg discomfort ratings at submaximal exercise did not change significantly. There was a significant inverse correlation between changes in leg discomfort and endurance time (r = − 0.88; p = 0.009) following 1-month of LUM/IVA. Conclusions Overall, 1-month of LUM/IVA did not increase endurance time or modify exertional dyspnea or leg discomfort ratings. However, individuals who experienced a reduction in leg discomfort following LUM/IVA had an improvement in endurance time. Future studies with a larger sample size are needed to verify these findings and to assess the long-term effects of LUM/IVA on exercise outcomes. Abstract

The introduction and rapid uptake of CFTR modulator therapy, in addition to other treatments, has significantly increased life expectancy in CF and provided more women the opportunity to consider and successfully be managed throughout pregnancy. There is however limited evidence to guide patient management and enable informed decision making. Here we report the experience to date from a large multidisciplinary Cystic Fibrosis quaternary referral center in managing patients on CFTR modulators in the peri- and post-partum periods. While women in this case series were advised to discontinue CFTR modulators during pregnancy, they would likely receive a very different message today.

Abstract

Abstract Background Connective tissue disease-associated interstitial lung disease (CTD-ILD) is associated with reduced quality of life and poor prognosis. Prior studies have not identified a consistent combination of variables that accurately predict prognosis in CTD-ILD. The objective of this study was to identify baseline demographic and clinical characteristics that are associated with progression and mortality in CTD-ILD. Methods Patients were retrospectively identified from an adult CTD-ILD clinic. The predictive significance of baseline variables on serial forced vital capacity (FVC), diffusion capacity (DLCO), and six-minute walk distance (6MWD) was assessed using linear mixed effects models, and Cox regression analysis was performed to assess impact on mortality. Results 359 patients were included in the study. Median follow-up time was 4.0 (IQR 1.5–7.6) years. On both unadjusted and multivariable analysis, male sex and South Asian ethnicity were associated with decline in FVC. Male sex, positive smoking history, and diagnosis of systemic sclerosis (SSc) vs. other CTD were associated with decline in DLCO. Male sex and usual interstitial pneumonia (UIP) pattern predicted decline in 6MWD. There were 85 (23.7%) deaths. Male sex, older age, First Nations ethnicity, and a diagnosis of systemic sclerosis vs. rheumatoid arthritis were predictors of mortality on unadjusted and multivariable analysis. Conclusion Male sex, older age, smoking, South Asian or First Nations ethnicity, and UIP pattern predicted decline in lung function and/or mortality in CTD-ILD. Further longitudinal studies may add to current clinical prediction models for prognostication in CTD-ILD.

Sex differences in morbidity and mortality have been reported in the cystic fibrosis (CF) population worldwide. However, it is unclear why CF women have worse clinical outcomes than men. In this review, we focus on the influence of female sex hormones on CF pulmonary outcomes and summarise data from in vitro and in vivo experiments on how oestrogen and progesterone might modify mucociliary clearance, immunity and infection in the CF airways. The potential for novel sex hormone-related therapeutic interventions is also discussed.

Abstract

The gene responsible for cystic fibrosis (CF) was discovered 25 years ago. This breakthrough has enabled a sophisticated understanding of how various mutations lead to specific alterations in the structure and function of the CF transmembrane regulator (CFTR) protein. Until recently, all therapies in CF were focused on ameliorating the downstream consequences of CFTR dysfunction. High-throughput drug screening approaches have yielded compounds that can modify CFTR structure and function, thus targeting the basic defect in CF. The present article describes the CFTR mutational classes, reviews mutation-specific therapies currently in late-phase clinical development, and highlights research opportunities and challenges with personalized medicine in CF.

Abstract

Immunoglobulin (Ig) G4-related lung disease is a fibroinflammatory entity that presents in protean ways. Diagnostically, IgG4-related lung disease requires a high clinical index of suspicion complemented by elevated serum IgG4 levels and/or biopsy that shows the characteristic pathological features. The disease is almost always responsive to systemic corticosteroids. However, relapse is common following their discontinuation. The authors present three cases to highlight the diverse clinical features, and to illustrate the diagnostic and therapeutic approaches to this disease.

Abstract

Background. The relative rarity and diversity of fibrotic interstitial lung disease (ILD) have made it challenging to study these diseases in single-centre cohorts. Here we describe formation of a multicentre Canadian registry that is needed to describe the outcomes of fibrotic ILD and to enable detailed healthcare utilization analyses that will be the cornerstone for future healthcare planning. Methods. The Canadian Registry for Pulmonary Fibrosis (CARE-PF) is a prospective cohort anticipated to consist of at least 2,800 patients with fibrotic ILD. CARE-PF will be used to (1) describe the natural history of fibrotic ILD, specifically determining the incidence and outcomes of acute exacerbations of ILD subtypes and (2) determine the impact of ILD and acute exacerbations of ILD on health services use and healthcare costs in the Canadian population. Consecutive patients with fibrotic ILD will be recruited from five Canadian ILD centres over a period of five years. Patients will be followed up as clinically indicated and will complete standardized questionnaires at each clinic visit. Prespecified outcomes and health services use will be measured based on self-report and linkage to provincial health administrative databases. Conclusion. CARE-PF will be among the largest prospective multicentre ILD registries in the world, providing detailed data on the natural history of fibrotic ILD and the healthcare resources used by these patients. As the largest and most comprehensive cohort of Canadian ILD patients, CARE-PF establishes a network for future clinical research and early phase clinical trials and provides a platform for translational and basic science research.

11. Evaluating Adult Cystic Fibrosis Care in BC: Disparities in Access to a Multidisciplinary Treatment Centre

Abstract

Background. Cystic fibrosis (CF) care that is delivered through dedicated, multidisciplinary CF clinics is believed to be partly responsible for improvements in the length and quality of life of persons with CF. We hypothesized patients living farthest from a CF clinic would be seen less frequently than recommended, which would result in reduced access to guideline-recommended care and poorer health outcomes. Methods. We performed a retrospective chart review of 168 patients who accessed CF care primarily through the St. Paul’s Hospital Adult CF Clinic. Subjects were stratified into four geographical groups according to the estimated one-way travel time by automobile from their home address to the clinic (360 mins). Results. There were no significant differences in pulmonary function, nutritional status, CF-related complications, or access to guideline-recommended CF pulmonary therapies between the four groups. Compared to the reference (150 mins) were less likely to be seen in the clinic quarterly as recommended by current CF care guidelines (p=0.002). Those in the farthest group (>360 mins) were at risk for more rapid decline in lung function compared to the reference group (FEV1% predicted annual change: −3.1%/year [95% CI −5.1 to −1.1] versus −0.9%/year [95% CI −1.6 to 0.1], resp., p=0.04). Conclusions. Access to CF care is a challenge for individuals who live outside Metro Vancouver and has health policy implications. Further initiatives should be undertaken to ensure equitable care for people living with CF.

13. Plasma sCD14 as a biomarker to predict pulmonary exacerbations in cystic fibrosis.

Abstract

BACKGROUND: One in four cystic fibrosis (CF) patients diagnosed with a pulmonary exacerbation will not recover their baseline lung function despite standard treatment. This highlights the importance of preventing such events. Clinical decision-making can be improved through a simple blood test that predicts individuals at elevated short-term risk of an exacerbation. METHODS: We obtained plasma samples from 30 stable CF patients from the St. Paul's Hospital Adult CF Clinic (Vancouver, Canada). For 15 patients, an additional plasma sample was obtained during an exacerbation. Soluble CD14 (sCD14) and C-reactive protein (CRP) were quantified using ELISA kits. Myeloperoxidase (MPO), interleukin(IL)-6, IL-1β, monocyte chemoattractant protein-1 (MCP-1), vascular endothelial growth factor (VEGF), and granulocyte colony-stimulating factor (G-CSF) were quantified using Luminex™ immunoassays. Stable state biomarker levels were examined in their ability to predict individuals that would experience a pulmonary exacerbation requiring intravenous (IV) antibiotics within 4 months. Paired stable and exacerbation plasma biomarker levels were also compared. RESULTS: sCD14 levels were significantly higher in patients that experienced a pulmonary exacerbation requiring IV antibiotics within 4 months (p = 0.001). sCD14 cut-off value of 1450 ng/mL was associated with an area under the curve of 0.91 (95% CI 0.83-0.99) for predicting an exacerbation within 4 months of a stable visit, with a sensitivity of 100% and specificity of 82%. Plasma sCD14 levels were significantly higher during exacerbations than during periods of clinical stability (p = 0.03). CONCLUSIONS: Plasma sCD14 is a promising biomarker for identifying CF patients who will exacerbate within 4 months of a stable visit but requires further study in larger, independent cohorts.

Abstract

Advances in cystic fibrosis management have significantly improved life expectancy in these patients. However, we are now faced with a growing number of long-term extrapulmonary consequences of this disease, including ophthalmic complications of diabetes in cystic fibrosis patients. We present a unique report that documents a case of diabetic papillopathy progressing to nonarteritic anterior ischemic optic neuropathy resulting in vision loss in a patient with CF and diabetes. It highlights the potentially devastating consequences of longstanding diabetes in CF patients.

Abstract

Acute exacerbations of interstitial lung disease present as clinical deteriorations, with progressive hypoxemia and parenchymal consolidation not related to infection, heart failure or thromboembolic disease. Following single lung transplantation, patients receive maintenance immunosuppression, which could mitigate the development of acute exacerbations in the native lung. A 66-year-old man with fibrotic, nonspecific interstitial pneumonitis presented with fever, hypoxemia and parenchymal consolidation limited to the native lung four years after single lung transplantation. Investigations were negative for infection, heart failure and thromboembolic disease. The patient worsened over the course of one week despite broad-spectrum antimicrobial therapy, but subsequently improved promptly with augmentation of prednisone dosed to 50 mg daily and addition of N-acetylcysteine. Hence, the patient fulfilled the criteria for a diagnosis of an acute exacerbation of pulmonary fibrosis in his native lung. Clinicians should consider acute exacerbation of parenchymal lung disease of the native lung in the differential diagnosis of progressive respiratory deterioration following single lung transplantation for pulmonary fibrosis.

Abstract

Rationale: Several common and rare genetic variants have been associated with idiopathic pulmonary fibrosis, a progressive fibrotic condition that is localized to the lung. Objectives: To develop an integrated understanding of the rare and common variants located in multiple loci that have been reported to contribute to the risk of disease. Methods: We performed deep targeted resequencing (3.69 Mb of DNA) in cases (n = 3,624) and control subjects (n = 4,442) across genes and regions previously associated with disease. We tested for associations between disease and 1) individual common variants via logistic regression and 2) groups of rare variants via sequence kernel association tests. Measurements and Main Results: Statistically significant common variant association signals occurred in all 10 of the regions chosen based on genome-wide association studies. The strongest risk variant is the MUC5B promoter variant rs35705950, with an odds ratio of 5.45 (95% confidence interval, 4.91-6.06) for one copy of the risk allele and 18.68 (95% confidence interval, 13.34-26.17) for two copies of the risk allele (P = 9.60 × 10-295). In addition to identifying for the first time that rare variation in FAM13A is associated with disease, we confirmed the role of rare variation in the TERT and RTEL1 gene regions in the risk of IPF, and found that the FAM13A and TERT regions have independent common and rare variant signals. Conclusions: A limited number of common and rare variants contribute to the risk of idiopathic pulmonary fibrosis in each of the resequencing regions, and these genetic variants focus on biological mechanisms of host defense and cell senescence.

Abstract

We report the first measurement of the neutrino-oxygen neutral-current quasielastic (NCQE) cross section. It is obtained by observing nuclear deexcitation $\gamma$-rays which follow neutrino-oxygen interactions at the Super-Kamiokande water Cherenkov detector. We use T2K data corresponding to $3.01 \times 10^{20}$ protons on target. By selecting only events during the T2K beam window and with well-reconstructed vertices in the fiducial volume, the large background rate from natural radioactivity is dramatically reduced. We observe 43 events in the $4-30$ MeV reconstructed energy window, compared with an expectation of 51.0, which includes an estimated 16.2 background events. The background is primarily nonquasielastic neutral-current interactions and has only 1.2 events from natural radioactivity. The flux-averaged NCQE cross section we measure is $1.55 \times 10^{-38}$ cm$^2$ with a 68\% confidence interval of $(1.22, 2.20) \times 10^{-38}$ cm$^2$ at a median neutrino energy of 630 MeV, compared with the theoretical prediction of $2.01 \times 10^{-38}$ cm$^2$., Comment: 10 pages, 6 figures

Abstract

The T2K experiment has observed electron neutrino appearance in a muon neutrino beam produced 295 km from the Super-Kamiokande detector with a peak energy of 0.6 GeV. A total of 28 electron neutrino events were detected with an energy distribution consistent with an appearance signal, corresponding to a significance of 7.3$\sigma$ when compared to 4.92 $\pm$ 0.55 expected background events. In the PMNS mixing model, the electron neutrino appearance signal depends on several parameters including three mixing angles $\theta_{12}$, $\theta_{23}$, $\theta_{13}$, a mass difference $\Delta m^2_{32}$ and a CP violating phase $\delta_{\mathrm{CP}}$. In this neutrino oscillation scenario, assuming $|\Delta m^2_{32}| = 2.4 \times 10^{-3}$ $\rm eV^2$, $\sin^2 \theta_{23} = 0.5$, and $\Delta m^2_{32} >0$ ($\Delta m^2_{32} <0$), a best-fit value of $\sin^2 2 \theta_{13}$ = $0.140^{+0.038}_{-0.032}$ ($0.170^{+0.045}_{-0.037}$) is obtained at $\delta_{\mathrm{CP}}=0$. Abstract

The T2K experiment studies oscillations of an off-axis muon neutrino beam between the J-PARC accelerator complex and the Super-Kamiokande detector. Special emphasis is placed on measuring the mixing angle theta_13 by observing electron neutrino appearance via the sub-dominant muon neutrino to electron neutrino oscillation, and searching for CP violation in the lepton sector. The experiment includes a sophisticated, off-axis, near detector, the ND280, situated 280 m downstream of the neutrino production target in order to measure the properties of the neutrino beam and to understand better neutrino interactions at the energy scale below a few GeV. The data collected with the ND280 are used to study charged- and neutral-current neutrino interaction rates and kinematics prior to oscillation, in order to reduce uncertainties in the oscillation measurements by the far detector. A key element of the near detector is the ND280 electromagnetic calorimeter (ECal), consisting of active scintillator bars sandwiched between lead sheets and read outwith multi-pixel photon counters (MPPCs). The ECal is vital to the reconstruction of neutral particles, and the identification of charged particle species. The ECal surrounds the Pi-0 detector (P0D) and the tracking region of the ND280, and is enclosed in the former UA1/NOMAD dipole magnet. This paper describes the design, construction and assembly of the ECal, as well as the materials from which it is composed. The electronic and data acquisition (DAQ) systems are discussed, and performance of the ECal modules, as deduced from measurements with particle beams, cosmic rays, the calibration system, and T2K data, is described., Comment: 45 pages, 29 PDF figures. Abstract

The T2K collaboration reports a precision measurement of muon neutrino disappearance with an off-axis neutrino beam with a peak energy of 0.6 GeV. Near detector measurements are used to constrain the neutrino flux and cross section parameters. The Super-Kamiokande far detector, which is 295 km downstream of the neutrino production target, collected data corresponding to $3.01 \times 10^{20}$ protons on target. In the absence of neutrino oscillations, $205 \pm 17$ (syst.) events are expected to be detected and only 58 muon neutrino event candidates are observed. A fit to the neutrino rate and energy spectrum assuming three neutrino flavors, normal mass hierarchy and $\theta_{23}\leq \pi/4$ yields a best-fit mixing angle $\sin^2(2\theta_{23})=1.000$ and mass splitting $|\Delta m^2_{32}| =2.44 \times 10^{-3}$ eV$^2$/c$^4$. If $\theta_{23}\geq \pi/4$ is assumed, the best-fit mixing angle changes to $\sin^2(2\theta_{23})=0.999$ and the mass splitting remains unchanged., Comment: 8 pages, 4 figures, revised version accepted for publication in Physical Review Letters

Abstract

The T2K collaboration: reports evidence for electron neutrino appearance at the atmospheric mass splitting, |\Delta m_{32}^2|=2.4x10^{-3} eV^2. An excess of electron neutrino interactions over background is observed from a muon neutrino beam with a peak energy of 0.6 GeV at the Super-Kamiokande (SK) detector 295 km from the beam's origin. Signal and background predictions are constrained by data from near detectors located 280 m from the neutrino production target. We observe 11 electron neutrino candidate events at the SK detector when a background of 3.3\pm0.4(syst.) events is expected. The background-only hypothesis is rejected with a p-value of 0.0009 (3.1\sigma), and a fit assuming \nu_{\mu}->\nu_e oscillations with sin^2(2\theta_{23})=1, \delta_{CP}=0 and |\Delta m_{32}^2|=2.4x10^{-3} eV^2 yields sin^2(2\theta_{13})=0.088^{+0.049}_{-0.039}(stat.+syst.)., Comment: 43 pages, 41 figures

Abstract

T2K has performed the first measurement of \nu{\mu} inclusive charged current interactions on carbon at neutrino energies of ~1 GeV where the measurement is reported as a flux-averaged double differential cross section in muon momentum and angle. The flux is predicted by the beam Monte Carlo and external data, including the results from the NA61/SHINE experiment. The data used for this measurement were taken in 2010 and 2011, with a total of 10.8 x 10^{19} protons-on-target. The analysis is performed on 4485 inclusive charged current interaction candidates selected in the most upstream fine-grained scintillator detector of the near detector. The flux-averaged total cross section is <\sigma_CC>_\phi =(6.91 +/- 0.13 (stat) +/- 0.84 (syst)) x10^{-39} cm^2/nucleon for a mean neutrino energy of 0.85 GeV.

Abstract

The Tokai-to-Kamioka (T2K) experiment studies neutrino oscillations using an off-axis muon neutrino beam with a peak energy of about 0.6 GeV that originates at the J-PARC accelerator facility. Interactions of the neutrinos are observed at near detectors placed at 280 m from the production target and at the far detector -- Super-Kamiokande (SK) -- located 295 km away. The flux prediction is an essential part of the successful prediction of neutrino interaction rates at the T2K detectors and is an important input to T2K neutrino oscillation and cross section measurements. A FLUKA and GEANT3 based simulation models the physical processes involved in the neutrino production, from the interaction of primary beam protons in the T2K target, to the decay of hadrons and muons that produce neutrinos. The simulation uses proton beam monitor measurements as inputs. The modeling of hadronic interactions is re-weighted using thin target hadron production data, including recent charged pion and kaon measurements from the NA61/SHINE experiment. For the first T2K analyses the uncertainties on the flux prediction are evaluated to be below 15% near the flux peak. Abstract

The T2K experiment is a long baseline neutrino oscillation experiment aiming to observe the appearance of {\nu} e in a {\nu}{\mu} beam. The {\nu}{\mu} beam is produced at the Japan Proton Accelerator Research Complex (J-PARC), observed with the 295 km distant Super- Kamiokande Detector and monitored by a suite of near detectors at 280m from the proton target. The near detectors include a magnetized off-axis detector (ND280) which measures the un-oscillated neutrino flux and neutrino cross sections. The present paper describes the outermost component of ND280 which is a side muon range detector (SMRD) composed of scintillation counters with embedded wavelength shifting fibers and Multi-Pixel Photon Counter read-out. Abstract

We report a measurement of muon-neutrino disappearance in the T2K experiment. The 295-km muon-neutrino beam from Tokai to Kamioka is the first implementation of the off-axis technique in a long-baseline neutrino oscillation experiment. With data corresponding to 1.43 10**20 protons on target, we observe 31 fully-contained single muon-like ring events in Super-Kamiokande, compared with an expectation of 104 +- 14 (syst) events without neutrino oscillations. The best-fit point for two-flavor nu_mu -> nu_tau oscillations is sin**2(2 theta_23) = 0.98 and |\Delta m**2_32| = 2.65 10**-3 eV**2. The boundary of the 90 % confidence region includes the points (sin**2(2 theta_23),|\Delta m**2_32|) = (1.0, 3.1 10**-3 eV**2), (0.84, 2.65 10**-3 eV**2) and (1.0, 2.2 10**-3 eV**2)., Comment: 7 pages, 4 figures

Abstract

Precise measurement of neutrino beam direction and intensity was achieved based on a new concept with modularized neutrino detectors. INGRID (Interactive Neutrino GRID) is an on-axis near detector for the T2K long baseline neutrino oscillation experiment. INGRID consists of 16 identical modules arranged in horizontal and vertical arrays around the beam center. The module has a sandwich structure of iron target plates and scintillator trackers. INGRID directly monitors the muon neutrino beam profile center and intensity using the number of observed neutrino events in each module. The neutrino beam direction is measured with accuracy better than 0.4 mrad from the measured profile center. Abstract

We report constraints on antineutrino oscillation parameters that were obtained by using the two MINOS detectors to measure the 7% muon antineutrino component of the NuMI neutrino beam. In the Far Detector, we select 130 events in the charged-current muon antineutrino sample, compared to a prediction of 136.4 +/- 11.7(stat) ^{+10.2}_{-8.9}(syst) events under the assumption |dm2bar|=2.32x10^-3 eV^2, snthetabar=1.0. Assuming no oscillations occur at the Near Detector baseline, a fit to the two-flavor oscillation approximation constrains |dm2bar|<3.37x10^-3 eV^2 at the 90% confidence level with snthetabar=1.0., Comment: Accepted for publication by Phys. Abstract

We report the results of a search for $\nu_{e}$ appearance in a $\nu_{\mu}$ beam in the MINOS long-baseline neutrino experiment. With an improved analysis and an increased exposure of $8.2\times10^{20}$ protons on the NuMI target at Fermilab, we find that $2\sin^2(\theta_{23})\sin^2(2\theta_{13})<0.12\ (0.20)$ at 90% confidence level for $\delta\mathord{=}0$ and the normal (inverted) neutrino mass hierarchy, with a best fit of $2\sin^2(\theta_{23})\sin^2(2\theta_{13})\,\mathord{=}\,0.041^{+0.047}_{-0.031}\ (0.079^{+0.071}_{-0.053})$. The $\theta_{13}\mathord{=}0$ hypothesis is disfavored by the MINOS data at the 89% confidence level., Comment: 5 pages, 3 figures

Abstract

The T2K experiment observes indications of $\nu_\mu\rightarrow \nu_e$ appearance in data accumulated with $1.43\times10^{20}$ protons on target. Six events pass all selection criteria at the far detector. In a three-flavor neutrino oscillation scenario with $|\Delta m_{23}^2|=2.4\times10^{-3}$ eV$^2$, $\sin^2 2\theta_{23}=1$ and $\sin^2 2\theta_{13}=0$, the expected number of such events is 1.5$\pm$0.3(syst.). Under this hypothesis, the probability to observe six or more candidate events is 7$\times10^{-3}$, equivalent to 2.5$\sigma$ significance. At 90% C.L., the data are consistent with 0.03(0.04)$<\sin^2 2\theta_{13}<$ 0.28(0.34) for $\delta_{\rm CP}=0$ and a normal (inverted) hierarchy., Comment: 20 pages, 6 figures, version published in PRL

Abstract

The T2K experiment is a long-baseline neutrino oscillation experiment. Its main goal is to measure the last unknown lepton sector mixing angle {\theta}_{13} by observing {\nu}_e appearance in a {\nu}_{\mu} beam. It also aims to make a precision measurement of the known oscillation parameters, {\Delta}m^{2}_{23} and sin^{2} 2{\theta}_{23}, via {\nu}_{\mu} disappearance studies. Other goals of the experiment include various neutrino cross section measurements and sterile neutrino searches. The experiment uses an intense proton beam generated by the J-PARC accelerator in Tokai, Japan, and is composed of a neutrino beamline, a near detector complex (ND280), and a far detector (Super-Kamiokande) located 295 km away from J-PARC. Abstract

Results are reported from a search for active to sterile neutrino oscillations in the MINOS long-baseline experiment, based on the observation of neutral-current neutrino interactions, from an exposure to the NuMI neutrino beam of $7.07\times10^{20}$ protons on target. A total of 802 neutral-current event candidates is observed in the Far Detector, compared to an expected number of $754\pm28\rm{(stat.)}\pm{37}\rm{(syst.)}$ for oscillations among three active flavors. The fraction $f_s$ of disappearing \numu that may transition to $\nu_s$ is found to be less than 22% at the 90% C.L., Comment: 5 pages, 3 tables, 2 figures. Published in Physical Review Letters

Abstract

This letter reports the first direct observation of muon antineutrino disappearance. The MINOS experiment has taken data with an accelerator beam optimized for muon antineutrino production, accumulating an exposure of $1.71\times 10^{20}$ protons on target. In the Far Detector, 97 charged current muon antineutrino events are observed. The no-oscillation hypothesis predicts 156 events and is excluded at $6.3\sigma$. The best fit to oscillation yields $\Delta \bar{m}^{2}=(3.36^{+0.46}_{-0.40}\textrm{(stat.)}\pm0.06\textrm{(syst.)})\times 10^{-3}\,\eV^{2}$, $\sin^{2}(2\bar{\theta})=0.86^{+0.11}_{-0.12}\textrm{(stat.)}\pm0.01\textrm{(syst.)}$. Abstract

Measurements of neutrino oscillations using the disappearance of muon neutrinos from the Fermilab NuMI neutrino beam as observed by the two MINOS detectors are reported. New analysis methods have been applied to an enlarged data sample from an exposure of $7.25 \times 10^{20}$ protons on target. A fit to neutrino oscillations yields values of $|\Delta m^2| = (2.32^{+0.12}_{-0.08})\times10^{-3}$\,eV$^2$ for the atmospheric mass splitting and $\rm \sin^2\!(2\theta) > 0.90$ (90%\,C.L.) for the mixing angle. Abstract

We searched for a sidereal modulation in the MINOS far detector neutrino rate. Such a signal would be a consequence of Lorentz and CPT violation as described by the Standard-Model Extension framework. It also would be the first detection of a perturbative effect to conventional neutrino mass oscillations. We found no evidence for this sidereal signature and the upper limits placed on the magnitudes of the Lorentz and CPT violating coefficients describing the theory are an improvement by factors of $20-510$ over the current best limits found using the MINOS near detector., Comment: 5 pages, 2 figures

Abstract

GENIE is a new neutrino event generator for the experimental neutrino physics community. The goal of the project is to develop a `canonical' neutrino interaction physics Monte Carlo whose validity extends to all nuclear targets and neutrino flavors from MeV to PeV energy scales. Currently, emphasis is on the few-GeV energy range, the challenging boundary between the non-perturbative and perturbative regimes, which is relevant for the current and near future long-baseline precision neutrino experiments using accelerator-made beams. The design of the package addresses many challenges unique to neutrino simulations and supports the full life-cycle of simulation and generator-related analysis tasks. GENIE is a large-scale software system, consisting of 120,000 lines of C++ code, featuring a modern object-oriented design and extensively validated physics content. Abstract

MINOS is a long baseline neutrino oscillation experiment that uses two detectors separated by 734 km. The readout systems used for the two detectors are different and have to be independently calibrated. To verify and make a direct comparison of the calibrated response of the two readout systems, test beam data were acquired using a smaller calibration detector. This detector was simultaneously instrumented with both readout systems and exposed to the CERN PS T7 test beam. Differences in the calibrated response of the two systems are shown to arise from differences in response non-linearity, photomultiplier crosstalk, and threshold effects at the few percent level. These differences are reproduced by the Monte Carlo (MC) simulation to better than 1% and a scheme that corrects for these differences by calibrating the MC to match the data in each detector separately is presented. The overall difference in calorimetric response between the two readout systems is shown to be consistent with zero to a precision of 1.3% in data and 0.3% in MC with no significant energy dependence., Comment: 25 pages, 8 figures
