Graves’ disease is an immune system disorder that results in the overproduction of thyroid hormones (hyperthyroidism). Thyroid disorders are a societal problem of great public concern because of their high prevalence. This problem can affect the well-being and quality of life of patients. The predisposing factors leading to this disease are not yet fully established and are likely to be interconnected in a complex way. Chemometric analysis allows for the detection of specific relationships between the medical parameter measurements obtained from the patients in an observation group, and the identification of patterns of similarity between these patients. It is not commonly used in clinical trials; however, it can provide reliable information which may help in creating more successful, individualised treatment strategies for established groups (patterns) of patients.The aim of this review is to summarize the latest knowledge about the risk factors for Graves’ disease and considerations about using the chemometric analysis in the study of the disease.

Contamination of the environment with toxic metals such as cadmium or lead is a worldwide issue. The accumulator of metals Cannabis sativa L. has potential to be utilized in phytoremediation, which is an environmentally friendly way of soil decontamination. Novel non-thermal plasma-based technologies may be a helpful tool in this process. Plasma activated water (PAW), prepared by contact of gaseous plasma with water, contains reactive oxygen and nitrogen species, which enhance the growth of plants. In this study, C. sativa was grown in a short-term toxicity test in a medium which consisted of plasma activated water prepared by dielectric barrier discharge with liquid electrode and different concentrations of cadmium or lead. Application of PAW on heavy metal contaminated C. sativa resulted in increased growth under Pb contamination as was determined by ecotoxicology tests. Furthermore, the PAW influence on the bioaccumulation of these metals as well as the influence on the nutrient composition of plants was studied primarily by applying Laser-induced breakdown spectroscopy (LIBS). The LIBS elemental maps show that C. sativa accumulates heavy metals mainly in the roots. The results present a new proof-of-concept in which PAW could be used to improve the growth of plants in heavy metal contaminated environment, while LIBS can be implemented to study the phytoremediation efficiency.

Heat stress is a major threat to global crop production, and understanding its impact on plant fertility is crucial for developing climate-resilient crops. Despite the known negative effects of heat stress on plant reproduction, the underlying molecular mechanisms remain poorly understood. Here, we investigated the impact of elevated temperature on centromere structure and chromosome segregation during meiosis in Arabidopsis thaliana. Consistent with previous studies, heat stress leads to a decline in fertility and micronuclei formation in pollen mother cells. Our results reveal that elevated temperature causes a decrease in the amount of centromeric histone and the kinetochore protein BMF1 at meiotic centromeres with increasing temperature. Furthermore, we show that heat stress increases the duration of meiotic divisions and prolongs the activity of the spindle assembly checkpoint during meiosis I, indicating an impaired efficiency of the kinetochore attachments to spindle microtubules. Our analysis of mutants with reduced levels of centromeric histone suggests that weakened centromeres sensitize plants to elevated temperature, resulting in meiotic defects and reduced fertility even at moderate temperatures. These results indicate that the structure and functionality of meiotic centromeres in Arabidopsis are highly sensitive to heat stress, and suggest that centromeres and kinetochores may represent a critical bottleneck in plant adaptation to increasing temperatures.

Abstract Introduction The antifibrotic drug nintedanib is used for the treatment of idiopathic pulmonary fibrosis (IPF). We analysed the effect of nintedanib on antifibrotic treatment outcome in real-world cohorts of Czech EMPIRE registry. Patients/methods Data of 611 Czech IPF subjects, 430 (70%) treated with nintedanib (NIN group), 181 (30%) with no-antifibrotic treatment (NAF group) were analysed. The influence of nintedanib on overall survival (OS), pulmonary function parameters as forced vital capacity (FVC) and diffusing lung capacity for carbon monoxide (DLCO), as well as GAP score (gender, age, physiology) and and CPI (composite physiological index) were investigated. Abstract Meiosis is a specialized cell division that halves the number of chromosomes in two consecutive rounds of chromosome segregation. In angiosperm plants is meiosis followed by mitotic divisions to form rudimentary haploid gametophytes. In Arabidopsis, termination of meiosis and transition to gametophytic development are governed by TDM1 and SMG7 that mediate inhibition of translation. Mutants deficient in this mechanism do not form tetrads but instead undergo multiple cycles of aberrant nuclear divisions that are likely caused by the failure to downregulate cyclin dependent kinases during meiotic exit. A suppressor screen to identify genes that contribute to meiotic exit uncovered a mutation in cyclin‐dependent kinase D;3 (CDKD;3) that alleviates meiotic defects in smg7 deficient plants. The CDKD;3 deficiency prevents aberrant meiotic divisions observed in smg7 mutants or delays their onset after initiation of cytokinesis, which permits formation of functional microspores. Although CDKD;3 acts as an activator of cyclin‐dependent kinase A;1 (CDKA;1), the main cyclin dependent kinase that regulates meiosis, cdkd;3 mutation appears to promote meiotic exit independently of CDKA;1. Furthermore, analysis of CDKD;3 interactome revealed enrichment for proteins implicated in cytokinesis, suggesting a more complex function of CDKD;3 in cell cycle regulation.

The Xerotyphlops vermicularis (Merrem, 1820) complex represents small, burrowing snakes that occur from the Balkan Peninsula to Afghanistan and generally they are mostly observed during the springtime. In the present study, we report new records of this snake (10 individuals observed) from Elenite village in coastal Bulgaria. According to current knowledge, they represent the northernmost locality of the species in the Balkan Peninsula. We expand the known distribution of X. vermicularis and help to fill information gaps.

In this work, we describe the Michael addition–cyclization reaction of 2-(2-nitrovinyl)phenol with two different reactive Michael donors, which lead to chiral benzopyran derivatives. Specifically, bifunctional amino-squaramides with one or two chiral units in the side chains were evaluated as catalysts in these transformations. Furthermore, the utility of selected green solvents as reaction media for these processes was also tested. The best result was achieved with methyl-cyclopentanone-2-carboxylate as the Michael donor in ethyl (–)-l-lactate with quinine-based amino-squaramide as catalyst (yield 72%, dr >99:1, ee 99%).

The Musashi family of RNA-binding proteins controls several biological processes including stem cell maintenance, cell division and neural function. Previously, we demonstrated that the C. elegans Musashi ortholog, msi-1, regulates forgetting via translational repression of the Arp2/3 actin-branching complex. However, the mechanisms controlling MSI-1 activity during the regulation of forgetting are currently unknown. Here we investigated the effects of protein phosphorylation on MSI-1 activity. We showed that MSI-1 function is likely controlled by alterations of its activity rather than its expression levels. Furthermore, we found that MSI-1 is phosphorylated and using mass spectrometry we identified MSI-1 phosphorylation at three residues (T18, S19 and S34). CRISPR-based manipulations of MSI-1 phosphorylation sites revealed that phosphorylation is necessary for MSI-1 function in both short- and long-term aversive olfactory associative memory. Thus, our study provides insight into the mechanisms regulating memory-related MSI-1 activity and may facilitate the development of novel therapeutic approaches. Author summary Understanding neural circuits and molecular mechanisms underlying learning and memory are the major challenges of neuroscience. It is a generally accepted model that a learning event causes modification of synapses; strengthening some within a circuit and weakening others (termed “synaptic plasticity”). A plastic nervous system requires not only the ability to acquire and store but also to forget new inputs. While learning and memory is widely investigated, clear-cut evidence for mechanisms involved in forgetting is still sparse. Previously, we demonstrated the role of the protein Musashi (MSI-1) in the active regulation of forgetting in the nematode C. elegans. Here we investigated the role of protein modification (phosphorylation) as a possible regulatory mechanism of the MSI-1 protein activity. We found that MSI-1 protein is modified at different positions and all of these modifications at the protein level contribute to the correct activity of the protein leading to active forgetting of short and long-term memories.

On land, the amphibians interact with the environment in a complex way-even small changes in the physiological conditions may significantly impact the behaviour and vice versa. In ectothermic tetrapods, the transition from inactive to active phase may be related to important changes in their thermal status. We studied the thermal ecology of adult Balkan spadefoots (Pelobates balcanicus Karaman, 1928) in northeastern Bulgaria. These toads spend the daytime buried between 10 and 15 cm in sandy substrates, and emerge after sunset. On the substrate, their thermal energy exchange is defined by the absence of heat flow from the sun. Secondary heat sources, like stored heat and infrared radiation from the soil play an important role for the thermal balance of the active spadefoot toads. At the beginning of their daily activity, we measured substrate temperature (at a depth of 11–12 cm), toad’s surface body temperature, and also provided thermal profiles of the animals and the substrate surface in their microhabitats. In animals which recently emerged from the substrate, the temperature was comparatively higher and was closer to that of the subsoil on the spot. After that, body temperature decreased rapidly and continued to change slowly, in correlation with air temperature. We detected a temperature gradient on the dorsal surface of the toads. On the basis of our measurements and additional data, we discuss the eventual role of air humidity and the effects of surface and skin water evaporation on the water balance and activity of the investigated toads.

The paper is dedicated to the research output presented at the jubilee 50th edition of the international scientific conference Synthesis and Analysis of Drugs 2022 (SAL), which was organized and hosted by the Faculty of Pharmacy of Masaryk University in Brno. Since its launch in 1971, when Jozef Čižmárik from the Faculty of Pharmacy of the Comenius University in Bratislava started organizing SAL, significant progress in medicinal chemistry has been documented every year through the experimental works of researchers from different scientific institutions, both universities and academies of sciences as well as companies. We must point out long-term guest of SAL, Miroslav Protiva, co-founder of the antihistamine dosulepin and antiemetic moxastine-teoclas. As already mentioned, SAL 2022 was held at the Faculty of Pharmacy in Brno, which has been part of the University of Veterinary and Pharmaceutical Sciences for 25 years but became part of Masaryk University in 2021. Hence, the year 2022 was unique not only because of the 50th anniversary of this meeting but also for this year’s conference was the first within this institution. What topics were discussed among SAL conference participants? The meeting addressed all issues related to drug research—their design, preparation, analysis, and detection methods, as well as their biological activity studies, ADME studies, etc. The multidisciplinary character of the research, including topics on pharmaceutical technology and innovative dosage form investigation, was an additional value of the conference and its outcomes. We would like to thank the organization team and all participants for both high-quality research presentations resulting in the output of this paper and for the informal and friendly atmosphere.

Working with digitized cultural heritage is an important aspect of education. This article offers research on the interaction of pupils and teachers with digitized cultural heritage processed in the Twine2 environment, focusing on exploring the potential use of digital nonlinear stories in the teaching of humanities in high schools. The aim of this study is to describe the design process that led to the creation of 16 innovative educational resources, and to present the results of their pilot testing and evaluation, which subsequently took place among participating teachers and pupils. For the evaluation, we used a mix of design methods, combining a quantitative and qualitative approach. Both the teachers and the pupils were asked about their experience and takeaways from the interaction with the resources offered. The findings show that presenting educational content in the form of a nonlinear interactive story is a suitable alternative to the traditional frontal educational approach, as both teachers and pupils perceived the resources as intuitive, appealing, and effective in helping to achieve the set educational goals. The study also highlights some key recommendations for future designers of nonlinear interactive stories, which emerged from the respondents’ feedback.

Meiosis in angiosperm plants is followed by mitotic divisions to form multicellular haploid gametophytes. Termination of meiosis and transition to gametophytic development is, in Arabidopsis, governed by a dedicated mechanism that involves SMG7 and TDM1 proteins. Mutants carrying the smg7-6 allele are semi-fertile due to reduced pollen production. We found that instead of forming tetrads, smg7-6 pollen mother cells undergo multiple rounds of chromosome condensation and spindle assembly at the end of meiosis, resembling aberrant attempts to undergo additional meiotic divisions. A suppressor screen uncovered a mutation in centromeric histone H3 (CENH3) that increased fertility and promoted meiotic exit in smg7-6 plants. The mutation led to inefficient splicing of the CENH3 mRNA and a substantial decrease of CENH3, resulting in smaller centromeres. The reduced level of CENH3 delayed formation of the mitotic spindle but did not have an apparent effect on plant growth and development. We suggest that impaired spindle re-assembly at the end of meiosis limits aberrant divisions in smg7-6 plants and promotes formation of tetrads and viable pollen. Furthermore, the mutant with reduced level of CENH3 was very inefficient haploid inducer indicating that differences in centromere size is not the key determinant of centromere-mediated genome elimination.

Background: The antifibrotic drugs nintedanib and pirfenidone are used for the treatment of idiopathic pulmonary fibrosis (IPF). We analysed the association of common profibrotic polymorphisms in MUC5B (mucin 5B, rs35705950) and DSP (desmoplakin, rs2076295) on antifibrotic treatment outcomes in IPF. Methods: MUC5B rs35705950 and DSP rs2076295 were assessed in IPF patients ( n = 210, 139 men/71 women) from the Czech EMPIRE registry and age- or sex-matched healthy individuals ( n = 205, 125 men/80 women). Measurement systems have to deal with processes in which, by the nature of the measured object or by the type of measurement itself, measurements are not repeatable in the sense that each sample unit can be measured only once. Such situations are usually referred as non-replicable measurement systems. The paper presents the approaches to GRR analysis in these cases. On the base of current development in this branch, the special attention is focused on the appropriate setup of GRR analysis and the choice which type of GRR design to use. The aim of the paper is to map out the current approaches to GRR analysis of non-replicable measurement systems with all the assumptions as for consistency, bias and temporal stability of objects. Subsequently, it is planned to verify these approaches in practice what is the opportunity to suggest the improving actions to obtain the results of high quality.

The main purpose of this work is to thoroughly describe the implementation protocol of laser-induced breakdown spectroscopy (LIBS) method in the plant analysis. Numerous feasibility studies and recent progress in instrumentation and trends in chemical analysis make LIBS an established method in plant bioimaging. In this work, we present an easy and straightforward phytotoxicity case study with a focus on LIBS method. We intend to demonstrate in detail how to manipulate with plants after exposures and how to prepare them for analyses. Moreover, we aim to achieve 2D maps of spatial element distribution with a good resolution without any loss of sensitivity. The benefits of rapid, low-cost bioimaging are highlighted.In this study, cabbage (Brassica oleracea L.) was treated with an aqueous dispersion of photon-upconversion nanoparticles (NaYF4 doped with Yb3+ and Tm3+ coated with carboxylated silica shell) in a hydroponic short-term toxicity test. After a 72-hour plant exposure, several macroscopic toxicity end-points were monitored. The translocation of Y, Yb, and Tm across the whole plant was set by employing LIBS with a lateral resolution 100 µm. The LIBS maps of rare-earth elements in B.oleracea plant grown with 50 μg/mL nanoparticle-treated and ion-treated exposures showed the root as the main storage, while the transfer via stem into leaves was minimal. On the contrary, the LIBS maps of plants exposed to the 500 μg/mL nanoparticle-treated and ion-treated uncover slightly different trends, nanoparticles as well as ions were transferred through the stem into leaves. However, the main storage organ was a root as well.

Abstract The article handles potential approaches to design and determination of total load capacity of foundation slabs and floors. The research is focused on punching shear failure of reinforced ground supported slab. The article presents detailed results of the experiment made and advanced numerical modelling based on nonlinear analysis and application of fracture-plastic model. The experiment made included a 2.0 × 1.95 m reinforced concrete slab-on-ground of 120 mm thickness. The experiment was followed by a parametric study of total load capacity calculation with nonlinear analysis which is supplemented by calculations based on existing design model code.

Abstract In this work, we proposed a new data acquisition approach that significantly improves the repetition rates of Laser-Induced Breakdown Spectroscopy (LIBS) experiments, where high-end echelle spectrometers and intensified detectors are commonly used. The moderate repetition rates of recent LIBS systems are caused by the utilization of intensified detectors and their slow full frame (i.e. echellogram) readout speeds with consequent necessity for echellogram-to-1D spectrum conversion (intensity vs. wavelength). Therefore, we investigated a new methodology where only the most effective pixels of the echellogram were selected and directly used in the LIBS experiments. Such data processing resulted in significant variable down-selection (more than four orders of magnitude). Samples of 50 sedimentary ores samples (distributed in 13 ore types) were analyzed by LIBS system and then classified by linear and non-linear Multivariate Data Analysis algorithms. The utilization of selected pixels from an echellogram yielded increased classification accuracy compared to the utilization of common 1D spectra.

In higher plants, germline differentiation occurs during a relatively short period within developing flowers. Understanding of the mechanisms that govern germline differentiation lags behind other plant developmental processes. This is largely because the germline is restricted to relatively few cells buried deep within floral tissues, which makes them difficult to study. To overcome this limitation, we have developed a methodology for live imaging of the germ cell lineage within floral organs of Arabidopsis using light sheet fluorescence microscopy. We have established reporter lines, cultivation conditions, and imaging protocols for high-resolution microscopy of developing flowers continuously for up to several days. We used multiview imagining to reconstruct a three-dimensional model of a flower at subcellular resolution. We demonstrate the power of this approach by capturing male and female meiosis, asymmetric pollen division, movement of meiotic chromosomes, and unusual restitution mitosis in tapetum cells. This method will enable new avenues of research into plant sexual reproduction.

In the early stages of the film (pre-cinematography), during the technological development of the kinescope (1865), or the moving photos, and later the flash photography (1873), the first attempts to record motion (biomechanics) appeared. The flash photography of the later period (protofilm) was replaced by the synthesis of motion (1878) when the social-cultural background also appeared on the moving photos, mainly focusing on selected themes reflecting the sports body and physicality in relation to the contemporary social atmosphere. The presented text selectively describes the development of body and body imaging in a limited period influenced by political-ideological mechanisms. However, it also functions as a factor that can influence society and bring about change. The documentary film cannot be unambiguously described as propaganda. The sport was not depicted solely from the perspective of the ideology of that time. But above all, mass exercises demonstrated the national community and state ideology.

Background: Patient adherence to an inhaled medication application technique (A-ApplT) represents a major health-care issue in patients with chronic obstructive pulmonary disease (COPD). However, there is a lack of studies evaluating this issue thoroughly. The aim of our study was to introduce a universal easy-to-use method of assessing the A-ApplT to chronic medication in moderate to very severe COPD individuals.Methods: The Czech Multicenter Research Database of COPD (COPD CMRD), a large observational prospective study, was used as a source of clinical data. A-ApplT was evaluated using our Five Steps Assessment. This measure is based on dichotomous evaluation of each of five predefined consecutive application technique steps and can be used in all settings for all currently available inhalation systems in COPD subjects.Results: A total of 546 participants (75.0% men; mean age 66.7 years; mean forced expiratory volume in 1s 44.7%) were available for analysis. This represents 69.6% of all patients recruited in the COPD CMRD. Less than one third of patients presented their application technique without any erroneous steps. The most problematic steps were breathing out completely in one breath immediately before inhalation (step No. 3), and the actual inhalation maneuver (step No. 4). The total number of errors was similar for dry powder inhalers and pressurized metered-dose inhalers.Conclusion: Our novel instrument, Five Steps Assessment, is comfortable for use in routine clinical practice to explore A-ApplT. The A-ApplT in real-life patients with non-mild COPD was inadequate and patients should be repeatedly trained by properly (re-)educated medical staff.

High-performance concrete (HPC) is subjected to wide attention in current research. Many research tasks are focused on laboratory testing of concrete mechanical properties with specific raw materials, where a mixture is prepared in a relatively small amount in ideal conditions. The wider utilization of HPC is connected, among other things, with its utilization in the construction industry. The paper presents two variants of HPC which were developed by modification of ordinary concrete used by a precast company for pretensioned bridge beams. The presented variants were produced in industrial conditions using common raw materials. Testing and comparison of basic mechanical properties are complemented with specialized tests of the resistance to chloride penetration. Tentative expenses for normal strength concrete (NSC) and HPC are compared. The research program was accomplished with a loading test of model experimental pretensioned beams with a length of 7 m made of ordinarily used concrete and one variant of HPC. The aim of the loading test was to determine the load–deformation diagrams and verify the design code load capacity calculation method. Overall, the article summarizes the possible benefits of using HPC compared to conventional concrete.

A five-month-old African jenny was presented with a history of exercise intolerance and syncopal episodes. Severe bradycardic arrhythmia due to a high-grade second-degree atrioventricular (AV) block with progression to complete AV block was diagnosed. The jenny underwent a transvenous single-chamber pacemaker implantation. The implantation procedure was performed in a lateral recumbency and the ventricular lead was inserted through the jugular vein. Positioning of the lead was guided by echocardiography. The pacemaker was programmed to VVI mode with a minimal ventricular rate of 40 pulses per minute, a pulse amplitude of 2.4 V, a pulse width of 0.5 ms and sensing amplitude of 2.5 mV. Short-term complications associated with the procedure included lead dislodgement and pacemaker pocket infection. The long-term outcome was satisfactory; the jenny showed improvement in heart function and quality of life after pacemaker implantation. The pulse generator replacement was performed twice (at nine-year intervals) and the intervention was always associated with a local inflammatory reaction around the pacing device. Cardiac examination 18 years after pacemaker implantation revealed no morphological changes in the heart; the electrode lead was still in the correct position and successful pacing and sensing of the ventricle were obtained. Regular follow-up checks are important to evaluate pacemaker function.

Fagus sylvatica Linnaeus (European beech), the ecologically and economically most important broadleaved tree species in the Czech Republic, was strongly reduced in the past. Today there are efforts to increase the proportion of beech to ensure optimal forest tree species composition. When extensively reintroducing beech, it is important to acquire more detailed knowledge of genetic diversity. Thirteen important beech populations in different stands in the territory of the Czech Republic were genotyped using 12 polymorphic nuclear microsatellite markers. The genotypic data from adult trees imply genetic differences between the populations. The estimated genetic diversity expressed as Shannon's information index ranged from 1.73 to 1.92. Thirteen beech populations showed excess homozygotes, as indicated by positive fixation index (F) values (F = 0.005-0.115). The pairwise FST values indicated low genetic differentiation between the 13 Czech beech populations, because they were greater than zero, that means they confirmed the presence of population structuring in Czech European beech. Not significant linear correlations were observed between genetic and geographic distances of the 13 beech populations studied on the basis of microsatellite markers. Twelve microsatellite markers were verified as highly polymorphic and suitable for genotyping of European beech populations.

The improving performance of the laser-induced breakdown spectroscopy (LIBS) triggered its utilization in the challenging topic of soft tissue analysis. Alterations of elemental content within soft tissues are commonly assessed and provide further insights in biological research. However, the laser ablation of soft tissues is a complex issue and demands a priori optimization, which is not straightforward in respect to a typical LIBS experiment. Here, we focus on implementing an internal standard into the LIBS elemental analysis of soft tissue samples. We achieve this by extending routine methodology for optimization of soft tissues analysis with a standard spiking method. This step enables a robust optimization procedure of LIBS experimental settings. Considering the implementation of LIBS analysis to the histological routine, we avoid further alterations of the tissue structure. Therefore, we propose a unique methodology of sample preparation, analysis, and subsequent data treatment, which enables the comparison of signal response from heterogenous matrix for different LIBS parameters. Additionally, a brief step-by-step process of optimization to achieve the highest signal-to-noise ratio (SNR) is described. The quality of laser–tissue interaction is investigated on the basis of the zinc signal response, while selected experimental parameters (e.g., defocus, gate delay, laser energy, and ambient atmosphere) are systematically modified.

(1) Background: Populus ×canescens (Aiton) Sm. is a fast-growing woody plant belonging to the family Salicaceae. Two poplar genotypes characterized by unique phenotypic traits (TP11 and TP20) were chosen to be characterized and tested for a physiological and transcriptomic response to Cd stress. (2) Methods: A comparative analysis of the effects of exposure to high cadmium (Cd) concentrations (10 µM and 100 µM) of TP11 and TP20 was performed. (3) Results: Neither of the tested Cd concentration negatively affected plant growth; however, the chlorophyll content significantly decreased. The potassium (K) content was higher in the shoots than in the roots. The magnesium concentrations were only slightly affected by Cd treatment. The zinc content in the shoots of TP20 was lower than that in the shoots of TP11. Cd accumulation was higher in the roots than in the shoots. After 10 days of exposure, 10 µM Cd resulted in comparable amounts of Cd in the roots and shoots of TP20. The most significant change in transcript amount was observed in endochitinase 2, 12-oxophytodienoate reductase 1 and phi classglutathione S-transferase. (4) Conclusions: Our study provided new insights for effective assessing the ability of different poplar genotypes to tolerate Cd stress and underlying Cd tolerance.

The aim of the study was to estimate the forage quality and mycotoxin contamination in the locality of Švýcárna (1304 m a.s.l.), where the cattle grazing after the long-term management cessation was introduced on the pasture area of 3.6 ha in 2012. The pasture was divided into two grazing plots: P1 (Nar) (dominance of Nardus stricta) and P2 (Des) (dominance of Deschampsia cespitosa). Samples of forage were taken in June, July, August and September 2014 and 2015 from five permanent plots situated in different places of locality. Samples were analysed on the content of basic nutrients and macro- and micro- elements. Further, the content of deoxynivalenol (DON), zearalenone (ZEA) and T-2/HT-2 toxin was determined using ELISA method. From June to September the mean content of crude fibre significantly (P < 0.05) increased (from 243.9 g.kg−1 DM to 331.7 g.kg−1 DM, respectively) and the content of crude protein significantly (P < 0.05) decreased (from 166.9 g.kg−1 DM to 89.2 g.kg−1 DM, respectively). Mean concentration of DON in P1 (Nar) was numerically higher than in P2 (Des), being 715 and 620.63 µg.kg−1, respectively (P < 0.05). Mean concentration of T-2/HT-2 toxin was 44.88 and 55.04 µg.kg−1 in P1 (Nar) and P2 (Des), respectively. The content of ZEA in P1 (Nar) of 54.65 µg.kg−1 was lower than in P2 (Des) being 78.72 µg.kg−1 (P < 0.05).

The effects of different levels of grazing utilization (two, three and four grazing cycles per year) and mineral fertilization (nil-fertilization; N100P30K60) on the botanical composition of permanent grasslands were studied in the locality of Rapotín (Czech Republic, 332 m a.s.l.) from 2003–2010. The vegetation of the experimental pasture was classified as Cynosurion. It was found that moderate treatment (three grazing cycles per year) without mineral fertilization showed the highest value of diversity index (DI = 6.08), and maximum dominance of legumes (Dmax = 9.1%), particularly Trifolium repens. The highest dominance of grasses (Dmax = 77.7%), mainly Dactylis glomerata and Elytrigia repens, was achieved with the fertilized treatment utilized in two grazing cycles per year. Based on RDA results, tested management treatments explained 26% of species composition variability, where effect of number of grazing cycles per year was five-times higher than effect of fertilization. We recommend grassland utilization in three grazing cycles per year as the most suitable way from the objective of both species diversity and botanical composition of pastures in similar site conditions. Pasture fertilization should be more controlled by careful consideration of individual pasture goals, actual nutrient status of the soil and possible environmental risks.

The approach to working capital management significantly affects the performance of companies. Nevertheless, this effect varies depending on the observed industry and company’s size and it may be assumed that it is also dependent on territory differences. The paper presents an empirical research aiming to identify particular links between net working capital and costs of the company. The outcomes indicate a relatively strong positive correlation between the variables, especially in case of inventory. Furthermore he ratio of financial costs to ordinary costs is low, as well as the impact of net working capital components on financial costs. It follows that a focus on collection period would not lead to significant savings. The findings appropriately complement Czech and foreign literature focused more on impact of net working capital or working capital on profitability indicators. Further studies concerning a more detailed analysis of the influence of net working capital on corporate costs are difficult to be found. The present research has been conducted on medium-sized companies located in the Czech Republic and manufacturing machinery and equipment. The obtained results suggest the most suitable area of focus for optimization of working capital in relation to costs for the types of companies defined above.

Voltammetric behavior of muscle relaxant dantrolene sodium (DAN) was studied and the voltammetric methods for its determination using polished and mercury meniscus modified silver solid amalgam electrodes (p-AgSAE and m-AgSAE) as well as using bismuth film electrode (BiFE, ex situ plating on GCE) have been proposed. These working electrodes represent the most commonly used alternatives to mercury ones which come wrongfully into disfavor because of alleged toxicity of mercury. Within this work, the obtained results of DAN determination have been completed by corresponding statistical parameters and also some electrochemical characteristics of AgSAEs and BiFE were assessed, especially in comparison with the mercury electrodes.

The objective of this research was to investigate occurrence of common couch grass (Elytrigia repens (L.) Desv. ex Nevski) under different grassland management by means of a small-plot trial. The trial was managed during 2003–2011 with four levels of utilisation: intensive (4 cuts per year, 1st cut on 15 May), medium intensive (3 cuts per year, 1st cut on 30 May), low intensive (2 cuts per year, 1st cut on 15 June), and extensive (2 cuts per year, 1st cut on 30 June). Four rates of fertilisation were applied for each utilisation treatment: nil-fertilisation, P30K60, N90P30K60, and N180P30K60 (pure nutrients). The intensively utilised treatments without N fertilisation showed the lowest mean percentage cover of couch (3.0 %, and 2.8 %, respectively). Further, we found a significant increase in couch grass cover (up to about 30 %) as a response to nitrogen fertilisation and it was most significant in grasslands utilised with two cuts per year. It can be concluded that grassland utilisation in three cuts per year and the dose 90 kg.ha−1 of nitrogen led to the maintenance of the occurrence of couch at the reasonable level up to 10–15 %.

The sequences of the closely linked barley dehydrin genes Dhn4 and Dhn7 are both known to vary allelically. Here we associated allelic constitution at Dhn4/7 with frost sensitivity across a panel of 30 diverse barley varieties. The combined presence of a 6 bp insertion in exon1 of Dhn4 and a 30 bp deletion in exon1 of Dhn7 was restricted to six-rowed winter and intermediate varieties characterised by relatively higher frost tolerance (12 genotypes; LT50 from -14.2°C to -15.6°C). The alternative combination was present with one exception (sixrowed winter variety Alissa) only in spring and two-rowed winter varieties (17 genotypes; LT50 from -10.0°C to -14.3°C). The genetic linkage between Dhn4 and Dhn7 identified e.g. in Dictoo and Morex varieties was verified by segregation analysis of F2 plants from a cross between two genotypes carrying different allelic combination of Dhn4 and Dhn7 genes (two-rowed spring variety Akcent × six-rowed winter variety Okal). The potential of the former allelic combination as a marker for enhanced frost tolerance was tested in a sample of F5 derivatives of a cross between the two-rowed winter type variety Monaco (Akcent allele combination) and the six-rowed winter type variety Okal. Plants with the Okal allele combination showed significantly higher frost tolerance than those with the alternative growth habit. The effect of ear type on frost tolerance was insignificant.

Although the character of countryside is still changing as a consequence of the historical development, agriculture (farming of the land) always represents one of its essential attributes. The authors of this paper analyze the current structure of agricultural workers, their views, opinions and attitudes toward their work, as well as the perception of agriculture by the public.

The purpose of this study is to investigate the time dependent growth of silica shells on CdTe quantum dots to get their optimum thicknesses for practical applications. The core/shell structured silica-coated CdTe quantum dots (CdTe/SiO2 QDs) were synthesized by the Ströber process, which used CdTe QDs co-stabilized by mercaptopropionic acid. The coating procedure used silane primer (3-mercaptopropyltrimethoxysilane) in order to make the quantum dots (QDs) surface vitreophilic. The total size of QDs was dependent on both the time of silica shell growth in the presence of sodium silicate, and on the presence of ethanol during this growth. The size of particles was monitored during the first 72 h using two principally different methods: Dynamic Light Scattering (DLS), and Scanning Electron Microscopy (SEM). The data obtained by both methods were compared and reasons for differences discussed. Without ethanol precipitation, the silica shell thickness grew slowly and increased the nanoparticle total size from approximately 23 nm up to almost 30 nm (DLS data), and up to almost 60 nm (SEM data) in three days. During the same time period but in the presence of ethanol, the size of CdTe/SiO2 QDs increased more significantly: up to 115 nm (DLS data) and up to 83 nm (SEM data). The variances occurring between silica shell thicknesses caused by different methods of silica growth, as well as by different evaluation methods, were discussed.

This article deals with the results of the research and development-led archaeological excavations at the execution sites in the Czech Republic and Lower Silesia (Poland). It summarises the gallows (their construction and materials used), the execution site, which also doubles as the burial place for the executed criminals and those who committed suicide. These deceased, who were excluded from the Christian cemetery, were buried here often without any care: they were thrown in various anatomical positions and orientations. That’s why this article also refers to the related issue of the interpretation of the medieval and modern atypical burials.

In 2003–2005 it was solved the issue about the nutrient balance in the permanent grasslands of Research Institute for Cattle Breeding Rapotín. In trial it was used these types of utilization: 1. Intensive – 4 cuts per year (1st cut up to 15th May, every next after 45 days), 2. Medium intensive – 3 cuts per year (1st cut from 16th to 31st May, every next after 60 days), 3. Low intensive – 2 cuts per year (1st cut up to 15th June, 2nd after 90 days), 4. Extensive – 2 cuts per year (1st cut from 16th to 30th June, 2nd after 90 days). There were following variants of the fertilization: A – no fertilization, B – P:K 30:60 kg.ha – 1, C – N:P:K 90:30:60 kg.ha – 1, D – N:P:K 180:30:60 kg.ha – 1. There was the considerable reaction of the permanent grassland on the nutrient application. The highest value of the coefficient of the neutral efficiency was by the fertilization N:P:K 90:30:60 kg of the pure nutrients. By the most of the variants we have noticed the negative nitrogen balance. The negative balance of the phosphorous was marked only by the unfertilized variants. All variants of the permanent grassland utilization had the negative potassium balance. The utilization had the significant influence (P0.01) to all observed indicators. The year had the significant influence too. From the point of view of the rational plant nutrition and of the environment protection from the contamination (especially from the nitrates) it appears as the most advisable the medium intensive type of the utilization with nitrogen dose: N:P:K 90:30:60.

In 2003 and 2004 was carried out the energy analysis of the different types of permanent grassland utilization on the Hrubý Jeseník locality. There were estimated values of the particular entrances of additional energy. Energy entrances moved according to the pratotechnologies from 2.17 GJ. ha–1 to 22.70 GJ.ha–1. The biggest share on energy entrances had fertilizers. It was 84.93% by the nitrogen fertilisation. The most energy benefit of brutto and nettoenergy was marked by the low intensive utilisation (33.40 GJ.ha–1 NEL and 32.40 GJ.ha–1 NEV on average). The highest value of energy efficiency (13.23%) was marked by the low intensive utilization of permanent grassland. By using of higher doses of industrial fertilizers has energy efficiency decreased. From view of energy benefit and intensiveness on energy entrances it appears the most available utilisation of permanent grassland with three cuts per year (first cut on May 31st at the latest, every next after 60 days) or two cuts per year (first cut on July 15th, next cuts after 90 days).

Tertiary amines 3-(dialkylamino)-2-hydroxypropyl 4-[(alkoxycarbonyl)amino]benzoates and their quaternary ammonium salts were synthesized. The final step of synthesis of quaternary ammonium salts was carried out by microwave-assisted synthesis. Software-calculated data provided the background needed to compare fifteen new resulting compounds by their physicochemical properties. The acid dissociation constant (pKa) and lipophilicity index (log P) of tertiary amines were determined; while quaternary ammonium salts were characterized by software-calculated lipophilicity index and surface tension. Biological evaluation aimed at testing acetylcholinesterase and butyrylcholinesterase-inhibiting activity of synthesized compounds. A possible mechanism of action of these compounds was determined by molecular modelling study using combined techniques of docking; molecular dynamics simulations and quantum mechanics calculations.
