Abstract This data descriptor presents a curated dataset for pathogen detection and identification (Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans) directly from whole-blood samples. The dataset was created using differential cell lysis combined with rapid extraction, digestion, and mass spectrometry-based proteomics. Our method offers a rapid diagnostic alternative to traditional culture, enabling timely disease management, such as sepsis. Highlighting our dataset’s uniqueness, it features a three-tier structure: Spectral Libraries of Pathogens for identifying peptide peaks for putative biomarkers; Spiked pathogen in blood MS data for biomarker panel optimization through varied concentration samples; and Parallel Reaction Monitoring (PRM) data from sepsis patients for validating our biomarker panel, achieving 83.3% sensitivity within seven hours without microbial enrichment culture. This dataset serves as a comprehensive reference for bioinformatic tool development and biomarker panel proposals, advancing microbial detection, antimicrobial resistance, and epidemiological studies.

RESUMO Foram mapeadas sete classes de uso e cobertura do solo da composição urbana por meio de imagens orbitais do satélite Quickbird para estudo de bacia hidrográfica situada em área urbana. Foi objetivo deste estudo gerar uma Carta de Uso e Ocupação do Solo em nível detalhado da Bacia do Arroio dos Pereiras, Irati-PR, as áreas das classes utilizadas e respectivos percentuais, fornecendo subsídios a estudos sobre possíveis ocorrências de enchentes na região central do Município de Irati, Paraná. Como resultado, foram obtidos os percentuais de 22,56% para a classe Florestas, 0,22% para Lagoas, 9,68% para Ruas e Calçadas, 4,84% para Áreas Permeáveis Dentro das Quadras, 5,73% para Áreas Impermeáveis Dentro das Quadras, 10,53% para Telhados e 46,45% para vegetação rasteira.

This paper introduces the Kabsch Marker Estimation Algorithm (KMEA), a new, robust multi-marker localization method designed for Autonomous Mobile Robots (AMRs) within Industry 4.0 (I4.0) settings. By integrating the Kabsch Algorithm, our approach significantly enhances localization robustness by aligning detected fiducial markers with their known positions. Unlike conventional methods that rely on a limited subset of visible markers, the KMEA uses all available markers, without requiring the camera’s extrinsic parameters, thereby improving robustness. The algorithm was validated in an I4.0 automated warehouse mockup, with a four-stage methodology compared to a previously established marker estimation algorithm for reference. On the one hand, the results have demonstrated the KMEA’s similar performance in standard controlled scenarios, with millimetric precision across a set of error metrics and a mean relative error (MRE) of less than 1%. On the other hand, KMEA, when faced with challenging test scenarios with outliers, showed significantly superior performance compared to the baseline algorithm, where it maintained a millimetric to centimetric scale in error metrics, whereas the other suffered extreme degradation. This was emphasized by the average reduced results of error metrics from 86.9% to 92% in Parts III and IV of the test methodology, respectively. These results were achieved using low-cost hardware, indicating the possibility of even greater accuracy with advanced equipment. The paper details the algorithm’s development, theoretical framework, comparative advantages over existing methods, discusses the test results, and concludes with comments regarding its potential for industrial and commercial applications by its scalability and reliability.

RESUMO O objetivo deste trabalho foi buscar associações entre os parâmetros de qualidade da água e a identificação de prováveis agentes poluidores em uma bacia hidrográfica do Sudoeste do Paraná. Foram espacializadas informações sobre a área de estudo, estabelecendo relações entre componentes do uso da terra rural e urbano, e os resultados apresentados pelas análises fisicoquímicas e biológicas realizadas em dois pontos amostrais do Rio Santa Rosa. Foram utilizadas ortoimagens do satélite SPOT 5 e uma carta do Serviço Autônomo Paranacidade. Foi utilizado o aplicativo de domínio público SPRING versão 5.1.6. Ao se comparar a qualidade da água com a Resolução CONAMA 357 (2005), para rios classe II, foram encontrados índices acima da normalidade para óleos e graxas, boro, ferro dissolvido, coliformes totais e coliformes termotolerantes. Assim, foram investigadas prováveis relações entre os índices encontrados acima do limite permitido e os possíveis agentes poluidores.

The pursuit of cosmetic ingredients with proven efficacy and safety that meet consumer needs drives the advancement of new products. Ascorbic acid (AA) is utilized in cosmetic products, predominantly for its potent antioxidant properties. Nonetheless, its instability compromises its efficacy. In this work, ascorbyl 2-O-glucoside persulfate (AAGS) was synthesized, characterized, and evaluated regarding its safety profile and potential bioactivities and the results were compared to AA and its glycoside AAG. Pre-formulation studies were performed to assess the stability of the compounds and their compatibility with typical excipients commonly used in topical formulations. AAGS did not affect the metabolic activity of keratinocyte, macrophage, and monocyte cell lines, up to 500 µM. AAGS also exhibited a non-prooxidant and non-sensitizing profile and anti-allergic activity by impeding the allergen-induced maturation of THP-1 cells. When compared to AA and AAG, AAGS was shown to be more stable at pH values between 5 and 7, as well as superior thermostability and photostability. AAGS demonstrated higher stability in metal solutions of Fe(II) and Mg(II) than AA. AAGS demonstrated similar DPPH radical scavenging activity compared to AA. These results provide useful information for the development of new AA derivatives, highlighting AAGS as a novel cosmetic ingredient.

Acute kidney injury (AKI) is a critical systemic complication caused by Bothrops envenoming, a neglected health problem in the Brazilian Amazon. Understanding the underlying mechanisms leading to AKI is crucial for effectively mitigating the burden of this complication. This study aimed to characterize the urinary protein profile of Bothrops atrox snakebite victims who developed AKI. We analyzed three groups of samples collected on admission: healthy subjects (controls, n = 10), snakebite victims who developed AKI (AKI, n = 10), and those who did not evolve to AKI (No-AKI, n = 10). Using liquid-chromatography tandem mass spectrometry, we identified and quantified (label-free) 1190 proteins. A panel of 65 proteins was identified exclusively in the urine of snakebite victims, with 32 exclusives to the AKI condition. Proteins more abundant or exclusive in AKI's urine were associated with acute phase response, endopeptidase inhibition, complement cascade, and inflammation. Notable proteins include serotransferrin, SERPINA-1, alpha-1B-glycoprotein, and NHL repeat-containing protein 3. Furthermore, evaluating previously reported biomarkers candidates for AKI and renal injury, we found retinol-binding protein, beta-2-microglobulin, cystatin-C, and hepcidin to be significant in cases of AKI induced by Bothrops envenoming. This work sheds light on physiological disturbances caused by Bothrops envenoming, highlighting potential biological processes contributing to AKI. Such insights may aid in better understanding and managing this life-threatening complication.

Objective: To evaluate the efficacy of the balloon spacer when used to reduce the radiation dose delivered to the rectum in prostate cancer patients undergoing external beam radiotherapy. Method: A single center retrospective analysis including 75 PC patients with localized T1-T3a disease who received balloon spacer followed by EBRT. Pre- and post-implantation computed tomography (CT) scans were utilized for treatment planning for standard EBRT (78–81 Gy in 1.8–2 Gy fractions). Rectal dosimetry was assessed using DVHs, and toxicities were graded with CTCAE v.4. Results: A median (IQR) prostate-rectum separation resulted in 1.6 cm (1.4–2.0) post balloon spacer implantation. Overall, 90.6 % (68/75) of patients had a clinically significant 25 % relative reduction in the rectal with a median relative reduction of 91.8 % (71.2–98.6 %) at rV70. Three (4.0 %) patients reported mild procedural adverse events, anal discomfort and dysuria. Within 90 days post-implantation, five patients (6.67 %) and 1 patient (1.33 %) reported grade 1 and grade 2 rectal toxicities (anal pain, constipation, diarrhea and hemorrhoids). Genitourinary (GU) grade 1 toxicity was reported in 37 patients (49.33 %), with only one patient (1.33 %) experiencing grade 2 GU toxicity. No grade ≥ 3 toxicity was reported. Conclusion: Balloon spacer implantation effectively increased prostate-rectum separation and associated with dosimetric gains EBRT for PC stage T1-T3a. Further controlled studies are required to ascertain whether this spacer allows for radiotherapy dose escalation and minimizes gastrointestinal (GI) toxicity.

The simulation results confirm the system’s exceptional precision and resilience in deep space missions, marking a significant advancement in astronautics and paving the way for future space exploration endeavors.

SpliceProt 2.0 is a public proteogenomics database that aims to list the sequence of known proteins and potential new proteoforms in human, mouse, and rat proteomes. This updated repository provides an even broader range of computationally translated proteins and serves, for example, to aid with proteomic validation of splice variants absent from the reference UniProtKB/SwissProt database. We demonstrate the value of SpliceProt 2.0 to predict orthologous proteins between humans and murines based on transcript reconstruction, sequence annotation and detection at the transcriptome and proteome levels. In this release, the annotation data used in the reconstruction of transcripts based on the methodology of ternary matrices were acquired from new databases such as Ensembl, UniProt, and APPRIS. Another innovation implemented in the pipeline is the exclusion of transcripts predicted to be susceptible to degradation through the NMD pathway. Taken together, our repository and its applications represent a valuable resource for the proteogenomics community.

Defect detection is a key element of quality control in today’s industries, and the process requires the incorporation of automated methods, including image sensors, to detect any potential defects that may occur during the manufacturing process. While there are various methods that can be used for inspecting surfaces, such as those of metal and building materials, there are only a limited number of techniques that are specifically designed to analyze specialized surfaces, such as ceramics, which can potentially reveal distinctive anomalies or characteristics that require a more precise and focused approach. This article describes a study and proposes an extended solution for defect detection on ceramic pieces within an industrial environment, utilizing a computer vision system with deep learning models. The solution includes an image acquisition process and a labeling platform to create training datasets, as well as an image preprocessing technique, to feed a machine learning algorithm based on convolutional neural networks (CNNs) capable of running in real time within a manufacturing environment. The developed solution was implemented and evaluated at a leading Portuguese company that specializes in the manufacturing of tableware and fine stoneware. The collaboration between the research team and the company resulted in the development of an automated and effective system for detecting defects in ceramic pieces, achieving an accuracy of 98.00% and an F1-Score of 97.29%.

Surface defect detection with machine learning has become an important tool in industries and a large field of study for researchers or workers in recent years. It is necessary to have a simplified source of information that helps us to better focus on one type of surface. In this systematic review, we present a classification for surface defect detection based on convolutional neural networks (CNNs) focused on surface types. Findings: Out of 253 records identified, 59 primary studies were eligible. Following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, we analyzed the structures of each study and the concepts related to defects and their types on surfaces. The presented review is mainly focused on finding a classification for the types of surfaces most used in industry (metal, building, ceramic, wood, and special). We delve into the specifics of each surface category, offering illustrative examples of their applications within both industrial and laboratory settings. Furthermore, we propose a new taxonomy of machine learning based on the obtained results and collected information. We summarized the studies and extracted the main characteristics such as type of surface, problem types, timeline, type of network, techniques, and datasets. Among the most relevant results of our analysis, we found that the metallic surface is the most used, as it is the one found in 62.71% of the studies, and the most prevalent problem type is classification, accounting for 49.15% of the total. Furthermore, we observe that transfer learning was employed in 83.05% of the studies, while data augmentation was utilized in 59.32%. Our findings also provide insights into the cameras most frequently employed, along with the strategies adopted to address illumination challenges present in certain articles and the approach to creating datasets for real-world applications. The main results presented in this review allow for a quick and efficient search of information for researchers and professionals interested in improving the results of their defect detection projects. Finally, we analyzed the trends that could open new fields of study for future research in the area of surface defect detection.

Background & Aims: Two recently developed composite models, the alpha-fetoprotein (AFP) score and Metroticket 2.0, could be used to select patients with hepatocellular carcinoma (HCC) who are candidates for liver transplantation (LT). The aim of this study was to compare the predictive performance of both models and to evaluate the net risk reclassification of post-LT recurrence between them using each model’s original thresholds. Methods: This multicenter cohort study included 2,444 adult patients who underwent LT for HCC in 47 centers from Europe and Latin America. A competing risk regression analysis estimating sub-distribution hazard ratios (SHRs) and 95% CIs for recurrence was used (Fine and Gray method). Harrell’s adapted c-statistics were estimated. The net reclassification index for recurrence was compared based on each model’s original thresholds. Results: During a median follow-up of 3.8 years, there were 310 recurrences and 496 competing events (20.3%). O presente artigo descreve a implementação da metodologia de aprendizagem baseada em problemas (ABP) no âmbito da unidade curricular de Publicidade e Relações Públicas, do 2.º ano da licenciatura em Marketing. A ABP é considerada uma abordagem global que tem sido reconhecida pelos académicos. Os resultados mostraram que a metodologia ABP tem resultados similares entre diferentes grupos. Todos os grupos de alunos estavam fortemente motivados, à exceção de um grupo que não demonstrou essas evidências por não ter a assiduidade suficiente para aproveitar o acompanhamento do professor. Em geral, todos os alunos puderam melhorar a colaboração entre os seus pares, bem como capacidades de comunicação. Demonstraram também evidências de uma postura ativa e de um trabalho autónomo. A inovação e a criatividade foram evidentes nos resultados finais. Em conclusão, a abordagem ABP pode influenciar os alunos de uma forma positiva.

O ruído é um problema da sociedade moderna e um poluente ambiental causado por atividades industriais, de tráfego, entre outras, podendo gerar problemas à saúde. Neste contexto, esse trabalho tem por finalidade avaliar o potencial de áreas verdes urbanas como barreira atenuadora de ruídos. Para isto, foi realizada uma análise de proximidade entre amostras de ruído mensurados em 10 locais da área urbana considerando a densidade de áreas verdes e a densidade de tráfego de veículos. Foi realizado o mapeamento por classe de níveis de tráfego urbano, das áreas verdes, no entorno de 10 pontos amostrais e calculadas as respectivas taxas de áreas verdes com uso de imagens orbitais de alta resolução espacial. Para as mesmas áreas foram calculadas densidades de tráfego urbano. Foi avaliada a influência atenuadora de ruídos devido a presença de maiores ou menores taxas de áreas verdes urbanas, caracterizadas como áreas verdes arbóreas e que possam funcionar como barreiras verticais verdes atenuadoras de ruídos, como também a interferência da frequência de veículos em função da densidade de tráfego veicular. Desta forma, buscou-se relações estatísticas entre os níveis de ruído encontrados e seus diferentes graus de urbanização. Os resultados demonstraram que as altas frequências de veículos, tanto a 100 como a 250 metros de proximidade dos pontos amostrais, influenciaram com maior intensidade de ruído e que as áreas verdes arbóreas reduziram marginalmente, porém tal redução não foi significativa.

Robotic competitions are an excellent way to promote innovative solutions for the current industries’ challenges and entrepreneurial spirit, acquire technical and transversal skills through active teaching, and promote this area to the public. In other words, since robotics is a multidisciplinary field, its competitions address several knowledge topics, especially in the STEM (Science, Technology, Engineering, and Mathematics) category, that are shared among the students and researchers, driving further technology and science. A new competition encompassed in the Portuguese Robotics Open was created according to the Industry 4.0 concept in the production chain. In this competition, RobotAtFactory 4.0, a shop floor, is used to mimic a fully automated industrial logistics warehouse and the challenges it brings. Autonomous Mobile Robots (AMRs) must be used to operate without supervision and perform the tasks that the warehouse requests. There are different types of boxes which dictate their partial and definitive destinations. In this reasoning, AMRs should identify each and transport them to their destinations. This paper describes an approach to the indoor localization system for the competition based on the Extended Kalman Filter (EKF) and ArUco markers. Different innovation methods for the obtained observations were tested and compared in the EKF. A real robot was designed and assembled to act as a test bed for the localization system’s validation. Thus, the approach was validated in the real scenario using a factory floor with the official specifications provided by the competition organization.

Erythrina velutina is a Brazilian native tree of the Caatinga (a unique semiarid biome). It is widely used in traditional medicine showing anti-inflammatory and central nervous system modulating activities. The species is a rich source of specialized metabolites, mostly alkaloids and flavonoids. To date, genomic information, biosynthesis, and regulation of flavonoids remain unknown in this woody plant. As part of a larger ongoing research goal to better understand specialized metabolism in plants inhabiting the harsh conditions of the Caatinga, the present study focused on this important class of bioactive phenolics. Leaves and seeds of plants growing in their natural habitat had their metabolic and proteomic profiles analyzed and integrated with transcriptome data. As a result, 96 metabolites (including 43 flavonoids) were annotated. Transcripts of the flavonoid pathway totaled 27, of which EvCHI, EvCHR, EvCHS, EvCYP75A and EvCYP75B1 were identified as putative main targets for modulating the accumulation of these metabolites. The highest correspondence of mRNA vs. protein was observed in the differentially expressed transcripts. In addition, 394 candidate transcripts encoding for transcription factors distributed among the bHLH, ERF, and MYB families were annotated. Based on interaction network analyses, several putative genes of the flavonoid pathway and transcription factors were related, particularly TFs of the MYB family. Expression patterns of transcripts involved in flavonoid biosynthesis and those involved in responses to biotic and abiotic stresses were discussed in detail. Overall, these findings provide a base for the understanding of molecular and metabolic responses in this medicinally important species. Moreover, the identification of key regulatory targets for future studies aiming at bioactive metabolite production will be facilitated.

The CTDI100 underestimates the dose indicator due the scattering beyond the scanning length of 100mm of the measurement. A proposed method to obtain the dose (including the “tail” of the dose distribution) is a measurement on a large phantom to estimate the dose indicator on an ideal infinitum large phantom (equilibrium dose). As the scanning length increases, the dose indicator grows asymptotically. Measuremente previously obteined with three tied phantoms of PMMA were adopted in the present work. It includes a set of length measured for different combinations of protocols of voltage, pitch, diameter and axis of measurement. Using an empirically characterized function of the experimental data, two methods to estimate the equilibrium dose were evaluated. One method is manipulating algebraically of the function, and another is the use of Levenberg-Marquardt least-squares. The comparative evaluation of both methods demonstrate that in higher values of the pitch results in a higher difference of both methods. The results demonstrate a good compatibility of the methods for the estimation of the quantity Deq. However, the results for alfa and Leq are significantly different when both methods are considered.

O processo de urbanização do município de Irati-PR ocorreu de forma intensa e acelerada, sendo inevitável a impermeabilização do solo por meio de edificações, calçamentos das ruas, passeios e pátios. Devido a esse processo, diversos fatores do meio natural são modificados, por exemplo, o solo, reduz a capacidade de infiltração, implicando no aumento da taxa de escoamento superficial e de sua velocidade, contribuindo significativamente para ocorrência de inundações. Diante disso, o objetivo do trabalho foi avaliar a verificar a efetividade do uso de imagens de ARP para análise da taxa de permeabilidade em um bairro da cidade de Irati, Paraná, utilizando imagens de altíssima resolução espacial. A técnica de vetorização sobre tela permitiu mapear as áreas permeáveis dentro dos lotes e estimar em quilômetros quadrados sua ocupação. Dispondo do percentual de permeabilidade do solo pode ser realizada a confrontação com a taxa de permeabilidade mínima exigida na legislação municipal. O resultado do estudo demonstrou que dos 82 lotes analisados 47,56% estão irregulares conforme a legislação. Por meio de resultados obtidos, foram avaliadas as condições de cada uma das quadras em relação à confrontação com a legislação vigente.

Polymer single-screw extrusion is a major industrial processing technique used to obtain plastic products. To assure high outputs, tight dimensional tolerances, and excellent product performance, extruder screws may show different design characteristics. Barrier screws, which contain a second flight in the compression zone, have become quite popular as they promote and stabilize polymer melting. Therefore, it is important to design efficient extruder screws and decide whether a conventional screw will perform the job efficiently, or a barrier screw should be considered instead. This work uses multi-objective evolutionary algorithms to design conventional and barrier screws (Maillefer screws will be studied) with optimized geometry. The processing of two polymers, low-density polyethylene and polypropylene, is analyzed. A methodology based on the use of artificial intelligence (AI) techniques, namely, data mining, decision making, and evolutionary algorithms, is presented and utilized to obtain results with practical significance, based on relevant performance measures (objectives) used in the optimization. For the various case studies selected, Maillefer screws were generally advantageous for processing LDPE, while for PP, the use of both types of screws would be feasible.

Localization is a crucial skill in mobile robotics because the robot needs to make reasonable navigation decisions to complete its mission. Many approaches exist to implement localization, but artificial intelligence can be an interesting alternative to traditional localization techniques based on model calculations. This work proposes a machine learning approach to solve the localization problem in the RobotAtFactory 4.0 competition. The idea is to obtain the relative pose of an onboard camera with respect to fiducial markers (ArUcos) and then estimate the robot pose with machine learning. The approaches were validated in a simulation. Several algorithms were tested, and the best results were obtained by using Random Forest Regressor, with an error on the millimeter scale. The proposed solution presents results as high as the analytical approach for solving the localization problem in the RobotAtFactory 4.0 scenario, with the advantage of not requiring explicit knowledge of the exact positions of the fiducial markers, as in the analytical approach.

When the COVID-19 pandemic hits Portugal in early March 2020, medical doctors, engineers and researchers, with the encouragement of the Northern Region Health Administration, teamed up to develop and build, locally and in a short time, a ventilator that might eventually be used in extreme emergency situations in the hospitals of northern Portugal. This letter tells you the story of Pneuma, a low-cost emergency ventilator designed and built under harsh isolation constraints, that gave birth to derivative designs in Brazil and Morocco, has been industrialized with 200 units being produced and is now looking forward to the certification as a medical device that will possibly support a go-to-market launch. Open intellectual property (IP), multidisciplinarity teamwork, fast prototyping and product engineering have shortened to a few months an otherwise quite longer idea-to-product route, clearly demonstrating that when scientific and engineering knowledge hold hands great challenges can be successfully faced.

Background & Aims: Patients with hepatocellular carcinoma (HCC) are selected for liver transplantation (LT) based on pre-LT imaging ± alpha-foetoprotein (AFP) level, but discrepancies between pre-LT tumour assessment and explant are frequent. Our aim was to design an explant-based recurrence risk reassessment score to refine prediction of recurrence after LT and provide a framework to guide post-LT management. Methods: Adult patients who underwent transplantation between 2000 and 2018 for HCC in 47 centres were included. A prediction model for recurrence was developed using competing-risk regression analysis in a European training cohort (TC; n = 1,359) and tested in a Latin American validation cohort (VC; n=1,085). Results: In the TC, 76.4% of patients with HCC met the Milan criteria, and 89.9% had an AFP score of ≤2 points. The recurrence risk reassessment (R3)-AFP model was designed based on variables independently associated with recurrence in the TC (with associated weights): ≥4 nodules (sub-distribution of hazard ratio [SHR] = 1.88, 1 point), size of largest nodule (3–6 cm: SHR = 1.83, 1 point; >6 cm: SHR = 5.82, 5 points), presence of microvascular invasion (MVI; SHR = 2.69, 2 points), nuclear grade >II (SHR = 1.20, 1 point), and last pre-LT AFP value (101–1,000 ng/ml: SHR = 1.57, 1 point; >1,000 ng/ml: SHR = 2.83, 2 points). Wolber’s c-index was 0.76 (95% CI 0.72–0.80), significantly superior to an R3 model without AFP (0.75; 95% CI 0.72–0.79; p = 0.01). Four 5-year recurrence risk categories were identified: very low (score = 0; 5.5%), low (1–2 points; 15.1%), high (3–6 points; 39.1%), and very high (>6 points; 73.9%). The R3-AFP score performed well in the VC (Wolber’s c-index of 0.78; 95% CI 0.73–0.83). Conclusions: The R3 score including the last pre-LT AFP value (R3-AFP score) provides a user-friendly, standardised framework to design post-LT surveillance strategies, protocols, or adjuvant therapy trials for HCC not limited to the Milan criteria. Lay summary: Considering discrepancies between pre-LT tumour assessment and explant are frequent, reassessing the risk of recurrence after LT is critical to further refine the management of patients with HCC. In a large and international cohort of patients who underwent transplantation for HCC, we designed and validated the R3-AFP model based on variables independently associated with recurrence post-LT (number of nodules, size of largest nodule, presence of MVI, nuclear grade, and last pre-LT AFP value). The R3-AFP model including last available pre-LT AFP value outperformed the original R3 model only based on explant features. The final R3-AFP scoring system provides a robust framework to design post-LT surveillance strategies, protocols, or adjuvant therapy trials, irrespective of criteria used to select patients with HCC for LT.

Considering the current importance watercourses quality conservation, it is important to establish relationships between parameters that enable evaluation of the origins of changes in water quality, allowing actions to mitigate them. However, it is important to improve the association of different variables and to take sufficient samples. This study associates usual techniques and parameters to analyse the water quality of an urban river from Paraná State, Brazil. For this, we used biological indicators (aquatic macroinvertebrates), physical-chemical (temperature, turbidity, true colour, pH, DO and BOD5,20) indicators and microbiological (faecal and total coliforms) indicators. These indicators were related to land use and occupation classes obtained from high resolution QuickBird 2 images. For this association, the surroundings (450 meters buffer) of three distinct points of the river were considered: I. Near the spring; II. In the downtown city; and III. In a residential neighbourhood. Different values of physical, chemical and microbiological variables were detected along the river, showing evident relationships between them and with the use and occupation of the urban and peri-urban space in the characterization of surface waters. The association design was able to detect the landscape effect on water quality in a coherent way and that these connections were mainly related to suppression of the riparian forest present in the surroundings, further demonstrating the importance of this vegetation for the maintenance of watercourse quality.

BACKGROUND Leishmaniasis is a neglected tropical disease caused by the parasite Leishmania braziliensis, commonly found in Brazil and associated with cutaneous and visceral forms of this disease. Like other organisms, L. braziliensis has an enzyme called glutamine synthetase (LbGS) that acts on the synthesis of glutamine from glutamate. This enzyme plays an essential role in the metabolism of these parasites and can be a potential therapeutic target for treating this disease. OBJECTIVES Investigate LbGS structure and generate structural models of the protein. METHODS We use the method of crosslinking mass spectrometry (XLMS) and generate structural models in silico using I-TASSER. FINDINGS 42 XLs peptides were identified, of which 37 are explained in a monomeric model with the other five indicating LbGS dimerization and pentamers interaction region. The comparison of 3D models generated in the presence and absence of XLMS restrictions probed the benefits of modeling with XLMS highlighting the inappropriate folding due to the absence of spatial restrictions. MAIN CONCLUSIONS In conclusion, we disclose the conservation of the active site and interface regions, but also unique features of LbGS showing the potential of XLMS to probe structural information and explore new drugs.

Solving real-world multi-objective optimization problems using Multi-Objective Optimization Algorithms becomes difficult when the number of objectives is high since the types of algorithms generally used to solve these problems are based on the concept of non-dominance, which ceases to work as the number of objectives grows. This problem is known as the curse of dimensionality. Simultaneously, the existence of many objectives, a characteristic of practical optimization problems, makes choosing a solution to the problem very difficult. Different approaches are being used in the literature to reduce the number of objectives required for optimization. This work aims to propose a machine learning methodology, designated by FS-OPA, to tackle this problem. The proposed methodology was assessed using DTLZ benchmarks problems suggested in the literature and compared with similar algorithms, showing a good performance. In the end, the methodology was applied to a difficult real problem in polymer processing, showing its effectiveness. The algorithm proposed has some advantages when compared with a similar algorithm in the literature based on machine learning (NL-MVU-PCA), namely, the possibility for establishing variable–variable and objective–variable relations (not only objective–objective), and the elimination of the need to define/chose a kernel neither to optimize algorithm parameters. The collaboration with the DM(s) allows for the obtainment of explainable solutions.

Developing innovative systems and operations to monitor forests and send alerts in dangerous situations, such as fires, has become, over the years, a necessary task to protect forests. In this work, a Wireless Sensor Network (WSN) is employed for forest data acquisition to identify abrupt anomalies when a fire ignition starts. Even though a low-power LoRaWAN network is used, each module still needs to save power as much as possible to avoid periodic maintenance since a current consumption peak happens while sending messages. Moreover, considering the LoRaWAN characteristics, each module should use the bandwidth only when essential. Therefore, four algorithms were tested and calibrated along real and monitored events of a wildfire. The first algorithm is based on the Exponential Smoothing method, Moving Averages techniques are used to define the other two algorithms, and the fourth uses the Least Mean Square. When properly combined, the algorithms can perform a pre-filtering data acquisition before each module uses the LoRaWAN network and, consequently, save energy if there is no necessity to send data. After the validations, using Wildfire Simulation Events (WSE), the developed filter achieves an accuracy rate of 0.73 with 0.5 possible false alerts. These rates do not represent a final warning to firefighters, and a possible improvement can be achieved through cloud-based server algorithms. By comparing the current consumption before and after the proposed implementation, the modules can save almost 53% of their batteries when is no demand to send data. At the same time, the modules can maintain the server informed with a minimum interval of 15 min and recognize abrupt changes in 60 s when fire ignition appears.

Background & Aims: Good outcomes after liver transplantation (LT) have been reported after successfully downstaging to Milan criteria in more advanced hepatocellular carcinoma (HCC). We aimed to compare post-LT outcomes in patients receiving locoregional therapies (LRT) before LT according to Milan criteria and University of California San Francisco downstaging (UCSF-DS) protocol and ‘all-comers’. Methods: This multicentre cohort study included patients who received any LRT before LT from Europe and Latin America (2000–2018). We excluded patients with alpha-foetoprotein (AFP) above 1,000 ng/ml. Competing risk regression analysis for HCC recurrence was conducted, estimating subdistribution hazard ratios (SHRs) and corresponding 95% CIs. Results: From 2,441 LT patients, 70.1% received LRT before LT (n = 1,711). Of these, 80.6% were within Milan, 12.0% within UCSF-DS, and 7.4% all-comers. Successful downstaging was achieved in 45.2% (CI 34.8–55.8) and 38.2% (CI 25.4–52.3) of the UCSF-DS group and all-comers, respectively. The risk of recurrence was higher for all-comers (SHR 6.01 [p

Nowadays, the automation of factory floors is necessary for extensive manufacturing processes to meet the ever-increasing competitiveness of current markets. The technological advances applied to the digital platforms have led many businesses to automate their manufacturing processes, introducing robotic manipulators collaborating with human operators to achieve new productivity, manufacturing quality, and safety levels. However, regardless of the amount of optimization implemented, some quality problems may be introduced in production lines with many products being designed and produced. This project proposes a solution for feature extraction that can be applied to automatic shape- and position-detection using a 2-dimension (2D) industrial laser to extract 3-dimension (3D) data where the movement of the item adds the third dimension through the laser’s beam. The main goal is data acquisition and analysis. This analysis will later lead to the generation of trajectories for a robotic manipulator. The results of this application proved reliable given their small measurement error values of a maximum of 2 mm.

Counterfeit medicine is still a crucial problem for healthcare systems, having a huge impact in worldwide health and economy. Medicine packages can be traced from the moment of their production until they are delivered to the costumers through the use of Data Matrix codes, unique identifiers that can validate their authenticity. Currently, many practitioners at hospital pharmacies have to manually scan such codes one by one, a very repetitive and burdensome task. In this paper, a system which can simultaneously scan multiple Data Matrix codes and autonomously introduce them into an authentication database is proposed for the Hospital Pharmacy of the Centro Hospitalar de Vila Nova de Gaia/Espinho, E.P.E. Relevant features are its low cost and its seamless integration in their infrastructure. The results of the experiments were encouraging, and with upgrades such as real-time feedback of the code’s validation and increased robustness of the hardware system, it is expected that the system can be used as a real support to the pharmacists.

Abstract Background RNA-binding proteins (RBPs) are well known as key factors in gene expression regulation in eukaryotes. These proteins associate with mRNAs and other proteins to form mRNP complexes that ultimately determine the fate of target transcripts in the cell. This association is usually mediated by an RNA-recognition motif (RRM). In the case of trypanosomatids, these proteins play a paramount role, as gene expression regulation is mostly posttranscriptional. Despite their relevance in the life cycle of Trypanosoma cruzi, the causative agent of Chagas’ disease, to date, few RBPs have been characterized in this parasite. Results We investigated the role of DRBD2 in T. cruzi, an RBP with two RRM domains that is associated with cytoplasmic translational complexes. We show that DRBD2 is an ortholog of the Gbp2 in yeast, an SR-rich protein involved in mRNA quality control and export. We used an immunoprecipitation assay followed by shotgun proteomics and RNA-seq to assess the interaction partners of the DRBD2-mRNP complex in epimastigotes. The analysis identified mostly proteins involved in RNA metabolism and regulation, such as ALBA1, ALBA3, ALBA4, UBP1, UBP2, DRBD3, and PABP2. The RNA-seq results showed that most of the transcripts regulated by the DRBD2 complex mapped to hypothetical proteins related to multiple processes, such as to biosynthetic process, DNA metabolic process, protein modification, and response to stress. Conclusions The identification of regulatory proteins in the DRBD2-mRNP complex corroborates the important role of DRBD2 in gene expression regulation in T. cruzi. We consider these results an important contribution to future studies regarding gene expression regulation in T. cruzi, especially in the field of RNA-binding proteins.

Dimorphandra gardneriana Tul. is a legume, the fruits of which are used in routine extraction by the pharmaceutical industry. The objective was to study the morphological aspects of seeds, germination, seedlings, and young plants of Dimorphandra gardneriana Tul. The study was conducted at the Laboratory of Seed Analysis (LAS), Center of Agricultural Sciences, Federal University of Paraíba (CCA-UFPB), Areia - PB. The external morphological characteristics observed of the seeds were color, texture, and consistency of the teguments; seed shape; hilum and micropyle position; color and thickness of raphe and other structures present; besides internal embryo characteristics (cotyledons, hypocotyl-radicle axis, plumule, and central cylinder); and presence of the endosperm. The seed imbibition curve was determined by the weight of the seeds over a period of 90 h, evaluated every 6 h, with germination being considered as the protrusion of the primary root, or the moment when the primary root breaks the integument, and normal seedlings were evaluated on the 5th day after sowing. Most of the morphological characteristics identified in the plants and seedlings of D. gardneriana resembled those observed for other species of the same genus. The seeds of D. gardneriana Tul., were medium sized, with an average length of 11.95 mm, width of 5.33 mm, and thickness of 4.3 mm, unitegmic with a hard tegument, and impermeable to water. The process of imbibition of the seeds was initially slow and the three-phase curve of imbibition was not clear. The germination was epigeic and phanerocotiledonar and began on the 5th day after sowing.

Anthropic activities as well as land use and occupation are closely linked to water and soil quality. An alternative to assess their influence on water quality in water bodies is through biomonitoring using Allium cepa as a bioindicator organism. This technique allows the detection of cytogenetic abnormalities in Allium cepa root meristematic cells after contact with analyzed water samples. Thus, we evaluated the genotoxic potential of water samples from the Rio das Antas (Antas River) along the urban perimeter of the city of Irati, Paraná, Brazil. With the aid of GeoEye Satellite high-resolution fused orbital images, we searched for possible relationships between the use and occupation of land around this river. Biomonitoring was performed at three different sampling points. Statistical equality between the negative control and Point 1 and between Points 2 and 3 was obtained using Fisher's test and a Principal Coordinate Analysis (PCoA). The former presented a low frequency of abnormalities chromosomes and the latter presented larger averages. In addition, Point 1 had strong influence of tree vegetation. Points 2 and 3, demonstrated a strong influence of urbanized area, with significant degradation of permanent preservation areas (áreas de preservação permanente – (APP). The results showed remarkable anthropogenic interference to the ecosystem. Furthermore, this indicates importance of an APP watercourses functioning to preserve the quality of the water resources.

Abstract Background Worldwide, breast cancer is the main cause of cancer mortality in women. Most cases originate in mammary ductal cells that produce the nipple aspirate fluid (NAF). In cancer patients, this secretome contains proteins associated with the tumor microenvironment. NAF studies are challenging because of inter-individual variability. We introduced a paired-proteomic shotgun strategy that relies on NAF analysis from both breasts of patients with unilateral breast cancer and extended PatternLab for Proteomics software to take advantage of this setup. Methods The software is based on a peptide-centric approach and uses the binomial distribution to attribute a probability for each peptide as being linked to the disease; these probabilities are propagated to a final protein p-value according to the Stouffer’s Z-score method. Results A total of 1227 proteins were identified and quantified, of which 87 were differentially abundant, being mainly involved in glycolysis (Warburg effect) and immune system activation (activated stroma). Additionally, in the estrogen receptor-positive subgroup, proteins related to the regulation of insulin-like growth factor transport and platelet degranulation displayed higher abundance, confirming the presence of a proliferative microenvironment. Conclusions We debuted a differential bioinformatics workflow for the proteomic analysis of NAF, validating this secretome as a treasure-trove for studying a paired-organ cancer type.

Abordaremos as formas de controle social realizadas pelas agências reguladoras, no Brasil, com ênfase ao caso das telecomunicações, exercido através da denominada Agência Nacional de Telecomunicações – Anatel, abordando os aspectos jurídicos, dos mecanismos utilizados como meio de controle social para acompanhar o dinamismo da sociedade nesse setor, sendo que destacamos os pontos positivos e negativos dessa participação popular, para melhor adequação do sistema ao que estabelece a Constituição Federal de 1988, bem como, a legislação extravagante, sobre serviços públicos, após as reformas constitucionais e administrativas, o que culminou na descentralização, com as privatizações e, consequentemente, a necessidade de regulamentação da prestação desses serviços públicos e, com base nessas reflexões apresentadas, propusemos algumas ações que poderiam ser adotadas para promover uma melhor participação da sociedade no processo regulatório, dando mais credibilidade ao papel exercido pela Anatel e definindo qual é o papel do cidadão consumidor, frente às dificuldades apresentadas.

Background. IgE mediates type I hypersensitivity reaction and can be found in the mucosa of organs affected by allergy. Acute appendicitis (AA) is a common disease, but its etiology remains poorly understood. Here, we investigated IgE deposition in histological sections of AA samples to test the hypothesis that an allergic reaction may substantially contribute to the pathophysiology of AA. Materials and Methods. In a retrospective study, we assessed the presence of IgE in appendicular specimens of histologically confirmed appendicitis and in the control group, comprised of negative appendicitis and incidental appendectomies, using a monoclonal antibody against human IgE. Samples from 134 appendectomies were included: 38 phlegmonous and 27 gangrenous appendicitis from the study group and 52 incidental appendectomies and 17 negative appendicitis from the control group. The slides were visualized by light microscopy, and a standard procedure was used to manually count the positive IgE staining cells. Results. IgE staining was present in the cells of all but 5 appendicular specimens. Cancer, a major world public health problem, is associated with chemotherapy treatments whose administration leads to secondary concerns, such as oral mucositis (OM). The OM disorder is characterized by the presence of ulcers in the oral mucosa that cause pain, bleeding, and difficulty in ingesting fluids and solids, or speaking. Bioactive compounds from natural sources have arisen as an effective approach for OM. This review aims to summarize the new potential application of different natural products in the prevention and treatment of OM in comparison to conventional ones, also providing a deep insight into the most recent clinical studies. Natural products, such as Aloe vera, Glycyrrhiza glabra, Camellia sinensis, Calendula officinalis, or honeybee crops, constitute examples of sources of bioactive compounds with pharmacological interest due to their well-reported activities (e.g., antimicrobial, antiviral, anti-inflammatory, analgesic, or wound healing). These activities are associated with the bioactive compounds present in their matrix (such as flavonoids), which are associated with in vivo biological activities and minimal or absent toxicity. Finally, encapsulation has arisen as a future opportunity to preserve the chemical stability and the drug bioa vailability of bioactive compounds and, most importantly, to improve the buccal retention period and the therapeutic effects.

The transdermal administration of nonsteroidal anti-inflammatory drugs (NSAIDs) is a valuable and safer alternative to their oral intake. However, most of these drugs display low water solubility, which makes their incorporation into hydrophilic biopolymeric drug-delivery systems difficult. To overcome this drawback, aqueous solutions of bio-based deep eutectic solvents (DES) were investigated to enhance the solubility of ibuprofen, a widely used NSAID, leading to an increase in its solubility of up to 7917-fold when compared to its water solubility. These DES solutions were shown to be non-toxic to macrophages with cell viabilities of 97.4% (at ibuprofen concentrations of 0.25 mM), while preserving the anti-inflammatory action of the drug. Their incorporation into alginate-based hydrogels resulted in materials with a regular structure and higher flexibility. These hydrogels present a sustained release of the drug, which is able, when containing the DES aqueous solution comprising ibuprofen, to deliver 93.5% of the drug after 8 h in PBS. Furthermore, these hydrogels were able to improve the drug permeation across human skin by 8.5-fold in comparison with the hydrogel counterpart containing only ibuprofen. This work highlights the possibility to remarkably improve the transdermal administration of NSAIDs by combining new drug formulations based on DES and biopolymeric drug delivery systems.

This paper presents a systematic mapping literature review about the mobile robotics competitions that took place over the last few decades in order to obtain an overview of the main objectives, target public, challenges, technologies used and final application area to show how these competitions have been contributing to education. In the review we found 673 papers from 5 different databases and at the end of the process, 75 papers were classified to extract all the relevant information using the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) method. More than 50 mobile robotics competitions were found and it was possible to analyze most of the competitions in detail in order to answer the research questions, finding the main goals, target public, challenges, technologies and application area, mainly in education.

The focus of interest for this article is the musical composition movement in Bahia, in so far as it enables a reflection on the interweaving of composition and cultural perspectives, not as isolated and autonomous domains, but as something inherent to composition. We will identify and reflect upon strategies to represent distance between distinct cultural poles, showing how they organize the musical form and fabric, in other words, the production of meaning. We start with an exemplary case provided by the piece Uma possível resposta op. 169 (1988) by the Brazilian composer (born in Switzerland), Ernst Widmer (1927-1990), and analyze similar strategies in other pieces, showing how the idea of establishing a distance which functions as a matrix of meaning responds to our interest in the connection between composition and culture, and permits a discussion of decolonization strategies present in the concert music produced in Bahia in recent decades.

Background: In patients with peripheral intravenous catheters (PIVCs), performing flushing is an essential procedure to maintain catheter patency and prevent complications. These PIVC related complications can lead to premature removal and therapeutics interruption, which implies the need of a new catheterization thus increasing patient discomfort and pain. Aims: To identify nursing practices related to the flushing procedure, namely: moment(s) of the flushing; the syringe size used; the flush solution, volume and technique; the knowledge and accomplishment of the recommended standards on flushing by nurses. Methods: A cross-sectional study was conducted between July and December 2017, with Brazilian and Portuguese nurses. An online questionnaire was developed based on the international recommendations on flushing procedure. Descriptive analysis was performed. Results: A total of 76 nurses answered the questionnaire. The majority of nurses (84.2%) performed flushing: the most common technique used was continuous syringe pressure (31.2%), with the push-pause technique being performed by 23.4% of the nurses. Despite the majority performs flushing at four distinct moments (after the PIVC insertion, before, between and after drug delivery), there are inconsistencies in flush solution, volume, and syringe size. The most used volume to perform flushing was 5 mL, filled using normal saline. Despite this, they also recognized the omission of this procedure due to time constrains, no familiarity with the procedure and unavailable material. Conclusions: This study identified that flushing procedure isn't always performed by nurses in their clinical practice. Also, several inconsistencies were observed between nurses that performed flushing, reflecting the lack of empirical evidence in this area of research.

BackgroundIn nursing practice, flushing the catheters pre and post-drug administration is considered an important clinical procedure to prevent complications, and requires the use of several syringes to comply with international standards of care. We envisioned an innovative double-chamber syringe that enables the filling and administration of both solutions. Following current international recommendations, the development of new medical devices should integrate Health Technology Assessment. The Human-centred design is usually used for that assessment purposes, as a method that actively include end-users in the devices development process.MethodApplication of the Human-Centred Design through the involvement of nurses in the initial stages of the device development in order to accomplish the initial stages of Technology Readiness Level. A multi-method approach was used, including literature/guidelines review, focus groups with end-users and expert panels.ResultsThe involvement of nurses enabled the definition of user requirements and contexts of use, as well as the evaluation of design solutions and prototypes in order to accomplish with usability and ergonomic features of the medical device.ConclusionsSignificant contributions were made regarding the final design solution of this innovative double-chamber syringe.

O objetivo deste estudo foi investigar a gestão das Associações Atléticas Acadêmicas - A.A.A. na ótica do gestor de esportes da A.A.A. Participaram do estudo 27 gestores ou representantes de esportes das A.A.A no município de São Paulo. Foi utilizado um questionário com perguntas abertas e fechadas de modo a estudar e compreender o perfil do gestor de esportes das A.A.A. tentando levantar sua identidade profissional e abrangência de seu trabalho, através da análise das respostas coletadas. Conclui-se que a média de idade dos gestores é de 23,4 anos, sendo que 88,9% deles ainda não concluíram a graduação, para uma gestão majoritariamente de um ou dois anos. Observou-se que 51,9% representam mais de um curso, 33,3% a Instituição de Ensino Superior e 14,8%, um único curso, com orçamentos anuais entre R$ 10.000,00 e R$ 60.000,00.

The knowledge of the areas susceptible to the occurrence of floods contributes to the urban planning and to the minimization of the effects to which the population is exposed. This study aimed to identify sites that are prone to floods in the urban area of the main hydrographic basin of the city of Irati, State of Paraná. For this mapping, we used a Geographic Information System (GIS) associated with methodologies including fuzzy logic and hierarchical analysis. For that, criteria for analysis were the altitude, slope, distance of water courses and degree of urbanization. The results showed that 3,645 km² of the Irati urban perimeter, about 13,8% of the study area, present some degree of susceptibility to floods. The generation of maps with degrees of susceptibility varying from 0 (null) to 1 (high) was a good visual indicator to identify points of greater or lesser risk of riverbed extravasation

En la sociedad actual, con Internet, hay infinidad de textos. Por eso resulta necesario seleccionar los más atractivos para el alumnado con el objetivo de fomentar el gusto por la lectura. Este trabajo analiza durante una semana cada uno de los enunciados que aparecen en las portadas de los periódicos deportivos de España (As, Marca, Mundo Deportivo y Sport) y Portugal (A Bola, O Jogo y Record). Con su análisis se pretende demostrar que la mayoría de los enunciados son retóricos, y que esto ocurre en ambos países. Se ha optado por los periódicos deportivos porque despiertan interés, y resulta un tema conocido para los más jóvenes. No hay que olvidar que los textos deportivos son tan creativos como los publicitarios (Guerrero Salazar, 2007). A través de una propuesta didáctica concreta se muestra cómo trabajar con títulos tan sugerentes como “Por todo lo alto” (Marca) y “¡¡Mamma mia!! (MD) o “Aviso Amarelo” (A Bola) o “Dragão à prova de ressaca” (O Jogo) para enseñar a escribir con la expresividad y emotividad. Requisitos que toda creación literaria necesita para tener éxito, para ser atractivo para los lectores.

Background and aims. Heterogeneous data has been reported regarding liver transplantation (LT) for hepatocellular carcinoma (HCC) in Latin America. We aimed to describe treatment during waiting list, survival and recurrence of HCC after LT in a multicenter study from Latin America.Material and methods. Patients with HCC diagnosed prior to transplant (cHCC) and incidentally found in the explanted liver (iHCC) were included. Imaging-explanted features were compared in cHCC (non-discordant if pre and post-LT were within Milan, discordant if pre-LT was within and post-LT exceeding Milan).Results. Overall, 435 patients with cHCC and 92 with iHCC were included. At listing, 81% and 91% of cHCC patients were within Milan and San Francisco criteria (UCSF), respectively. Five-year survival and recurrence rates for cHCC within Milan, exceeding Milan/within UCSF and beyond UCSF were 71% and 16%; 66% and 26%; 46% and 55%, respectively. Locoregional treatment prior to LT was performed in 39% of cHCC within Milan, in 53% beyond Milan/within UCSF and in 83% exceeding UCSF (p < 0.0001). This treatment difference was not observed according to AFP values ( 1,000 ng/mL 64%; p = 0.12). Discordant imaging-explanted data was observed in 29% of cHCC, showing lower survival HR 2.02 (CI 1.29; 3.15) and higher recurrence rates HR 2.34 when compared to AFP 1,000 ng/mL at listing was independently associated with a higher 5-year recurrence rate and a HR of 3.24 when compared to AFP

The Automatic Dependent Surveillance Broadcast (ADS-B) protocol can be used as a complement to Radar-based Air Traffic Control (ATC), or as a replacement in areas where the latter is impractical. However, ADS-B messages do not include any integrity or authenticity thus they can be easily tampered using inexpensive hardware and open source software. Modifying ADS-B message format is infeasible because it would render the already large implemented base obsolete. This paper proposes ADS-Bsec, which addresses the ADS-B limitations while being compatible with the installed base. ADS-Bsec provides the missing integrity and authenticity into ADS-B packets using a key management infrastructure without altering the existing packet format. This paper also argues for including the proposed framework as a component of the NextGen infrastructure based on our experience in prototyping efforts using cognitive radios, cryptography and multilateration together with existing radar-based ATC systems.
