Mephedrone is a psychoactive drug that increases dopamine, serotonin and noradrenaline levels in the central nervous system via interaction with transporters or monoamines. The aim of the presented study was to assess the role of the GABA-ergic system in the expression of mephedrone-induced reward. For this purpose, we conducted (a) a behavioral evaluation of the impact of baclofen (a GABAB receptors agonist) and GS39783 (a positive allosteric modulator of GABAB receptors) on the expression of mephedrone-induced conditioned place preference (CPP) in rats, (b) an ex vivo chromatographic determination of the GABA level in the hippocampi of rats subchronically treated with mephedrone and (c) an in vivo evaluation of GABA hippocampal concentration in rats subchronically administered with mephedrone using magnetic resonance spectroscopy (MRS). The results show that GS39783 (but not baclofen) blocked the expression of CPP induced by (20 mg/kg of) mephedrone. The behavioral effect was consistent with chromatographic analysis, which showed that mephedrone (5 and 20 mg/kg) led to a decrease in GABA hippocampal concentration. Altogether, the presented study provides a new insight into the involvement of the GABA-ergic system in the rewarding effects of mephedrone, implying that those effects are at least partially mediated through GABAB receptors, which suggests their potential role as new targets for the pharmacological management of mephedrone use disorder.

The SOLARIS synchrotron located in Krakow, Poland, is a third-generation light source operating at medium electron energy. The first synchrotron light was observed in 2015, and the consequent development of infrastructure lead to the first users’ experiments at soft X-ray energies in 2018. Presently, SOLARIS expands its operation towards hard X-rays with continuous developments of the beamlines and concurrent infrastructure. In the following, we will summarize the SOLARIS synchrotron design, and describe the beamlines and research infrastructure together with the main performance parameters, upgrade, and development plans.

The countermovement jump test (CMJ) is commonly used as an indicator of the anaerobic capacity of the lower limbs. The main objective of this study was to evaluate the relationship between the CMJ with arm swing and the Wingate anaerobic test (WAnT) among female volleyball and football players. A group of first league female volleyball club players (VG, n = 12, age = 24.2 years), a group of first league female football club players (FG, n = 12, age = 20.6 years), and a control group (CG, n = 10, age = 20.4 years) participated in this research. The measurements of selected somatic features were carried out. The CMJ and WAnT were used to assess the explosive strength and maximum muscle power of the lower extremities. An analysis of variance test demonstrated differences between the VG and FG in terms of height (p p p r = .82 and r = .57, respectively. There was not statistically significant differences between values of these coefficients. The obtained results showed a potentially lean body mass influence on the results of the CMJ in VG. In women’s football and volleyball, the results of the CMJ are strongly correlated with the power parameters registered with the WAnT. In volleyball, players’ coaches should include a measurement of lean body mass in the assessment peak power by vertical jump tests.

The present study examined the validity of the force-velocity (F-V) mechanical parameters of the lower limbs as a predictors of the competitive performance in junior male speed climbing. The second aim was to evaluate F-V characteristics in competitive speed climbers in the junior male category. Ten junior male speed climbers (age: 18±0.5 years; BH: 174.7±4.5 cm; BW: 66.4±6.6 kg; with mean best climbing time from the competition BCT: 7.29±0.66 s) were participants in the research. All they were participants in the Youth European Cup in speed climbing. F-V characteristics were measured using a cycle ergometer test according to the Vandewalle’s protocol. To evaluate of the validity between BCT (criterion) and mechanical parameters (practical measures) from F-V test a standard error of the estimate (expressed as a raw and standardized typical error, TE) and correlation coefficients (rx,y) were used. Moreover, the validity of BCT as a potentially predictor of final competition rank (as a criterion for BCT) was evaluated. Additionally the mean value and standard deviation were calculated for all measured variables. Validity of the BCT as a competition place predictor was largely overestimated (TE=0.73 and rx,y=0.81, p p≤0.05). Accuracy of the BCT prediction for somatic and mechanical variables were very largely error biased. The standardized values of TE for these variables were: 1.93÷7.25. The rx,y were in the range of -0.18 to -0.46. The level of the mechanical variables from the F-V cycle ergometer as a predictors of the BCT in speed climbing may be largely error biased. Application of the cycle ergometer F-V test of prediction BCT values in speed climbing may have low practical value. Moreover, BCT variable may be a poor predictor of the final competition result. Coaches should take these suggestions into account during preparations to speed climbing competitions.

Mephedrone is a widely used drug of abuse, exerting its effects by interacting with monoamine transporters. Although this mechanism has been widely studied heretofore, little is known about the involvement of glutamatergic transmission in mephedrone effects. In this study, we comprehensively evaluated glutamatergic involvement in rewarding effects of mephedrone using an interdisciplinary approach including (1) behavioural study on effects of memantine (non-selective NMDA antagonist) on expression of mephedrone-induced conditioned place preference (CPP) in rats; (2) evaluation of glutamate concentrations in the hippocampus of rats following 6 days of mephedrone administration, using in vivo magnetic resonance spectroscopy (MRS); and (3) determination of glutamate levels in the hippocampus of rats treated with mephedrone and subjected to MRS, using ion-exchange chromatography. In the presented research, we confirmed priorly reported mephedrone-induced rewarding effects in the CPP paradigm and showed that memantine (5 mg/kg) was able to reverse the expression of this effect. MRS study showed that subchronic mephedrone administration increased glutamate level in the hippocampus when measured in vivo 24 h (5 mg/kg, 10 mg/kg and 20 mg/kg) and 2 weeks (5 mg/kg and 20 mg/kg) after last injection. Ex vivo chromatographic analysis did not show significant changes in hippocampal glutamate concentrations; however, it showed similar results as obtained in the MRS study proving its validity. Taken together, the presented study provides new insight into glutamatergic involvement in rewarding properties of mephedrone. Major depressive disorder is a stress-related disease associated with brain metabolic dysregulation in the glutamine-glutamate/γ-aminobutyric acid (Gln-Glu/GABA) cycle. Recent studies have demonstrated that microbiome-gut-brain interactions have the potential to influence mental health. The hypothesis of this study was that Lactobacillus rhamnosus JB-1 (LR-JB1™) dietary supplementation has a positive impact on neuro-metabolism which can be quantified in vivo using magnetic resonance spectroscopy (MRS). A rat model of depressive-like disorder, chronic unpredictable mild stress (CUMS), was used. Baseline comparisons of MRS and behavior were obtained in a control group and in a stressed group subjected to CUMS. Of the 22 metabolites measured using MRS, stressed rats had significantly lower concentrations of GABA, glutamate, glutamine + glutathione, glutamate + glutamine, total creatine, and total N-acetylaspartate (tNAA). Stressed rats were then separated into 2 groups and supplemented with either LR-JB1™ or placebo and re-evaluated after 4 weeks of continued CUMS. The LR-JB1™ microbiotic diet restored these metabolites to levels previously observed in controls, while the placebo diet resulted in further significant decrease of glutamate, total choline, and tNAA. LR-JB1™ treated animals also exhibited calmer and more relaxed behavior, as compared with placebo treated animals. In summary, significant cerebral biochemical downregulation of major brain metabolites following prolonged stress were measured in vivo using MRS, and these decreases were reversed using a microbiotic dietary supplement of LR-JB1™, even in the presence of continued stress, which also resulted in a reduction of stress-induced behavior in a rat model of depressive-like disorder.

The aim of the study was to evaluate changes in the central visual pathways during the early and advanced stages of bilateral normal-tension glaucoma (NTG). Methods: The studied groups constituted patients with bilateral normal-tension glaucoma of the same stage (n = 45) and age-matched healthy volunteers (n = 17). All patients underwent ophthalmic examination and examination on a 1.5 Tesla Magnetic Resonance Scanner (Optima 360, GE Healthcare). Volume and cortical thickness analyses were performed using the open-source automated software package FreeSurfer. Results: There was a significant difference in lateral geniculate nuclei volume between the control and advanced glaucoma groups in the right hemisphere (p = 0.03) and in the left hemisphere between the early and advanced glaucoma patients (p = 0.026). The optic chiasm volume differed significantly between the control and advanced NTG groups (p = 0.0003) and between early and advanced glaucoma patients (p = 0.004). Mean cortical thickness analysis revealed a significant increase in values in the advanced glaucoma group in the right Brodmann area 17 (BA17) (p = 0.007) and right BA18 (p = 0.049) as compared to early NTG. In the left BA18 area, the mean thickness of the cortex in the early glaucoma group was significantly lower than in the control group (p = 0.03). Conclusions: The increase in the grey matter thickness in the V1 region with more-advanced glaucoma stages may reflect compensatory hypertrophy. Additionally, the regions of the brain early affected during glaucoma with reduced thickness were the right lateral occipital gyrus and left lingual gyrus. The most prominent change during the course of glaucoma was the increase in grey matter thickness in the right cuneus.

The work describes the physical basis of the chemical exchange saturation transfer (CEST) technique; it presents the beginnings of the implementation of the method and its possible applications. The principles of correct data acquisition and possible solutions used during the design of the CEST sequence are shown. The main problems related to data analysis are indicated, and an example Z-spectrum from in vivo study of the rat brain is introduced. Furthermore, the parameters related to spectrum analyses such as magnetisation transfer asymmetry (MTRasym) and amide proton transfer asymmetry (APTasym) are presented. In the following part, different types of the CEST method often mentioned in the literature are discussed. Subsequently, the possible applications of the CEST method in both clinical and experimental practice are described.

The aim of the study was to assess the volume of the lateral geniculate nucleus (LGN) in patients with open-angle glaucoma in 7Tesla MRI and to evaluate its relation to RNFL thickness and VF indices. Material and methods. The studied group consisted of 20 open-angle glaucoma patients with bilaterally the same stage of glaucoma (11 with early glaucoma and nine with advanced glaucoma) and nine healthy volunteers from the Department of Diagnostics and Microsurgery of Glaucoma, Medical University of Lublin, Poland. Circumpapillary RNFL-thickness measurements were performed using OCT in all patients and visual fields were performed in the glaucoma group. A 7Tesla MRI was performed to assess the volume of both lateral geniculate bodies. Results. The LGN volume varied significantly between groups from 122.1 &plusmn, 14.4 mm3 (right LGN) and 101.6 &plusmn, 13.3 mm3 (left LGN) in the control group to 80.2 &plusmn, 17.7 mm3 (right LGN) and 71.8 &plusmn, 14.2 mm3 (left LGN) in the advanced glaucoma group (right LGN p = 0.003, left LGN p = 0.018). However, volume values from early glaucoma: right LGN = 120.2 &plusmn, 26.5 mm3 and left LGN = 103.2 &plusmn, 28.0 mm3 differed significantly only from values from the advanced group (right LGN p = 0.006, left LGN p = 0.012), but not from controls (right LGN p = 0.998, left LGN p = 0.986). There were no significant correlations between visual field indices (MD (mean deviation) and VFI (visual field index)) and LGN volumes in both glaucoma groups. Significant correlations between mean RNFL (retinal nerve fiber layers) thickness and corresponding and contralateral LGN were observed for the control group (corresponding LGN: p = 0.064, contralateral LGN: p = 0.031) and early glaucoma (corresponding LGN: p = 0.017, contralateral LGN: p = 0.008), but not advanced glaucoma (corresponding LGN: p = 0.496, contralateral LGN: p = 0.258). Conclusions. The LGN volume decreases in the course of glaucoma. These changes are correlated with RNFL thickness in early stages of glaucoma and are not correlated with visual field indices.

Cel Celem badań było dokonanie analizy arktycznego sporu terytorialnego pomiędzy Danią a Kanadą toczonego o wsypę Hans. Badanie polegało na kompletnym ujęciu przyczyn sporu, jego przebiegu, a także implikacji na przyszłość. Metoda badań Spór terytorialny o wyspę Hans został zbadany w opraciu o analizę historyczną oraz analizę źródeł pierwotnych i wtórnych. Wyniki Badanie wykazało, iż arktyczny spór terytorialny o wyspę Hans ma historycznie długi przebieg i jak dotąd nie udało się znaleźć jego rozwiązania. Istnieje kilka propozycji rozwiązania sporu, jednak żadna z nich obecnie nie jest brana pod uwagę. Bardzo możliwe, że w przyszłości w rozwiązanie sporu włączy się Organizacja Narodów Zjednoczonych oraz społeczność międzynarodowa. Na przebieg sporu mogą mieć wpływ zmiany klimatyczne postępujace w Arktyce. Wnioski Spór terytorialny o wyspę Hansa różni się od innych sporów. Przebiega za pomocą dyplomatycznych przepychanek niejednokrotnie z elementami humorystycznymi przez co był określony mianem ''wojny whiskey''.

Purpose The aim of the work was to share the practical experience of preclinical and clinical proton magnetic resonance spectroscopy (1HMRS) studies conducted using a 7-Tesla magnetic field strength scanner, taking into account the specificity of both settings in the context of translational research. Material and methods 1HMRS volunteer studies conducted using a Discovery 950 GE 7T scanner, were carried out with PRESS sequence, and a VOI measuring 2.0 × 2.0 × 2.0 cm3 placed in the white matter at the parietal occipital lobe. Rodent spectra obtained using a 7T Bruker were measured with PRESS, with a VOI 2.0 × 2.0 × 5.5 mm3 placed over the hippocampus. Results 1HMRS data from humans and rats show that the brain spectra obtained in the same field are characterised by a similar neurochemical structure and spectral resolution. Spectra obtained from rats demonstrate the following metabolites: NAA, Glu, Gln, Ins, Cho, Cr, PCr, Tau, GABA, Lac, NAAG, and Asp. In turn, spectra from humans allowed estimation of the following metabolites: Ala, NAA, Glu, Gln, Ins, Cho, Cr, PCr, Tau, GABA, Lac, NAAG, and Asp. Signals from Gln, Glu with chemical shift around 2.4 ppm, from Cr, PCr, and GABA at 3 ppm, and signals from Cho and Tau at approximately 3.2 ppm, can be properly separated and estimated both in humans and in rats. Conclusions These results are promising in terms of broadening the knowledge of many neurological diseases by inducing them on animal models and then transferring this knowledge to clinical practice. In spite of this, important distinctions in the technical aspects and methodological differences of high-field 1HMRS in both preclinical and clinical conditions should be taken into account.

Current research on speed climbing at the senior level shows that speed capabilities are a significant factor determining climbing time. The main purpose of our research was to evaluate the level of speed capabilities among young speed climbers and to examine the relationship between the level of speed capabilities and the results achieved during an international competition. The subjects of the research were girls and boys in the following age categories: Female Youth A (age=16.6), Female Youth B (age=14.5 years), Male Youth A (age=16.87 years) and Male Youth B (age=14.8 years). Measurements of somatic features were included: body height, body weight, percentage of fat tissue and BMI. Speed capabilities were measured with Wingate Test. The significance test of differences showed no significant differentiation within climbing times between the groups of girls and the groups of boys. A strong correlation between climbing time and relative peak power was noticed in the Female Youth B group. The correlation between peak power and climbing time was lower in the other groups. Research showed that in speed climbing, as career progresses, it is importance of other factors rather than speed capabilities that may increase. It can be assumed that with an increase in training experience, motor skills become more essential.
