A recovery approach within mental health services has gained momentum. Its meaning for adults with intellectual disabilities recovering from mental health disorders is less understood. Peoples' experiences of recovery were explored to help inform recovery-focused recommendations for clinical practise. A qualitative design using interpretative phenomenological analysis was applied. Nine interviews with people with intellectual disabilities who had experienced mental health disorders were conducted. Two themes that emerged focusing on entry to service and the recovery experience. Subthemes for entry to service included unfair treatment, valuing information and managing expectations. The recovery experience subthemes were therapeutic alliance, self-management, emotional development, autonomy, connectedness, positive identity and a belief in recovery. Hearing peoples' experiences directly allowed the current themes to emerge in the context of living with lifelong disabilities. This article adds to the sparse literature and highlights considerations for recovery-based interventions for people with intellectual disabilities. Previous research has highlighted a lack of continuity of care when young people with a neurodevelopmental condition make the transition from children's to adult specialist healthcare services. A lack of planning, consistency, and availability of adult services has been found to lead to; increased anxiety, poor health outcomes, reduced support and some young people not receiving healthcare. The majority of transition research has focused on what health professionals consider important in the transition process, rather than focusing on the experiences of the young people and those closest to them. Our objective was to gather evidence from young people (and their families) who had experienced transition from children's to adult specialist healthcare services through semi-structured interviews. Volunteers were recruited from two London boroughs. All young people were aged between 18 and 25 years with a neurodevelopmental condition (Attention Deficit Hyperactivity Disorder, Autism Spectrum Disorder and/or an Intellectual Disability). Overall, we interviewed six young people with support from a family member. Five further family members were interviewed on behalf of the young person. In total, ten semi-structured interviews were transcribed verbatim and analysed using Interpretative Phenomenological Analysis. Four themes emerged from the analysis: (a) Parents as advocates, (b) Availability of adult's specialist health and social care services, (c) Lack of information sharing and (d) Transition as a binary, abrupt change. Our findings suggest the transition experience could be improved by changing service specifications to incorporate assessment and handover across the age range of 16-20 years. Additionally, statutory services should understand and provide the coordination role now offered by parents in transition. We suggest future research could evaluate the feasibility of a patient-owned online information sharing tool with information about relevant services for young people and their families.

Aim The main aim of this study was to understand what health and wellbeing initiatives are helpful for healthcare workers' stress and psychological distress during the Covid-19 pandemic. Background Healthcare workers are at increased risk of poor mental health during health emergencies; understanding support required for healthcare workers' is of paramount importance. Methods Participants were healthcare workers at a health and social care organisation (N = 159). The study included an online questionnaire including an evaluation of health and wellbeing initiatives and measures of perceived stress and psychological distress. Results The highest rated resources were counselling, personal protective equipment (PPE), and Covid-19 testing. Those who accessed yoga reported significantly less stress and psychological distress than those who did not access yoga. Conclusions Healthcare workers with higher stress and psychological distress felt less supported by their organisation, less listened to and less involved in organisational decisions. Implications for nursing management Practical implications are discussed such as forward planning for health emergencies (e.g., PPE supply), accessible Covid-19 testing as well as budgeting for counselling services and exercise classes. In addition, targeted support for those diagnosed with Covid-19 is recommended, alongside involvement of staff members in organisational decisions.

Finger millet blast, caused by Magnaporthe grisea, is the most important disease of finger millet in East Africa. Diseased plants are significantly less productive, and most cultivars grown by farmers are susceptible to the disease. Fungicide application is an option for disease management; however, smallholder farmers cannot afford the cost. Host plant resistance is therefore the most viable option for managing the disease. Eighty-one finger millet germplasm accessions from East Africa were evaluated for resistance to blast disease, in natural and inoculated trials. Three accessions (G18, G43, and G67) were identified as resistant to all the three progressive stages of blast: leaf, neck, and panicle. However, one (G3) and four (G15, G16, G60, and G70) accessions were only resistant to leaf and neck blast, respectively. Two resistant (G39 and G43) and 12 moderately resistant (G3, G7, G11, G20, G23, G27, G31, G33, G36, G66, G74, and G81) accessions to blast attained grain yields >2.0 t/ha. These accessions varied in time to maturity, plant height, and grain color, which will enable farmers to select accessions appropriate to their target agro-ecological zones and desired end uses. East African finger millet germplasm has high potential as a source of blast-resistant accessions that could be evaluated for direct production and/or for blast-resistance breeding.

General and specific environmental adaptation of genotypes is the main goal of breeders. However, genotype-by-environment (G × E) interaction complicates the identification of genotypes for release. This study aimed at analyzing the effects of G × E interaction on the expression of important cassava traits using two multivariate analyses: additive main effects and multiplicative interaction (AMMI) and genotype stability index (GSI). Total carotene content (TCC), postharvest physiological deterioration (PPD), and reaction to viral diseases were significantly affected by G × E interaction effects. The low percent (%) variation due to genotype for cassava brown streak disease (CBSD) explained the influence of environment on CBSD expression. The % variation due to genotype for TCC was higher (96%) than variation due to environment (1.7%) and G × E interaction (2.4%) indicating a low interaction effect of environment on TCC accumulation. The % variation due to genotype was higher than % variation due to environment for all traits but CBSD root necrosis and CBSD on stems, indicating the influence of environment on the severity of the viral diseases. These findings indicate that screening for disease resistance requires multi-environment trials, whereas a single-environment trial suffices to screen for total carotene content.

When creating a test platform, the primary focus is often on the selection of test and measurement instrumentation or the test programming environment. Although it is important to ensure that the selected instrumentation can provide the proper stimulus and measurement capabilities, it is also important to consider how you are going to cable and interface the test platform to the Unit Under Test (UUT). High accuracy measurement devices can be undone by a poorly performing interface. Here are some examples; • UUTs incorporating wireless communication will need a test platform to measure high frequency and low power signals. Selecting the wrong cable or not accounting for insertion loss on connectors can severely impact the accuracy of testing. • Measurement devices which are used to test a wide number of products can degrade their instrument's connectors from frequent changing of the test interface or cabling. This can be mitigated by using external test interfaces. In these applications it is important to consider the lifetime of the interconnect used and how easily it can be repaired on-site. Apart from manual bench top testing, automated test applications will involve some signal management, or switching. This adds a whole layer of potential factors that could limit the performance of any test solution if all factors are not taken into account. In all but the simplest test scenarios, test resources will need to be shared between multiple UUT points requiring some kind of switching system and interfacing. This interfacing between test instrumentation, switching system and the UUT could involve connector to connector and connector to UUT wiring. Also, there may be a test fixture interface that is either manually or automatically operated. This requires careful consideration as the cumulative effect of all these factors may significantly degrade the ability to test the UUT to the desired specifications. Another factor to consider is that the testing application may involve adverse environmental conditions. This paper will discuss aspects of test interfacing. While we cannot cover all possible methods in this short paper, we will look at considerations for selecting a test fixture interface, cables and connectors, mass interconnect and RF/Microwave applications. Please note that while we are using some Pickering products as examples, these recommendations apply to any test vendor or platform.

One of the attempts by farmers in counteracting the devastating effects of cassava brown streak disease (CBSD) on yield and quality of cassava is early harvesting. However, most varieties grown by farmers are often late bulking which increases the disease severity while on the other hand early harvesting results in significant yield losses. Farmers, therefore, need early storage root bulking cassava varieties in order to reduce the time to harvest leading to a faster rate of return to investment, while at the same time avoiding devastating effects of CBSD on yield and quality of cassava. The study was, therefore, conducted to identify high-yielding and early storage root bulking cassava genotypes as well as traits associated with early storage root bulking and estimate yield loss if any due to early harvesting. The overall aim was to generate information that would guide future improvement programmes for high-yielding and early-bulking cassava varieties in Malawi and other countries facing similar challenges. Trials were implemented using a square lattice design with three replications at two locations for two growing seasons with three harvest intervals (6, 9 and 12 months after planting, MAP). High yields were obtained of up to 9.5 t/ha at 6 and 17.8 t/ha at 9 MAP. Furthermore, the study revealed that yields obtained at 9 MAP were higher than those obtained at 12 MAP for some genotypes which suggests that such genotypes would be considered as early storage root bulking. Simple correlation analysis identified harvest index, storage root number, storage root diameter and storage root length as the selection criteria to achieve high fresh storage root yield (t/ha) and dry mass yield (t/ha). Path coefficient analysis allocated harvest index and shoot mass as the major selection criteria in improving fresh storage yield and dry mass yield. The study suggests that both source and sink capacities were important for determining early yield. Therefore, these two traits are the key determinants of early storage root bulking and should be used when selecting early-bulking cultivars and indirectly selecting for storage root number, storage root diameter and storage root length.

Alternaria blight (Alternaria spp.) is an important sweetpotato disease in Uganda causing yield losses of over 50 % in susceptible genotypes. In Uganda, Alternaria bataticola and Alternaria alternata are the major species with A. bataticola the more aggressive of the two. The most effective control measure for this disease is the use of resistant genotypes. This study was conducted to determine the inheritance of resistance to Alternaria blight and the general and specific combining abilities of the available germplasm. There is little information on farmers’ knowledge about cassava (Manihot esculenta Crantz) brown streak disease (CBSD), despite extensive studies on incidences and severities. The objective of this study was to assess farmers’ knowledge of CBSD diagnosis and management. The study was conducted in three districts of Malawi by administering semi-structured interviews in combination with disease incidence and severity surveys. Farmers’ knowledge of disease diagnosis and management was associated with CBSD incidence and severity. High levels of knowledge about CBSD were observed in areas with high disease incidence. Only 10.1% of the farmers were capable of identifying the foliar symptoms of the disease. On average, 75.0% and 71.7% of the farms had leaf and storage root incidences, respectively. At harvest, 88.3% of the farmers’ fields exhibited storage root necrosis. Physiological postharvest deterioration (PPD) and late bulking are among the traits that make cassava an unattractive crop in many environments. This study aimed at assessing the main constraints of cassava production, the effects of late bulking, the losses due to PPD and the factors affecting adoption of new cultivars in Rwanda. A participatory rural appraisal (PRA) and a baseline survey were conducted in March-May 2014 in three agro-ecological zones in the country using a multistage sampling method. Cassava was grown on 0.29 ha out of 0.69 ha total average land possession per household. The majority of cassava farmers (59.1 %) practised intercropping as their land holding is small. Average yield was 21.8 t ha−1. A number of constraints was identified, particularly the lack of clean cuttings, viral diseases, late bulking cultivars, drought, limited information and knowledge, weathered soils, insufficient fertilizers, land shortage, lack of markets and effective storage techniques. Loss due to PPD was estimated at 11.9 % of total production per year. Piecemeal harvest and underground storage of roots were the main practices used to delay PPD. Change in colour and taste, rotting, difficulty in removing skin and increase of fibres in the flesh were the farmers’ methods for assessing PPD. Time to harvest varied from district to district and was attributed to genetic x environment interactions. The use of late bulking varieties and the lack of yield production of other crops resulted in reduced food availability and potential food crises. Farmer preferences, information and extension services, performance, quality, market acceptability and cutting production influenced the adoption of new cassava cultivars. Thus, breeding objectives targeting the end user preferences could enhance the adoption of new cultivars.

Cassava (Manihot esculenta Crantz) brown streak disease (CBSD) is currently one of the major constraints to sustained cassava production in Malawi. Its economic impact is mainly manifested in the storage roots where it causes pitting, necrosis and constriction. CBSD can effectively be managed by using resistant varieties as well as early harvesting especially if the varieties are early bulking (high storage root yield). However, development of resistant and high storage root bulking varieties requires an understanding of gene action controlling inheritance of the two traits. Currently, there is very little information in Malawi regarding the inheritance pattern and relative importance of general (GCA) and specific combining ability (SCA) of these two traits. Therefore, a study was conducted to determine mode of gene action as well as importance of GCA and SCA in the genetic control of CBSD resistance and storage root yield. Information generated is essential in the selection of parents and breeding strategies for an effective breeding programme. Cassava is the principal staple root crop, providing a major source of calories for rural and urban households in Luapula province of Zambia. However, the yields on smallholder farms are relatively low largely due to disease infections. The study was therefore conducted to establish farmers’ perceptions, knowledge and management of cassava mosaic disease (CMD), which is one of the major diseases of cassava, and to establish farmers’ preferred traits, constraints and assess sources of cassava cuttings for planting. Focus group discussions (FGD) and structured interviews involving 156 farmers in Mwense, Mansa and Samfya districts were conducted from December 2008 to March 2009. Knowledge of CMD was limited among the respondents. Only 2.4% of the respondents were aware of the disease despite high CMD incidence in farmers’ fields. The majority of the farmers were aware of the importance of insect pests; however, they could not differentiate between damages due to diseases or insect pests. Sixteen cassava genotypes comprising introductions, local landraces and improved genotypes were evaluated for two seasons in Mansa, Zambia, for their reaction to cassava mosaic disease (CMD). The study was conducted in 2009/10 and 2010/11 seasons to evaluate the reaction of cassava cultivars to CMD and agronomic traits. Cassava mosaic disease severity and leaf retention were scored at 6 months after planting (MAP) and data on yield and yield components were recorded at harvest (7 MAP). Significant genotype x season interaction for CMD, harvest index, fresh root yield, biomass, plant height, root size and leaf retention was recorded. Bangweulu, Kalaba, Chikula, Mwakamoya and Chila-7 were the most susceptible genotypes over the two seasons. Mweru, Kampolombo, TMS190, TMS3001, Tanganyika and Nalumino had low severity scores. Harvest index ranged from 0.36 (Mwakamoya) to 0.55 (Chila-7) for the combined seasons. Chila-7 had the highest fresh root yield with a mean of 0.87 kg plant-1 for the combined seasons. The resistant genotypes might be used to improve the CMD resistance of local cultivars through hybridisation.

Physiological postharvest deterioration (PPD) of cassava is the main constraint affecting its nutritional and economical values. PPD is induced by wounds when detaching storage roots from mother plant during harvesting. It is accelerated by the reactive oxygen species (ROS) such as oxygen ion (O2) and peroxide (O2)−2. The carotenoid content and its antioxidant property can help in extending shelf life of cassava storage roots. The primary mode of action of carotenoids as antioxidant is to quench singlet oxygen. Cassava breeding was reported to successfully introgress and improve carotenoid content into cassava. The two types of phytoene synthase (PSY) enzymes (PSY1 and PSY2) are key regulators of carotenoids accumulation in cassava. Carotenoids formation and accumulation in cassava storage roots are induced by a single nucleotide polymorphism in PSY2 which causes a non-conservative amino acid exchange. This single nucleotide polymorphism in PSY gene is co-segregated with β-carotene in cassava storage roots, a phenomenon that could help to unravel the mechanism of introgression of carotenoids into cassava. This article investigates breeding feasibility for improving quality of cassava landraces in developing countries.

Cassava green mite (CGM) Mononychellus tanajoa (Bondar) (Acari: Tetranychidae) is a major arthropod pest causing significant loss in the yields of storage roots and planting materials of cassava in Zambia. Its control has been mainly based on the use of exotic predatory mites as biological control agents, which unfortunately, have not established well in Zambia due to the lack of suitable host genotypes and harsh weather conditions. The current study was aimed at breeding cassava for improvement of morphological traits that are associated with resistance to CGM, which can also enable cassava genotypes to provide shelter and ensure continuous survival of natural enemies of CGM, and to determine the inheritance of these traits by assessing combining ability and therefore the type of gene action involved in their expression. Cassava storage roots start deteriorating 1 to 3 days after harvest due to postharvest physiological deterioration (PPD) which reduces the potential of cassava as a commercial crop. The objective of this study was to examine the extent of genetic variability in cassava for PPD. Genetic variability is important in selecting suitable genotypes for crop improvement. Twelve cassava genotypes were evaluated in a randomized complete block design at three different locations in Uganda for five different harvest times, and PPD assessment was done 7 days after each harvest time. Genotype, location, harvest time, genotype × harvest time and harvest time × location were highly significantly different for PPD. Sunflower (Helianthus annuus L.) production in South Africa depends on natural rainfall which is variable and unpredictable, hence the need to develop drought tolerant sunflower genotypes to stabilise production under random stress environments. An exploratory research study was undertaken to determine the effectiveness of using moisture stress selection environments and three secondary traits, head diameter (HD), stem diameter (SD) and stay green canopy (SG) in developing drought tolerant sunflower cultivars with high oil yield (OY) in the presence of drought stress. A total of 84 testcross hybrids (TCHs) developed by crossing two testers to 42 female cytoplasmic male sterile (CMS) inbred lines were evaluated in nine environments for oil yield and other agronomic traits. The environments were classified as random drought stress environments (RSE), managed drought stress environments (MSE) and non-stress environments (NSE). Type A and type B genetic correlations based on broad sense heritability (H2) and narrow sense heritability (h2), respectively were estimated to quantify indirect selection efficiency. Type A genetic correlations indicated that SG had the potential to be used as a secondary trait to indirectly improve OY. The indirect selection efficiency (ISE) for OY via SG using genetic correlation based on H2 were 0.79, 0.82 and 0.78 in the RSE, MSE and NSE, respectively and genetic correlation based on h2 were 0.67, 0.98, and 0.93 in the RSE, MSE and NSE, respectively. In both cases, selection in the MSE had the highest ISE. Estimates of indirect selection based on type B genetic correlation indicated that indirect selection for OY in the MSE and NSE for the RSE was as effective as direct selection for OY in the RSE based on the additive genetic correlations of 0.96 obtained in both selection environments. Overall, the results from this exploratory study should inform the development of breeding strategies to improve the drought tolerance of sunflower cultivars grown in South Africa and associated environments.

Cassava ( Manihot esculenta Crantz) is an important root crop worldwide. It exhibits substantial differential genotypic responses to varying environmental conditions, a phenomenon termed genotype × environment interaction (GEI). A significant GEI presents challenges in the selection of superior genotypes. The objective of this study was to examine the effect of genotype, environment and GEI on early fresh storage root yield (FSRY) and related traits in cassava. Accordingly, 12 cassava genotypes were evaluated in a randomised complete block design at three contrasting locations (Jinja, Nakasongola and Namulonge) in Uganda. Trials were harvested nine months after planting and the data collected were analysed using the additive main effects and multiplicative interaction (AMMI) model. The AMMI analysis of variance showed significant variation among genotypes for early FSRY and all other traits assessed. Locations were significantly different for all traits except for cassava brown streak disease root necrosis. The GEI effect was non-significant for early FSRY, but significant for other traits. For early FSRY, 48.5% of the treatment sum of squares was attributable to genotypes, 27.3% to environments, and 24.1% to GEI, indicating a predominance of genotypic variation for this trait. Predominance of genotypic variation was also observed for all the other traits. A majority of the genotypes (67%) had low interaction effects with locations for early FSRY, with Akena, CT2, CT4 and NASE14 being the most stable genotypes for the trait. Significant negative correlation was observed between cassava mosaic disease severity and early FSRY and storage root number, indicating significant negative effects of cassava mosaic disease on early FSRY and stability in cassava. The information generated will inform future selection initiatives for superior early-yielding cassava genotypes combining resistance to cassava mosaic and brown streak diseases in Uganda.

Such information is important in the selection of parents and breeding strategies for an effective breeding programme. The objective of this study was to estimate the general combining ability (GCA) of nine cassava parents and their specific combining ability (SCA) for early SRY and disease resistance traits, as well as to determine the gene action controlling these traits. Thirty-six full-sib cassava families were generated from a 9 × 9 half- diallel mating design and were evaluated in two distinct environments in Uganda using a 3 × 12 row by column design. Family, GCA and SCA effects and their interactions with environments were significantly different for most traits, indicating, respectively significant differences in the mean performances of the families, additive and non-additive gene action in the expression of the traits, and the non-additive influence of the environments. The relative importance of additive and non-additive gene action varied between traits, indicating the need for specific breeding strategies for each trait. Parents with desirable GCA effects for most traits were developed from cassava introductions from South America, highlighting their importance and possibility of widening genetic variability of African cassava germplasm. The GCA effects for the parents did not always correlate with their per se performance, implying that selection of parents based on their per se performance may not necessarily lead to development of superior hybrids. © 2014 Elsevier B.V. In this study, genetic diversity in 340 finger millet accessions from Kenya, Tanzania and Uganda and 15 minicore accessions was assessed using 23 single-sequence repeat markers and five qualitative traits. Nineteen markers were polymorphic with a mean polymorphic information content value of 0.606 and a range of 0.035–0.889, with allele size ranging from 148 to 478. A total of 195 alleles were detected (range of 3–23 and average of 10.3 alleles per locus), with 57.7% being rare and 17.4% being private. Differentiation between the accessions of the three countries was weak, with most of the genetic diversity being explained by variability within the countries and subregions than by that among the countries and subregions. The highest genetic diversity was observed in the Kenyan accessions (0.638 ± 0.283) and the least in the Ugandan accessions (0.583 ± 0.264). Combining ability is one of the most important information breeders use to identify superior inbred lines on the basis of their performance in hybrid combinations. The objectives of our study were (i) to quantify the importance of general combining ability (GCA) and specific combining ability (SCA) variances for seed yield, oil content and oil yield; and (ii) estimate GCA and SCA effects of seed yield, oil content and oil yield of inbred lines developed from advanced cycle pedigree breeding populations in sunflower. A total of 109 female S3 cytoplasmic male sterile (CMS) lines from four bi-parental populations in advanced cycle pedigree breeding were crossed with two testers to form 218 testcross hybrids (TCHs). The TCHs were then evaluated in three environments. Variance component analysis results showed predominance of σ2gca over σ2sca for seed yield and oil yield indicating that superior TCHs can be identified based on positive and significant GCA effects of the female lines. For oil content σ2sca was predominant over σ2gca indicating that selecting for TCHs with high oil content would be best among line × tester combinations and not among female S3CMS lines per se. The proportion of GCA and SCA effects in the best five TCHs in each breeding population also confirmed the predominance of GCA effects over SCA effects for seed yield and oil yield while for oil content both GCA and SCA effects appear to be important, with SCA effects having more influence than GCA. The best selection strategy would therefore be to capture the GCA in the early stages of inbreeding and then SCA for the few unique combinations when lines are almost fixed.

Structure-specific DNA endonucleases have critical roles during DNA replication, repair and recombination, yet they also harbor the potential for causing genome instability. Controlling these enzymes may be essential to ensure efficient processing of ad hoc substrates and to prevent random, unscheduled processing of other DNA structures, but it is unknown whether structure-specific endonucleases are regulated in response to DNA damage. Here, we uncover DNA damage-induced activation of Mus81-Eme1 Holliday junction resolvase in fission yeast. This novel regulation requires both Cdc2CDK1 and Rad3ATR-dependent phosphorylations of Eme1. Mus81-Eme1 activation prevents gross chromosomal rearrangements in cells lacking the BLM-related DNA helicase Rqh1. We propose that linking Mus81-Eme1 DNA damaged-induced activation to cell cycle progression ensures efficient resolution of Holliday junctions that escape dissolution by Rqh1-TopIII while preventing unnecessary DNA cleavages.

Cassava mosaic disease (CMD) is caused by cassava mosaic begomoviruses (CMBs), which are transmitted by the whitefly (Bemisia tabaci) in sub-Saharan Africa. CMD and the pathogenic viruses associated with cassava were assessed in a country-wide survey in seven provinces of Zambia between April and May 2009. CMD incidence was highest in Northwestern (71.2%) and lowest in Western (34.3%) provinces. Disease symptoms were severe in Eastern (3.94) and Lusaka (3.88), moderate in Central (3.54), Luapula (3.48) and Northern (3.31) and mild in Northwestern (3.01) and Western (2.50) provinces. In addition, the numbers of adult whitefly were assessed on cassava fields, and found to be highest in Lusaka (2.12) and lowest in Central (0.02) province. The early generation selection of cassava quantitative and qualitative traits saves breeding resources as it can shorten breeding schemes. Inheritance analysis provides important breeding information for developing new improved varieties. This study aimed at developing an F1 segregating cassava population and determining mode of gene action of pulp colour and selected traits at early generation selection (F1 seedling and clones).The 15 families exhibited significant (P 0.05) phenotypic variation between offspring. The general combining ability (GCA) was significant for all traits except cassava brown streak disease on leaves, whereas specific combining ability (SCA) was significant for all evaluated traits. The Garukansubire and Gitamisi genotypes were the best general combiners for improving fresh storage root yield, while G1 and G2 were the best general combiners for improved carotenoid (yellow/orange pulp colour) and delayed physiological postharvest deterioration. The pulp colour had the highest GCA/SCA ratio and percent sum of squares due to GCA.The 15 F1 families exhibited essential genetic diversity for cassava improvement. The expression of most cassava traits was controlled by both additive and non-additive gene action. The study elucidated the role of dominance effects over the additive effects for the evaluated traits. However, the pulp colour was predominantly controlled by additive gene action. As a plant breeding programme matures there is a general tendency to recycle elite inbred parents, a strategy known as advanced cycle pedigree breeding. The challenge with this approach is not only to maintain genetic variability in the base breeding populations but also their usefulness for further genetic advancement in developing experimental hybrids with better performance than the commercial hybrids on the market. To assess the genetic variability and usefulness of breeding populations in sunflower (Helianthus annuus L.), 109 new inbred lines across four breeding populations Pop1, Pop2, Pop3 and Pop4 in advanced cycle pedigree breeding scheme were testcrossed to two testers to form testcross hybrid (TCH) groups: Pop1TC, Pop2TC, Pop3TC and Pop4TC. Moderate to high genetic variability along with high heritability were obtained for seed yield and oil yield within and across all TCH groups. Heritability for oil content was low to high ranging from 0.36 (Pop2TC) to 0.81 (Pop1TC). Genetic advance (GA%) with a 10 % selection intensity ranged from 36 % (Pop2TC) to 42 % (Pop1TC) for seed yield; 38 % (Pop3TC) to 43 % (Pop1TC) for oil yield; and 1.3 % (Pop2TC) to 5.1 % (Pop1TC) for oil content. To quantify the commercial potential of experimental TCHs, founder parent heterosis (FPH), mid-standard heterosis (MSH) and high standard heterosis (HSH) were calculated. Out of a total of 218 experimental TCHs evaluated, 28 had positive FPH, MPH and HSH values for oil yield representing a 13 % selection intensity as is usually applied in early generation testing.

Information on genetic inheritance of farmer preferred traits is fundamental in selection of parents and breeding strategies for an effective participatory plant breeding. Forty sibs representing each of the 24 full-sib families generated using 6 × 4 North Carolina II mating design from ten popular cassava varieties grown in mid-altitude tropical climatic conditions were evaluated in two distinct environments using α-lattice design. Crosses, general and specific combining ability (GCA and SCA) effects and their interaction with environment were significant for most traits evaluated indicating significant genotypic, additive, non-additive gene action and their interaction with environment. Significance of genotypic, GCA, SCA effects and their interaction with environments imply selection of superior genotypes and parents should be based on multi-location evaluation. The presence of both additive and non-additive gene action for most preferred traits indicates the need to have specific breeding strategies that exploits both gene actions. However, the importance of additive over non-additive gene action varied between traits indicating the need specific breeding approaches for these traits. Over 50% of variability within the crosses for all traits evaluated except height to first branching, plant height and branching level was due to SCA effect indicating the importance of non-additive gene action. The GCA effects for the parents did not generally correlate with their per se performance also the best performing crosses were not always developed from parents with high GCA effects. These implied selection of parents based on their per se performance may not necessarily lead to development of superior hybrids.

Genetic improvement for seed yield and oil-content in sunflower cultivars was initiated in the early 1970s in South Africa. Since then no study has been carried out to assess the progress and contribution of new cultivars to seed yield improvement to justify continued investment into breeding new cultivars. The aim of this study was therefore to quantify the contribution of new cultivars to seed yield and associated traits in sunflower over four decades of breeding in South Africa. Two data-sets were used in this study: (1) side-by-side evaluation of historical and current sets of popular cultivars in the same environment under one set of trial management practices; and (2) yield trends in commercial farmers’ fields based on annual production estimates. The absolute genetic gain (yield increase per year) for seed yield in the side-by-side trials ranged from 18 to 32 kg ha−1 year−1 with a mean of 24 kg ha−1 year−1. In contrast, the absolute seed yield increase under commercial production was 12 kg ha−1 year−1. The estimated relative genetic gain for seed yield based on side-by-side trials was 1.5% year−1 and the relative gain in seed yield per year under commercial production was 1.9% year−1. The contribution of new cultivars to total seed yield progress in sunflower were 56.3% for the period 1970–1989; 23.9% from 1990 to 2009 and the mean over the four decades under consideration 1970–2009 was 41.6%. Positive increases in genetic gains for oil yield, oil content and number of seeds per head were also obtained although the gain in oil content was relatively low.

Breeding for tolerance to soybean rust is potentially one of the solutions to combating soybean rust; however, very little progress in tolerance breeding has been documented. Fourteen pre-selected genotypes were evaluated in a sprayed versus unsprayed split-plot trial conducted over three seasons for their tolerance to soybean rust. Different assessments of tolerance were compared for their effectiveness in discriminating between genotypes. Conventional methodology using a yield loss % index for assessing tolerance produced results which were poorly correlated with each other over seasons, which likely accounts for the historic lack of breeding success. This study has used the superiority measure (Pi) of Lin and Binns and the ecovalence statistic (Wi) of Wricke in a biplot to identify the most tolerant genotypes. A novel statistic (WiPi) has been generated from this biplot which facilitates the simultaneous selection for general performance and yield stability of genotypes under a range of rust stress conditions.

Most models of homologous recombination invoke cleavage of Holliday junctions to explain crossing over. The Mus81·Eme1 endonuclease from fission yeast and humans cleaves Holliday junctions and other branched DNA structures, leaving its physiological substrate uncertain. We report here that Schizosaccharomyces pombe mus81 mutants have normal or elevated frequencies of gene conversion but 20- to 100-fold reduced frequencies of crossing over. Thus, gene conversion and crossing over can be genetically separated, and Mus81 is required for crossing over, supporting the hypothesis that the fission yeast Mus81·Eme1 protein complex resolves Holliday junctions in meiotic cells.

Functional studies strongly suggest that the Mus81-Eme1 complex resolves Holliday junctions (HJs) in fission yeast, but in vitro it preferentially cleaves flexible three-way branched structures that model replication forks or 3' flaps. Here we report that a nicked HJ is the preferred substrate of endogenous and recombinant Mus81-Eme1. Cleavage occurs specifically on the strand that opposes the nick, resulting in resolution of the structure into linear duplex products. Resolving cuts made by the endogenous Mus81-Eme1 complex on an intact HJ are quasi-simultaneous, indicating that Mus81-Eme1 resolves HJs by a nick and counternick mechanism, with a large rate enhancement of the second cut arising from the flexible nature of the nicked HJ intermediate. Recombinant Mus81-Eme1 is ineffective at making the first cut. We also report that HJs accumulate in a DNA polymerase alpha mutant that lacks Mus81, providing further evidence that the Mus81-Eme1 complex targets HJs in vivo.

Genome integrity is protected by Cds1 (Chk2), a checkpoint kinase that stabilizes arrested replication forks. How Cds1 accomplishes this task is unknown. We report that Cds1 interacts with Rad60, a protein required for recombinational repair in fission yeast. Cds1 activation triggers Rad60 phosphorylation and nuclear delocalization. A Rad60 mutant that inhibits regulation by Cds1 renders cells specifically sensitive to replication fork arrest. Genetic and biochemical studies indicate that Rad60 functions codependently with Smc5 and Smc6, subunits of an SMC (structural maintenance of chromosomes) complex required for recombinational repair. These studies indicate that regulation of Rad60 is an important part of the replication checkpoint response controlled by Cds1. We propose that control of Rad60 regulates recombination events at stalled forks.

Mus81, a fission yeast protein related to the XPF subunit of ERCC1-XPF nucleotide excision repair endonuclease, is essential for meiosis and important for coping with stalled replication forks. These processes require resolution of X-shaped DNA structures known as Holliday junctions. We report that Mus81 and an associated protein Eme1 are components of an endonuclease that resolves Holliday junctions into linear duplex products. Mus81 and Eme1 are required during meiosis at a late step of meiotic recombination. The mus81 meiotic defect is rescued by expression of a bacterial Holliday junction resolvase. These findings constitute strong evidence that Mus81 and Eme1 are subunits of a nuclear Holliday junction resolvase.

The protein kinase Chk1 enforces the DNA damage checkpoint. This checkpoint delays mitosis until damaged DNA is repaired. Chk1 regulates the activity and localization of Cdc25, the tyrosine phosphatase that activates the cdk Cdc2. Here we report that Mik1, a tyrosine kinase that inhibits Cdc2, is positively regulated by the DNA damage checkpoint. Mik1 is required for checkpoint response in strains that lack Cdc25. Long-term DNA damage checkpoint arrest fails inΔmik1 cells. DNA damage increases Mik1 abundance in a Chk1-dependent manner. Ubiquitinated Mik1 accumulates in a proteasome mutant, which indicates that Mik1 normally has a short half-life. Thus, the DNA damage checkpoint might regulate Mik1 degradation. Mik1 protein and mRNA oscillate during the unperturbed cell cycle, with peak amounts detected around S phase. These data indicate that regulation of Mik1 abundance helps to couple mitotic onset to the completion of DNA replication and repair. Coordinated negative regulation of Cdc25 and positive regulation of Mik1 ensure the effective operation of the DNA damage checkpoint.

The IL-2 growth hormone is the major growth factor of activated T lymphocytes during a developing immune response. IL-2 is required not only for cell cycle progression but also to protect Ag-activated T cells from programmed cell death. In several cell types, activation of NF-κB and/or activating protein-1 (AP-1) has been demonstrated to be extremely important in blocking apoptosis. To determine whether either or both of these transcription factors are involved in cell survival or cell cycle progression in response to IL-2, primary human T cells responsive to the growth factor were analyzed for NF-κB and AP-1 activation. The current study clearly demonstrates that IL-2 does not induce IκBα degradation or NF-κB activation in primary human T cells that respond to IL-2 by entering the cell cycle and avoiding apoptosis. Similarly, IL-2 neither activates JNK nor increases AP-1 binding activity to a consensus o-tetradecanoylphorbol 13-acetate (TPA) response element. On the other hand, the growth factor does induce the activation of STAT3 and STAT5 in these cells, as has been previously demonstrated. These data show that neither NF-κB nor AP-1 activation is required for IL-2-mediated survival or cell cycle progression in activated primary human T cells.

Short-lasting unilateral neuralgiform headache attacks with conjunctival injection and tearing (SUNCT) or with cranial autonomic symptoms (SUNA) are primary headaches characterized by frequent attacks of severe headaches in association with cranial autonomic features. Patients with chronic SUNCT or SUNA have unremitting symptoms that necessitate prolonged use of medical preventive treatments, many of which are prone to causing side effects. They can be medically intractable, in which case neurally destructive or cranially invasive surgical treatments can be offered, though these have hitherto yielded conflicting results. Occipital nerve stimulation (ONS) offers a nondestructive and relatively low risk surgical alternative.To assess the efficacy and safety of ONS in chronic, medically intractable SUNCT and SUNA patients.Prospective open-label study.Nine medically intractable, chronic SUNCT and SUNA patients were implanted with electrodes for bilateral occipital nerve stimulation. Data were collected prospectively for pre- and postimplantation headache characteristics, including frequency, intensity and duration of attacks. Diaries were used to assess headache improvement.At a median follow-up of 38 months (range 24-55 months), all but one patient reported substantial improvement. Four patients became pain-free, 3 were almost pain-free (96 - 98% improvement), and one had a marked reduction in attack frequency and severity (81% improvement). After an initial rapid improvement, the maximum benefit of ONS was attained after a lag of a few months. Device malfunction was followed by recurrence or worsening of the attacks within a few days in most patients. Adverse events included lead migration, exposure of the electrode, and pain due to muscle recruitment over the leads. One patient developed hemicrania continua one month after implantation and was successfully treated with indomethacin.ONS appears to offer an effective and safe treatment option, without significant morbidity, for medically intractable SUNCT and SUNA. Given the variable results with cranially invasive or neurally destructive surgery, ONS might be considered the surgical treatment of choice for medically intractable SUNCT and SUNA.

With the reality of global climate change there is a need to exploit the variation in the germplasm in order to develop genotypes adapted to these changes. This requires breeding and selection of crops at strategically selected locations along a rainfall/temperature gradient to enable farmers select desired cultivars. Eighty one finger millet germplasm lines from East Africa were evaluated in eight environments spread across Kenya, Tanzania and Uganda for adaptation, grain yield stability using the additive main effects and multiplicative interaction (AMMI) ANOVA and Genotype and Genotype x Environment (GGE) models and blast reaction under artificial and natural inoculation. Lanet 2012 long rains, Serere 2012 long rains and Miwaleni 2012 long rains were found to be the most discriminating environments for the low temperature, sub-humid mid-altitude and dry lowland areas, respectively. Alupe 2012 long rains was the ideal environment for blast selection. Seven genotypes were identified for yield stability across the eight environments whereas nine genotypes had specific adaptation. Nine genotypes were identified with resistance to three blast types. However, one and two genotypes had high resistance only to leaf and neck blast, respectively. Two resistant and 12 moderately resistant genotypes to blast attained the highest grain yields and had varied maturity, plant heights and grain colour. This will provide farmers the opportunity to select genotypes appropriate to their target agro-ecologies with desired end-uses. The East African finger millet germplasm has high potential as a source of climate smart high yielding and blast resistant genotypes for direct production and/or breeding.

Tako-tsubo cardiomyopathy is a cardiac syndrome precipitated by profound emotional stress and anxiety, particularly in middle-aged women. It presents as a mimic of acute myocardial infarction, but coronary angiography shows normal coronary arteries and a characteristic left ventriculogram resembling an “octopus pot”. The condition seems to have a favourable prognosis. Initially described in Japan, and with many names in the literature, it is being increasingly recognised in the West owing to early coronary angiography and primary coronary intervention, accounting for up to 1 in 30 primary cases of angioplasty in some institutions. A typical case is described, and the clinical features, pathophysiology and management reviewed.

Short-interval intracortical inhibition (SICI) decreases during voluntary contraction of the target muscle. It is unknown whether this effect also occurs with postural contractions. We have compared the effects of voluntary and postural contractions on SICI in the soleus (SOL) muscle. We applied transcranial magnetic stimuli (TMS) in subjects under three tasks: sitting at rest (Rest), sitting while activating the SOL muscle (Voluntary), or standing quietly (Postural). In control trials, we applied suprathreshold TMS to obtain unconditioned motor-evoked potentials (MEPs). In test trials, the same TMS was preceded by a subthreshold TMS at different interstimulus intervals (ISIs), to obtain a conditioned MEP. SICI and intracortical facilitation (ICF) were expressed as the decrease or increase in MEP size relative to unconditioned MEPs. There was significant effect of task in mean SICI or mean ICF in SOL. Mean SICI in SOL was 52% in Rest and decreased to 21% in Voluntary and 15% in Postural. Mean ICF in SOL was 132% and decreased to 113% in Voluntary and to 108% in Postural. Mean SICI in SOL was not different in Voluntary and Postural tasks. There was no effect of task in mean SICI or mean ICF in TA. Our results indicate that decrease of SICI with muscle contraction occurs to a similar extent with tonic voluntary and postural activation, suggesting that those contractions require a similar type of cortical involvement. However, it cannot be excluded that some part of the SICI reduction with muscle contraction depends on changes in segmental excitability.

The purpose of this study was to assess the prevalence and correlates of comorbid diabetes and Post-Traumatic Stress disorder(PTSD)and potential relationships between PTSD and diabetes outcomes. Male patients enrolled in a VA primary care database (N = 73,270) were classified as having diabetes from pharmacy records (N = 14,438) and grouped into those with diagnoses of PTSD with depression (N = 649), PTSD-only (N = 480), Depression-only (N = 1696), Other psychiatric diagnosis (N = 736), or No psychiatric diagnosis (N = 10,877) based on the Purpose of Visit diagnoses in the medical record. Outcomes included glycemic control (HbA1c), cholesterol and triglycerides. Correlates were age, substance use disorder, other psychiatric diagnosis, number of primary care encounters, and medications. The prevalence of comorbid diabetes and PTSD was 8% (n = 1129). Of these, 57% (n = 649) had comorbid depression. Patients with PTSD and depression had higher rates of substance use disorder and higher cholesterol and LDL. Patients with depression had poorer glycemic control. Patients with PTSD and depression weighed more and had higher BMI than patients with neither diagnosis. Thus, male diabetes patients with PTSD and depression may be vulnerable to substance use disorders and to weight/lipid problems that can affect health. Depression is a likely contributor to poor glycemic control. Careful screening for mental health comorbidities is needed for diabetes patients.

What promotes motor recovery from stroke? To date, studies of recovery from stroke have shown alterations in function in various cortical areas, including the contralesional (unaffected) motor cortex (M1) [1–5]. However, whether these changes contribute to recovery or are mere epiphenomena remains unclear [6, 7]. We therefore sought evidence that the ipsilateral M1 can compensate for dysfunction of the contralateral M1. We recorded the change in force production during a finger-tapping task in response to acute disruption of M1 function by repetitive transcranial magnetic stimulation (rTMS). Neither control (occipital) nor ipsilateral M1 rTMS lead to a change in tapping force. RTMS over contralateral M1 had a short-lived effect and induced changes in ipsilateral M1 excitability around the time that these behavioral effects abated, consistent with delayed compensation by the ipsilateral M1. Simultaneous bilateral M1 stimulation, designed to prevent compensation by the ipsilateral M1, had a large and prolonged effect on tapping force. This is the first demonstration that the ipsilateral primary motor cortex is capable of functionally significant compensation for focal contralateral cortical dysfunction in the adult human and provides a rational basis for interventional treatments aimed at promoting functional compensation in unaffected cortical areas after stroke.
