1. Implementing numerical algorithms to optimize the parameters in Kampmann–Wagner Numerical (KWN) precipitation models

Abstract: Abstract The Kampmann–Wagner Numerical (KWN) model of precipitation is a powerful tool to simulate the precipitation of the second phase considering the nucleation, growth, and coarsening. Some quantities such as interfacial energy and nucleation site number density are required to accomplish the simulation. Practically, those quantities are hard to measure in the experiment directly, and the derivation of those quantities through modeling can also be costly. In this work, we hereby adopt the minimization algorithm implemented in the open-source Scipy Python package to derive that important information in terms of very limited experimental data. The convergence and robustness of different algorithms are discussed. Among those algorithms, the Nelder–Mead and Powell algorithms are successfully applied to optimize multiple parameters during KWN modeling. This work will shed light on the design of experiments/processes and facilitate integrated computational materials engineering (ICME).

3. Towards a Pedagogics of Epiphany

Abstract: This article begins by focusing on critiques of the ideology and practice of neoliberalism in universities, nation-wide and globally. The argument proceeds to analyse alternative or counter-neoliberal strategies of reading, writing, and teaching propounded by selected scholars. My analysis reveals that, although these scholars argue the necessity of developing and putting into practice pedagogical strategies that contest those that are driven by the neoliberal agenda, they do little more than gesture toward implementation of these. By way of redressing this lack, I present a case study that indicates specific tactics for “slipping under the neoliberal radar”. This leads to a discussion of what I consider to be a crucial academic concern – the practice, or the art, of reading, as expressed by George Steiner (1978), John Williams in his novel Stoner (1965), and Éammon Dunne in an analysis of this novel (2016). It is my intention to show how these texts inspired me to concoct what I call a pedagogics of epiphany. I conclude my argument by explaining what I mean by this concept, and the ways in which its implementation might reflect a simultaneously radical and circumspect orientation toward the ongoing project of “doing academia differently”.

Abstract: Abstract In the development of innovative technology products, companies of all sizes are being encouraged to innovate responsibly and regulators are encouraged to adapt their regulatory systems to be smarter, more proportionate and adaptive to the needs of innovative technologies. The British Standards Institution Responsible Innovation (RI) Guide (Publicly Available Specification [PAS] 440) is an industry‐wide standard relevant to both these policy trends. It supports companies by providing a framework to demonstrate the balance between the potential benefits and harms and, if necessary, to take action to maximise the benefits and/or minimise the harms. It includes guidance on engagement with stakeholders and will codify what stakeholders can expect from companies undertaking responsible innovation, paving the way to more harmonious relationships among stakeholders with differing interests and values. A cross‐sectoral survey of innovative companies showed that 90% favoured the development of such a standard. PAS 440 was also trialled in two early‐stage biotechnology companies and its expected benefits included contributing to coordinated responsible behaviour along a supply chain; better company and stakeholder understanding of the product properties; supporting decision‐making on whether or not to start a company; considering the risks of not developing the product and avoiding reputational risks. Benefits were expected to be increasingly significant as the RI standard becomes widely adopted.

5. A novel brain-penetrant oral UGT8 inhibitor decreases in vivo galactosphingolipid biosynthesis in murine Krabbe disease

Abstract: Krabbe disease is a rare, inherited neurodegenerative disease due to impaired lysosomal β-galactosylceramidase (GALC) activity and formation of neurotoxic β-galactosylsphingosine (‘psychosine’). We investigated substrate reduction therapy with a novel brain-penetrant inhibitor of galactosylceramide biosynthesis, RA 5557, in twitcher mice that lack GALC activity and model Krabbe disease. This thienopyridine derivative selectively inhibits uridine diphosphate-galactose glycosyltransferase 8 (UGT8), the final step in the generation of galactosylceramides which are precursors of sulphatide and, in the pathological lysosome, the immediate source of psychosine. Administration of RA 5557, reduced pathologically elevated psychosine concentrations (72–86%) in the midbrain and cerebral cortex in twitcher mice: the inhibitor decreased galactosylceramides by about 70% in midbrain and cerebral cortex in mutant and wild type animals. Exposure to the inhibitor significantly decreased several characteristic inflammatory response markers without causing apparent toxicity to myelin-producing cells in wild type and mutant mice; transcript abundance of oligodendrocyte markers MBP (myelin basic protein) and murine UGT8 was unchanged. Administration of the inhibitor before conception and during several breeding cycles to mice did not impair fertility and gave rise to healthy offspring. Nevertheless, given the unchanged lifespan, it appears that GALC has critical functions in the nervous system beyond the hydrolysis of galactosylceramide and galactosylsphingosine. Our findings support further therapeutic exploration of orally active UGT8 inhibitors in Krabbe disease and related galactosphingolipid disorders. The potent thienopyridine derivative with effective target engagement here studied appears to have an acceptable safety profile in vivo; judicious dose optimization will be needed to ensure efficacious clinical translation.

Abstract: Abstract Background Biobanks provide an important foundation for genomic and personalised medicine. In order to enhance their scientific power and scope, they are increasingly becoming part of national or international networks. Public trust is essential in fostering public engagement, encouraging donation to, and facilitating public funding for biobanks. Globalisation and networking of biobanking may challenge this trust. Methods We report the results of an Australian study examining public attitudes to the networking and globalisation of biobanks. The study used quantitative and qualitative methods in conjunction with bioethical analysis in order to determine factors that may contribute to, and threaten, trust. Results Our results indicate a generally high level of trust in biobanks and in medical research more broadly. Key factors that can reduce perceived trustworthiness of biobanks are commercialisation and involvement in global networking. Conclusions We conclude that robust ethical oversight and governance standards can both promote trust in global biobanking and ensure that this trust is warranted.

Abstract: Stigma and isolation are common in people with tuberculosis (TB). Social isolation contributes to reduced health outcomes and TB treatment adherence. Stigma and the drivers of isolation in people with Drug-Resistant (DR)-TB may include modifiable advice and practices of family and Health Care Workers (HCW). This study aimed to understand the drivers of isolation and stigma from the perspective of people with DR-TB in Vietnam. A greater understanding of stigma and isolation is important to identify and balance patients' needs and disease transmission risk. In-depth interviews were conducted with 12 people with DR-TB and seven HCWs who care for people with DR-TB in two provinces in Vietnam. Interviews were audio-recorded, transcribed verbatim and translated to English. Data collection and analysis were conducted simultaneously. The data were then analysed using a thematic framework approach. Stigma and extended isolation were common experiences among people with DR-TB. To mitigate stigma, people with DR-TB used the local term 'lao lực' to describe their condition to others which is believed to be a less infectious and less stigmatising type of TB. This study identified that although HCW informed people with DR-TB of when they were no longer infectious and isolation was no longer required, their infection control advice was not always consistent. Despite knowing they were no longer infectious, most people with DR-TB continued to self-isolate to minimise the perceived repercussions of societal stigma, to protect their 'thể diện' (honour, prestige, reputation), and eliminate all risk of transmitting DR-TB to their family. This study identified three interconnected drivers of self-isolation in Vietnam, including fear of infecting others, fear of stigmatization, and to protect family reputation. TB control programmes need to better understand the social aspects of DR-TB to enable them to better support patients. Educating HCW to provide evidence-based infection control advice is vital.

In this New Journal of Physics 2020 Plasma Accelerator Roadmap, perspectives from experts in this field provide a summary overview of the field and insights into the research needs and developments for an international audience of scientists, including graduate students and researchers entering the field.

Abstract: This paper states 16 principles for the long term retention and preservation of digital geographic information. The paper is mainly aimed at public sector geographic information providers in Europe (particularly those involved in mapping and cadastre) with the intention of highlighting the significance of fundamental concepts for digital geographic data archiving. Geographic information providers are mainly mapping agencies, but also archives preserving geographic data among a wider range of digital information. A supplementary objective is that the paper may provide useful information for providers of all types of geographic information right around the world. Abstract: Increased metabolism is a requirement for tumor cell proliferation. To understand the dependence of tumor cells on fatty acid metabolism, we evaluated various nodes of the fatty acid synthesis pathway. Using RNAi we have demonstrated that depletion of fatty-acid synthesis pathway enzymes SCD1, FASN, or ACC1 in HCT116 colon cancer cells results in cytotoxicity that is reversible by addition of exogenous fatty acids. This conditional phenotype is most pronounced when SCD1 is depleted. We used this fatty-acid rescue strategy to characterize several small-molecule inhibitors of fatty acid synthesis, including identification of TOFA as a potent SCD1 inhibitor, representing a previously undescribed activity for this compound. Reference FASN and ACC inhibitors show cytotoxicity that is less pronounced than that of TOFA, and fatty-acid rescue profiles consistent with their proposed enzyme targets. Two reference SCD1 inhibitors show low-nanomolar cytotoxicity that is offset by at least two orders of magnitude by exogenous oleate. One of these inhibitors slows growth of HCT116 xenograft tumors. Our data outline an effective strategy for interrogation of on-mechanism potency and pathway-node-specificity of fatty acid synthesis inhibitors, establish an unambiguous link between fatty acid synthesis and cancer cell survival, and point toward SCD1 as a key target in this pathway.

12. Constraining the Physical Properties of Stellar Coronal Mass Ejections with Coronal Dimming: Application to Far-ultraviolet Data of є Eridani

Abstract: Coronal mass ejections (CMEs) are a prominent contributor to solar system space weather and might have impacted the Sun's early angular momentum evolution. A signal diagnostic of CMEs on the Sun is coronal dimming: a drop in coronal emission, tied to the mass of the CME, that is the direct result of removing emitting plasma from the corona. We present the results of a coronal dimming analysis of Fe XII 1349 Å and Fe XXI 1354 Å emission from ϵ Eridani ( ϵ Eri), a young K2 dwarf, with archival far-ultraviolet observations by the Hubble Space Telescope's Cosmic Origins Spectrograph. Following a flare in 2015 February, ϵ Eri's Fe XXI emission declined by 81 ± 5%. Although enticing, a scant 3.8 minutes of preflare observations allows for the possibility that the Fe XXI decline was the decay of an earlier, unseen flare. Dimming nondetections following each of three prominent flares constrain the possible mass of ejected Fe XII-emitting (1 MK) plasma to less than a few × 1015 g. This implies that CMEs ejecting this much or more 1 MK plasma occur less than a few times per day on ϵ Eri. On the Sun, 1015 g CMEs occur once every few days. For Eri, the mass-loss rate due to CME-ejected 1 MK plasma could be < 0.6 ṀꙨ well below the star's estimated 30 ṀꙨ mass-loss rate (wind + CMEs). The order-of-magnitude formalism we developed for these mass estimates can be broadly applied to coronal dimming observations of any star.

Abstract: A mean-field model has been developed to simulate curvature-driven grain growth by exploring the evolution of grain size distribution under arbitrary thermal histories. The model was integrated into precipitation module TC-PRISMA, so that the pinning effect of the concurrently precipitated particles on the growing grains can be considered by a modified, location-specific Zener model. The developed model was validated against analytical calculations, and then applied to real alloy systems, fed with assessed grain boundary energy and mobility data. Its capabilities, limitations, and directions to improvements have been discussed.

Abstract: Although there is an extensive literature on the efficacy of the low carbohydrate diet (LCD) for weight loss and in the management of type 2 diabetes, concerns have been raised that the LCD may increase cardiovascular disease (CVD) risk by increasing the level of low-density lipoprotein cholesterol (LDL-C). We have assessed the value of LDL-C as a CVD risk factor, as well as effects of the LCD on other CVD risk factors. We have also reviewed findings that provide guidance as to whether statin therapy would be beneficial for individuals with high LDL-C on an LCD.Multiple longitudinal trials have demonstrated the safety and effectiveness of the LCD, while also providing evidence of improvements in the most reliable CVD risk factors. Recent findings have also confirmed how ineffective LDL-C is in predicting CVD risk.Extensive research has demonstrated the efficacy of the LCD to improve the most robust CVD risk factors, such as hyperglycemia, hypertension, and atherogenic dyslipidemia. Our review of the literature indicates that statin therapy for both primary and secondary prevention of CVD is not warranted for individuals on an LCD with elevated LDL-C who have achieved a low triglyceride/HDL ratio.

23. Causes of Late Cretaceous subduction termination below South China and Borneo: Was the Proto-South China Sea underlain by an oceanic plateau?

Abstract: The South China, Indochina, and Borneo margins surrounding the South China Sea contain long-lived arcs that became inactive at approximately 85 Ma, even though an embayment of oceanic crust (the ‘Proto-South China Sea’) remained in the intervening region. This oceanic crust eventually subducted in the Cenozoic below Borneo and the Cagayan arc, while the modern South China Sea opened in its wake. To investigate the enigmatic cessation of Mesozoic subduction below South China and Borneo, we studied a fragment of oceanic crust and overlyinƒg trench-fill sediments that accreted to NW Borneo during the final stages of subduction. Based on radiolarian biostratigraphy of cherts overlying the pillow basalts and detrital zircon geochronology of the trench-fill, we constrained the minimum age of the oceanic crust during accretion to 40 Ma. This shows that subduction cessation was not related to ridge subduction. Geochemical analysis of pillow basalts revealed an enriched mid-ocean ridge basalt signature comparable to oceanic plateaus. Using paleomagnetism, we show that this fragment of oceanic crust was not part of the Izanagi Plate but was part of a plate (the ‘Pontus’ Plate) separated from the Izanagi Plate by a subduction zone. Based on the minimum 40 Ma age of the oceanic crust and its geochemistry, we suggest that Mesozoic subduction below South China and Borneo stopped when an oceanic plateau entered the trench, while the eastern plate margin with the Izanagi Plate remained active. We show how our findings offer opportunities to restore plate configurations of the Panthalassa-Tethys junction region.

24. High Energy, High Pulse Rate Laser Operation Using Crystalline Adhesive-Free Bonded Yb:YAG Slabs

Abstract: We report on the successful amplification of 10 ns pulses to 10 J energy at 10 Hz in a DiPOLE laser amplifier using crystalline Yb:YAG/Cr:YAG composite slabs manufactured using adhesive-free bonding (AFB) technology. We demonstrate that bonded slabs are suitable for operation in high energy cryogenic laser amplifiers. We also report on frequency doubling of the beam amplified in the bonded slabs. When the pulse energy of the output infrared beam is set to 5 J, a pulse energy of 3.9 J is achieved in the green (corresponding to 78% conversion efficiency). Results demonstrate that AFB technology is suitable for producing large-sized gain material slabs and can overcome current limitations in the manufacture of large-aperture gain material pieces. We believe this work will facilitate energy scaling of high energy lasers where aperture scaling of optical elements is not achievable via conventional manufacturing techniques.

25. The origin of early Archean barite deposits on the Kaapvaal and Pilbara cratons

Abstract: Sulfate minerals are rare in the geological record prior to the oxygenation of the Earth’s atmosphere circa 2.4 billion years ago (Ga). An exception to this are a few isolated occurrences of early Archean (3.6-3.2 Ga) barite (BaSO4), hosted in volcano-sedimentary rocks in South Africa, India and Western Australia. The origin of these barite deposits is controversial, despite having been studied over decades. Here, we combine field observations and geochemical data from a multi-year investigation into barite occurrences on the Kaapvaal and Pilbara cratons to derive a holistic model for the formation of early Archean barite. Studied deposits include the 3.52 Ga Londozi deposit in Eswatini and the 3.49 Ga North Pole deposit in Western Australia that are hosted in volcanic rocks, and the 3.26-3.23 Ga Barite Valley deposit in South Africa and possibly time-equivalent but little-known Cooke Bluff deposit in Western Australia that are found in sedimentary successions. Our field observations indicate that barite is closely associated with chert on both the Kaapvaal and the Pilbara cratons, although the scale of barite mineralization is much larger in the Pilbara and cross-cutting barite veins are only observed at North Pole and Cooke Bluff. These findings suggest that the fluids from which the chert precipitated are the same as the fluids from which the barite formed, and geochemical data support an origin for these barium-rich fluids that is related to low-temperature hydrothermal circulation of seawater. Barite precipitation could have been triggered by silica removal from these fluids. The ubiquity of chert in the early rock record suggests that these settings may have been common in the early Archean and that barite formation was therefore limited by sulfate abundance, and could only occur in settings where hydrothermal circulation and local sulfate enrichment occurred together.

Abstract: Lunar IceCube is a NASA nanosatellite mission designed to prospect, locate, and measure volumes and composition of water ice deposits and other volatiles on the Moon in support of future exploitation by robots and human explorers. Lunar IceCube is one of several public -private partnerships selected under NASA's Next Space Technologies for Exploration Partnerships Broad Agency Announcement for the development of advanced exploration systems. Among the first small satellites planned to explore deep space, Lunar IceCube, along with the 12 other Exploration Mission One (EM -1) missions will help lay a foundation for future small-scale planetary missions. In addition to providing useful scientific data, Lunar IceCube will help inform NASA's strategy for sending humans farther into the solar system.
