In this paper, I consider how Chinese gamers modify both ideologies and technologies on the gaming platform Steam. Working from Mayfair Yang’s notion of “socialist space” and Lefebvre’s “representational spaces,” I argue that these gamers seek to transform their representational spaces into reality by “modding” the content and code of computer games and archiving roguish discourses on the tactical safe space of the Steam Workshop. Because of these modders’ paratextual activity, a China-specific version of the Steam client will be released in the near future, thus censoring Steam as is in mainland China. To better understand this “China Steam” swap and the relationship between modding and censorship, I employ a diachronic approach, examining historical precedents of other “propaganda archives” in both premodern and modern eras. Moreover, I engage with fan fiction studies to further conceptualize modding and censorship, fusing de Certeau’s “strategies and tactics” with Abigail De Kosnik’s “rogue archives” and thus putting forth the framework of a tactical safe space. After laying out this approach, I share the process and findings of my “guerilla netnography” of Chinese modders on the Left for Dead 2 (L4D2) Workshop, detailing three categories of mods and their associated paratexts. Finally, I turn to Steven J. Jackson and Cuban thinkers and scholars to show how modding is a part of a larger sociopolitical narrative of repair and inventiveness in post-socialist contexts such as the PRC, in which modders turn the socialist ideology of the state upon itself.

Fusion genes are structural variants that arise when the coding region of two previously independent genes become joined together; fusion genes can be a major driving cause of cancer development. Many gene fusion events have been classified as clinically-relevant and are targets for both diagnostic applications, such as TMPRSS2-ERG in prostate cancer, and therapeutic applications, such as CD74-ROS1 and EML4-ALK in non-small cell lung cancer. Given the potential clinical benefits of early detection, there is significant interest in developing highly sensitive and accurate diagnostic assays to detect cancer using these gene fusions as biomarkers. A significant barrier to the development of these diagnostic assays is the lack of a reliable and renewable sample source harboring gene fusions of interest for use in assay development and analytical validation. Here, we describe the development and performance of a new fusion reference material: the Twist Pan-cancer RNA Fusion Control, a highly-multiplexed positive control designed to be spiked into a selected RNA background or as a stand-alone positive control. The Twist Pan-cancer RNA Fusion Control provides a wide selection of 80+ common and curated cancer targets for analytical validation and assay development. The synthetic RNAs are designed centered around the known and documented fusion sites with 750 nt of RNA 5′- of and 3′- of the fusion junction, suitable for short read sequencing and qPCR experiments. Pooled synthetic fusion RNAs are quantified at known molar ratios, NGS QC’d for uniformity, and ddPCR’d to establish a highly precise pool concentration. Finally, in fusion detection NGS experiments, we observe over a 95% recall rate of fusion events. The Pan-cancer RNA Fusion Control is designed to serve as a spike-in or stand-alone positive control and specifically pairs well with a clinically-relevant target enrichment (TE) panel within the Twist Targeted Enrichment for Gene Expression solution. This control sample positive for RNA fusions can be applied in both qPCR and NGS workflows to validate panel/probe sets, establish limits of detection, and monitor ongoing assay performance. Introduction: The objective of this paper is to demonstrate techniques on how to improve hip flexibility and mobility, especially for track and field athletes. It also seeks to explain the underlying importance of hip flexibility and mobility. When athlete’s hips are stiff and inflexible, muscle imbalance may occur and increase the risk of injury. Muscle imbalance in the hip often causes knee and hip pain, due to the hip flexors and quadriceps overworking in order to compensate for weaker muscles. The hip mobility and flexibility exercises illustrated here are designed to significantly improve the range of motion (ROM) of the hips. Enhanced ROM can improve motor performance, skill execution, and even prevent injury. Excellent hip extension improves the ability to assist the hip in developing power. Methods: 9 Hip Mobility and 6 Hip Flexibility exercises are described in this paper. The exercises are to be performed 3× per week with at least 24 hours break in-between to achieve optimum results. Conclusion: This paper sought to outline the importance of hip mobility and flexibility, in particular for non-contact-sport athletes (such as those in track and field), and their effects on athletic performance and injury prevention. After proximately 6 weeks, the ROM of the hip will be expected to improve significantly.

Es muss grundsatzlich zwischen Unfallen wahrend der Ausubung des Pferdesports und dem blosen Umgang mit dem Pferd differenziert werden, da hier unterschiedliche Verletzungsmuster zu beachten sind. Im Rahmen der Unfallpravention kommen der Aufklarung uber potenzielle Unfallrisiken sowie der fundierten Ausbildung von Reiter und Pferd besondere Bedeutung zu.

Acipenser iridovirus-European (AcIV-E) is an important pathogen of sturgeons. Two variants differing by single-nucleotide polymorphisms (SNP) in the Major Capsid Protein gene have been described, but without any indication as to their prevalence in farms. To facilitate epidemiological studies, we developed a high-resolution melting (HRM) assay to distinguish between two alleles (var1 and var2) differing by five point substitutions. The HRM assay detected as little as 100 copies of plasmids harboring cloned sequences of var1 and var2, which have melting temperatures (Tm) differing by only 1 °C. The assay was specific of AcIV-E as demonstrated by the absence of signal when testing a related, yet distinct, virus as well as DNA from an AcIV-E-negative sturgeon sample. Experiments with mixtures of two distinct plasmids revealed abnormal melting curve patterns, which showed dips just before the main melting peaks. These dips in the curves were interpreted as the dissociation of heteroduplexes fortuitously created during the PCR step. Screening AciV-E-positive field samples of Russian sturgeons from three farms revealed the presence of var2, based on the Tm. However, for a few samples, the melting curves showed patterns typical of var2 as the dominant viral genome, mixed with another minor variant which proved to be var1. In conclusion, HRM is a simple method to screen for AcIV-E var1 and var2 and can be used on a large scale in Europe to trace these two variants which likely represent two genetic lineages.
