1. Multiple invasions and predation: The impact of the crayfish Cherax quadricarinatus on invasive and native snails

Abstract

Abstract The pace of biological invasions has increased in recent decades, leading to multiple invasions and the potential dominance of invasive species, destabilizing local ecological networks. This provides opportunities to study new ecological species interactions, including predation. Tropical freshwaters have been particularly concerned by aquatic invasions and we focused here on the Martinique island (Lesser Antilles). We examined the predator–prey relationships involving invasive Thiarid snails (Tarebia granifera and Melanoides tuberculata) and the native Neritina punctulata, both confronted with a newcomer predator, the redclaw crayfish (Cherax quadricarinatus). We conducted several mesocosm experiments to assess the impact of crayfish predation on snail survival and the passive and active antipredator responses of snails. A first experiment indicated snail survival rates between 50% and 80%, depending on crayfish size and sex. Notably, there was a negative correlation between snail survival and male crayfish size and the predation method (shell crushing vs. “body sucking”) varied with crayfish size. The second experiment suggested no refuge size for snails, with both very small (5 mm) unable to escape predation, regardless of crayfish size (from 77 to 138 mm) or sex. Finally, we investigated the escape behavior of Thiarids regarding three crayfish cues. Melanoides tuberculata tend to bury in the substrate and T. granifera to climb up aquarium walls, what was expected from their shell morphologies, and both responding to crayfish cues within minutes. Overall, C. quadricarinatus proves to be an efficient snail predator with limited escape options for snails, potentially contributing to the decline of certain snail populations in Martinique. This omnivorous predator might impact other native species across different groups, including shrimps and fish. Our study underscores the urgent need for monitoring efforts, solidifying the redclaw crayfish reputation as a dangerous invasive species for freshwater macrobenthic faunas worldwide.

Abstract

Abstract Background 5-Methylcytosine (5mC) is an important epigenetic mark in eukaryotes. Little information about its role exists for invertebrates. To investigate the contribution of 5mC to phenotypic variation in invertebrates, alteration of methylation patterns needs to be produced. Here, we apply new non-nucleoside DNA methyltransferase inhibitors (DNMTi) to introduce aleatory changes into the methylome of mollusk species. Results Flavanone inhibitor Flv1 was efficient in reducing 5mC in the freshwater snails Biomphalaria glabrata and Physa acuta, and to a lesser degree, probably due to lower stability in sea water, in the oyster Crassostrea gigas. Flv1 has no toxic effects and significantly decreased the 5mC level in the treated B. glabrata and in its offspring. Drug treatment triggers significant variation in the shell height in both generations. A reduced representation bisulfite-sequencing method called epiGBS corroborates hypomethylation effect of Flv1 in both B. glabrata generations and identifies seven Differential Methylated Regions (DMR) out of 32 found both in Flv1-exposed snails and its progeny, from which 5 were hypomethylated, demonstrating a multigenerational effect. By targeted bisulfite sequencing, we confirmed hypomethylation in a locus and show that it is associated with reduced gene expression. Conclusions Flv1 is a new and efficient DNMTi that can be used to induce transient and heritable modifications of the epigenetic landscape and phenotypic traits in mollusks, a phylum of the invertebrates in which epigenetics is understudied.

Abstract

We describe here a protocol for the generation of sequence-ready libraries for population epigenomics studies, and the analysis of alignment results. We show that the protocol can be used to monitor chromatin structure changes in populations when exposed to environmental cues. The protocol is a streamlined version of the Assay for transposase accessible chromatin with high-throughput sequencing (ATAC-seq) that provides a positive display of accessible, presumably euchromatic regions. The protocol is straightforward and can be used with small individuals such as daphnia and schistosome worms, and probably many other biological samples of comparable size (~10,000 cells), and it requires little molecular biology handling expertise.

Classical biomonitoring techniques have focused primarily on measures linked to various biodiversity metrics and indicator species. Next-generation biomonitoring (NGB) describes a suite of tools and approaches that allow the examination of a broader spectrum of organizational levels—from genes to entire ecosystems. Here, we frame 10 key questions that we envisage will drive the field of NGB over the next decade. While not exhaustive, this list covers most of the key challenges facing NGB, and provides the basis of the next steps for research and implementation in this field. These questions have been grouped into current- and outlook-related categories, corresponding to the organization of this paper.

9. Color polymorphism and conspicuousness do not increase speciation rates in Lacertids

Abstract

Conspicuous body colors and color polymorphism have been hypothesized to increase rates of speciation. Conspicuous colors are evolutionary labile, and often involved in intraspecific sexual signaling and thus may provide a raw material from which reproductive isolation can easily evolve, while polymorphism could favor rapid evolution of new lineages through morphic speciation. Here, we investigated the influence of the presence/absence of conspicuous colorations, and of color polymorphism on the speciation of Lacertids. We used several state-dependent diversification models, and showed that, regardless of the methods, conspicuous colorations and color polymorphism were not related to species speciation. While the lack of correlation between conspicuous colorations and speciation rates is in line with most of the literature testing this hypothesis, the results for color polymorphism contradict previous studies, and question the generality of the morphic speciation hypothesis.

Abstract

The complexity of food webs and how it depends on environmental variables is a long-standing question in ecology. Food-chain length depends on many factors, such as productive space, disturbance and spatial processes. It is not clear though how food-chain length should vary with adaptive evolutionary changes of the constitutive species. Assuming that each trophic level is subject to a competition-colonization trade-off, we model the adaptive evolution of their colonization rates and its consequences on equilibrium occupancies and food-chain length. When colonization rates are allowed to evolve, longer food chains can persist. Extinction, perturbation and habitat loss all affect the evolutionary-stable colonization rates, but trade-off strength (costs of dispersal) has a major role: weaker trade-offs yield longer chains. Our results suggest a strong link between within-trophic level competition, spatial occupancies and food-chain length. Although these eco-evo dynamics partly alleviate the constraint on food-chain length in metacommunities, we show it is no magic bullet: the highest, most vulnerable, trophic levels are also those that least benefit from evolution. Our model generates qualitative predictions regarding how trait evolution affects the response of communities to disturbance and habitat loss. This highlights the importance of eco-evolutionary dynamics at metacommunity level in determining food-chain length.

Abstract

International audience; Metacommunity structure reflects the interplay of various processes, including niche filtering, extinction/colonization and interspecific interactions. Spatial patterns of species distributions are often analyzed to infer these processes. However, such inferences rely on often unrealistic equilibrium assumptions, and remain ambiguous, as different processes can produce similar patterns. Temporal data may improve these inferences. For example, stochastic species turnover may occur in local communities, while, on the long run, temporal changes are kept within limits set by locally available niches. Our objective is to explore how the joint analysis of spatial and temporal patterns can clarify the contribution of different processes to metacommunity structure. We recorded the occurrences of 21 freshwater mollusc species, and environmental data, in 250 sites over 17 successive years in a network of ponds in Guadeloupe (Lesser Antilles). We analyzed variation in α and β-diversities in space and time, and used a joint-species distribution mode to characterize species–environment and species–species relationships. Local communities showed pronounced temporal variation reflecting both imperfect species detection and true stochastic species turnover. On the long term however, local communities were largely controlled by niche filtering along two main environmental gradients, one driven by site connectivity, the other by hydrological stability and aquatic vegetation. Two gastropod clades, caenogastropods and pulmonates, showed contrasted spatio–temporal distributions resulting from different responses to these gradients, and these distributions seemed little altered by interspecific competition. Our study illustrates the benefit of using spatiotemporal metacommunity data to discern long-term impacts of niche filtering and species interactions behind short-term stochasticity.

Abstract

When environmental conditions are unpredictable, expressing alternative phenotypes spreads the risk of failure, a mixed strategy called bet-hedging. In the southern part of its range, the Parsley Frog Pelodytes punctatus breeds both in autumn and in spring. Our aim was to study the breeding phenology and reproductive success associated with the use of those two seasonal niches to understand how this breeding strategy can be maintained. Field surveys revealed that breeding phenology was typically bimodal with a higher breeding effort in autumn. More importantly, in spring, the survival rate of offspring was severely reduced by the presence of autumn tadpoles, indicating a clear priority effect. However, the autumn cohort often failed to survive over winter, in which case spring cohorts were often successful. Based on those results, we constructed a model in which females can allocate a variable portion of eggs to each season and added a priority effect. We conclude that the existence of the two breeding seasons may indeed constitute a bet-hedging strategy.

Abstract

We present an unusual case of an intra-abdominal collection which evidenced a rare etiology and raises diagnostic particularities. Background. Fish bones ingestion is frequent, but seldom followed by complications. Those are often reported at specific sites. Objectives. This case report emphasizes the unusual presentation and site localization of a colonic perforation by a small fish bone, in the context of limited radiological accuracy at the diagnostic phase. Case presentation. A 37 year old male was admitted to the gastroenterology ward with upper and left sided abdominal pain associated with fever and marked fatigue. His medical history was marked by a sleeve gastrectomy in 2010 for obesity. Abdominal signs and elevated acute inflammatory syndrome on blood tests were followed by computer tomography which revealed a pericolic mass near the left splenic flexure. The pain and fever increased in intensity, so a laparotomy was proposed. Intraoperatively, a tumor-like lesion was found and a resection with oncologic limits was performed. Microscopic examination of the specimen revealed a fish bone, but only after surgery did the patient confirm that he had eaten fish meal the week before. The post-operative period was uneventful. Conclusion. Fish bones remain some of the most frequently ingested alimentary foreign bodies; they may cause atypical clinical presentations, frequently omitted by the patients themselves if symptoms appear delayed. They could also lead to possible high-risk complications which need to be addressed by surgeons.

Abstract

Although metacommunity ecology has been a major field of research in the last decades, with both conceptual and empirical outputs, the analysis of the temporal dynamics of metacommunities has only emerged recently and still consists mostly of repeated static analyses. Here we propose a novel analysis framework to assess metacommunity processes using path analyses of spatial and temporal diversity turnovers. We detail the principles and practical aspects of this framework and apply it to simulated datasets to illustrate its ability to decipher the respective contributions of entangled drivers of metacommunity dynamics. We then apply it to four real datasets. Empirical results support the view that metacommunity dynamics may be generally shaped by multiple ecological processes acting in concert, with environmental filtering being variable across both space and time. These results reinforce our call to go beyond static analyses of metacommunities that are blind to the temporal part of environmental variability. Abstract

International audience; Cytoplasmic male-sterility (CMS) is a form of genetic conflict over sex determination that results from differences in modes of inheritance between genomic compartments 1-3. Indeed, maternally-transmitted (usually mitochondrial) genes, sometimes enhance their transmission by suppressing the male function in a hermaphroditic organism, to the detriment of biparentallyinherited nuclear genes. As a result, these hermaphrodites become functionally female, and may coexist with regular hermaphrodites in so-called gynodioecious populations 3. CMS has been known in plants since Darwin's times 4 , but is previously unknown in the animal kingdom 5-8. We relate the first observation of CMS in animals. It occurs in a freshwater snail population, where some individuals appear unable to sire offspring in controlled crosses and show anatomical, physiological and behavioral characters consistent with a suppression of the male function. Male-sterility is associated with a mitochondrial lineage that underwent a spectacular acceleration of DNA substitution rates, affecting the entire mitochondrial genome-this acceleration concerns both synonymous and nonsynonymous substitutions and therefore results from increased mitogenome mutation rates. Consequently, mitochondrial haplotype divergence within the population is exceptionally high, matching that observed between snail taxa that diverged 475 million years ago. This result is reminiscent of similar accelerations in mitogenome evolution observed in plant clades where gynodioecy is frequent 9,10 , both being consistent with arms-race evolution of genome regions implicated in CMS 11,12. Our study shows that genomic conflicts can trigger independent evolution of similar sex determination systems in plants and animals, and dramatically accelerate molecular evolution.

WOS:000779278700001; International audience; Characterizing the diversity of demographic strategies among species can inform research in topics such as trait syndromes, community stability, coexistence, and ecological succession. However, this diversity can depend on the spatial scale considered: at the landscape scale, species often form metapopulations, that is sets of local, sometimes short-lived, populations, inhabiting discrete habitat patches. Metapopulation dynamics are most frequently analyzed in individual species or pairs of interacting species because of the large amount of data required for multiple species, and because species vary in their perceptions of what constitutes a favorable or unfavorable habitat. Here we evaluate, using a case study, whether a metapopulation model can be used to generate accurate estimates of demographic parameters and to describe the diversity of dynamics, responses to environment, and prospects of long-term persistence in a guild of species inhabiting a common fragmented landscape. We applied this approach to a guild of 22 mollusk species that inhabit freshwater habitats on two islands of Guadeloupe, to compare metapopulation dynamics among species. We analyzed a 15-year time series of occupancy records for 278 sites using a multistate occupancy model that estimated colonization and extinction rates as a function of site-specific and year-specific environmental covariates, then used model results to simulate future island metapopulation dynamics. Despite the diverse array of metapopulation trajectories—a mix of species with either stable, increasing, declining, or fluctuating metapopulations—and the inherent challenges associated with such data (e.g., imperfect detection, spatial and temporal heterogeneity), our model accurately captured among-patch variation in suitability for many mollusk taxa. The dynamics of rare species or species with habitat preferences not fully captured by the retained set of covariates were less well described. For several species, we detected a negative correlation between extinction and colonization. This variation in habitat suitability created species-specific extinction-resistant pockets in the landscape. Our comparative analysis also revealed that species had distinct strategies for metapopulation dynamics, such as “fast-turnover” species with both a high proportion of occupied sites and a high rate of site extinction in the landscape.

Abstract

Plant sexual dimorphism is thought to evolve in response to sex-specific selection associated with competition for access to mates or resources, both of which may be density dependent. In wind-pollinated plants in particular, vegetative traits such as plant size and architecture may influence resource acquisition and both pollen dispersal and receipt, with potential conflict between these two components of fitness. We evaluated the role of plant density in shaping plant traits by measuring evolutionary responses in experimental populations of the sexually dimorphic wind-pollinated plant Mercurialis annua. After three generations of evolution, we observed divergence between high- and low-density populations in several vegetative traits, whereas there was no divergence for reproductive traits. A reversal in the direction of sexually dimorphic traits expressed in young plants evolved in both low- and high-density populations compared to the original population (stored as seeds). Compared to the source population, males at high density evolved to be taller when young, whereas at low density young females tended to become smaller. These results demonstrate that a simple change in plant density can induce age-dependent and sex-specific evolution in the ontogeny of vegetative organs, and illustrates the power of experimental evolution for investigating plant trait evolution.

Abstract

Coexistence with related species poses evolutionary challenges to which populations may react in diverse ways. When exposed to similar environments, sympatric populations of two species may adopt similar phenotypic trait values. However, selection may also favour trait divergence as a way to reduce competition for resources or mates. The characteristics of external body parts, such as coloration and external morphology, are involved to varying degrees in intraspecific signalling as well as in the adaptation to the environment and consequently may be diversely affected by interspecific interactions in sympatry. Here, we studied the effect of sympatry on various colour and morphological traits in males and females of two related newt species Lissotriton helveticus and L. vulgaris. Importantly, we did not only estimate how raw trait differences between species respond to sympatry, but also the marginal responses after controlling for environmental variation. We found that dorsal and caudal coloration converged in sympatry, likely reflecting their role in adaptation to local environments, especially concealment from predators. In contrast, aspects of male and female ventral coloration, which harbours sexual signals in both species, diverged in sympatry. This divergence may reduce opportunities for interspecific sexual interactions and the associated loss of energy, suggesting reproductive character displacement (RCD). Our study emphasizes the contrasting patterns of traits involved in different functions and calls for the need to consider this diversity in evolutionary studies.

Abstract

Background 5-Methylcytosine (5mC) is an important epigenetic mark in eukaryotes. Little information about its role exists for invertebrates. To investigate the contribution of 5mC to phenotypic variation in invertebrates, alteration of methylation patterns needs to be produced. Here, we apply new non-nucleoside DNA methyltransferase inhibitors (DNMTi) to introduce aleatory changes into the methylome of mollusk species. Results Flavanone inhibitor Flv1 was efficient in reducing 5mC in the freshwater snails Biomphalaria glabrata and Physa acuta, and to a lesser degree, probably due to lower stability in sea water, in the oyster Crassostrea gigas. Flv1 has no toxic effects and significantly decreased the 5mC level in the treated B. glabrata and in its offspring. Drug treatment triggers significant variation in the shell height in both generations. A reduced representation bisulfite-sequencing method called epiGBS corroborates hypomethylation effect of Flv1 in both B. glabrata generations and identifies seven Differential Methylated Regions (DMR) out of 32 found both in Flv1-exposed snails and its progeny, from which 5 were hypomethylated, demonstrating a multigenerational effect. By targeted bisulfite sequencing, we confirmed hypomethylation in a locus and show that it is associated with reduced gene expression. Conclusions Flv1 is a new and efficient DNMTi that can be used to induce transient and heritable modifications of the epigenetic landscape and phenotypic traits in mollusks, a phylum of the invertebrates in which epigenetics is understudied.

Abstract

Understanding how environmental change affects genetic variances and covariances of reproductive traits is key to formulate firm predictions on evolutionary responses. This is particularly true for sex-specific variance in reproductive success, which has been argued to affect how populations can adapt to environmental change. Our current knowledge on the impact of environmental stress on sex-specific genetic architecture of fitness components is still limited and restricted to separate-sexed organisms. However, hermaphroditism is widespread across animals and may entail interesting peculiarities with respect to genetic constraints imposed on the evolution of male and female reproduction. We explored how food restriction affects the genetic variance-covariance (G) matrix of body size and reproductive success of the simultaneously hermaphroditic freshwater snail Physa acuta. Our results provide strong evidence that the imposed environmental stress elevated the opportunity for selection in both sex functions. However, the G matrix remained largely stable across the tested food treatments. Importantly, our results provide no support for cross-sex genetic correlations suggesting no strong evolutionary coupling of male and female reproductive traits. We discuss potential implications for the adaptation to changing environments and highlight the need for more quantitative genetic studies on male and female fitness components in simultaneous hermaphrodites. Abstract

Plant sexual dimorphism is thought to evolve in response to sex-specific selection associated with competition for access to mates or resources, both of which will often be density-dependent. In wind-pollinated plants in particular, vegetative traits can have an important influence on both resource acquisition and the pollen dispersal and receipt, with potential conflict between these two components of fitness. We evaluated the role of plant density in shaping plant traits by measuring evolutionary responses in experimental populations of the sexually dimorphic wind-pollinated plant Mercurialis annua. After three generations of evolution, we observed divergence between high- and low-density populations in several vegetative traits, whereas there was no divergence for reproductive traits. A reversal in the direction of sexually dimorphic traits expressed in young plants evolved in both low- and high-density populations compared to the original population (stored as seeds). Compared to the source population, males at high density evolved to be taller when young, whereas at low density young females tended to become smaller. These results demonstrate that a simple change in plant density can induce rapid, age-dependent and sex-specific evolution in the ontogeny of vegetative organs, and illustrates the power of experimental evolution for investigating plant trait evolution.

version 2; peer review: 2 approved; International audience; We describe here a protocol for the generation of sequence-ready libraries for population epigenomics studies, and the analysis of alignment results. We show that the protocol can be used to monitor chromatin structure changes in populations when exposed to environmental cues. The protocol is a streamlined version of the Assay for transposase accessible chromatin with high-throughput sequencing (ATAC-seq) that provides a positive display of accessible, presumably euchromatic regions. The protocol is straightforward and can be used with small individuals such as daphnia and schistosome worms, and probably many other biological samples of comparable size (~10,000 cells), and it requires little molecular biology handling expertise.

Abstract

International audience; Biomonitoring ecosystems is necessary in order to evaluate risks and to efficiently manage ecosystems and their associated services. Agrosystems are the target of multiple stressors that can affect many species through effects cascading along food webs. However, classic biomonitoring, focused on species diversity or indicator species, might be a poor predictor of the risk of such whole-ecosystem perturbations. Thanks to high-throughput sequencing methods, however, it might be possible to obtain sufficient information about entire ecological communities to infer the functioning of their associated interaction networks, and thus monitor more closely the risk of the collapse of entire food webs due to external stressors. In the course of the ‘next-generation biomonitoring’ project, we collectively sought to experiment with this idea of inferring ecological networks on the basis of metabarcoding information gathered on different systems. We here give an overview of issues and preliminary results associated with this endeavour and highlight the main difficulties that such next-generation biomonitoring is still facing. Going from sampling protocols up to methods for comparing inferred networks, through biomolecular, bioinformatic, and network inference, we review all steps of the process, with a view towards generality and transferability towards other systems. © 2022 Elsevier Ltd

Abstract

Cryptic species can present a significant challenge to the application of systematic and biogeographic principles, especially if they are invasive or transmit parasites or pathogens. Detecting cryptic species requires a pluralistic approach in which molecular markers facilitate the detection of coherent taxonomic units that can then be analyzed using various traits (e.g., internal morphology) and crosses. In asexual or self-fertilizing species, the latter criteria are of limited use. We studied a group of cryptic freshwater snails (genusGalba)from the family Lymnaeidae that have invaded almost all continents, reproducing mainly by self-fertilization and transmitting liver flukes to humans and livestock. We aim to clarify the systematics, distribution and phylogenetic relationships of these species with an integrative approach that includes morphology (shell and reproductive anatomy), molecular markers, wide-scale sampling across America, and data retrieved from GenBank (to include Old World samples). Our phylogenetic analysis suggests that the genusGalbaoriginated ca. 22 Myr ago and today comprises six clusters of species. Four of these clusters (G.truncatula,G.cubensis/viator,G.humilisandG.schirazensis) are morphologically cryptic and constitute species or species complexes with wide variation in their genetic diversity, geographic distribution and invasiveness. The other two clusters constitute a single species or a species complex (Galba cousini/meridensis) that demonstrate more geographically restricted distributions and exhibit an alternative morphology more phylogenetically derived than the cryptic one. Further genetic studies are required to clarify the status of bothG. cousini/meridensisandG.cubensis/viator. We emphasize that noGalbaspecies should be identified without molecular markers and that additional sampling is required, especially in North America, Eurasia and Africa to clarify remaining questions in systematics and biogeography. We also discuss several hypotheses that can explain crypsis inGalba, such as convergence and morphological stasis, and hypothesize a role for stabilizing selection in amphibious (rather than strictly freshwater) habitats.

Abstract

The distribution of neutral genetic variation in subdivided populations is driven by the interplay between genetic drift, migration, local extinction and colonization. The influence of environmental and demographic factors has also been increasingly examined in empirical studies, but generally focusing on a single species. An open question is whether these factors will similarly, or idiosyncratically, affect a guild of species occupying the same, though exhibiting different traits, mating systems and histories. We addressed it by comparing the population genetic structure of the four most common species of hermaphroditic freshwater snails in Guadeloupe (Lesser Antilles), which occupy a network of patchily distributed sites experiencing temporal variation in water availability. We analyzed microsatellite variability in 21 to 43 populations per species, and built predictions on how several environmental and demographic variables, quantified from a long-term annual survey, as well as species traits, may affect the distribution of genetic variation. These species displayed similarities, such as fairly high levels of variation, but with marked differences among sites, as well as strong genetic differentiation and limited isolation by distance, which can be explained by passive dispersal (strong role of site connectivity), extinction/colonization dynamics and variation in local population size. They also exhibit differences, largely due to the mating system with less genetic diversity and more genetic differentiation in the two selfing species when compared to the two outcrossing ones. These differences can also be attributed to interspecific interactions resulting from the ongoing invasion of Guadeloupe by one of the species studied, which affects the demography of other species, and, to a limited extent, to local environmental factors. Our comparative approach shows both differences and uniqueness in the way species occupy the same landscape, and provides a possible entry to interspecific interactions in community assembly.

Abstract

Niche partitioning is the most studied factor structuring communities of competing species. In fragmented landscapes, however, a paradox can exist: different taxa may competitively dominate different types of habitat patches, resulting in a form of spatial niche partitioning, yet differences in long-term distributions among species can appear surprisingly small. This paradox is illustrated by an emblematic metacommunity - that of Daphnia spp. in rockpools on the Finnish Baltic coast, where three species compete with each other, have distinct ecological preferences, yet largely overlap in long-term distributions. Here we examine how metacommunity models that explicitly estimate species-specific demographic parameters can solve the apparent paradox. Our research confirms previous studies that local extinction rates are influenced by environmental variables in a strong and species-specific way and are considerably increased by interspecific competition. Yet, our simulations show that this situation exists alongside interspecific differences in realized niches that are, overall, small, and identified three main explanations for this compatibility. Our results illustrate how state-space modelling can clarify complex metacommunity dynamics and explain why local competition and niche differentiation do not always scale up to the landscape level.

Abstract

Phenotypic plasticity is an important feature of biological systems that is likely to play a major role in the future adaptation of organisms to the ongoing global changes. It may allow an organism to produce alternative phenotypes in responses to environmental cues. Modifications in the phenotype can be reversible but are sometimes enduring and can even span over generations. The notion of phenotypic plasticity was conceptualized in the early 20thcentury by Richard Woltereck. He introduced the idea that the combined relations of a phenotypic character and all environmental gradients that influence on it can be defined as “norm of reaction”. Norms of reaction are specific to species and to lineages within species, and they are heritable. He postulated that reaction norms can progressively be shifted over generations depending on the environmental conditions. One of his biological models was the water-fleedaphnia. Woltereck proposed that enduring phenotypic modifications and gene mutations could have similar adaptive effects, and he postulated that their molecular bases would be different. Mutations occurred in genes, while enduring modifications were based on something he called theMatrix. He suggested that this matrix (i) was associated with the chromosomes, (ii) that it was heritable, (iii) it changed during development of the organisms, and (iv) that changes of the matrix could be simple chemical substitutions of an unknown, but probably polymeric molecule. We reasoned that the chromatin has all postulated features of this matrix and revisited Woltereck’s classical experiments withdaphnia. We developed a robust and rapid ATAC-seq technique that allows for analyzing chromatin of individual daphnia and show here (i) that this technique can be used with minimal expertise in molecular biology, and (ii) we used it to identify open chromatin structure in daphnia exposed to different environmental cues. Our result indicates that chromatin structure changes consistently in daphnia upon this exposure confirming Woltereck’s classical postulate.

Abstract

Recent insights in evolutionary biology have shed light on epigenetic variation that interacts with genetic variation to convey heritable information. An important characteristic of epigenetic changes is that they can be produced in response to environmental cues and passed on to later generations, potentially facilitating later genetic adaptation. While our understanding of epigenetic mechanisms in vertebrates is rapidly growing, our knowledge about invertebrates remains lower, or is restricted to model organisms. Mollusks in particular, are a large group of invertebrates, with several species important for ecosystem function, human economy and health. In this review, we attempt to summarize the literature on epigenetic and intergenerational studies in mollusk species, with potential importance for adaptive evolution. Our review highlights that two molecular bearers of epigenetic information, DNA methylation and histone modifications, are key features for development in mollusk species, and both are sensitive to environmental conditions to which developing individuals are exposed. Further, although studies are still scarce, various environmental factors (e.g. predator cues, chemicals, parasites) can induce intergenerational effects on the phenotype (life-history traits, morphology, behaviour) of several mollusk taxa. More work is needed to better understand whether environmentally-induced changes in DNA methylation and histone modifications have phenotypic impacts, whether they can be inherited through generations and their role in intergenerational effects on phenotype. Such work may bring insights into the potential role of epigenetic in adaptation and evolution in mollusks.

Abstract

We studied the population genetic structure of the freshwater snail Galba schirazensis (Küster, 1862), a potential vector of infectious diseases such as fascioliasis. Galba schirazensis has now a worldwide distribution but a poorly known origin because this species has been distinguished only recently from the morphologically similar and cosmopolitan Galba truncatula (O.F. Müller, 1774). We developed specific microsatellite markers and sequenced a mitochondrial gene (cytochrome oxidase subunit I (CO1)) to study individuals of G. schirazensis from the Old World and the New World. We found very low genetic diversity within populations, no heterozygotes, and marked population structure — a pattern observed in other highly selfing lymnaeid species with recently enlarged distributions as a result of biological invasions. The total lack of observed heterozygosity in the few populations of G. schirazensis that displayed some allelic diversity suggests high selfing rates. We also found that the center of diversity, and by extension the origin area of this species, should be found in the New World, whereas Old World populations should rather result from a recent introduction of a genetically uniform population. The microsatellite markers developed here will help to clarify the history of expansion of G. schirazensis and might help to understand its role as a potential vector of infectious diseases. Abstract

International audience; Biological invasions offer interesting situations for observing how novel interactions between closely related, formerly allopatric species may trigger phenotypic evolution in situ. Assuming that successful invaders are usually filtered to be competitively dominant, invasive and native species may follow different trajectories. Natives may evolve traits that minimize the negative impact of competition, while trait shifts in invasives should mostly reflect expansion dynamics, through selection for colonization ability and transiently enhanced mutation load at the colonization front. These ideas were tested through a large-scale common-garden experiment measuring life-history traits in two closely related snail species, one invasive and one native, co-occurring in a network of freshwater ponds in Guadeloupe. We looked for evidence of recent evolution by comparing uninvaded or recently invaded sites with long-invaded ones. The native species adopted a life history favoring rapid population growth (i.e., increased fecundity, earlier reproduction, and increased juvenile survival) that may increase its prospects of coexistence with the more competitive invader. We discuss why these effects are more likely to result from genetic change than from maternal effects. The invader exhibited slightly decreased overall performances in recently colonized sites, consistent with a moderate expansion load resulting from local founder effects. Our study highlights a rare example of rapid life-history evolution following invasion.

Abstract

Batesian mimicry, with its highly colorful species and astonishing mimic-model resemblance, is a canonical example of evolution by natural selection. However, Batesian mimicry could also occur in inconspicuous species and rely on subtle resemblance. Although potentially widespread, such instances have been rarely investigated, such that the real frequency of Batesian mimicry has remained largely unknown. To fill this gap, we developed a new approach using deep learning to quantify the resemblance between putative mimics and models from photographs. We applied this method to quantify the frequency of Batesian mimicry in Western Palearctic snakes. Potential mimics were revealed by an excess of resemblance with sympatric venomous snakes compared to random expectations. We found that 8% of the non-venomous species were potential mimics, among which all were imperfect mimics. This study is the first to quantify the frequency of Batesian mimicry in a whole community of vertebrates, and shows that even concealed species can be reliably identified as potential models. Our approach should prove useful to detect mimicry in other communities, and more generally it highlights the benefits of deep learning for quantitative studies of phenotypic resemblance.

Abstract

Bateman's principles posit that male fitness varies more, and relies more on mate acquisition, than female fitness. While Bateman's principles should apply to any organism producing gametes of variable sizes, their application to plants is potentially complicated by the high levels of polyandry suspected for plants, and by variation in the spatial distribution of prospective mates. Here we quantify the intensity of sexual selection by classical Bateman metrics using two common gardens of the wind-pollinated dioecious plant Mercurialis annua . Consistent with Bateman's principles, males displayed significantly positive Bateman gradients (a regression of fitness on mate number), whereas the reproductive success of females was independent of their ability to access mates. A large part of male fitness was explained by their mate number, which in turn was associated with males' abilities to disperse pollen. Our results suggest that sexual selection can act in plant species in much the same way as in many animals, increasing the number of mates through traits that promote pollen dispersal.

Abstract

International audience; Sexual selection may contribute to the evolution of plant sexual dimorphism by favoring architectural traits in males that improve pollen dispersal to mates. In both sexes, larger individuals may be favored by allowing the allocation of more resources to gamete production (a ‘budget’ effect of size). In wind‐pollinated plants, large size may also benefit males by allowing the liberation pollen from a greater height, fostering its dispersal (a ‘direct’ effect of size). To assess these effects and their implications for trait selection, we measured selection on plant morphology in both males and females of the wind‐pollinated dioecious herb Mercurialis annua in two separate experimental common gardens at contrasting density. In both gardens, selection strongly favored males that disperse their pollen further. Selection for pollen production was observed in the high‐density garden only and was weak. In addition, male morphologies associated with increased mean pollen dispersal differed between the two gardens, as elongated branches were favored in the high‐density garden while shorter plants with longer inflorescence stalks were favored in the low‐density garden. Larger females were selected in both gardens. Our results point to the importance of both a direct effect of selection on male traits that affect pollen dispersal, and, to a lesser extent, a budget effect of selection on pollen production.

Abstract

The diversity of animal visual displays has intrigued scientists for centuries. Sexual selection theory has explained some of this diversity, yet most of this effort has focused on simple aspects of signal design, such as color. The evolution of complex patterns that characterize many sexual displays remains largely unexplained. The field of empirical aesthetics, a subdiscipline of cognitive psychology, has shown that humans are attracted to visual images that match the spatial statistics of natural scenes. We investigated whether applying this result to animals could help explain the diversification of complex sexual signaling patterns. We used Fourier analysis to compare the spatial statistics of body patterning in ten species of darters (Etheostoma spp.), a group of freshwater fishes with striking male visual displays, with those of their respective habitats. We found a significant correlation between the spatial statistics of darter patterns and those of their habitats for males, but not for females. Our results suggest that visual characteristics of natural environments can influence the evolution of complex patterns in sexual signals.

Abstract

1. Heterozygosity fitness correlations (HFCs) have been used extensively to explore the impact of inbreeding on individual fitness. Initially, most studies used small panels of microsatellites, but more recently with the advent of next generation sequencing, large SNP datasets are becoming increasingly available and these provide greater power and precision to quantify the impact of inbreeding on fitness. 2. Despite the popularity of HFC studies, effect sizes tend to be rather small. One reason for this may be a low variation in inbreeding level across individuals. Using genetic markers, it is possible to measure variance in inbreeding through the strength of correlation in heterozygosity across marker loci, termed identity disequilibrium (ID). 3. ID can be quantified using the measure g2 which is also a central parameter in HFC theory that can be used within a wider framework to estimate the direct impact of inbreeding on both marker heterozygosity and fitness. However, no software exists to calculate g2 for large SNP datasets nor to implement this framework. 4. inbreedR is an R package that provides functions to calculate g2 based on microsatellite and SNP markers with associated p-values and confidence intervals. Within the framework of HFC theory, inbreedR also estimates the impact of inbreeding on marker heterozygosity and fitness. Moreover, we implemented easy-to-use simulations to explore the precision and magnitude of estimates based on different numbers of genetic markers. We hope this package will facilitate good practice in the analysis of HFCs and help to deepen our understanding of inbreeding effects in natural populations.

Abstract

Basic models of mating-system evolution predict that hermaphroditic organisms should mostly either cross-fertilize, or self-fertilize, due to self-reinforcing coevolution of inbreeding depression and outcrossing rates. However transitions between mating systems occur. A plausible scenario for such transitions assumes that a decrease in pollinator or mate availability temporarily constrains outcrossing populations to self-fertilize as a reproductive assurance strategy. This should trigger a purge of inbreeding depression, which in turn encourages individuals to self-fertilize more often and finally to reduce male allocation. We tested the predictions of this scenario using the freshwater snail Physa acuta, a self-compatible hermaphrodite that preferentially outcrosses and exhibits high inbreeding depression in natural populations. From an outbred population, we built two types of experimental evolution lines, controls (outcrossing every generation) and constrained lines (in which mates were often unavailable, forcing individuals to self-fertilize). After ca. 20 generations, individuals from constrained lines initiated self-fertilization earlier in life and had purged most of their inbreeding depression compared to controls. However, their male allocation remained unchanged. Our study suggests that the mating system can rapidly evolve as a response to reduced mating opportunities, supporting the reproductive assurance scenario of transitions from outcrossing to selfing.

Abstract

Sexual selection operates through consecutive episodes of selection that ultimately contribute to the observed variance in reproductive success between individuals. Understanding the relative importance of these episodes is challenging, particularly because the relevant postcopulatory fitness components are often difficult to assess. Here, we investigate different episodes of sexual selection on the male sex function, by assessing how (precopulatory) mating success, and (postcopulatory) sperm-transfer efficiency and sperm-fertilizing efficiency contribute to male reproductive success. Specifically, we used a transgenic line of the transparent flatworm, Macrostomum lignano, which expresses green fluorescent protein (GFP) in all cell types, including sperm cells, enabling in vivo sperm tracking and paternity analysis. We found that a large proportion of variance in male reproductive success arose from the postcopulatory episodes. Moreover, we also quantified selection differentials on 10 morphological traits. Testis size and seminal vesicle size showed significant positive selection differentials, which were mainly due to selection on sperm-transfer efficiency. Overall, our results demonstrate that male reproductive success in M. lignano is not primarily limited by the number of matings achieved, but rather by the ability to convert matings into successful fertilizations, which is facilitated by producing many sperm.

Abstract

Sex allocation theory predicts that simultaneous hermaphrodites evolve to an evolutionary stable resource allocation, whereby any increase in investment to male reproduction leads to a disproportionate cost on female reproduction and vice versa. However, empirical evidence for sexual trade-offs in hermaphroditic animals is still limited. Here, we tested how male and female reproductive traits evolved under conditions of reduced selection on either male or female reproduction for 40 generations in a hermaphroditic snail. This selection favors a reinvestment of resources from the sex function under relaxed selection toward the other function. We found no such evolutionary response. Instead, juvenile survival and male reproductive success significantly decreased in lines where selection on the male function (i.e., sexual selection) was relaxed, while relaxing selection on the female function had no effect. Our results suggest that most polymorphisms under selection in these lines were not sex-antagonistic. Rather, they were deleterious mutations affecting juvenile survival (thus reducing both male and female fitness) with strong pleiotropic effects on male success in a sexual selection context. These mutations accumulated when sexual selection was relaxed, which supports the idea that sexual selection in hermaphrodites contributes to purge the mutation load from the genome as in separate-sex organisms.

Abstract

Modeling the dynamics of competition and coexistence between species is cru- cial to predict long-term impacts of invasive species on their native congeners. However, natu- ral environments are often fragmented and variable in time and space. In such contexts, regional coexistence depends on complex interactions between competition, niche differentia- tion and stochastic colonization–extinction dynamics. Quantifying all these processes at land- scape scale has always been a challenge for ecologists. We propose a new statistical framework to evaluate metapopulation parameters (colonization and extinction) in a two-species system and how they respond to environmental variables and interspecific competition. It requires spatial surveys repeated in time, but does not assume demographic equilibrium. We apply this model to a long-term survey of two snails inhabiting a network of freshwater habitats in the West Indies. We find evidence of reciprocal competition affecting colonization or extinction rates, modulated by species-specific sensitivity to environmental variables. Simulations using model estimates allow us to predict species dynamics and explore the role of various coexis- tence mechanisms described by metacommunity theory in our system. The two species are pre- dicted to stably coexist, because niche partitioning, source–sink dynamics and interspecific differences in extinction–colonization parameters all contribute to reduce the negative impacts of competition. However, none of these mechanisms is individually essential. Regional coexis- tence is primarily facilitated by transient co-occurrence of the two species within habitat patches, a possibility generally not considered in theoretical metacommunity models. Our framework is general and could be extended to guilds of several competing species.

Abstract

Theory and empirical data showed that two processes can boost selection against deleterious mutations, thus facilitating the purging of the mutation load: inbreeding, by exposing recessive deleterious alleles to selection in homozygous form, and sexual selection, by enhancing the relative reproductive success of males with small mutation loads. These processes tend to be mutually exclusive because sexual selection is reduced under mating systems that promote inbreeding, such as self‐fertilization in hermaphrodites. We estimated the relative efficiency of inbreeding and sexual selection at purging the genetic load, using 50 generations of experimental evolution, in a hermaphroditic snail (Physa acuta). To this end, we generated lines that were exposed to various intensities of inbreeding, sexual selection (on the male function), and nonsexual selection (on the female function). We measured how these regimes affected the mutation load, quantified through the survival of outcrossed and selfed juveniles. We found that juvenile survival strongly decreased in outbred lines with reduced male selection, but not when female selection was relaxed, showing that male‐specific sexual selection does purge deleterious mutations. However, in lines exposed to inbreeding, where sexual selection was also relaxed, survival did not decrease, and even increased for self‐fertilized juveniles, showing that purging through inbreeding can compensate for the absence of sexual selection. Our results point to the further question of whether a mixed strategy combining the advantages of both mechanisms of genetic purging could be evolutionary stable. Fil: Noël, Elsa. Abstract

International audience; A molecular tool described here allows in one step for specific discrimination among three cryptic freshwater snail species (genus Galba) involved in fasciolosis transmission, a worldwide infectious disease of humans and livestock. The multiplex PCR approach taken targets for each species a distinctive, known microsatellite locus which is amplified using specific primers designed to generate an amplicon of a distinctive size that can be readily separated from the amplicons of the other two species on an agarose gel. In this way, the three Galba species (G. cubensis, G. schirazensis, and G. truncatula) can be differentiated from one another, including even if DNA from all three were present in the same reaction. The accuracy of this new molecular tool was tested and validated by comparing multiplex PCR results with species identification based on sequences at mitochondrial and nuclear markers. This new method is accurate, inexpensive, simple, rapid, and can be adapted to handle large sample sizes. It will be helpful for monitoring invasion of Galba species and for developing strategies to limit the snail species involved in the emergence or re-emergence of fasciolosis.

Abstract

[Departement_IRSTEA]Territoires [TR1_IRSTEA]SEDYVIN; International audience; Community genetics aims at understanding how within-species variation, species diversity and environmental factors interact to shape community assembly. An approach that emerged a few years ago has been to quantify the correlation between the neutral genetic diversity of a focal species and species diversity of the surrounding community (species–genetic diversity correlations, or SGDCs). We here review this approach and discuss its interpretative framework in a community ecology context. First, we show that the case for mostly positive SGDCs is probably overstated due to publication bias – only 11% are significantly positive, a fraction comparable to the significantly negative ones. This suggests that variation in area and connectivity among habitat patches, theoretically leading to positive SGDCs, is not the only factor affecting SGDCs. Second, building upon previous contributions, we propose a general framework to identify the multiple factors underpinning SGDCs, and argue that it will help deepen our understanding of community assembly, especially with regard to the ecological factors playing at metacommunity scale. Our framework distinguishes between site and community factors which can affect SGDCs either positively or negatively, depending on whether the focal species and the rest of the community are similar or dissimilar, in terms of realized niches and dispersal abilities. Empirical studies should thus go beyond simply computing SGDCs, and we provide statistical methods (e.g. structural equation modelling) to decompose SGDCs into the multiple contributions of site and community factors. As an example, we use a published dataset (freshwater snail metacommunity), and show how the role of focal population size on SGDCs had hitherto not been detected. We further discuss how considering several focal species and various delimitations of the community may help one to identify clusters of ecologically similar species. We eventually highlight the benefit that SGDC studies would get from integrating beta-diversities.

Abstract

International audience; Why do some species successfully invade new environments? Which of these invasive species will alter or even reshape their new environment? The answers to these questions are simultaneously critical and complex. They are critical because invasive species can spectacularly alter their new environment, leading to native species extinctions or loss of important ecosystem functions that fundamentally reduce environmental and societal services. They are complex because invasion success in a novel environment is influenced by various attributes embedded in natural landscapes—biogeographical landscape properties, abiotic environmental characteristics, and the relationship between the invasive species and the resident species present in the new environment. We explore whether a condensed record of the relationships among species, in the form of a network, contains the information needed to understand and predict invasive species success and subsequent impacts. Applying network theory to study invasive species is a relatively novel approach. For this reason, much research will be needed to incorporate existing ecological properties into a network framework and to identify which network features hold the information needed to understand and predict whether or not an invasive species is likely to establish or come to dominate a novel environment. This paper asks and begins to answer the 14 most important questions that biologists must address to integrate network analysis into the study of invasive species. Answering these questions can help ecologists produce a practical monitoring scheme to identify invasive species before they substantially alter native environments or to provide solutions to mitigate their harmful impacts.

Abstract

International audience; Self-fertilization is widely believed to be an ''evolu-tionary dead end'' [1, 2], increasing the risk of extinction [3] and the accumulation of deleterious mutations in genomes [4]. Strikingly, while the failure to adapt has always been central to the dead-end hypothesis [1, 2], there are no quantitative genetic selection experiments comparing the response to positive selection in selfing versus outcrossing populations. Here we studied the response to selection on a morphological trait in laboratory populations of a hermaphroditic, self-fertile snail under either selfing or outcrossing. We applied both treatments to two types of populations: some having undergone frequent selfing and purged a substantial fraction of their mutation load in their recent history [5], and others continuously maintained under outcrossing. Populations with a history of outcrossing respond faster to selection than those that have experienced selfing. In addition, when self-fertilization occurs during selection, the response is initially fast but then rapidly slows, while out-crossing populations maintain their response throughout the experiment. This occurs irrespective of past selfing history, suggesting that high levels of inbreeding depression, contrary to expectation [6], do not set strong limits to the response to selection under inbreeding, at least at the timescale of a few generations. More surprisingly, pheno-typic variance is consistently higher under selfing, although it quickly becomes less responsive to selection. This implies an increase in non-heritable variance, hence a breakdown of developmental canalization [7] under selfing. Our findings provide the first empirical support of the short-term positive and long-term negative effects of selfing on adaptive potential.

Abstract

The differential plasticity hypothesis suggests that sexual dimorphism in dioecious plants could evolve in response to sex-specific resource requirements for reproduction (i.e., high carbon requirements for ovules and high nitrogen demands for pollen). When resources become limiting during growth, males and females should, therefore, adjust their allocation to resource-harvesting organs differently. To investigate the potential for plants to respond to resource limitation late in life and to test the differential plasticity hypothesis, we grew male and female individuals of the annual wind-pollinated plant Mercurialis annua in a common garden. Late in the growth season, we simulated a change in competition by decreasing plant density in half of the replicates. We measured both allocation to vegetative and reproductive traits and analyzed the relative allocation to reproduction vs. growth. Males and females differentially adjusted their resource allocation in response to varying plant densities, despite the fact that they were reproductively mature. Males maintained the same relative allocation of resource to reproductive vs. vegetative tissues at both densities. In contrast, females reduced vegetative growth proportionally less than seed production at the higher density. Our results highlight the dynamic nature of allocation decisions taken by plants, which respond quickly and in a sexually dimorphic way to changes in their competitive circumstances. The existence of resource ‘currencies’ limiting male and female functions differently have potentially led to the evolution of sex-specific strategies of resource acquisition and deployment, with females conserving resources for vegetative organs to ensure their future carbon-rich reproduction. Abstract

The freshwater snail Galba cubensis (Pfeiffer, 1839) has a large distribution in the Americas. Despite being an intermediate host of Fasciola hepatica - the trematode causing fasciolosis in livestock and humans - its population genetics have never been studied. We isolated and characterized 15 microsatellite loci in G. cubensis to evaluate its genetic diversity, population-genetic structure and mating system. We tested the microsatellite loci in 359 individuals from 13 populations of G. cubensis from Cuba, Guadeloupe, Martinique, Puerto Rico, Venezuela, Colombia and Ecuador. We also tested cross-amplification in three closely related species: G. truncatula, G. viator and G. neotropica. We found that G. cubensis has a similar population structure to other selfing lymnaeids that live in temporary habitats: low genetic diversity, large departure from Hardy-Weinberg equilibrium, marked population structure and high selfing rate. We found that seven and six loci amplified in G. truncatula and G. viator, respectively, and that all 15 loci amplified in G. neotropica. This last finding suggests a close relatedness between G. cubensis and G. neotropica, probably being conspecific and synonymous. This new set of microsatellite markers will be a useful tool to study the genetic diversity of this snail species across a large geographical range and, consequently, to understand the emergence and re-emergence of fasciolosis in the Americas., Centro de Estudios Parasitológicos y de Vectores, Facultad de Ciencias Naturales y Museo
