Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Células B-1, um subtipo de linfócitos B, estão presentes nos tecidos linfoides e nas cavidades peritoneal e pleural. Apresentam a capacidade de produzir anticorpos naturais e induzíveis, secretam citocinas, podem fagocitar patógenos, migrar para foco inflamatório inespecífico e modular a resposta imunológica. No entanto, aspectos da fisiologia dessas células como o seu papel em situação de estresse (por exemplo na privação e/ou restrição de sono), ainda não foram esclarecidos. Dessa forma, esse trabalho teve por objetivo avaliar a ativação e diferenciação de células B-1 murinas em contexto de restrição de sono. Para tanto, grupos de camundongos machos da linhagem C57BL/6 foram submetidos ao protocolo de 18h de restrição de sono por 21 dias, sendo cada grupo infectado por diferentes micro-organismos ao vigésimo dia: Escherichia coli (bactéria), Candida albicans e Paracoccidioides brasiliensis (fungos). Então, células B-1 peritoneais desses animais foram purificadas e avaliadas quanto expressão de marcadores de ativação (CD80, CD86, CD40 e MHC-II), produção de moléculas microbicidas (NO e ROS) bem como expressão dos genes de citocinas (IL-6, IL-10, IL-12 e TNF-α), TLRs (2, 6, 9) e fatores de comprometimento linfoide e mieloide (EBF, E2A, IL-7R, G-csfr, M-csfr e Spi1). Nossos resultados mostraram que para todos os modelos de infecção, a restrição de sono estimulou em pouco ou nada a expressão de CD40, CD80, CD86, e MHC-II em células B-1. A produção de moléculas microbicidas por sua vez foi maior nos animais restritos de sono infectados com P. brasiliensis (NO) e C. albicans (ROS). Ainda, os resultados da expressão de genes de TLRs mostraram que nos modelos infectados com fungos houve aumento na expressão de TLR-2. Foi observado também que a expressão dos genes de citocinas foi maior em animais restritos infectados com P. brasiliensis (IL-12) e C. albicans (IL-12, IL-6 e TNF-α). Os dados de expressão dos genes de comprometimento de linhagem mostraram que para todos os modelos de infecção, foi observada polarização de células B-1 para perfil mieloide nos animais restritos de sono. Em conjunto, os resultados obtidos trazem importantes informações acerca das células B-1 e sugerem que a restrição de sono por si só pode influenciar a ativação e diferenciação dessas células, porém tais mudanças dependem do tipo de estímulo de infecção, ressaltando ainda mais a importância de estudos que avaliem alterações imunológicas decorrentes da restrição do sono. B1 cells are a subtype of B lymphocytes distributed in lymphoid tissues and peritoneal and pleural cavities. These cells produce natural and inducible antibodies, secrete cytokines, phagocyte pathogens, migrate to nonspecific inflammatory focus and are able to modulate the immune response. However, the physiological aspects of B1 cells in stress condition (sleep restriction, for example), are poorly known. Thus, the present study aims to evaluate murine B1 cells physiology, activation and differentiation in sleep restriction context. For that, male C57BL/6 mice were submitted to 18 h sleep restriction protocol for 21 days. Then, mice were infected by a different microorganism on the twentieth day: Escherichia coli (bacteria), Candida albicans and Paracoccidioides brasiliensis (fungi). After 24 h, peritoneal B1 cells were purified and evaluated for activation markers expression (CD80, CD86, CD40 and MHCII), cytokines (IL6, IL10, IL12 e TNF), TLRs (2, 6 and 9) and lymphoid and myeloid commitment factors (EBF, E2A, IL7R, Gcsfr, Mcsfr and Spi1). Our results showed that sleep restriction have no effect on expression of CD40, CD80, CD86, and MHCII in B1 cells. Microbicidal molecules were higher in sleeprestricted animals infected with P. brasiliensis (NO) and C. albicans (ROS). Furthermore, TLRs genes expression results showed that fungi infected models can lead to an increase in TLR2 expression in sleeprestricted animals. It was also observed that cytokines gene expression were higher in restricted animals infected with P. brasiliensis (IL12) and C. albicans (IL12, IL6 and TNF). To all infection models (P. brasiliensis, C. albicans and E. coli), a myeloid profile polarization was observed in sleeprestricted animals. Altogether, our results bring valuable information about B1 cells, suggesting that sleep restriction alone can influence B1 cell activation and differentiation, but such changes depend on infection stimulus, highlighting the importance of more studies that assess immunological changes resulting due sleep restriction.

A infecção murina pelo T. cruzi Sylvio X10/4, um parasita de baixa virulência, possibilita que camundongos CD28KO infectados sobrevivam por um longo período de tempo. Esta característica permite estudar o impacto da ausência de CD28 sobre vários aspectos da resposta dos linfócitos T e B. No presente trabalho, nosso principal objetivo foi verificar, em camundongos CD28KO e C57BL/6 (WT, utilizado como controle), infectados ou não pelo parasita, quais aspectos da resposta imune são preservados na ausência deste co-receptor. Comparados aos camundongos controle C57BL/6, os camundongos CD28KO se mostraram suscetíveis à infecção pelo T cruzi Sylvio X10/4, resultando na morte dos animais no decorrer de um amplo período, que variou de algumas semanas a quase um ano. Os camundongos CD28KO exibiram taxas mais elevadas de parasitemia subpatente do que os camundongos C57BL/6, bem como infiltrados inflamatórios mais intensos e duradouros no coração (miocárdio e pericárdio). Acompanhando os camundongos CD28KO em diferentes momentos da infecção, encontramos uma expansão inicial do compartimento linfocitário seguida de sua contração com o início da fase crônica, não destacando nenhuma diferença significativa na frequência e número total de linfócitos T e B no baço, se comparadas à dos camundongos C57BL/6 infectados. Além disso, observamos que linfócitos TCD4 e TCD8 de camundongos CD28KO infectados produzem IFNgamma. Por outro lado, apesar de observarmos uma diminuição da produção de IL-2 pelos linfócitos TCD4 e TCD8 de camundongos CD28KO infectados, é inegável que a produção desta citocina ocorre na ausência de CD28. Diferentemente, no dia 16 da infecção pelo T. cruzi Sylvio X10/4, a análise dos linfócitos T auxiliares foliculares, TFH (do inglês Follicular Helper T cells), mostrou que, ao contrário do observado no camundongo C57BL/6, não houve geração destas células nos camundongos CD28KO infectados. Em forma análoga, não observamos nestes animais a presença da população de células B de Centros Germinativos, GC-B (do inglês, Germinal Center B cells). A nível sérico, observamos que na fase aguda da infecção, além de anticorpos parasitaespecíficos da classe IgM, os camundongos CD28KO infectados produzem níveis discretos, embora significativos, de anticorpos IgG2c frente ao T. cruzi , semelhantes aos dos camundongos C57BL/6 neste momento da infecção. Porém, na fase crônica, enquanto o nível de IgG2c frente ao T. cruzi teve um grande aumento no soro do camundongo C57BL/6 infectado, no camundongo CD28KO infectado, o seu nível sérico permaneceu análogo àquele observado na fase aguda. Os resultados do presente estudo levantam a possibilidade de que anticorpos anti-T cruzi da classe IgM, assim como aqueles da classe IgG (produzidos em pequenas quantidades nos camundongos CD28KO infectados devido à ausência dos elementos necessários para a mutação somática), poderiam ser suficientes para determinar o controle parcial das formas circulantes do parasita, permitindo a sobrevivência do camundongo CD28KO por muito tempo. Isto seria possível no caso de infecção pelo T. cruzi Sylvio X10/4 por se tratar de um parasita que determina níveis subpatentes de parasitemia. No presente trabalho discutimos a possível origem desses anticorpos extrafoliculares, procurando desvendar o quadro denominado ativação policlonal do sistema imunológico, presente em diversos processos infecciosos e ainda pouco esclarecido. The murine infection by T. cruzi Sylvio X10/4, a low virulence parasite, allows infected CD28KOmice to survive for a long period of time. This characteristic permits us to study the impact of the absence of CD28 on various aspects of the T and B lymphocyte response. In the present study, our main objective was to verify, in CD28KO and C57BL/6 (WT, used as control) mice, infected or not by the parasite, which aspects of the immune response are preserved in the absence of CD28. Compared to control C57BL/6 mice, CD28KO mice were susceptible to infection by T. cruzi Sylvio X10/4, resulting in the animals\' death over a wide period, which ranged from a few weeks to almost a year. CD28KO mice exhibited higher rates of subpatent parasitemia than C57BL/6 mice, as well as more intense and lasting inflammatory infiltrates in the heart (myocardium and pericardium). Accompanying the CD28KO mice at different times of the infection, we found an initial expansion of the lymphocyte compartment followed by its contraction with the beginning of the chronic phase, not displaying significative differences in the frequency and the total number of T and B spleen lymphocytes, when compared to those in the infected C57BL/6 mice. In addition, we observed that both TCD4 and TCD8 lymphocytes from infected CD28KO mice produce IFNgamma. Moreover, although we observed a decrease in IL-2 production by TCD4 and TCD8 lymphocytes from infected CD28KO mice, it is undeniable that the production of this cytokine occurs in the absence of CD28. Differently, on day 16 of the infection by T. cruzi Sylvio X10/4, the analysis of follicular auxiliary T lymphocytes, TFH (from the English Follicular Helper T cells) showed, that, unlike that observed in the infected C57BL/6 mouse, there was no formation of these cells in infected CD28KO mice. Similarly, we did not observe in these animals the presence of the Germinal Center B cell population, GC-B (from the English, Germinal Center B cells). At the serum level, we observed that in the acute phase of infection, in addition to parasitespecific antibodies of the IgM class, infected CD28KO mice produce discrete, although significant, levels of IgG2c antibodies to T. cruzi , similar to that of C57BL/6 mice at this time ofinfection. However, in the chronic phase, while the level of IgG2c against T. cruzi displayed alarge increase in the serum of the infected C57BL/6 mouse, in the infected CD28KO mouse, itslevels did not differ from that observed in the acute phase. The results of the present study raise the possibility that anti-T cruzi antibodies of the IgM class, as well as those of the IgG class (produced in small quantities in infected CD28KO mice due to the absence of the necessary elements for the somatic mutation), could be sufficient todetermine the partial control of the circulating forms of the parasite, allowing the survival of the CD28KO mouse for a long time. This would be possible in the case of infection by T. cruzi Sylvio X10/4, as it is a parasite that determines subpatent levels of parasitemia. In the present work, we discussed the possible origin of these extra-follicular antibodies, seeking to unravel the picture called polyclonal activation of the immune system, present in several infectiousprocesses and still poorly clarified.

A resposta imune desencadeada em pacientes com leishmaniose tegumentar Americana (LTA) é predominantemente celular. Assim, o estudo dos mecanismos imunológicos responsáveis pela formação, progressão e cura desta infecção é de grande importância, considerando a deformação que ela pode causar em indivíduos infectados. Este estudo investigou a expressão de mRNA de IFN-?, IL-10, IL-27, TNF-?, TGF-? e IL-6 em 40 biopsias de parafina (FFPE) fixadas em formalina de pacientes com diagnóstico clínico e laboratorial para LTA. Um grupo de 10 biópsias FFPE coletadas de pacientes com diagnóstico clínico de leishmaniose mucosa (LM) foi utilizado para o controle da evolução da infecção. Inicialmente, foi padronizado a extração de RNA, a síntese de cDNA e a escolha do gene endógeno em 9 biopsias frescas e 8 FFPE. Após essas padronizações, as moléculas de RNA de fragmentos de biopsias parafinadas, foram extraídas com kit específico, tratadas com DNase, quantificadas por fluorimetria e sintetizadas para cDNA. A expressão relativa de cada citocina foi determinada (em duplicata) por PCR em tempo real. As reações foram normalizadas frente ao padrão de expressão do gene endógeno GAPDH, que foi previamente padronizado. O padrão de expressão de cada gene alvo foram determinados segundo a fórmula do \"CT comparativo\" (2-??CT), na qual calcula-se quantas vezes mais ocorre a expressão do gene alvo em relação a indivíduos normais. Também incluído nesse estudo 5 amostras de biopsia de pele de indivíduos normais (grupo IV). Os resultados foram expressos em média de quantificação relativa, a análise estatística foi realizada utilizando foi determinada por Teste t não pareado e teste F para comparação de variâncias, onde três grupos de pacientes foram formados. Grupo I, 35 pacientes com LTA, cuja expressão relativa de IFN-? foi inferior a 100. Grupo II, 5 pacientes com LTA cuja expressão relativa de IFN-? foi superior a 100. Grupo III, 10 pacientes com leishmaniose mucosa. Os níveis de expressão para IFN-? foram 33,34; 214,1 e 129,6 vezes maiores para os Grupos I, II e III, respectivamente do que as amostras do controle normal. Para o TNF-? foram 11,22; 2,37 e 4,59 vezes superiores ao normal nos Grupos I, II e III, respectivamente. Para IL-10 foram 5,14; 11,54 e 2,51 vezes superiores aos valores normais nos Grupos I, II e III, respectivamente. Para IL-27 foram 15,81; 85,29 e 17,29 vezes superiores ao normal nos Grupos I, II e III, respectivamente. Para o TGF-? foram 5,38; 4,33 e 4,72 vezes superiores ao normal nos Grupos I, II e III, respectivamente. Para a IL-6 foram 5,92; 5,69 e 2,09 vezes superiores aos valores normais nos Grupos I, II e III, respectivamente. Estes resultados foram semelhantes a resultados de estudos que analisam a expressão de citocinas já sintetizadas. No geral, os resultados de expressão gênica das citocinas estudadas sugerem que a exacerbação da resposta imune do perfil Th1 (IFN-?) poderia levar ao surgimento de lesões mais graves nos pacientes dos Grupos II e III. Os níveis elevados de mRNA de IL-10, observado nos pacientes do Grupo II podem promover baixa ativação macrofágica, colaborando com a replicação do parasita e assim culminar com a progressão da doença. A expressão de mRNA de IL-27 não interferiu na expressão de mRNA de IFN-? nas células dos pacientes com leishmaiose tegumentar ou leishmaniose mucocutânea. Em conclusão, estes resultados podem sugerir que os pacientes do Grupo II podem desenvolver formas mais graves de LTA quando comparados aos do Grupo I. The immune response triggered in patients with American cutaneous leishmaniasis (ACL) is predominantly cellular. Thus, the study of the immunological mechanisms responsible for the formation, progression and cure of this infection is of great importance, considering the deformation that it can cause in infected individuals. This study investigated the expression of IFN-?, IL-10, IL-27, TNF-?, TGF-? and IL-6 mRNA in 40 formalin-fixed paraffin-embedded paraffin biopsies (FFPE) from patients with clinical and laboratory diagnosis for LTA. A group of 10 FFPE biopsies collected from patients with clinical diagnosis of mucosal leishmaniasis (MCL) was used to control the evolution of the infection. Initially, RNA extraction, cDNA synthesis and the choice of the endogenous gene were standardized in 9 fresh biopsies and 8 FFPE. After these standardizations, the RNA molecules of fragments of paraffin-shaped biopsies were extracted with a specific kit, treated with DNase, quantified by fluorimetry and synthesized for cDNA. The relative expression of each cytokine was determined (in duplicate) by real-time PCR. Reactions were normalized against the expression pattern of the endogenous GAPDH gene, which was previously standardized. The expression pattern results for each target gene were determined by the \"comparative CT\" (2-??CT) formula, in which it is calculated how many times more the expression of the target gene occurs in relation to normal individuals. Also included in this study 5 skin biopsy samples from normal subjects (group IV). The results were expressed as mean relative quantification, statistical analysis was performed using was determined by unpaired t test and F test for comparison of variances, where three groups of patients were formed. Group I, 35 patients with ACL, whose relative expression of IFN-? was less than 100. Group II, 5 patients with ACL whose relative expression of IFN-? was greater than 100. Group III, 10 patients with mucosal leishmaniasis. The expression levels for IFN-? were 33,34; 214,1 and 129,6 times higher for Groups I, II and III, respectively than the samples from the normal control. For TNF-? were 11,22; 2,37 and 4,59 times higher than normal in Groups I, II and III, respectively. For IL-10 were 5,14; 11,54 and 2,51 times higher than the normal values in Groups I, II and III, respectively. For IL-27, 15,81; 85,29 and 17,29 times higher than normal in Groups I, II and III, respectively. For TGF-? were 5,38; 4,33 and 4,72 times higher than normal in Groups I, II and III, respectively. For IL-6 were 5,92; 5,69 and 2,09 times higher than the normal values in Groups I, II and III, respectively. These results were similar to the results of studies that analyze the expression of cytokines already synthesized. In general, the gene expression results of the cytokines studied suggest that the exacerbation of the Th1 profile (IFN-?) immune response could lead to more serious lesions in patients in Groups II and III. Elevated levels of IL-10 mRNA observed in Group II patients may promote low macrophage activation, assisting in the replication of the parasite and thus culminating in the progression of the disease. Expression of IL-27 mRNA did not interfere in the expression of IFN-? mRNA in the cells of patients with tegumentary leishmaiosis or mucocutaneous leishmaniasis. In conclusion, these results may suggest that patients in Group II may develop more severe forms of ACL when compared to those in Group I.

Parasitos do gênero Leishmania causam um espectro de doenças chamadas de leishmaniose. Para compreender de melhor forma a imunobiologia da Leishmania, o estudo de outras células envolvidas no processo de infecção do parasito se torna importante. As células B-1 são encontradas principalmente nas cavidades peritoneal e pleural, tendo como característica a capacidade de se diferenciar em células fagocíticas. Este trabalho teve como objetivo avaliar o papel das células B-1 na infecção por L. (L.) amazonensis. Os resultados mostraram que, assim como os macrófagos, os B-1CDPs são infectados pelo mecanismo de fagocitose e permitem a multiplicação da Leishmania no seu interior. Além disso, os vacúolos parasitóforos formados nos B-1CDPs apresentaram ser maiores do que dos macrófagos. Portanto, os dados com B1CDPs sugerem que estas células podem possuir um papel relevante na infecção por L. (L.) amazonensis, podendo ser considerados células hospedeiras importantes durante a infecção devido à incapacidade de responder de maneira eficaz para a eliminação dos parasitos. The genus Leishmania parasites cause a spectrum of diseases called leishmaniasis. To better understand the immunobiology of Leishmania, the study of other cells involved in parasite infection process becomes important. The B-1 cells are found predominantly in the peritoneal and pleural cavities, whose feature consist on an ability to differentiate into phagocytic cells. This study aimed to evaluate the role of B-1 cells in the infection by L. (L.) amazonensis. The results showed that, like macrophages, B-1CDPs are infected by a mechanism of phagocytosis and allow the multiplication of Leishmania within. Furthermore, the parasitophorous vacuoles in the B-1CDPs showed to be larger than those formed in the macrophages. Therefore, B1CDPs can have an important role in infection by L. (L.) amazonensis and can be considered important host cells during infection due to inability to respond effectively to the elimination of parasites.

A cromoblastomicose é uma micose subcutânea, com alto índice de morbidade, sendo Fonsecaea pedrosoi (F. pedrosoi) considerado o maior agente etiológico dessa micose, caracterizando uma doença crônica, geralmente confinada na pele e tecidos subcutâneos. Raramente os indivíduos apresentam cura dessa doença, pois as terapias contemporâneas mostram-se deficientes e poucos trabalhos relatam a relação parasito-hospedeiro. As células dendríticas (DCs) são especializadas na apresentação de antígenos para linfócitos T naive induzindo respostas imunes primárias. Diante disso, propomos estudar a capacidade migratória de DCs após infecção com conídios de F. pedrosoi, uma vez que o processo de migração dessas células está intimamente ligado com a sua função sobre as células T, levando ao desenvolvimento de uma resposta imune adaptativa protetora. O fenótipo de DCs foi avaliado através de células obtidas dos linfonodos poplíteos, inguinais e patas de camundongos BALB/c após 12, 24 e 72 horas de infecção com conídios do fungo. Células obtidas foram marcadas com anticorpos específicos e analisadas por citometria de fluxo. Após 24 e 72 horas de infecção verificamos uma diminuição significativa na porcentagem de DCs nas patas, e um aumento significativo dessas células nos linfonodos após 72 horas. A expressão de marcadores de superfície como CCR7 e moléculas co-estimulatórias, mostraram-se diminuídas nas células obtidas das patas. Como no processo de imunofenotipagem podemos analisar DCs vindas de diversos locais, para melhor avaliar a capacidade migratória das DCs, células das patas foram marcadas in vivo injetando-se subcutaneamente o corante CFSE juntamente com conídios do fungo. Verificamos que após 12 e 72 horas, as DCs das patas dos animais infectados, migraram para os linfonodos regionais. Assim constatamos que o fungo F. pedrosoi é capaz de induzir a migração de macrófagos e neutrófilos para o sítio de infecção em poucas horas, leva ao aumento de células B nos linfonodos após 12 e 72 horas de infecção, e também leva ao aumento de linfócitos T CD4+ tanto no local de infecção quanto nos linfonodos. Os resultados também comprovam que o fungo F. pedrosoi foi capaz de ativar as DCs, induzindo sua migração para os linfonodos regionais. The chromoblastomycosis is a subcutaneous mycosis with a high morbidity rate, Fonsecaea pedrosoi (F. Pedrosoi) being the largest etiologic agent of this mycosis, featuring a chronic disease, usually confined to the skin and subcutaneous tissues. Rarely do people have cure for this disease, because the therapies shown to be deficient contemporary and few studies report the host-parasite relationship. Dendritic cells (DCs) are specialized in presenting antigens to naïve T lymphocytes inducing primary immune responses. Therefore, we propose to study the migratory capacity of DCs after infection with conidia of F. pedrosoi, since the migration of these cells is intimately linked to its function on T cells, leading to development of a protective adaptive immune response. The phenotype of DCs was evaluated using cells obtained from popliteal lymph nodes, sub cutaneous tissue of BALB/c mice after 12, 24 and 72 hours of infection with conidia of the fungus. Cells were labeled with specific antibodies and analyzed by flow cytometry. After 24 and 72 hours of infection we found a significant decrease in the percentage of DCs in the sub cutaneous tissue, and a significant increase of these cells in the lymph nodes after 72 hours. The expression of surface markers such as CCR7 and costimulatory molecules, were reduced in cells obtained from the sub cutaneous tissue. Like the process of immunophenotyping we can analyze DCs coming from various locations, to better assess the migratory capacity of DCs, cells were stained paws in vivo by injecting dye subcutaneously with CFSE conidia of the fungus. We found that after 12 and 72 hours, DCs sub cutaneous tissue of infected animals migrated to regional lymph nodes. We found that the fungus F. pedrosoi is capable of inducing migration of macrophages and neutrophils to the site of infection within a few hours, leads to increased B-cells in lymph nodes after 12 and 72 hours of infection, and also leads to an increase of CD4 + T lymphocytes in both the site of infection and in the lymph nodes. The results also show that the fungus F. pedrosoi was able to activate the DCs, inducing their migration to regional lymph nodes.

A paracoccidioidomicose é uma enfermidade sistêmica causada principalmente pelo fungo Paracoccidioides brasiliensis. Recentemente, observou-se que a capacidade fagocítica dos macrófagos alveolares (MA) em animais diabéticos está diminuída em comparação aos MA de animais sadios, e que a insulina estimula a atividade fagocítica em MA oriundos de animais diabéticos e sadios por mecanismos diferentes. Neste projeto, usando um modelo de carência relativa de insulina (diabetes mellitus experimental), estudamos a intervenção da insulina em um modelo de infecção sistêmica. Após aprovação do comitê de ética (protocolo CEUA/FCF/421), camundongos machos da linhagem C57BL/6 (diabéticos, 60 mg/kg aloxana/10 dias, e seus respectivos controles) receberam injeção intratraqueal contendo suspensão de leveduras de P. brasiliensis ou volume equivalente de PBS estéril. Animais dos grupos de tratamento receberam insulina pela via subcutânea diariamente por 12 dias. Nas amostras, foram avaliados: a) o número de células dos lavados peritoneal (LPe) e broncoalveolar (LBA), o leucograma, e a glicemia (monitor de glicose); b) os níveis séricos de insulina no soro pela técnica de ELISA; c) as concentrações de citocinas (TNF-α, IL-6, IL-4, IL-10, IL-12, CINC-1, CINC-2, CINC-3) nos LPe e LBA e nos homogenatos (ELISA). Após incubação de 55 dias, comparados aos controles (2.9±0,4g e 192±7.5 mg/dL), animais tornados diabéticos (0.87 ± 0,25 g e 570,1 ± 9,27mg/dL) apresentaram redução no ganho de massa corpórea durante o período de experimentação e elevados níveis de glicose sanguínea. O tratamento de insulina reduziu os níveis de glicose (547±36,8mg/dL vs.323,6±36,9mg/dL), embora não o suficiente para tornar os animais normoglicêmicos. Comparados aos controles, animais diabéticos apresentaram número reduzido de leucócitos no LPe (2.2 x106 ± 0.2cells/mm3 vs 1.3 x106 ± 0.1cells/mm3) e, no LBA, reduzidas concentrações de CINC-2 (662,3±73,8pg/mL vs 312,7±114,7pg/mL), CINC-1 (115,5.0±25,5pg/mL vs 88,3±24,7pg/mL) e IL-10(320,9±58,4pg/mL vs 161,0±59,4pg/mL) depois da infecção. O tratamento com insulina restaurou a concentração de leucócitos nos LPe de animais diabéticos, mas não no LBA. Os dados sugerem que a insulina modula a produção/liberação de citocinas, sem alterar a migração de leucócitos para LBA e restaurando a migração destes para LPe durante o curso da paracoccidioidomicose. Paracoccidioidomycosis is a systemic disease mainly caused by Paracoccidioides brasiliensis fungus that interact with antigen-presenting cells, changing its main biological functions. Recently it was observed that the phagocytic capacity of these cells in diabetic animals for IgG opsonized targets is decreased compared to healthy animals, and that insulin stimulates the phagocytic activity in alveolar macrophages in from diabetic and healthy animals, by different mechanisms. In this project, using a model of relative lack of insulin (experimental diabetes mellitus), we studied the intervention of insulin in a model of systemic infection. After approval by the committee of ethics (protocol CEUA/FCF/421), C57BL/6 male diabetic (60mg/kg aloxan/10days) mice and their respective controls were subjected to intratracheal injection of suspension of P. brasiliensis or an equivalent volume of TBS sterile. In the forty-third day, insulin-treated mice were treated subcutaneously daily with insulin for 12 days at 6 P.M. We evaluated: a) the number of cells of the peritoneal(PeL) and bronchoalveolar (BAL) fluids, the leucogram and the glucose levels; b) the levels of insulin on the serum by the technique of ELISA; c) the levels of cytokines (TNF-α, IL-6, IL-4, IL-10, IL-12, CINC-1, CINC-2, CINC-3) in the BAL and PeL fluid, and on organ homogenates. After 55 days, relative to controls (2.9±0,4g and 192±7.5 mg/dL)), mice rendered diabetic (0.87 ± 0,25 g and 570,1 ± 9,27mg/dL) exhibited a reduction in body weight gain during the experimental period and sharply elevated blood glucose levels. Treatment of diabetic animals with insulin induced a reduction in blood glucose levels (547±36,8mg/dL vs.323,6±36,9mg/dL), but it was not sufficient to reduce glycemia to control values. In addition, relative to controls, infected diabetic mice exhibited a reduction in the number of leukocytes into the PeL fluid ((2.2 x106 ± 0.2cells/mm3 vs 1.3 x106 ± 0.1cells/mm3) and reduced BAL concentrations of CINC-2 (662,3±73,8pg/mL vs 312,7±114,7pg/mL), CINC-1 (115,5.0±25,5pg/mL vs 88,3±24,7pg/mL) and IL-10 (320,9±58,4pg/mL vs 161,0±59,4pg/mL) after P. brasiliensis infection. Treatment of diabetic mice with insulin restored concentrations of leukocytes in the PeL fluid but not in the BAL. Data presented suggest that insulin modulates the production/release of cytokines but not leukocyte migration to the BAL while restoring this paramether on the PeL during the course of P. brasiliensis fungus-induced PCM.

Paracoccidioidomicose (PCM) é uma micose sistêmica causada por Paracoccidioides brasiliensis (P. brasiliensis), que acomete preferencialmente o pulmão, sendo este um dos poucos órgãos que está em continua e intensa interação com o meio ambiente e o sistema imune. O grande desafio para o sistema imune pulmonar é discriminar o que é nocivo e reagir de acordo. As células dendríticas (DCs) são apresentadoras de antígenos \"profissionais\" capazes de fazer o processamento do antígeno e a apresentação de peptídeos a linfócitos T e são ideais para manter este equilíbrio delicado entre a tolerância e uma resposta imune efetora. Sabendo da importância dessas células no sistema imune e que a infecção por P. brasiliensis ataca preferencialmente o pulmão, células de lavado broncoalveolar (BAL), após infecção experimental com leveduras de P. Brasiliensis, foram analisadas. Nós observamos um significante aumento de DCs no BAL após 24hrs da infecção intratraqueal com 104 leveduras de P. brasiliensis. A caracterização das DCs mostraram que essas células expressaram CD11c, MHC-II e DEC205. A fim de verificar a capacidade de migração das DCs, as mesmas foram diferenciadas a partir de células de medula óssea e pulsadas com leveduras de P. brasiliensis, marcadas com CFSE e injetadas intratraquealmente em camundongos. Como controle negativo, utilizamos PBS e DCs não pulsadas com o fungo. Os resultados mostraram, que após 12 horas de infecção, as DCs (CFSE+CD11c+), migraram para os linfonodos torácicos. No entanto, a quantidade destas células diminui discretamente após 24 horas de infecção. Além disso, não observamos DCs nos linfonodos regionais, quando analisamos os grupos controle. Dessa forma, as células pulmonares também foram marcadas in vivo injetando-se intratraquealmente o corante CFSE juntamente com leveduras de P. brasiliensis. Verificamos que após 12 horas, as DCs pulmonares, dos animais infectados com o fungo, migraram para os linfonodos regionais. Esses resultados comprovam que o fungo P. brasiliensis foi capaz de ativar as DCs, induzindo sua migração para linfonodos regionais, e induziu nesse órgão uma resposta por células T evidenciada pela produção preferencial de IL-10. They are one of the few organs that are in constant and intense interaction with the environment and the immune system. The great challenge for the pulmonary immune system is to discriminate what is harmful and react accordingly. Dendritic cells (DCs) are \"professionals\" antigen-presenting cells and they can do the antigen processing. They can also do the presentation of peptides to T lymphocytes and are ideal for maintaining this delicate balance between tolerance and an effector immune response. Knowing the importance of these cells in the immune system and that the infection by P. brasiliensis preferentially attacks the lungs, cells from bronchoalveolar lavage (BAL), after experimental infection with yeasts of P. Brasiliensis, were analyzed. We observed a significant increase of DCs in BAL after 24 hours of intratracheal infection with 104 of yeast cells of P. brasiliensis. The characterization of these cells showed that DCs expressed CD11c, MHC-II and DEC205. In order to verify the ability of migration of DCs, they were differentiated from bone marrow cells and pulsed with yeasts of P. brasiliensis, they were also labeled with CFSE and injected intratracheally into mice. As a negative control, we used PBS and DCs that were not pulsed with the fungus. The results showed that after 12 hours of infection, the DCs (CFSE + CD11c +), migrated to the thoracic lymph nodes. However, the quantity of these cells decreases slightly after 24 hours of infection. Furthermore, we observed no DCs in regional lymph nodes when we analyzed the control groups. Thus, the lung cells were also labeled in vivo by injecting the CFSE dye intratracheally with yeasts of P. brasiliensis. We found that after 12 hours, pulmonary DCs of the animals infected with the fungus migrated to regional lymph nodes. These results indicate that the fungus P. brasiliensis was able to activate DCs, inducing their migration to regional lymph nodes, and inducing a response in that organ by T cells as evidenced by preferential production of IL-10.
