Preparation of Zymoseptoria tritici isolates for extraction of chromosomes via pulsed-field electrophoresis Pat Martinez 1990 (Bruce McDonald lab) (Simplified from Cooley, et al, Curr. Genet. 13:383-389.)

Presented here are the results from a field study concerning the measurement of O 3 and N 2 O produced by corona discharge from a grounded metal tube with a sharp point during thunderstorms. We observed as much as a tenfold increase in the 03 concentration as well as a four percent increase in the N 2 O concentration as a result of corona discharge. The concentrations of both O 3 and N 2 O returned to the original ambient levels after the thunderstorm ended.

An alternative method for the determination of mercury (Hg) in solid samples is presented in this article. Gaseous Hg is evolved when a solid sample is heated above 500°C in the presence of oxygen. The evolved Hg is collected on silver wool and is analyzed using cold vapor methods. We have found greater than 95% Hg recovery with various sediment and ash standards.

The formation of nitrous oxide (N 2 O) by point to plane corona discharge exhibits a polarity dependence with the negative point producing 4.2 ± 0.2 times more N 2 O than found during similar positive point discharges. These results were obtained under ambient temperature and pressure. For the discharge currents used, ±5, ±10, and ±15 μA , N 2 O formation was found to be linear with current for both the negative and positive point discharge. Consistent with previously reported values, we found that negative point produced 7.2 ±0.2 times more ozone (O 3 ) than positive point discharges, under the same conditions. We also found that with negative point discharge, only one N 2 O molecule was produced for every 90±10 O 3 molecules; one N 2 O molecule for every 50 ± 5 O 3 molecules during positive point discharge.

Nitrous acid was the only soluble species initially formed when the effluent of electrical discharges in air was passed through distilled water. The nitrous acid was about 5 % of the total amount of NO x (NO + NO 2 ) produced by these electrical discharges.
