The health effects of (poly)phenols (PPs) depend upon their bioavailability that, in general, is very low and shows a high interindividual variability. The low bioavailability of PPs is mainly attributed to their low absorption in the upper gastrointestinal tract as a result of their low water solubility, their presence in foods as polymers or in glycosylated forms, and their tight bond to food matrices. Although many studies have investigated how technological and biotechnological processes affect the phenolic composition of fruits and vegetables, limited information exists regarding their effects on PP bioavailability in humans. In the present review, the effect of food processing (mechanical, thermal, and non-thermal treatments), oral-delivery nanoformulations, enzymatic hydrolysis, fermentation, co-administration with probiotics, and generation of postbiotics in PP bioavailability have been overviewed, focusing in the evidence provided in humans. The presence of benzene, toluene, ethylbenzene and xylenes (BTEX) in dyes, paints and pigments is not still regulated but might be an indicator of contamination due to the well-known health risk of these compounds. An analytical procedure based on headspace sorptive extraction using a polydimethylsiloxane stir bar, was developed for the gas chromatographic determination of BTEX in finger paint samples. Univariate and multivariate designs were used to optimize experimental parameters, including equilibrium time, desorption time, saline effect and sample mass. Thirty minutes were necessary to absorb the analytes in the polydimethylsiloxane stir bar followed by a 5 min thermal desorption step. Limits of quantification of between 0.036 and 0.12 ng g−1 depending on the compound, were obtained. BTEX were found in the finger paint samples at levels between 0.14 and 12 ng g−1.

El principal objetivo de esta Tesis Doctoral ha sido el desarrollo de una plataforma analítica complementaria utilizando diferentes técnicas de tratamiento y preconcentración de la muestra y determinación analítica con diversas técnicas instrumentales para conseguir la identificación y cuantificación de contaminantes orgánicos en muestras de distinta naturaleza. Los compuestos orgánicos y las matrices estudiadas han sido seleccionadas para la resolución de retos científicos y tecnológicos que actualmente suscitan interés en la sociedad. Las técnicas de microextracción para el tratamiento de muestra tienen por objeto simplificar la primera etapa del proceso analítico, incrementando, al mismo tiempo, la sensibilidad de los procedimientos. La aplicación de técnicas miniaturizadas permite catalogar los métodos desarrollados como respetuosos con el medio ambiente, considerando el cumplimiento de los principios de la Química Analítica Verde. La separación de los compuestos se ha llevado a cabo utilizando técnicas cromatográficas, seleccionando en función de las características físico-químicas de los analitos, la cromatografía de gases (GC) o la cromatografía líquida (LC). Por otra parte, cuando se ha aplicado GC, se han utilizado sistemas de inyección clásicos o mediante una unidad de desorción térmica (TDU). La plataforma desarrollada propone la espectrometría de masas (MS) para la detección de los contaminantes, siendo un objetivo de gran importancia la elección de la fuente de ionización y el analizador más adecuados, así como las condiciones óptimas de trabajo que proporcionen la máxima sensibilidad y selectividad. Por tanto, se presentan distintas vías complementarias para la determinación de compuestos orgánicos a nivel de trazas, permitiendo la identificación inequívoca y la cuantificación a muy bajos niveles de concentración, para cumplir con los límites establecidos en la legislación vigente. La metodología de trabajo para el desarrollo de la presente Tesis Doctoral incluye por un lado la optimización de las etapas de preparación de muestra, separación cromatográfica y detección. Por otra parte, cada método optimizado se ha validado de acuerdo con las guías internacionales, obteniendo las distintas características analíticas de calibración, así como los parámetros de precisión y exactitud. Finalmente, los métodos analíticos son aplicados al análisis de muestras reales. El trabajo presentado se ha estructurado de acuerdo a la técnica miniaturizada aplicada en el tratamiento de la muestra. Así, las primeras secciones se centran en la aplicación de la microextracción dispersiva líquido-líquido (DLLME), que ha proporcionado muy buenos resultados en cuando a rapidez, sensibilidad y selectividad, permitiendo obtener factores de preconcentración muy elevados con un consumo muy bajo de disolventes orgánicos y de muestra. La extracción por adsorción sobre barras agitadoras (SBSE) permite trabajar en ausencia de disolventes orgánicos, permitiendo la transferencia cuantitativa de los analitos preconcentrados hasta el sistema cromatográfico mediante la unidad de desorción térmica. Se ha utilizado también la extracción en fase sólida magnética (MSPE), que permite la adsorción de los analitos sobre nanopartículas magnéticas funcionalizadas, resaltando su fácil aislamiento utilizando un campo magnético externo. Finalmente, la técnica de extracción en fase sólida dispersiva (DSPE) se aplicó con objeto de simplificar la matriz de las muestras de los detergentes, cuya complejidad no permite el análisis directo. Así, se han optimizado y validado, siguiendo las guías internacionales, procedimientos analíticos para la determinación de ftalatos, alquilfenoles y éteres glicólicos como contaminantes en productos de limpieza doméstica; pesticidas de última generación, insecticidas (piretroides, nicotinoides) y fungicidas (triazoles, oxazoles y estrobilurinas) en muestras agrícolas como aguas, suelos, frutas y verduras; bisfenoles, parabenos y compuestos dicarbonílicos (glioxal y metilglioxal) en orina; hidrocarburos aromáticos policíclicos en tejidos humanos procedentes de autopsias; alquilfenoles como contaminantes en alimentos (zumos y alimentos infantiles) y compuestos orgánicos volátiles como los BTEX en pinturas. The main objective of this Doctoral Thesis was the development of a complementary analytical platform using different techniques of sample treatment and preconcentration and analytical determination with different separation techniques to achieve the identification and quantification of organic contaminants in different matrices. All the developed methods are applied to analytes and samples whose nature concerns society and which may be regarded as scientific and technological challenges that can be solved. The microextraction techniques simplify the first stage of the analytical process, the sample treatment, meanwhile increasing the sensitivity of the procedures. The application of miniaturized techniques allows to identify the developed methods as environmentally friendly and comply with the principles of Green Analytical Chemistry. The separation of the compounds was carried out using chromatographic techniques, selecting liquid chromatography (LC) or gas chromatography (GC) depending on physical-chemical characteristics of the analytes. Classic injection systems or thermal desorption (TD) injection were used when GC was selected. The developed platform proposes mass spectrometry (MS) for the detection of pollutants, where the most important parameters are the choice of the most suitable ionization source and analyzer, as well as the optimal work conditions that provide maximum sensitivity and selectivity. Therefore, different complementary ways are presented for the determination of organic compounds at trace levels, allowing correctly identification and quantification at very low concentration levels, in order to comply with the limits established in the current European Legislation. The methodology applied in the development of this Doctoral Thesis includes, on the one hand, the optimization of the sample preparation, chromatographic separation and detection stages; and, on the other hand, each optimized method has been validated according to the international guidelines, obtaining the analytical characteristics of the method such as linearity, precision and accuracy parameters. Finally, the analytical methods are applied to the analysis of real samples. The presented Doctoral Thesis was structured according to miniaturized technique applied in the sample treatment. Thus, the first sections focus on the application of dispersive liquid-liquid microextraction (DLLME), which provided the best results in terms of speed, sensitivity and selectivity, obtaining very high preconcentration factors with a very low consumption of organic and sample solvents. The extraction by stir bar sorptive extraction (SBSE) works in the absence of organic solvents, allowing the quantitative transfer of the preconcentrated analytes into the chromatographic system using a thermal desorption unit. Magnetic solid phase extraction (MSPE) was also used, which allows the adsorption of analytes on functionalized magnetic nanoparticles. One of its advantage is the easy isolation using an external magnetic field. Finally, solid phase dispersive extraction (DSPE) technique was applied in order to simplify the matrix of cleaning products, whose complexity does not allow direct analysis. Thus, analytical procedures have been optimized and validated, following the international guides, for the determination of phthalates, alkylphenols and glycol ethers in household cleaning products, new generation pesticides, insecticides (pyrethroids, nicotinoids) and fungicides (triazols, oxazols and strobilurins) in agricultural samples such as water, soils, fruits and vegetables; bisphenols, parabens and α-dicarbonilic compounds (glyoxal and methylglyoxal) in urine; polycyclic aromatic hydrocarbons in human tissues from autopsies; alkylphenols as food contaminants (fruit juices and infant formulas) and volatile organic compounds (BTEX) in finger paints.
