Abstract

The greater amberjack, Seriola dumerili (Risso, 1810), is a promising candidate for the diversification of European aquaculture production, but inconsistent reproduction in captivity prevents commercial production. Recent studies showed that greater amberjack confined in sea cages exhibited scarce gonad development and early interruption of gametogenic activity during the reproductive season. The aim of the present study was to improve our understanding of the observed impairment of spermatogenesis. Adult wild and captive-reared males were sampled during 3 different phases of the reproductive cycle: early gametogenesis (EARLY; late April to early May), advanced gametogenesis (ADVANCED; late May to early June), and spawning (SPAWNING; late June to July). Spermatogonial stem cells and proliferating germ cells were identified through the immunohistochemical localization of Pou5f1 and proliferating cell nuclear antigen, respectively. Apoptotic germ cells were identified throughout the terminal deoxynucleotidyl transferase-mediated 2’-deoxyuridine 5’-triphosphate nick end labeling method. Sperm quality of captive-reared fish was evaluated using computer-assisted sperm analysis. Captive-reared males exhibited seminiferous lobules of a smaller diameter, a precocious and progressive decrease of spermatogonial mitosis, and a high level of apoptosis at the beginning of the reproductive season, concomitant with a many-fold higher 17β-estradiol plasma concentration. The motile spermatozoa percentage of captive greater amberjack was lower than in other teleosts, and a drastic decrease of spermatozoa motility duration, velocity, and ATP content occurred along the reproductive season. An abnormal increase of sperm concentration as well as an increase of dead spermatozoa occurred during the SPAWNING phase, probably because of lack of sperm hydration and ejaculation and consequent sperm ageing. The present study demonstrates the extreme susceptibility of greater amberjack to rearing stress and underscores the need for improvement of the rearing and handling procedures to ameliorate gametogenesis dysfunctions in commercial aquaculture production.

Abstract

Gli AA. effettuano un’analisi dello scenario relativo alla produzione bio a livello mondiale, europeo e nazionale. In particolare indagano se in Italia l’evoluzione del numero degli operatori del settore risulta in linea con la ritrovata richiesta dei consumatori di poter disporre di un numero crescente di detti prodotti. Tale analisi è stata ulteriormente approfondita al fine di cogliere le eventuali diversità di diffusione del biologico sul territorio italiano, distinto nelle tre macro-aree geografiche Nord, Centro e Sud negli 2000-2007. Per analizzare tali fenomeni sono stati presi in esame le serie storiche dei dati ufficiali disponibili, aggiornati al 2008, provenienti dal Ministero delle Politiche Agricole, Alimentari e Forestali (Mipaaf) e dal Sistema d’informazione nazionale sull’agricoltura biologica (SINAB). L’analisi dei dati ha consentito di rilevare come alla crescita degli Operatori Bio abbia contribuito il Sud più delle restanti due macroaree in cui il Paese può essere suddiviso. Il contributo medio dato alla crescita degli operatori è stato, infatti, del 60% per il Sud, del 17% per il Centro e del 23% per il Nord. Tali dati dimostrano la particolare attenzione accordata al settore del biologico al Sud, costituendo, pertanto, punto di forza per lo sviluppo economico di un’area geografica storicamente in ritardo rispetto al resto della nazione

Abstract

The reproductive strategy of an unexploited population of Arca noae from the salt-water Bizerte Lagoon (Tunisia, western Mediterranean), including its tissue plasticity, was studied. In total 309 individuals, collected monthly from October 2013 to September 2014, were examined; 142 were females, 42 were males and five were hermaphrodites. They were used in histological and immunohistochemical (stem marker: Pou5f1; proliferation marker: proliferating cell nuclear antigen (PCNA)) analyses of gonads and adjacent tissues (N = 189) as well as to compute the monthly condition index (N = 120). Water salinity, temperature and chlorophyll a concentration were recorded. Ripe ovaries were observed in two discrete periods, October–November and April–August. Both gonad ripenings were followed by spawning periods, November–April and July–September. The mature oocyte density showed that the first spawning period was less vigorous than the second one. These data also indicated that A. noae is a multiple spawner. Five cases of protandric hermaphroditism occurred from December to April. Gonad tissue was strictly associated and intermingled with the digestive gland and mantle muscle fibres. Seasonal variations were observed in the relative proportions of digestive gland and gonads: the former predominated when the latter regressed (March) and vice versa (peak in June). Seasonal transitions from germinal to somatic tissue and vice versa were hypothesised to occur through transdifferentiation mechanisms based on the activity of stem and proliferating cells. The condition index roughly increased along with gonad ripening and decreased during the spawning periods, although it did not run parallel to gonad evolution, because it also depended on chlorophyll a concentration, a proxy for phytoplankton density. The condition index was significantly correlated, by multiple regression, to both mature oocyte density and chlorophyll a concentration. Arca noae appears to have evolved a flexible reproductive strategy that makes it capable of exploiting diverse environmental conditions, which also involves tissue transdifferentiation. Abstract

Melanomacrophage centres ( MMCs), located in different organs of non-mammalian vertebrates, play a role in the destruction, detoxification or recycling of endogenous and exogenous materials. Cytochrome P450 monoxygenase 1 A ( CYP1A) is involved in xenobiotics biotransformation, and its liver expression is considered as a biomarker for detecting exposure to environmental pollutants. Atlantic bluefin tuna ( ABFT), Thunnus thynnus L., liver samples were collected from: wild animals caught in the eastern Atlantic; juveniles reared in the central Adriatic; juveniles reared in the northern Adriatic; adults reared in the western Mediterranean. The samples were processed for basic histology, histochemistry and for CYP1 A immunodetection. An unexpected high density of MMCs, containing ferric iron and lipofuscin-ceroids, was detected in the juveniles sampled in the northern Adriatic Sea. These individuals showed also a strong anti- CYP1 A immunopositivity in hepatocytes and in the epithelium of bile ducts. This study supports the utility of MMCs as biomarkers of fish 'health status' and gives concern for a potential contaminant accumulation in ABFT. [ABSTRACT FROM AUTHOR]
