Introdução e Objetivo: O consumo excessivo de álcool continua como uma das principais causas de doença hepática em todo o mundo. Os microRNAs são pequenos RNAs não codificantes que atuam no nível pós-transcricional para regular a expressão de seus respectivos RNAs mensageiros. Na atualidade, estão sendo utilizados para diagnóstico e prognóstico, bem como para futuras terapias gênicas de diversas doenças, inclusive doenças hepáticas. Entre os modelos para os estudos de doenças hepáticas, o zebrafish vem se destacando por possuir muitas vantagens. O objetivo deste estudo foi avaliar a expressão hepática e sérica de miR-122, miR-155 e miR-217 em um modelo de exposição crônica ao etanol em zebrafish adulto. Métodos: Zebrafish adultos de ambos os sexos, foram divididos em dois grupos (n=281): Grupo Etanol (GE) exposto a 0,5% v/v na água do aquário e Grupo controle (GC), sem exposição ao etanol. Após 28 dias os animais foram eutanasiados. No tecido hepático foram realizadas análises histopatológicas, quantificação de lipídios e triglicerídeos e a avaliação das citocinas inflamatórias il-1β, il-10 e tnf-α. A expressão de miR-122, miR-155 e miR-217 foi quantificada no tecido hepático e no soro. Resultados: Após os 28 dias de exposição ao etanol, as análises histopatológicas mostraram lesão hepática e deslocamento dos núcleos dos hepatócitos no grupo GE, confirmada pela quantificação de acúmulos de lipídios A expressão gênica de il-1β mostrou aumento no GE, entretanto il-10 e tnf-α não apresentaram diferenças entre os grupos. A análise da expressão hepática de miR-122 e miR-155 foram aumentadas no GE, porém miR-217 hepático não apresentou diferença entre os grupos. A análise de expressão circulante de miR-155 e miR-217 apresentou aumento significativo no GE, porém miR-122 circulante não apresentou diferença entre os grupos. Conclusão: A exposição crônica ao etanol em zebrafish adulto promove dano hepático, acúmulo de lipídios nos 6 hepatócitos e variação nas citocinas inflamatórias do fígado. A expressão dos microRNAs circulantes e hepáticos é aumentada pela indução com etanol. Estes achados sugerem que os microRNAs juntamente com este modelo é adequado para estudos de mecanismos, fisiopatogenia e até terapêutica da DHA. The aim of this study was to evaluate the hepatic and serum expression of miR-122, miR-155 and miR-217 in a model of chronic exposure to ethanol in adult zebrafish. Methods: Adults zebrafish wild type of both sexes were divided into two groups (n = 281): Ethanol Group (EG), exposed to 0.5% v/v in the aquarium water and Control Group (CG), without exposure to ethanol. Expression of miR-122, miR-155 and miR-217 was quantified in liver tissue and serum. Results: After 28 days of exposure to ethanol, histopathological analysis showed hepatic lesion and displacement of hepatocyte nuclei in the EG, confirmed by the quantification of lipid accumulations. The gene expression of il-1β showed increase in EG, but il-10 and tnf-α showed no differences between groups. Analysis of hepatic expression of miR-122 and miR-155 was increased in EG, but hepatic miR-217 showed no difference between groups. The analysis of circulating expression of miR-155 and miR-217 presented a significant increase in the EG, but miR-122 circulating did not present difference between the groups. Conclusion: Chronic ethanol exposure in adult zebrafish produces hepatic damage, lipids accumulation in hepatocytes and variation in the inflammatory cytokines of the liver. The expression of circulating and hepatic 8 microRNAs is altered by induction with ethanol. These results indicate that miRNA, as well as the adult model of zebrafish, are potential tools for the study of mechanisms, pathophysiology and even ALD therapy.and possible therapeutic targets.
