We demonstrate a systematic method for solving the Hamilton-Jacobi equation for general relativity with the inclusion of matter fields. The generating functional is expanded in a series of spatial gradients. Each term is manifestly invariant under reparameterizations of the spatial coordinates (``gauge-invariant''). At each order we solve the Hamiltonian constraint using a conformal transformation of the 3-metric as well as a line integral in superspace. This gives a recursion relation for the generating functional which then may be solved to arbitrary order simply by functionally differentiating previous orders. - Abstract
During the onset of the COVID-19 pandemic, the COVIDiSTRESS Consortium launched an open-access global survey to understand and improve individuals’ experiences related to the crisis. A year later, we extended this line of research by launching a new survey to address the dynamic landscape of the pandemic. This survey was released with the goal of addressing diversity, equity, and inclusion by working with over 150 researchers across the globe who collected data in 48 languages and dialects across 137 countries. The resulting cleaned dataset described here includes 15,740 of over 20,000 responses. The dataset allows cross-cultural study of psychological wellbeing and behaviours a year into the pandemic. It includes measures of stress, resilience, vaccine attitudes, trust in government and scientists, compliance, and information acquisition and misperceptions regarding COVID-19. Open-access raw and cleaned datasets with computed scores are available. Just as our initial COVIDiSTRESS dataset has facilitated government policy decisions regarding health crises, this dataset can be used by researchers and policy makers to inform research, decisions, and policy.

The mutational landscape is shaped by many processes. Genic regions are vulnerable to mutation but are preferentially protected by transcription-coupled repair1. In microorganisms, transcription has been demonstrated to be mutagenic2,3; however, the impact of transcription-associated mutagenesis remains to be established in higher eukaryotes4. Here we show that ID4—a cancer insertion–deletion (indel) mutation signature of unknown aetiology5 characterized by short (2 to 5 base pair) deletions —is due to a transcription-associated mutagenesis process. We demonstrate that defective ribonucleotide excision repair in mammals is associated with the ID4 signature, with mutations occurring at a TNT sequence motif, implicating topoisomerase 1 (TOP1) activity at sites of genome-embedded ribonucleotides as a mechanistic basis. Such TOP1-mediated deletions occur somatically in cancer, and the ID-TOP1 signature is also found in physiological settings, contributing to genic de novo indel mutations in the germline. Thus, although topoisomerases protect against genome instability by relieving topological stress6, their activity may also be an important source of mutations in the human genome. We thank S. A 6-month baseline period was used to assess patient characteristics. Patients with visits to an urologist, without visits to an oncologist, were assigned to the urology cohort, and patients with visits to an oncologist, with or without visits to an urologist, were assigned to the oncology cohort. Treatment patterns, health care resource utilization, and costs during a variable follow-up period were compared between cohorts using descriptive statistics and Lin's regression.Results: The urology cohort had fewer comorbid illnesses (P < 0.001) and patients were less likely to have other cancers during baseline (P < 0.001) or to die during follow-up (P = 0.004) compared with the oncology cohort. The oncology cohort patients were significantly more likely to have a claim for hormones (74.5% vs 61.1%; P < 0.001), chemotherapy (46.9% vs 10.2%, P < 0.001), and radiation (22.3% vs 3.7%, P < 0.0001) over follow-up. Mean unadjusted health care costs were higher in the oncology vs the urology cohort (US$31,896 vs US$15,318, respectively; P < 0.001). At 6 years follow-up, cumulative adjusted CRPC-specific costs were significantly higher among patients treated by oncologists with chemotherapy than among patients treated by urologists.Conclusion: CRPC patients treated by oncologists had greater use of hormones, chemotherapy, and radiation; higher percentages of patients with inpatient stays, emergency room, and ambulatory visits; and higher health care costs, than patients treated by urologists.Keywords: prostatic neoplasms, castration, treatment protocols, oncology, urology

- Abstract
During the onset of the COVID-19 pandemic, the COVIDiSTRESS Consortium launched an open-access global survey to understand and improve individuals’ experiences related to the crisis. A year later, we extended this line of research by launching a new survey to address the dynamic landscape of the pandemic. This survey was released with the goal of addressing diversity, equity, and inclusion by working with over 150 researchers across the globe who collected data in 48 languages and dialects across 137 countries. The resulting cleaned dataset described here includes 15,740 of over 20,000 responses. The dataset allows cross-cultural study of psychological wellbeing and behaviours a year into the pandemic. It includes measures of stress, resilience, vaccine attitudes, trust in government and scientists, compliance, and information acquisition and misperceptions regarding COVID-19. Open-access raw and cleaned datasets with computed scores are available. Just as our initial COVIDiSTRESS dataset has facilitated government policy decisions regarding health crises, this dataset can be used by researchers and policy makers to inform research, decisions, and policy. © 2022, The Author(s).

- Abstract
Formulation is central to clinical practice. Formulation has a factor weighing, pattern recognition and explanatory hypothesis modelling focus. Formulation attempts to make sense of why a person presents in a certain state at a certain time and context, and how that state may be best managed to enhance mental health, safety and optimal change. Inherent to the clinical need for formulation is an appreciation of the complexities, uncertainty and limits of applying theoretical concepts and symptom, diagnostic and risk categories to human experience; or attaching meaning or weight to any particular factor in an individual's history or mental state without considering the broader biopsychosocial and cultural context. With specific reference to suicide prevention, this paper considers the need and potential for the computer linguistic community to be both cognisant of and ethically contribute to the clinical formulation process.

29. Emotional stress classification using spiking neural networks

- Abstract
Objective: This study examined the data modeling capability of spiking neural networks (SNN) in classifying stressed versus relaxed brain states using electroencephalogram (EEG) data. The input spatiotemporal dynamics were explored to obtain further knowledge regarding the two-brain states. Method: A publicly available EEG data set for emotion analysis using psychological signals (DEAP) collected from 32 participants (50% females) with an average age of 26.9 is used in this study. Firstly, data extraction is performed using a criterion that defines stress and relaxation states using self-reported valence and arousal scores. Two hundred eight such extracted samples were selected to train and evaluate a novel three-layer feedforward SNN. This SNN consisted of leaky-integrate and fire neurons and learned from incoming data using spike-time-dependent plasticity (STDP) and dynamically evolving SNN algorithms. The SNN performance was evaluated using both fivefold cross-validation and a 60:40 training testing split. To explore input spatiotemporal dynamics, a specialized SNN architecture for brain data processing named NeuCube was used. Results: The highest-performing model of the novel SNN algorithm produced 88% average accuracy (F1 score: 86.21%, Matthews correlation coefficient: 0.78). This SNN outperformed traditional machine learning (ML) techniques without the use of manual feature extraction. Moreover, the input dynamics revealed higher prefrontal activation during relaxation states compared to stress states. Conclusions: The results showed the capability of the SNN algorithm to recognize stressed and relaxed states of the brain, using temporal learning techniques. Furthermore, the findings obtained from NeuCube suggested a potential approach for brain data analysis, setting SNNs apart from black box approaches used for brain data processing.
