Abstract

The first generation of ELT instruments includes an optical-infrared high-resolution spectrograph, indicated as ELT-HIRES and recently christened ANDES (ArmazoNes high Dispersion Echelle Spectrograph). ANDES consists of three fibre-fed spectrographs ([U]BV, RIZ, YJH) providing a spectral resolution of $\sim$100,000 with a minimum simultaneous wavelength coverage of 0.4-1.8 $\mu$m with the goal of extending it to 0.35-2.4 $\mu$m with the addition of a U arm to the BV spectrograph and a separate K band spectrograph. It operates both in seeing- and diffraction-limited conditions and the fibre feeding allows several, interchangeable observing modes including a single conjugated adaptive optics module and a small diffraction-limited integral field unit in the NIR. Modularity and fibre-feeding allow ANDES to be placed partly on the ELT Nasmyth platform and partly in the Coud\'e room. ANDES has a wide range of groundbreaking science cases spanning nearly all areas of research in astrophysics and even fundamental physics. Among the top science cases, there are the detection of biosignatures from exoplanet atmospheres, finding the fingerprints of the first generation of stars, tests on the stability of Nature's fundamental couplings, and the direct detection of the cosmic acceleration. The ANDES project is carried forward by a large international consortium, composed of 35 Institutes from 13 countries, forming a team of almost 300 scientists and engineers which include the majority of the scientific and technical expertise in the field that can be found in ESO member states., Comment: SPIE astronomical telescope and instrumentation 2024, in press

Abstract

The first generation of E-ELT instruments will include an optical-infrared High Resolution Spectrograph, conventionally indicated as EELT-HIRES, which will be capable of providing unique breakthroughs in the fields of exoplanets, star and planet formation, physics and evolution of stars and galaxies, cosmology and fundamental physics. A 2-year long phase A study for EELT-HIRES has just started and will be performed by a consortium composed of institutes and organisations from Brazil, Chile, Denmark, France, Germany, Italy, Poland, Portugal, Spain, Sweden, Switzerland and United Kingdom. Abstract

PLATO 2.0 has recently been selected for ESA's M3 launch opportunity (2022/24). Providing accurate key planet parameters (radius, mass, density and age) in statistical numbers, it addresses fundamental questions such as: How do planetary systems form and evolve? Are there other systems with planets like ours, including potentially habitable planets? The PLATO 2.0 instrument consists of 34 small aperture telescopes (32 with 25 sec readout cadence and 2 with 2.5 sec candence) providing a wide field-of-view (2232 deg2) and a large photometric magnitude range (4-16 mag). It focusses on bright (4-11 mag) stars in wide fields to detect and characterize planets down to Earth-size by photometric transits, whose masses can then be determined by ground-based radial-velocity follow-up measurements. Asteroseismology will be performed for these bright stars to obtain highly accurate stellar parameters, including masses and ages. The combination of bright targets and asteroseismology results in high accuracy for the bulk planet parameters: 2%, 4-10% and 10% for planet radii, masses and ages, respectively. The planned baseline observing strategy includes two long pointings (2-3 years) to detect and bulk characterize planets reaching into the habitable zone (HZ) of solar-like stars and an additional step-and-stare phase to cover in total about 50% of the sky. PLATO 2.0 will observe up to 1,000,000 stars and detect and characterize hundreds of small planets, and thousands of planets in the Neptune to gas giant regime out to the HZ. It will therefore provide the first large-scale catalogue of bulk characterized planets with accurate radii, masses, mean densities and ages. This catalogue will include terrestrial planets at intermediate orbital distances, where surface temperatures are moderate. R.; Zechmeister, M.; Zimara, J., The first generation of ELT instruments includes an optical-infrared high resolution spectrograph, indicated as ELT-HIRES and recently christened ANDES (ArmazoNes high Dispersion Echelle Spectrograph). ANDES consists of three fibre-fed spectrographs (UBV, RIZ, YJH) providing a spectral resolution of similar to 100,000 with a minimum simultaneous wavelength coverage of 0.4-1.8 mu m with the goal of extending it to 0.35-2.4 mu m with the addition of a K band spectrograph. It operates both in seeing- and diffraction-limited conditions and the fibre-feeding allows several, interchangeable observing modes including a single conjugated adaptive optics module and a small diffraction-limited integral field unit in the NIR. Its modularity will ensure that ANDES can be placed entirely on the ELT Nasmyth platform, if enough mass and volume is available, or partly in the Coude room. ANDES has a wide range of groundbreaking science cases spanning nearly all areas of research in astrophysics and even fundamental physics. Among the top science cases there are the detection of biosignatures from exoplanet atmospheres, finding the fingerprints of the first generation of stars, tests on the stability of Nature's fundamental couplings, and the direct detection of the cosmic acceleration. The ANDES project is carried forward by a large international consortium, composed of 35 Institutes from 13 countries, forming a team of more than 200 scientists and engineers which represent the majority of the scientific and technical expertise in the field among ESO member states., The Italian effort for ANDES is supported by the Italian National Institute for Astrophysics (INAF). Abstract

International audience; HIRES will be the high-resolution spectrograph at optical and near-infrared (NIR) wavelengths for ESO's Extremely Large Telescope (ELT). It will consist of three fibre-fed spectrographs providing a wavelength coverage of 0.4–1.8 µm (with a goal of 0.35–1.8 µm) at a spectral resolution of ~ 100 000. Fibre-feeding allows HIRES to have several interchangeable observing modes, including a single-conjugate adaptive optics (SCAO) module and a small diffraction-limited integral field unit in the NIR. It will therefore be able to operate in both seeing- and diffraction-limited modes. HIRES will address a wide range of science cases spanning nearly all areas of research in astrophysics and even fundamental physics. Some of the top science cases will be the detection of biosignatures from exoplanet atmospheres, finding the fingerprints of the first generation of stars (Pop III), tests on the stability of Nature's fundamental couplings, and the direct detection of the cosmic acceleration. The HIRES consortium is composed of more than 30 institutes from 14 countries, forming a team of more than 200 scientists and engineers.

14. The Icebreaker Mission to Search for Life on Mars

Abstract

The search for evidence of life on Mars is the ultimate motivation for its scientific exploration. The results from the Phoenix mission indicate that the high N. latitude ice-rich regolith at low elevations is likely to be a recently habitable place on Mars [Stoker et al., 2010]. The near-surface ice likely provided adequate water activity during periods of high obliquity, 3 to 10 Myr ago. Carbon dioxide and nitrogen are present in the atmosphere, and nitrates may be present in the soil. Together with iron in basaltic rocks and perchlorate in the soil they provide carbon and energy sources, and oxidative power to drive metabolism. Furthermore, the presence of organics is possible, as thermally reactive perchlorate would have prevented their detection by Viking and Phoenix. The Mars Icebreaker Life mission [McKay et al., 2013] focuses on the following science goals: (1) Search for biomolecular evidence of life; (2) Search for organic matter from either exogeneous or endogeneous sources using methods that are not effected by the presence of perchlorate; (3) Characterize oxidative species that produced reactivity of soils seen by Viking; and 4) Assess the habitability of the ice bearing soils. The Icebreaker Life payload (Figure 1) includes a 1-m rotary percussive drill that brings cuttings samples to the surface where they are delivered to three instruments (Fig. 1), the Signs of Life Detector (SOLID) [Parro et al., 2011] for biomolecular analysis, Laser Desorption Mass Spectrometer (LDMS) [??? 2015]) for broad spectrum organic analysis, and Wet Chemistry Laboratory (WCL) [Hecht et al., 2009] for detecting soluble species of nutrients and reactive oxidants. The Icebreaker payload fits on the Phoenix spacecraft and can land at the well-characterized Phoe-nix landing site in 2020 in a Discovery-class mission.

Abstract

HIRES will be the high-resolution spectrograph at optical and near-infrared (NIR) wavelengths for ESO’s Extremely Large Telescope (ELT). It will consist of three fibre-fed spectrographs providing a wavelength coverage of 0.4–1.8 µm (with a goal of 0.35–1.8 µm) at a spectral resolution of ~ 100 000. Fibre-feeding allows HIRES to have several interchangeable observing modes, including a single-conjugate adaptive optics (SCAO) module and a small diffraction-limited integral field unit in the NIR. It will therefore be able to operate in both seeing- and diffraction-limited modes. HIRES will address a wide range of science cases spanning nearly all areas of research in astrophysics and even fundamental physics. Some of the top science cases will be the detection of biosignatures from exoplanet atmospheres, finding the fingerprints of the first generation of stars (Pop III), tests on the stability of Nature’s fundamental couplings, and the direct detection of the cosmic acceleration. The case for life on Mars grows stronger. Investigations at Gale Crater by Curiosity have revealed fine-grained sedimentary rocks inferred to represent an ancient lake environment suited to support life. In addition, Curiosity tentatively found a heterogeneous distribution of organic carbon within these sediments, consistent with the detection of native organic C in Mars meteorites. Furthermore, modern potentially habitable environments have been recognized on Mars including the N. Polar region visited by Phoenix, gully features suggesting modern water flows, and RSLs that occur seasonally suggest liquid processes. The time is ripe for missions to Mars incorporating a search for biochemical evidence of life.

Abstract

Among the potential martian molecular targets are those supplied by meteoritic carbonaceous chondrites such as amino acids and polyaromatic hydrocarbons (PAHs), or true biomarkers by any hypothetical Martian biota (aromatic amino acids, steroid and triterpene molecules, peptides). Heat extraction and pyrolysis based methods currently used in planetary exploration are highly aggressive and very often modify the target molecules making a cumbersome task their identification. We have developed and validated a mild, non-destructive, multiplex competitive/inhibitory microarray immunoassay and its implementation in the SOLID (Signs of Life Detector) instrument for simultaneous detection of several organic molecules relevant for Mars exploration and environmental monitoring. A multiplex inhibitory immunoassay with a set of highly specific and sensitive antibodies capable to distinguish between D and L aromatic amino acids (Phe, Tyr, Trp), benzo[a]pyrene (BAP), pentachlorophenol or sulfone-containing aromatics, was validated in SOLID instrument for the analysis of kerogen samples as analogues of refractory organic material in carbonaceous chondrites or even Mars organics. Most of the antibodies exhibited sensitivities at 1-10 ppb level and some of them even at ppt. The multiplex immunoassay allowed the detection of BAP as well as aromatic sulfones in a water/methanol extract of an early cretaceous sample (c.a. 100 My) enriched in type IV kerogen. No L/D aromatic amino acids were detected, reflecting the high maturity and the absence of chemical groups. The results demonstrated once more the feasibility of multiplex inhibitory immunoassays and its potential use for in situ analytical instruments in planetary exploration and environmental monitoring.

Abstract

Polar ecosystems are areas very vulnerable to both natural or anthropogenic forcings. Knowledge of the microbial assemblage inhabiting these ecosystems can improve current understanding of their functioning and response to environmental changes. Within the SPONGEPOP project funded by the 7th European Framework INTERACT programme, two seasonal surveys (May and July 2014) were performed at 9 different stations along the Pasvik river (Fennoscandia). An antibody microarray approach using Life Detector Chips (LDC) was applied to evaluate the benthic microbial community, its metabolic profiles and seasonal variations. Three different microchips were used, specific for the whole community, psychrophylic or cyanobacterial populations. LDC allowed a quick characterization of the benthic microbial community, revealing its great physiological versatility. Seasonal differences were found particularly at inner as well as outer stations. Whole community chip showed a microflora particularly active during the warmer months, as shown by the highest number of positive reactions. In May, high activity was found at the innermost stations, while in July no clear spatial trend was observed. Proteins were the most frequently metabolized compounds, followed by nucleotides and small molecules. High Shannon diversity indices (H) were found for the whole community at the innermost stations in May, while in July higher values at the outermost ones were reached. Psychrophiles reacted with high intensity with a great number of compounds at the outer stations during May; in this period, high H values characterized these microorganisms. In July, when temperature increased, their responses were sensibly reduced. Cyanobacteria showed a few number of positive responses, mostly at the inner stations of the river in May. In summer, only spots of reactivity against Microcystis novacekii and M. aeruginosa were detected.

Abstract

In this study, we report the detection and characterization of the organic minerals weddellite (CaC2O4 · 2H2O) and whewellite (CaC2O4 · H2O) in the hyperarid, Mars-like conditions of the Salar Grande, Atacama desert, Chile. Weddellite and whewellite are commonly of biological origin on Earth and have great potential for preserving records of carbon geochemistry and possible biological activity on Mars if they are present there. Weddellite and whewellite have been found as secondary minerals occurring inside the lower detrital unit that fills the Salar Grande basin. The extremely low solubility of most oxalate minerals inhibits detection of oxalate by ion chromatography (IC). Crystalline oxalates, including weddellite and whewellite, were detected by X-ray diffraction (XRD). The association of weddellite with surface biota and its presence among subsurface detrital materials suggest the potential of a biological origin for Salar Grande weddellite and whewellite. In this regard, biological activity is uniquely capable of concentrating oxalates at levels detectable by XRD. The complementary detection of oxalate-bearing phases through IC in the upper halite-rich unit suggests the presence of a soluble oxalate phase in the basin that is not detected by XRD. The formation, transport, and concentration of oxalate in the Salar Grande may provide a geochemical analogue for oxalate-bearing minerals recently suggested to exist on Mars.

Abstract

Adequate read filtering is critical when processing high-throughput data in marker-gene-based studies. Sequencing errors can cause the mis-clustering of otherwise similar reads, artificially increasing the number of retrieved Operational Taxonomic Units (OTUs) and therefore leading to the overestimation of microbial diversity. Sequencing errors will also result in OTUs that are not accurate reconstructions of the original biological sequences. Herein we present a novel and sensitive sequence filtering algorithm that minimizes both problems by calculating the exact error-probability distribution of a sequence from its quality scores. In order to validate our method, we quality-filtered thirty-seven publicly available datasets obtained by sequencing mock and environmental microbial communities with the Roche 454, Illumina MiSeq and IonTorrent PGM platforms, and compared our results to those obtained with previous approaches such as the ones included in mothur, QIIME and UPARSE. Our algorithm retained substantially more reads than its predecessors, while resulting in fewer and more accurate OTUs. This improved sensitiveness produced more faithful representations, both quantitatively and qualitatively, of the true microbial diversity present in the studied samples. Furthermore, the method introduced in this work is computationally inexpensive and can be readily applied in conjunction with any existent analysis pipeline.

Abstract

Similarities between the Atacama Desert (Chile) and Mars include extreme aridity, highly oxidizing chemistry, and intense ultraviolet radiation that promoted the photochemical production of perchlorates and nitrates. Concentration of these ions under hyperarid conditions led to the formation of nitrate- and perchlorate-bearing deposits in ephemeral lakes, followed by later deposition of chlorides and sulfates. At some locations, such as the Salar Grande, hypersaline deposits have remained unaltered for millions of years. We conducted a drilling campaign in deposits of the Salar to characterize the preservation state of biological molecules. A 5 m deep discontinuous core was recovered and subjected to multitechnique analysis including the antibody microarray-based biosensor LDChip300 and the SOLID (Signs Of Life Detector) instrument, complemented by geophysical, mineralogical, geochemical, and molecular analysis. We identified two units based on the mineralogy: the upper one, from the surface to ~320 cm depth characterized by a predominance of halite and anhydrite, and the lower one, from 320 to 520 cm, with a drop in halite and anhydrite and an enrichment in nitrate and perchlorate. Organic compounds including biomolecules were detected in association with the different depositional and mineralogical units, demonstrating the high capacity for molecular preservation. Hypersaline environments preserve biomolecules over geologically significant timescales; therefore, salt-bearing materials should be high-priority targets for the search for evidence of life on Mars. Abstract

Actigraphy is a method for monitoring the movements of the nondominant arm, and the technology has found applications ranging from clinical devices to smart wristbands. Time series obtained from actigraphy data is used in chronobiology to define the sleep-wake cycle, as well as in sleep medicine to evaluate an individual’s sleep quality. In the study described in this paper, an algorithm based on recurrence quantification analysis (RQA) was applied to a time series obtained from a commercial actigraph, which was used to collect raw data alongside polysomnography (PSG), generally considered as the gold standard for assessing sleep quality. The central hypothesis is that transitions between sleep and wakefulness are not purely random events, but are strongly influenced by two internal processes: the homeostatic pressure and the circadian cycle. On the basis of this premise, application of RQA to time series as an estimator of this system should lead to improved results and allow more reliable investigations than a purely empirical approach. To compare the results from the RQA algorithm and those from PSG, we present a detailed statistical analysis involving a bias evaluation of the two methods following an approach suggested by Bland and Altman, a comparison of data processed using the kappa coefficient, and a comparison of consolidated sleep quality data using the p -value.
