4. Multi‐Satellite Data Assimilation for Large‐Scale Hydrological‐Hydrodynamic Prediction: Proof of Concept in the Amazon Basin.

Abstract

Satellite remote sensing enhances model predictions by providing insights into terrestrial and hydrological processes. While data assimilation techniques have proven promising, there is a lack of standardized and effective approaches for integrating multiple observations simultaneously. This study presents a novel assimilation framework, the multi‐observation local ensemble‐Kalman‐filter (MoLEnKF), designed to effectively integrate multiple variables, even at scales different than the model. Evaluation of MoLEnKF in the Amazon River basin includes assimilation experiments with remote sensing data only, including water surface elevation (WSE), terrestrial water storage (TWS), flood extent (FE), and soil moisture (SM). MoLEnKF demonstrates improvements in a scenario where regions lack in‐situ hydroclimatic records and when assuming uncertainties of large‐scale hydrologic‐hydrodynamic models. Assimilating WSE outperforms daily discharge and water‐level estimations, achieving 38% and 36% error reduction, respectively. However, the monthly evapotranspiration estimate achieves the greatest error reduction by assimilating SM with 11%. MoLEnKF always remains in second position in a ranking of error and uncertainty reduction, providing an intermediate condition, being able to holistically outperform univariate experiments. MoLEnKF also outperform state‐of‐the‐art models in many cases. This study suggests potential improvements, urging exploration of correlations between assimilated variables and adaptive localization methods based on seasonality. The flexibility and the elegant way of expressing the LEnKF equations by MoLEnKF facilitates their application with different types of variables, compatible with large‐scale hydrologic‐hydrodynamic models and missions such as SWOT. Its robustness ensures easy replicability worldwide, facilitating hydrological reanalysis and improved forecasting, establishing MoLEnKF as a valuable tool for the scientific community in hydrological research. Plain Language Summary: The use of satellites to collect information from far away helps us to understand how water behaves on the continents. But combining all this data with uncertain computer models is complicated. This study introduces a new method called multi‐observation local ensemble‐Kalman‐filter (MoLEnKF) to combine many different kinds of data at once. We tested MoLEnKF in the Amazon River basin, using satellite data on water levels, terrestrial water storage, flood extent and soil moisture. MoLEnKF by using all these observations at the same time obtained better results holistically than the individual experiments, improving our ability to predict aspects such as the amount of discharge, water level and evapotranspiration. This study is a step forward and could be really useful for understanding and predicting water‐related phenomena worldwide, especially in a context of scarce or no availability of in‐situ observations. Abstract

ALK-negative anaplastic large cell lymphoma (ALCL) comprises subgroups harboring rearrangements of DUSP22 (DUSP22- R) or TP63 (TP63-R). Two studies reported 90% and 40% 5-year overall survival (OS) rates in 21 and 12 DUSP22-R/TP63- not rearranged (NR) patients, respectively, making the prognostic impact of DUSP22-R unclear. Here, 104 newly diagnosed ALK-negative ALCL patients (including 37 from first-line clinical trials) from the LYSA TENOMIC database were analyzed by break-apart fluorescence in situ hybridization assays for DUSP22-R and TP63-R. There were 47/104 (45%) DUSP22-R and 2/93 (2%) TP63-R cases, including one DUSP22-R/TP63-R case. Abstract

Recent years have seen the development of 1-D and 2-D regional-scale hydrological-hydrodynamic models, which differ greatly from reach-scale applications in terms of subgrid assumptions, parameterization, and applied resolution. Although 1-D and 2-D comparisons have already been performed at reach and local scales, model differences at regional scale are poorly understood. Moreover, there is a need to improve the coupling between hydrological and hydrodynamic models. It is addressed here by applying the MGB model at 1-D and 2-D dimensions for the whole similar to 700,000 km(2) Negro basin (Amazon), which presents different wetland types. Long-term continuous simulations are performed and validated with multisatellite observations of hydraulic variables. Results showed that both approaches are similarly able to estimate discharges and water levels along main rivers, especially considering parameter uncertainties, but differ in terms of flood extent and volume and water levels in complex wetlands. In these latter, the diffuse flow and drainage patterns were more realistically represented by the 2-D scheme, as well as wetland connectivity across the basin. The 2-D model led to higher drainage basinwide, while the 1-D model was more sensitive to hydrodynamic parameters for discharge and flood extent and had a similar sensitivity for water levels. Finally, tests on the coupling between hydrologic and hydrodynamic processes suggested that their representation in an online way is less important for tropical wetlands than model dimensionality, which largely impacts water transfer and repartition. Plain Language Summary Hydrologic-hydrodynamic numerical models are powerful tools for improving water resources and disaster management and for the understanding of hydrological processes. Last years have faced the development of regional-scale applications (i.e., areas between 10(4) to 10(6) km(2)), with approaches at both 1-D and 2-D dimensions, that is, by representing surface water transport in either one (along rivers) or two horizontal directions (in order to represent flood propagation in wetlands). In this study, we present a regional-scale application of the MGB model at both 1-D and 2-D dimensions for the Negro River Basin in the Amazon and compare its capability to simulate processes at the basin scale and for different wetland types. We show that current 1-D and 2-D models are similarly capable to represent discharges and water levels along main rivers, but for more complex wetlands, the 2-D models are required, especially for estimating water levels. We also show that for tropical wetlands, the consideration of feedbacks between hydrologic and hydrodynamics is less important than the dimensionality of the model (i.e., if it is 1-D or 2-D). New lessons on the functioning of complex river-wetland systems in the Amazon are presented, in special consideration of the current availability of remote sensing data sets.

Abstract

BACKGROUND: Our aim was to evaluate whether the cell of origin (COO) as defined by the Hans algorithm and MYC/BCL2 coexpression, which are the two main biological risk factors in elderly patients treated with rituximab, cyclophosphamide, doxorubicin hydrochloride, vincristine, and prednisolone (R-CHOP), maintain their prognostic value in a large prospective clinical trial. PATIENTS AND METHODS: We evaluated 285 paraffin-embedded samples from patients (60-80 years of age) enrolled in the Lymphoma Study Association trial LNH03-6B who were treated with R-CHOP. We correlated the COO defined by the transcriptome according to the Wright algorithm with that defined by the Hans algorithm in a subset of 62 tumors with available frozen tissue samples. RESULTS: The non-germinal center B-cell-like phenotype according to the Hans algorithm and BCL2 expression (but not MYC and BCL2 coexpression) predicted worse progression-free survival [hazard ratio (HR)=1.78, P = 0.003 and HR = 1.79, P = 0.003, respectively] and overall survival (HR = 1.85, P = 0.005 and HR = 1.67, P = 0.02, respectively) independently of the International Prognostic Index. The correlation between the Hans algorithm and the Wright algorithm was 91%, with an almost perfect concordance according to a kappa test (0.81). CONCLUSIONS: Our results suggest that immunohistochemically defined COO remains a useful tool for predicting prognosis in diffuse large B-cell lymphoma when performed under optimized standardized conditions and that BCL2 expression may help to identify elderly patients at risk for relapse and who could potentially respond to anti-BCL2 targeted agents. In this prospective phase III trial, the coexpression of MYC and BCL2 does not appear to predict worse survival. CLINICAL TRIAL NUMBER: NCT00144755.
