Abstract

We use X-ray phase-contrast tomography (XPCT) in a multi-organ approach to identify early imaging markers predictive of multiple sclerosis (MS) in EAE animal model. As the majority of neurodegenerative diseases, MS is characterized by a progressive accumulation of biological deficits across different organs and systems. A simultaneous imaging of different disease-relevant networks and a multiscale imaging, ranging from the single cell through to the organ as a whole, are required to provide complete reliable information. XPCT offers the unprecedented possibility to investigate structural and cellular alterations at brain, gut, and eye levels in a multiscale approach. The comparison between naive mice and EAE-affected mice sacrificed at different time points and the correlation of the data from different organs and different time points, unveils the identification of early changes in organs possible predictive of the disease.

Abstract

The still unmet ability of following the fate of specific cells or molecules within the whole body would give an outstanding breakthrough in the comprehension of disease mechanisms and in the monitoring of therapeutic approaches. Our idea is to push forward the bio-nanotechnology to a level where it serves the most advanced 3D bio-medical imaging to provide a multi-scale imaging ranging from the whole organ down to the cellular level, enabling high-resolution visualization of disease-relevant cells within the whole disease-altered biological context. We present here the first proof-of-concept of a novel tomography procedure, Immuno-Histo-X-ray Phase Contrast Tomography (XPCT) that combines cutting-edge XPCT, which provides detailed image of the whole organ, with molecular imaging at the cellular level, identifying the relevant cells via an immunohistochemistry-based approach. We combine metal nanoparticles and a single-domain antibody that target relevant cells. Abstract

Introduction: In the study of neurodegenerative diseases, the possibility to follow the fate of specific cells or molecules within the whole body would be a milestone to better understand the complex evolution of disease mechanisms and to monitor the effects of therapies. The techniques available today do not allow the visualization of disease-relevant cells within the whole tridimensional biological context at high spatial resolution. Methods: Here we show the results from the first validation steps of a novel approach: by combining the conjugate nanobodies anti-glial fibrillary acidic protein (GFAP) and metal-nanoparticles (i.e. 2 nm gold NP) with X-ray phase contrast tomography (XPCT) we would be able to obtain a tridimensional visualization and identification of cells of interest together with the surrounding tissue and the vascular and neuronal networks. Results: By exploiting the X-ray attenuation properties of metal nanoparticles and the specific targeting capabilities of nanobodies, we could give XPCT the specificity it presently lacks, making it no longer a pure morphological but a molecular and targeted imaging technique. In our case, we synthesized and characterized Gold-NP/GFAP nanobody to target the astrocytes of mouse brain. Discussion: The results of the first tests presented in this paper have provided us with information on the feasibility of the approach, encouraging us to carry out further experiments in order to achieve the ultimate goal of setting up this new imaging technique.

6. Distribution of star-forming complexes in dwarf irregular galaxies

Abstract

We study the distribution of bright star-forming complexes in a homogeneous sample of 72 late-type (``irregular'') dwarf galaxies located within the 10 Mpc volume. Star-forming complexes are identified as bright lumps in B-band galaxy images and isolated by means of the unsharp-masking method. For the sample as a whole the radial number distribution of bright lumps largely traces the underlying exponential-disk light profiles, but peaks at a 10 percent smaller scale length. Moreover, the presence of a tail of star forming regions out to at least six optical scale lengths provides evidence against a systematic star formation truncation within that galaxy extension. Considering these findings, we apply a scale length-independent concentration index, taking into account the implied non-uniform random spread of star formation regions throughout the disk. The number profiles frequently manifest a second, minor peak at about two scale lengths. Relying on a two-dimensional stochastic self-propagating star formation model, we show these secondary peaks to be consistent with triggered star formation; for a few of the brighter galaxies a peculiar peak distribution is observed that is conceivably due to the onset of shear provided by differential rotation. On scales between 100 and 1000 pc, and by taking into account exponential-disk structure, bright lumps reveal cluster dimensions between 1.3 and 2, with a weak trend to higher dimensions for brighter galaxies. Interpreting cluster dimension in terms of porosity of a self-similar intragalactic medium, we derive a relation between current star formation rate, scale length, and porosity., Comment: 16 pages, accepted for publication in Astronomy & Astrophysics

7. Supernova type Ia luminosities, their dependence on second parameters, and the value of H_0

Abstract

A sample of 35 SNe Ia with good to excellent photometry in B and V, minimum internal absorption, and 1200 < v < \approx 30000 km/s is compiled from the literature. As far as their spectra are known they are all Branch-normal. For 29 of the SNe Ia also peak magnitudes in I are known. The SNe Ia have uniform colors at maximum, i.e. =-0.012 mag (sigma=0.051) and =-0.276 mag (sigma=0.078). In the Hubble diagram they define a Hubble line with a scatter of $\sigma_M$=0.21-0.16 mag, decreasing with wavelength. The scatter is further reduced if the SNe Ia are corrected for differences in decline rate Delta_m_15 or color (B-V). A combined correction reduces the scatter to sigma<=0.13 mag. After the correction no significant dependence remains on Hubble type or galactocentric distance. The Hubble line suggests some curvature which can be differently interpreted. A consistent solution is obtained for a cosmological model with Omega_M=0.3, Omega_Lambda=0.7, which is indicated also by much more distant SNe Ia. Absolute magnitudes are available for eight equally blue (Branch-normal) SNe Ia in spirals, whose Cepheid distances are known. If their well defined mean values of M_B, M_V, and M_I are used to fit the Hubble line to the above sample of SNe Ia one obtains H_0=58.3 km/s/Mpc, or, after adjusting all SNe Ia to the average values of Delta_m_15 and (B-V), H_0=60.9 km/s/Mpc. Various systematic errors are discussed whose elimination tends to decrease H_0. The finally adopted value at the 90-percent level, including random and systematic errors, is H_0=58.5 +/- 6.3 km/s/Mpc. Several higher values of H_0 from SNe Ia, as suggested in the literature, are found to depend on large corrections for variations of the light curve parameter and/or on an unwarranted reduction of the Cepheid distances of the calibrating SNe Ia., Comment: 42 pages, including 9 figures; submitted to ApJ

Abstract

Cepheids are the pillar of the extragalactic distance scale, but their reach in distance is not sufficient to calibrate H_0. Yet HST has provided Cepheid distances to eight galaxies which have produced SNe Ia. The latter are used as nearly perfect standard candles to carry the distance scale to 30,000 km/s, giving H_0 = 58 +/- 5. Abstract

Colorectal and glioblastoma cancer stem-like cells (CSCs) are essential for translational research. Cell line authentication by short tandem repeat (STR) profiling ensures reproducibility of results in oncology research. This technique enables to identify mislabeling or cross-contamination of cell lines. In our study, we provide a reference dataset for a panel of colorectal and glioblastoma CSCs that allows authentication. Each cell line was entered into the cell Line Integrated Molecular Authentication database 2.1 to be compared to the STR profiles of 4485 tumor cell lines. This article also provides clinical data of patients from whom CSCs arose and data on the parent tumor stage and mutations. STR profiles and information of our CSCs are also available in the Cellosaurus database (ExPASy) as identified by unique research resource identifier codes.

Abstract

With the increasing number of research biobanks and the importance of their role in supporting medical and biological research, the development and sharing of biobanking best practices and benchmarking standards has become paramount. To promote outstanding biobank services for research, the Research Biobank of the Year Competition (RBYC) has been inaugurated by the European, Middle-Eastern, and African Society for Biopreservation and Biobanking (ESBB) in October 2013. The procedures for the call and evaluation procedure, including the newly developed scoring system, are presented here. The statistics and evaluation results of the first year's applications, as well as the experiences of the jury are reported here, and improvements for the RBYC in subsequent years are proposed. Beyond offering a unique benchmarking opportunity for biobanks, the RBYC is discussed as a novel tool to enhance biobank quality, transparency, usage, connectivity, innovation, and sustainability.

Abstract

The QDF (Quality of life, Depression and cognitive Function) is a project elaborated and implemented by a Research Network of the nursing personnel of the cardiological services which promoted and run the GISSI-HF trial. It is a "companion study" which included up to 1/3 of all the GISSI-HF centres.To describe the distribution and evaluation of QDF variables measured with standardised instruments (Kansas City Cardiomiopathy Quality of Life questionnaire, KCCQ; Geriaric Depression scale GDS, Mini Mental State Examination MMSE), to verify whether and how far their knowledge could influence clinical decisions; to assess the prognostic impact of QDF values measured at entrance on mortality and morbidity. The study protocol included also a comprehensive evaluation of QDF variables independent from the questionnaires, by the nurses in charge of the study patients (the results of this part of the protocol are not included in this report).The QDF data were collected at baseline, 6 months, 1, 2 and 3 years through two self administered questionnaires (KCCQ, GDS), while the MMSE only for patients70 years. The questionnaires were administered to the patients who accepted to participate. A specific goal of the project focuses on the degree of independence between the results of the questionnaires and the judgement of the nurses. This contribution presents the operational implementation of the study protocol and general framework.

Abstract

To document the degree of reliability of validated and mainly qualitative questionnaires most often used in research settings and/or projects, in a broad network of unselected cardiological centres; to assess whether the quality of data could be compatible with their use not only as descriptive but also in prognostic scores including mainly "hard" clinical variables.Eighty-three patients were included. Compliance with the self-administration of the Kansas City Cardiomiopathy Questionnaire (KCCQ) and the Geriatric Depression Scale (GDS) was very high throughout the study (KCCQ 97.7% baseline and 85.8% at three years). As expected more difficulties were encountered with the Mini Mental State Examination (administered to 80.6% patients baseline and to 54.3% at three years). Main reasons for not administering questionnaires was ascribed to organizational and patients problems.It is worth noticing that the study was conducted by nurses without economical incentives. The questionnaires were well completed with minimal missing data. If questionnaires are well presented to patients, a routine assessment of quality of life and depression could be feasible in everyday care.

Abstract

Aim of this paper is to explore and quanti-quantitatively assess whether QoL as measured with the Kansas City Cardiomiopathy Questionnaire (KCCQ) summary score could be considered as an independent relevant component of clinical prognostic score of morbidity and mortality and identify patients at risk for death or admissions.Sixty-three per cent NYHA II and 39.4% NYHA III-IV patients experience a good Qol (score75). Risk factors for "not good" (75) QoL are age, NYHA class III-IV, diabetes, COPD and previous hospitalizations. NYHA II and III-IV patients with not good QoL experience an higher mortality than patients with a good QoL in the same classes. NYHA II patients with not good QoL experience the same 1 year readmission rates as NYHA III-IV patients with good QoL.Quality of life scores identify patients with different risk of mortality and readmissions within the same NYHA class. The prognostic value of KCCQ summary scores could identify candidates for disease management in whom better targeted care strategies may reduce hospitalizations and prevent deaths.

Abstract

This contribution describes the patients included for the QDF (Quality of life, Depression and Cognitive functions) study and explores the distribution and time trends of scores of the three dimensions.The large majority of patients are males (78.3%), and mean age is 67.2 +/- 10.7 years (9.2%80 years). Patients in NYHA class III-IV experience more depression and a worse QoL as compared to the less severly clinically affected counterparts in NYHA class. Although QoL and depression are influenced by the same variables, not all patients with bad QoL are depressed and viceversa. Mean scores of cognitive functioning are not influenced by the same variables that influence QoL and depression.The regular measurement of QoL, depression and cognitive function is recommended by some authors. It seems to be confirmed that the three dimensions need to be explored to disentangle the specific informative value that each of them could add to clinical measures.

Abstract

The prevalence of cognitive impairment (CI) is reported to be higher in heart failure patients than in the general population.To describe the prevalence of CI and to assess its prognostic value on the increased risk of hospitalizations and death over the medium term (3 years).MMSE was administered to 620/745 heart failure patients70 years. Up to 35% of the tested population were classified as mild CI and 12% as moderate. The severity of baseline clinical conditions, as expressed as NYHA classes, do not appear to be associated to different degrees of CI. The subgroup of patients with moderate CI includes higher proportion of people with the lowest educational level, of feminine gender, of higher age (80). Even in a population closely monitored as the one included in the GISSI-HLF trial, it is worth to be underlined that the presence of moderate CI appears to be strongly prognostic of an increased burden of care (hospitalizations), and of death over the whole observation period of the study (3 years).The presence of even mild or moderate CI is prognostic for increased mortality and readmissions for patients of the same NYHA class. In the absence of effective intervention strategies, research should concentrate on the yield, if any, of a systematic screening for CI in heart failure patients.

Abstract

Depression is common in patients with heart failure but its causal and/or prognostic role with respect to the cardiac condition in still a matter of research.To describe the prevalence of depression and to assess its specific prognostic value in terms of increased risk of hospitalizations or death.Overall, 27.5% of 1495 patients experience symptoms of depression. Risk factors for depression are female sex, age70, higher NYHA class, previous hospitalizations and comorbidities. Depressed patients of the same NYHA class are at higher risk for 1 and 3 years mortality and all causes hospital admissions.Depression, assessed with a simple and easy to administer instrument, can add information to clinical variables in the identification of patients at increased risk of mortality and readmissions within the same NYHA class.

Abstract

The child's discomfort and the cost of overnight hospitalization are clear disadvantages of prolonged esophageal pH monitoring. The aim of this study was to verify the reliability of short recording versus 24-h testing in a pediatric series with symptoms suggestive of gastroesophageal reflux (GER) disease. A 24-h pH monitoring performed on 160 patients with either gastroenterological symptoms (n = 61), respiratory problems (n = 58), or emesis plus respiratory problems (n = 41) was reviewed. Regardless of clinical presentation, children were also classified according to age : 10% were considered positive in patients 5% were considered positive in other age groups. Negative predictive values of the short recording RI ranged from 71 to 90%. Positive predictive values ranged from 50 to 83% ; it was unreliable for children
