Abstract

Background: Insulin resistance (IR) in skeletal muscle is a key feature of the pre-diabetic state, hypertension, dyslipidemia, cardiovascular diseases and also predicts type 2 diabetes. However, the underlying molecular mechanisms are still poorly understood. Methods: To explore these mechanisms, we related global skeletal muscle gene expression profiling of 38 non-diabetic men to a surrogate measure of insulin sensitivity, i.e. homeostatic model assessment of insulin resistance (HOMA-IR). Results: We identified 70 genes positively and 110 genes inversely correlated with insulin sensitivity in human skeletal muscle, identifying autophagy-related genes as positively correlated with insulin sensitivity. Replication in an independent study of 9 non-diabetic men resulted in 10 overlapping genes that strongly correlated with insulin sensitivity, including SIRT2, involved in lipid metabolism, and FBXW5 that regulates mammalian target-of-rapamycin (mTOR) and autophagy. The expressions of SIRT2 and FBXW5 were also positively correlated with the expression of key genes promoting the phenotype of an insulin sensitive myocyte e.g.PPARGC1A. Conclusions: The muscle expression of 180 genes were correlated with insulin sensitivity. These data suggest that activation of genes involved in lipid metabolism, e.g.SIRT2, and genes regulating autophagy and mTOR signaling, e.g.FBXW5, are associated with increased insulin sensitivity in human skeletal muscle, reflecting a highly flexible nutrient sensing.

Abstract

Probiotics are linked to positive regulatory effects on the immune system. The aim of the study was to examine the association between the exposure of probiotics via dietary supplements or via infant formula by the age of 1 year and the development of celiac disease autoimmunity (CDA) and celiac disease among a cohort of 6520 genetically susceptible children. Use of probiotics during the first year of life was reported by 1460 children. Time-to-event analysis was used to examine the associations. Overall exposure of probiotics during the first year of life was not associated with either CDA (n = 1212) (HR 1.15; 95%CI 0.99, 1.35; p = 0.07) or celiac disease (n = 455) (HR 1.11; 95%CI 0.86, 1.43; p = 0.43) when adjusting for known risk factors. Intake of probiotic dietary supplements, however, was associated with a slightly increased risk of CDA (HR 1.18; 95%CI 1.00, 1.40; p = 0.043) compared to children who did not get probiotics. It was concluded that the overall exposure of probiotics during the first year of life was not associated with CDA or celiac disease in children at genetic risk.

Abstract

35th European Photovoltaic Solar Energy Conference and Exhibition; 2005-2009, Regular photovoltaic (PV) system inspections have become a challenge with the significant growth in the number of modules and peak power capacity of PV installations. Image acquisition using drones, based on visual, thermographic, and more recently luminescence, can be a viable solution for large-scale PV inspections. As luminescence can provide a highly detailed and accurate PV module failure diagnosis, the development of a daylight electroluminescence (EL) imaging capability is of high importance. EL imaging performed in the field during the day requires the enhancement of the relatively weak luminescence signal over the noise from the sun. This is accomplished by image averaging and background subtraction, which requires the highly accurate registering of the of individual module images. A sequential EL image acquisition at high frame rates in continuous motion at different angles will be the real-world situation for a drone-based PV inspection in daylight, and to account for this movement while maintaining high quality images, several image processing steps must be developed. With this motivation, here we describe and perform EL image processing on a module with different faults to assure quality of the EL images in different motion speeds.

Abstract

Burn tragedy is a significant hazard to human life and properties. Automatic fire alarm device allows for real-time tracking, monitoring and intimation. Once the fire happens, it gives early warning and helps to the fire loss. It is versatile security and alarm system which can be used by individuals, corporations or industrial establishments. The concept behind this project is to have a quick, easy, accurate and low-cost fire sensing and location-intimating device for users to get support in critical situations. The system can be positioned at any remote location that the consumer can quickly access with the aid of GSM technology. In this it is proposed that NodeMCU acquire signals from different sensors and control to manage communication with property owner. This is carried out by sending SMS immediately to owner in initial stage and to fire brigade in critical stage to resolve fire hazard. Different sensors incorporated within system are smoke, fire and flammable gas sensor. It helps to estimate specific threshold values in hazardous condition and alarms the buzzer accordingly. Abstract

33rd European Photovoltaic Solar Energy Conference and Exhibition; 2016-2020, The goal of this work is to develop outdoor defect detection imaging and understand fully its challenges and limitations. The imaging is based on luminescence strategies that will be used for fast and accurate UAV-based inspection system for PV power plants. We studied electroluminescence (EL) acquisition under natural light conditions during several times of the day, under high sun irradiation, to unveil the sunlight noise characteristics towards an InGaAs detector. In order to bring more freedom to a drone-based inspection, we also show the preliminary results of a laser-line based photoluminescence (PL) strategy as a viable method for an outdoor module PL imaging system.

Abstract

Many research articles on various helical gearbox stages are accessible in reputable journals. The majorities of earlier research, despite the fact that triple step helical gearboxes have several benefits, were confined on single step and double step helical gears. Lately, most researching experts who work in this field have started paying close attention to the importances of three stage helical gearboxes. Throughout this research, authors seek to carry out an exhaustive evaluation of the current triple step helical transmission articles for examining the most widely used survey designs, approaches, tools & techniques. The above investigation expansively analysed 315 journal articles on helical gearbox various stages and then used content analysis procedure with an inductive method of research for carrying out an organized review of 45 literatures on three step helix transmission box that appeared in different scientific publications over the preceding two decades. Apart from this; the study presents a broad assessment of currently utilized materials, several future possibilities and problems in employing different materials for various parts of gearbox and its casing. This important research finding indicates there’s an enormous potentiality towards dominating the area of research as well as continuing more chances for exploring growth of three stage helical gearboxes, which provides novel career opportunities for both experts and industrial professionals.

Abstract

In Electric Spark Machining, choice of manufacturing constraints is a necessary feature. This Electrical Spark Machining is an advanced process in all manufacturing processes. For the manufacturing of sophisticated parts, which require accurate machining that time Electric Discharge Machining used. A super alloy, heat-treated tool steel, ceramics, heat resistance steels etc. which are tough to machining. For this, we use Electric Discharge Machining which also known as Electric Spark Machining. Incoloy alloy set in the group of super stainless steels. The main element of this Incoloy alloy is nickel-chromium-iron with other extra copper, molybdenum metal. High solidity at high peak temperature and super resist of erosion is the main feature of Incoloy alloy. Vast application of Incoloy 800 is like Heat exchangers, Carburizing equipments, Heating elements and nuclear steam generator tubing. In our research, we use hollow, flat, convex and concave electrode profile for machining of Incoloy 800.MRR and TWR are the parameters we analyze after the experiment.

Abstract

Healthy first-degree relatives with heredity of type 2 diabetes (FH+) are known to have metabolic inflexibility compared with subjects without heredity for diabetes (FH-). In this study, we aimed to test the hypothesis that FH+ individuals have an impaired response to exercise compared with FH-. Sixteen FH+ and 19 FH- insulin-sensitive men similar in age, peak oxygen consumption ((V) over dot(O2 peak)), and body mass index completed an exercise intervention with heart rate monitored during exercise for 7 mo. Before and after the exercise intervention, the participants underwent a physical examination and tests for glucose tolerance and exercise capacity, and muscle biopsies were taken for expression analysis. The participants attended, on average, 39 training sessions during the intervention and spent 18.8 MJ on exercise. (V) over dot(O2 peak)/kg increased by 14%, and the participants lost 1.2 kg of weight and 3 cm waist circumference. Given that the FH- group expended 61% more energy during the intervention, we used regression analysis to analyze the response in the FH+ and FH- groups separately. Exercise volume had a significant effect on (V) over dot(O2 peak), weight, and waist circumference in the FH- group, but not in the FH+ group. After exercise, expression of genes involved in metabolism, oxidative phosphorylation, and cellular respiration increased more in the FH- compared with the FH+ group. This suggests that healthy, insulin-sensitive FH+ and FH- participants with similar age, (V) over dot(O2 peak), and body mass index may respond differently to an exercise intervention. The FH+ background might limit muscle adaptation to exercise, which may contribute to the increased susceptibility to type 2 diabetes in FH+ individuals.

A single surgeon (NMR) performed all operations. Eyes that received the XEN implant concomitant with a glaucoma drainage device insertion or that were lost to 6-month follow-up were excluded. Intraocular pressure (IOP) change, change in glaucoma medications, frequency of slit lamp revision procedures, and frequency of secondary glaucoma surgeries were the primary outcomes compared between groups.Results: Fifty eyes that underwent ab interno placement and 30 eyes that underwent ab externo placement were studied. The ab interno cohort demonstrated a mean IOP reduction of 8.4 ± 1.7 mmHg (28.6% decrease) by 12 months, compared to a mean reduction of 12.8 ± 3.0 mmHg (40.1% decrease) in the ab externo group (p = 0.208). Mean reduction in medication use was 1.81 ± 0.29 medications in the ab interno group and 1.86 ± 0.37 in the ab externo group (p = 0.913). By 12 months, 58% of ab interno eyes had required 5-fluorouracil injection compared to 36.7% of ab externos (p = 0.105). Bleb needling was applied to 42% and 26.7% of the eyes, respectively (p = 0.231). A second glaucoma surgery was necessary for 20% of the ab interno cohort and 10% of the ab externo cohort (p = 0.351).Conclusion: There were no differences in outcomes between ab interno and ab externo placement of the XEN Gel Stent. Abstract

Chromosomal abnormalities occur in 0.1% to 0.2% of live births, and the most common clinically significant aneuploidy among live-born infants is Down syndrome (trisomy 21). Other sonographically detectable aneuploidies include trisomy 13, 18, monosomy X, and triploidy. Second-trimester ultrasound scan detects 2 types of sonographic markers suggestive of aneuploidy. Markers for major fetal structural abnormalities comprise the first type; the second type of markers are known as "soft markers" of aneuploidy. These latter markers are nonspecific, often transient, and can be readily detected during the second-trimester ultrasound. The most commonly studied soft markers of aneuploidy include a thickened nuchal fold, rhizomelic limb shortening, mild fetal pyelectasis, echogenic bowel, and echogenic intracardiac focus and choroid plexus cyst. There is a great deal of interest in the ultrasound detection of aneuploidy, as evidenced by the large number of publications in the literature on this topic. Unfortunately, studies evaluating the significance of the soft markers of aneuploidy vary widely and show contradictory results. In this article, we review the most common ultrasonographic soft markers used to screen aneuploidy and discuss ultrasonographic technique and measurement criteria for the detection of soft markers. We also review the clinical relevance of soft markers to aneuploidy risk assessment and evidence-based strategies for the management of affected pregnancies with each of these markers in light of current literature.

Abstract

Context: First-degree relatives of patients with type 2 diabetes (FH+) have been shown to have decreased energy expenditure and decreased expression of mitochondrial genes in skeletal muscle. In previous studies, it has been difficult to distinguish whether mitochondrial dysfunction and differential regulation of genes are primary (genetic) or due to reduced physical activity, obesity, or other correlated factors. Objective: The aim of this study was to investigate whether mitochondrial dysfunction is a primary defect or results from an altered metabolic state. Design: We compared gene expression in skeletal muscle from 24 male subjects with FH and 26 without FH matched for age, glucose tolerance, VO2peak (peak oxygen uptake), and body mass index using microarrays. Additionally, type fiber composition, mitochondrial DNA content, and citrate synthase activity were measured. The results were followed up in an additional cohort with measurements of in vivo metabolism. Results: FH+ vs. FH- subjects showed reduced expression of mitochondrial genes (P = 2.75 x 10(-6)), particularly genes involved in fatty acid metabolism (P = 4.08 x 10(-7)), despite similar mitochondrial DNA content. Strikingly, a 70% reduced expression of the monoamine oxidase A(MAOA) gene was found in FH+ vs. FH- individuals (P = 0.0009). Down-regulation of the genes involved in fat metabolism was associated with decreased in vivo fat oxidation and increased glucose oxidation examined in an additional cohort of elderly men. Conclusions: These results suggest that genetically altered fatty acid metabolism predisposes to type 2 diabetes and propose a role for catecholamine-metabolizing enzymes like MAOA in the regulation of energy metabolism. Abstract

The goal of this research is the assessment of the validity of existing three dimensional numerical programs in the prediction of the flow fields about general three dimensional hypersonic bodies. A detailed experimental research program was performed in which surface and flow field pressures were mapped. The results of the experimental work were compared with existing inviscid programs. Improvements were made on the existing numerical methods to include angle of attack. A summary of this work is presented.

Abstract

Flow field properties over a hypersonic vehicle are measured and compared with results from the numerical analysis of inviscid supersonic flow theory. Good agreement between theory and experiment at zero angle of attack, and significant discrepancy at angles of attack greater than or equal to 5 deg are found. The experiments indicate that turbulent boundary layer without flow separation exists over the model surface at zero angle of attack. Vortex lift-off, flow separation, and shock boundary layer interaction occur over the leeward surface, and near the wing-fuselage junction with angles of attack greater than or equal to 5 deg. Improvements of the existing numerical method to compute hypersonic cruise vehicle flow field at large angle of attack are presented.
