Abstract

Over the past century, evidence has emerged that endocrine disrupting chemicals (EDCs) have an impact on reproductive health. An increased frequency of reproductive disorders has been observed worldwide in both wildlife and humans that is correlated with accidental exposures to EDCs and their increased production. Epidemiological and experimental studies have highlighted the consequences of early exposures and the existence of key windows of sensitivity during development. Such early in life exposures can have an immediate impact on gonadal and reproductive tract development, as well as on long-term reproductive health in both males and females. Traditionally, EDCs were thought to exert their effects by modifying the endocrine pathways controlling reproduction. Advances in knowledge of the mechanisms regulating sex determination, differentiation and gonadal development in fish and rodents have led to a better understanding of the molecular mechanisms underlying the effects of early exposure to EDCs on reproduction. In this manuscript, we review the key developmental stages sensitive to EDCs and the state of knowledge on the mechanisms by which model EDCs affect these processes, based on the roadmap of gonad development specific to fish and mammals.

Abstract

Soils, nutrients and other factors support human food production. The loss of high-quality soils and readily minable nutrient sources pose a great challenge to present-day agriculture. A comprehensive scheme is required to make wise decisions on system’s sustainability and minimize the risk of crop failure. Soil quality provides useful indicators of its chemical, physical and biological status. Tools of precision agriculture and high-throughput technologies allow acquiring numerous soil and plant data at affordable costs in the perspective of customizing recommendations. Large and diversified datasets must be acquired uniformly among stakeholders to diagnose soil quality and plant nutrition at local scale, compare side-by-side defective and successful cases, implement trustful practices and reach high resource-use efficiency. Machine learning methods can combine numerous edaphic, managerial and climatic yield-impacting factors to conduct nutrient diagnosis and manage nutrients at local scale where factors interact. Compositional data analysis are tools to run numerical analyses on interacting components. Fractal models can describe aggregate stability tied to soil conservation practices and return site-specific indicators for decomposition rates of organic matter in relation to soil tillage and management. This chapter reports on machine learning, compositional and fractal models to support wise decisions on crop fertilization and soil conservation practices.

Abstract

The [Ca.sup.2+] affinity and permeation of the epithelial [Ca.sup.2+] channel (ECaC1) were investigated after expression in Xenopus oocytes. ECaC1 displayed anomalous mole-fraction effects. Extra-cellular [Ca.sup.2+] and [Mg.sup.2+] reversibly inhibited ECaC1 whole cell [Li.sup.+] currents: I[C.sub.50] = 2.2 [+ or -] 0.4 [micro]M (n = 9) and 235 [+ or -] 35 [micro]M (n = 10), respectively. These values compare well with the [Ca.sup.2+] affinity of the L-type voltage-gated [Ca.sup.2+] ([Ca.sub.v]1.2) channel measured under the same conditions, suggesting that high-affinity [Ca.sup.2+] binding is a well-conserved feature of epithelial and voltage-gated [Ca.sup.2+] channels. Neutralization of D550 and E535 in the pore region had no significant effect on [Ca.sup.2+] and [Mg.sup.2+] affinities. In contrast, neutralization of D542 significantly decreased [Ca.sup.2+] affinity (I[C.sub.50] = 1.1 [+ or -] 0.2 mM, n = 6) and [Mg.sup.2+] affinity (I[C.sub.50] > 25 [+ or -] 3 mM, n = 4). Despite a 1,000-fold decrease in [Ca.sup.2+] affinity in D542N, [Ca.sup.2+] permeation properties and the [Ca.sup.2+]-to-[Ba.sup.2+] conductance ratio remained comparable to values for wild-type ECaCi. Together, our observations suggest that D542 plays a critical role in [Ca.sup.2+] affinity but not in [Ca.sup.2+] permeation in ECaCi.
