Abstract

In Indiana and Minnesota the state education agency, state higher education agency, and the state workforce agency have collaborated to develop career and technical education courses intended to improve high school students' college and career readiness. These agencies partnered with the Regional Educational Laboratory Midwest to examine whether high school graduates in each state who completed a large number of career and technical education courses in a single career-oriented program of study (concentrators) had different college and workforce outcomes from graduates who completed fewer (samplers) or no career and technical education courses (nonparticipants). The study found that in the 2012/13-2017/18 graduation cohorts, male graduates were more likely to be concentrators than female graduates, and graduates who received special education services were more likely to be concentrators than those who did not receive services. Graduates who were not proficient in reading in grade 8 also were more likely to become concentrators than those who were proficient. Graduates who attended urban and suburban schools were more likely than students who attended town and rural schools to be nonparticipants. Concentrators were less likely than samplers and nonparticipants with similar characteristics to enroll in college, but the differences reflect mainly enrollment in four-year colleges. Concentrators were more likely to enroll in two-year colleges. Concentrators also were less likely than similar samplers and nonparticipants to complete a bachelor's degree within four to six years. Finally, compared with similar samplers and nonparticipants, concentrators were employed at higher rates in the first five years after high school and had higher earnings. This study brief discusses why the study was completed, what was studied and how, findings, and implications. 4. Do College and Career Readiness and Early College Success in Indiana Vary Depending on Whether Students Attend Public, Charter, or Private Voucher High Schools? Study Brief. REL 2021-071

Abstract

Indiana has a robust portfolio of high school options, including traditional public schools, charter schools, and private schools that accept Indiana Choice Scholarships (private voucher schools). This study identified the type of high school enrollment (high school type and voucher status) among the student populations of four cohorts (who were in grade 9 in 2010/11-2013/14) and examined those students' performance on indicators of college and career readiness and early college success. The study found that charter school students and recipients of private school vouchers (voucher recipients) were more likely than other students to be Black or to be eligible for the national school lunch program (an indicator of low socioeconomic status). In addition, students in private voucher schools who did not receive vouchers (nonvoucher students) performed similarly to or better than students in traditional public schools and charter school students on most indicators of college and career readiness, after other factors were adjusted for. And voucher recipients performed similarly to or better than students in traditional public schools on most indicators of college and career readiness, after other factors were adjusted for, whereas charter school students performed similarly to or worse than students in traditional public schools. Finally, nonvoucher students performed better than students in all other types of high school enrollment on one indicator of early college success, completing all attempted credits in the first year, after other factors were adjusted for. State and district administrators, charter school leaders and authorizers, and leaders of private voucher schools could use these findings to inform the design of policies to improve college and career readiness and early college success. 5. Understanding the Teacher Pipeline for Indiana's K-12 Public Schools. Study Brief. REL 2021-065

Abstract

Leaders at the Indiana Commission for Higher Education and the Indiana Department of Education are concerned about teacher shortages and want a better understanding of the teacher pipeline for the state's K-12 public schools. This study tracked 11,080 students who first enrolled in an Indiana public college or university in 2010/11, 2011/12, or 2012/13 and pursued a bachelor's degree in education at any point in college. Among those entrants 41 percent completed a bachelor's degree in education by 2017/18. Of those degree completers 55 percent earned an initial instructional license, and of those licensed, 69 percent entered teaching in an Indiana public school. Compared with the initial group of students entering education programs, students who completed a bachelor's degree, those who earned an initial instructional license, and those who entered teaching in an Indiana public school were less likely to be from a racial/ethnic minority group or to have been eligible for the national school lunch program in high school. Students who entered an education program in their third year of college or later were less likely to complete a bachelor's degree in education than students who entered in their first year, after other differences in student characteristics and the characteristics of the first college they attended were adjusted for. Students who received an Indiana 21st Century Scholarship in their first year of college or who received financial aid beyond their first year were more likely to complete a bachelor's degree in education, while students who received a Pell Grant were less likely. Finally, students who started at a two-year college and transferred to a four-year college were more likely to complete a bachelor's degree in education than students who started at a four-year college. To date, however, no studies have analyzed the use of this tool in patients with persistent symptoms following coronavirus disease 2019, a condition known as Long COVID.Purpose: To analyze the effectiveness of motivational interviewing with regard to the adherence to telerehabilitation for Long COVID using a mobile application. As a secondary objective, factors related to greater motivation before and after the motivational interviewing techniques were analyzed.Patients and Methods: This longitudinal design substudy used a sample of 52 adult patients with Long COVID participating in the intervention group of a randomized clinical trial. This trial examined the effectiveness of a telerehabilitation program for this population using a mobile application. This program included three motivational interviews to achieve maximum treatment adherence. In this study, the main variables were motivation and adherence to application use. Sociodemographic and clinical data, personal constructs, and affective state were also collected. Subsequently, a descriptive, correlational, and regression statistical analysis was performed using the SPSS Statistics program.Results: The median motivation prior to the first motivational interview was 8 (IQR 2), the median at the end of the last motivational interview was 8.5 (IQR 2.75), and the change in motivation levels after the three motivational interviews was 0.5 (IQR 1). Affective state and final motivation scores were predictors of greater adherence to telerehabilitation treatment.Conclusion: A high level of motivation after participating in motivational interviewing appears to be related to higher levels of adherence to telerehabilitation in patients with Long COVID. Abstract

Silicon-substituted hydroxyapatite (SiHA) macroporous scaffolds have been prepared by robocasting. In order to optimize their bone regeneration properties, we have manufactured these scaffolds presenting different microstructures: nanocrystalline and crystalline. Moreover, their surfaces have been decorated with vascular endothelial growth factor (VEGF) to evaluate the potential coupling between vascularization and bone regeneration. In vitro cell culture tests evidence that nanocrystalline SiHA hinders pre-osteblast proliferation, whereas the presence of VEGF enhances the biological functions of both endothelial cells and pre-osteoblasts. The bone regeneration capability has been evaluated using an osteoporotic sheep model. In vivo observations strongly correlate with in vitro cell culture tests. Those scaffolds made of nanocrystalline SiHA were colonized by fibrous tissue, promoted inflammatory response and fostered osteoclast recruitment. These observations discard nanocystalline SiHA as a suitable material for bone regeneration purposes. On the contrary, those scaffolds made of crystalline SiHA and decorated with VEGF exhibited bone regeneration properties, with high ossification degree, thicker trabeculae and higher presence of osteoblasts and blood vessels. Considering these results, macroporous scaffolds made of SiHA and decorated with VEGF are suitable bone grafts for regeneration purposes, even in adverse pathological scenarios such as osteoporosis., Comment: 23 pages

Abstract

Macroporous scaffolds made of a SiO2-CaO-P2O5 mesoporous bioactive glass (MBG) and epolycaprolactone (PCL) have been prepared by robocasting. These scaffolds showed an excellent in vitro biocompatibility in contact with osteoblast like cells (Saos 2) and osteoclasts derived from RAW 264.7 macrophages. In vivo studies were carried out by implantation into cavitary defects drilled in osteoporotic sheep. The scaffolds evidenced excellent bone regeneration properties, promoting new bone formation at both the peripheral and the inner parts of the scaffolds, thick trabeculae, high vascularization and high presence of osteoblasts and osteoclasts. In order to evaluate the effects of the local release of an antiosteoporotic drug, 1% (%wt) of zoledronic acid was incorporated to the scaffolds. Abstract

Introducción: La homofobia es un importante problema social, en particular para los adolescentes. La discriminación y el desprecio a que son sometidos aquellos adolescentes homosexuales o los que tienen dudas en torno a su identidad sexual, tiene consecuencias adversas en todo su desarrollo ulterior. Detectar y enfrentar conductas homofóbicas en los colegios, resulta clave si pretendemos una sociedad justa y sana, donde la convivencia con la diversidad se respete. La UNESCO invita a todas las sociedades a enfrentar estas conductas desde la ética individual y las políticas públicas en ambientes escolares. Objetivos: Conocer la prevalencia de homofobia en los adolescentes escolarizados de un colegio de la localidad de suba en la ciudad de Bogotá e identificar los métodos preventivos. Metodología: Se aplicó una encuesta presencial auto diligenciada y dicotómica a estudiantes de sexto a once grados, indagando por variables sociodemográficas y utilizando un test cuantitativo, en donde el total de su sumatoria indica si es o no homofóbico. Resultados: Se encuestaron 843 estudiantes. El 24,9% de la población encuestada no son homofóbicos, el 49,1% aún tienen algunos prejuicios y el 26% son homofóbicos. Se compararon los homofóbicos versus los no homofóbicos entre sexo femenino y masculino obteniendo como relaciones significativas un OR de 0.25, IC 95% Li 0.16 y Ls 0.37, también se comparó la homofobia versus la no homofobia entre edades de 13-15 años con 10-12 años obteniendo OR de 0.43, Li 0,27 y Ls 0,70, entre edades de 16-19 años con 10-12 años obteniendo OR de 0.3, Li 0.17 y Ls 0.51 y finalmente entre edades de 16-19 años con 13-15 años obteniendo OR 0.68, Li 0.42 y Ls 1.09. Conclusión: La homofobia es un rechazo que se puede generar de manera involuntaria a través del desprecio y la discriminación, ya que no se tiene en común la misma orientación social, pensamientos o ideologías. Se encontró que el 26% de los adolescentes escolarizados son homofóbicos. Se evidenció mayores actitudes homofóbicas en el sexo masculino que el femenino y se observó que entre menos edad entre los adolescentes, hay más actitudes homofóbicas.

41. Characterisation of a biosensor surface for use with scale-down technologies in the process monitoring of Escherichia coli antibody fragment production

Abstract

This thesis illustrates the need to consider overall process performance through the use of biosensors, scale-down and low-volume analytical technologies to achieve processes that account for cell engineering, upstream processing and its effects on downstream processing performance; the upstream/downstream interface. An Escherichia coli fed-batch fermentation expressing antibody fragment (Fab’) was used as a system representative of current manufacturing challenges. Sensor surface characterisation using dual polarisation interferometry (DPI) was explored and a step-wise fabrication and characterisation of a multi-layer DPI based biosensor was carried out. A chemical characterisation of the sensor surface was performed ex-situ using x-ray photoelectron spectroscopy (XPS) and secondary ion mass spectrometry (SIMS) to investigate the chemical identity and homogeneity of the chemical layers. The studies uncovered UV/ozone cleaning as the method of choice in comparison with surface acid washing and the use of hydrophilic surfaces as optimal for antibody recognition in comparison to hydrophobic surfaces. The studies highlighted the use of DPI as a sensor characterisation tool. Biological characterisation assessed the potential of the sensor as a monitoring tool. Following characterisation, method development ensued, to quantify antibody fragments via interaction with protein G. The surfaces were assessed through non-specific binding studies and challenged with complex fermentation materials. The characterised biosensors were utilised to assess Fab’ product loss and Fab’ quality during bioprocessing. The biosensor was shown to be sensitive down to 1.7 µg/mL of Fab’ with low interference due to non-specific binding (NSB) and good percentage recovery in 4-fold diluted extracellular broth. The sensor surfaces were then used in conjunction with scale-down tools for primary recovery and low-volume analytical technologies to obtain insights into the interaction between cells from the upstream process and downstream processing operations. For the process studied, the net yield at the end of the primary recovery, which takes into account both product losses during fermentation and centrifugation, was the critical parameter due to the intercellular nature of the product. Studies showed that 30 h was the optimal harvest time with regard to product loss. To benefit from increased product titres at 40 h and above requires that cell strength and integrity be improved. The effect of cell engineering on the growth and productivity of the upstream process as well as its effect on cell integrity at harvest was conducted. This was conducted through the assessment of the growth and productivity impacts of periplasmic nuclease expression in an Escherichia coli Fab' fragment production strain. This modification allows the removal of DNA upon cell disruption. The Co-expression of staphylococcal nuclease and antibody product in the same sub-cellular compartment showed no impact on the growth characteristics and Fab’ yield of the host strain. However, introduction of the periplasmic nuclease did perturb kinetics of Fab’ production; increasing the rate of intracellular Fab’ accumulation and concomitantly increasing in the rate of appearance of Fab’ in the external medium. This thesis highlights the need to consider the effects of upstream processing on primary recovery and shows how harvest point determination is a critical process parameter to assess during the design of a bioprocess.

Abstract

Individuals with minor ischaemic stroke and intracranial occlusion are at increased risk of poor outcomes. Intravenous thrombolysis with tenecteplase might improve outcomes in this population. We aimed to test the superiority of intravenous tenecteplase over non-thrombolytic standard of care in patients with minor ischaemic stroke and intracranial occlusion or focal perfusion abnormality.

Abstract

Network analysis is currently used in a myriad of contexts: from identifying potential drug targets to predicting the spread of epidemics and designing vaccination strategies, and from finding friends to uncovering criminal activity. Despite the promise of the network approach, the reliability of network data is a source of great concern in all fields where complex networks are studied. Here, we present a general mathematical and computational framework to deal with the problem of data reliability in complex networks. In particular, we are able to reliably identify both missing and spurious interactions in noisy network observations. Remarkably, our approach also enables us to obtain, from those noisy observations, network reconstructions that yield estimates of the true network properties that are more accurate than those provided by the observations themselves. Our approach has the potential to guide experiments, to better characterize network data sets, and to drive new discoveries.

Abstract

We use agent-based modeling to investigate the effect of conservatism and partisanship on the efficiency with which large populations solve the density classification task--a paradigmatic problem for information aggregation and consensus building. We find that conservative agents enhance the populations' ability to efficiently solve the density classification task despite large levels of noise in the system. In contrast, we find that the presence of even a small fraction of partisans holding the minority position will result in deadlock or a consensus on an incorrect answer. Our results provide a possible explanation for the emergence of conservatism and suggest that even low levels of partisanship can lead to significant social costs., Comment: 11 pages, 5 figures

Abstract

The Cox regression analysis is used to determine the relationship between a dependent variable and covariates in survival analysis involving censored data. The proportional hazards assumption is one of the most important assumptions of Cox regression. Outliers may have a strong influence on the Cox regression model's parameter estimates and lead to violation of the proportional hazard assumption. Therefore, having outliers in the data set is a problem for researchers. In this case, robust estimations are commonly used to infer the parameters in a more robust way. However, we explore a new approach consisting of considering an outlier as missing data and replacing it by the multiple imputation method. The aim of this study is to compare these two methods through simulation. Furthermore, an analysis of a lung cancer data set is considered for illustration. According to the results of the study carried out based on simulated data sets and a real data set, the multiple imputation method, which is a missing data analysis method, solves the problem of outliers better than the robust estimation method, as the outcome is closer to the results obtained through original data.
