Abstract Krill are vital as food for many marine animals but also impacted by global warming. To learn how they and other zooplankton may adapt to a warmer world we studied local adaptation in the widespread Northern krill (Meganyctiphanes norvegica). We assemble and characterize its large genome and compare genome-scale variation among 74 specimens from the colder Atlantic Ocean and warmer Mediterranean Sea. The 19 Gb genome likely evolved through proliferation of retrotransposons, now targeted for inactivation by extensive DNA methylation, and contains many duplicated genes associated with molting and vision. Analysis of 760 million SNPs indicates extensive homogenizing gene-flow among populations. Nevertheless, we detect signatures of adaptive divergence across hundreds of genes, implicated in photoreception, circadian regulation, reproduction and thermal tolerance, indicating polygenic adaptation to light and temperature. The top gene candidate for ecological adaptation was nrf-6, a lipid transporter with a Mediterranean variant that may contribute to early spring reproduction. Such variation could become increasingly important for fitness in Atlantic stocks. Our study underscores the widespread but uneven distribution of adaptive variation, necessitating characterization of genetic variation among natural zooplankton populations to understand their adaptive potential, predict risks and support ocean conservation in the face of climate change.

Abstract The study of species groups in which the presence of interspecific hybridization or introgression phenomena is known or suspected involves analysing shared bi-parentally inherited molecular markers. Current methods are based on different categories of markers among which the classical microsatellites or the more recent genome wide approaches for the analyses of thousands of SNPs or hundreds of microhaplotypes through high throughput sequencing. Our approach utilizes intron-targeted amplicon sequencing to characterise multi-locus intron polymorphisms (MIPs) and assess genetic diversity. These highly variable intron regions, combined with inter-specific transferable loci, serve as powerful multiple-SNP markers potentially suitable for various applications, from species and hybrid identification to population comparisons, without prior species knowledge. We developed the first panel of MIPs highly transferable across fish genomes, effectively distinguishing between species, even those closely related, and populations with different structures. MIPs offer versatile, hypervariable nuclear markers and promise to be especially useful when multiple nuclear loci must be genotyped across different species, such as for the monitoring of interspecific hybridization. Moreover, the relatively long sequences obtained ease the development of single-locus PCR-based diagnostic markers. This method, here demonstrated in teleost fishes, can be readily applied to other taxa, unlocking a new source of genetic variation.

Haptic feedback holds the potential to enhance the engagement and expressivity of future digital and electric musical instruments. This study investigates the impact of artificial vibration on the perceived quality of a silent electric cello. We developed a haptic cello prototype capable of rendering vibration signals of varying degree of congruence with the produced sound. Experienced cellists participated in an experiment comparing setups with and without vibrotactile feedback, rating them on preference, perceived power, liveliness, and feel. Results show nuanced effects, with added vibrations moderately enhancing feel and liveliness, and significantly increasing perceived power when using vibrations obtained from the pickup at the cello’s bridge. High uncertainty in our statistical model parameters underscores substantial individual differences in the participants responses, as commonly found in qualitative assessments, and highlights the importance of consistent feedback in the vibrotactile and auditory channels. Our findings contribute valuable insights to the intersection of haptics and music technology, paving the way for creating richer and more engaging experiences with future musical instruments.

Abstract Background Chronic migraine (CM) negatively impacts the quality of life of 2 to 4% of pediatric patients. In adults, CM is frequently linked to medication overuse headache (MOH), but there is a much lower prevalence of MOH in children. A suboptimal response to acute therapies may lead to their reduced use, thus preventing MOH development in children and adolescents. The frequency of patients with CM who do not respond to acute therapies was examined in the present study. We investigated whether the prevalence of MOH was different between responders and non-responders. We also examined whether patients receiving prophylactic therapy had an improved response to acute therapy. Finally, we investigated if there was a difference in the frequency of psychiatric comorbidities between responders and non-responders. Methods We retrospectively analysed clinical data of all chronic pediatric migraineurs under the age of 18 referred to the Headache Centre at Bambino Gesù Children Hospital in June 2021 and February 2023. ICHD3 criteria were used to diagnose CM and MOH. We collected demographic data, including the age at onset of migraine and the age of the CM course. At baseline and after 3 months of preventive treatment, we evaluated the response to acute medications. Neuropsychiatric comorbidities were referred by the children’s parents during the first attendance evaluation. Results Seventy patients with CM were assessed during the chosen period. Paracetamol was tried by 41 patients (58.5%), NSAIDs by 56 patients (80.0%), and triptans by 1 patient (1.4%). Fifty-one participants (73%) were non-responder to the abortive treatment. The presence of MOH was detected in 27.1% of the whole populations. Regarding our primary aim, MOH was diagnosed in 29% of non-responder patients and 22% of responders (p > 0.05). All patients received preventative treatment. In line with current trends and challenges in the field of cultural heritage, the research presented here is the result of the V.I.T.A. project: an initiative on digital cultural heritage that involves museum operators in the digitization and development of digital applications for various works of art and ethnographic elements from the Civic Museums of Ascoli Piceno. The project includes the entire digitization workflow, ranging from three-dimensional modeling to immersive virtual experiences for museums. The work testifies to a fruitful collaboration between universities and cultural institutions, which has led the Marche museum to become a virtuous example in the field of digital transformation of heritage.

Today growing interest to accomplish the authorities’ nutritional and health claim is oriented towards vegetable-based α-linolenic acid (ALA) enriched foods.The aim of this work was to develop a cracker (CKO) enriched with Camelina seed (Camelina sativa L.) oil (CO), a well-known ALA and linoleic acid (LA) source. A fast HPLC-WVD method was set up and validated to quantify ALA and LA in CO and CKO. CKO was thought as bread alternatives that meets the health claim of European Commission regulation on cardiovascular well-being.Results indicated an ALA content in CO of 27.06 ± 1.65 g/100 g, with an ALA/LA ratio higher than two; differently, ALA content was of 2.10 ± 0.15 g/100 g in CKO, with lower ratio due to LA contribution of other ingredients. Cooking process did not affect PUFAs content in CO, confirming its stability when incorporated in food products. ALA bioaccessibility was lower than 34 % in raw oil, while increased when CO was incorporated in CKO (40.06 ± 0.83 %), thus indicating a protecting activity within the food matrix. These results confirmed that CKO could represent a suitable food to deliver camelina oil essential fatty acids, even if an additional optimization of the formulation is required to further improve ALA bioaccessibility.

The proteoleaginous plants demand has seen significant growth, leading to an expansion of the sunflower (Helianthus annuus, L.) and industrial hemp (Cannabis sativa, L.) cultivation area in Italy. However, by-products obtained during seed oil extraction and agricultural residues are often unused due to the absence of a receptive market and nearby processing centers. The carbon footprint (CF) methodology was used to compare the two supply chains considering the soil incorporation of all crop residues and by-products. The boundary of the supply chains analyzed includes all the agricultural processes that occur during cultivation and the subsequent oil extraction phase. Furthermore, research explored the direct and indirect environmental benefits of incorporating by-products into the soil, in terms of reducing the need for mineral fertilizers to restore soil fertility due to the nutrients contained in the buried biomass, and the potential carbon sequestration achievable. Results show that 1 kg of sunflower and hemp oil release 4.49 kg CO2-eq and 23.34 kg CO2-eq, respectively. Agriculture represents the most impacting phase and, in particular, fertilization, tillage and harvest are responsible for high emissions. The different results between the two supply chains can be attributed mainly to yield and extraction efficiency. The use of by-products as amended in the soil (avoided fertilizers) contributes to a reduction of greenhouse gas (GHG) emissions by −0.53 kg CO2-eq and −7.87 kg CO2-eq per kg of sunflower and hemp oils, respectively. Additionally, the sequestration of carbon in biomass can result in a further reduction of −1.16 and −33.6 kg CO2-eq per kg of sunflower and hemp oil, respectively. In summary, sunflower oil production emits 74 % less CO2 than hemp oil. However, if all crop biomass is buried, hemp has the potential to be more sustainable. This phenomenon depends on many factors such as soil type, climate, and farming practices. The study outcomes can aid policymakers, farmers, and the agribusiness to make informed decisions on promoting and expanding sustainable sunflower and hemp cultivation in Italy.

Abstract Tumor recognition by T cells is essential for antitumor immunity. A comprehensive characterization of T cell diversity may be key to understanding the success of immunomodulatory drugs and failure of PD-1 blockade in tumors such as multiple myeloma (MM). Here, we use single-cell RNA and T cell receptor sequencing to characterize bone marrow T cells from healthy adults (n = 4) and patients with precursor (n = 8) and full-blown MM (n = 10). Large T cell clones from patients with MM expressed multiple immune checkpoints, suggesting a potentially dysfunctional phenotype. Dual targeting of PD-1 + LAG3 or PD-1 + TIGIT partially restored their function in mice with MM. We identify phenotypic hallmarks of large intratumoral T cell clones, and demonstrate that the CD27− and CD27+ T cell ratio, measured by flow cytometry, may serve as a surrogate of clonal T cell expansions and an independent prognostic factor in 543 patients with MM treated with lenalidomide-based treatment combinations.

Angulated-screw channels (ASCs) allow the clinician to employ screw-retained restorations in almost all cases, as the access hole can be moved away from the vestibular portion of the crown, where it would jeopardize the final esthetic result. The objective of this study was to compare screw-retained restorations employing ASCs with restorations cemented on angled abutments. In this study, 30 subjects, equally divided into two groups: group 1 (cemented restorations on angulated abutments) and group 2 (screw-retained restorations adopting ASCs), were treated and retrospectively compared after 2 years using the pink esthetic score (PES) and the white esthetic score (WES). All restorations were in use at the last follow-up, with a survival rate of 100%. Three mechanical complications were observed (2 chippings and 1 crown came loose), with a success rate of 93% in group 1 and 87% in group 2 (p > 0.05). No statistically significant differences were reported regarding the esthetic outcome; the marginal bone loss (MBL) showed better results for the screw-retained restorations, both at the distal aspect (group 1 = 0.98 mm ± 0.16; group 2 = 0.45 mm ± 0.06; p = 0.006) and at the mesial aspect (group 1 = 1.04 ± 0.27; group 2 = 0.45 ± 0.005; p < 0.001). From an esthetical perspective, screw-retained restorations with ASCs and cemented restorations on angulated abutments are both effective means of restoring implants; both have excellent esthetic outcomes, but screw-retained restorations have reduced bone loss when compared to cemented ones but are more prone to mechanical complications. Still, our results must be cautiously observed given the reduced dimension of our sample. Larger studies are needed to confirm our findings.

Myelin oligodendrocyte glycoprotein antibody disease (MOGAD) is characterized by multiple phenotypic conditions such as acute disseminated encephalomyelitis, optic neuritis, and myelitis. MOGAD’s spectrum is expanding, with potential symptoms of increased intracranial pressure that are similar to idiopathic intracranial hypertension (IIH). We report a boy with new-onset continuous headache and a brain MRI at onset suggesting idiopathic intracranial hypertension (IIH). The patient showed resistance to treatment with acetazolamide and, after one month, developed optic neuritis in the left eye. Laboratory tests documented positive MOG antibodies (anti-MOG) in the serum. The final diagnosis was MOGAD, with the initial symptoms resembling IIH.

This review summarizes the relationship between diet, the gut microbiome, and migraine. Key findings reveal that certain dietary factors, such as caffeine and alcohol, can trigger migraine, while nutrients like magnesium and riboflavin may help alleviate migraine symptoms. The gut microbiome, through its influence on neuroinflammation (e.g., vagus nerve and cytokines), gut–brain signaling (e.g., gamma-aminobutyric acid), and metabolic function (e.g., short-chain fatty acids), plays a crucial role in migraine susceptibility. Migraine can also alter eating behaviors, leading to poor nutritional choices and further exacerbating the condition. Individual variability in diet and microbiome composition highlights the need for personalized dietary and prebiotic interventions. Epidemiological and clinical data support the effectiveness of tailored nutritional approaches, such as elimination diets and the inclusion of beneficial nutrients, in managing migraine. More work is needed to confirm the role of prebiotics, probiotics, and potentially fecal microbiome translation in the management of migraine. Future research should focus on large-scale studies to elucidate the underlying mechanisms of bidirectional interaction between diet and migraine and develop evidence-based clinical guidelines. Integrating dietary management, gut health optimization, and lifestyle modifications can potentially offer a holistic approach to reducing migraine frequency and severity, ultimately improving patient outcomes and quality of life.

Globe artichoke (Cynara cardunculus var. scolymus L.) is widely cultivated in the Mediterranean area and Italy is one of the largest producers. A great issue is represented by its high amount of by-product, mainly consisting of external bracts and stems, but also of residual leaves, stalks, roots, and seeds. Artichoke by-products are rich in nutrients (carbohydrates and proteins) and bioactive compounds (polyphenols and terpenes) and represent potential ingredients for foodstuffs, functional foods, and food supplements, due to their functional and biological properties. In fact, artichoke by-products’ components exhibit many beneficial effects, such as dyspeptic, prebiotic, antioxidant, anti-inflammatory, antiglycative, antimicrobial, anticarcinogenic, and hypolipidemic properties. Therefore, they can be considered potential food ingredients useful in reducing the risk of developing metabolic and age-related disorders. This work summarizes the economic and environmental impact of the recovery and valorization of artichoke by-products, focusing on rheological, physical, and biological properties of the different components present in each by-product and their different food applications.

Background: Multiple sclerosis (MS) is increasing in the pediatric population and, as in adults, symptoms vary among patients. In children the first manifestations can sometimes overlap with acute neurological symptoms. Urological symptoms have not been much studied in childhood. We shared our experience with MS urological manifestation in children. Methods: This article is a retrospective evaluation of all children with MS, according to the Krupp criteria, who also present with urological symptoms. We collected demographic and clinical history, the MR localization of demyelinating lesions, urological symptoms, and exams. Results: We report on six MS pediatric cases with urological manifestation. Urinary symptoms, characterized by urinary incontinence in five patients and urinary retention in one patient, appeared in a different time frame from MS diagnosis. Urodynamic exams showed both overactive and underactive bladder patterns. Treatment was defined according to lower urinary tract dysfunction, using clean intermittent catheterization, oxybutynin, and intradetrusor Onabotulinum Toxin-A injection. A low acceptance rate of invasive evaluation and urological management was observed. Conclusions: The MS diagnosis was traumatic for all our patients. We believe it is important to address urological care in young people from the time of diagnosis for prompt management; it could be useful to include a pediatric urologist in multidisciplinary teams.
