Second messenger cyclic adenosine monophosphate (cAMP) has been found to regulate multiple mitochondrial functions, including respiration, dynamics, reactive oxygen species production, cell survival and death through the activation of cAMP-dependent protein kinase A (PKA) and other effectors. Several members of the large family of A kinase anchor proteins (AKAPs) have been previously shown to locally amplify cAMP/PKA signaling to mitochondria, promoting the assembly of signalosomes, regulating multiple cardiac functions under both physiological and pathological conditions. In this review, we will discuss roles and regulation of major mitochondria-targeted AKAPs, along with opportunities and challenges to modulate their functions for translational purposes in the cardiovascular system.

Accumulating evidence suggests that modifications of gut function and microbiota composition might play a pivotal role in the pathophysiology of several cardiovascular diseases, including heart failure (HF). In this study we systematically analysed gut microbiota composition, intestinal barrier integrity, intestinal and serum cytokines and serum endotoxin levels in C57BL/6 mice undergoing pressure overload by transverse aortic constriction (TAC) for 1 and 4 weeks. Compared to sham-operated animals, TAC induced prompt and strong weakening of intestinal barrier integrity, long-lasting decrease of colon anti-inflammatory cytokine levels, significant increases of serum levels of bacterial lipopolysaccharide and proinflammatory cytokines. TAC also exerted effects on microbiota composition, inducing significant differences in bacterial genera inside Actinobacteria, Firmicutes, Proteobacteria and TM7 phyla as shown by 16S rDNA sequencing of fecal samples from TAC or sham mice. These results suggest that gut modifications represent an important element to be considered in the development and progression of cardiac dysfunction in response to TAC and support this animal model as a valuable tool to establish the role and mechanisms of gut-heart crosstalk in HF. Evidence arising in this field might identify new treatment options targeting gut integrity and microbiota components to face adverse cardiac events.

The main metabolic abnormalities promoting cardiac dysfunction are the resistance to the meta- bolic actions of insulin in heart tissue, compensatory hyperinsulinemia, and the progression of hyperglycemia. Two stages of diabetic cardiomyopathy are described: an early stage characterized by left ventric- ular hypertrophy and impaired diastolic function, and a late stage characterized by cardiac fibrosis and systolic dysfunction. The occurrence of HF in patients affected by DM significantly impacts on quality of life, manifesta- tion of new symptoms and worsening of existing symptoms, and on long-term prognosis. Notargettreatmentshavebeentestedindiabeticcardiomyopathy,thusclinicaltrialsareneededto define the role of available HF therapies or to find new therapeutic targets for this clinical condition.

A-kinase anchoring proteins (AKAPs) transmit signals cues from seven-transmembrane receptors to specific sub-cellular locations. Mitochondrial AKAPs, encoded by the akap1 gene, have been shown to modulate mitochondrial function and reactive oxygen species (ROS) productionin the heart. In the present study we hypothesized that akap1 might be crucial to preserve mitochondrial function and structure, and cardiac responses to myocardial ischemia. To test this, eight-week-old akap1 global knockout mice (akap1 -/-) of either gender or their wild-type (wt) littermates underwent myocardial infarction (MI) by permanent left coronary artery ligation. Age and gender matched mice of either genotype underwent a left thoracotomy without coronary ligation and were usedas controls (sham). Twenty-four hours after coronary ligation, despite similar areas-at-risk among the groups, akap1 -/-mice displayed larger infarct size. Seven days after MI, cardiac function and survival were significantly reduced in akap1 -/- compared to WT, while cardiac fibrosis was significantly increased. Akap1 deletion was associated with remarkable mitochondrial structural abnormalities at electron microscopy, increased ROS production and reduced mitochondrial function after MI. These alterations were associated with enhanced cardiac mitophagy and apoptosis. Autophagy inhibition by 3-methyladenine significantly reduced apoptosis and ameliorated cardiac dysfunction following MI in akap1 -/-mice. These results demonstrate that akap1 deficiency promotes cardiac mitochondrial aberrations and mitophagy, enhancing infarct size, pathological cardiac remodeling and mortality under ischemic conditions. Thus, mitochondrial AKAPs might represent important players in the development of post-ischemic cardiac remodeling and novel therapeutic targets.

I probiotici vengono proposti come modulatori dell’infiammazione sia del tratto gastro-enterico che vulvo-vaginale. In questo lavoro viene indagato l’effetto di Lactobacillus crispatus, ceppo potenzialmente probiotico, sulla risposta immune innata di cellule epiteliali HeLa nei confronti della Candida albicans e sulla modulazione dell’espressione dei Toll-like receptors (TLR), della produzione di β-defensine e di citochine. La C. albicans è coinvolta in disordini infettivi ed è riconosciuta come un patogeno emergente, causa di candidosi umane. La protezione dell’ospite contro l’infezione da C. albicans è complessa e include differenti “steps” del sistema di difesa immune. Diverse recenti ricerche hanno tentato di dimostrare il significativo ruolo giocato dall’immunità innata nel prevenire le infezioni da C. albicans delle mucose. Noi abbiamo studiato l’interazione tra C. albicans e cellule epiteliali umane usando monostrati di cellule HeLa in coltura. La C. albicans era capace di aderire alle cellule epiteliali ma, il numero di batteri diminuiva significativamente in presenza di L. crispatus. Quando le cellule epiteliali venivano messe a contatto con C. albicans, esse esprimevano alti livelli di TLR-2/4 correlati all’aumentata produzione di IL-8 e di β-defensine 2/3. Il pretrattamento delle cellule HeLa con L. crispatus determinava una diminuita virulenza della C. albicans come evidenziato dalla diminuita capacità di crescita e di aderenza nelle cellule epiteliali. In aggiunta si è dimostrato una “down-regulation” dell’espressione TLR-2/4 e della produzione IL-8 e β-defensine 2/3. In conclusione i nostri risultati mostrano che L. crispatus stimola le difese delle cellule epiteliali nell’infezione da C. albicans attraverso il coinvolgimento dell’espressione TLR-2/4, IL-8 e β-defensine 2/3 e quindi i risultati ottenuti supportano il potenziale probiotico di questo Lactobacillus come agente anti-infettivo soprattutto verso la C. albicans.

La parodontite è un'infezione polimicrobica multifattoriale caratterizzata da un processo infiammatorio distruttivo indotta da uno squilibrio tra la virulenza batterica e le difese dell'ospite. I batteri della placca sottogengivale possono invadere il dento-parodonto e passare ai tessuti più profondi, come il legamento parodontale e l'osso alveolare. Cellule umane del legamento parodontale (HPLC) contribuiscono alla risposta immunitaria nella cavità orale e possono produrre citochine che aumentano la risposta infiammatoria. Porphyromonas gingivalis, bacillo aerobio Gram-negativo è stato coinvolto come agente eziologico fondamentale nell'iniziazione e progressione della parodontite. Questo patogeno produce diversi fattori di virulenza che stimolano le cellule HPLC a produrre mediatori infiammatori e citochine proinfiammatorie. Questi mediatori ulteriormente attivano le cellule parodontali a costruire eccessive risposte infiammatorie dell'ospite, con conseguente progressione della malattia periodontale. Polifenoli naturali come il resveratrolo risultano avere proprietà battericide contro i batteri patogeni, inibendo la crescita di alcuni batteri e mostrando attività chemo-preventiva, anti-infiammatoria e antiossidante. Per simulare le condizioni in vivo, come quelli presenti nei siti di infezioni parodontali, nel presente studio abbiamo utilizzato un modello di cellule HPLC stimolate con lipopolisaccaride (LPS) di P. gingivalis. Per determinare se il resveratrolo può interferire con l’attività di LPS di P. gingivalis e può ridurre la produzione di molecole pro-infiammatorie, abbiamo esaminato l’effetto del batterio sulla produzione di citochine IL-1, IL-6, IL-8, IL-12 e TNF-α e sulla produzione di NO in cellule HPLC. I risultati ottenuti mostrano che il trattamento con il resveratrolo in cellule HPLC stimolate con lipopolisaccaride (LPS) di P. gingivalis induce una minore espressione di NO che è dose e tempo dipendente, correlata ad una maggiore efficienza di cellule HPLC infette e ad una diminuzione della produzione di citochine proinfiammatorie in cellule HPLC stimolate da LPS di P. gingivalis. Questi risultati suggeriscono la capacità del resveratrolo di determinare effetti immunomodulatori, potendo trovare quindi possibili applicazioni terapeutiche per il trattamento della parodontite.
