The aim of this study was to evaluate the association of different concentrations of Trolox® and the addition of a fixed concentration of DHA in the freezing of semen of Mangalarga Marchador stallions. The objective of this study was to determine the scrotal thermographic profile and to verify the influence of temperature and humidity of the humid tropical climate on testicular temperature and seminal quality of Mangalarga Marchador stallions. The thermal profiles of the proximal, middle, and distal zones of the testicles and total surface temperature (TSTT) were recorded using an FLIR E60bx thermal imager. The average air temperature (°C) and relative humidity (%) were obtained 1, 5, 9, 33, and 66 days before semen collection and showed a mean value of 26.5±2.4 and 80.4±6.0 respectively. The scrotal surface temperature was close to 34°C and there was no variation with the age of the stallion, reproductive activity, and characteristics of the ejaculate (P>0.05). Este trabalho foi realizado com objetivo de avaliar se o intervalo entre a inseminacao e ovulacao pode influenciar a taxa de prenhez e a razao sexual secundaria da prole de femeas zebuinas submetidas a inseminacao artificial em tempo fixo (IATF). Foram sincronizados 469 femeas, entre novilhas e vacas, alocadas aleatoriamente em tres grupos experimentais: IA42, IA54 e IA66, inseminadas respectivamente as 42, 54 e 66 horas apos a remocao do implante de progesterona. As taxas de concepcao diferiram entre as categorias ( p < 0,05), mas nao entre os tratamentos resultando em 28% e 45% no grupo IA42, 34% e 47% no IA54 e finalmente 33% e 48% no IA66 para novilhas e vacas respectivamente. Os resultados demonstraram que o momento da inseminacao nao alterou a taxa de concepcao nem a razao sexual secundaria das crias, o que permite maleabilidade no momento de inseminacao apos sincronizacao e a busca por alternativas para manipular a razao sexual.

RESUMO A utilização da gema de ovo dificulta a padronização de meios diluidores e apresenta riscos biológicos. Assim, este estudo avaliou diferentes concentrações da lipoproteína de baixa densidade (LDL), em substituição à gema de ovo, para a confecção de diluentes para criopreservação espermática em ovinos. Foram utilizados um diluidor controle (CTR= 20% de gema de ovo) e cinco tratamentos, substituindo-se a gema pelas diferentes proporções de LDL (T1=6%; T2=8%; T3=12%; T4=16%; T5=20%), todos à base de TRIS-glicerol. Para o estudo, utilizaram-se dois ejaculados, de seis reprodutores da raça Santa Inês. Sessenta dias após a criopreservação, as amostras foram descongeladas e avaliadas subjetivamente quanto à motilidade total (MT, %) e progressiva (MP, %), ao vigor (1-5) e à integridade funcional (choque hisposmótico com água destilada, %) e estrutural (corante supravital eosina, %) das membranas espermáticas. Knowledge about reproduction of white-lipped peccary is of great importance to assist with the conservation of this species and enable its rational use in captivity. This study aimed to evaluate the effect of ACP-103®, ACP-116® and BTS semen extenders on sperm viability during cooling of Tayassu pecari semen. Five ejaculates from four adult males were chilled. The animals were submitted to the protocols of sedation and anesthesia for semen collection by the electroejaculation method. After collection, the semen was macro- and microscopically assessed and diluted to reach 35x106 spermatozoa/mL in each of the three different extenders tested. The fresh-extended semen was packed in a BotuFLEX® thermal box to keep samples at 15°C for 24 hours. After cooling, the following semen parameters were analyzed: sperm motility, functional and structural integrity of sperm membranes, mitochondrial activity, chromatin condensation, and the thermoresistance test was performed. RESUMO Queixada (Tayassu pecari) é um mamífero neotropical, classificado como vulnerável devido à caça e à destruição de seu habitat. Em razão das dificuldades na reprodução em cativeiro, técnicas de reprodução assistida podem ser aplicadas em programas de repovoamento da espécie. Dessa forma, objetivou-se identificar o melhor protocolo de teste hiposmótico (HOST) para avaliar a integridade funcional dos espermatozoides de queixada. O sêmen de quatro machos adultos foi coletado com auxílio de eletroejaculador após contenção física e protocolo de sedação e anestesia. O sêmen foi avaliado quanto às características macro e microscópicas e diluído nas seguintes soluções hiposmóticas: água destilada (0mOsmol/L), sacarose (50, 100, 150mOsm/L) e frutose (50, 100, 150mOsm/L). Cada amostra foi incubada em duplicata, e uma sofreu fixação em solução de citrato de sódio formolizado a 4%. Duzentos espermatozoides foram avaliados por amostra e classificados em reativos ou não ao HOST. Todas as soluções testadas foram semelhantes em identificar o percentual de espermatozoides reativos, independentemente de a amostra ser ou não fixada (P>0,05). Dessa forma, pode-se usar água destilada como HOST por este apresentar resultados similares e por ser um teste mais barato.

Objetivou-se avaliar a motilidade, cinética e integridade das membranas de espermatozoides ovinos criopreservados em diluidores contendo 8% de LDL com antioxidantes enzimáticos em diferentes concentrações. Quatro carneiros da raça Santa Inês foram utilizados para formar quatro pools de sêmen (cada pool contendo ejaculados provenientes dos quatro carneiros, totalizando quatro ejaculados por animal). Cada pool de sêmen foi dividido em oito alíquotas para os seguintes tratamentos: 1) Tris- glicose-glicerol (TGG) + (16%) gema de ovo (controle 1); 2) TGG + 8% (g/L) LDL (controle 2); 3) TGG + 8% LDL + catalase 100 U/mL; 4) TGG + 8% LDL + catalase 200 U/mL; 5) TGG + 8% LDL + superóxido dismutase 100 U/mL; 6) TGG + 8% LDL + superóxido dismutase 200 U/mL; 7) TGG + 8% LDL + glutationa reduzida 5 mM; and 8) TGG + 8% LDL + glutationa reduzida 10 mM. As amostras foram envasadas em palhetas de 0,25 mL, resfriadas (-0,25 °C/min), mantidas a 5 °C por duas horas e em seguida congeladas (-25 °C/ min) usando uma máquina de congelar TK4000 ® . Resumo A ovulação em gatas é induzida por um reflexo neuroendócrino atribuído à estimulação mecânica dos receptores sensoriais durante o coito. Esta estimulação pode ser simulada com auxílio do swab vaginal, desencadeando a pseudogestação. Objetivou-se verificar a eficiência da indução de ovulação com swab, a fim de estabelecer um tratamento contraceptivo natural para felinos domésticos, bem como os efeitos sobre o útero do uso repetido dessa técnica. Na primeira fase do trabalho, foram avaliados 12 animais em três ciclos estrais consecutivos. No primeiro ciclo (T1), houve estimulação vaginal com swab. No segundo ciclo (T2), foi utilizado macho vasectomizado para cópula. No último ciclo (T3), a ovulação foi acompanhada sem estímulo (controle). Na segunda etapa do trabalho, 13 gatas foram submetidas a sucessivos estados de pseudogestação com intuito de verificar os efeitos da estimulação mecânica sobre o útero. A confirmação da ovulação em todas as etapas do trabalho foi realizada por meio da mensuração dos níveis de progesterona. A estimulação vaginal com swab apresentou resposta similar à obtida por monta natural (P>0,05). Algumas gatas apresentaram modificações uterinas discretas; no entanto, nenhum desses achados foi considerado de relevância patológica. Desta forma, a indução de ovulação com swab mostrou-se segura e sem efeitos colaterais expressivos. Abstract A neuroendocrine reflex attributed to the mechanical stimulation of sensory receptors during copulation induces ovulation in cats. This stimulation can be simulated with a vaginal swab, triggering pseudopregnancy. The efficiency of ovulation induction with swab was evaluated in order to establish a natural contraceptive treatment for domestic cats as well as to analyze the effect on uterus of the repeated use of this technique. At the first phase of the study, 12 animals were evaluated in three consecutive estrous cycles. In the first cycle (T1), vaginal swab stimulation was applied. In the second cycle (T2), a vasectomized male was used. In the third cycle (T3), ovulation was accompanied unstimulated (control). At the second phase of the study, 13 female cats were induced to successive pseudopregnancy states to check the effects on the uterus. Confirmation of ovulation in the two phases of the study was carried out by measuring progesterone serum levels. The vaginal swab stimulation showed a similar response to that obtained by natural mating (P>0.05). Some cats showed mild uterine changes without pathological relevance. Thus, ovulation induction with swab is secure and without significant side effects.

The present study evaluated the effect of storage conditions on the LDL efficacy for cryopreserving ovine sperm. In this way, we compared egg yolk extender with three different forms for LDL storing, LDL diluted in Tris-glucose extender and stored in frozen (i) and freeze-dried (ii) states and LDL stored pure and added into the extender prior to use (iii). We also tested the effect of two storage temperatures (−20 and −80 °C) and three storage times (30, 60, 120 d). Frozen and freeze-dried extenders containing LDL, as well as LDL stored pure, improved post-thaw sperm quality. Storage temperatures did not influence negatively the cryoprotectiveness of LDL extenders. Furthermore, lyophilised LDL extenders stored at −20 °C were more effective in preserving sperm longevity than the other extenders stored at −20 °C. Finally, LDL extenders stored for 30 and 120 d were more efficient than 60 d in preserving ram sperm freezability.

As preguiças-de-coleira (Bradypus torquatus) são mamíferos arborícolas da família Bradypodidae. Podem ser encontradas nos trechos de Mata Atlântica do Brasil e a maior diversidade genética de suas populações ocorre em matas do sul da Bahia. A observação desses animais na natureza é muito difícil, pois passam a maior parte da vida escondidos no denso emaranhado das copas, por isso, dados sobre aspectos reprodutivos são escassos e não existem informações sobre ciclo estral dessa espécie. Este trabalho teve por objetivo identificar as células do epitélio vaginal da preguiça-de-coleira (Bradypus torquatus) como forma de viabilizar o uso dessa técnica para estudar as fases do ciclo estral desses animais. As amostras para citologia vaginal foram obtidas de quatro preguiças de coleira que habitavam áreas de Mata Atlântica do sul da Bahia. Após captura manual do animal, procedeu-se a coleta de material biológico, introduzindo uma escova ginecológica estéril, na comissura dorsal da vulva. Para cada amostra foram feitos dois esfregaços rotacionando a extremidade da escova sobre cada lâmina de vidro, fazendo-se em geral três impressões lineares. O esfregaço foi imediatamente corado pelo método Panótico rápido (Laborclin®). Nas preguiças BT033, BT065 e BT042 foi possível identificar respectivamente 30%, 33% e 7% de células parabasais (PB); 56%, 22% e 10% de células intermediárias pequenas (IP); 6%, 18% e 6% de células intermediárias grandes (IG); 2%, 13% e 24% de células superficiais nucleadas (SN); 6%, 14% e 53% de células superficiais anucleadas (SA). Na preguiça BT464 foi possível fazer duas coletas com intervalo de 13 meses. Os dados da primeira e segunda coleta foram, respectivamente: 6% e 17,5 de células PB, 5% e 25% de células IP, 11% e 15,5% de células IG, 8% e 19,5% de células SN e 70% e 22,5% de células SA. Enfatiza-se que as porcentagens de células do epitélio vaginal variaram entre indivíduos e também na mesma preguiça. Isto sugere que a citologia vaginal possa ser uma ferramenta de avaliação do ciclo estral em preguiça-de-coleira. Maned sloths (Bradypus torquatus) are arboreal mammals of the family Bradypodidae. They can be only found in the Atlantic coast forest of Brazil and its most genetically diverse populations occur in forests of southern Bahia. The observation of these animals in the wild is very difficult as they spend most of their lifetime hidden in the dense forest canopy. Data on their reproductive aspects are scarce, and there is none information about their estrous cycle. This research aimed at identifying the vaginal epithelial cells of maned sloths (Bradypus torquatus) as a possible way to study the phases of the estrous cycle of this animal. The samples for vaginal cytology were obtained from four free ranging maned sloths living in a protected area of coastal forest in the South of Bahia. The sterile gynecological brush was inserted up to the necessary distance to reach the pelvic channel. For each sample two smears were made by rotating the tip of the brush onto each glass slide, producing in general three linear impressions. Staining was performed using rapid Panotic Kit (Laborclin®). Maned sloths BT033, BT065, and BT042 presented, respectively, 30%, 33%, and 7% of parabasal epithelial cells (PB); 56%, 22%, and 10% of small intermediate cells (IP); 6%, 18%, and 6% of large intermediate cells (IG); 2%, 13%, and 24% of superficial epithelial cell with a nucleus (SN); 6%, 14%, and 53% of anucleated superficial epithelial cell (AS). Two cell samples were collected for maned sloth BT464 with a 13 months interval. Cytological differences were observed between the two samples (1st and 2nd): 6% and 17,5% of PB cells, 5% and 25% of IP cells, 11% and 15,5% of IG cells, 8% and 19,5% of SN cells and 70% and 22,5% of AS cells, respectively. It's interesting to remark that the percentage of vaginal epithelial cells varied among sloths and also for the same animal. This result suggests that vaginal cytology of maned sloth can be used as a tool to evaluate of estrous cycle.

Estudou-se o efeito de diferentes concentrações de gema de ovo, açúcares e tampões nos diluidores sobre a viabilidade espermática pós-descongelação de sêmen eqüino. Os diluidores foram: D1 = 10% de gema de ovo e acréscimo de 50% de lactose e glicose-EDTA em relação ao D5; D2= 10% de gema de ovo e redução de 50% de lactose e glicose-EDTA em relação ao D5; D3= 20% de gema de ovo e acréscimo de 50% de lactose e glicose-EDTA em relação ao D5; D4= 20% de gema de ovo e redução de 50% de lactose e glicose-EDTA em relação ao D5; D5= 20% de gema de ovo e composição de lactose e glicose-EDTA segundo Martin et al. (1979). Cinco ejaculados de seis garanhões foram utilizados para testar os diluidores contendo diferentes agentes crioprotetores: 3,5% de etilenoglicol e 5% de acetamida. Após diluição final nos diferentes diluidores, o sêmen foi submetido a diferentes protocolos de congelação: com e sem resfriamento prévio. Após descongelação em banho-maria a 75ºC, avaliaram-se a motilidade, o vigor, e a integridade estrutural e funcional das membranas espermáticas. A redução na concentração de gema de ovo de 20% para 10%, nos diluidores contendo a mesma concentração de açúcares e tampões, não alterou a capacidade crioprotetora dos diluidores com etilenoglicol ou acetamida. O aumento na concentração de açúcares e tampões no diluidor lactose-EDTA-gema de ovo não acrescentou efeito crioprotetor ao etilenoglicol e acetamida. Diluidores com maior osmolaridade tenderam a preservar melhor o sêmen eqüino congelado com etilenoglicol ou acetamida. The objective was to evaluate: (1) the influence of the length of an equilibrium period done at room temperature on cooled semen diluted in extender containing different low density lipoproteins (LDL) concentration in replacement of whole egg yolk, (2) the effect of different concentration of LDL in replacement of egg yolk in extenders for freezing ram sperm and (3) the influence of the length of an equilibrium period after cooling to 5 °C on maintenance of post-freezing sperm viability. In Experiment I, semen samples were diluted in extender containing 15% egg yolk or 2, 4 or 8% LDL (v/v) in replacement of the egg yolk. Diluted semen was subjected to 0, 30, 60, or 120 min of equilibrium at room temperature and thereafter cooled in the refrigerator (−0.13 °C/min) to reach 5 °C in 2.5 h. Motility assessments were performed on re-warmed samples (38 °C) at 1 h interval for 3 h; the percentage of sperm with functional integrity of the plasma membrane was determined based on a hypoosmotic swelling test (HOST). There was no interaction (P > 0.05) between extenders and duration of pre-cooling equilibrium for the evaluated characteristics. The whole yolk extender preserved motility similar to 8% LDL (P > 0.05) and better preserved the motility than 2 or 4% LDL extenders (P < 0.05). The plasma membrane integrity was evenly preserved among extenders (P > 0.05). In Experiment II, extenders of Experiment I were used to freeze ram semen immediately after final dilution, with neither pre nor post cooling equilibrium period. The following parameters were evaluated after thawing (38 °C/30 s): sperm longevity (incubation 38 °C/3 h), functional and structural integrity of sperm membranes (HOST and fluorescent probes) and kinetics of sperm by computer analysis (CASA). Extenders with 4% LDL or 8% LDL had similar (P > 0.05) responses than that obtained with whole egg yolk for all evaluated parameters. The 2% LDL extender was less efficacious (P < 0.05). In Experiment III, semen samples diluted in whole yolk, 4% LDL or 8% LDL were allowed to equilibrate after cooling to 5 °C for 1 or 2 h before freezing. After thawing (38° C/30 s), samples were subjected to the same tests as in Experiment II. Equilibration for 2 h increased (P < 0.05) post thaw sperm motility (regardless of the extender). In conclusion, using ram semen, LDL (4% or 8%) replaced effectively whole egg yolk (15%), maximizing its effectiveness when equilibration length at 5 °C before freezing was of 2 h.

The aim of this study was to test different concentrations of low density lipoprotein (LDL) in replacement of whole egg yolk in extenders preserved in the aqueous or lyophilized form, for ram sperm cryopreservation using two cooling curves (-40°C/min from 5 to –140°C and nitrogen vapor). One ejaculate from six Santa Ines rams was collected. Each ejaculate was divided into nine different diluents as follows: Tris-16% yolk (control), and Tris with 2, 4, 6 and 8% fresh LDL, and criopreserved in the aqueous or lyophilized form. The samples were diluted to a final concentration of 100 x 106 sperm/mL and filled into 0.25 ml straws. After thaw, sperm cells were evaluated for motility and sperm kinetics (CASA), and submitted to the hypoosmotic swelling test and the evaluation of the structural integrity of sperm membranes using fluorescent dyes (CFDA: PI), as well as sperm morphology and longevity. The experimental design was randomized blocks, and results were submitted to ANOVA and the averages were compared using the Scott-Knott test. There were no differences in progressive motility and functional and structural integrity of the membrane evaluated when different concentrations of aqueous or lyophilized low density lipoproteins or egg yolk were added to the extender (P>0.05). As for the velocity of sperm movement, the control medium had some kinetic scores similar to the extender containing LDL, both aqueous and lyophilized (P> 0.05). Results were similar between cooling curves. Therefore, we conclude that the media containing all concentrations of LDL, aqueous or lyophilized, were able to protect the ram sperm cells during the cryopreservation process, as whole egg yolk did.

Egg yolk is the most widely used cryoprotectant in the composition of extenders for cryopreservation of mammalian spermatozoa; yet, efforts have been made to find ways to substitute it due to the possibility of transporting pathogenic microorganisms, the lack of standardization, and the presence of substances that inhibit metabolic exchanges or decrease the motility of sperm. The protective effect of egg yolk was attributed to the presence of low density lipoproteins (LDL) that prevent cholesterol efflux by increasing membrane stability and resistance to low temperatures. (Moussa et al. 2002 Theriogenology 57, 1695–1706) demonstrated that the purification of LDL is possible, thereby allowing its use as a replacement for integral egg yolk in extender. Then, this study aimed to evaluate the effect of substitution of egg yolk by different LDL concentrations during cryopreservation of ram semen. A total of 6 sheep, 3 ejaculates per animal, were collected. After collection, the ejaculates were evaluated and diluted in different extenders: E1) Tris, glucose, 15% egg yolk, and 5% glycerol; E2) Tris, glucose, 5% LDL (Moussa et al. 2002 Theriogenology 57, 1695–1706), and 5% glycerol; E3) Tris, glucose, 10% LDL, and 5% glycerol; and E4) Tris, glucose, 20% LDL, and 5% glycerol. The samples were adjusted to a final concentration of 600 × 106 sptz mL–1 and filled into 0.25-mL straws and frozen in a TK 4000® machine. After thawing, sperm motility and spermatic vigor were evaluated, and the test of thermo-resistance (TTR) was conducted. Functional and structural integrity of the spermatic membrane were evaluated through the hypoosmotic test and the use of fluorescent dyes. The kinetic parameters of sperm were assessed by the computerized system (CASA). The statistical analysis was performed using the statistical program SAS (Statistical Analysis System), and the averages were compared using the Duncan multiple test. Este trabalho teve como objetivos avaliar a eficiência de duas formas de adição da dimetilformamida a 5%, utilizando-se como meio diluidor base o INRA 82 modificado, e de duas curvas de congelamento na preservação espermática eqüina, avaliada in vitro. Objetivou-setambém verificar o grau de lesão dos espermatozóides durante as diversas etapas do processo de criopreservação. Um ejaculado de nove garanhões foi utilizado para testar quatro tratamentos: adição do crioprotetor, em única etapa, sem resfriamento prévio (T1- controle); adição do crioprotetor, em múltiplas etapas, sem resfriamento prévio (T2); adição do crioprotetor, em única etapa, com resfriamento prévio (T3); e adição do crioprotetor, em múltiplas etapas, com resfriamento prévio (T4). O sêmen foi envasado em palhetas de 0,5 mL e congelado três centímetros acima da lâmina do nitrogênio líquido e posteriormente submerso no mesmo. O descongelamento foi realizado a 52°C por dez segundos, seguido da imersão em banho-maria a 37°C por mais trinta segundos. Após a adição do crioprotetor, em única ou múltiplas etapas,foram avaliados os parâmetros espermáticos de motilidade espermática total, progressiva e vigor do movimento espermático, em microscopia óptica (aumento de 400X). A integridade funcional da membrana plasmática foi avaliada por meio do teste hiposmótico. Após o descongelamento, foram avaliados os parâmetros de motilidade espermática total, motilidade espermática progressiva, vigor do movimento espermático e integridade estrutural das membranasespermáticas por meio de sondas fluorescentes (carboxifluoresceína e iodeto de propídeo). Os espermatozóides foram submetidos ao teste de termo-resistência. Imediatamente após a adição do crioprotetor, em única ou múltiplas etapas, não foram observadas diferenças entre ostratamentos (p>0,05). Após o descongelamento, foi observada superioridade da adição do crioprotetor, em múltiplas etapas, em todos os parâmetros avaliados, independentemente da curva de congelamento adotada, com ou sem resfriamento controlado prévio (p
