Hepatitis E virus (HEV) is recognized as an emerging zoonosis. Pigs and wild boars are considered the main reservoirs of zoonotic HEV-3 and HEV-4 genotypes. In Europe, autochthonous human cases of hepatitis E, mainly associated with HEV-3 and consumption of raw or undercooked pig and wild boar liver/meat, have increased over the last decades. From 2016 to 2024, during several hunting seasons, we conducted a molecular and serological longitudinal survey on the circulation of HEV in Maremman wild boar (Italian subspecies/ecotype, Sus scrofa majori) and fallow deer (Dama dama) populations in a protected area in Central Italy. During the study period, 346 livers (256 from wild boar, 90 from fallow deer), 161 serum (127 from wild boar, 34 from fallow deer), and 23 meat juice (11 from wild boar, 12 from fallow deer) samples were collected. Serum and meat juice samples were tested using a commercial ELISA test for the detection of total anti-HEV antibodies. An estimated serological prevalence of 28.3% (39/138) in wild boar and 21.7% (10/46) in fallow deer was found. The 346 liver samples were tested using a HEV Real-Time RT-PCR for the detection of HEV-RNA. Thirty-one wild boar (12%) and four fallow deer (4.4%) livers were found positive. Phylogenetic analysis of 11 partial ORF2 sequences from wild boar confirmed the HEV3 heterogeneity in this species, revealing different strains (3f, 3c) circulating over the years. The detected subtypes are among the most commonly detected in Italy and our strains showed a high correlation with human and wild boar Italian strains. Although the studied area is a fenced natural reserve, the presence of different strains over time suggests the probable virus introduction from the external. Our results confirm fallow deer susceptibility to the infection, and that wild boar could be considered the main wild HEV reservoir. This is also the first study demonstrating the infection in the so-called Italian subspecies/ecotype Maremman wild boar. Moreover, our results corroborate that the consumption of undercooked or raw liver from both wild boar and fallow deer, or the direct contact with these animals, could represent a zoonotic risk.

Abstract Background To date, there is a scarcity of information and literature on Macaca maura health status relative to viral diseases. The objectives of the present study were to investigate on the potential spread of enteric and non-enteric viruses shed in the environment through a wild macaque feces and to understand the possible interrelation in the spread of zoonotic viruses in a poorly studied geographical area, the Sulawesi Island. This study will also contribute providing useful information on potential threats to the health of this endangered species. Methods The sampling was conducted between 2014 and 2016 in the Bantimurung Bulusaraung National Park, in the south of the Sulawesi Island and non-invasive sampling methods were used to collect fresh stools of the M. maura, one of the seven macaque species endemic to the island of Sulawesi, Indonesia. The population under study consisted in two wild, neighboring social macaque groups with partially overlapping home ranges; twenty-four samples were collected and examined using negative staining electron microscopy and a panel of PCR protocols for the detection of ten RNA and two DNA viruses. Results Viral particles resembling parvovirus (5 samples), picornavirus (13 samples) and calicivirus (13 samples) were detected by electron microscopy whereas the PCR panel was negative for the 12 viruses investigated, except for one sample positive for a mosquito flavivirus. The results did not correlate with animal sex; furthermore, because all of the animals were clinically healthy, it was not possible to correlate feces consistency with viral presence. Conclusions As information on viral infections in wild moor macaques remains limited, further studies are yet required to identify the fecal–oral and blood transmitted potentially zoonotic viruses, which may infect the moor macaque and other macaque species endemic to the South Sulawesi Island.

Salmonella is a Gram-negative enteric bacterium responsible for the foodborne and waterborne disease salmonellosis, which is the second most reported bacterial zoonosis in humans. Many animals are potential sources of salmonellosis, including dogs, cats, and other pets. We report the case of an outbreak of salmonellosis in a family in central Italy, affecting two children and involving their three dogs as carriers. One of the children needed medical care and hospitalisation. Isolation and analysis of stool samples from the sibling and the animals present in the house were carried out. Serotyping allowed the identification of S. enterica subsp. enterica serovar Typhimurium in its monophasic variant for all the isolates. The results of whole-genome sequencing confirmed that the strains were tightly related. The minimum inhibitory concentration (MIC) test documented the resistance to ampicillin, sulfamethoxazole, and tetracycline. The origin of the zoonotic outbreak could not be assessed; however, the case study showed a clear passage of the pathogen between the human and non-human members of the family. The possibility of a transmission from a dog to a human suggests the need for further studies on the potential ways of transmission of salmonellosis through standard and alternative feed.

Community seedbanks promote conservation and the use of crop genetic diversity, as well as supporting farmer seed systems. This study analyses seed flow and access to crop genetic diversity over time in the Nakaseke, Rubaya, and Kibuga seedbanks of Uganda. The modes of operation of the banks were compared through scrutinizing records of crops and varieties being conserved, quantities of seed distributed, to whom, and quantities returned. The Nakaseke seed bank distributed the highest varietal diversity of common bean (Phaseolus vulgaris) and groundnuts (Arachis hypogaea L.), whereas the Rubaya seedbank distributed the highest quantity of common bean seed, followed by the Kibuga seedbank. There were no significant differences between the type of variety of seed, quantities of seed accessed, and seed returned to the seedbanks by women and men—except for the Nakaseke seedbank, where women returned significantly higher quantities of common bean seed. The Kibuga and Rubaya seedbanks dealt with individual farmers, whereas the Nakaseke seedbank dealt with individual farmers and groups. The extent to which core functions were achieved by a particular seedbank depended on the mode of operation, including actors, management, degree of development, socio-economic setting, among others. Further research is recommended to unpack these factors and come up with the most appropriate combinations for greater seedbank effectiveness.

Foodborne diseases (FBDs) represent a worldwide public health issue, given their spreadability and the difficulty of tracing the sources of contamination. This report summarises the incidence of foodborne pathogens and toxins found in food, environmental and clinical samples collected in relation to diagnosed or suspected FBD cases and submitted between 2018 and 2020 to the Food Microbiology Unit of the Istituto Zooprofilattico Sperimentale del Lazio e della Toscana (IZSLT). Data collected from 70 FBD investigations were analysed: 24.3% of them started with an FBD diagnosis, whereas a further 41.4% involved clinical diagnoses based on general symptomatology. In total, 5.6% of the 340 food samples analysed were positive for the presence of a bacterial pathogen, its toxins or both. Among the positive samples, more than half involved meat-derived products. Our data reveal the probable impact of the COVID-19 pandemic on the number of FBD investigations conducted. In spite of the serious impact of FBDs on human health and the economy, the investigation of many foodborne outbreaks fails to identify the source of infection. This indicates a need for the competent authorities to continue to develop and implement a more fully integrated health network.

Botulism in cattle is rarely reported in Italy. This study describes an outbreak of botulism in a dairy herd in Central Italy in September 2012, and the notably high mortality rate it caused. Differential diagnoses involving toxicology and bacteriology, and electrolyte imbalances, all proved negative. A multiplex polymerase chain reaction (PCR) for detecting the BoNT gene led to the identification of the causative agent as Clostridium botulinum type DC. The presence of the toxin was confirmed subsequently via mouse bioassay. Initially, the peracute deaths and ambiguous clinical signs delayed the diagnosis and, as a result, impeded identification of the source of the infection on the farm. The severity of the outbreak demonstrates that screening for animal botulism should always form part of the diagnostic protocols used to investigate sudden peracute deaths without apparent cause in livestock.

El objetivo de la investigación fue determinar la reacción de 14 genotipos de musáceas frente a Mycosphaerella fijensis, agente causal de la Sigatoka negra. Se condujeron dos experimentos en el Departamento de Protección Vegetal de la Estación Experimental P ichilingue del Instituto Nacional de Investigaciones Agropecuarias (INIAP), Ecuador. Los genotipos evaluados fueron: banano Orito (AA), Guineo de jardín, Williams, Filipino, Gros Michel, Mulato y Morado, del genoma (AAA), y los plátanos Limeño, Maqueño, Do minico, Dominico - Hartón, Dominico gigante, Dominico negro y Barraganete, del genoma AAB. Se usaron dos métodos de inoculación: a) in vitro sobre fragmentos de hoja con una suspensión de conidios y b) inoculación de pl á ntulas de ocho semanas de edad con una suspensión micelial. En el primer caso se midió el periodo de incubación (PI) y el número de lesiones a los 14 y 20 días después de la inoculación (DDI). En invernadero, se registró el PI, tiempo de evolución de síntomas (TES) y tiempo de desarrollo de la enfermedad (TDE). Se determinaron diferencias estadísticas significativas (p ≤ 0 , 05) para todas las variables evaluadas; los cultivares Orito (AA) y Limeño (AAB) calificaron como resistentes, mientras que todos los demás fueron susceptibles, aunque mostra ron un rango de variabilidad en la respuesta a las inoculaciones .

In Europe, foodborne transmission has been clearly associated to sporadic cases and small clusters of hepatitis E in humans linked to the consumption of contaminated pig liver sausages, raw venison, or undercooked wild boar meat. In Europe, zoonotic HEV-genotype 3 strains are widespread in pig farms but little information is available on the prevalence of HEV positive pigs at slaughterhouse. In the present study, the prevalence of HEV-RNA positive pigs was assessed on 585 animals from 4 abattoirs located across Italy. Twenty-one pigs (3.6%) tested positive for HEV in either feces or liver by real-time RT-PCR. In these 21 pigs, eight diaphragm muscles resulted positive for HEV-RNA. Among animals collected in one abattoir, 4 out of 91 plasma tested positive for HEV-RNA. ELISA tests for the detection of total antibodies against HEV showed a high seroprevalence (76.8%), confirming the frequent exposure of pigs to the virus. The phylogenetic analyses conducted on sequences of both ORF1 and ORF2 fragments, shows the circulation of HEV-3c and of a novel unclassified subtype. This study provides information on HEV occurrence in pigs at the slaughterhouse, confirming that muscles are rarely contaminated by HEV-RNA compared to liver, which is the most frequently positive for HEV.

Banana and plantain (Musa spp.) are very important crops in Ecuador. Agricultural production systems based on a single cultivar and high use of external inputs to increase yields may cause changes in the landscape structure and a loss in biodiversity. This loss may be responsible for a decrease in the complexity of arthropod food webs and, at the same time, related to a higher frequency and range of pest outbreaks. Very little is known either about the ecological mechanisms causing destabilization of these systems or the importance of the diversity of natural enemies to keep pests under control. Few studies have focused on this issue in tropical ecosystems. Here, we address this problem, comparing two Musa-based agroecosystems (monocultivar and mixed-species plantations) at two sites in Ecuador (La Maná and El Carmen) with different precipitation regimes. The diversity of soil macro fauna, represented by arthropods, was established, as indicators of the abovementioned disturbances. Our ultimate goal is the optimization of pest management by exploring more sustainable cropping systems with improved soil quality. Arthropod abundance was higher in the mixed system at both localities, which was clearly associated with the quality of the soils. In addition, we found Hymenoptera species with predatory or parasitic characteristics over the pests present in the agroecosystems under study. These highly beneficial species were more abundant at the locality of La Maná. The mixed type of production system provides plant diversity, which favors beneficial arthropod abundance and permits lower agrochemical application without yield penalties in comparison to the monoculture. These findings will help in the design of Musa-based agroecosystems to enhance pest control.

The aim of this study was to determine the prevalence of Shiga toxin-producing Escherichia coli in fresh beef marketed in 2017 in 13 regions of Italy, to evaluate the potential risk to human health. According to the ISO/TS 13136:2012 standard, 239 samples were analysed and nine were STEC positive, from which 20 strains were isolated. The STEC-positive samples were obtained from Calabria (n = 1), Campania (n = 1), Lazio (n = 2), Liguria (n = 1), Lombardia (n = 1) and Veneto (n = 3). All STEC strains were analysed for serogroups O26, O45, O55, O91, O103, O104, O111, O113, O121, O128, O145, O146 and O157, using Real-Time PCR. Three serogroups were identified amongst the 20 strains: O91 (n = 5), O113 (n = 2), and O157 (n = 1); the O-group for each of the 12 remaining STEC strains was not identified. Six stx subtypes were detected: stx1a, stx1c, stx2a, stx2b, stx2c and stx2d. Subtype stx2c was the most common, followed by stx2d and stx2b. Subtype stx2a was identified in only one eae-negative strain and occurred in combination with stx1a, stx1c and stx2b. The presence in meat of STEC strains being potentially harmful to human health shows the importance, during harvest, of implementing additional measures to reduce contamination risk.

Human campylobacteriosis remains the most commonly reported gastrointestinal disease in Europe and Campylobacter (C.) jejuni and C. coli are the two species most frequently involved in such foodborne disease. Based on the sampling plan established in the region of Lazio (Central Italy) the aim of our work was to investigate the occurrence of Campylobacter spp. in poultry meat preparations collected by the local veterinary authority at retail shops and processing plants. We also observed whether various factors such as animal species or type of product affected the isolation rate. Occurrence was significantly lower than previous surveys (12/209, 5.7%) and chicken meat was more contaminated than turkey meat.

In recent years, the incidence of foodborne diseases caused by Shiga toxin-producing Escherichia coli (STEC) has increased globally. For this reason, within the specific regional control plan for the detection of STEC in food products in Italy, the presence of STEC in unpasteurised milk cheeses was investigated. In total, 203 samples obtained from March 2011 to December 2013 were analysed, with two standard methods (ISO 16654:2001 and ISO 13136:2012). Two strains of E. coli O157 were isolated (2/161, 1.2%) but did not carry any virulence-associated genes and 22 stx-positive samples (22/146, 15.1%) were detected in enrichment cultures, mostly from ovine cheeses. Only two strains isolated from different ovine cheeses carried stx gene and none of these was eae-positive. This study confirms the presence of stx-positive E. coli and suggests that this type of food cannot be excluded as a potential vehicle of STEC.

During the wild boars hunting season (2012-2013), 6508 adults were shot in Lazio Region, Viterbo territory. Out of the 6508, 393 subjects were sampled to be tested for the detection of Salmonella spp., Yersinia enterocolitica, E. coli VTEC and Trichinella spp. For this purpose, diaphragm and skeletal muscles samples were collected after the shooting, and tested at the Istituto Zooprofilattico Sperimentale Lazio and Toscana laboratories, Rome, Italy. Samples were tested respectively by polymerase chain reaction, enzyme linked immunofluorescent assay, microbiological and parasitological assays. The positivity percentages are 3.6% for Salmonella spp., 14.8% (out of 230 samples tested) for Yersinia enterocolitica, 4.6% for E.coli VTEC (vtx 1 and 2 genes). Each positive sample for vtx 1 and 2 was tested to isolate Verotoxigenic E.coli strains; none of them was positive for the same virulence genes. All diaphragm samples were negative to the trichinoscopic test. Every bacterial strain was identified serologically by seroagglutination test.
