Brachypodium distachyon was proposed as new model plant for grasses because of its small genome and its phylogenetic position between rice and Triticeae crops. Since, a range of genetic and genomic resources has been developed and the whole genome sequence is now available. We use Brachypodium as a model system for studying how to modify grass crops for increased biofuel production made from lignocellulosic cell walls. We are involved in developing many resources for Brachypodium: TILLING collection, natural accessions, different methods of phenotyping, and tools and method for gene transfer via Agrobacterium tumefaciens. Some lignin mutants from the monolignol biosynthesis pathway were already identified by phenotyping and TILLING, and their functional analysis is underway notably through complementation by overexpression of concerned gene in mutant background.

La transformation via Agrobacterium et la régénération par embryogenèse somatique est la méthode la plus couramment utilisée pour l'obtention de cotonniers transgéniques. Bien que le cotonnier soit au quatrième rang des plantes transgéniques cultivées dans le monde, en terme de surface, l'obtention de cotonniers transformés est encore un processus qui présente plusieurs limites majeures. Dans la plupart des méthodes publiées, le délai de régénération est encore le plus souvent de 8-10 mois auxquels il faut ajouter 5 à 6 mois pour obtenir les graines T1. Le taux de conversion des embryons somatiques en plantules est faible et on observe sur les plantes transférées en sol, différentes variations (développement floral, stérilité totale ou partiel) induites par la culture in vitro. Différentes études ont été conduites pour améliorer les performances du procédé de régénération de cotonniers transformés: elles portent sur le choix de la souche d'A. tumefaciens, la séquence de milieux de culture, les conditions de développement des embryons somatiques. Les rendements des différentes étapes depuis l'inoculation des explants par la souche d'Agrobacterium jusqu'à l'obtention de graines issues des transformants primaires sont présentés. Les cultures embryogènes sont anergiées et peuvent se multiplier pendant de nombreux mois sur des milieux dépourvus de substances de croissance. Une différence très significative du taux d'AIA endogène des cals primaires et des souches embryogènes a été mise en évidence et permet d'expliquer le caractère anergié de ces cultures. Le procédé est largement génotype dépendant et nous travaillons avec une variété, la variété Coker 310, qui présente de bonnes potentialités de régénération. Le matériel végétal est constitué d'explants d' hypocotyles issus de germinations réalisées en conditions aseptiques. Bien qu'il s'agisse de lignées pures, au sens du sélectionneur, nous observons une forte variabilité de réponse entre lots de graine selon leur origine ainsi qu'au sein d'un même lot. Une sélection effectuée sur la base des performances en embryogenèse a permis de réduire cette variabilité et d'augmenter le taux d'embryogenèse. De plus, il a été possible, en sélectionnant les graines utilisées, d'obtenir la néoformation d'embryons sur des milieux dépourvus de substances de croissance.

La cellulose est un polymère semi-cristallin naturel, renouvelable et biodégradable issue principalement des fibres de coton et de bois. Cependant, son utilisation pour des applications techniques est limitée par la structure spécifique des fibres. En effet, dans son état naturel, la cellulose ne peut être fondue comme les polymères thermoplastiques. Une étape de dissolution des fibres est nécessaire avant la mise en oeuvre sous forme de fil, de film ou encore de dérivés tels que les colles. Cette étude traite des mécanismes de gonflement et de dissolution des fibres de cellulose. Le gonflement et la dissolution des fibres de cellulose dépendent fortement de la qualité du solvant utilisé. Cinq principaux modes de gonflement et de dissolution peuvent être identifiés en fonction de la qualité du solvant dans lequel la fibre est plongée [1]. Plusieurs explications ont été proposées pour décrire le phénomène de gonflement par ballonnement observé dans les solvants modérés (mode 2 et 3). Selon Ott et al. [2], lors du gonflement dans un solvant modéré, la paroi primaire (paroi externe) de la fibre, considérée comme peu soluble, se déchire et glisse le long de la fibre pour former des zones où la fibre ne peut gonfler. Afin d'illustrer cette hypothèse, nous avons réalisé des observations microscopiques à haute résolution qui révèlent très clairement la présence de trois régions (Voir figure ci-dessous). En étudiant le gonflement et la dissolution de fibres de coton à différents stades de maturité (de l'élongation de la paroi primaire au dépôt de la paroi secondaire), nous avons montré que la dissolution de la paroi primaire est très difficile à obtenir même dans de bons solvants. A des stades de maturité plus avancés, la paroi secondaire déposée (paroi interne) se dissout par l'intérieur et gonfle. La paroi primaire se déchire pour former les sections non gonflées et les hélices, et limite donc le gonflement et la dissolution de la fibre. Cette proposition de mécanisme est illustrée par des observations microscopiques à haute résolution et des images en microscopie électronique à balayage. Après élimination de la paroi primaire par traitement enzymatique, la dissolution des fibres de cotons et de bois débute par un gonflement homogène et non par ballonnement. Les sections non gonflées et les hélices ne sont plus visibles. Cette étude révèle la difficulté à dissoudre les fibres de cellulose naturelles du fait de leur structure spécifique et montre le potentiel des nouvelles méthodes d'activation des fibres de cellulose par traitement enzymatiques pour faciliter la dissolution. One of the limitations to the use of cellulose is the difficulty involved in the processing and fabrication because of the specific structure of the fibres. In most cases, cellulose cannot be used in its natural form. It has to be dissolved. The focus of this paper is to study the swelling and dissolution mechanisms of cellulose fibres. The swelling and dissolution mechanisms of cellulose fibres strongly depend on the quality of the solvent. Five dissolution modes can be observed when a free floating cellulose fibre is in a solvent [1]. Several explanations have been proposed for the ballooning phenomenon observed in moderate quality solvent (mode 2 and 3). According to Ott et al. [2], during swelling in a solvent, the primary wall (external wall), considered as poorly soluble, cracks, then slides along the fibre to form protective regions where the fibre cannot swell. In order to highlight this analysis, we have performed high resolution observations by optical microscopy These observations reveal very clearly the presence of three main regions (See figure below). By studying the dissolution mechanisms of cotton fibres at different development stages (from elongation of the primary wall to the deposit of the secondary wall), we have shown that the dissolution of the primary wall is very difficult to obtain even in good quality solvent. At later stages, the secondary wall deposited (internal wall) dissolves from the inside and swells. The primary wall cracks under pressure to form unswollen sections and helices, and prevents the swelling and the dissolution of the fibre in some zones. The mechanism proposed is illustrated by high resolution microscopy observations and scanning electron microscopy images. After the removal of the primary wall by enzymatic treatment, the dissolution of wood and cotton fibres occurs with a homogeneous swelling instead of ballooning. Helices and unswollen sections are not visible any more. This study reveals that natural cellulose fibres do not dissolve easily due to their specific wall structure and shows the potential of new "activation" methods by enzymatic treatment to ease the dissolution. Cotton fibers are single-celled structures that differentiate from epidermal cells of the outer integument of the seed coat that elongate, fill with secondary cell wall cellulose, mature and die. Studies, which include the analysis of gene expression throughout fiber development and the detection of fiber quality QTLs, are underway in many laboratories to identify loci arid/or genes of interest that might explain the underlying mechanisms that determine cotton fiber development and quality. The model plant Arabidopsis thaliana is being used to investigate the molecular basis of many important biological functions, some of which, such as cell differentiation, cell elongation, and cellulose biosynthesis and deposition are relevant to the development of cotton fiber cells and orthologous genes active in these processes might provide useful candidates for genes that determine fiber quality traits. Cellulose synthesis, in particular, could play an important role in determining certain aspects of cotton fiber quality, such as fiber length, strength, at- fineness. Recent advances in understanding the synthesis of cellulose hove come from the analysis of A. thaliana cellulose-deficient mutants, and the isolation of a number of genes involved in this process, including the 10 members of the catalytic sub-unit of the cellulose synthase (CESA). Database searching has identified a number of cotton EST CESA orthologs that cluster with A. thaliana sequences rather than together, allowing the identification of orthologs for five of the 10 A. thaliana CESA isoforms. Furthermore, one totton EST formed a branch on its own, possibly indicating a specialized function for this gene in fiber development. Other genes involved in cellulose biosynthesis, as well as genes involved in cell elongation or in maintaining polarized cell expansion have been identified in A. thaliana. and for most of these genes, cotton orthologs can be found among fiber ESTs. These orthologs represent useful candidate genes to develop molecular markers for genetic mapping, fiber quality QTL analysis, and diversity studies in cotton and could be the targets for transformation experiments aimed at understanding their respective contribution to fiber quality, thus opening the way to the genetic engineering of cotton for the modification of fiber traits.

Cette mission fait suite à celle de janvier 2003, effectuée afin de prendre connaissance de la méthode de transformation du cotonnier par injection d'ADN plasmidique dans les fleurs et mettre en place la collaboration avec le laboratoire du Professeur Jia Shirong du BRI (Biotechnology Research Center du CAAS de Beijing). De même que lors de cette première mission, l'ensemble des frais de séjour ont été pris en charge par le laboratoire du Dr Jia Shirong. Transgenic cotton plants were amongst the first genetically modified (GM) plants to be released on the market in 1995/1996. Since then, the acreage devoted to GM cotton worldwide has increased dramatically, reaching 5.3 million hectares in the year 2000. This figure represents 16% of the world cotton acreage. In the United States of America, GM cotton represents more than 70% of the national cotton acreage, whereas in continental China, the proportion of transgenic cotton is approximately 10%. Other countries, such as Australia, Argentina, Mexico, and South Africa have already adopted this technology, while others are awaiting a proper legislation (or modifications thereof) to introduce these plants. Many more countries are in the process of evaluating GM cotton in the field. The rapid growth in the acreage devoted to GM cotton (from 700-800 000 ha in 1996 to more than 5 million ha in 2000) seems to indicate than farmers adopting this technology must somehow benefit from its use. The fact that these varieties have been grown on a large scale for 5 years allows a preliminary analysis of their financial and ecological impact. Despite important variations from year to year and from place to place, adopters of the GM technology seem to have benefited financially from transgenic varieties. The issue of GM plants is in the midst of a passionate debate in particular in Europe (but also in other countries), and various points regarding this technology have been raised. These include, but are not restricted to, public health, the impact on the environment (the development of toxin-resistant insects, (trans) gene flow...), as well as socio-economical issues. It is probable that the areas devoted to GM cotton will grow within the next few years through the opening of new markets, as well as through a more divers offer. Indeed, a number of companies and institutions are presently testing new transgenic cotton plants whose agronomic or technological properties have been modified through genetic engineering. Even though some of these products may never reach the market, it is probable that others will in the more or less near future.

Genetic research of CIRAD cotton programme, involves germplasm management, breeding, biotechnology, and methodological studies. It is being carried out joinly with partners from developing countries.

Cotton production in Thailand has been decreased by nearly 50% over the last fifteen years. The cotton bollworm, #Helicoverpa armigera# Hubner is the key pest in Thai cotton cropping system. One strategy to circumvent this problem is to transfer to cotton plants genes encoding for #Bacillus thuringiensis# endotoxins which have been identified as very toxic towards this insect. Techniques were developed for embryogeni callus induction, transformation and regeneration of Sri Somrong 60, a Thai cotton variety. Somatic embryogenesis was obtained by culturing hypocotyl explants on to zeatin and NAA containing MS medium and subculturing on to hormone free medium. This method yielded 23-50% embryogenic callus induction. Transformation of Sri Somrong 60 with synthetic crylA(b) gene was accomplished by using #Agrobacterium tumefaciens# strain C 58: pG2260. The selection of resistant calli which were obtained at frequencies varying from 3 to 50%, was accomplished on kanamycin containing medium. The molecular analysis of recovered plants showed the integration of the transferred gene. Bioassays conducted with leaves of different transgenic plant lines showed different level of resistance towards #Helicoverpa armigera# first instar larvae. The association of B.t genes with other tolerance characters existing in the cotton germplasm is in progress using conventional breeding techniques.

Protoplasts were isolated from callus, cotyledons, hypocotyls and leaves of cotton (Gossypium hirsutum L. var. Coker 310) using different digestion media. Protoplast yield per gram of fresh mass of leaves, callus cotyledons and hypocotyls was 22.2, 5.5, 1.7 and 1. 1 million respectively. Protoplasts were most viable in basic medium in presence of thidiazuro 0.663 gm/1 and 2,4,5-trichlorophenoxy acetic acid 7 mg/1. Hypocotyl and leaf-derived protoplasts formed microcolonies. Addition of CaC12 and glutamine to the basic medium improved protoplast survival, and division of hypocotyl and leaf-derived protoplasts. The growth conditions, such as the light and carbon source of donor plant were the important factors for protoplast isolation and culture.

Plants expression genes encoding -endotoxins from #Bacillus thuringiensis# (Bt) have triggered interest for the control of insect pest. Numerous plant species have been transformed with genes encoding various toxins. The first transformation experiments conducted with bacterial genes showed that their level of expression in plants is too low to confer adequate protection. To circumvent these problems, Bt toxins genes have been modified or resynthetized, dramatically improving their level of expression and the protection afforded. Despite these improvements, problems remain : the control of less susceptible insects and the durable deployment of transgenic plants have yet to be fully addressed.

Plants expressing genes encoding -endotoxins (cry genes) from the bacterium #Bacillus thuringiensis# (Bt) have triggered great interest for the control of agronomically important insect pests. A wide variety of plant species has been transformed with genes coding for various toxins aimed mainly at Lepidopteran and Coleopteran pests. The first transformation experiments conducted with bacterial genes (wild-type or native genes) have shown that the level of expression of these genes in plants is usually too low to confer good protection. To circumvent these problems, Bt toxin genes have been partially modified or totally re-synthesized, dramatically improving their level of expression in plants. Despite these improvements, some problems remain. In particular, the control of less susceptible insects, and that of the safe deployment of transgenic plants in relation to the emergence of toxin-resistant insects has yet to be addressed.

La résistance aux insectes est un objectif majeur pour l'amélioration variétale du cotonnier, la lutte mettant en jeu 30 % du marché mondial des insecticides chimiques. La première démarche a consisté à introduire et à faire exprimer dans la plante des gènes issus de Bacillus thuringiensis, bactérie productrice de toxine entomopathogénique. Les études sur l'expression dans le génome sont en cours. La deuxième priorité consiste dans la stratégie contre l'apparition de résistance à la toxine chez l'insecte ; d'où le recours préconisé à l'emploi de plusieurs gènes de résistance, associés à un gène codant pour des inhibiteurs de protéases. Ce programme de recherche est conduit par une entente CIRAD-INRA

Procédé de régénération du cocotier à partir d'explants comprenant les étapes de: induction de la formation de cals à partir d'explants, induction de l'embryogenèse sur les cab, obtention de structures embryonnaires maturation des embryons, caulogenèse et rhizogenèse. La maturation des embryons est effectuée par repiquage de ces structures sur un milieu dépourvu de charbon actif et d'auxine, et comprenant au moins une cytokinine.

Le transfert de gènes codant pour des protéines entomopathogènes permet la création de variétés de riz et de cotonnier résistant à certains insectes ravageurs. Le premier axe développé est l'utilisation de gènes codant pour des toxines "insecticides" de la bactérie Bacillus thuringiensis. Les premières obtentions de cotonniers transformés datent de 1987, à partir de la transformation via Agrobacterium tumefaciens et de la régénération par embryogenèse somatique. Quelques lignées ont confirmé au champ des propriétés insecticides. Le CIRAD, en collaboration avec l'INRA, conduit des recherches sur la transformation du cotonnier pour la résistance à Heliothis armigera et à Spodoptera littoralis. Des cotonniers transgéniques ayant intégré un gène de B.t. ou exprimant un gène d'inhibiteur de protéase ont été obtenus et ont produit des graines. Plus de quatre ans après les premières obtentions de dicotylédones transgéniques, des riz transgéniques ont été produits. L'application au transfert de résistance aux foreurs de tiges pour les riz méditerranéens et tropicaux est engagée par le CIRAD. La méthode de transfert utilise la transformation des protoplastes.
