IntroductionInfantile hypotonia with psychomotor retardation and characteristic facies-1 (IHPRF1, MIM#615419) is a rare, birth onset, autosomal recessive disorder caused by homozygous or compound heterozygous truncating variants in NALCN gene (MIM#611549) resulting in a loss-of-function effect.MethodsWe enrolled a new IHPRF1 patients’ cohort in the framework of an international multicentric collaboration study. Using specialized in silico pathogenicity predictors and ad hoc structural analyses, we assessed the mechanistic consequences of the deleterious variants retrieved on NALCN structure and function.ResultsTo date 38 different NALCN variants have been retrieved from 33 different families, 26 from unrelated and 22 from related patients. We report on five new IHPRF1 patients from four different families, harboring four newly identified and one previously retrieved variant that exhibited a markedly significant functional impact, thereby compromising the functionality of the protein complex.DiscussionBy widening the functional spectrum of biallelic variants affecting the NALCN gene, this article broadens the IHPRF1 syndrome’s genotype-phenotype correlation and gives new insight into its pathogenic mechanism, diagnosis, and clinical management.

Relatively little is known about the way vision is used to guide locomotion in the natural world. What visual features are used to choose paths in natural complex terrain? To answer this question, we measured eye and body movements while participants walked in natural outdoor environments. We incorporated measurements of the three-dimensional (3D) terrain structure into our analyses and reconstructed the terrain along the walker’s path, applying photogrammetry techniques to the eye tracker’s scene camera videos. Combining these reconstructions with the walker’s body movements, we demonstrate that walkers take terrain structure into account when selecting paths through an environment. We find that they change direction to avoid taking steeper steps that involve large height changes, instead of choosing more circuitous, relatively flat paths. Our data suggest walkers plan the location of individual footholds and plan ahead to select flatter paths. These results provide evidence that locomotor behavior in natural environments is controlled by decision mechanisms that account for multiple factors, including sensory and motor information, costs, and path planning.

Abstract Hydropower expansion is increasingly responsible for connectivity and biodiversity loss in freshwater ecosystems. The Amazon basin, which supports the highest level of freshwater biodiversity globally, faces such unplanned expansion. Here, we demonstrate and quantify the impacts of two major hydroelectric dams on the Madeira River on the trans‐Amazonian movements of one apex predatory catfish (Brachyplatystoma rousseauxii) performing the longest migration known in freshwaters. Using otolith microchemistry, we show that above the dams, the former basin‐wide homing migration between the breeding grounds in the upper Madeira and the nursery in the Amazon estuary has now been replaced by residency. We found evidence suggesting downstream migration of juvenile fish past dams into the Amazon estuary and that some of them home back to the Madeira. However, we did not find evidence that the fish homing back from the estuary can access their breeding grounds in the upper Madeira; they remain blocked below the dams. Our results provide undisputable evidence that the conservation of the species primarily requires the reestablishment of connectivity with the construction of efficient fishways.

Plant leaves can be used to determine the atmospheric deposition of organic contaminants, including polycyclic aromatic hydrocarbons (PAHs), to assess the contamination status of an area. The purpose of this study was to develop an analytical method for the determination of PAHs deriving from atmospheric deposition using Malva sylvestris leaves. Analytes were recovered from the leaves of the plant using cyclohexane as an organic solvent and subsequent sonication. The percentage recoveries (R%) were good (from 65.8 ± 3.2 to 104.2 ± 16.9), together with the instrumental analytical parameters, including correlation coefficients (r) ≥ 0.995 for all PAHs. The instrumental analysis was carried out using GC-MS in total ion current and single ion monitoring at the same time. Real samples taken from urban environments have shown that they are not always the most contaminated. At the Palermo site, leaves were observed to have high amounts of PAHs due to the deposition of dust generated by combustion processes that occurred near the sampling site. Further studies are recommended to compare the use of plants and classical sampling systems for monitoring the atmospheric deposition of key contaminants toxic to human health.

This research investigates the dynamics of contaminant exposure in European hake (Merluccius merluccius, L. 1758) from the Adriatic Sea (Central Mediterranean Sea) by examining the levels of total mercury (THg) and polycyclic aromatic hydrocarbons (PAHs) in the muscle fish tissues. The study explores the correlations between these pollutants and somatic indexes to identify the early warning signals of pollution and ecological effects. The levels of pollutants are influenced by season and sex. Lipids appear to have a minimal effect on the PAH levels, whereas they exhibit a positive correlation with mercury levels in the muscle. No significant relationships between the pollutants and condition indexes were observed, except for a positive correlation between THg and the gonadosomatic index, indicating a potential impact on the reproductive health of fish. In contrast, PAHs showed no meaningful correlation with condition indexes. Differences in contaminant accumulations and lipid levels between sexes reflect variations in metabolic activity, reproductive costs, and adaptive strategies to seasonal changes and energy demands. This study highlights the importance of long-term monitoring to improve pollution management, environmental conservation, and the protection of marine organisms’ health.

Fisheries management requires improvement in scientific knowledge to ensure sustainable exploitation of important commercial species and population renewal. Within this context, this study focused on the reproductive biology of spot tail mantis shrimp, Squilla mantis, in the Central Mediterranean Sea, aiming to understand the reproductive pattern of this species and validate the macroscopic maturity scale through histological analysis. A multi-year sampling was performed from 2016 to 2020 by a commercial fishing fleet in the Northern Central Adriatic Sea (GSA 17), and a total of 2206 individuals were collected. The monthly average value of the total sex ratio of S. mantis was 0.524 ± 0.044 (mean ± SEM) in favor of females, which dominated the population from September to April. The proposed 5 stage macroscopic maturity scale was validated histologically through histological analysis, confirming synchronous ovarian development. The somatic indexes (GSI and K Fulton) and monthly incidence of macroscopic ovarian maturity stages highlighted a protracted reproductive season from winter to spring (January–May). Although the length-weight relationship showed a similar growth trend between genders, males reached a bigger size in terms of carapace length (C.L.) and dominated the population from 32 mm (C.L.). The macroscopic L50 estimated was 25.94 mm (C.L.).

Small synthetic oligodeoxynucleotides (ODNs) can mimic microbial nucleic acids by interacting with receptor systems and promoting immunostimulatory activities. Nevertheless, some ODNs can act differently on the plasmacytoid dendritic cell (pDC) subset, shaping their immunoregulatory properties and rendering them suitable immunotherapeutic tools in several clinical settings for treating overwhelming immune responses. We designed HIV–1–derived, DNA- and RNA-based oligonucleotides (gag, pol, and U5 regions) and assessed their activity in conferring a tolerogenic phenotype to pDCs in skin test experiments. RNA-but not DNA-oligonucleotides are capable of inducing tolerogenic features in pDCs. Interestingly, sensing the HIV–1–derived single-stranded RNA-gag oligonucleotide (RNA-gag) requires both TLR3 and TLR7 and the engagement of the TRIF adaptor molecule. Moreover, the induction of a suppressive phenotype in pDCs by RNA-gag is contingent upon the induction and activation of the immunosuppressive enzyme Arginase 1. Thus, our data suggest that sensing of the synthetic RNA-gag oligonucleotide in pDCs can induce a suppressive phenotype in pDCs, a property rendering RNA-gag a potential tool for therapeutic strategies in allergies and autoimmune diseases.

Desmosterolosis is a rare sterol biosynthesis disorder characterized by multiple congenital anomalies, failure to thrive, severe developmental delay, progressive epileptic encephalopathy, and elevated levels of desmosterol caused by biallelic mutations of DHCR24 encoding 3-β-hydroxysterol Δ-24-reductase. DHCR24 is regarded as the key enzyme of cholesterol synthesis in the metabolism of brain cholesterol as it catalyzes the reduction of the Δ-24 double bond of sterol intermediates during cholesterol biosynthesis. To date, 15 DHCR24 variants, detected in 2 related and 14 unrelated patients, have been associated with the desmosterolosis disorder. Here, we describe a proband harboring the never-described DHCR24 homozygous missense variant NM_014762.4:c.506T>C, NP_055577.1:p.M169T, whose functional validation was confirmed through biochemical assay. By using molecular dynamics simulation techniques, we investigated the impact of this variant on the protein stability and interaction network with the flavin adenine dinucleotide cofactor, thereby providing a preliminary assessment of its mechanistic role in comparison to all known pathogenic variants, the wild-type protein, and a known benign DHCR24 variant. This report expands the clinical and molecular spectra of the DHCR24-related disorder, reports on a novel DHCR24 deleterious variant associated with desmosterolosis, and gives new insights into genotype–phenotype correlations.

The tryptophan-degrading enzyme indoleamine 2,3-dioxygenase 1 (IDO1) is a plastic immune checkpoint molecule that potently orchestrates immune responses within the tumor microenvironment (TME). As a heme-containing protein, IDO1 catalyzes the conversion of the essential amino acid tryptophan into immunoactive metabolites, called kynurenines. By depleting tryptophan and enriching the TME with kynurenines, IDO1 catalytic activity shapes an immunosuppressive TME. Accordingly, the inducible or constitutive IDO1 expression in cancer correlates with a negative prognosis for patients, representing one of the critical tumor-escape mechanisms. However, clinically trialed IDO1 catalytic inhibitors disappointed the expected anti-tumor efficacy. Interestingly, the non-enzymatic apo-form of IDO1 is still active as a transducing protein, capable of promoting an immunoregulatory phenotype in dendritic cells (DCs) as well as a pro-tumorigenic behavior in murine melanoma. Moreover, the IDO1 catalytic inhibitor epacadostat can induce a tolerogenic phenotype in plasmacytoid DCs, overcoming the catalytic inhibition of IDO1. Based on this recent evidence, IDO1 plasticity was investigated in the human ovarian cancer cell line, SKOV-3, that constitutively expresses IDO1 in a dynamic balance between the holo- and apo-protein, and thus potentially endowed with a dual function (i.e., enzymatic and non-enzymatic). Besides inhibiting the catalytic activity, epacadostat persistently stabilizes the apo-form of IDO1 protein, favoring its tyrosine-phosphorylation and promoting its association with the phosphatase SHP-2. In SKOV-3 cells, both these early molecular events activate a signaling pathway transduced by IDO1 apo-protein, which is independent of its catalytic activity and contributes to the tumorigenic phenotype of SKOV-3 cells. Overall, our findings unveiled a new mechanism of action of epacadostat on IDO1 target, repositioning the catalytic inhibitor as a stabilizer of the apo-form of IDO1, still capable of transducing a pro-tumorigenic pathway in SKOV-3 tumor. This mechanism could contribute to clarify the lack of effectiveness of epacadostat in clinical trials and shed light on innovative immunotherapeutic strategies to tackle IDO1 target.

Indoleamine 2,3 dioxygenase 1 (IDO1), a leader tryptophan-degrading enzyme, represents a recognized immune checkpoint molecule. In neoplasia, IDO1 is often highly expressed in dendritic cells infiltrating the tumor and/or in tumor cells themselves, particularly in human melanoma. In dendritic cells, IDO1 does not merely metabolize tryptophan into kynurenine but, after phosphorylation of critical tyrosine residues in the non-catalytic small domain, it triggers a signaling pathway prolonging its immunoregulatory effects by a feed-forward mechanism. We here investigated whether the non-enzymatic function of IDO1 could also play a role in tumor cells by using B16-F10 mouse melanoma cells transfected with either the wild-type Ido1 gene (Ido1WT) or a mutated variant lacking the catalytic, but not signaling activity (Ido1H350A). As compared to the Ido1WT-transfected counterpart (B16WT), B16-F10 cells expressing Ido1H350A (B16H350A) were characterized by an in vitro accelerated growth mediated by increased Ras and Erk activities. Faster growth and malignant progression of B16H350A cells, also detectable in vivo, were found to be accompanied by a reduction in tumor-infiltrating CD8+ T cells and an increase in Foxp3+ regulatory T cells. Our data, therefore, suggest that the IDO1 signaling function can also occur in tumor cells and that alternative therapeutic approach strategies should be undertaken to effectively tackle this important immune checkpoint molecule.

Marfan syndrome (MIM: # 154700; MFS) is an autosomal dominant disease representing the most common form of heritable connective tissue disorder. The condition presents variable multiorgan expression, typically involving a triad of cardiovascular, eye, and skeletal manifestations. Other multisystemic features are often underdiagnosed. Moreover, the disease is characterized by age related penetrance. Diagnosis and management of MFS in the adult population are well-described in literature. Few studies are focused on MFS in the pediatric population, making the clinical approach (cardiac and multiorgan) to these cases challenging both in terms of diagnosis and serial follow-up. In this review, we provide an overview of MFS manifestations in children, with extensive revision of major organ involvement (cardiovascular ocular and skeletal). We attempt to shed light on minor aspects of MFS that can have a significant progressive impact on the health of affected children. MFS is an example of a syndrome where an early personalized approach to address a dynamic, genetically determined condition can make a difference in outcome. Applying an early multidisciplinary clinical approach to MFS cases can prevent acute and chronic complications, offer tailored management, and improve the quality of life of patients.

We evaluated the accuracy of the quantitative and semiquantitative analysis in detecting regional atrophy patterns and differentiating mild cognitive impairment patients who remain stable (aMCI-S) from patients who develop Alzheimer’s disease (aMCI-AD) at clinical follow-up. Baseline magnetic resonance imaging was used for quantitative and semiquantitative analysis using visual rating scales. Visual rating scores were related to gray matter thicknesses or volume measures of some structures belonging to the same brain regions. Receiver operating characteristic (ROC) analysis was performed to assess measures’ accuracy in differentiating aMCI-S from aMCI-AD. Comparing aMCI-S and aMCI-AD patients, significant differences were found for specific rating scales, for cortical thickness belonging to the middle temporal lobe (MTL), anterior temporal (AT), and fronto-insular (FI) regions, for gray matter volumes belonging to MTL and AT regions. ROC curve analysis showed that middle temporal atrophy, AT, and FI visual scales showed better diagnostic accuracy than quantitative measures also when thickness measures were combined with hippocampal volumes. Semiquantitative evaluation, performed by trained observers, is a fast and reliable tool in differentiating, at the early stage of disease, aMCI patients that remain stable from those patients that may progress to AD since visual rating scales may be informative both about early hippocampal volume loss and cortical thickness reduction.

WFS1 spectrum disorder (WFS1-SD) is a rare monogenic neurodegenerative disorder whose cardinal symptoms are childhood-onset diabetes mellitus, optic atrophy, deafness, diabetes insipidus, and neurological signs ranging from mild to severe. The prognosis is poor as most patients die prematurely with severe neurological disabilities such as bulbar dysfunction and organic brain syndrome. Mutation of the WFS1 gene is recognized as the prime mover of the disease and responsible for a dysregulated ER stress signaling, which leads to neuron and pancreatic β-cell death. There is no currently cure and no treatment that definitively arrests the progression of the disease. GLP-1 receptor agonists appear to be an efficient way to reduce elevated ER stress in vitro and in vivo, and increasing findings suggest they could be effective in delaying the progression of WFS1-SD. Here, we summarize the characteristics of GLP-1 receptor agonists and preclinical and clinical data obtained by testing them in WFS1-SD as a feasible strategy for managing this disease.

Src is a protein tyrosine kinase commonly activated downstream of transmembrane receptors and plays key roles in cell growth, migration, and survival signaling pathways. In conventional dendritic cells (cDCs), Src is involved in the activation of the non-enzymatic functions of indoleamine 2,3-dioxygenase 1 (IDO1), an immunoregulatory molecule endowed with both catalytic activity and signal transducing properties. Prompted by the discovery that the metabolite spermidine confers a tolerogenic phenotype on cDCs that is dependent on both the expression of IDO1 and the activity of Src kinase, we here investigated the spermidine mode of action. We found that spermidine directly binds Src in a previously unknown allosteric site located on the backside of the SH2 domain and thus acts as a positive allosteric modulator of the enzyme. Besides confirming that Src phosphorylates IDO1, here we showed that spermidine promotes the protein–protein interaction of Src with IDO1. Overall, this study may pave the way toward the design of allosteric modulators able to switch on/off the Src-mediated pathways, including those involving the immunoregulatory protein IDO1.

Abstract Background Malan syndrome (MALNS) is a recently described ultrarare syndrome lacking guidelines for diagnosis, management and monitoring of evolutive complications. Less than 90 patients are reported in the literature and limited clinical information are available to assure a proper health surveillance. Results A multidisciplinary team with high expertise in MALNS has been launched at the “Ospedale Pediatrico Bambino Gesù”, Rome, Italy. Sixteen Italian MALNS individuals with molecular confirmed clinical diagnosis of MALNS were enrolled in the program. For all patients, 1-year surveillance in a dedicated outpatient Clinic was attained. The expert panel group enrolled 16 patients and performed a deep phenotyping analysis directed to clinically profiling the disorder and performing critical revision of previously reported individuals. Some evolutive complications were also assessed. Previously unappreciated features (e.g., high risk of bone fractures in childhood, neurological/neurovegetative symptoms, noise sensitivity and Chiari malformation type 1) requiring active surveillance were identified. A second case of neoplasm was recorded. No major cardiovascular anomalies were noticed. An accurate clinical description of 9 new MALNS cases was provided. Conclusions Deep phenotyping has provided a more accurate characterization of the main clinical features of MALNS and allows broadening the spectrum of disease. A minimal dataset of clinical evaluations and follow-up timeline has been proposed for proper management of patients affected by this ultrarare disorder.

Mucinous epithelial ovarian cancer (mEOC) is a rare subtype of epithelial ovarian cancer, characterized by poor responses to standard platinum-based chemotherapy. Polo-like kinase 1 (PLK1) is a key regulator of mitosis and cell cycle progression and its inhibition has been recently identified as a target in mEOC. In this study, we aimed to identify further therapeutic targets in mEOC using a CRISPR/Cas9 library targeting 3015 genes, with and without treatment with onvansertib, a PLK1 inhibitor. We identified twelve genes associated with cell survival (ZC2HC1C, RPA2, KIN17, TUBG1, SMC2, CDC26, CDC42, HOXA9, TAF10, SENP1, MRPS31, and COPS2) and three genes (JUND, CARD9, and BCL2L2) in synthetic lethality with onvansertib treatment. We validated that SENP1 downregulation is important for the growth of mEOC cells through esiRNA interference and the use of a pharmacological inhibitor Momordin Ic. The downregulation of CARD9 and BCL2L2 combined with subtoxic doses of onvansertib interfered with mEOC cell growth. Interestingly, the combination of navitoclax, an inhibitor of BcL2 family members including BCL2L2, was synergistic in all four of the mEOC cell lines tested and substantially induced cell death through apoptosis. These data support the use of a combination of navitoclax and onvansertib as a new therapeutic strategy for mEOC. [ABSTRACT FROM AUTHOR]

Context: The invasive wels catfish is spreading to many European waterbodies and is the subject of controversy concerning its environmental impact. Aims: The objective was to investigate its life-history traits (growth and reproduction) for the first time in southern France. Methods: A culling action was used to assess the key life-history traits, using sections of pectoral spines for age and growth estimations, and gonad macroscopic examination for the reproduction parameters. Key results: The biggest individual was a 227-cm male. The age was accurately estimated from clear translucent marks on spines, with a maximum of 13 years for both sexes. The growth appeared to be extremely rapid and was the highest at old ages recorded from both the native and the invasive distribution range. The length and age at first sexual maturity, 70.1 cm for 4–5-year-old females, and 54.1 cm for 2–3-year-old males, were the lowest recorded in Europe, indicating an early maturity. Conclusions: The life-history parameters (rapid growth, early maturation, high fecundity) showed a very high potential for adaptation and colonisation. Implications: The cost and management required to regulate the species appear not easily feasible, particularly when combined with its high growth rate. Notwithstanding the increased interest in wild edible plants, little is known on how some domestic thermal processes can affect their content. The aim of this study was to investigate the amounts of minerals, B1 and B2 vitamins, tocols, and carotenoids in raw, boiled, and steamed wild edible plants, namely, Sonchus asper (L.) Hill s.l., Sonchus oleraceus L., Cichorium intybus L., and Beta vulgaris L. var cicla. All vegetables were confirmed as high sources of lutein (from 6 to 9 mg/100 g) and β-carotene (from 2 to 5 mg/100 g). Quite high amounts of violaxanthin and neoxanthin were found. Alfa-tocopherol and γ-tocopherol were the main tocols, with same contents in raw and processed vegetables (about 2.5 mg/100 g). The most abundant macro element and trace element were, respectively, potassium and iron. B1 and B2 vitamins were found in low amounts in almost all plants, with the exception of thiamine in Beta vulgaris (about 1.6 mg/100 g). Boiling led to a significant loss of minerals (up to 60%) and B-group vitamins (up to 100%), while, among carotenoids, it only affected violaxanthin levels (up to 90%). Steamed vegetables showed only a slight reduction, about 20%, in β-carotene and lutein, with a marked decrease in violaxanthin and neoxanthin. One hundred grams of all fresh and cooked plants can be claimed as a source of vitamin A and E.

The European eel (Anguilla anguilla) is endangered due to its peculiar life-history cycle, fishing pressure and difficulty in global population management. To improve our understanding of the population dynamics and refine conservation policies, an extended mark and recapture experiment and glass eel stocking were conducted in the River Rhone Delta (France) over 8 years. Around 1100 yellow eels were PIT-tagged and released in 2007, 2.5 kg of glass eels were released each year from 2008 to 2012, and the population was monitored using fishing between 2007 and 2015. After capture or recapture, the body parameters, sex and maturity were assessed. Age was estimated from otolith growth rings. At the end of the experiment, silver eels were between 352 and 875 mm long (age 17 to 185 months) and yellow eels between 170 and 868 mm long (age 12 to 123 months). Age estimates were validated using mark and recapture and showed 16% age underestimates and 5% inaccurate ages. The growth rates were extremely variable with lengths ranged by 2-fold at a given age. These results highlight the difficulty of eel population management, at least in the Mediterranean area. L'anguille europeenne (Anguilla anguilla) est menacee en raison de son cycle biologique singulier, de la pression de peche et de la difficulte que presente la gestion de sa population. Pour une meilleure comprehension de la dynamique de sa population et de la politique de conservation, une vaste experience de marquage-recapture et le deversement de civelles a ete realisee dans le delta du Rhone (France) sur une periode de huit ans. Quelques 1100 anguilles jaunes ont ete marquees avec des PIT-tags et relachees en 2007, 2,5 kg de civelles ont ete relaches chaque annee de 2008 a 2012, et la population a fait l'objet d'une surveillance par peche de 2007 a 2015. Apres capture ou recapture, les parametres corporels, le sexe et la maturite ont ete enregistres. L'age a ete estime a partir d'anneaux de croissance des otolites. A la fin de l'experience, les anguilles argentees faisaient de 352 a 875 mm de long (leurs ages allant de 17 a 185 mois) et les anguilles jaunes, de 170 a 868 mm de long (12 a 123 mois). La validation des ages estimes par marquage et recapture a revele que 16 % des ages etaient sous-estimes et 5 % etaient inexacts. Les taux de croissance etaient extremement variables, les longueurs pouvant varier du simple au double pour un age donne.
