Abstract

In recent years, excessive use of antibiotics has been raising its head to a serious problem all around the world, as pathogens become drug-resistant and create challenges to the medical field. This failure of the most potent antibiotics that kill pathogens increases the thirst for researchers to look for another way of killing pathogens. It has led to the findings of antimicrobial peptide, which are the most potent peptides to destroy pathogens. This review gives special emphasis to the usage of marine bacteria and other microorganisms for antimicrobial peptide (AMP), which are eco-friendly as well as a developing class of natural and synthetic peptides with a wide spectrum of targets to pathogenic microbes. Consequently, a significant attention has been paid mainly to (i) the structure and types of antimicrobial peptides and (ii) mode of action and mechanism of antimicrobial peptide resistance to pathogens. In addition to this, the designing of AMPs has been analyzed thoroughly for reducing toxicity and developing better potent AMP. It has been done by the modified unnatural amino acids by amidation to target the control of biofilm and persister cells.

Abstract

Cancer is one of the most deadly diseases on the planet. Over the past decades, numerous antineoplastic compounds have been discovered from natural resources such as medicinal plants and marine species as part of multiple drug discovery initiatives. Notably, several marine flora (e.g. Ascophyllum nodosum, Sargassum thunbergii) have been identified as a rich source for novel cytotoxic compounds of different chemical forms. Despite the availability of enormous chemically enhanced new resources, the anticancer potential of marine flora and fauna has received little attention. Interestingly, numerous marine-derived secondary metabolites (e.g., Cytarabine, Trabectedin) have exhibited anticancer effects in preclinical cancer models. Most of the anticancer drugs obtained from marine sources stimulated apoptotic signal transduction pathways in cancer cells, such as the intrinsic and extrinsic pathways. This review highlights the sources of different cytotoxic secondary metabolites obtained from marine bacteria, algae, fungi, invertebrates, and vertebrates. Furthermore, this review provides a comprehensive overview of the utilisation of numerous marine-derived cytotoxic compounds as anticancer drugs, as well as their modes of action (e.g., molecular target). Finally, it also discusses the future prospects of marine-derived drug developments and their constraints.

Abstract

Background and Aim: Rheumatoid arthritis (RA) is a prevalent and debilitating disease that affects the joints. The study was intended to evaluate the anti-arthritic activity of aerial parts of Heliotropium zeylanicum (EEHZ). EEHZ constituting phytoconstituents to treat adjuvant induced arthritic rats to minimize the side effects. Materials and Methods: The anti-arthritic activity study was carried out by using adjuvant induced model of Wistar albino strain rats. EEHZ was injected at different doses such as 100 and 300 mg/kg/i.p., and the study was compared with standard drug dexamethazone (5 g/kg). The results obtained from the above methods were subjected to statistical analysis. Results: The study was conducted various hematological parameters and histopathological sections were carried out. The plant has various phytoconstituents such as reducing sugars, saponins, starch, steroids and terpenoids. Significantly one among those phytoconstituent lessens to oedema and arthritis of the rodents. The EEHZ showed the maximum inhibitory activity at (300 mg/kg/i.p.) by dose dependent manner. These inhibitions were statistically significant (P < 0.01-0.001). Conclusion: From these results indicate that EEHZ is a bioactive agent and having significant results in anti-inflammatory and anti-arthritic action by inhibition of the exudation, and leukocytes recruitment into the inflamed tissues and bone regeneration calcium deposition. Abstract

The preponderance of helminth contagions is constrained to tropical regions and can cause massive vulnerability to malnutrition, anemia, pneumonia, and eosinophilia. Many human parasitic diseases cause severe illness in endemic populations. The helminths transmitting through the gastro-intestinal tract may develop resistance to anthelmintic drugs. The phytotherapy, anthelmintic, and antimicrobial efficacy of Dodonaea viscosaleaf solvent extracts were examined. Phytochemical screening was carried out by ultraviolet (UV) and Fourier transform infrared spectroscopy (FTIR). The anthelmintic activity was performed against a South Indian earth worm as a model by measuring the paralytic time. The antibacterial activity was performed against Bacillus subtilis, Escherichia coli, Klebsiella pneumoniae, and Staphylococcus aureus, which stimulate the stomach helminths. Phytochemical screening revealed the presence of chemical compounds from the UV spectrum, and alkenes and aromatic compounds were confirmed by FTIR. Higher concentrations of the D. viscosaextract showed a rapid paralytic effect with a rapid death rate and histopathology. The zone-of-inhibition study indicated the potent antibacterial activity of the ethanolic and methanolic extracts of D. viscosaagainst different bacterial species. The current research revealed that D. viscosahas significant anthelmintic and antibacterial activities and it can be used for further elucidation and characterization.
