Purpose -- The purpose of this paper is to assess the current status of special libraries of Bangladesh and their ability to adapt information technology (IT) in various library operations and services. Design/methodology/approach -- The study is based on the case study method. A structured questionnaire was distributed among the special libraries of Bangladesh to uncover the status of using IT by those libraries. Findings -- Being a developing country, Bangladesh is confronted with certain problems to introduce IT-based services and other facilities in the special libraries. The study has revealed the embryonic state of IT in the special libraries of Bangladesh. Originality/value -- The paper is an original case study on the application of IT in the Special Libraries of Bangladesh. It will help web researchers to obtain a precise picture of the application of IT in these libraries. Purpose --The purpose of this paper is to look at the prolific growth of electronic resources during the last decade. The standards and strategies are sought to access and organize the abundance of e-resources and to ensure free flow of information to the user's community. This survey attempts to measure the quantum of e-resources accessed in the academic ambience of Business School libraries in Orissa (India). Moreover, the study attempts to examine to what extent Electronic Information Services (EIS) are offered to users of Business School libraries in the State with an opinion pool of the librarians of the respective Business Schools. Furthermore, the study aims to highlight problems and constraints faced by the information professionals in accessing e-resources and delivering electronic information services with some constructive suggestions and remedial measures for the ensuing bottle-neck. Design/methodology/approach -- The paper highlights the availability of e-resources and critically examines the quantum of their use in Business School libraries of Orissa and the crucial role played by the information professionals in providing electronic information services to users through the findings based on responses from structured questionnaires. The SPSS package is used for analysis of data. Findings -- The paper arrives at the conclusion that the internet-based e-resources are being well used compared with CD-ROM databases. Premier web search options like Google and Yahoo! are the most frequently used search engines, while the other searching options are less used. Availability of some key online databases are exclusively confined to only a selected few B-School libraries of the State. Practical implications -- The paper restricts the study exclusively to e-resources as the type of material and the librarians of 25 major Business School libraries of Orissa (India) as the respondents in its scope of discussion. Originality/value -- The paper sheds some light on the use of e-resources in Business School libraries of Orissa (India) with some constructive suggestions for the development of the platform of electronic resources and services. Purpose-The purpose of the study is to find out the trends of web-based information seekers at Sambalpur University, India since the internet is more helpful than the library in the present electronic era.Design/methodology/approach-The study is based on the case study method. A structured questionnaire was distributed among the relevant researchers at Sambalpur University in order to ascertain their web searching habits.Findings-The application of is ever increasing-to the extent that people believe that electronic material will eventually replace the traditional library and users need not go there to find and collect the information they need. The study revealed, however, that in one Indian university, traditional library and printed materials are still more effective to researchers than web-based information and resources.Originality/value-This is an original case study on web-based information retrieval trends of university researchers. This paper will help web researchers to obtain a clear-cut picture of the information seeking and retrieval trends of the researchers of an Indian university which will serve as a role model for the universities of the Third World. Osteoporosis is well noted to be a universal ailment that realization impaired bone mass and micro architectural deterioration thus enhancing the probability of fracture. Despite its high incidence, its management remains highly demanding because of the multifactorial pathophysiology of the disease. This review highlights recent findings in the management of osteoporosis particularly, gene expression and hormonal control. Some of the newest approaches regarding the subject are described, including single-cell RNA sequencing and long non-coding RNAs. Also, the review reflects new findings on hormonal signaling and estrogen and parathyroid hormone; patient-specific approaches due to genetic and hormonal variation. Potential new biomarkers and AI comprised as factors for improving the ability to anticipate and manage fractures. These hold great potential of new drugs, combination therapies and gene based therapies for osteoporosis in the future. Further studies and cooperation of scientists and clinicians will help to apply such novelties into practical uses in the sphere of medicine in order to enhance the treatment of patients with osteoporosis.

Artificial intelligence (AI) is one of the fastest-growing fields in various industries, including engineering, architecture, medical and clinical research, aerospace, and others. AI, which is a combination of machine learning (ML), deep learning (DL), and human intelligence (HI), is revolutionizing drug discovery and development by making it more cost-effective and efficient. It is also being used in fields such as medicinal chemistry, molecular and cell biology, pharmacology, pharmacokinetics, formulation development, and toxicology. AI plays a crucial role in clinical testing by enhancing patient stratification, patient sample evaluation, and trial design, assisting in the identification of biomarkers, determining efficacy criteria, dose selection, trial length, and target patient population selection. The primary objective of this study is to emphasize the importance of AI in clinical trials and drug development, while also exploring the existing challenges and potential advancements in AI within the healthcare industry. A comprehensive literature review was conducted, covering the period from 1998 to 2023. The Science Direct, PubMed, and Google Scholar databases were searched for relevant information. A variety of publications, including Research Gate, Nature, MDPI, and Springer Link, provided pertinent data. This study aimed to gain a deeper understanding of the use of AI in clinical research and drug development, as well as its potential and limitations. We also discuss the benefits and main data limitations of the traditional trial and drug development approach. AI approaches are currently being used to overcome research obstacles and eliminate conceptual or methodological limitations. After discussing possible obstacles and coping mechanisms, we provide several recommendations to help individuals understand the challenges and difficulties associated with clinical research and drug development. Background: Hereditary nonsyndromic hearing loss (NSHL) is an extremely heterogeneous disorder, both genetically and clinically. Myosin VI ( MYO6 ) pathogenic variations have been reported to cause both prelingual and postlingual forms of NSHL. Postlingual autosomal dominant cases are often overlooked for genetic etiology in clinical setups. In this study, we used next-generation sequencing (NGS)-based targeted deafness gene panel assay to identify the cause of postlingual hearing loss in an Indian family. Methods: The proband and his father from a multigenerational Indian family affected by postlingual hearing loss were examined via targeted capture of 129 deafness genes, after excluding gap junction protein beta 2 ( GJB2 ) pathogenic variants by Sanger sequencing. NGS data analysis and co-segregation of the candidate variants in the family were carried out. The variant effect was predicted by in silico tools and interpreted following American College of Medical Genetics and Genomics-Association for Molecular Pathology guidelines. Results: A novel heterozygous transversion c.3225T>G, p.(Tyr1075*) in MYO6 gene was identified as the disease-causing variant in this family. This stop-gained variant is predicted to form a truncated myosin VI protein, which is devoid of crucial cargo-binding domain. PCR-RFLP screening in 200 NSHL cases and 200 normal-hearing controls showed the absence of this variant indicating its de novo nature in the population. Furthermore, we reviewed MYO6 variants reported from various populations to date. Conclusions: To the best of our knowledge, this is the first family with MYO6 -associated hearing loss from an Indian population. The study also highlights the importance of deafness gene panels in molecular diagnosis of GJB2 -negative pedigrees, contributing to genetic counseling in the affected families.

An accurate assessment of nitrate leaching is important for efficient fertiliser utilisation and groundwater pollution reduction. However, past studies could not efficiently model nitrate leaching due to utilisation of conventional algorithms. To address the issue, the current research employed advanced machine learning algorithms, viz., Support Vector Machine, Artificial Neural Network, Random Forest, M5 Tree (M5P), Reduced Error Pruning Tree (REPTree) and Response Surface Methodology (RSM) to predict and optimize nitrate leaching. In this study, Urea Super Granules (USG) with three different coatings were used for the experiment in the soil columns, containing 1 kg soil with fertiliser placed in between. Statistical parameters, namely correlation coefficient, Mean Absolute Error, Willmott index, Root Mean Square Error and Nash-Sutcliffe efficiency were used to evaluate the performance of the ML techniques. In addition, a comparison was made in the test set among the machine learning models in which, RSM outperformed the rest of the models irrespective of coating type. Neem oil/ Acacia oil(ml): clay/sulfer (g): age (days) for minimum nitrate leaching was found to be 2.61: 1.67: 2.4 for coating of USG with bentonite clay and neem oil without heating, 2.18: 2: 1 for bentonite clay and neem oil with heating and 1.69: 1.64: 2.18 for coating USG with sulfer and acacia oil. The research would provide guidelines to researchers and policymakers to select the appropriate tool for precise prediction of nitrate leaching, which would optimise the yield and the benefit-cost ratio., (© 2024. Otosclerosis (OTSC) is a focal and diffuse bone disorder of the human middle ear characterized by abnormal bone growth and deposition at the stapes' footplate. This hinders the transmission of acoustic waves to the inner ear leading to subsequent conductive hearing loss. The plausible convections for the disease are genetic and environmental factors with yet an unraveled root cause. Recently, exome sequencing of European individuals with OTSC revealed rare pathogenic variants in the Serpin Peptidase Inhibitor, Clade F (SERPINF1) gene. Here, we sought to investigate the causal variants of SERPINF1 in the Indian population. The gene and protein expression was also evaluated in otosclerotic stapes to ameliorate our understanding of the potential effect of this gene in OTSC. A total of 230 OTSC patients and 230 healthy controls were genotyped by single-strand conformational polymorphism and Sanger sequencing methods. By comparing the case controls, we identified five rare variants (c.72 C > T, c.151 G > A, c.242 C > G, c.823 A > T, and c.826 T > A) only in patients. Four variants c.390 T > C (p = 0.048), c.440-39 C > T (p = 0.007), c.643 + 9 G > A (p = 0.035), and c.643 + 82 T > C (p = 0.005) were found to be significantly associated with the disease. Down-regulation of SERPINF1 transcript level in otosclerotic stapes was quantified by qRT-PCR, ddPCR and further validated by in situ hybridization. Similarly, reduced protein expression was observed by immunohistochemistry and immunofluorescence in otosclerotic stapes that corroborate with immunoblotting of patients' plasma samples. The transition of the Earth's climate from one zone to another is one of the major causes behind biodiversity loss, rural-urban migration, and increasing food crises. The rising rate of arid-humid zone transition due to climate change has been substantially visible in the last few decades. However, the precise quantification of the climate change-induced rainfall variation on the climate zone transition still remained a challenge. To solve the issue, the Representative Grid Location-Multivariate Adaptive Regression Spline (RGL-MARS) downscaling algorithm was coupled with the Koppen climate classification scheme to project future changes in various climate zones for the study area. It was observed that the performance of the model was better for the humid clusters compared to the arid clusters. It was noticed that, by the end of the 21 st century, the arid region would increase marginally and the humid region would rise by 24.28-36.09% for the western province of India. In contrast, the area of the semi-arid and semi-humid regions would decline for the study area. It was observed that there would be an extensive conversion of semi-humid to humid zone in the peripheral region of the Arabian sea due to the strengthening of land-sea thermal contrast caused by climate change. Similarly, semi-arid to arid zone conversion would also increase due to the inflow of dry air from the Arabian region. Background: Green strategy involves the design, synthesis, processing, and use of chemical substances by eliminating the generation of chemical hazards. This approach focuses on atom economy, use of safer solvents or chemicals, consumption of energy, and decomposition of the chemical substances to non-toxic materials which are eco-friendly., Objective: So, the microwave irradiated heating method is considered a green and sustainable technique for the development of novel heterocyclic scaffold-like isoxazole derivatives via chalcones. Isoxazole derivatives play a vital role due to their diverse pharmacological activities such as antibiotic (Sulfamethoxazole, Cloxacillin, Flucloxacillin, Cycloserine), anti-fungal (Drazoxolon), Antirheumatic (Leflunomide), antidepressant (Isocarboxazid), antineoplastic (Acivicin), anticonvulsant (Zonisamide), antipsychotic (Risperidone) and anti-inflammatory drugs (Valdecoxib), etc. Methods: The isoxazole derivatives were synthesized with the help of microwave irradiation that follows green chemistry protocol., Results: The titled compounds were subjected to antiepileptic evaluation to determine their therapeutic potential., Conclusion: The use of microwave radiation enhances the rate of the reaction which leads to high selectivity with improved product yields in comparison with the traditional heating methods. Microwave radiation is used as a heating source during the synthesis of heterocyclic compounds. The heating mechanisms involved in microwave-induced synthesis include dipolar polarization and ionic conduction. This heating technology follows the green protocol as it involves the use of recyclable organic solvents during synthesis. The microwave heating approach offers a faster rate of reaction, easier work-up procedure, and higher product yield with purity and also reduces environmental pollution. So, microwave heating is applied as a sustainable technology for the efficient production of pyrimidine compounds as one of the heterocyclic moieties. Pyrimidine is a six-membered nitrogenous heterocyclic compound that plays a significant role due to several therapeutic applications. Otosclerosis (OTSC) is the primary form of conductive hearing loss characterized by abnormal bone remodelling within the otic capsule of the human middle ear. A genetic association of the RELN SNP rs3914132 with OTSC has been identified in European population. Previously, we showed a trend towards association of this polymorphism with OTSC and identified a rare variant rs74503667 in a familial case. Here, we genotyped these variants in an Indian cohort composed of 254 OTSC cases and 262 controls. We detected a significant association of rs3914132 with OTSC (OR = 0.569, 95%CI = 0.386-0.838, p = 0.0041). To confirm this finding, we completed a meta-analysis which revealed a significant association of the rs3914132 polymorphism with OTSC (Z = 6.707, p<0.0001) across different ethnic populations. Linkage analysis found the evidence of linkage at RELN locus (LOD score 2.1059) in the OTSC family which has shown the transmission of rare variant rs74503667 in the affected individuals. To understand the role of RELN and its receptors in the development of OTSC, we went further to perform a functional analysis of RELN/reelin. Here we detected a reduced RELN (p = 0.0068) and VLDLR (p = 0.0348) mRNA levels in the otosclerotic stapes tissues. Furthermore, a reduced reelin protein expression by immunohistochemistry was confirmed in the otosclerotic tissues. Electrophoretic mobility shift assays for rs3914132 and rs74503667 variants revealed an altered binding of transcription factors in the mutated sequences which indicates the regulatory role of these variations in the RELN gene regulation. Subsequently, we showed by scanning electron microscopy a change in stapes bone morphology of otosclerotic patients. In conclusion, this study evidenced that the rare variation rs74503667 and the common polymorphism rs3914132 in the RELN gene and its reduced expressions that were associated with OTSC., Competing Interests: The authors have declared that no competing interests exist.

The global incidence of cancer and cancer-related mortality is expected to rise in recent years despite advancements in cancer diagnosis and therapeutics. Increasing evidences of decrypting molecular mechanisms underlying cancer progression have commanded the tremendous development of synthetic anticancer drugs. With limitations in the current conventional cancer therapeutic approaches, the non-nutritive dietary phytochemicals have emerged as potent modulators of apoptosis and autophagy associated key signaling pathways in various cancer cells. The dynamic regulation of apoptosis and autophagy by phytochemicals in cancer are identified as promising therapeutic candidates with minimal cytotoxicity and enhanced biological activity. Dietary phytochemicals and their synthetic analogs have exhibited potency in the modulation of apoptosis and autophagy in several cancer cells as individuals or in combination with pre-existing FDA (Food and Drug Administration) approved anticancer drugs. In the current generation of medical science, developing precision and personalized medicine and their consumption as food supplements will hold high prevalence in cancer therapeutics. Hence understating the impact of dietary phytochemicals on human health and their molecular mechanism will thrive a new horizon in cancer therapeutics. Background: Otitis media (OM) is a middle ear inflammatory complex disorder involving genetic and environmental factors. It onsets during childhood and often recurs and perplexes in genetically susceptible patients. Previously, murine models had shown the association of ISL LIM homeobox 1 (ISL1) gene with otitis media with effusion., Aim: To investigate the association of ISL1 genetic variants with otitis media., Subjects and Methods: A total of 285 cases and 277 controls were recruited for the study. The entire coding region of ISL1 gene was genotyped using Sanger sequencing or single-strand conformation polymorphism methods. Genotype, haplotype, in silico analysis, and linkage disequilibrium analysis were performed., Results: The variants rs2303751 (c.504A>G) and rs121913540 (c.513G>A) were associated with OM, and the OR (95%CI) was 0.74 (0.57-0.95) and 0.43 (0.20-0.91), respectively. Besides, the rs2303751 AA genotype was associated with elevated eosinophil numbers in OM when compared to controls. The 5 SNP haplotype analysis of SNPs c.-492A>G, c.504A>G, c.513G>A, c.576C>T, and c.*651A>T revealed A-A-G-C-A to be a risk haplotype in females whereas the 3 SNP haplotype analysis of SNPs c.504A>G, c.513G>A, and c.567C>T suggested G-A-C as protective and A-G-C to be a risk haplotype for otitis media., Conclusion: Ours is the first report which shows a significant association of ISL1 variants (rs2303751 and rs121913540) with hearing-related disorder like otitis media in humans. These results implicate the possible role of ISL1 gene in the etiopathology of otitis media. Green synthetic protocol refers to the development of processes for the sustainable production of chemicals and materials. For the synthesis of various biologically active compounds, energy-efficient and environmentally benign processes are applied, such as microwave irradiation technology, ultrasound-mediated synthesis, photo-catalysis (ultraviolet, visible and infrared irradiation), molecular sieving, grinding and milling techniques, etc. Thesemethods are considered sustainable technology and become valuable green protocol to synthesize new drug molecules as theyprovidenumerous benefits over conventional synthetic methods.Based on this concept, oxadiazole derivatives are synthesized under microwave irradiation technique to reduce the formation of byproduct so that the product yield can be increased quantitatively in less reaction time. Hence, the synthesis of drug molecules under microwave irradiation follows a green chemistry approach that employs a set of principles to minimize or remove the utilization and production of hazardous toxic materials during the design, manufacture and application of chemical substances.This approach plays a major role in controlling environmental pollution by utilizing safer solvents, catalysts, suitable reaction conditions and thereby increases the atom economy and energy efficiency. Oxadiazole is a five-membered heterocyclic compound that possesses one oxygen and two nitrogen atoms in the ring system.Oxadiazole moiety is drawing considerable interest for the development of new drug candidates with potential therapeutic activities including antibacterial, antifungal, antiviral, anticonvulsant, anticancer, antimalarial, antitubercular, anti-asthmatic, antidepressant, antidiabetic, antioxidant, antiparkinsonian, analgesic and antiinflammatory, etc. This review focuses on different synthetic approaches of oxadiazole derivatives under microwave heating method and study of their various biological activities.

Background: Otosclerosis (OTSC) is a genetically heterogeneous disorder, characterized by abnormal bone growth in the middle ear, affecting the stapes bone. Previous studies have shown that single nucleotide polymorphisms (SNPs) of the COL1A1, BMP2, and BMP4 genes are linked to susceptibility of OTSC, musculoskeletal degenerative diseases, and bone remodeling. Aims : To evaluate the genetic association and expression levels of COL1A1 , BMP2 , and BMP4 genes with OTSC in the Indian population. Methods : A total of 320 otosclerotic and 320 control samples were screened for four SNPs (rs1107946, rs11327935, rs2269336, and rs1800012) of the COL1A1 gene; rs3178250 of the BMP2 gene; and rs17563 of the BMP4 gene using single-strand conformation polymorphism analysis, and restriction fragment length polymorphism analyses. Genotypic, haplotypic, and linkage disequilibrium analyses were performed to assess the potential associations of these SNPs with OTSC. COL1A1 , BMP2 , and BMP4 mRNA expression levels were analyzed by semiquantitative RT-PCR and real-time PCR. Results : Genotypes of two SNPs, rs1800012 and rs17563, were found to be associated with OTSC (the rs1800012 GT genotype, p  = 0.0022, OR = 0.481; and the rs17563 TC genotype, p  = 0.0225, OR = 1.471). Haplotypic analyses revealed that the COL1A1 haplotype G-T-C-T ( p  = 0.021) was significantly increased among controls. Functional studies revealed an unexpected decrease in mRNA expression of COL1A1 but an increased expression of the BMP2 and BMP4 genes in otosclerotic stapes tissues. Conclusions : Our findings suggest that OTSC is a heterogeneous disorder, but that the GT genotype of the rs1800012 locus is protective and that the TC genotype at the rs17563 locus is a risk factor. In addition, our studies indicate that changes in the expression of the COL1A1 , BMP2 , and BMP4 genes may contribute to the genetic susceptibility of OTSC by regulating their mRNA levels.

Otosclerosis (OTSC) is a late-onset hearing disorder characterized by increased bone turnover in the otic capsule. Disturbed osteoprotegerin expression has been found in the otosclerotic foci which may have an important role in the pathogenesis of OTSC. To identify the genetic risk factors, we sequenced the coding region and exon-intron boundaries of the OPG gene in 254 OTSC patients and 262 controls. Sequence analysis identified five known polymorphisms c.9C>G, c.30+15C>T, c.400+4C>T, c.768A>G, and c.817+8A>C. Testing of these SNPs revealed sex specific association with c.9C>G in males and c.30+15C>T in females after multiple correction. Furthermore, meta-analysis provided evidence of association of the c.9C>G polymorphism with OTSC. In secondary analysis, we investigated the mRNA expression of OPG and associated genes RANK and RANKL in otosclerotic tissues compared to controls. Expression analysis revealed significantly missing/reduced OPG expression only in otosclerotic tissues. However, the signal sequence polymorphism c.9C>G has shown no effect on OPG mRNA expression. In conclusion, our results suggest that the risk of OTSC is influenced by variations in the OPG gene along with other factors which might regulate its altered expression in otosclerotic tissues. Otosclerosis (OTSC) is a common form of acquired hearing loss resulting from disturbed bone remodeling in the otic capsule of the middle ear. Transforming growth factor-beta1 (TGFB1) produced by osteoblasts is the most abundant growth factor in human bone. Previous studies have shown the contribution of single-nucleotide polymorphisms (SNPs) in TGFB1 toward the risk of developing OTSC in some ethnic populations. The present study was aimed at investigating the genetic association and expression profiles of TGFB1 in OTSC patients. Two SNPs (c.-800G > A and c.-509C > T) in the promoter region and three SNPs (c.29T > C, c.74G > C, and c.788C > T) in the coding region were genotyped in 170 cases and 170 controls. The genetic association analysis revealed the significant association between c.-509C > T (p = 0.0067; odds ratio [OR] = 1.562; 95% confidence interval [CI], 1.140-2.139) and OTSC. The increased minor allele "T" frequency in cases (0.42) compared to controls (0.31) indicates its possible role in the etiology of the disease. The minor allele frequencies for the SNPs c.-800G > A, c.29T > C, and c.74G >C were similar among the cases (0.04, 0.47, and 0.08, respectively) and controls (0.05, 0.42, 0.07, respectively). We found that c.788C > T was monomorphic in this population. Interestingly, a four-locus haplotype (G-T-T-G) from these SNPs was found to be significantly associated with OTSC (p = 0.0077). We identified a de novo heterozygous mutation c.-832G > A in the promoter region of TGFB1 in 1 patient. In a secondary analysis, we investigated the possibility of abnormal TGFB1 expression and irregular bone growth in OTSC by expression analysis of TGFB1 mRNA in disease tissue compared to control. We found relatively increased expression of TGFB1 mRNA in the stapes tissues of cases compared to controls (p = 0.0057). In conclusion, this study identified a risk variant c.-509C > T and a risk haplotype G-T-T-G in the TGFB1 gene that contribute toward the susceptibility to OTSC., (© 2013 American Society for Bone and Mineral Research.)

Otosclerosis (MIM 166800) is primarily a metabolic bone disorder of the otic capsule, which leads to bony fixation of the stapedial footplate in the oval window; it is among the most common causes of acquired hearing loss. The etiology of this disease is largely unknown, although epidemiological studies suggest the involvement of both genetic and environmental factors. Recently, a reelin gene, SNP rs3914132, located in intron 2, was shown to be associated with otosclerosis in a European population. When we sequenced blood DNA samples of 85 individuals with otosclerosis and 85 controls, four SNPs of this gene: rs3914131 (P = 0.6463), rs3914132 (P = 0.1822), rs9641319 (P = 0.7371), and rs10227303 (P = 0.5669) were not significantly associated with this disease. In one familial case, a novel variant (C/T) at contig position 2923488 was found to be inherited by the proband and affected family members.

Mutations in the GJC3 gene are known to cause nonsyndromic hearing impairment (NSHI). In this study, we screened for mutations in the connexin 29 (Cx29) gene in peripheral blood collected from patients with NSHI. DNA was extracted from peripheral blood cells of 123 NSHI patients and 127 normal-hearing control subjects. Coding regions of Cx29 were amplified by polymerase chain reaction using primer pairs flanking both exons. Sequences were analyzed and compared with the published Cx29 sequence. On comparison with control subjects, only one patient and her normal-hearing mother showed a novel heterozygous variant in exon 1 c.569T>A (p. Ile190Asn), which most likely represents a rare polymorphism. From the study, we conclude that mutations in the Cx29 gene do not play a role in the causation of NSHI in Indian population.
