Fibroblast growth factor 21 (FGF21) acts as an anti‐atherosclerotic agent. However, the specific mechanisms governing this regulatory activity are unclear. Autophagy is a highly conserved cell stress response which regulates atherosclerosis (AS) by reducing lipid droplet degradation in foam cells. We sought to assess whether FGF21 could inhibit AS by regulating cholesterol metabolism in foam cells via autophagy and to elucidate the underlying molecular mechanisms. In this study, ApoE−/− mice were fed a high‐fat diet (HFD) with or without FGF21 and FGF21 + 3‐Methyladenine (3MA) for 12 weeks. Our results showed that FGF21 inhibited AS in HFD‐fed ApoE−/− mice, which was reversed by 3MA treatment. Moreover, FGF21 increased plaque RACK1 and autophagy‐related protein (LC3 and beclin‐1) expression in ApoE−/− mice, thus preventing AS. However, these proteins were inhibited by LV‐RACK1 shRNA injection. Foam cell development is a crucial determinant of AS, and cholesterol efflux from foam cells represents an important defensive measure of AS. In this study, foam cells were treated with FGF21 for 24 hours after a pre‐treatment with 3MA, ATG5 siRNA or RACK1 siRNA. Our results indicated that FGF21‐induced autophagy promoted cholesterol efflux to reduce cholesterol accumulation in foam cells by up‐regulating RACK1 expression. Interestingly, immunoprecipitation results showed that RACK1 was able to activate AMPK and interact with ATG5. Taken together, our results indicated that FGF21 induces autophagy to promote cholesterol efflux and reduce cholesterol accumulation in foam cells through RACK1‐mediated AMPK activation and ATG5 interaction. These results provided new insights into the molecular mechanisms of FGF21 in the treatment of AS.

BACKGROUND Nectin-4 is overexpressed in several human malignant tumors. This study aimed to investigate the expression of Nectin-4 in esophageal cancer tissues compared with adjacent normal esophageal tissue and its association with clinicopathological parameters and prognosis. MATERIAL AND METHODS Nectin-4 expression in esophageal cancer tissues was compared with adjacent normal esophageal tissue from 94 patients using immunohistochemistry, Western blot, and quantitative reverse transcription-polymerase chain reaction (RT-qPCR). The chi-squared (χ²) test and Fisher's exact test compared categorical variables. The log-rank test and Kaplan-Meier survival analysis assessed the relationship between Nectin-4 expression and overall survival (OS). Univariate and multivariate Cox proportional risk models compared Nectin-4 expression, patient prognosis, and clinicopathological parameters. Diabetic retinopathy is the leading cause of blindness, yet its treatment is very limited. Anti-VEGF drug has been widely applied in ocular disease, but its effects on diabetic retinopathy and the underlying mechanism have remained to be fully explored. To elucidate the role of anti-VEGF treatment, we sought to determine the effects of bevacizumab on diabetic neurovascular changes extending from the 3rd to 9th week with induced diabetes in adult rats. The retinal neurovascular changes included increased expression of VEGF, nNOS, iNOS, eNOS, and NO in the course of diabetes progression. In diabetic rats given bevacizumab injection, the ganglion cell loss and alterations of retinal thickness were ameliorated. In this connection, the immunofluorescence labeling of the above biomarkers was noticeably decreased. Along with this, Western blotting confirmed that bevacizumab treatment was associated with a decrease of VEGF, Flk-1, and cAMP response element binding and protein kinase C protein expression. The present results suggest that bevacizumab treatment in the early stage of the retinopathy may ameliorate the lesions of retinopathy, in which VEGF/Flk-1 signaling has been shown here to play an important role.

Based on the measurements from the US National Data Buoy Center 3-m discus buoy site No. 44004 (38.5°N, 70.47°W) from January 1 to March 31 of 2003, with the COARE algorithm (Version 3.0), the results from four parameterization schemes developed recently for sea surface aerodynamic roughness length were compared with each other. Calculations of frictional speedu*, drag coefficientCd and wind stress τ indicate that the calculated frictional velocities from the four schemes (8.50%–16.20%, the normalized standard error estimate, or NSEE), the computed drag coefficients and wind stress (respectively 15.08%–28.67% and 17.26%–50.59% NSEE) are reasonable. Schemes YT96 and GW03 are consistent. The O02 scheme gives over-estimated values foru* andCd. Schemes TY01 and GW03 display discontinuous characteristics in handling young wave data.

Objective: To investigate and analyze the typical CT findings of salivary gland lymphoepithelial carcinoma. Methods: CT findings in 8 patients with lymphoepithelial carcinoma (LEC) in salivary gland were studied retrospectively. Surgical resection was performed in all cases. Results: 8 cases were subdivided to primary tumor and secondary tumor. In primary tumor group, 5 were localized in the parotid gland, 2 were found in the submandibular glands; 6 lesions had with homogeneous density, 1 was associated with cystic degeneration and 1 with calcification; the margins of lesions in 5 cases were poorly defined, while well-defined in 2. On enhanced CT: obvious enhancement was achieved in all of the 7 solitary lesions, among which 4 were homogenously enhanced while the other 3 had heterogeneous enhancement. In the 2 cases of nodules in deep lobe, retromandibular vein was affected; 7 patients had positive Epstein-Barr virus (EBV) test, with Ki-67 measurements ranged from 40% to 80%. 1 patient had LEC secondary to Benign Lymphoepithelial Lesion (BLEL), with involvement of bilateral parotid glands; the lesion was manifested as multiple differently-sized nodules, with partial cystic change, all of the nodules had clear margins, and obvious ring-shape enhancement was seen in the nodules with cystic change; the patient with secondary LEC had negative EBV test and a Ki-67 value of 20%. Conclusion: In most cases, LEC was primary and occurred in the parotid glands. The probable diagnosis could be made based upon Dual-phase contrast-enhanced CT scan findings combined with positive expression of EBV.
