Contains fulltext : 287914.pdf (Publisher’s version ) (Open Access), Since the beginning of 2020, a remarkably low incidence of respiratory virus hospitalizations has been reported worldwide. We prospectively evaluated 587 children, aged <12 years, admitted for respiratory tract infections from 1 September 2021 to 15 March 2022 in four Italian pediatric hospitals to assess the burden of respiratory viruses during the COVID-19 pandemic in Italy. At admission, a Clinical Respiratory Score was assigned and nasopharyngeal or nasal washing samples were collected and tested for respiratory viruses. Total admissions increased from the second half of October 2021 to the first half of December 2021 with a peak in early November 2021. The respiratory syncytial virus (RSV) incidence curve coincided with the total hospitalizations curve, occurred earlier than in the pre-pandemic years, and showed an opposite trend with respect to the incidence rate of SARS-CoV-2. Our results demonstrated an early peak in pediatric hospitalizations for RSV. SARS-CoV-2 may exhibit a competitive pressure on other respiratory viruses, most notably RSV.

Abstract

In this study we evaluated the antimycotic activity of Achillea ageratum essential oil against Candida albicans. The principal components of the essential oil were yomogi alcohol, 1,8-cineole, artemisia treine and artemisia alcohol. Most strains of C. albicans tested had an MIC 2.25 [micro]g/ml and an MFC between 2.25 and 4.5 [micro]g/ml; the MIC and MFC were low when compared to fluconazole.We also demonstrated that the essential oil was able to affect the major virulence factors of C. albicans; the inhibition of germ tube formation at the sub-inhibitory concentration of 0.29 [micro]g/ml and the inhibition of biofilm formation by over 60% at a concentration of 9 [micro]g/ml. In conclusion, Achillea ageratum essential oil showed a good activity in vitro against C. albicans and may be potentially used as anticandida agent.To support this, further studies are in preparaton to confirm these results. Abstract

Treatment of systemic infection with Candida albicans with a combination of an antifungal agent (i.e. fluconazole) and a thymus-derived immunostimulant (i.e. thymosin α1 (Tα1)) in mice immunosuppressed by morphine treatments was investigated. In normal mice, fluconazole given after infection with 106C. albicans cells was more effective than in mice treated with morphine. Combination treatment with fluconazole and Tα1 prolonged survival and reduced the fungal burden in the kidneys of immunosuppressed mice. We also investigated the influence of this combined treatment on killing properties of polymorphonuclear leucocytes (PMN) and natural killer (NK) cell activity, inhibited by morphine administrations. Treatment with Tα1 or fluconazole as single agents promoted a recovery of normal NK cell activity and intracellular killing of C. albicans by PMN, while the combination significantly increased both of these responses, probably through the modulation of lymphokine production. Our data suggest that the additive effect of Tα1 and fluconazole is due to a direct antifungal action and activation of the immunocompetence. [ABSTRACT FROM AUTHOR]

Abstract

Herpes simplex virus infections are prevalent viral infections in humans. HSVs are also the most common cause of sporadic viral encephalitis (HSE). Magnetic resonance is the imaging method of choice for HSE because it provides the most sensitive method for detecting early lesions. The objective of this study is to set-up and in vitrotest an experimental contrast agent specific for antigens present on HSV-infected cells, bound with a paramagnetic agent detectable by MR imaging. A selected anti-HSV HrFab was labelled with Alexa Fluor 488, 125I and Gd3+Cl6. In order to assess anti-HSV affinity and specificity, ELISA assays were performed. Vero cells infected with HSV strains were visualized by MRI using anti-HSV HrFab/Gd3+Cl6complex. Results of the ELISA tests demonstrated that the anti-HSV HrFab labelled with Gd3+Cl6showed similar affinity for the antigens while the 125I immunoconjugate showed reduced affinity. MRI confirmed high affinity and specificity of antibody for the detection of HSV infections.
