Abstract

Predator–prey relationships can influence community processes, and a rich prey spectrum is important to favour carnivore conservation, as well as to buffer single prey towards intensive predation. Antipredator behavioural responses can occur and can be dynamic in time and space, which may generate counter‐responses in predators. However, data are scarce on their role in modulating carnivore diet and behaviour. Data are especially needed for European landscapes that are largely anthropized and have been recently recolonized by large carnivores. In a protected area in central Italy recently recolonized by the wolf and hosting a rich community of wild ungulates, we studied the interactions between this predator and three ungulate species. At the initial stage of wolf recovery, the fallow deer and the wild boar were the main prey, while the roe deer was a minor food item. Through camera‐trapping and predator food habits, we assessed temporal changes in wolf–prey relationships throughout 5 years (2017–2022). Wolf detection rates were spatially associated with those of fallow deer and wild boar, but shrub cover was positively related to predator and negatively to prey, suggesting possible prey avoidance of sites with lower visibility and greater predation risk. Throughout the years, the fallow deer increased its diurnal activity, with a decreasing temporal overlap with the predator. The wolf showed crepuscular/nocturnal activity, with an increased synchronization with the wild boar, which replaced the fallow deer as first prey. No support for major spatiotemporal responses was reported for wild boar and roe deer. With the ongoing recovery of carnivores across Europe, conservation priorities may emphasize the need to maintain an efficient ecological role of predators. Our results support the role of antipredator responses in modulating predator behaviour and diet and emphasize the importance of a diverse spectrum of wild prey to ensure the conservation of the ecological role of carnivores. Abstract

In (Ferrucci, Pacini and Sessa, 1995) an extended form of resolution, called Reduced SLD resolution (RSLD), is introduced. In essence, an RSLD derivation is an SLD derivation such that redundancy elimination from resolvents is performed after each rewriting step. It is intuitive that redundancy elimination may have positive effects on derivation process. However, undesiderable effects are also possible. In particular, as shown in this paper, program termination as well as completeness of loop checking mechanisms via a given selection rule may be lost. The study of such effects has led us to an analysis of selection rule basic concepts, so that we have found convenient to move the attention from rules of atom selection to rules of atom scheduling. A priority mechanism for atom scheduling is built, where a priority is assigned to each atom in a resolvent, and primary importance is given to the event of arrival of new atoms from the body of the applied clause at rewriting time. This new computational model proves able to address the study of redundancy elimination effects, giving at the same time interesting insights into general properties of selection rules. As a matter of fact, a class of scheduling rules, namely the specialisation independent ones, is defined in the paper by using not trivial semantic arguments. As a quite surprising result, specialisation independent scheduling rules turn out to coincide with a class of rules which have an immediate structural characterisation (named stack-queue rules). Then we prove that such scheduling rules are tolerant to redundancy elimination, in the sense that neither program termination nor completeness of equality loop check is lost passing from SLD to RSLD., Comment: 53 pages, to appear on TPLP

Abstract

Acylation stimulating protein (ASP) is a hormone produced by adipocytes and is of importance for the storage of energy as fat. We examined whether ASP might also have effects on islet function. In clonal INS-1 cells, ASP dose-dependently augmented glucose-stimulated insulin secretion. The lowest effective dose of ASP at 10 mmol/l glucose was 5 micro mol/l. The effect was glucose-dependent because ASP did not increase insulin secretion at 1 mmol/l glucose but had clear effect at 10 and 20 mmol/l glucose. Similarly, ASP augmented glyceraldehyde-induced insulin secretion but the hormone did not enhance insulin secretion in response to depolarization by 20 mmol/l of KCl. ASP-induced insulin secretion was completely abolished by competitive inhibition of glucose phosphorylation by glucokinase with 5-thio-glucose and was partially inhibited by the calcium channel blocker, nifedipine, and by the protein kinase C inhibitor, GF109203. Furthermore, thapsigargin, an inhibitor of Ca(2+)-ATPase in the endoplasmic reticulum, did not affect ASP-induced insulin secretion. ASP (>5 micro mol/l) also augmented glucose-stimulated insulin secretion from islets isolated from C57BL/6J mice, and intravenous administration of ASP (50 nmol/kg) augmented the acute (1 and 5 min) insulin response to intravenous glucose (1 g/kg) in C57BL/6J mice. This was accompanied by an increased rate of glucose disposal. Minimal model analyses of data derived from the intravenous glucose tolerance test revealed that whereas ASP augmented insulin secretion, the hormone did not affect insulin sensitivity (S(I)) or glucose effectiveness (S(G)). We conclude that ASP augments glucose-stimulated insulin secretion through a direct action on the islet beta cells. The effect is dependent on glucose phosphorylation, calcium uptake and protein kinase C. Stimulation of insulin secretion by ASP in vivo results in augmented glucose disposal.

Abstract

IntroductionChagas disease is caused by theTrypanosoma cruziinfection. It is a neglected tropical disease with considerable impact on the physical, psychological, familiar, and social spheres. The Valle Alto of Cochabamba is a hyperendemic region of Bolivia where efforts to control the transmission of the disease have progressed over the years. However, many challenges remain, above all, timely detection and health-care access.MethodsFollowing the Science Shop process, this bottom-up research emerged with the participation of the civil society from Valle Alto and representatives of the Association ofCorazones Unidos por el Chagasfrom Cochabamba. The aim of this study is to explore the social determinants in the living realities of those affected by Chagas disease or the silent infection and how families in the Valle Alto of Cochabamba cope with it. An interdisciplinary research team conducted a case study of the life stories of three families using information from in-depth interviews and performed a descriptive qualitative content analysis and triangulation processes.FindingsFindings provide insights into social circumstances of the research subjects’ lives; particularly, on how exposure toTrypanosoma cruziinfection affects their daily lives in terms of seeking comprehensive health care. Research subjects revealed needs and shared their experiences, thus providing an understanding of the complexity of Chagas disease from the socioeconomic, sociocultural, political, and biomedical perspectives. Results enlighten on three dimensions: structural, psychosocial, and plural health system. The diverse perceptions and attitudes toward Chagas within families, including the denial of its existence, are remarkable as gender and ethnocultural aspects. Findings support recommendations to various stakeholders and translation materials.ConclusionsIntersectional disease management and community involvement are essential for deciding the most appropriate and effective actions. Education, detection, health care, and social programs engaging family units ought to be the pillars of a promising approach.

24. Management of Small Bowel Obstruction (SBO) in older adults (>80 years): a propensity scorematched analysis on predictive factors for a (un) successful non-operative management (NOM).

Abstract

OBJECTIVE: Small Bowel Obstruction (SBO) is a common emergency in older patients. The most appropriate treatment strategy is still matter of debate. The aim of this study was to compare a non-operative management (NOM) vs. a surgical procedure for patients ≥ 80 years with SBO. PATIENTS AND METHODS: All patients ≥ 80 years admitted to our Emergency Department (ED) for SBO between January 1st, 2015, and December 31st, 2020 were included in this study. In order to correct for baseline covariates and factors associated to clinical management, we used a 1:1 propensity score matching (PSM) analysis. The primary outcome was to compare the overall in-hospital mortality. Secondary outcomes included occurrence of major complications and in-hospital length of stay (LOS). RESULTS: A total of 561 patients were enrolled. After propensity score matching (PSM) analysis, 302 patients (151 each group) were included in the analysis. Mortality did not differ between the two groups. After PSM mechanical ventilation, sepsis, cumulative major complications, and LOS were significantly higher in the operative treatment group [15.9% vs. 1.5%, 9.4% vs. 4.1%, 27.6% vs. 19.2%, and 9.4 (6.4-14.3) days vs. 8.1 (4.5-13.3) days, respectively; p<0.001, p=0.013, p=0.025, and p=0.003, respectively]. CONCLUSIONS: In patients ≥ 80 years with SBO, a NOM could yield similar results, in terms of overall mortality, compared to a surgical management. Thus, particularly in patients with multiple comorbidities or functional impairments, a conservative approach should always be considered. [ABSTRACT FROM AUTHOR]

Abstract

Plasma gamma-glutamyltransferase (GGT) was suggested to reflect the level of systemic oxidative stress. Oxidative stress induces changes in arterial structure and function and contributes to the development of hypertension. Therefore, GGT may be associated with arterial remodeling and blood pressure (BP) increment, even in absence of disease. To test this hypothesis, we evaluated, in 825 healthy subjects at low cardiometabolic risk, the associations of plasma GGT with carotid artery intima-media thickness (IMT), luminal diameter and prehypertension; in 154 subjects was evaluated also the association with aortic stiffness (cfPWV). Associations were controlled for insulin sensitivity, C-reactive protein, and life-style habits. In the main population, BP was remeasured after 3 years. Carotid diameter and cfPWV, but not IMT, were directly and independently related to plasma GGT. Subjects with prehypertension (N = 330) had higher GGT as compared with subjects with normal BP (22 [14] vs 17 [11] IU/L; adjusted P = 0.001), and within prehypertensive subjects, those who developed hypertension during 3 years had higher GGT than those without incident hypertension (27 [16] vs 21 [14] IU/L; adjusted P < 0.05). Objective Women with polycystic ovary syndrome (PCOS) have higher circulating levels of C-reactive protein, but the relationship between inflammation and endocrine function in PCOS remains poorly understood. Thus, this study aimed to investigate the association between low-grade inflammation and sex hormones in women with PCOS. Design and Patients A comprehensive panel of biomarkers of inflammation was measured in serum of 63 women with PCOS using proximity extension assay technology. Associations of 65 biomarkers with sex hormones were assessed without and with adjustment for age and body mass index (BMI). Abstract

Glucagon-like peptide-1 (GLP-1) plays a significant role in glucose homeostasis through its incretin effect on insulin secretion. However, GLP-1 also exhibits extrapancreatic actions, and in particular its possible influences on insulin sensitivity are controversial. To study the dynamic action of GLP-1 on insulin sensitivity, we applied advanced statistical modeling methods to study glucose disappearance in mice that underwent intravenous glucose tolerance test with administration of GLP-1 at various dose levels. In particular, the minimal model of glucose disappearance was exploited within a population estimation framework for accurate detection of relationships between glucose disappearance parameters and GLP-1. Minimal model parameters were estimated from glucose and insulin data collected in 209 anesthetized normal mice after intravenous injection of glucose (1 g/kg) alone or with GLP-1 (0.03-100 nmol/kg). Insulin secretion markedly increased, as expected, with increasing GLP-1 dose. However, minimal model-derived indexes, i.e., insulin sensitivity and glucose effectiveness, did not significantly change with GLP-1 dose. Instead, fractional turnover rate of insulin action [[P.sub.2] = 0.0207 [+ or -] 24.3% (min) at zero GLP-1 dose] increased steadily with administered GLP-1 dose, with significant differences at 10.4 nmol/kg ([P.sub.2] = 0.040 [+ or -] 15.5%, P = 0.0046) and 31.2 nmol/kg ([P.sub.2] = 0.050 [+ or -] 29.2%, P = 0.01). These results show that GLP-1 influences the dynamics of insulin action by accelerating insulin action following glucose challenge. Abstract

In phenotype experiments in mice, determination of dynamic insulin sensitivity often uses the insulin tolerance test. However, the interpretation of this test is complicated by the counterregulation occurring at low glucose. To overcome this problem, we determined the dynamic insulin sensitivity after inhibition of endogenous insulin secretion by diazoxide (25 mg/kg) in association with intravenous administration of glucose plus insulin (the DSGIT technique). Estimation of insulin sensitivity index ([S.sub.r]) by this technique showed good correlation to [S.sub.1] from a regular intravenous glucose tolerance test (r = 0.87; P < 0.001; n = 15). With DSGIT, we evaluated dynamic insulin sensitivity in mice with a rat insulin promoter ([beta]-cell-targeted) dominant-negative mutation of hepatic nuclear factor (HNF)-l[alpha] [RIP-DN HNF-l[alpha] (Tg) mice]. When insulin was administered exogenously at the same dose in Tg and wild-type (WT) mice, plasma insulin levels were higher in WT, indicating an increased insulin clearance in Tg mice. When the diazoxide test was used, different doses of insulin were therefore administered (0.1 and 0.15 U/kg in WT and 0.2 and 0.25 U/kg in Tg) to achieve similar insulin levels in the groups. Minimal model analysis showed that [S.sub.I] was the same in the two groups (0.78 [+ or -] 0.21 x [10.sup.-4] min*[pmol.sup.-1]*[l.sup.-1] in WT vs. 0.60 [+ or -] 0.11 in Tg; P = 0.45) as was the glucose elimination rate (P = 0.27). We conclude that 1) the DSGIT technique determines the in vivo dynamic insulin action in mice, 2) insulin clearance is increased in Tg mice, and 3) chronic islet dysfunction in RIP-DN HNF-l[alpha] mice is not compensated with increased insulin sensitivity. glucose effectiveness; glucose tolerance; minimal modeling; diazoxide; insulin action
