The sub-Antarctic Magellan Ecoregion is a unique biogeographic area located in the southern end of South America with notable marine flora and high endemism. Brown macroalgae, particularly Macrocystis pyrifera, dominate the coastlines and are considered critical ecosystem engineers, providing shelter, food, and reproduction sites for various species. Additionally, they are essential components used in the food industry and biomedicine due to their lipids, amino acids, and fiber content. In this study, we determined the fatty acid content in different thallus structures (holdfast, stipes and fronds) of Macrocystis pyrifera collected in Rinconada Bulnes (53°35ʼ47.76” S; 70°56ʼ08.52” W) in the spring of 2021. The stipes had a highest total lipid content (3.73%) than the fronds (2.74%). The fatty acid profile showed higher values of monounsaturated fatty acids in the stipe (Ʃ 43.0%) and holdfast (Ʃ 41.7%), while fronds displayed higher values of polyunsaturated fatty acids (Ʃ 32.4%). This study provides insights into the nutritional and biomedical potential of M. pyrifera and highlights the importance of understanding the distribution of fatty acids in different thallus structures.

Inflammatory bowel diseases (IBDs), which include ulcerative colitis (UC) and Crohn’s disease (CD), cause chronic inflammation of the gut, affecting millions of people worldwide. IBDs have been frequently associated with an alteration of the gut microbiota, termed dysbiosis, which is generally characterized by an increase in abundance of Proteobacteria such as Escherichia coli, and a decrease in abundance of Firmicutes such as Faecalibacterium prausnitzii (an indicator of a healthy colonic microbiota). The mechanisms behind the development of IBDs and dysbiosis are incompletely understood. Using samples from colonic biopsies, we studied the mucosa-associated intestinal microbiota in Chilean and Spanish patients with IBD. In agreement with previous studies, microbiome comparison between IBD patients and non-IBD controls indicated that dysbiosis in these patients is characterized by an increase of pro-inflammatory bacteria (mostly Proteobacteria) and a decrease of commensal beneficial bacteria (mostly Firmicutes). Notably, bacteria typically residing on the mucosa of healthy individuals were mostly obligate anaerobes, whereas in the inflamed mucosa an increase of facultative anaerobe and aerobic bacteria was observed. We also identify potential co-occurring and mutually exclusive interactions between bacteria associated with the healthy and inflamed mucosa, which appear to be determined by the oxygen availability and the type of respiration. Finally, we identified a panel of bacterial biomarkers that allow the discrimination between eubiosis from dysbiosis with a high diagnostic performance (96% accurately), which could be used for the development of non-invasive diagnostic methods. In the pathogen Typanosoma cruzi, the calcium ion (Ca2+) regulates key processes for parasite survival. However, the mechanisms decoding Ca2+ signals are not fully identified or understood. Here, we investigate the role of a hypothetical Ca2+-binding protein named TcCAL1 in the in vitro life cycle of T. cruzi. Results showed that the overexpression of TcCAL1 fused to a 6X histidine tag (TcCAL1-6xHis) impaired the differentiation of epimastigotes into metacyclic trypomastigotes, significantly decreasing metacyclogenesis rates. When the virulence of transgenic metacyclic trypomastigotes was explored in mammalian cell invasion assays, we found that the percentage of infection was significantly higher in Vero cells incubated with TcCAL1-6xHis-overexpressing parasites than in controls, as well as the number of intracellular amastigotes. Additionally, the percentage of Vero cells with adhered metacyclic trypomastigotes significantly increased in samples incubated with TcCAL1-6xHis-overexpressing parasites compared with controls. In contrast, the differentiation rates from metacyclic trypomastigotes to axenic amastigotes or the epimastigote proliferation in the exponential phase of growth have not been affected by TcCAL1-6xHis overexpression. Based on our findings, we speculate that TcCAL1 exerts its function by sequestering intracellular Ca2+ by its EF-hand motifs (impairing metacyclogenesis) and/or due to an unknown activity which could be amplified by the ion binding (promoting cell invasion). This work underpins the importance of studying the kinetoplastid-specific proteins with unknown functions in pathogen parasites.

La caracterización de las prioridades presupuestarias en la pesca y acuicultura son importantes para optimizar la asignación y ejecución de sus recursos presupuestarios en Perú en beneficio de sus actores. Con base en la metodología del Estimado del Apoyo a la Pesca (FSE, por sus siglas en inglés), este documento caracteriza las prioridades de inversión y gasto público y analiza su consistencia con los instrumentos de gestión en el sector. Se identificó que el valor del apoyo a este sector se incrementó de PEN 235,2 millones en 2016 a PEN 376,4 millones en 2020. Además, a pesar de la volatilidad en la inversión pública, se destacó los esfuerzos orientados a investigación y desarrollo, ordenamiento pesquero e infraestructura. No obstante, todavía persisten varios problemas en el sector, a pesar de que el apoyo de Perú en relación con su PBI no es bajo en comparación a otros países que son potencias pesqueras. Ante estos hallazgos, parece ser que hay un problema respecto al uso poco eficiente de los recursos presupuestarios.

Background Anti-PD-(L)1 blocking agents can induce immune-related adverse events (irAEs), which can compromise treatment continuation. Since circulating leukocyte-platelet (PLT) complexes contribute to inflammatory and autoimmune diseases, we aimed to analyze the role of these complexes as predictors of irAEs in non-small cell lung cancer (NSCLC) patients receiving anti-PD-(L)1. Materials and methods Twenty-six healthy donors (HD) and 87 consecutive advanced NSCLC patients treated with anti-PD-(L)1 were prospectively included. Percentages of circulating leukocyte-PLT complexes were analyzed by flow cytometry and compared between HD and NSCLC patients. The association of leukocyte-PLT complexes with the presence and severity of irAEs was analyzed. Results NSCLC patients had higher percentages of circulating leukocyte-PLT complexes. Higher percentages of monocytes with bound PLT (CD14 + PLT +) were observed in patients who received prior therapies while CD4 + T lymphocytes with bound PLT (CD4 + PLT +) correlated with platelets counts. The CD4 + PLT + high percentage group presented a higher rate of dermatological irAEs while the CD4 + PLT + low percentage group showed a higher rate of non-dermatological irAEs (p < 0.001). A lower frequency of grade >= 2 irAEs was observed in the CD4 + PLT + high percentage group (p < 0.05). Patients with CD4 + PLT + low and CD14 + PLT + high percentages presented a higher rate of grade >= 3 irAEs and predominantly developed non-dermatological irAEs (p < 0.01). Conclusions Our results suggest that circulating leukocyte-PLT complexes and the combination of CD4 + PLT + and CD14 + PLT + percentages can be used as a predictive biomarker of the development and severity of irAEs in advanced NSCLC patients receiving anti-PD-(L)1 agents.

Background: Diarrheagenic Escherichia coli (DEC) strains are a main cause of diarrhea worldwide in children under 5 years old. DEC virulence is strongly regulated by environmental conditions and metabolites produced by the gut microbiota in the intestinal tract. In this study, we evaluated changes in gut microbiota-metabolome in children with or without diarrhea produced by DEC pathotypes. Goal: To determine gut microbiota composition and metabolome in stool samples obtained from healthy children and children with diarrhea positive for DEC pathotypes. Methods: We analyzed a total of 16 age-paired stool samples: 8 diarrheal samples positive for one DEC pathotype and 8 stool samples from healthy children. To identify the microbiota composition, we sequenced the V3-V4 region of the 16S rRNA and determined operational phylogenetic units (OPU). OPU were then used to predict metabolic pathways using the PICRUSt2 software. The presence of metabolites in stool samples was determined by LC-MS. A correlation analysis was performed with the main genera from each group and main metabolites. Bacteria associated with variance of main metabolites were identified using the MIMOSA2 software. Results: DEC and healthy groups showed a statistically different microbiota composition. A decrease in Firmicutes together with an increase in Bacteroidetes and Proteobacteria was found in the DEC group compared to the healthy group. Metabolic pathway predictions based on microbiota diversity showed that pathways involved in histidine and L-ornithine metabolism were significantly different between groups. A total of 88 metabolites detected by LC-MS were included in the metabolome analysis. We found higher levels of histamine and lower levels of ornithine in DEC samples than in the healthy group. Histamine and L-ornithine were associated with a specific microbiota species and the corresponding metabolic pathways. Conclusion: Stool samples from healthy children and children positive for DEC displayed a differential metabolome and microbiota composition. A strong correlation between a gut microbiota species and certain metabolites, such as histamine and L-ornithine, was found in the DEC group. This information might be useful to identify mechanisms and signaling molecules involved in the crosstalk between microbiota and DEC pathotypes.

The global shortage of reagents and kits for nucleic acid extraction and molecular detection of SARS-CoV-2 requires new cost-effective strategies for the diagnosis of suspected COVID-19 cases, especially in countries that need to increase detection capacity. Pooled nucleic acid testing has been extensively used as a cost-effective strategy for HIV, HepB, HepC and influenza. Also, protocols dispensing of RNA extraction appears as an attractive option for detection of SARS-CoV-2. In this study, we found that pooling of 5 samples showed that CT variations were in the range of 1.0-4,5 units, with less likelihood of a false negative result. Results of the sample without nucleic acid ex-traction, was unsatisfactory, with a significant increase in CT values, and thus for risk of a false negative result. In conclusion, pooling nasopharyngeal samples with both automated and manual extraction proved reliable, and thus a potential efficient alternative for the diagnosis of suspected COVID-19 in developing countries.

espanolAntecedentes: En Chile, el 16 % de la poblacion son mayores de 60 anos; de ellos el 25 % tiene algun grado de dependencia funcional y el 10 % dependencia severa. En este contexto, una de las problematicas de los equipos de salud es la definicion de objetivos de cuidado cuando el paciente no los puede manifestar, decision que podria facilitarse definiendo directrices anticipadas. Material y metodos: Estudio transversal, descriptivo. Incluyo pacientes de 40-60 anos que recibieron atencion entre septiembre-octubre de 2017 en el Hospital San Jose, ubicado en Santiago de Chile. Se aplico una encuesta sobre el conocimiento de riesgo de dependencia funcional y sus voluntades anticipadas. Resultados: 150 pacientes, edad media 51,4 anos, 56 % son solteros/divorciados, promedio 1,2 hijos y escolaridad media 46 %. Las patologias mas prevalentes fueron hipertension arterial del 50 % y diabetes del 30 %. El 40 % tenia autopercepcion de salud regular. El 50 % refirio conocer sus patologias cronicas y riesgo de dependencia asociado. Respecto a las preferencias anticipadas en el hipotetico caso de dependencia severa en el futuro, el 60 % escogeria un familiar como cuidador, el 72 % preferiria vivir en domicilio propio y el 55,3 % rechazaria reanimacion en caso de parada cardiorrespiratoria. No se encontro correlacion estadistica entre escolaridad y conocimiento de estado de salud, riesgo de dependencia asociado a sus enfermedades ni tampoco con la decision de recibir reanimacion en caso de parada cardiorrespiratoria. Conclusiones: De nuestra muestra podemos interpretar que existe una brecha respecto a lo que significa envejecimiento satisfactorio, manejo adecuado de patologias cronicas potencialmente invalidantes y elaboracion de directrices anticipadas por quienes las padecen. EnglishBackground: In Chile, 16 % of the population are over the age of 60; of these, 25 % have some degree of functional dependence and 10 % have severe dependence. In this context, one of the problems of health teams is the definition of objectives of care when the patient cannot express them, a decision that could facilitate defining directives. Material and methods: A cross-sectional, descriptive study. It included patients aged 40-60 years who received care between September and October of 2017 at San Jose Hospital, Santiago de Chile. A survey on the knowledge of their functional dependence risk was administered, covering their advance preferences. Results: A total of 150 patients participated with an average age of 51.4 years; 56 % were single/divorced. They had 1.2 children on average, and a mean level of schooling of 46 %. The most prevalent diseases were hypertension (50 %) and diabetes (30 %). Forty percent perceived their health status as fair; 50 % claimed to be aware of their chronic diseases and their associated risk of dependence. Regarding the advance preferences, in the hypothetical situation of severe dependence, 60 % would choose a relative as caregiver, 72 % would prefer to live in their own home, and 55.3 % rejected resuscitation in case of cardiac arrest. There was no statistical correlation between schooling and awareness of health status, risk of dependence associated with their illnesses, or the decision to receive resuscitation in case of cardiac arrest. Conclusions: From our sample we may interpret that there is a gap regarding the meaning of successful aging, correct management of potentially invalidating chronic diseases, and preparation of advance directives by those who suffer from them.

Vertical agriculture has been showed strong advantages in the production culture. At the same time, have contributed in the conditionate of architecture of the cities. Many troubles can be treated with the implantation of vertical farm in the cities. Between the most important are: obtain vegetables in site, as a consequence it is not necessary transportation. If the production is a hydroponics, there are not drainage contamination. Water use is strong reduced, etc. The idea is to establishment a utility model, slow cost and easy to operate, able to integrate in the wall of each small house and produce during all year some species of vegetables. The objective of this work is to analyze beginning from a technological point of view and sustainability, and evaluate conditions to adapt the system “vertical farm” in the big city (Mexico city). This analysis pretends contribute to the adaptability from traditional farm to “indoor farm” with emphasis in the efficient use of the resources such as water, soil, energy and environment. All together considering economic issues and the effort in order to change process at local scale.

The Patagonian toothfish Dissostichus eleginoides is one of the most important fisheries from the Southern Ocean. The biology of this species is relatively well studied and some nutritionals issues have also been reported; however there is no information about the composition of the bacterial community of the gastrointestinal tract, which is essential to characterize the microbiota of this fish. The bacterial flora of D. eleginoides is here described for the first time using culturable methods. By applying traditional culture-based techniques and 16S rDNA sequencing methods it was possible to characterize the families Vibronaceae and Moraxellaceae, which were mainly represented by Vibrio and Psychrobacter, respectively. This Patagonian fish shows a microbiota very similar to other cold waters fishes.

Objectives Ki-67 is a proliferation marker in prostate cancer. A prognostic RNA signature was developed to characterize prostate cancer aggressiveness. The aim was to evaluate prognostic correlation of CCP and Ki-67 with biochemical failure (BF), and survival in high-risk prostate cancer patients (pts) treated with radiation therapy (RT). Methods CCP score and Ki-67 were derived retrospectively from pre-treatment paraffin-embedded prostate cancer tissue of 33 men diagnosed from 2002 to 2006. CCP score was calculated as an average expression of 31 CCP genes. Ki-67 was determined by IHC. Single pathologist evaluated all tissues. Factors associated to failure and survival were analyzed. Results Median CCP score was 0.9 (-0-1 – 2.6). CCP 0: 1 pt; CCP 1: 19 pts; CCP 2: 13 pts. Median Ki-67 was 8.9. Ki-67 cutpoint was 15.08%. BF and DSM were observed in 21% and 9%. Ki-67 ≥ 15% predicted BF ( p = 0.043). With a median follow-up of 8.4 years, 10-year BF, OS, DM and DSM for CCP 1 vs. CCP 2 was 76–71% ( p = 0.83), 83–73% ( p = 0.86), 89–85% ( p = 0.84), and 94–78% ( p = 0.66). On univariate, high Ki-67 was correlated with BF ( p = 0.013), OS ( p = 0.023), DM ( p = 0.007), and DSM ( p = 0.01). On Cox MVA, high Ki-67 had a BF trend ( p = 0.063). High CCP score was not correlated with DSM. Conclusions High Ki-67 significantly predicted outcome and provided prognostic information. CCP score may improve accuracy stratification. We did not provide prognostic correlation of CCP and DSM. It should be validated in a larger cohort of pts.

The Scapula Reposition Test (SRT) is proposed to determine if a relationship exists between scapular dyskinesis and shoulder pathology. The purpose of this study was to evaluate intra-rater and inter-rater reliability of the SRT in subjects with rotator cuff tendinopathy and scapular dyskinesis. In addition, we compared subjective strength findings from the test to an objective measure made by dynamometry. The SRT was independently and randomly performed by two physical therapists in 42 subjects. The percent agreement, Cohen&rsquo, s kappa (&kappa, ), maximum attainable &kappa, prevalence and bias indexes, and prevalence-adjusted-bias-adjusted kappa were used as intra- and inter-rater reliability estimates. Finally, the point-biserial correlation coefficient (rpb) was used for correlation analysis of objective and subjective strength findings. A moderate intra-rater (&kappa, = 0.43, CI 95%, 0.14 to 0.73, p = 0.004) and poor inter-rater (&kappa, = 0.08, CI 95%, &minus, 0.22 to 0.38, p = 0.61) agreement was found. Subjective strength changes during SRT and dynamometry were poorly correlated (rpb = 0.137, 0.175 to 0.423, p = 0.389). The SRT cannot be recommended for clinical practice. More studies evaluating its reliability are needed as well as further research on the capability of a rater to manually detect strength changes.

e23525 Background: Leiomyosarcomas (LMS) are soft tissue sarcomas that derive from smooth muscle cells and can arise anywhere in the body (most frequently in extremities, uterus or retroperitoneum). The clinical prognostic factors are well established but molecular factors influencing prognostic are unknown. The DMD gene, which codifies for the dystrophin protein, has been proposed as tumour suppressor gene in LMS. The aim of our study is to evaluate dystrophin expression in LMS and its relation with the patient prognosis. Methods: A total of 103 patients (pts) with LMS were analysed. Clinical and pathological data were collected retrospectively from patients’ electronic board system. Dystrophin expression was analysed by immunohistochemistry (IHC) in paraffin embedded tissue LMS samples using the Novocastra NCL-DYS2 (Leica Biosystems). Semiquantitative assessment of the staining was classified from score 0 to 3 (0 = no dystrophin expression, 1 = low expression, 2 = moderate expression, 3 = high expression). Results: Of all 103 pts, 70 (68%) had localized disease and the majority of tumours were larger than 5 cm (75%). The most frequent locations of primary LMS were extremities (n = 31; 30.1%), uterus (n = 23; 23.2%) and retroperitoneum (n = 21; 20.4%). Most LMS were high grade (G): 7 pts G1 (6.8%), 35 pts G2 (34%) 2 and 53 pts G3 (51.5%). 50 of all grade LMS (48.6%) had loss of dystrophin expression (score 0), 17 (16.5%) had score 1, 23 (22.3%) score 2 and 10 (9.7%) score 3. Loss/low dystrophin expression measured as score 0 or 1, was more frequent in grade 3 LMS compared to grade 2 or grade 1 (77.4% vs. 57.1% vs. 16.7%; p = 0.005). There were no differences in dystrophin loss between localized or metastatic disease at diagnosis (64.2% vs. 72.2%; p > 0.05). In our series, loss or reduced dystrophin expression did not correlate with the risk of relapse in localized patients or overall survival in metastatic patients. Conclusions: Loss of dystrophin expression is a common event in LMS. Loss/low dystrophin expression is more frequent in grade 3 LMS compared to grade 2 and grade 1 LMS. Loss of dystrophin expression did not correlate with risk of relapse or overall survival in our series. Additional genetic evaluations to study DMD in LMS are underway.

1582 Background: Despite the use of clinical eligibility criteria and mutation predictive models, a great proportion of families are negative for germline mutations in BRCA1/2 genes. Traditionally, risk assessment of inconclusive results included the recommendation of high-risk surveillance protocol, the update of incident cancer cases in the family and the consideration of additional testing to rule out the possibility of phenocopy. More recently, next generation sequencing multigene panels have become a standard practice in cancer genetics clinics worldwide. We addressed the value of multigene panel retesting of BRCA1/2 negative HBOC families in our institution. Methods: After genetic counseling session and informed consent, a total of 137 individuals (119 probands and 18 extra cancer-affected relatives) from distinct BRCA1/2 negative families were retested using a panel containing 11 breast and ovarian cancer susceptibility genes ( BRCA1/2, PALB2, ATM, CHEK2, PTEN, TP53, STK11, BRIP1, RAD51C, RAD51D). Results: According to the BOADICEA model, the remaining probability of mutation in BRCA1/2 or PALB2 genes in our cohort was 5.5% (0.1-61). The reasons for considering retesting were the addition of any incident cancer diagnosis in 33 cases (24%), a prior study with a low sensitivity screening technique (dHPLC) in 6 families (5%) and the expansion of the study to other putative breast and ovarian susceptibility genes in 98 families (71%). Overall, 3 pathogenic (2 BRCA2, 1 CHEK2) and 8 likely pathogenic variants (1 BRCA2, 4 CHEK2 and 3 ATM) were found. The prevalence was 8%. The detection rate among 19 families with a > 10% remaining probability of mutation in BRCA1/2 and PALB2 genes was 26%. The 3 clinically significant variants in BRCA2 were detected in 2 families and 1 updated cancer family history (BOADICEA remaining probability of 59, 61 and 12%, respectively). Cascade testing was subsequently done in 15 relatives resulting 8 in mutation carriers and 9 true negatives. Conclusions: Our results support the value of updating cancer incident cases and considering expanded panels in selected families.

Introduction: Compared to bovine formula (BF), breast milk (BM) has unique properties. In the newborn intestine, there is a homeostatic balance between the counterparts of the immune system, which allows a physiological inflammation, modulated by the gut microbiota. Many studies have attempted to understand the effect of BF vs. BM, and the changes in the gut microbiota, but few also focus on intestinal inflammation. Methods: We conducted a cohort study of newborn infants during their first 3 months. In stool samples taken at 1 and 3 months (timepoints T1 and T3), we quantified calprotectin, IL-8 and α1-antitrypsin by ELISA and we evaluated the expression of IL8 and IL1β genes by RT-qPCR. To determine the microbiota composition, the 16S rRNA gene was amplified and sequenced using 454 pyrosequencing. Sequences were clustered into operational taxonomic units (OTUs). Results: In total 15 BM and 10 BF infants were enrolled. In the BM group, we found calprotectin and α1-antitrypsin levels were significantly elevated at T3 compared to T1; no differences were found between T1 and T3 in the BF group. A comparison between the BM and BF groups showed that calprotectin levels at T1 were lower in the BM than the BF group; this difference was not observed at T3. For IL-8 levels, we found no differences between groups. A gene expression analysis of the IL8 and IL1β genes showed that infants from the BF group at T1 have a significantly increased expression of these markers compared to the BM group. Gut microbiota analyses revealed that the phylum Bacteroidetes was higher in BM than BF, whereas Firmicutes were higher in BF. A redundancy analysis and ANOVA showed BM has a community structure statistically different to BF at T1 but not at T3. Compared to BF, BM at T1 showed a higher representation of Enterococcus, Streptococcus, Enterobacter, Lactococcus, and Propionibacterium. Conclusions: We found a basal state of inflammation in the infants' intestine based on inflammation markers. One month after birth, infants receiving BF exhibited higher levels of inflammation compared to BM.

The Patagonian toothfish or Chilean sea bass (Dissostichus eleginoides), found in the Southern Ocean surrounding Antarctica, is an important fishery species for Chile. This high-value species is regarded as overfished, making it an attractive target for aquaculture. When developing a reproduction program for any aquaculture species, it is important to implement genetic tools to evaluate diversity, inbreeding, and parentage. We calculated genetic diversity and paternity/maternity exclusion probabilities based on five commonly-used microsatellite loci in a natural population of Patagonian toothfish from southern Chile (n = 34) in order to evaluate the potential utility of these five markers in stock management. The observed number of alleles per locus (Na) and observed heterozygosities (H O) are within range as described by studies performed in other sub-Antarctic regions. All five loci were strongly polymorphic, with H O > 0.6 and Na > 7, and paternity exclusion probabilities were high (PE > 0.99), indicating the potential utility of these loci in paternity/maternity exclusion analyses of Patagonian toothfish.

Resumen Fundamento y objetivo Analizar la posible utilidad y capacidad de la procalcitonina (PCT) para pronosticar la existencia de bacteriemia en los pacientes con neumonia adquirida en la comunidad (NAC) producida o no por Streptococcus pneumoniae (S. pneumoniae). Pacientes y metodo Estudio observacional, prospectivo y descriptivo de pacientes diagnosticados de NAC en un servicio de urgencias (SU). Se recogieron variables sociodemograficas, de comorbilidad, el indice de Charlson, grados del Pneumonia Severity Index y criterios de NAC grave, estudios microbiologicos y determinaciones analiticas y de los biomarcadores (PCT y proteina C reactiva). Se realizo seguimiento durante 30 dias. Se calculo el poder pronostico y el rendimiento diagnostico de bacteriemia originada o no por S. pneumoniae. Resultados Se incluyeron 474 pacientes. Los hemocultivos fueron positivos en 85 casos (17,9%), 75 de ellos con aislamiento de S. pneumoniae (88,4%) (en 5 junto con otro patogeno). Para la prediccion de bacteriemia (causada o no por S. pneumoniae) la PCT obtiene un area bajo la curva Receiver Operating Characteristic de 0,988 (intervalo de confianza del 95% 0,908-0,995; p 98%, con coeficiente de probabilidad positivo > 10. Los serotipos de S. pneumoniae mas frecuentes fueron 19A, 7F, 1 y 3. Los valores medios mayores de PCT se encontraron en los serotipos 7F, 19A, 3 y 1, con diferencias significativas con el resto (p = 0,008). Los serotipos con mayor porcentaje de sepsis grave-shock septico fueron 3, 1 y 19A, los de mortalidad a los 30 dias fueron 1, 3 y 19A y los de afectacion multilobar o bilateral, los serotipos 3, 19A y 6A. Conclusiones En los pacientes con NAC en el SU la PCT consigue un gran rendimiento diagnostico para descartar o sospechar bacteriemia y para orientar la etiologia de la NAC por S. pneumoniae. Los serotipos 1, 3, 19A y 7F se presentan con mayor frecuencia, respuesta inflamatoria sistemica y gravedad clinica.

One of the main problems facing Atlantic halibut hatcheries is the high mortality in the early stages of larval development. Several factors could be involved, for example: water quality, diseases or abnormalities, such as deformities occurring in the yolk sac larvae prior to exogenous feeding. The aim of this study was to identify differences in bacterial flora associated with yolk sac larvae with oral deformity. We also aimed to establish whether there is any relationship between bacterial strains and the "gaping jaws" syndrome. During our study, 74 bacterial isolates were obtained using three different nutrient media: Marine Agar, R2A and TCBS. Some of these bacteria were characterized using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) and 16S rRNA sequencing. The immune response in larvae exhibiting the "gaping jaws" condition was measured by real time PCR. Our results showed significant differences in bacterial flora between normal and gaping larvae. The gaping yolk sac larvae were predominantly colonized by members of the families Vibrionaceae and Flavobacteriaceae. Bacteria belonging to the Bacillus and Pseudoalteromonas genera were also present but less frequent. It was not possible to associate a type or group of bacteria directly related to "gaping". Strikingly, larvae with gaping jaws had an increase in the expression of two immune related genes, like hepcidin and chemokine (MIP-1s). These results indicate activation of the immune response in larvae with "gaping jaws" syndrome and this response could be related to bacteria isolated from gaping condition.

e16047 Background: Management of stage I SGCC depends on pathological findings after orchiectomy. Four risk-adapted strategies were sequentially applied in a single institution during a 24-year (yr) period according to national guidelines. Here, we compare treatment burden and outcomes of each of them. Methods: From 1/1994 to 1/2018, 208 patients with stage I SGCC were prospectively included in 4 cohorts. Those without risk criteria underwent close surveillance. Patients received active treatment as follow: Group 1: 1994-1999, only patients with T > pT1 received 2 cycles of carboplatin (CBDCA AUC7 x2); Group 2: 1999-2003, patients received CBDCA AUC7 x2 if either tumor size > 4cm or rete testis invasion; Group 3 : 2004-2009, CBDCA AUC7 x2 if both tumor size > 4cm and rete testis invasion were present; Group 4 : ≥2010, CBDCA AUC7 x1 if either tumor size > 4cm or rete testis invasion. Kaplan Meier and log-rank tests were used to evaluate disease-free survival (DFS), Kruskal-Wallis test to compare amount of chemotherapy received per patient. Results: At a median follow-up of 108 months [range 3-423], 19 (9.1%) relapses had occurred. Global 3 and 5-yr DFS were 92.3% and 90%. All relapsing patients were rendered disease-free with 4 cycles of cisplatin (CDDP) - etoposide. Table 1 summarizes results by cohort: Conclusions: A risk-adapted program provided an overall specific survival of 100%. A clinically significant difference in RR was observed when 1 or 2 courses of CBDCA were given. In our series and considering treatment burden, vascular invasion was a better criteria for patient selection to adjuvant chemotherapy, showing a similar DFS but a lower no of total platinum cycles per patient.[Table: see text]

Increased atmospheric CO2 emissions are inducing changes in seawater carbon chemistry, lowering its pH, decreasing carbonate ion availability and reducing calcium carbonate saturation state. This phenomenon, known as ocean acidification, is happening at a faster rate in cold regions, i.e., polar and sub-polar waters. The larval development of Arbacia dufresnei from a sub-Antarctic population was studied at high (8.0), medium (7.7) and low (7.4) pH waters. The results show that the offspring from sub-Antarctic populations of A. dufresnei are susceptible to a development delay at low pH, with no significant increase in abnormal forms. Larvae were isometric between pH treatments. Even at calcium carbonate (CaCO3) saturation states (of both calcite and aragonite, used as proxies of the magnesium calcite) A. dufresnei potential to poleward migrate and further colonize southern regions.

Los metodos de interpolacion en hidrologia son importantes para conocer y transferir informacion de un sitio a otro; por ejemplo caudales medios anuales. Es comun utilizar el Analisis de Componentes Principales (ACP) para estimar y conocer series de tiempo de variables hidrologicas en sitios sin registros. En este articulo se construyen ecuaciones regionales de transferencia de informacion hidrologica, a partir de los datos anuales de caudales medios. Asimismo, se analiza la relacion espacial que existe entre los parametros de un modelo tipo AR(p) con las caracteristicas orograficas de la region. La validacion del modelo se efectua comparando los datos medidos, en una estacion previamente eliminada del analisis; con los estimados a partir de las ecuaciones regionales. Los resultados indican que es un procedimiento sencillo y confiable para transferir informacion hidrologica de un sitio a otro cuando existen escasos registros o nulos.

To analyze the usefulness and ability of procalcitonin (PCT) to predict the presence of bacteremia in patients with community-acquired pneumonia (CAP) caused by Streptococcus pneumoniae (S. pneumoniae) or other bacteria.This is an observational, prospective and descriptive study involving patients who were diagnosed with CAP in our Emergency Department. Data collected included socio-demographic and comorbidity variables, Charlson index, stage in the Pneumonia Severity Index and criteria of severe NAC, microbiologic studies and biomarker determinations (PCT and C reactive protein). The follow-up was carried out during 30 days to calculate the predictive power and the diagnostic performance for bacteremia caused or not by S. pneumoniae.Four hundred and seventy-four patients were finally included in the study. Blood cultures were positive in 85 individuals (17.9%) and S. pneumoniae was identified as the responsible pathogen in 75 of them (88.4%) (in 5 cases together with another agent). The area under the Receiver Operating Characteristic curve for PCT to predict bacteremia (caused by S. pneumoniae or not) was 0.988 (95% confidence interval 0.908-0.995; P.001) and, considering a cut-off value≥0.95ng/mL, the negative predictive value and the positive likelihood ratio were98% and10, respectively. The most frequently isolated serotypes of S. pneumoniae were 19A, 7F, 1 and 3. The highest mean levels of PCT were found in serotypes 7F, 19A, 3 and 1, which showed statistically significant differences with regard to the others serotypes considered (P=.008). Serotypes associated with the highest percentage of severe sepsis-septic shock, 30-days mortality and multi-lobe or bilateral affection were 3, 1 and 19A; 1, 3 and 19A; and 3, 19A and 6A, respectively.PCT had a remarkable diagnostic ability to discard or suspect bacteremia and to guide the etiology of CAP caused by S. pneumoniae. Serotypes 1, 3, 19A and 7F showed greater frequency, systemic inflammatory response and clinical severity.
