1. Novel meriolin derivatives potently inhibit cell cycle progression and transcription in leukemia and lymphoma cells via inhibition of cyclin-dependent kinases (CDKs)

Abstract: Abstract A key feature of cancer is the disruption of cell cycle regulation, which is characterized by the selective and abnormal activation of cyclin-dependent kinases (CDKs). Consequently, targeting CDKs via meriolins represents an attractive therapeutic approach for cancer therapy. Meriolins represent a semisynthetic compound class derived from meridianins and variolins with a known CDK inhibitory potential. Here, we analyzed the two novel derivatives meriolin 16 and meriolin 36 in comparison to other potent CDK inhibitors and could show that they displayed a high cytotoxic potential in different lymphoma and leukemia cell lines as well as in primary patient-derived lymphoma and leukemia cells. In a kinome screen, we showed that meriolin 16 and 36 prevalently inhibited most of the CDKs (such as CDK1, 2, 3, 5, 7, 8, 9, 12, 13, 16, 17, 18, 19, 20). In drug-to-target modeling studies, we predicted a common binding mode of meriolin 16 and 36 to the ATP-pocket of CDK2 and an additional flipped binding for meriolin 36. We could show that cell cycle progression and proliferation were blocked by abolishing phosphorylation of retinoblastoma protein (a major target of CDK2) at Ser612 and Thr82. Moreover, meriolin 16 prevented the CDK9-mediated phosphorylation of RNA polymerase II at Ser2 which is crucial for transcription initiation. This renders both meriolin derivatives as valuable anticancer drugs as they target three different Achilles’ heels of the tumor: (1) inhibition of cell cycle progression and proliferation, (2) prevention of transcription, and (3) induction of cell death.

2. Novel 4-alkoxy Meriolin Congeners Potently Induce Apoptosis in Leukemia and Lymphoma Cells

Abstract: Meriolins (3-(pyrimidin-4-yl)-7-azaindoles) are synthetic hybrids of the naturally occurring alkaloids variolin and meridianin and display a strong cytotoxic potential. We have recently shown that the novel derivative meriolin 16 is highly cytotoxic in several lymphoma and leukemia cell lines as well as in primary patient-derived lymphoma and leukemia cells and predominantly targets cyclin-dependent kinases (CDKs). Here, we efficiently synthesized nine novel 2-aminopyridyl meriolin congeners (3a–3i), i.e., pyrimeriolins, using a one-pot Masuda borylation-Suzuki coupling (MBSC) sequence, with eight of them bearing lipophilic alkoxy substituents of varying length, to systematically determine the influence of the alkoxy sidechain length on the biological activity. All the synthesized derivatives displayed a pronounced cytotoxic potential, with six compounds showing IC50 values in the nanomolar range. Derivatives 3b–3f strongly induced apoptosis and activated caspases with rapid kinetics within 3–4 h in Jurkat leukemia and Ramos lymphoma cells. The induction of apoptosis by the most potent derivative 3e was mediated by the intrinsic mitochondrial death pathway, as it was blocked in caspase-9 deficient and Apaf-1 knockdown Jurkat cells. However, as recently shown for meriolin 16, derivative 3e was able to induce apoptosis in the Jurkat cells overexpressing the antiapoptotic protein Bcl-2. Since tumor cells often inactivate the intrinsic mitochondrial apoptosis pathway (e.g., by overexpression of Bcl-2), these meriolin congeners represent promising therapeutic agents for overcoming therapeutic resistance.

Abstract: Chicken Parvovirus (ChPV) belongs to the genus Aveparvovirus and is implicated in enteric diseases like runting–stunting syndrome (RSS) in poultry. In RSS, chicken health is affected by diarrhea, depression, and increased mortality, causing significant economic losses in the poultry industry. This study aimed to characterize the ChPV genomes detected in chickens with RSS through a metagenomic approach and compare the molecular and evolutionary characteristics within the Aveparvovirus galliform1 species. The intestinal content of broiler flocks affected with RSS was submitted to viral metagenomics. The assembled prevalent genomes were identified as ChPV after sequence and phylogenetic analysis, which consistently clustered separately from Turkey Parvovirus (TuPV). The strain USP-574-A presented signs of genomic recombination. The selective pressure analysis indicated that most of the coding genes in A. galliform1 are evolving under diversifying (negative) selection. Protein modeling of ChPV and TuPV viral capsids identified high conservancy over the VP2 region. The prediction of epitopes identified several co-localized antigenic peptides from ChPV and TuPV, especially for T-cell epitopes, highlighting the immunological significance of these sites. However, most of these peptides presented host-specific variability, obeying an adaptive scenario. The results of this study show the evolutionary path of ChPV and TuPV, which are influenced by diversifying events such as genomic recombination and selective pressure, as well as by adaptation processes, and their subsequent immunological impact.

Abstract: There is growing interest in emerging viruses that can cause serious or lethal disease in humans and animals. The proliferation of cloacal virome studies, mainly focused on poultry and other domestic birds, reveals a wide variety of viruses, although their pathogenic significance is currently uncertain. Analysis of viruses detected in wild birds is complex and often biased towards waterfowl because of the obvious interest in avian influenza or other zoonotic viruses. Less is known about the viruses present in the order Passeriformes, which comprises approximately 60% of extant bird species. This review aims to compile the most significant contributions on the DNA/RNA viruses affecting passerines, from traditional and metagenomic studies. It highlights that most passerine species have never been sampled. Especially the RNA viruses from Flaviviridae, Orthomyxoviridae and Togaviridae are considered emerging because of increased incidence or avian mortality/morbidity, spread to new geographical areas or hosts and their zoonotic risk. Arguably poxvirus, and perhaps other virus groups, could also be considered “emerging viruses”. However, many of these viruses have only recently been described in passerines using metagenomics and their role in the ecosystem is unknown. Finally, it is noteworthy that only one third of the viruses affecting passerines have been officially recognized.

Abstract: Our understanding about viruses carried by wild animals is still scarce. The viral diversity of wildlife may be best described with discovery-driven approaches to the study of viral diversity that broaden research efforts towards non-canonical hosts and remote geographic regions. Birds have been key organisms in the transmission of viruses causing important diseases, and wild birds are threatened by viral spillovers associated with human activities. However, our knowledge of the avian virome may be biased towards poultry and highly pathogenic diseases. We describe and compare the fecal virome of two passerine-dominated bird assemblages sampled in a remote Neotropical rainforest in French Guiana (Nouragues Natural Reserve) and a Mediterranean forest in central Spain (La Herrería). We used metagenomic data to quantify the degree of functional and genetic novelty of viruses recovered by examining if the similarity of the contigs we obtained to reference sequences differed between both locations. In general, contigs from Nouragues were significantly less similar to viruses in databases than contigs from La Herrería using Blastn but not for Blastx, suggesting that pristine regions harbor a yet unknown viral diversity with genetically more singular viruses than more studied areas. Additionally, we describe putative novel viruses of the families Picornaviridae, Reoviridae and Hepeviridae. These results highlight the importance of wild animals and remote regions as sources of novel viruses that substantially broaden the current knowledge of the global diversity of viruses.

7. Chorotypes, zones for the conservation of Scarabaeoidea, and representativity in protected areas of El Salvador.

Abstract: Ecological niche models and species distribution models (SDMs) are tools that can define a species' niche based on presence records and identify those areas where appropriate ecological conditions converge as zones of potential distribution for the species. SDMs help to optimise conservation efforts, particularly important in countries with ecological and economical problems. We constructed SDMs by means of the Maxent algorithm for 160 species of scarab beetles (Scarabaeoidea) to identify chorotypes and important zones for conservation in El Salvador, and to evaluate richness representativity within the System of Protected Natural Areas (SANP). The chorotypes were identified with UPGMA method, with a matrix of presence‐absence in ≈3 km2, and important zones for conservation were identified with the Zonation algorithm. The richness representation in the SANP was done with EstimateS, comparing the estimated by Chao 2 ± confidence intervals (95%) in the same number of cells (n = 811). We identified five chorotypes: (C1) lowland and middleland species, (C2) species widely distributed, (C3) species with an affinity for mid‐elevations, (C4) species limited to lowlands, and (C5) species limited to high montane areas. Most important conservation zones are located in montane regions. As in other tropical areas, the volcanic and montane slopes of El Salvador represent key areas for conserving biodiversity. Species richness in the SANP was not significantly greater than that outside the system; therefore, we recommend continuous prioritisation and amplification of the protected natural heritage of the country, with emphasis on the mountainous area of the territory. Abstract: IntroductionThe standardization of sampling protocols is imperative for robustly studying any taxonomic group. Replicable methods allow the comparison of data between different spatial and temporal studies. In the case of dung beetles, one of the best-studied indicator groups in analyses of environmental disturbance, a wide range of collection methodologies are used, from basic pitfall traps to more complex or complementary methods such as mini-Winkler extractor. Also, different types of attractive baits, sampling effort, durations, and designs are used in dung beetle studies. Variations in methodological approaches are particularly noted in the Neotropics, which may be related to the vast number of biological strategies and behavior of dung beetles that inhabit this region. A lack of methodological unification for the Neotropical region makes a cross-sectional analysis of the information impossible.MethodsWe performed a compilation and analytical review of the existing literature for dung beetle sampling in the Neotropics, discussing the most used methodologies, their advantages and disadvantages, and specific cases in which particular models are more efficient.ResultsPitfall traps baited with human excrement are the most common sampling method, but there is a wide range of models and variations in the structure of this trap. The complementary effect generated by flight interception traps, light traps, and direct collections, particularly within microhabitats, is exciting for the potential of finding new species. Some methodologies, such as mini-Winkler extractor, fogging, or very specific baits, are infrequently used.DiscussionThere was a lack of inclusion of spatial and temporal variation among studies. Therefore, it is necessary to consider broader sampling windows, which include different spatial scales, seasons, and years. Finally, we propose a standard protocol for sampling dung beetles in the Neotropics, depending on each objective, and including a basic methodology for obtaining complete local inventories.

Abstract: The checklist of the species of fungus beetle families Endomychidae and Anamorphidae from El Salvador is updated. Only six species of Endomychidae and one Anamorphidae have been previously recorded from the country, with specific localities reported only for two of them. The study of specimens collected in El Salvador over 60 years ago by Jan and Bohumila Bechyné and held in the Royal Belgian Institute of Natural Sciences, Brussels, allowed us to have a better idea on the diversity and distribution of these beetles in the country. Eight species of Endomychidae and one of Anamorphidae are reported here, including three first records for El Salvador: Anidrytus guatemalae Arrow, Epipocus bifidus Gerstaecker and Stenotarsus ovalis Arrow. Additionally, first records for species of Endomychidae from countries of the Neotropical region are provided: Anidrytus batesi Strohecker from Peru, A. ovatulus Gerstaecker from Guyana, Ephebus hirtulus Strohecker from Paraguay, Epopterus vacuus Gerstaecker from Costa Rica and Acinaces unicolor Tomaszewska from Peru. Abstract: The urbanization process has accelerated in the last century and is estimated to grow by 1.1% each year, causing urban centers to become increasingly more common. Urban green areas are important, because they provide needed shelter and food for birds allowing them to survive and adapt to urbanized landscapes. In the city of San Salvador, the campus of the University of El Salvador includes an area with numerous trees and shrubs. The objective of this research was to update the knowledge of the avifauna that feeds, rests, and nests in this area. From February 2017 to January 2018 we conducted a monthly survey of the birds on the campus. Additionally, we reviewed the literature and inspected available collections to create a historical and updated the inventory of the birds on the campus. We registered 64 species, of which 14 constitute new records for the site. The greatest number of observations was of Brotogeris jugularis, Zenaida asiatica and Quiscalus mexicanus. The total richness of the University of El Salvador increased to 100 species. We also highlight the presence of three locally threatened species; Eupsittula canicularis, Psittacara strenuus and B. jugularis and Amazona auropalliata a globally endangered species.

Abstract: Dung removal by macrofauna such as dung beetles is an important process for nutrient cycling in pasturelands. Intensification of farming practices generally reduces species and functional diversity of terrestrial invertebrates, which may negatively affect ecosystem services. Here, we investigate the effects of cattle-grazing intensification on dung removal by dung beetles in field experiments replicated in 38 pastures around the world. Within each study site, we measured dung removal in pastures managed with low- and high-intensity regimes to assess between-regime differences in dung beetle diversity and dung removal, whilst also considering climate and regional variations. The impacts of intensification were heterogeneous, either diminishing or increasing dung beetle species richness, functional diversity, and dung removal rates. The effects of beetle diversity on dung removal were more variable across sites than within sites. Dung removal increased with species richness across sites, while functional diversity consistently enhanced dung removal within sites, independently of cattle grazing intensity or climate. Our findings indicate that, despite intensified cattle stocking rates, ecosystem services related to decomposition and nutrient cycling can be maintained when a functionally diverse dung beetle community inhabits the human-modified landscape.

Abstract: La creciente urbanización ha propiciado que los ecosistemas naturales sean constantemente sustituidos por paisajes urbanos, proceso muy notorio en El Salvador, debido a su pequeña extensión territorial (21.041 km²) y su alta densidad poblacional (291 hab/km²). Realizamos un inventario en 12 áreas verdes urbanas, con diferentes tamaños, formas y distancia con respecto al área boscosa más grande de la zona metropolitana, basados en la teoría de biogeografía de islas de McArthur y Wilson (1967). Evaluamos si la riqueza, diversidad y equititavidad de aves se relacionaba con el tamaño y la distancia de las áreas verdes y si la forma de las mismas tenía algún efecto sobre la riqueza de aves. Registramos un total de 20 especies de aves y las clasificamos según su dieta (generalista y especialista). Observamos que la distancia no influyó en la riqueza de aves y que no hubo interacción entre las variables tamaño y distancia, pero que sí influyó el tamaño del área verde. La riqueza de aves con dieta restringida incremento en las áreas verdes más circulares que en las de forma más irregular. Concluimos que en el centro urbano de San Salvador, la presencia de áreas verdes grandes y circulares aporta más a la riqueza de aves de dieta especialista, que áreas de similar tamaño pero de forma irregular. Sin embargo, áreas pequeñas aportan más a la riqueza de aves que poseen esa dieta, si su forma es más circular

Abstract: The purpose of this article is to explore employee age as a moderating factor in the relationship between leader contingent reward behavior (CRB) and work engagement. In doing so, the authors seek to provide a more nuanced understanding of the mediating role of work engagement in the negative effect of leader CRB on turnover intention.,The authors used online surveys among a sample of employees of a retail company in Chile to capture individual perceptions about supervisor CRB, work engagement and turnover intention. To test the authors’ hypotheses, they modeled a first-stage moderated mediation effect using Hayes’ Process macro.,The authors’ results confirm the hypothesis that the negative effect of leader CRB on employee turnover intention is partially mediated by employee work engagement. Interestingly, age was a significant moderator of the mediation effect only for individuals working at headquarters, but not for employees working in stores.,This study expands current knowledge about how the leadership–engagement relationship can predict organizational outcomes, including age as a boundary condition. Following the job demands-resources theory, the authors also prove that conceptualizing leader CRB as a job resource can benefit the integration of leadership and work engagement research. The authors’ findings may help organizational researchers and practitioners acknowledge contextual differences in understanding the combined effects of leadership styles and work engagement.,El proposito de este articulo es explorar la edad del empleado como un factor moderador en la relacion entre el comportamiento de recompensa contingente del lider, engagement laboral, e intencion de renuncia.,Usamos encuestas en linea, en una muestra de empleados de una empresa de retail en Chile, para capturar las percepciones individuales sobre los comportamientos de recompensa contingente de los supervisores, el engagement laboral, y la intencion de renuncia. Para probar nuestras hipotesis modelamos un efecto de mediacion moderada de primera-etapa utilizando el macro Process de Hayes.,Los resultados confirman la hipotesis de que las recompensas contingentes del lider estan negativamente relacionadas con la intencion de renuncia. Este efecto esta parcialmente mediado por el nivel de engagement laboral del empleado. Curiosamente, la edad fue un moderador significativo del efecto de mediacion solo para quienes trabajaban en la oficina central, pero no para quienes trabajan en las tiendas.,Nuestro estudio amplia el conocimiento actual sobre como la relacion liderazgo-compromiso puede predecir resultados organizacionales, incluyendo la edad como condicion de borde. Basados en la teoria de las demandas y recursos del trabajo (job demands-resources theory), mostramos que, al conceptualizar el comportamiento de recompensa contingente del lider como un recurso laboral, se puede beneficiar la investigacion sobre la integracion del liderazgo y el engagement laboral. Abstract: Parque Bicentenario in the Area Natural Protegida El Espino-Bosque Los Pericos, considered the "last green lung of San Salvador", is the most important forested area in central El Salvador. We performed a survey to provide the first inventory of scarab beetles (Scarabaeidae) of the park. Collections were conducted monthly for one year (August 2018-July 2019) by using baited pitfall, aerial, and ultraviolet light traps. Four subfamilies, nine tribes, 14 genera, 20 species (8% of the total richness of the family in El Salvador), and 1,584 individuals were captured. The most abundantly collected species in the park were the scarabaeines Onthophagus batesi Howden and Cartwright, Onthophagus belorhinus (Bates), Dichotomius centralis (Harold), Coprophanaeus corythus (Harold), Dichotomius yucatanus (Bates), and Coprophanaeus boucardi (Nevinson).

Abstract: Introduction: Oral status has a clear impact on systemic health, a key component of successful aging. Awareness of oral disorders permits early diagnoses and treatments. This study investigates oral disorders awareness among community-dwelling elders. Methods: Community-based survey through face-to-face interviews undertaken in four cities at busy commercial and administrative areas. Results: A total of 789 people were recruited, mostly nonsmoker (60.3%) males (56.9%) with basic education (65.6%). The most frequently recalled disorder was bruxism, followed by periodontal diseases and ulcerations/aphthae. Caries was mentioned by 3.91%. Dentate participants were more aware of dental-related disorders, such as bruxism ( p = .001) and periodontitis ( p = .010), except for caries ( p = .100). Caries awareness was related to educational level ( p = .018) and smoking ( p = .020), as was oral cancer ( p = .019) with former smokers more aware of this neoplasm. Education was strongly related with periodontal awareness ( p = .001). Conclusion: Awareness of the main oral disorders is worryingly poor among the elderly population and immediate action is required by taking advantage of the position of family nurses in the treatment of systemic chronic disorders and through educational interventions on this population subgroup to ease preventive behaviors, favor early diagnoses, and promote simpler, more effective treatments with a lower financial burden.
