Light-sheet microscopy is a powerful method for imaging small translucent samples in vivo, owing to its unique combination of fast imaging speeds, large field of view, and low phototoxicity. This chapter briefly reviews state-of-the-art technology for variations of light-sheet microscopy. We review recent examples of optogenetics in combination with light-sheet microscopy and discuss some current bottlenecks and horizons of light sheet in all-optical physiology. We describe how 3-dimensional optogenetics can be added to an home-built light-sheet microscope, including technical notes about choices in microscope configuration to consider depending on the time and length scales of interest.

Fast fashion continuously promotes the mass consumption of clothes and, because of a non-linear economy, the disposal of textile waste. The generation of textile waste is expected to increase further and has already been shown to significantly contribute to the release of fibrous microplastics (FMPs) into the environment. FMPs are the primary microplastic (MP) in the air, and FMP (and MP) contamination is a global concern to address to prevent negative impacts on human health and the environment. The collection of textile waste microfibers during the textile manufacturing processes, i.e., finishing of fabrics, and valorization in construction materials such as fiber-reinforced cementitious composites (FRCs), is here proposed to limit FMPs spread. A thorough characterization of the textile waste microfibers allowed the description of their size, composition, and the main features required for their use in FRCs (density, water absorption, water release in time). Microfiber contents 0-4 wt% and three different microfiber conditions were considered in Portland cement: water-saturated, NaOH-treated, and untreated. The increase in microfiber content significantly promoted the physical-mechanical properties and insulating power of unreinforced Portland cement. And water saturation and NaOH treatment were more successful in reducing linear shrinkage and increasing toughness. For the first time, this work showed a practical mitigation approach of FMPs through the valorization of textile waste to manufacture sustainable and efficient FRCs. Notably, the results displayed that the same amount of FMP fallout in Paris daily can be mitigated per ton of cement paste to create valuable and promising construction materials. Adaptive optics can improve the performance of optical systems and devices by correcting phase aberrations. While in most applications wavefront sensing is employed to drive the adaptive optics correction, some microscopy methods may require sensorless optimization of the wavefront. In these cases, the correction is performed by describing the aberration as a linear combination of a base of influence functions, optimizing an image quality metric as a function of the coefficients. The influence functions base is generally chosen to either efficiently represent the adaptive device used or to describe generic wavefronts in an orthogonal fashion. A rarely discussed problem is that most correction bases have elements which introduce, together with a correction of the aberration, a shift of the imaging field of view in three dimensions. While simple methods to solve the problem are available for linear microscopy methods, nonlinear microscopy techniques such as multiphoton or second harmonic generation microscopy require non-trivial base determination. In this paper, we discuss the problem, and we present a method for calibrating a shift-less base on a spatial light modulator for two-photon microscopy.

Quantunque la riflessione sull’importanza di perseguire gli obiettivi di sviluppo sostenibile si sia ampliata e potenziata in epoca relativamente recente, da ben più tempo le tradizioni religiose nello sviluppo plurisecolare delle loro dottrine e prassi, ciascuna secondo il proprio peculiare ordine assiologico, promuovono i valori che presiedono alla cura del creato, alla dignità della persona, al bene comune. Il contributo che le religioni possono offrire agli obiettivi dell’Agenda 2030 delle Nazioni Unite coinvolge non solo il piano pratico, ma soprattutto quello di comprensione critica, nel senso di offrire motivazioni forti per una presa di coscienza seria e un coinvolgimento autentico, per dare sostanza e sostegno etico e spirituale a un nuovo modello culturale di sviluppo integrale che promuova nella complessità degli aspetti ecologici e antropologici, e quindi anche religiosi, il rispetto di tutti gli esseri viventi; lo sfruttamento responsabile delle risorse naturali; il consolidamento di relazioni sociali improntate alla solidarietà, all’inclusione e alla promozione delle persone; l’impostazione dei rapporti produttivi e commerciali secondo modalità autenticamente umane. Il rapporto tra religioni e sviluppo sostenibile viene analizzato in merito a tre ambiti del programma d’azione: “Ambiente e alimentazione (obiettivi 2, 12 e 15 Agenda ONU 2030)”; “Comunità umane solidali e inclusive (obiettivi 4, 10, 11 e 16 Agenda ONU 2030)”; “Vita umana, lavoro e sviluppo tecnologico (obiettivi 3, 8 e 9 Agenda ONU 2030)”. Per ogni settore sono individuati alcuni obiettivi strategici e su questi traguardi riflettono, in primo luogo gli esperti sotto il profilo scientifico, per illustrare il significato e i contenuti dei singoli obiettivi, e in secondo luogo gli studiosi o i rappresentanti delle diverse tradizioni religiose, per precisare il valore attribuito a questi obiettivi dalla dottrina confessionale e sottolineare il contributo che intende dare per la loro promozione.
