Abstract To gain a deeper understanding of the flowering pattern and reproductive characteristics of Epimedium sagittatum, to enrich the research on the flower development of E. sagittatum and its reproductive regulation, and to screen the methods suitable for the rapid detection of pollen viability of E. sagittatum and to promote its cross-breeding. The characteristics of its flower parts were observed, recorded and measured, and the pollen viability of E. sagittatumwas determined by five methods, including TTC staining, I2-KI staining, red ink staining, peroxidase method and in vitro germination method. The flowering process of E. sagittatum can be divided into five stages: calyx dehiscence, bract spathe, petal outgrowth, pollen dispersal, and pollination and withering. The results of I2-KI staining and peroxidase method were significantly higher than those of other methods; the in vitro germination method was intuitive and accurate, but the operation was complicated and time-consuming; the red ink staining method was easy to operate and had obvious staining effect, and the results were the closest to those of the in vitro germination method; and it was found that the pollen of E. sagittatum was not as effective as the in vitro germination method at the bud stamen stage, the flower stigma and the flower bud. It was also found that the pollen viability and germination rate of E. sagittatum pollen were higher in the three periods of bud spitting, petal adductor and pollen dispersal. Comparing the five methods, the red ink staining method was found to be a better method for the rapid detection of pollen viability; the best pollination periods of E. sagittatum were the bud stamen stage, petal adductor stage, and pollen dispersal stage of flowers at the peak of bloom. This study on the flowering and fruiting pattern of E. sagittatum, and the related mechanism of sexual reproduction, can be used as a reference for the next step of research on the breeding of E. sagittatum.

SUMMARY: The pollen viability directly affects the pollination process and the ultimate grain yield of rice. Here, we identified that the MORN motif‐containing proteins, OsMORN1 and OsMORN2, had a crucial role in maintaining pollen fertility. Compared with the wild type (WT), the pollen viability of the osmorn1 and osmorn2 mutants was reduced, and pollen germination was abnormal, resulting in significantly lower spikelet fertility, seed‐setting rate, and grain yield per plant. Further investigation revealed that OsMORN1 was localized to the Golgi apparatus and lipid droplets. Lipids associated with pollen viability underwent alterations in osmorn mutants, such as the diacylglyceride (18:3_18:3) was 5.1‐fold higher and digalactosyldiacylglycerol (18:2_18:2) was 5.2‐fold lower in osmorn1, while the triacylglycerol (TG) (16:0_18:2_18:3) was 8.3‐fold higher and TG (16:0_18:1_18:3) was 8.5‐fold lower in osmorn2 than those in WT. Furthermore, the OsMORN1/2 was found to be associated with rice cold tolerance, as osmorn1 and osmorn2 mutants were more sensitive to chilling stress than WT. The mutants displayed increased hydrogen peroxide accumulation, reduced antioxidant enzyme activities, elevated malondialdehyde content, and a significantly decreased seedling survival rate. Lipidomics analysis revealed distinct alterations in lipids under low temperature, highlighting significant changes in TG (18:2_18:3_18:3) and TG (18:4_18:2_18:2) in osmorn1, TG (16:0_18:2_18:2) and PI (17:2_18:3) in osmorn2 compared to the WT. Therefore, it suggested that OsMORN1 and OsMORN2 regulate both pollen viability and cold tolerance through maintaining lipid homeostasis. Sporopollenin, as the main component of the pollen exine, is a highly resistant polymer that provides structural integrity under unfavourable environmental conditions. Tetraketone α‐pyrone reductase 1 (TKPR1) is essential for sporopollenin formation, catalyzing the reduction of tetraketone carbonyl to hydroxylated α‐pyrone. The functional role of TKPR1 in male sterility has been reported in flowering plants such as maize, rice, and Arabidopsis. However, the molecular cloning and functional characterization of TKPR1 in cotton remain unaddressed. In this study, we identified 68 TKPR1s from four cotton species, categorized into three clades. Transcriptomics and RT‐qPCR demonstrated that GhTKPR1_8 exhibited typical expression patterns in the tetrad stage of the anther. GhTKPR1_8 was localized to the endoplasmic reticulum. Moreover, ABORTED MICROSPORES (GhAMS) transcriptionally activated GhTKPR1_8 as indicated by luciferase complementation tests. GhTKPR1_8‐knockdown inhibited anther dehiscence and reduced pollen viability in cotton. Additionally, overexpression of GhTKPR1_8 in the attkpr1 mutant restored its male sterile phenotype. Rising temperature affects agricultural production, causing food insecurity. Thus, the objective of this study was to evaluate how increased temperature influences pollen viability, photosynthetic and enzymatic responses, and their consequences on the final yield of cowpea cultivars. The cultivars BRS Acauã, BRS Guariba, BRS Gurguéia, and BRS Pajeú were used, kept in growth chambers under two temperature regimes: 24.8–30.8–37.8 °C and 20–26–33 °C. The cultivars BRS Acauã, BRS Guariba, and BRS Pajeú showed prolonged flowering and greater flower abortion, at 23.58%, 34.71%, and 25.55%, respectively, under an increase of 4.8 °C in temperature. This increase also reduced the viability of BRS Acauã and BRS Pajeú pollen by 34 and 7%, respectively. Heating increased stomatal opening and transpiration but reduced chlorophyll content. The enzymatic response varied according to cultivars and temperature. Changes in photosynthetic and enzymatic activities contribute to reducing pollen viability and productivity. BRS Acauã was the most affected, with an 82% reduction in the number of seeds and a 70% reduction in production. BRS Gurguéia maintained its production, even with an increase of 4.8 °C, and can be selected as a cultivar with the potential to tolerate high temperatures as it maintained pollen viability, with less flower abortion, with the synchrony of physiological and biochemical responses and, consequently, greater production. Understanding pollen and ovule fertility as factors influencing fruit and seed set is important in cassava breeding. Extended daylength with red light (RL) and plant growth regulators (PGRs) have been used to induce flowering and fruit set in cassava without any reference to effects on pollen viability or ovule fertilizability. This study investigated the effects of field-applied RL and PGR on pollen viability and ovule fertilizability. Panels of cassava genotypes with early or moderate flowering responses were used. RL was administered from dusk to dawn. Two PGRs, 6-benzyl adenine (BA), a cytokinin and silver thiosulphate (STS), an anti-ethylene, were applied. Pollen viability was assessed based on pollen grain diameter, in vitro stainability, in vivo germinability, ovule fertilizability, and ploidy level. Treating flowers with RL increased the pollen diameter from 145.6 in control to 148.5 µm in RL, 78.5 to 93.0% in stainability, and 52.0 to 56.9% in ovule fertilizability in treated female flowers. The fruit set also increased from 51.5 in control to 71.8% in RL-treated female flowers. The seed set followed a similar trend. The ploidy level of pollen from RL-treated flowers increased slightly and was positively correlated with pollen diameter (R2 = 0.09 *), ovule fertilization (R2 = 0.20 *), fruit set (R2 = 0.59 *), and seed set (R2 = 0.60 *). Treating flowers with PGR did not affect pollen diameter but increased stainability from 78.5% in control to 82.1%, ovule fertilizability from 42.9 to 64.9%, and fruit set from 23.2 to 51.9% in PGR-treated female flowers. Combined BA + STS application caused the highest ovule fertilizability, fruit, and seed set efficiency. These results show that RL and PGR treatments increase pollen viability and ovule fertilizability. This is important for planning pollination strategies in cassava breeding programmes.

In vitro pollen germination is considered the most efficient method to assess pollen viability. The pollen germination frequency and pollen tube length, which are key indicators of pollen viability, should be accurately measured during in vitro culture. In this study, a Mask R-CNN model trained using microscopic images of tree peony (Paeonia suffruticosa) pollen has been proposed to rapidly detect the pollen germination rate and pollen tube length. To reduce the workload during image acquisition, images of synthesized crossed pollen tubes were added to the training dataset, significantly improving the model accuracy in recognizing crossed pollen tubes. At an Intersection over Union threshold of 50%, a mean average precision of 0.949 was achieved. The performance of the model was verified using 120 testing images. The R 2 value of the linear regression model using detected pollen germination frequency against the ground truth was 0.909 and that using average pollen tube length was 0.958. Abstract Pollen viability, dispersion ability and longevity during deep-freezer storage were studied in three Juniperus taxa distributed in Slovakia. All these characteristics of pollen are closely related to the pollination and/or fertilization success of the junipers in nature. Pollen viability varied considerably between the three populations of J. communis and one population of each, J. sibirica and J. communis var. intermedia. Pollen germination rate ranged between 40.75% and 75.06%, and pollen tube length between 30.32 and 40.41 µm. A clear tendency indicates a higher germination rate of J. communis pollen from lower altitudes and reduced germination of J. sibirica and J. communis var. intermedia pollen from higher altitudes. The dispersion potential of the J. communis pollen during its shedding seems relatively low. In 2021, pollen cloud density was diluted at 68.1% at the 4 m distance from the test shrub, in 2022 of 52.1% at the 17 m distance from the pollen source. A deep-freezer storage of juniper pollen in a double-walled polyethylene bag with silica gel was not efficient enough, as indicated by the drop of pollen germination rate of 31.2% in J. communis and of 79.4% in J. sibirica during a 1-year storage period at − 81 °C.

Lavender (Lavandula spp.) is one of the most widely grown essential oil crops in the world. This study aimed to determine the nuclear DNA contents using flow cytometry, pollen viability using TTC (2,3,5-Triphenyl Tetrazolium Chloride) and IKI (Iodine Potassium Iodide) tests, pollen production quantities using hemacytometric method, and seed retention rates per spike and flower of lavender (L. angustifolia var. Raya) and lavandin (L. intermedia var. Super) grown under ecological conditions in Isparta province of Turkey. The nuclear DNA contents were 2.11 and 2.54 pg 2C-1, respectively in the lavender and lavandin cultivar. The flowers of the lavender cultivar produced abundant pollen grains (average 5800 pollen per flower and 1450 pollen per anther) with high viability (60.65-65.05%) and seed retention rate per spike (91.57% on average). The lavandin cultivar, which had very low pollen viability (1.08-3.32%) and pollen grains (average of 2350 pollen per flower and 587.5 pollen per anther) gave very low seed retention rates per spike (0.60% on average). While each flower had four ovaries with the potential to produce four nutlets, lavandin flowers produced only trace numbers (0.15% on average) of seeds. As a result, the lavandin cultivar had more nuclear DNA content, longer stem and spike, smaller size but more numerous flowers, less and lighter anthers, lower pollen grains and viability, and vey few inviabile seeds compared to the lavender cultivar. In species like lisianthus, uncertainty persists regarding the optimal methods for assessing pollen quality, which is crucial for evaluating pollen parent fertility. This study seeks to identify the most reliable techniques for this purpose. Fresh and dead pollen from four lisianthus (Eustoma grandiflorum) varieties was used. The dead pollen was obtained by thermal inactivation. Five chemical staining methods (iodine-potassium iodide, 2,3,5-triphenyltetrazolium chloride - TTC, lactophenol cotton blue, safranin, acetocarmine) were employed to assess pollen viability, and two biological methods (Petri dishes, hanging drops) were used to determine the germination rate. Four solid medium cultures were employed in Petri dishes, while the hanging drop utilised four liquid medium cultures. Thirteen tests were conducted for each variety, evaluating fresh and dead pollen. The study found significant variations in pollen quality among lisianthus varieties and methods. Fresh pollen showed viability rates ranging from 56.87% to 99.41% and germination rates from 0.20% to 45.11%. TTC exhibited the lowest viability rate across all varieties, while the highest germination rate was observed in the liquid culture medium with only boric acid and PEG1500. Notably, TTC was the sole viability method that did not stain dead pollen, and no germination occurred in any method for dead pollen. TTC is the most reliable staining method, and a liquid culture medium with boric acid and PEG1500 effectively determines lisianthus pollen quality. • Comprehensive phenology from the onset of the primordial bud to the senescence phase, is documented for Macrotyloma uniflorum. • Floral organs exhibits ultrastructural variation in Macrotyloma uniflorum. • Macrotyloma uniflorum accessions from Republic of Benin are non-echinate; other accessions exhibit spinuliferous pollen. • There is a positive correlation between days to flowering and most floral traits. • Pollens of Macrotyloma uniflorum are viable during the day. The absence of information on the reproductive biology of some nutritive legumes has been an obstacle to their genetic improvement. Gathering information on reproductive biology contributes significantly to the interest of breeders to have adequate tools for initiating the improvement program through crosses among high performing genotypes. To contribute to this, we examined the flowering phenology, pollen biology, stigma receptiveness at bud dehiscence and anthesis phases, and the in-vitro germination of the pollens in horsegram (Macrotyloma uniflorum). Pollens of horsegram were found to be either tricolporate or triporate, echinates in all accessions except in the collections from Benin. Ovaries in horsegram showed great variation in the number of ovules and activities of peroxidase indicated the receptiveness of stigma at bud dehiscence phase and during anthesis. A high number of ovules per ovary was found in accessions from Australia (Leichardt 1 and 2), Mali (LeichardtMali) and India (CPI22679). Pollen viability analysis implies the possibility of better results when pollinating during the day and immediately after the flower opens. The study provides information on the reproductive biology of horsegram for future hybridization program in order to enhance the performance of the crop and increase production. Plants in the Gesneriaceae family are appreciated for their decorative leaves and flowers, ease of cultivation, and shade tolerance. Sexual hybridisation has long been carried out for producing novel hybrids. However, scientific knowledge is lacking on the correct timing of pollination in this family. This study was conducted to elucidate the optimal timing for pollination by screening pollen viability and stigma receptivity throughout the flowering period in eight gesneriad taxa. Pollen viability was evaluated by the in vitro germination test and stigma receptivity was based on stigma morphology and pistil length. The flowering duration varied from 10 days in Alsobia to 18 days in Streptocarpus. While the pollens of Episcia, Kohleria, Saintpaulia, Sinningia, and Smithiantha had totally lost viability 2–5 days before the flowers withered, a slightly contrasting situation was observed in the cases of Alsobia, Deinostigma, and Streptocarpus, where a small portion of pollen grains remained viable towards the end of the flowering period. The highest pollen germination rate was recorded from 1 day in Alsobia, Episcia, and Kohleria, to 9 days in Deinostigma. The reduction in pollen germination was rapid in Alsobia, Saintpaulia, and Smithiantha, and moderate in the remaining taxa. The greatest pollen tube growth occurred at 1–3 days after flower opening and decreased rapidly in all the taxa tested. The longest pistil of 2.03–3.50 cm was observed at 3–8 days of anthesis depending on the plant tested. The findings in this study suggest that higher pollination success may be achieved using pollen grains of newly opened flowers and stigmas of mature flowers in this family. [ABSTRACT FROM AUTHOR]

Modern agricultural biotechnologies, such as those derived from genetic modification, are solutions that can enable an increase in food production, lead to more efficient use of natural resources, and promote environmental impact reduction. Crops with altered genetic materials have been extensively subjected to safety assessments to fulfill regulatory requirements prior to commercialization. The Brazilian National Technical Biosafety Commission (CTNBio) provides provisions for commercial release of transgenic crops in Brazil, including requiring information on pollen dispersion ability as part of environmental risk assessment, which includes pollen viability and morphology studies. Here we present the pollen viability and morphology of non-transgenic conventional materials, single-event genetically modified (GM) products, and stacked GM products from soybean, maize and cotton cultivated in Brazil. Microscopical observation of stained pollen grain was conducted to determine the percentage of pollen viability as well as pollen morphology, which is assessed by measuring pollen grain diameter. The pollen viability and diameter of GM soybean, maize and cotton, evaluated across a number of GM events in each crop, were similar to the conventional non-GM counterparts. Pollen characterization data contributed to the detailed phenotypic description of GM crops, supporting the conclusion that the studied events were not fundamentally different from the conventional control. Pollen grains represent the male gametes of seed plants and their viability is critical for sexual reproduction in the plant life cycle. Palynology and viability studies have traditionally been used to address a range of botanical, ecological and geological questions, but recent work has revealed the importance of pollen viability in invasion biology as well. Here, we report an efficient visual method for assessing the viability of pollen using digital holographic microscopy (DHM). Imaging data reveal that quantitative phase information provided by the technique can be correlated with viability as indicated by the outcome of the colorimetric test. We successfully test this method on pollen grains of Lantana camara, a well-known alien invasive plant in the tropical world. Our results show that pollen viability may be assessed accurately without the usual staining procedure and suggest potential applications of the DHM methodology to a number of emerging areas in plant science.

Terminal heat stress during reproductive stage in wheat (Triticum aestivum L.) causes pollen grain sterility and has a drastic impact on wheat crop production. Finding genotypes with high pollen viability under heat stress is crucial to cope with the impact of climate change through developing heat-tolerant cultivars. To assess the effect of terminal heat stress on pollen viability in a panel of spring wheat genotypes (N = 200), RCBD (randomized complete block design) field trials were conducted under normal and heat stress conditions for two consecutive years (2020–2021 and 2021–2022). Analysis of variance showed significant variation in genotypes, treatments, and genotype × treatment interaction. Fifty and 46 genotypes were categorized as heat tolerant (HSIpv < 0.5) in the first and second year, respectively. Twelve genotypes, namely, Chenab-70, Pari-73, Pak-81, MH-21, Punjab-76, NIFA-Aman, NUWYT-63, Swabi-1, Nisnan-21, Frontana, Amin-2000, and Pirsabak-2004, were found to be heat tolerant across the years. The violin plot displayed a trend of improvement in heat tolerance (HSIpv < 0.5) over the period of time in many modern wheat varieties. However, some modern wheat varieties released after 2001 such as Janbaz-09 (57%), Ghazi-2019 (57%), and Sindhu-16 (43%) had very low pollen viability under heat stress conditions. The results of phenotypic coefficient of variance (PCV%), genotypic coefficient of variance (GCV%), broad sense heritability (h2bs), and genetic advance (GA) suggested the major contribution of genetic factors in controlling pollen viability trait. Higher values of h2bs and GA under heat stress conditions suggested pollen viability as a heat tolerance trait controlled by additive genetic effects. Taken together, these results suggested pollen viability as a useful trait for selection in early generations under elevated temperatures. The genotypes identified as heat tolerant in both years can be used as genetic resources for breeding cultivars with higher pollen viability under elevated temperature conditions.

Maize pollen is highly sensitive to heat and drought, but few studies have investigated the combined effects of heat and drought on pollen viability. In this study, pollen's structural and physiological characteristics were determined after heat, drought, and combined stressors. Furthermore, integrated metabolomic and transcriptomic analyses of maize pollen were conducted to identify potential mechanisms of stress responses. Tassel growth and spikelet development were considerably suppressed, pollen viability was negatively impacted, and pollen starch granules were depleted during anthesis under stress. The inhibitory effects were more significant due to combined stresses than to heat or drought individually. The metabolic analysis identified 71 important metabolites in the combined stress compared to the other treatments, including sugars and their derivatives related to pollen viability. Transcriptomics also revealed that carbohydrate metabolism was significantly altered under stress. Moreover, a comprehensive metabolome-transcriptome analysis identified a central mechanism in the biosynthesis of UDP-glucose involved in reducing the activity of sucrose synthase SH-1 (shrunken 1) and sus1 (sucrose synthase 1) that suppressed sucrose transfer to UDP-glucose, leading to pollen viability exhaustion under stress. In conclusion, the lower pollen viability after heat and drought stress was associated with poor sucrose synthase activity due to the stress treatments.

Background: The research was conducted to screen rice cultivars with resistance ability to high-temperature conditions and analyze the relationship between pollen viability and filled grain of native Indonesia rice cultivars. Methods: The rice seeds were germinated, sown into a tray and then transplanted into pots filled with sandy loam and compost 3:1. After 20% of flowering, four pots of each cultivar were kept under ambient temperature (28-32°C) and the other four pots were transferred to high-temperature (33-38°C) conditions. Pollen stainability and pollen germination were conducted using IKI solution and by germinating them under in vitro conditions respectively before being viewed under the light microscope. Yield characters were analyzed to obtain a relationship with pollen viability. Result: Percentage of pollen germination, filled and empty grains of rice cultivars significantly differed between the two conditions by t-test. Ciherang and Sigeudop were two resistant cultivars for high-temperature treatment, with filled grain percentage more than 85%. There was a strong relationship between pollen germination and filled grains at ambient temperature (r=0.95; p<0.01) and high temperature (r=0.94; p<0.01). Oil tea (Camellia oleifera Abel.) is one of the most important woody edible oil tree species in China, with intraspecific polyploid. In order to study the variation in pollen size and vigor of C. oleifera at ploidy level, four ploidy covers a total of 32 types of Camellia pollens as the material for the experiment. The results showed that the pollen sizes of diploid, tetraploid, hexaploidy, and octaploid were positively correlated with the ploidy level. Pollen viability of C. oleifera was determined by fluorescein diacetate (FDA) dye solution staining and medium containing 10% sucrose, 0.01% boric acid, and 1% agar germination in vitro, which indicated that the pollen viability and germination rate of the hexaploid were relatively high among the four ploidy levels, at 79.69% and 71.78% respectively. The pollen vigor of diploid NR-3, tetraploid DP43, hexaploid CJ-12, and octoploid YNYC-1 was higher than that of other materials with the same ploidy level. Pollen grains, the male gametophytes for reproduction in higher plants, are vulnerable to various stresses that lead to loss of viability and eventually crop yield. A conventional method for assessing pollen viability is manual counting after staining, which is laborious and hinders high-throughput screening. We developed an automatic detection tool (PollenDetect) to distinguish viable and nonviable pollen based on the YOLOv5 neural network, which is adjusted to adapt to the small target detection task. Compared with manual work, PollenDetect significantly reduced detection time (from approximately 3 min to 1 s for each image). Meanwhile, PollenDetect can maintain high detection accuracy. When PollenDetect was tested on cotton pollen viability, 99% accuracy was achieved. Furthermore, the results obtained using PollenDetect show that high temperature weakened cotton pollen viability, which is highly similar to the pollen viability results obtained using 2,3,5-triphenyltetrazolium formazan quantification. PollenDetect is an open-source software that can be further trained to count different types of pollen for research purposes. Thus, PollenDetect is a rapid and accurate system for recognizing pollen viability status, and is important for screening stress-resistant crop varieties for the identification of pollen viability and stress resistance genes during genetic breeding research. Rising temperature affects agricultural production, causing food insecurity. Thus, the objective of this study was to evaluate how increased temperature influences pollen viability, photosynthetic and enzymatic responses, and their consequences on the final yield of cowpea cultivars. The cultivars BRS Acauã, BRS Guariba, BRS Gurguéia, and BRS Pajeú were used, kept in growth chambers under two temperature regimes: 24.8–30.8–37.8 °C and 20–26–33 °C. The cultivars BRS Acauã, BRS Guariba, and BRS Pajeú showed prolonged flowering and greater flower abortion, at 23.58%, 34.71%, and 25.55%, respectively, under an increase of 4.8 °C in temperature. This increase also reduced the viability of BRS Acauã and BRS Pajeú pollen by 34 and 7%, respectively. Heating increased stomatal opening and transpiration but reduced chlorophyll content. The enzymatic response varied according to cultivars and temperature. Changes in photosynthetic and enzymatic activities contribute to reducing pollen viability and productivity. BRS Acauã was the most affected, with an 82% reduction in the number of seeds and a 70% reduction in production. BRS Gurguéia maintained its production, even with an increase of 4.8 °C, and can be selected as a cultivar with the potential to tolerate high temperatures as it maintained pollen viability, with less flower abortion, with the synchrony of physiological and biochemical responses and, consequently, greater production.

• The pollen viability, storage and germination results showed significant differences among the species of Casuarina and between stains (acetocarmine and aniline blue), storage duration, temperature and germination media (BA and BK medium). • The female flowers had a life span of about 10 days, and the stigma receptivity started 3 days after anthesis and peaked between the fourth and sixth days. • The receptivity period was linked to distinct morphological characteristics of the stigma, which can be used as a guide for hand pollination. • Successful storage of pollen and knowledge of the stigma receptivity time help breeders perform controlled pollination, leading to a high level of fruit and seed set. Casuarina species (Casuarinaceae) are nitrogen-fixing multipurpose trees planted extensively throughout the tropics for wood production and environmental services. Increasing productivity of plantations through breeding is underway in China, India and Thailand. The current focus of breeding is to produce interspecific hybrids possessing a combination of specifies-specific characters of forestry importance. Earlier hybridization attempts were less successful mainly due to insufficient knowledge on floral and pollen biologyand lack of efficient hand pollination techniques. The differences in the floral characters and their influence on pollen and seed output in three Casuarina species (C. equisetifolia, C. junghuhniana and C. cristata) were studied. Pollen from six clones were stored at three temperature levels and tested for viability from 3 to 45 days after storage using acetocarmine and aniline blue stains. In vitro germination of stored pollen was testedwith two germination media from 3 to 30 days. Female flowers were control pollinated from 1 to 7 days after anthesis and stigma receptivity was determined based on presence of germinating pollen. There was a significant effect of clone, stain, storage temperature and storage period on the viability of pollen. Pollen viability and in vitro germination reduced drastically when stored at room temperature for three days but was high under low temperature storage. While the viability of pollen stored at 4°C and -20 °C comparable up to 15 days of storage, freezer storage showed better viability at 45 days. In vitro germination was higher with the broad-based Brewbaker-Kwack medium than the basic sucrose-boron medium. The stigma was receptive from the third to seven days after anthesis and the peak period was on fourth and fifth days for C. equisetifolia and C. junghuhniana and fifth and sixth days for C. cristata. The receptivity level of the stigma was associated with distinct morphological features during lifespan of the female flower. Pollen stored under low temperatures can be used to cross parents with dissimilar flowering phenology within the same flowering season and conducting hand pollination during when the stigma is at its peak receptivity will increase hybridization success. • Pollen morphology of eight coconut accessions differed in origin, stature, breeding behaviour and tolerance to abiotic stresses were characterised with scanning electron microscope. • Variations in pollen size, shape and exine ornamentation pattern was observed between the coconut genotypes. • Pollen physiological features such as pollen viability, germination and pollen tube length and pollen nutrient composition were evaluated for the eight coconut genotypes. • The results obtained revealed differences in pollen morphological features which can potentially be used for cultivar identification and the information on pollen viability, germination and pollen mineral contents amongst genotypes can support conservation and crop improvement studies of the genotypes. The micro-morphological characteristic of the pollen grain is one of the most important diagnostic traits use for identification and characterization of plants even to the accession levels. In this study, pollen grains characterization of eight different coconut (Cocos nucifera L.) accessions were carried out. Morphometric observations and structure analysis of the pollen grains were performed using light and scanning electron microscopy. The variation in length and width observed ranged from 55.06 -71.25 µm and 24.38 -36.27 µm amongst the accessions. The pollen grains exhibited an ellipsoidal to elliptical shape with the exine ornamentation ranging from granulate to reticulate. Pollen viability, germination and pollen tube length were significantly different amongst the tested accessions. While pollen viability varied from 92.35 to 96.5%, germination rate and pollen tube length (after 2 h) ranged from 16.05 - 31.59% and 234.95 μm to 414.96 μm respectively. Pollen mineral composition from different accessions also varied significantly with Malayan Orange Dwarf (MOD) recording higher phosphorus (0.93%), calcium (0.64%) and magnesium (0.38%) content while highest potassium (1.01%), sodium (0.71%), iron (149.35 ppm) zinc (119.59 ppm) and boron (45.05 ppm) content was noticed in San Ramon Tall (SNRT). This investigation highlights the fact that these pollen morphological features could be potentially used for differentiating the selected accessions thereby supporting their conservation and crop improvement studies.
