Plant cytoplasmic male sterility (CMS) is maternally inherited and often manifested as aborted pollen development, but the molecular basis of abortion remains to be identified. To facilitate an investigation of CMS in cotton, the complete sequence of cotton mitochondrial (mt) genome for CMS-D2 line ZBA was determined. The mt genome was assembled as a single circular molecule with 634,036 bp in length. A total of 194 ORFs, 36 protein-coding genes, six rRNAs, and 24 tRNAs were identified. Several chimeric genes encoding hypothetical proteins with transmembrane domains were identified. Among them, a previously unknown chimeric gene, orf610a, which is composed of atp1 and a 485-bp downstream sequence of unknown nature, was identified. RT-PCR and qRT-PCR validation indicated that orf610a was expressed specifically in a sterile line. Ectopic expression of orf610a in yeast resulted in excessive accumulation of reactive oxygen species and reduction in ATP content, in addition to inhibition of cellular growth. Transgenic A. thaliana overexpressing orf610a fused with a mitochondrial targeting peptide displayed partial male sterility. Interaction between ORF610a and the nuclear-encoded protein RD22 indicated an association between ORF610a and pollen abortion. Positive feedback during transcriptional regulation between nuclear regulatory factors and the mt CMS gene may account for the male sterility of ZBA. [ABSTRACT FROM AUTHOR]

In animal and yeast system, subcellular fractionation has been widely used in studies of protein localization and organelle proteomics. Alternatively, it has not been an effective way of study in plant system because of some experimental limitations. The main aim of this study is to optimize subcellular and subnuclear fractionation of wild-type of Arabidopsis thaliana ecotype Colombia (Col-0) leaves by comparing three different methods to isolate nuclear membrane. In the study, we at first optimized nuclear washing steps to remove chloroplast contents from nuclear fractions. By optimizing speed of centrifugation and chemical component of the nuclear washing buffer, purified nuclear fractions was obtained. After measurement of protein amount for each fraction, purity of the fractions was analyzed by western blot assay with some specific cell compartment markers such as anti-Histone3 for nuclear fraction and anti-Rubisco for cytoplasmic fraction. Also, lactate dehydrogenase enzyme assay was used to confirm purity of the fractions. Then, subnuclear fractionation was done to isolate purified nuclear membrane. Three different methods were used to separate the nuclear membrane from whole purified nucleus. Nuclear compartment markers such as anti-Histone3 and anti-Fibrillarin was used in this step. In the study, Inner Nuclear Membrane protein (AtSUN2) was used as a nuclear membrane marker. All things considered, we conclude that the method with DNase digestion and high centrifugation speed (first method) is a more effective way in separation of nuclear membrane fractions because the low centrifugal speed (second method) does not appear to be sufficient for separating the nuclear membrane, and the third method does not seem to be a very effective way as it requires a high centrifugal speed. Rice yield is closely related to plant leaf shape and chlorophyll content. In this study, we isolated and identified a narrow and rolled leaf mutant, temporarily named nrl3 with darker green leaves. Histological analysis showed that nrl3 has a reduced number of vascular bundles and undergoes abnormal abaxial sclerenchymatous cell differentiation. The NRL3 mutant phenotype was controlled by a single recessive gene, fine-mapped to a 221 kb interval between Indel3 and RM2322 on Chr3. There are 42 ORF in this interval. Sequencing identified an SNP mutant leading to a premature stop in ORF 18, the candidate gene. Bioinformation analysis indicated that NRL3 encodes a novel protein with unknown function. NRL3 is localized in cytoplasm, membrane and nucleus. Expression analysis of nrl3 showed that genes involved in chlorophyll synthesis were significantly up-regulated while those involved in chlorophyll degradation and programmed cell death (PCD) were significantly down-regulated. The expression levels of photosynthesis genes were also affected. Y2H and BIFC assays indicated that NRL3 interacts directly with NAL9/VYL to regulate leaf morphology in rice. Electric signaling and Ca2+ waves were discussed to occur in systemic wound responses. Two new overlapping scenarios were identified: (i) membrane depolarization in two special cell types followed by an increase in systemic cytoplasmic Ca2+ concentration ([Ca2+] cyt), and (ii) glutamate sensed by GLUTAMATE RECEPTOR LIKE proteins and followed by Ca2+-based defense in distal leaves. Following cytokinesis in plants, Endoplasmic MTs (EMTs) assemble on the nuclear surface, forming a radial network that extends out to the cell cortex, where they attach and incorporate into the cortical microtubule (CMT) array. We found that in these post-cytokinetic cells, the MT-associated protein CLASP is enriched at sites of EMT-cortex attachment, and is required for stable EMT tethering and growth into the cell cortex. Loss of EMT-cortex anchoring in clasp-1 mutants results in destabilized EMT arrays, and is accompanied by enhanced mobility of the cytoplasm, premature vacuolation, and precocious entry into cell elongation phase. Thus, EMTs appear to maintain cells in a meristematic state by providing a structural scaffold that stabilizes the cytoplasm to counteract actomyosin-based cytoplasmic streaming forces, thereby preventing premature establishment of a central vacuole and rapid cell elongation. Light energy is essential for photosynthetic energy production and plant growth. Chloroplasts in green tissues convert energy from sunlight into chemical energy via the electron transport chain. When the level of light energy exceeds the capacity of the photosynthetic apparatus, chloroplasts undergo a process known as photoinhibition. Since photoinhibition leads to the overaccumulation of reactive oxygen species (ROS) and the spreading of cell death, plants have developed multiple systems to protect chloroplasts from strong light. Recent studies have shown that autophagy, a system that functions in eukaryotes for the intracellular degradation of cytoplasmic components, participates in the removal of damaged chloroplasts. Previous findings also demonstrated an important role for autophagy in chloroplast turnover during leaf senescence. In this review, we describe the turnover of whole chloroplasts, which occurs via a type of autophagy termed chlorophagy. We discuss a possible regulatory mechanism for the induction of chlorophagy based on current knowledge of photoinhibition, leaf senescence and mitophagy--the autophagic turnover of mitochondria in yeast and mammals. Many scientific findings have been reported on the beneficial function of reactive oxygen species (ROS) in various cellular processes, showing that they are not just toxic byproducts. The double-edged role of ROS shows the importance of the regulation of ROS level. We report a gene, rrsRLK (required for ROS-scavenging receptor-like kinase), that encodes a cytoplasmic RLK belonging to the non-RD kinase family. The gene was identified by screening rice RLK mutant lines infected with Xanthomonas oryzae pv. oryzae (Xoo), an agent of bacterial leaf blight of rice. The mutant (1rrsRLK) lacking the Os01g02290 gene was strongly resistant to many Xoo strains, but not to the fungal pathogen Magnaporthe grisea. 1rrsRLK showed significantly higher expression of OsPR1a, OsPR1b, OsLOX, RBBTI4, and jasmonic acid-related genes than wild type. We showed that rrsRLK protein interacts with OsVOZ1 (vascular one zinc-finger 1) and OsPEX11 (peroxisomal biogenesis factor 11). In the further experiments, abnormal biogenesis of peroxisomes, hydrogen peroxide (H2O2) accumulation, and reduction of activity of ROS-scavenging enzymes were investigated in 1rrsRLK. These results suggest that the enhanced resistance in 1rrsRLK is due to H2O2 accumulation caused by irregular ROS-scavenging mechanism, and rrsRLK is most likely a key regulator required for ROS homeostasis in rice. Abstract: Sorghum is one of the pioneering cereal crops where cytoplasmic male sterility (CMS) was successfully exploited for mass production of F1 hybrid seed. Mapping genes for fertility restoration (Rf) is an important aspect of understanding the molecular basis of fertility restoration in crop plants. In this study, we fine‐mapped a fertility restoration locus, Rf2 of sorghum reported earlier (Jordan, Mace, Henzell, Klein, & Klein, ), involving two F2 populations (296A × RS29 and 296A × DSV1) and newly developed SSR markers delimited Rf2 locus to 10.32‐kb region on chromosome 2. The Rf2 locus was tightly linked with two new SSRs, MS‐SB02‐3460 (0.14 cM) and MS‐SB02‐3466 (0.75 cM) on both sides, and hosted only one gene (Sobic.002G057050) of PPR gene family. Another new SSR marker developed in the study, MS‐SB02‐37912, forms the part of PPR gene and could act as a perfect marker in marker‐assisted breeding for fertility restoration involving Rf2 in sorghum breeding. Abstract: Adoption of pigeonpea hybrids in central and southern India is showing high impact with on‐farm yield advantages of >30%. The hybrid pigeonpea technology, the first in any legume crop, is based on a cytoplasmic‐nuclear male‐sterility (CMS) system. For a long‐term sustainability of hybrid programme, it is imperative that both nuclear diversity and cytoplasmic diversity are maintained among hybrid parents. In this context, a continuous search for new CMS‐inducing cytoplasms is necessary. This paper reports detection of maternal inheritance of male sterility in the progeny derived from a natural hybrid between a wild relative [Cajanus lineatus (W. & A.) Maesen comb. nov.] of pigeonpea and an unknown pigeonpea [Cajanus cajan (L.) Millsp.] genotype. In the present study, the male sterility was maintained up to BC7F1 generation by an advanced breeding pigeonpea line ICPL 99044. This male sterility inducing cytoplasm of C. lineatus was tagged as A6. In future, this CMS genetic stock can be used to develop a range of new pigeonpea hybrids with high yield and adaptation. Abstract: In Brassica oleracea, production of F1 hybrid seeds mainly makes use of the improved Ogura cytoplasmic male sterile (CMS) line. However, reliance on one particular line is a risk, and it would be advantageous to develop other CMS lines. In this study, we transferred Diplotaxis erucoides cytoplasm to B. oleracea cultivars using an alloplasmic B. rapaCMS line as a bridge plant to avoid incompatibility between donor and recipient plants. The new B. oleraceaCMS lines, which were derived by four generations of backcrossing, had small rudimentary anthers with no pollen grain and showed complete male sterility. There was no functional defect in other floral organs, and the ability to receive normal pollen did not appear to be impaired. Moreover, the B. oleraceaCMS lines carrying D. erucoides cytoplasm had larger leaf areas and a normal plastochron. As a consequence, the B. oleraceaCMS lines carrying D. erucoides cytoplasm have the potential to be valuable alternatives for use in commercial B. oleracea hybrid seed production. Diallel crosses in an unbalanced scheme were carried out in Jatropha to (i) evaluate the additive and non-additive genetic components; (ii) select parents through the general combining ability; (iii) estimate the specific combining ability used in the crosses; and (iv) verify the existence of the maternal effect and inbreeding depression. The experiment was carried out in a complete diallel scheme with four progenitors, unbalanced for the number of crosses. The experimental design consisted of a randomized block, with 5 replications and 3 plants per plot. The following characteristics were evaluated: stem diameter (SD), number of branches (NB), plant height (PH), canopy projection on the row (CPR), canopy projection between rows (CPB), mass of hundred grains (MHG) and grain yield (GY). Estimates of variances were obtained using the method of restricted maximum likelihood, while breeding values were estimated by the best linear unbiased prediction. It was concluded that the additive effect was predominant in the genetic control for SD, CPR, and CPB; the dominance effect was predominant for PH, NB, and GY; there was a cytoplasmic effect and nuclear genes of the female parent for all evaluated traits; parents 107 and 190 are promising for reducing the size and increasing the grain yield; there was inbreeding depression for SD and GY; and the favorable crosses for increasing GY were 190x107 and 190x190. Spatiotemporal changes in cellular calcium (Ca2+) concentrations are essential for signal transduction in a wide range of plant cellular processes. In legumes, nuclear and perinuclear-localized Ca2+ oscillations have emerged as key signatures preceding downstream symbiotic signaling responses. Förster resonance energy transfer (FRET) yellow-based Ca2+ cameleon probes have been successfully exploited to measure the spatiotemporal dynamics of symbiotic Ca2+ signaling in legumes. Although providing cellular resolution, these sensors were restricted to measuring Ca2+ changes in single subcellular compartments. In this study, we have explored the potential of single fluorescent protein-based Ca2+ sensors, the GECOs, for multicolor and simultaneous imaging of the spatiotemporal dynamics of cytoplasmic and nuclear Ca2+ signaling in root cells. Single and dual fluorescence nuclear and cytoplasmic-localized GECOs expressed in transgenic Medicago truncatula roots and Arabidopsis thaliana were used to successfully monitor Ca2+ responses to microbial biotic and abiotic elicitors. In M. truncatula, we demonstrate that GECOs detect symbiosis-related Ca2+ spiking variations with higher sensitivity than the yellow FRET-based sensors previously used. Additionally, in both M. truncatula and A. thaliana, the dual sensor is now able to resolve in a single root cell the coordinated spatiotemporal dynamics of nuclear and cytoplasmic Ca2+ signaling in vivo. The GECO-based sensors presented here therefore represent powerful tools to monitor Ca2+ signaling dynamics in vivo in response to different stimuli in multi-subcellular compartments of plant cells. Pyrophosphate (PPi) is produced as byproduct of biosynthesis in the cytoplasm, nucleus, mitochondria and chloroplast, or in the tonoplast and Golgi by membrane-bound H + -pumping pyrophosphatases (PPv). Inorganic pyrophosphatases (E.C. 3.6.1.1; GO:0004427) impulse various biosynthetic reactions by recycling PPi and are essential to living cells. Soluble and membrane-bound enzymes of high specificity have evolved in different protein families and multiple pyrophosphatases are encoded in all plant genomes known to date. The soluble proteins are present in cytoplasm, extracellular space, inside chloroplasts, and perhaps inside mitochondria, nucleus or vacuoles. The cytoplasmic isoforms may compete for PPi with the PPv enzymes and how PPv and soluble activities are controlled is currently unknown, yet the cytoplasmic PPi concentration is high and fairly constant. Manipulation of the PPi metabolism impacts primary metabolism and vice versa , indicating a tight link between PPi levels and carbohydrate metabolism. These enzymes appear to play a role in germination, development and stress adaptive responses. In addition, the transgenic overexpression of PPv has been used to enhance plant tolerance to abiotic stress, but the reasons behind this tolerance are not completely understood. Sorghum is the important cereal crop around the world and hence understanding and utilizing the genetic variation in sorghum accessions are essential for improving the crop. The D2 statistics was applied to assess the diversity among 41 B-lines and 43 Rlines of sorghum identified from minicore collection. The analysis of variance revealed significant differences among the genotypes for all the characters under study. The genotypes were grouped into 13 clusters, where cluster III comprised maximum of 14 genotypes followed by cluster IV with 7 genotypes each among R-lines whereas B-lines grouped into 6 clusters with cluster I with 22 genotypes followed by cluster II with 9 genotypes. Inter-cluster distance was maximum between the clusters IX and X with 648.07 among R-lines and in B-lines maximum intercluster distance was between I and VI with 1136.00 which indicated that genotypes included in these clusters may give heterotic response and thus better segregants. Regarding genetic divergence of the genotypes, panicle weight (32.07%), SPAD readings at maturity stage (29.76) and 100 seed weight (20.49%) contributed major share among R-lines and incase of B-lines panicle weight (49.49%), primaries per panicle (25.13%) and 100 seed weight (10.77%) contributed high towards genetics. Autophagy is a major pathway for degradation and recycling of cytoplasmic material, including individual proteins, aggregates, and entire organelles. Autophagic processes serve mainly survival functions in cellular homeostasis, stress adaptation and immune responses but can also have death-promoting activities in different eukaryotic organisms. In plants, the role of autophagy in the regulation of programmed cell death (PCD) remained elusive and a subject of debate. More recent evidence, however, has resulted in the consensus that autophagy can either promote or restrict different forms of PCD. Here, we present latest advances in understanding the molecular mechanisms and functions of plant autophagy and discuss their implications for life and death decisions in the context of developmental and pathogen-induced PCD. Key message: Engineered chromosomes 1BS and 1RS offer a new alternative in the development of hybrid systems in bread wheat and triticale. Abstract: In the cytoplasmic male sterility system for hybrid wheat based on the cytoplasm of Triticum timopheevi fertility restoration is difficult, with few good restorer genes available. In the system based on the cytoplasms of Aegilops kotschyi, Ae. uniaristata and Ae. mutica, essentially all chromosomes 1B carry locus Rf that restores male fertility; male sterility manifests itself in wheats with the 1RS.1BL translocation where 1BS chromosome arm is missing. To generate male sterile wheats without the 1RS.1BL translocation, the 1BS arm was cytogenetically engineered to replace the segment with Rf with two short inserts of rye chromatin. Conversely, to enhance fertility restoration by doubling the number of restorers present for eventual use in wheat and triticale, a region of 1BS with Rf was inserted into 1RS. Alloplasmic wheats with Rf removed were completely male sterile; alloplasmic wheats with engineered 1RS carrying Rf and without normal 1B were male fertile. An exception to the ubiquitous presence of Rf is T. spelta var. duhamelianum; four accessions tested in this study gave inconsistent results but some did not restore male fertility. Engineered chromosomes 1BS and 1RS and chromosomes 1B of T. spelta offer a new alternative for practical application of a cytoplasmic male sterility system in the development of hybrid wheat and hexaploid triticale. In sexual organisms, division of the zygote initiates a new life cycle. Although several genes involved in zygote division are known in plants, how the zygote is activated to start embryogenesis has remained elusive. Here, we showed that a mutation in ZYGOTE-ARREST 3 (ZYG3) in Arabidopsis led to a tight zygote-lethal phenotype. Map-based cloning revealed that ZYG3 encodes the transfer RNA (tRNA) ligase AtRNL, which is a single-copy gene in the Arabidopsis genome. Expression analyses showed that AtRNL is expressed throughout zygotic embryogenesis, and in meristematic tissues. Using pAtRNL::cAtRNL-sYFP-complemented zyg3/zyg3 plants, we showed that AtRNL is localized exclusively in the cytoplasm, suggesting that tRNA splicing occurs primarily in the cytoplasm. Analyses using partially rescued embryos showed that mutation in AtRNL compromised splicing of intron-containing tRNA. Mutations of two tRNA endonuclease genes, SEN1 and SEN2, also led to a zygote-lethal phenotype. Grain size and shape are two crucial traits that influence grain yield and grain appearance in rice. Although several factors that affect grain size have been described in rice, the molecular mechanisms underlying the determination of grain size and shape are still elusive. In this study we report that WIDE AND THICK GRAIN 1 ( WTG1) functions as an important factor determining grain size and shape in rice. The wtg1-1 mutant exhibits wide, thick, short and heavy grains and also shows an increased number of grains per panicle. WTG1 determines grain size and shape mainly by influencing cell expansion. WTG1 encodes an otubain-like protease, which shares similarity with human OTUB1. Biochemical analyses indicate that WTG1 is a functional deubiquitinating enzyme, and the mutant protein (wtg1-1) loses this deubiquitinating activity. WTG1 is expressed in developing grains and panicles, and the GFP- WTG1 fusion protein is present in the nucleus and cytoplasm. Overexpression of WTG1 results in narrow, thin, long grains due to narrow and long cells, further supporting the role of WTG1 in determining grain size and shape. Thus, our findings identify the otubain-like protease WTG1 to be an important factor that determines grain size and shape, suggesting that WTG1 has the potential to improve grain size and shape in rice. Transmission electron microscopy was used to reveal a layer of multiply folded membranes that closely surrounded the tannin-accumulating vacuole in cells of honeysuckle petal trichomes. A huge amount of tannins were deposited in the peripheral region and the center of the vacuole. The prolific membranes extended to the tannins deposited along the vacuole periphery. It was difficult to distinguish the vacuole membrane, and it seemed as if it was the layer of multiply folded membranes that separated the vacuole lumen from the cytoplasm. In addition, there were also membrane assemblies in the cytoplasm away from the vacuole, which were continuous with the proliferated membranes bordering the vacuole. Therefore, the tannin-accumulating vacuole was in close association with a very large network of proliferated membranes. The occurrence of such a layer of multiply folded membranes around the tannin-accumulating vacuole might be a structural strategy for improvement of the efficiency of vacuolar accumulation of tannins. Nine CMS analogues from different cytoplasmic sources and their common maintainer were crossed with four perfect restorers from PET-1 source to develop of 40 sunflower hybrids. The experiment was conducted during spring season 2011 and 2012 two years continuously, under two different water environments, normal irrigated and water stress environment, where irrigation was stopped after the anthesis. The analysis of variance revealed significant differences among the sources for day to flowering, days to maturity and seed yield under both the environments, both the years and pooled over the years. The variance due to GCA and SCA showed that gene action was additive for maturity whereas, non-additive genetic effect was observed for days to flowering and seed yield under both the environments. The CMS analogues E002-91A (H. annuus), PKU-2A (H. annuus), and ARG-3A (H. argophyllus) were recorded as good combiner for early flowering in both the environments. The CMS analogs E002-91A and ARG-3A were observed good combiners for early flowering, early maturity and high seed yield, whereas, NC-41B from conventional CMS source H. petiolaris (PET-1) was recorded good combiner for late flower and early maturity and poor combiner for seed yield under both the environments. Among testers P69R was recorded good combiner for late flowering under normal water environment while, in water stress it was good combiner for early flowering but it was not good for seed yield. RCR-8297 was recorded for good combiner for late maturity and high seed yield under both the environments. The SCA estimates were highest in cross E002-91A× RCR-8297 and PKU-2A × P124R and ARG-6A × RCR-8297 for early flowering and early maturity under both the environments. The general (GCA) and specific (SCA) combining abilities among eight cytoplasmic male sterile lines and six restorers as testers were estimated using line × tester method to assess the potential of the materials in the sunflower (Helianthus annuus L.) breeding program. Test cross progenies along with their parents were evaluated in two sowing dates. Both lines and testers had significant differences for all the traits except seeds per head. The lines: LA2 and LA7 were superior combiners with significant positive general combining ability (GCA) effects for seed yield and most of the yield components. Among the testers: RF1 and RF6 were good combiners for seed yield. Most of the lines and testers with significant positive GCA effects for seed yield had significant positive GCA effect for oil yield. LA7 × RF6 had significant positive specific combining ability (SCA) effects for seed width, seed yield and oil yield. The crosses LA4 × RF1 and LA7 × RF6 had significant SCA effects for seed yield and oil yield. Estimating degree of dominance more than one for all the traits except seeds per head and percentage of kernel indicate the importance of non additive genetic effects for them; therefore for improving these traits, hybrid breeding will be more effective. Betalains replace anthocyanins as color pigments in most families of Caryophyllales. Unlike anthocyanins, betalains are derived from tyrosine via three enzymatic steps: hydroxylation of L-tyrosine to L-3,4-dihydroxyphenylalanine (L-DOPA; step 1), and conversion of L-DOPA to betalamic acid (step 2), and to cyclo-DOPA (cDOPA; step 3). The principal enzymes responsible for these reactions have been elucidated at the molecular level, but their subcellular localizations have not been explored; hence, the intracellular compartments wherein betalains are biosynthesized remain unknown. Here, we report on the subcellular localization of these principal enzymes. Bioinformatic predictors and N- and C-terminal GFP tagging in transgenic tobacco, showed that Beta vulgaris CYP76AD1 which mediates both steps 1 and 3, DODA1 that catalyzes step 2, and CYP76AD6 which also mediates step 1, were similarly localized to the cytoplasm and nucleus (although the P450s were also weakly present in the endoplasmic reticulum). These two compartments were also the principal locations of Mirabilis jalapa cDOPA5GT. The cytoplasmic and nuclear co-localization of these key enzymes in tobacco suggests that betalains are biosynthesized in the cytoplasm and/or nucleus of betalain-containing plant cells. Elucidation of the subcellular compartmentation of betalain biosynthesis will facilitate the bioengineering of the betalain biosynthetic pathway in non-betalain-containing plants. Plants are constantly challenged by various environmental stresses, including high salinity and drought, and they have evolved defense mechanisms to counteract the deleterious effects of these stresses. The plant hormone ABA regulates plant growth and developmental processes and mediates abiotic stress responses. Here, we report the identification and characterization of a novel CaWDP1 (Capsicum annuum) protein. The expression of CaWDP1 in pepper leaves was induced by ABA, drought and NaCl treatments, suggesting its role in the abiotic stress response. CaWDP1 proteins show conserved sequence homology with other known WDP1 proteins, and they are localized in the nucleus and cytoplasm. We generated CaWDP1-silenced peppers via virus-induced gene silencing (VIGS). We evaluated the responses of these CaWDP1-silenced pepper plants and CaWDP1-overexpressing (OX) transgenic Arabidopsis plants to ABA and drought. CaWDP1-silenced pepper plants displayed enhanced tolerance to drought stress, and this was characterized by low levels of leaf water loss in the drought-treated leaves. In contrast to CaWDP1-silenced plants, CaWDP1-OX plants exhibited an ABA-hyposensitive and drought-susceptible phenotype, which was accompanied by high levels of leaf water loss, low leaf temperatures, increased stomatal pore size and low expression levels of stress-responsive genes. To predict the subcellular location of ascorbate peroxidase 1 (APX1) protein in Arabidopsis thaliana, a plasmid expressing APX1-enhance green fluorescent protein (EGFP) fusion protein was constructed, transformed into Agrobacterium tunwfaciens GV3101, and transiently expressed in tobacco leaves. The localization of APX1 protein in the ceHs was determined by detecting the distribution of the fusion protein in tobacco epidermal cells. Twenty eight rice genotypes were selected for identification of fertility restoration for three CMS lines (Pusa 6A, IR79156A and IR68897A) with WA cytoplasm. On the basis of pollen fertility (%) and spikelet fertility (%), restoration of fertility was identified. Out of the 28 genotypes, 5 genotypes (Dantheshwari, Akshyadhan, IR36, URG-42 and URG-30) showed complete restoration for two CMS lines i.e. Pusa 6A and IR79156A, URG-1 showed complete sterility for three CMS lines (Pusa 6A, IR79156A and IR68897A) and Anjali showed complete sterility for two CMS lines (Pusa 6A and IR79156A). Four CMS lines and eight elite testers were crossed in 4 x 8 half diallel to obtain 32 hybrids. These lines were used to estimate the heterosis values, general combining ability (GCA) of parents and specific combining ability (SCA) effect for grain yield and its components. The research was conducted in November 2010 to March 2011 at Sukabumi. Thirty-two hybrids and their parental lines were planted using randomized complete block design with three replications. The characters of yield components and yield were observed. The variances due to SCA were larger than the variance due to GCA for characters plant height, panicle length, number of filled grain per panicle, number of unfilled grain per panicle, total grain per panicle, filled grain percentage per panicle and grain yield. Those characters were controlled by non-additive genes. BI485A and BI599A lines were good general combiner for number of productive tiller per plant, filled grain per panicle and filled grain percentage per panicle, while a good tester combiner were IR53942, CRS8, CRS9, SMD10 and SMD15. The yielding character was controlled by overdominant gene action, dominant x dominant or epistasis. The highest specific combining ability for grain yield was achieved by BI855A/SMD11. The best hybrid rice derived from WA male sterile lines was BI485A/IR53942 (10.21t/ha), while the best hybrids derivated from Gambiaca male sterile lines was BI855A/SMD11 (9.88 t/ha) and Kalinga derivated hybrid was BI665A/IR53942 (9.15 t/ha). Two major conducting tissues in plants, phloem and xylem, are composed of highly specialized cell types adapted to long distance transport. Sieve elements (SEs) in the phloem display a thick cell wall, callose-rich sieve plates and low cytoplasmic density. SE differentiation is driven by selective autolysis combined with enucleation, after which the plasma membrane and some organelles are retained. By contrast, differentiation of xylem tracheary elements (TEs) involves complete clearance of the cellular components by programmed cell death followed by autolysis of the protoplast; this is accompanied by extensive deposition of lignin and cellulose in the cell wall. Plant defensins are small, basic, cysteine-rich peptides, belonging to the antimicrobial peptide superfamily, commonly found in the plant kingdom. In this study, we cloned and characterized a plant defensin gene from Jatropha curcas (JcDef). JcDef carried conserved receptor binding sites and a cysteine motif, and it was phylogenetically grouped together with defensin Ec-AMP-D2-like in Elaeis guineensis. JcDef is localized to cytoplasm and highly expressed in young tissues with fast metabolism such as cotyledons and stem apexes. Plant tissue growth requires the interdependent cellular processes of cytoplasmic growth, cell wall extension and cell division, but the feedbacks that link these processes are poorly understood. Recent papers have revealed developmentally regulated coupling between plant cell growth and progression through both mitotic cycles and endocycles. Modeling has given insight into the effects of cell geometry and tissue mechanics on the orientation of cell divisions. Developmental inputs by auxin have been highlighted in the control of cell turgor, vacuole function and the microtubule dynamics that underlies oriented growth and division. The East Himalaya-Hengduan Mountains (EH-HM) region has a high biodiversity and harbors numerous endemic alpine plants. This is probably the result of combined orographic and climate oscillations occurring since late Tertiary. Here, we determined the genetic structure and evolutionary history of alpine oak species (including Quercus spinosa, Quercus aquifolioides, and Quercus rehderiana) using both cytoplasmic-nuclear markers and ecological niche models (ENMs), and elucidated the impacts of climate oscillations and environmental heterogeneity on their population demography. Our results indicate there were mixed genetic structure and asymmetric contemporary gene flow within them. The ENMs revealed a similar demographic history for the three species expanded their ranges from the last interglacial (LIG) to the last glacial maximum (LGM), which was consistent with effective population sizes changes. Effects of genetic drift and fragmentation of habitats were responsible for the high differentiation and the lack of phylogeographic structure. Our results support that geological and climatic factors since Miocene triggered the differentiation, evolutionary origin and range shifts of the three oak species in the studied area and also emphasize that a multidisciplinary approach combining molecular markers, ENMs and population genetics can yield deep insights into diversification and evolutionary dynamics of species. The cytoplasm of laticifers, which are plant cells specialized for rubber production and defense against microbes and herbivores, is a latex. Although laticifers share common functions, the protein constituents of latexes are highly variable among plant species and even among organs. In this study, transcriptomic and proteomic analyses of Euphorbia tirucalli 's ( Euphorbiaceae ) latex were conducted to determine the molecular basis of the laticifer's functions in this plant. The hybrid de novo assembly of Illumina mRNA-seq and expressed sequence tags obtained by Sanger's sequencing revealed 26,447 unigenes. A unigene similar to Arabidopsis embryo-specific protein 3 (AT5G62200), which is a PLAT domain-containing protein, and rubber elongation factor showed the highest expression levels. The proteome analysis, studied by liquid chromatography – mass spectrometry with the de novo assembled unigenes as the database, revealed 161 proteins in the latex, 107 of which were not detected in the stem. A gene ontology analysis indicated that the laticifer's proteome was enriched with proteins related to proteolysis, phosphatase, defense against various environmental stresses and lipid metabolisms. D-mannose-binding lectin, ricin (which lacked the N-terminal conserved ribosome-inactivating protein domain), chitinase and peroxidase were highly accumulated, as confirmed by two-dimensional polyacrylamide gel electrophoresis. Thus, the lectins and chitinase may be the major defensive proteins against pests, and the other defense-related proteins and transcripts detected in latex may work in coordination with them. In the vast majority of sexual life cycles, fusion between single-celled gametes is directly followed by nuclear fusion, leading to a diploid zygote and a lifelong commitment between two haploid genomes. Mushroom-forming basidiomycetes differ in two key respects. First, the multicellular haploid mating partners are fertilized in their entirety, each cell being a gamete that simultaneously can behave as a female, i.e. contributing the cytoplasm to a zygote by accepting nuclei, and a male gamete, i.e. only donating nuclei to the zygote. Second, after gamete union, the two haploid genomes remain separate so that the main vegetative stage, the dikaryon, has two haploid nuclei per cell. Only when the dikaryon produces mushrooms, do the nuclei fuse to enter a short diploid stage, immediately followed by meiosis and haploid spore formation. So in basidiomycetes, gamete fusion and genome mixing (sex) are separated in time. The 'living apart together' of nuclei in the dikaryon maintains some autonomy for nuclei to engage in a relationship with a different nucleus. We show that competition among the two nuclei of the dikaryon for such 'extramarital affairs' may lead to genomic conflict by favouring genes beneficial at the level of the nucleus, but deleterious at that of the dikaryon. The study used four tomato varieties, 03786 (susceptible), 05237 (susceptible), C2985 (resistant) and Wva536 (resistant) with different resistance, to formulate combinations according to Griffing (I) complete diallel cross method. And Botrytis cinerea was inoculated to investigate the incidence of gray mold. The results showed that: 1 there were significant differences in disease resistance between die rent varieties and different, combinations. 2 There were significant differences in general combining ability and special combining ability between the tested varieties, and Wva536 and C2985 had a higher general combining ability effect, and were both relatively good parents when formulating resistant combinations. Combinations Wva536 x 03786 and Wva536 x 05237 had the highest special combining ability, and thus have the value of further research and utilization. 3 The additive effect in resistance inheritance was located at the primary position, and partial dominance also existed as well as cytoplasmic effect. 4 The broad-sense heritability and narrow-sense heritability were, respectively, 89.73% and 82.15%, which were higher, indicating that the parents had a greater effect on offsprings. The experiment involved 7 different cms analogues as a female parent and 6 restores lines as male parent to synthesis of a set of these 42 hybrids. The experiment was conducted during spring season 2012 in Line×Tester breeding design and evaluated for combining ability of 7 morphological, physiological and important sunflower yield components traits. The results based on GCA effects of parents for different characters revealed that among the lines DV-10A and among testers P93R, P143R and RHA83R6 were found to be the good general combiners for seed yield and oil content with early maturing except P143R, which is good combiner for flowering and late maturity. The crosses CMS-XA×RHA83R6 and PRUN-29A×RHA83R6 were found to be superior and exhibited highest SCA effects with highest mean values for oil content but for seed yield having negative SCA effects with average for seed yield. Key message: Using DArT analysis, we demonstrated that all Solanum × michoacanum (+) S. tuberosum somatic hybrids contained all parental chromosomes. However, from 13.9 to 29.6 % of the markers from both parents were lost in the hybrids. Abstract: Somatic hybrids are an interesting material for research of nucleus-cytoplasm interaction and sources of new nuclear and cytoplasmic combinations. Analyses of genomes of somatic hybrids are essential for studies on genome compatibility between species, its evolution and are important for their efficient exploitation. Diversity array technology (DArT) permits analysis of the composition of nuclear DNA of somatic hybrids. The nuclear genome compositions of 97 Solanum × michoacanum (+) S. tuberosum [ mch (+) tbr] somatic hybrids from five fusion combinations and 11 autofused 4 x mch were analyzed for the first time based on DArT markers. Out of 5358 DArT markers generated in a single assay, greater than 2000 markers were polymorphic between parents, of which more than 1500 have a known chromosomal location on potato genetic or physical map. DArT markers were distributed along the entire length of 12 chromosomes. We noticed elimination of markers of wild and tbr fusion components. The nuclear genome of individual somatic hybrids was diversified. Mch is a source of resistance to Phytophthora infestans. From 97 mch (+) tbr somatic hybrids, two hybrids and all 11 autofused 4 x mch were resistant to P. infestans. The analysis of the structure of particular hybrids' chromosomes indicated the presence of markers from both parental genomes as well as missing markers spread along the full length of the chromosome. Markers specific to chloroplast DNA and mitochondrial DNA were used for analysis of changes within the organellar genomes of somatic hybrids. Plants have evolved a unique way of partitioning the cytoplasm of dividing cells: Instead of forming a contractile ring that constricts the plasma membrane, plant cells target membrane vesicles to the plane of division where the vesicles fuse with one another to form the partitioning membrane. Plant cytokinesis starts in the centre and progresses towards the periphery, culminating in the fusion of the partitioning membrane with the parental plasma membrane. This membrane dynamics is orchestrated by a specific cytoskeletal array named phragmoplast that originates from interzone spindle remnants. Lack of variability in sorghum is the main reason for the stagnation in crop improvement including low heterosis. Studies on somaclonal mutations of sorghum have led to the development of a diverse axillary branched grain sorghum mutant, Sb ABM of postrainy season variety A-1 (Annigeri-1) which bears multiple panicles. Considering its developmental significance and phenotypic diversity, this mutant was subjected to detailed genetic analysis. To exploit its genetic potentiality, the mutant was involved in a full diallel study along with ruling varieties to assess the combining ability and to quantify the magnitude of heterosis. Sb ABM performed exceptionally well in hybrid combinations for all the traits. Mean squares due to genotypes were highly significant for all the traits. The results on general combining ability revealed that Sb ABM was significantly better general combiner for all the traits studied. Majority of the hybrids involving Sb ABM as parental line exhibited high degree of heterosis for most of the traits, finally contributing to overall plant's vigour. Mature seeds are an ultimate physiological status that enables plants to endure extreme conditions such as high and low temperature, freezing and desiccation. Seed longevity, the period over which seed remains viable, is an important trait not only for plant adaptation to changing environments, but also, for example, for agriculture and conservation of biodiversity. Reduction of seed longevity is often associated with oxidation of cellular macromolecules such as nucleic acids, proteins and lipids. Seeds possess two main strategies to combat these stressful conditions: protection and repair. The protective mechanism includes the formation of glassy cytoplasm to reduce cellular metabolic activities and the production of antioxidants that prevent accumulation of oxidized macromolecules during seed storage. The repair system removes damage accumulated in DNA, RNA and proteins upon seed imbibition through enzymes such as DNA glycosylase and methionine sulfoxide reductase. In addition to longevity, dormancy is also an important adaptive trait that contributes to seed lifespan. Studies in Arabidopsis have shown that the seed-specific transcription factor ABSCISIC ACID-INSENSITIVE3 (ABI3) plays a central role in ABA-mediated seed dormancy and longevity. Seed longevity largely relies on the viability of embryos. Nevertheless, characterization of mutants with altered seed coat structure and constituents has demonstrated that although the maternally derived cell layers surrounding the embryos are dead, they have a significant impact on longevity. Exceptionally well-preserved Miocene pollen from the Bochnia salt mine of southern Poland is reported herein. The halite deposits within the salt mine belonging to Late Badenian (Miocene) marine evaporites originated in the Paratethys. Rounded and angular structures are present inside pollen grains. On the basis of the similarity with plasmolyzed pollen grains of modern plants, these structures are considered to represent cytoplasms plasmolyzed in the condensed brine prior to fossilization. Two forms of plasmolyzed cytoplasms (concave and convex) can be observed in modern pollen. Both are distinguished in the investigated fossil material. In porate and colporate grains the shape of the plasmolyzed cellular content is concave while in inaperturate it is convex. The plasmolysis form depends on the type of apertures and pollen shape. The percentage of pollen with fossilized cytoplasms within individual taxa is a valuable environmental indicator, as it depends on the proximity of the pollen-producing plant assemblages to the depositional setting. Interspecific hybridization can be considered an accelerator of evolution, otherwise a slow process, solely dependent on mutation and recombination. Upon interspecific hybridization, several novel interactions between nuclear and cytoplasmic genomes emerge which provide additional sources of diversity. The magnitude and essence of intergenomic interactions between nuclear and cytoplasmic genomes remain unknown due to the direction of many crosses. This study was conducted to address the role of nuclear-cytoplasmic interactions as a source of variation upon hybridization. Wheat ( Triticum aestivum) alloplasmic lines carrying the cytoplasm of Aegilops mutica along with an integrated approach utilizing comparative quantitative trait locus (QTL) and epigenome analysis were used to dissect this interaction. The results indicate that cytoplasmic genomes can modify the magnitude of QTL controlling certain physiological traits such as dry matter weight. Furthermore, methylation profiling analysis detected eight polymorphic regions affected by the cytoplasm type. In general, these results indicate that novel nuclear-cytoplasmic interactions can potentially trigger an epigenetic modification cascade in nuclear genes which eventually change the genetic network controlling physiological traits. These modified genetic networks can serve as new sources of variation to accelerate the evolutionary process. In this study, the atp8 gene was cloned from the cytoplasmic male sterile (CMS) line UG93A and its maintainer line UG93B in kenaf. Its DNA sequence analysis showed that atp8 containing 480-bp, encoding 159 amino acid residues, and a 9-bp insertion was found at the 3′flanking sequence in UG93A compared with UG93B. The cDNA sequence of atp8 analyzed by RT-PCR indicated that there were five loci edited, but six loci edited in UG93B. The editing frequencies were higher in sterile cytoplasm than in fertile cytoplasm. The relative expression of atp8 analyzed by real-time PCR showed that the expressed level of atp8 in UG93A was lower than that of its maitainer UG93B and its F hybrid UG93A/992 (a restore line). Furthermore, based on the difference of the 9-bp differences at the 3′flanking sequence of atp8 between UG93A and UG93B, a molecular marker specific to male sterile cytoplasm was developed, which can be used for indentifying whether any germplasm of kenaf is male sterile cytoplasm or male fertile cytoplasm. We characterized the function of the rice cytosolic hexokinase OsHXK7 ( Oryza sativa He xo kinase 7), which is highly upregulated when seeds germinate under O2-deficient conditions. According to transient expression assays that used the promoter:luciferase fusion construct, OsHXK7 enhanced the glucose (Glc)-dependent repression of a rice α-amylase gene ( RAmy3D) in the mesophyll protoplasts of maize, but its catalytically inactive mutant alleles did not. Consistently, the expression of OsHXK7, but not its catalytically inactive alleles, complemented the Arabidopsis glucose insensitive2-1 ( gin2-1) mutant, thereby resulting in the wild type characteristics of Glc-dependent repression, seedling development, and plant growth. Interestingly, OsHXK7-mediated Glc-dependent repression was abolished in the O2-deficient mesophyll protoplasts of maize. This result provides compelling evidence that OsHXK7 functions in sugar signaling via a glycolysis-dependent manner under normal conditions, but its signaling role is suppressed when O2 is deficient. The germination of two null OsHXK7 mutants, oshxk7-1 and oshxk7-2, was affected by O2 deficiency, but overexpression enhanced germination in rice. A study of latex harvesting technologies, appropriate to the opening of tapping of Hevea brasiliensis clone GT 1 having moderate metabolism, was undertaken in south eastern Côte d'Ivoire. Rubber trees were planted in 1989 at a density of 510 trees/ha and harvesting was started in 1995. Ten different latex harvesting systems were tested and monitored for seven years. The parameters measured were rubber yield, isodiametric growth of the trunk, physiological profile and sensitivity to tapping panel dryness. The results showed that tapping with hormonal stimulation highly activated the metabolism of latex vessels. They also indicated that the tapping opening at the height of 1.20m from the ground level of downward tapping showed high yield than the opening at 0.70m in upward tapping and not followed by downward tapping for a minimum period equivalent to four years. Half-spiral upward tapping from the fifth year certainly induces very high yield. However, a high rate of tapping panel dryness and signs of early physiological fatigue were observed, probably due to over-exploitation. The different hormonal stimulation strategies caused little damage on the growth of the trunk and the physiological parameters of the latex. The latex harvesting system which is most suitable for clone GT 1, at opening is S/2 d4 6d/7 12 m/12.ET2.5% Pa1 (1) 4/y at 1.20m above ground. Lippia alba (Verbenaceae) is an aromatic shrub recently described as a new tropical polyploid species with five distinct chromosome numbers (2 n = 30, 38, 45, 60, and 90) with 2 n = 30, 45, and 60 being the most frequent ones. Cytomixis is a cellular process known as the migration of genetic material mainly between meiocytes through cytoplasmic connections or cytomictic channels. This phenomenon has been reported in various plant families such as Leguminosae, Brassicaceae, Poaceae, Apocynaceae, Liliaceae, Rutaceae, and others. The transference of genetic material between pollen mother cells (PMCs), by cytomictic channels, induces the formation of unbalanced and unreduced (2 n) gametes, and is considered a possible source of aneuploid and polyploid plants. Here, we describe for the first time, the occurrence of cytomixis in meiotic cells of L. alba (tetraploid cytotype) analyzing data obtained from meiotic behavior assays. In addition, the pollen size and viability were also evaluated. A high index of irregularities during meiosis was observed as well as unviable pollen with heterogeneous size. Approximately 80 % of zygotene cells showed genetic material exchange. Allopolyploids, formed by hybridization and chromosome doubling, face the immediate challenge of having duplicated nuclear genomes that interact with the haploid and maternally inherited cytoplasmic (plastid and mitochondrial) genomes. Most of our knowledge of the genomic consequences of allopolyploidy has focused on the fate of the duplicated nuclear genes without regard to their potential interactions with cytoplasmic genomes. As a step toward understanding the fates of nuclear-encoded subunits that are plastid-targeted, here we examine the retention and expression of the gene encoding the small subunit of Ribulose-1, 5-bisphosphate carboxylase/oxygenase (Rubisco; rbcS) in multiple populations of allotetraploid Tragopogon miscellus (Asteraceae). These polyploids formed recently (~80 years ago) and repeatedly from T. dubius and T. pratensis in the northwestern United States. Examination of 79 T. miscellus individuals from 10 natural populations, as well as 25 synthetic allotetraploids, including reciprocally formed plants, revealed a low percentage of naturally occurring individuals that show a bias in either gene (homeolog) loss (12%) or expression (16%), usually toward maintaining the maternal nuclear copy of rbcS. For individuals showing loss, seven retained the maternally derived rbcS homeolog only, while three had the paternally derived copy. All of the synthetic polyploid individuals examined (S0 and S1 generations) retained and expressed both parental homeologs. The plant-specific protein motor class myosin XI is known to function in rapid bulk flow of the cytoplasm (cytoplasmic streaming) and in organellar movements. Recent studies unveiled a wide range of physiological functions of myosin XI motors, from intracellular motility to organ movements. Arabidopsis thaliana has 13 members of myosin XI class. In vegetative organs, myosins XIk, XI1, and XI2 primarily contribute to dynamics and spatial configurations of endoplasmic reticulum that develops a tubular network in the cell periphery and thick strand-like structures in the inner cell regions. Myosin XI-i forms a nucleocytoplasmic linker and is responsible for nuclear movement and shape. In addition to these intracellular functions, myosin XIf together with myosin XIk is involved in the fundamental nature of plants; the actin–myosin XI cytoskeleton regulates organ straightening to adjust plant posture. Male sterile (MS) plants fail to produce functional anthers, pollen, or male gametes. This condition is useful in sunflower hybrid seed production. A MS form identified in an invasive population of H. annuus located in Las Malvinas was characterized by 37 morphological variables, six mitochondrial genes ( atpA, atp6, coxIII, orfb, orfB and orfC) associated with cytoplasmic male sterility (CMS), five SSR loci associated with ms9, ms10, and ms11 nuclear male sterility genes, and the HRGO2 marker associated with the Rf1 fertility restoration gene for CMS-PET1. Restoration of MS condition was evaluated through crosses with 13 sunflower inbred lines. Las Malvinas site was described regarding its population size, spatial distribution, and male sterility occurrence. Ten years after the first observation, population size was over 250,000 individuals. The adoption of this species as ornamental plant in home gardens has contributed its expansion. The low frequency of MS plants found in natural conditions (0.4 %) is concordant with a CMS form in combination with restoration genes. Wild individuals harboring the CMS-PET1 cytoplasm and individuals carrying a different mitochondrial genome were found. The Rf1 restorer gene for CMS-PET1 was not detected. The new type of male sterility was different than CMS-PET1 in pollen morphology; absence of orfC; and different response to crosses with inbred lines. Molecular differences were found with CMS-RES1 and CMS-MAX1. The male sterility could not be explained by differences in the cytoplasmic or nuclear regions studied.
