In the present study, we describe a natural outbreak of carp edema virus disease (CEVD) in koi carp, concentrating on clinical manifestation, gross and microscopic pathology, immunological parameters, viral diagnostics, and phylogenetic analysis. Examination of white blood cell parameters showed increased monocyte and decreased lymphocyte counts in CEV-affected fish compared to healthy control fish. Regarding immune system functioning, the present work shows, for the first time, enhanced phagocytic activity in CEV-affected fish. Respiratory burst of phagocytes was strongly increased in diseased fish, the increase being attributed to an increased phagocyte count rather than enhancement of their metabolic activity. Disease conditions that involve multiple predisposing or contributing factors, or manifest as low performance and/or low-level mortality, can pose a diagnostic challenge that requires an interdisciplinary approach. Reaching a diagnosis may also be limited by a lack of available clinical profile parameter reference ranges to discriminate healthy fish from those affected by specific disease conditions. Here, we describe our experience investigating poorly performing rainbow trout (Oncorhynchus mykiss) in an intensive recirculation aquaculture, where reaching a final diagnosis of nephrocalcinosis was not as straightforward as one would wish. To list the issues making the diagnosis difficult, it was necessary to consider the creeping onset of the problem. Further diagnostic steps needed to ensure success included obtaining comparative data for fish blood profiles and water quality from both test and control aquacultural systems, excluding infections with salmonid pathogenic agents and evaluating necropsy findings. The present study aimed at the aetiological diagnosis of skin tumours of roe deer (Capreolus capreolus) in the Czech Republic. A total of 33 roe deer specimens showing skin masses were sampled for histopathology, virus detection and identification using polymerase chain reaction (PCR) and deoxyribonucleic acid sequence (DNA) analysis, and to investigate fibropapilloma-associated levels of metallothionein, zinc and oxidative stress in 2012 and 2013. Ticks (Ixodes ricinus) and deer keds (Lipoptena cervi) were also collected and pooled from sampled game specimens. Ticks found on dogs in hunting grounds under study were pooled into another sample. Skin tumours, ranging from 2 to 10 per inspected specimen and 2 to 5 cm in size, were classified as fibropapillomas by histopathology. All 33 viral-infection-suspected skin samples from roe deer in South Moravia and South Bohemia were PCR positive. The nucleotide sequences of PCR products were 100% homologous to the Western roe deer papillomavirus 1 isolate CcPV-1. Ticks and deer keds from positive roe deer and ticks from dogs were PCR and DNA sequence positive for the roe deer papillomavirus. Viral DNA was also demonstrated in one blood sample from a roe deer female. Differences in metallothionein, zinc, taurine and electrochemical index among samples from lesions and normal skin of affected roe deer and negative controls from papillomavirus-non-infected animals were non-significant. While we have demonstrated circulation of specific roe deer papillomavirus in the Czech Republic that results in multiple fibropapillomatous skin tumours, many ecological and epidemiological issues of this wildlife disease still remain unanswered.

This study reported on the possibility of intoxications of non-target wild animals associated with use of bromadiolone as the active component of rodenticides with anticoagulation effects. A laboratory test was done with earthworms were exposed to bromadiolone-containing granules under the conditions specified in the modified OECD 207 guideline. No mortality of earthworms was observed during the fourteen days long exposure. When the earthworms from the above test became a part of the diet of common voles in the following experiment, no mortality of consumers was observed too. However, electrochemical analysis revealed higher levels of bromadiolone in tissues from earthworms as well as common voles compared to control animals. There were determined comparable levels of bromadiolone in the liver tissue of common voles after primary (2.34 +/- 0.10 mu g/g) and secondary (2.20 +/- 0.53 mu g/g) intoxication. Therefore, the risk of secondary intoxication of small mammalian species feeding on bromadiolone-containing earthworms is the same as of primary intoxication through baited granules. Bromadiolone bio- accumulation in the food chain was monitored using the newly developed analytical procedure based on the use of a liquid chromatography coupled with electrochemical detector (HPLC-ED). The HPLC-ED method allowed to determine the levels of bromadiolone in biological samples and is therefore suitable for examining the environmental hazards of this substance.

Práce se zabývá možností použití lineárních Hallových snímačů vyráběných jako integrované obvody pro konstrukci absolutních snímačů polohy a natočení. Hallovy snímače ve spojení s permanentními magnety umožňují rozměrově malou konstrukci. V této práci je popsán návrh a řešení absolutního lineárního snímače polohy pro lineární pohon Stewartovy platformy a absolutních snímačů natočení pro robotický manipulátor Mini-swing. Použití těchto snímačů přináší mnoho výhod, ty hlavní jsou minimální rozměry a vysoká rozlišovací přesnost., This work deals with possibilities of use of linear Hall sensors which are made as integrated circuit for construction of absolute sensors of position and rotary position. Hall sensors with permanent magnets allow a smaller construction. In this work there is described a design and solution of absolute linear sensor of position for linear drive of Stewart's platform and absolute sensors of rotary position for robotic manipulator Mini-swing. Use of these sensors adds many advantages, primary minimal size and high resolution.
