Abstract

Background Intrinsic capacity (IC) is a concept related to functionality that reflects healthy aging. ATPase inhibitory factor 1 (IF1) is a multifaceted protein that regulates mitochondrial oxidative phosphorylation (OXPHOS), and may be involved in IC. The objective of this study is to investigate the association between plasma levels of IF1 and IC changes in community-dwelling older adults. Methods Community-dwelling older adults from the Multidomain Alzheimer Preventive Trial (MAPT Study) were enrolled in this study. A composite IC score was calculated based on 4 IC domains: locomotion, psychological dimension, cognition, and vitality (with data available annually over 4 years of follow-up). Secondary analyses were conducted on the sensory domain (with data available only for 1 year of follow-up). Mixed-model linear regression adjusted for confounders was conducted. Results A total of 1 090 participants with usable IF1 values were included in the study (75.3 ± 4.4 years; 64% females). Compared to the lowest quartile, both the low– and high–intermediate IF1 quartiles were found to be cross-sectionally associated with greater composite IC scores across 4 domains (βlow–intermediate, 1.33; 95% confidence interval [CI] 0.06–2.60 and βhigh–intermediate, 1.78; 95% CI 0.49–3.06). In the secondary analyses, the highest quartile was found to be associated with a slower decline in composite IC scores across 5 domains over 1 year (βhigh 1.60; 95% CI 0.06–3.15). The low– and high–intermediate IF1 quartiles were also found to be cross-sectionally associated with greater locomotion (βlow–intermediate, 2.72; 95% CI 0.36–5.08) and vitality scores (βhigh–intermediate, 1.59; 95% CI 0.06–3.12), respectively. Conclusions This study is the first to demonstrate that levels of circulating IF1, a mitochondrial-related biomarker, are associated with IC composite scores in both cross-sectional and prospective analyses among community-dwelling older adults. Abstract

BackgroundIntrinsic capacity (IC) is a function-related concept that reflects healthy aging. Identifying IC biomarkers is an essential step to slow down functional declines at early stages. ATPase inhibitory factor 1 (IF1) is a multifaceted protein that regulates mitochondrial oxidative phosphorylation (OXPHOS), thereby affecting cellular energy production.ObjectiveTo investigate the association between plasma levels of IF1 and IC changes over four years in community-dwelling older adults.MethodsCommunity-dwelling older adults aged over 70 years at risk of cognitive decline from the Multidomain Alzheimer Preventive Trial (MAPT Study) were enrolled in this study. A composite IC score was calculated (ranging from 0 to 100; higher is better) over time using data on the following IC domains: locomotion, psychological dimension, cognition, vitality, and sensory ability (vision and hearing; assessed over one year only). Plasma levels of IF1 were assessed one year after the beginning of the study. Mixed-model linear regression adjusted for confounders was performed.ResultsA total of 1090 participants had usable IF1 values (mean age 75.3 ± 4.4 years; 64% females). The mean values of plasma IF1 and IC (4 domains) were 565.62 ± 251.92 ng/mL and 74.85 ± 8.43, respectively. Compared to the lowest quartile, low- and high-intermediate IF1 quartiles were cross-sectionally associated with greater composite IC scores of four domains (βlow-intermediate, 1.33; 95% CI 0.06– 2.60 and βhigh-intermediate, 1.78; 95% CI 0.49–3.06), and the highest quartile associated with a slower decline in composite IC scores of five domains over one year (βhigh 1.60; 95% CI 0.06– 3.15). The low- and high-intermediate IF1 quartiles were cross-sectionally associated with greater locomotion (βlow-intermediate, 2.72; 95% CI 0.36–5.08) and vitality scores (βhigh-intermediate, 1.59; 95% CI 0.06–3.12), respectively.ConclusionThis study is the first to report circulating IF1 levels as a mitochondrial-related biomarker associated with IC composite scores in cross-sectional and prospective analyses in community-dwelling older adults. Further research is needed to confirm these findings, in particular, to determine a potential cut-off defining optimal plasma IF1 levels and to unravel the potential mechanisms that can explain these associations.

Abstract

Late‐life aging is often associated with appetite reduction and weight loss. Physical activity (PA) may prevent these processes, but the molecular mechanisms involved remain elusive. The present study investigated the putative mediating aspect of growth differentiation factor 15 (GDF‐15), a stress signalling protein involved in aging, exercise and appetite control, on the association between PA and late‐life‐associated weight loss. One thousand eighty‐three healthy adults (63.8% women) aged 70 years and over who participated in the Multidomain Alzheimer Preventive Trial were included. Bodyweight (kg) and PA levels (square root of metabolic equivalent of task‐min/week) were assessed repeatedly from baseline to the 3‐year visit, whereas plasma GDF‐15 (pg/mL) was measured at the 1‐year visit. Multiple linear regressions were performed to test the association between first‐year mean PA level, 1‐year visit GDF‐15 concentration and subsequent bodyweight changes. Mediation analyses were used to investigate whether GDF‐15 mediated the association between first‐year mean PA levels and consecutive bodyweight changes. Multiple regression analyses demonstrated that higher first‐year mean PA levels significantly predicted lower GDF‐15 and bodyweight at 1 year (B = −2.22; SE = 0.79; P= 0.005). In addition, higher 1‐year visit GDF‐15 levels were associated with faster subsequent bodyweight loss (Time × GDF‐15 interaction B = −0.0004; SE = 0.0001; P= 0.003). Mediation analyses confirmed that GDF‐15 mediated the association between first‐year mean PA levels and subsequent bodyweight changes (mediated effect ab = 0.0018; bootstrap SE = 0.001; P< 0.05) and revealed that mean PA had no direct effect on subsequent bodyweight changes (c′ = 0.006; SE = 0.008; P> 0.05). This study suggests that GDF‐15 may be one of the molecules mediating the link between PA and late‐life weight loss, but mechanistic studies are necessary to further support the present findings.

Abstract

Although the iron-heart disease hypothesis is prevalent, the epidemiological findings are incongruent. The relationship of serum ferritin with early cardiovascular disease (CVD), particularly atheroscterosis, has not been evaluated extensively, particularly with accounting for inflammation. We examined this association in a case-control study of 124 age- and sex-matched pairs embedded in the population-based random sample (MONICA survey) in Southwest France, taking into account inflammation status. Cases had [greater than or equal to] 2 carotid atherosclerotic plaques and controls had none. Inflammation was assessed using several markers, including serum [alpha]-1 acid glycoprotein (AGP) and high sensitivity C-reactive protein. There was an interaction of inflammation with group (case/control) for serum ferritin. In adults without elevated AGP, serum ferritin was significantly greater in atherosclerotic cases than in adults in the control group. In models adjusted for CVD risk factors, the odds of atherosclerosis increased with the increase in serum ferritin in individuals without elevated AGP; for every 10-[micro]g/L increase in serum ferritin, the risk for atherosclerosis increased by 3% (odds ratio [95% CI]: 1.03 [1.01-1.06]). In conclusion, carotid atherosclerosis was positively associated with serum ferritin in individuals free from subclinical inflammation based on AGP. Abstract

International audience; HDL-Cholesterol (HDL-C) is not an accurate surrogate marker to measure the cardioprotective functions of HDL in coronary artery diseases (CAD) patients. Hence, measurement of other HDL-related parameters may have prognostic superiority over HDL-C. In this work, we examined the predictive value of HDL particles profile for long-term mortality in CAD patients and to compare its informative value to that of HDL-C and apoA-I. HDL particles profiles were measured by nuclear magnetic resonance (NMR) spectroscopy in 214 male participants with stable CAD (45-74 years). Median follow up was 12.5 years with a 36.4% mortality rate. Cardiovascular mortality accounted for 64.5%. Mean concentrations of total HDL particles (HDL-P), small-sized HDL (SHDL-P) and apoA-I were lower in deceased than in surviving patients whereas no difference was observed according to HDL-C and large HDL particles. All NMR-HDL measures were correlated between themselves and with other HDL markers (HDL-C, apoA-I and LpA-I). In a multivariate model adjusted for cardiovascular risk factors and bioclinical variables, HDL-P and SHDL-P displayed the strongest inverse association with all-cause and cardiovascular mortality. Weaker associations were recorded for apoA-I. Based on our results, we conclude that HDL particle profile measured by NMR spectroscopy should be considered to better stratify risk in population at high risk or in the setting of pharmacotherapy.

Abstract

The biochemical and physical properties of remnant high-density lipoprotein2 (HDL2) and prebeta1-HDL, which were generated by hepatic lipase, were examined via density gradient ultracentrifugation. The results revealed that the production of remnant-HDL2 particles, which display fluidity of the lipid environment and modifications of apoA-1 conformation, is attributed to hepatic lipase. It was also suggested by the results that hepatic lipase is not responsible for the promotion of the direct conversion of HDL2 to HDL3.

Abstract

Studies have shown that omega-3 polyunsaturated fatty acids (PUFAs) are associated with brain, cardiovascular, and immune function, as well as physical performance and bone health in older adults. So far, few studies have investigated the associations between PUFA status and performance-based tests of physical function. The objective of this study was to investigate the associations between the omega-3 PUFA levels (eicosapentaenoic acid (EPA), docosahexaenoic acid (DHA)) in red blood cell (RBC) membranes and physical performance, in a sample of community-dwelling older adults. This is a longitudinal observational study using data from the Multidomain Alzheimer's Disease Trial (MAPT), a randomized, placebo-controlled trial. Four hundred participants from MAPT placebo group with available PUFA data were included. Omega-3 PUFA levels in RBC membranes were measured at baseline, and their percentage of total RBC membrane fatty acid content was calculated. We dichotomized the standardized omega-3 PUFA levels in RBC membranes as low (lowest quartile) vs. high (three upper quartiles). Gait speed (in m/s) and short physical performance battery (SPPB) score (range from 0 to 12, higher is better) were used to assess physical performance at baseline and after 6, 12, 24, and 36 months. Participants were 75.2 (± 4.3) years old and 68% were female. Bivariate analyses found that the characteristic of the participants in the lowest quartile of omega-3 PUFA levels (Q1) and those in the three upper quartiles (Q2-Q4) was not different at baseline; only those in Q1 were slightly older. In an unadjusted model, the difference in gait speed after 3 years of follow-up was significant (- 0.09 ± 0.03 m/s; p = 0.008) between participants in Q1 and those in Q2-Q4. In a model adjusted for age, gender, educational level, cognitive function, depressive status, body mass index, physical activity, grip strength, and their time interaction, this difference remained clinically relevant (- 0.07 ± 0.04 m/s; p = 0.075). No difference between the two groups was found for the SPPB score development over 3 years. Older adults with subjective memory complaints and in the lower quartile of omega-3 have a faster decline on gait speed compared to people in the three upper quartiles. Other longitudinal studies are needed to explore this association and to examine mechanisms.

Abstract

Omega-3 (n-3) and 6 (n-6) polyunsaturated fatty acids (PUFAs) have been associated with reduced cognitive decline in observational studies. Hence, we examined the cross-sectional associations between cortical β-amyloid (Aβ) and erythrocyte membrane PUFAs in 61 non-demented elderly individuals reporting subjective memory complaints from the Multidomain Alzheimer Preventive Trial (MAPT) placebo arm. Cortical-to-cerebellar standard uptake value ratios were obtained using [18F] florbetapir positron emission tomography (PET). Fatty acids were measured in erythrocyte membranes by gas chromatography. Associations were explored using adjusted multiple linear regression models and were considered significant at p ≤ 0.005 after correction for multiple testing (10 comparisons). We found no significant associations between cortical Aβ and erythrocyte membrane PUFAs. The associations closest to significance after adjustment were those between Aβ and erythrocyte membrane arachidonic acid (No apolipoprotein E status adjustment: B-coefficient, 0.03; CI, 0.01,0.05; p = 0.02. Furthermore, the association between Aβ and erythrocyte membrane arachidonic acid seemed to be specific to Apolipoprotein E e4 non-carriers (B-coefficient 0.03, CI: 0.00,0.06, p = 0.03, n=36). In contrast, no association was found between Aβ and erythrocyte membrane linoleic acid in Apolipoprotein E e4 stratified analysis. Investigating the relationships between Aβ and PUFAs longitudinally would provide evidence as to whether fatty acids, particularly arachidonic acid and linoleic acid, might modulate cognition through Aβ-dependent mechanisms. Abstract

An in vitro study was undertaken to determine the high- and low-affinity sites of the remnant plasma high-density lipoprotein 2 (HDL2) eliminated by the liver and compare the HDL2 interaction with the hepatocyte cell line from those triglyceride rich HDL2. The levels of plasma HDL are inversely related to the occurrence of atherosclerosis and coronary heart disease. Results revealed that remnant HDL2 binded to two classes of sites in contrast with the triglyceride rich HDL2 and the latter was able to compete for the high-affinity binding site.

Abstract

The role of apolipoprotein A-I (apoA-I) in the uptake of high-density lipoprotein 3 (HDL3) into human hepatoma cells (HepG2) at 37 degrees Centigrade is discussed. Binding assays, kinetic assays and inhibition experiments indicate the role of clathrin-coated vesicles in the internalization of HDL3. These vesicles are formed by the interaction of apoA-I and a cell surface protein. Blocking the formation of these vesicles by treating cells with saccharose hypertonic shock inhibits uptake of HLD3 by HepG2.

Abstract

The human preovulatory follicular fluid, which has only high-density lipoprotein (HDL) as a lipoprotein class and a substantial amount of pre-beta-1 HDL, was studied to determine the characteristics of pre-beta-1 HDL. The apparent molecular mass of pre-beta-1 HDL was found to be 61.6 kDa. They contain very little lipids compared to follicular fluid HDL. Cholesteryl ester and triacylglycerol molecules made up 8 to 9% of the molecular weight.
