Background: Exosomes produced by mesenchymal stem cells (MSCs) have vascular generative properties and are considered new effective candidates for the treatment of bone defects as alternatives to cell therapy. Improving the pro-regenerative function and efficacy of exosomes has been a popular research topic in the field of orthopaedics. Methods: We prepared mesoporous bioactive glass (mBG) microspheres via the template method. The ionic products of mBGs used to treat MSCs were extracted, and the effects of exosomes secreted by MSCs on osteoblast (OB) and macrophage (MP) behaviour and bone defect repair were observed in vivo (Micro-CT, H&E, Masson, and immunofluorescence staining for BMP2, COL1, VEGF, CD31, CD163, and iNOS). Results: The mBG spheres were successfully prepared, and the Exo-mBG were isolated and extracted. Compared with those in the blank and Exo-Con groups, the proliferation and osteogenic differentiation of OBs in the Exo-mBG group were significantly greater. For example, on Day 7, OPN gene expression in the Ctrl-Exo group was 3.97 and 2.83 times greater than that in the blank and Exo-mBG groups, respectively. In a cranial defect rat model, Exo-mBG promoted bone tissue healing and angiogenesis, increased M2 macrophage polarisation and inhibited M1 macrophage polarisation, as verified by micro-CT, H&E staining, Masson staining and immunofluorescence staining. These effects may be due to the combination of a higher silicon concentration and a higher calcium-to-phosphorus ratio in the mBG ionic products. Conclusion: This study provides insights for the application of exosomes in cell-free therapy and a new scientific basis and technical approach for the utilisation of MSC-derived exosomes in bone defect repair. The translational potential of this article: Our study demonstrated that exosomes produced by mBG-stimulated MSCs have excellent in vitro and in vivo bone-enabling and immunomodulatory functions and provides insights into the use of exosomes in clinical cell-free therapies.

In order to solve the problem of many coal remains, easy to spontaneous combustion and difficult to be sealed in the goaf near the two ends of roadway of the working face in Shendong Mining Area, this paper proposes a new type of inorganic paste fire preventing and extinguishing filling material, which can efficiently and quickly fill the upper and lower corners of the working face. Fly ash, slag and P·O·42.5 silicate cement, which are easily accessible in the vicinity of Shendong Mining Area, are used as the main raw materials of the paste, and the physical properties and chemical compositions of the materials are investigated through indoor tests, and the optimal ratio of the paste and the amount of additives are determined through the paste liquidity test, the paste solidification test, the paste expansion test and the paste filling strength test. The test results show that: the slag base paste has good self-expansion, the self-expansion rate can reach 5.6% in 7 days, and the initial setting time is short, which can meet the strength requirements of the project. The best proportioning scheme of the paste fireproofing material is cement : fly ash : slag = 1 : 4 : 20, and the mass fraction is 78%, and the addition amount of J85 accelerator is 6%. The engineering application shows that, after using the preferred paste filling material for corner filling and sealing, the air leakage in the mining airspace is reduced to 30% of that before filling, the CO volume fraction is reduced from 120×10−6 to 46.5×10−6, the O2 volume fraction is reduced to less than 10% at a point of about 80 m in depth of the strike length of the mining area, and the strike length of the dispersion zone + oxidation zone in the goaf is shortened from 160 m before filling to 80 m.

The Silk Road Economic Belt has become a pivotal region for China's sustainable development, focusing on the balance between carbon emissions and ecological resilience. Understanding the coordination between these factors is crucial for advancing China's westward economic shift and optimizing urban and demographic structures. This study investigates the coupling and coordination dynamics between carbon emissions and ecological resilience across 15 key cities in the Silk Road Economic Belt, using data from 2008 to 2018. By applying a ''pressure-state-response'' framework, the ecological resilience of the region is quantitatively assessed. Spatial disparities and the dynamic evolution of coupling coordination are analyzed through the Gini coefficient and kernel density estimation. The findings reveal significant regional variations in ecological resilience and a decline in coupling coordination over time. Shifts in the patterns of coordination indicate opportunities for enhancing resource utilization and environmental management. This research provides theoretical insights for policymakers to improve green development strategies, optimize industrial structures, and plan urban green spaces more effectively within the Silk Road Economic Belt.

Super-gravity field was reported to strongly intensify the buoyancy-driven flow of liquid during solidification, affecting the solute convection and morphology of eutectic structure in binary alloys. In ternary eutectic alloys, certain two-phase regions with a coarse structure can usually be detected concurrently along with three-phase eutectics, leading to the formation of heterogeneous structure with limited ductility. Currently, Al–Ce–Ni ternary eutectic alloy was chosen as a modal system, which was reported for the first time to be solidified under super-gravity field with high volume fraction of three-phase eutectics. Meanwhile, the formation of ultrafine-fibrous interconnected Al11Ce3–Al3Ni eutectic clusters with more “chains” and higher spatial connectivity was detected mainly due to the relatively higher nucleation rate. The high volume fraction and refinement of interconnected Al11Ce3–Al3Ni eutectic clusters with excellent thermal stability could contribute to enhancement of ductility and tensile strength at elevated-temperature. Interestingly, the strength retention at 300 °C and 400 °C were able to reach up to ∼68.1 % and ∼33.2 %, respectively.

Abstract Reservoir stimulation using hydraulic fracturing technology is one of the most common stimulation measures to develop tight oil and gas reservoirs, and the conductivity of fractures after fracturing is the key to the stimulation effect of oil fields. In this paper, the laboratory simulation system of fracture conductivity is used to study the long‐term fracture conductivity under the displacement of formation water and fracturing fluid. The experimental results show that the fracture conductivity decreases with the increase of displacement time under the displacement of formation water and fracturing fluid, and the decrease of fracture conductivity under the displacement of fracturing fluid with time is greater than that of formation water displacement because of the plugging effect of fracturing fluid residue on pore channels. The use of formation water and fracturing fluid circulation displacement can delay the decline rate of conductivity with time. The conductivity increases with the increase of sand concentration, proppant particle size, and elastic modulus, and decreases with the increase of closure pressure. In addition, by studying the influence of elastic modulus on fracture width, it is found that the smaller the elastic modulus is, the more serious the embedding phenomenon of proppant is, and the smaller the fracture width is.

Abstract Accurate and efficient molecular recognition plays a crucial role in the fields of molecular detection and diagnostics. Conventional trial‐and‐error‐based molecular recognition approaches have always been challenged in distinguishing minimal differences between targets and non‐targets, such as single nucleotide polymorphisms (SNPs) of oligonucleotides. To address these challenges, here, a novel concept of dynamic addressing analysis is proposed. In this concept, by dissecting the regions of the target and creating a corresponding recognizer, it is possible to eliminate the inaccuracy and inefficiency of recognition. To achieve this concept, a Dynamic Addressing Molecular Robot (DAMR), a DNA‐based dynamic addressing device is developed which is capable of dynamically locating targets. DAMR is designed to first bind to the conserved region of the target while addressing the specific region dynamically until accurate recognition is achieved. DAMR has provided an approach for analyzing low‐resolution targets and has been used for analyzing SNP of miR‐196a2 in both cell and serum samples, which has opened new avenues for effective and efficient molecular recognition.

The massive combustion of fossil fuels has resulted in excessive emissions of carbon dioxide (CO2), which has become an urgent environmental problem worldwide. Post-combustion capture technology is one of the most promising carbon capture technologies with rapid growth potential. Ionic materials hold great promise as superior adsorbents for post-combustion capture, offering advantages such as increased adsorption capacity, adjustable adsorption rate, and enhanced operational stability. This review mainly summarized the rational design of pure ionic materials and ion hybrid materials for carbon capture. The relationship between the structural design of materials and the control of capacity, rate, and stability of carbon capture was focused on. A complete description of several emerging ionic materials and their potential for carbon capture is also given. Simultaneously, novel control concepts and rapid screening techniques are introduced. Finally, the article discusses the problems and solutions for the use of ionic materials in future post-combustion capture technology. This work deepens our comprehension of the construction and control of ionic materials, offering valuable insights for the advancement of industrial carbon capture.

We reported a novel method to prepare Al–Si/SiC composites infiltrated under super-gravity field without Mg addition. Noted that centrifugal force acted as the main driving force to overcome the penetration resistance, making it feasible to obtain the Al–Si/SiC composite with high relative density (≥99.3 %). The interfacial reaction between Al and SiC was retarded by Si addition under super-gravity field. Interestingly, the rod-like eutectic Si was detected at Al–SiC interface with semi-continuous distribution, leading to the excellent bending strength (342.6 ± 1.0 MPa), enhanced thermal conductivity (173 W/m K at 373 K), and relatively low coefficient of thermal expansion (8.90 × 10−6 K−1 ranged from 323 to 373 K). The interfacial regulation and fracture mechanism were discussed in details.

Catalysts that can rapidly degrade tetracycline (TC) in water without introducing secondary ion pollution have always been challenging. Herein, a cobalt-based catalyst (CoOx@P-C) is prepared so that CoOx quantum particles (5–10 nm) are uniformly distributed on a linear substrate, and the outer layer is covered with a shell (P-C). The quantum particles of CoOx provide many active sites for the reaction, which ensures the efficient degradation effect of the catalyst, and 30 mg/L TC can be completely degraded in only 5 min. The shell of the quantum particles' outer layer can effectively reduce ions' extravasation. The combination of the shell-like structure and the linear substrate greatly enhances the catalysis's stability and ensures that the catalyst is prepared into a film for practical application. The high catalytic activity of CoOx@P-C is mainly due to the following factors: (1) Uniformly distributed ultra-small nanoparticles can provide many active sites. (2) The microenvironment formed by the core-shell structure enhances not only catalytic stability but also provides the driving force to improve the reaction rate. (3) The composite of CoOx and P-C core-shell structure can accelerate electron transfer and generate many reactive oxygen species in a short time, which makes TC degrade extremely rapidly.

Heat treatments with controlled cooling rates have been used to investigate orthorhombic (O) phase precipitation and its variant selection (VS) in Ti–22Al–25Nb alloy, a latest Ti2AlNb-based intermetallic alloy with excellent comprehensive properties. This study uncovers the misorientation types of different O variants mainly belong to two types with a sharing angle of 90° and distinct rotated axis of or . A notable lamellar structure, colony side plates (CSPs), frequently develop on both sides of adjoining B2 grains, which always have a detrimental effect on the final properties. This structure significantly influenced by VS, variants having (001) planes that are paralleled to the common {110} plane of adjacent B2 grains are selected in colony plates on both sides. If the axis of the two B2 grains coincidentally close as well, the colony plates may appear to penetrate the GB. Moreover, the CSPs structure could form on condition of much slow cooling rate after holding in rather high temperature. These findings could provide insights on how to control processing conditions and tailor the lamellar structure to obtain the desired properties.

This study aimed to assess the impact of dietary selenoprotein extracts from Cardamine hupingshanensis (SePCH) on the growth, hematological parameters, selenium metabolism, immune responses, antioxidant capacities, inflammatory reactions and intestinal barrier functions in juvenile largemouth bass (Micropterus salmoides). The base diet was supplemented with four different concentrations of SePCH: 0.00, 0.30, 0.60 and 1.20 g/Kg (actual selenium contents: 0.37, 0.59, 0.84 and 1.30 mg/kg). These concentrations were used to formulate four isonitrogenous and isoenergetic diets for juvenile largemouth bass during a 60-day culture period. Adequate dietary SePCH (0.60 and 1.20 g/Kg) significantly increased weight gain and daily growth rate compared to the control groups (0.00 g/Kg). Furthermore, 0.60 and 1.20 g/Kg SePCH significantly enhanced amounts of white blood cells, red blood cells, platelets, lymphocytes and monocytes, and levels of hemoglobin, mean corpuscular volume and mean corpuscular hemoglobin in the hemocytes. In addition, 0.60 and 1.20 g/Kg SePCH increased the mRNA expression levels of selenocysteine lyase, selenophosphate synthase 1, 15 kDa selenoprotein, selenoprotein T2, selenoprotein H, selenoprotein P and selenoprotein K in the fish liver and intestine compared to the controls. Adequate SePCH not only significantly elevated the activities of antioxidant enzymes (Total superoxide dismutase, catalase, glutathione reductase, glutathione peroxidase), the levels of total antioxidant capacity and glutathione, while increased mRNA transcription levels of NF-E2-related factor 2, Cu/Zn-superoxide dismutase, catalase, glutathione reductase and glutathione peroxidase. However, adequate SePCH significantly decreased levels of malondialdehyde and H2O2 and the mRNA expression levels of kelch-like ECH-associated protein 1a and kelch-like ECH-associated protein 1b in the fish liver and intestine compared to the controls. Meanwhile, adequate SePCH markedly enhanced the levels of immune factors (alkaline phosphatase, acid phosphatase, lysozyme, complement component 3, complement component 4 and immunoglobulin M) and innate immune-related genes (lysozyme, hepcidin, liver-expressed antimicrobial peptide 2, complement component 3 and complement component 4) in the fish liver and intestine compared to the controls. Adequate SePCH reduced the levels of pro-inflammatory cytokines (tumour necrosis factor-α, interleukin 8, interleukin 1β and interferon γ), while increasing transforming growth factor β1 levels at both transcriptional and protein levels in the liver and intestine. The mRNA expression levels of mitogen-activated protein kinase 13 (MAPK 13), MAPK14 and nuclear factor kappa B p65 were significantly reduced in the liver and intestine of fish fed with 0.60 and 1.20 g/Kg SePCH compared to the controls. Histological sections also demonstrated that 0.60 and 1.20 g/Kg SePCH significantly increased intestinal villus height and villus width compared to the controls. Furthermore, the mRNA expression levels of tight junction proteins (zonula occludens-1, zonula occludens-3, Claudin-1, Claudin-3, Claudin-5, Claudin-11, Claudin-23 and Claudin-34) and Mucin-17 were significantly upregulated in the intestinal epithelial cells of 0.60 and 1.20 g/Kg SePCH groups compared to the controls. In conclusion, these results found that 0.60 and 1.20 g/Kg dietary SePCH can not only improve growth, hematological parameters, selenium metabolism, antioxidant capacities, enhance immune responses and intestinal functions, but also alleviate inflammatory responses. This information can serve as a useful reference for formulating feeds for largemouth bass.

Abstract Indoxyl sulfate is a protein-bound uremic toxin synthesized from indole that cannot be efficiently removed by the hemodialysis method and thus becomes a key risk factor for the progression of chronic kidney disease. Here, we develop a non-dialysis treatment strategy to fabricate an ultramicroporous olefin-linked covalent organic framework with high crystallinity in a green and scalable fashion for selectively removing the indoxyl sulfate precursor (i.e., indole) from the intestine. Various analyses show that the resulting material exhibits excellent gastrointestinal fluid stability, high adsorption efficiency, and good biocompatibility. Notably, it realizes the efficient and selective removal of indole from the intestine and significantly attenuates serum indoxyl sulfate level in vivo. More importantly, the selective removal efficacy of indole is substantially higher than that of the commercial adsorbent AST-120 used in the clinic. The present study opens up a new avenue to eliminate indoxyl sulfate by a non-dialysis strategy and further expands the in vivo applications of covalent organic frameworks.

Glass ceramics (GCs) containing PbS quantum dots (QDs) are prepared for temperature sensing. Broadband emissions are detected in the GCs when PbS QDs are precipitated from the glasses, and emissions centers are modulated from 1250 nm to 1960 nm via heat treatments. The emission centers of GCs exhibit blue-shifts when environment temperatures increase from room temperature to 210 °C. Importantly, the shift values of emission centers increase linearly with the test temperature, which is beneficial for applications in temperature sensing. A temperature sensor based on PbS QDs GC is heat-treated at 500 °C for 10 h, possesses the highest sensitivity of 0.378 nm/°C, and exhibits excellent stability and repeatability at high temperatures (up to 210 °C). Moreover, GC fibers are fabricated by using the GCs as the fiber core. The sensitivity of the temperature-sensing sensor of the GC fibers is also demonstrated and the sensitivity is as high as 0.558 nm/°C. The designed PbS QDs GCs provide a significant materials base for the manufacturing of fluorescent temperature sensors and the GC fibers offer significant opportunities for temperature detection in complex, integrated and compact devices.

This study investigated the effects of glucose (GLU), tapioca starch (TS), gelatinized tapioca starch (GTS), potato starch (PS) and gelatinized potato starch (GPS) on growth and physiological responses in juvenile largemouth bass Micropterus salmoides. After 8 weeks, fish fed with starch diets had better weight gain and growth rates. Counts of red blood cells and monocytes were increased in the PS and GPS groups, compared to GLU group. Contents of serum triglyceride and total cholesterol were markedly elevated in the TS, PS and GPS groups. There were lower levels of serum glucose, insulin and cholecystokinin, and higher agouti-related peptide contents in the PS group compared to GLU group. PS and GPS could enhance glycolysis and TCA cycle by increasing their enzyme activities and transcriptional levels. Additionally, starch sources markedly heightened mRNA levels of key genes involved in the respiratory electron transport chain. Additionally, elevated mRNA levels of key antioxidant genes were shown in the TS and GTS groups. Moreover, TS and PS could promote immunity by upregulating transcriptional levels of the complement system, lysozyme and hepcidin. Taken together, starch exhibited better growth via increasing glycolysis and TCA cycle compared with GLU, and PS could improve antioxidant and immune capacities in largemouth bass.

This study evaluated effects of high starch (20%) on hematological variations, glucose and lipid metabolism, antioxidant ability, inflammatory responses, and histopathological lesions in largemouth bass. Results showed hepatic crude lipid and triacylglycerol (TAG) contents were notably increased in fish fed high starch. High starch could increase counts of neutrophils, lymphocytes, monocytes, eosinophils, and basophils and serum contents of TAG, TBA, BUN, and LEP (p < 0.05). There were increasing trends in levels of GLUT2, glycolysis, gluconeogenesis, and LDH in fish fed high starch through the AKT/PI3K signal pathway. Meanwhile, high starch not only triggered TAG and cholesterol synthesis, but mediated cholesterol accumulation by reducing ABCG5, ABCG8, and NPC1L1. Significant increases in lipid droplets and vacuolization were also shown in hepatocytes of D3–D7 groups fed high starch. In addition, high starch could decrease levels of mitochondrial Trx2, TrxR2, and Prx3, while increasing ROS contents. Moreover, high starch could notably increase amounts of inflammatory factors (IL-1β, TNF-α, etc.) by activating NLRP3 inflammasome key molecules (GSDME, caspase 1, etc.). In conclusion, high starch could not only induce metabolic disorders via gluconeogenesis and accumulation of glycogen, TAG, and cholesterol, but could disturb redox homeostasis and cause inflammatory responses by activating the NLRP3 inflammasome in largemouth bass.

This study investigated the effects of fish oil (FO), soybean oil (SO), rapeseed oil (RO), peanut oil (PO) and lard oil (LO) on growth, immunity and muscle quality in juvenile largemouth bass. After 8 weeks, the results showed that FO and RO could increase weight gain and serum alkaline phosphatase and apelin values compared with LO (p < 0.05). Except lower crude lipid contents, higher amounts of n-3 polyunsaturated fatty acids (15.83% and 14.64%) were present in the dorsal muscle of the FO and RO groups. Meanwhile, FO and RO could heighten mRNA levels of immune defense molecules (lysozyme, hepcidin, and transforming growth factor β1) compared with PO (p < 0.05). While SO could increase potential inflammatory risk via rising counts of white blood cells, platelets, neutrophils and monocytes, and mRNA levels of interleukins (IL-1β, IL-8, IL-12 and IL-15), FO and RO could improve hardness, chewiness and springiness through increasing amounts of hydroxyproline, collagen and lysyl oxidase, and mRNA levels of collagen 1α2 and prolyl hydroxylase in the fish dorsal muscle. Moreover, FO and RO could improve firmness through increasing glycogen and glycogen synthase 1 levels when compared with LO (p < 0.05). Therefore, these results could provide dietary lipid source references during the feeding process of adult largemouth bass.

Background: Osteoarthritis (OA) is the most common degenerative joint disease, mainly affecting the elderly worldwide, for which the drug treatment remains a major challenge. Low-grade inflammation plays a pivotal role in OA onset and progression. Exploration of notable anti-inflammatory and disease-modifying drugs on human samples could facilitate the evaluation of therapeutic strategies for OA. Methods: The anti-inflammatory drug 5-aminosalicylic acid (5-ASA) is a first-line drug for ulcerative colitis (UC), however no study has explored the effects of 5-ASA on articular chondrocytes. In this work, both in vitro (chondrocyte pellets) and ex vivo (osteochondral explants) human inflammatory OA models were applied to evaluate the effects of 5-ASA. Results: In the inflammatory pellet model, 5-ASA remarkably downregulated the gene expression of interleukin-6 (IL-6), and cyclooxygenase-2 (COX-2) while upregulating proteoglycan 4 (PRG4) and cartilage oligomeric matrix protein (COMP) gene expression. Total glycosaminoglycan (GAG) synthesis by pellets was markedly increased in 5-ASA-treated groups compared with the inflammatory group. In conditioned medium, inflammatory mediators (IL-8, nitric oxide) were markedly inhibited upon 5-ASA treatment. Moreover, histological staining showed 5-ASA retained proteoglycan content and inhibited degradation of extracellular matrix (ECM) core components, aggrecan (ACAN) and collagen type II (COL2). In the inflammatory explant model, 5-ASA mitigated signs of OA development by reducing inflammatory mediators and GAG loss. Conclusions: These findings suggest that 5-ASA has anti-inflammatory and pro-anabolic effects on human chondrocyte pellet and osteochondral explant inflammatory OA models. The translational potential of this article: Disease-modifying OA drugs are an unmet clinical need for the treatment of OA. Our study explored and demonstrated the anti-inflammatory and protective effects of 5-ASA on in vitro and ex vivo human inflammatory OA models, showing its translational potential for OA treatment.

Jet impinging device is designed for decay heat removal on horizontal fuel rods in a low temperature heating reactor. An experimental system with a fuel rod simulator is established and experiments are performed to evaluate water film covering capacity, within 0.0287–0.0444 kg/ms mass flow rate, 0–164.1 kW/m2 heating flux and 13.8–91.4°C feeding water temperature. An effective method to obtain the film coverage rate by infrared equipment is proposed. Water film flowing patterns are recoded and the film coverage rates at different circumference angles are measured. It is found the film coverage rate decreases with heating flux during single-phase convection, while increases after onset of nucleate boiling. Besides, film coverage rate is found affected by Marangoni effect and film accelerating effect, and surface wetting is significantly facilitated by bubble behavior. Based on the observed phenomenon and physical mechanism, dry-out depth and initial dry-out rate are proposed to evaluate film covering potential on a heating surface. A model to predict film coverage rate is proposed based on the data. The findings would have reliable guide and important implications for further evaluation and design of decay heat removal system of new reactors, and could be helpful for passive containment cooling research.

Abstract Background The single-cell platform provided revolutionary way to study cellular biology. Technologically, a sophistic protocol of isolating qualified single cells would be key to deliver to single-cell platform, which requires high cell viability, high cell yield and low content of cell aggregates or doublets. For musculoskeletal tissues, like bone, cartilage, nucleus pulposus and tendons, as well as their pathological state, which are tense and dense, it’s full of challenge to efficiently and rapidly prepare qualified single-cell suspension. Conventionally, enzymatic dissociation methods were wildly used but lack of quality control. In the present study, we designed the rapid cycling enzymatic processing method using tissue-specific enzyme cocktail to treat different human pathological musculoskeletal tissues, including degenerated nucleus pulposus (NP), ossifying posterior longitudinal ligament (OPLL) and knee articular cartilage (AC) with osteoarthritis aiming to rapidly and efficiently harvest qualified single-cell suspensions for single-cell RNA-sequencing (scRNA-seq). Results We harvested highly qualified single-cell suspensions from NP and OPLL with sufficient cell numbers and high cell viability using the rapid cycling enzymatic processing method, which significantly increased the cell viability compared with the conventional long-time continuous digestion group (P < 0.05). Bioanalyzer trace showed expected cDNA size distribution of the scRNA-seq library and a clear separation of cellular barcodes from background partitions were verified by the barcode-rank plot after sequencing. T-SNE visualization revealed highly heterogeneous cell subsets in NP and OPLL. Unfortunately, we failed to obtain eligible samples from articular cartilage due to low cell viability and excessive cell aggregates and doublets. Conclusions In conclusion, using the rapid cycling enzymatic processing method, we provided thorough protocols for preparing single-cell suspensions from human musculoskeletal tissues, which was timesaving, efficient and protective to cell viability. The strategy would greatly guarantee the cell heterogeneity, which is critical for scRNA-seq data analysis. The protocol to treat human OA articular cartilage should be further improved.

Agricultural soils undergo periods of saturation followed by desiccation throughout the course of a growing season. It is believed that these periods of wetting and drying influence soil structure and may affect the rate of soil detachment. Thus, an experiment was conducted to investigate the influence of a disturbed soil (soil sieved to simulate tillage) subjected to various wetting and drying cycles, on soil bulk density and the resistance to soil detachment with runoff. Seven treatments consisting of wetting and drying cycles ranging from 0 to 6 cycles were evaluated under laboratory conditions using an experimental flume apparatus. A Richards growth model proposed for predicting the influence of wetting and drying on soil detachment was also evaluated. Results showed that the soil bulk density increased as the number of wetting and drying cycles increased. The soil detachment rate decreased as the number of wetting and drying cycles increased. Moreover, initial soil detachment (occurring as soon as runoff began) rates were high for 1 to 3 wetting and drying cycles, while the rate of initial detachment decreased after the third cycle. For example, soils with two and three wetting and drying cycles took 6.5 and 7 min to reach the maximum 1 cm souring depth, respectively, while the soils subjected to four or more wetting and drying cycles did not reach the maximum 1 cm depth during the 15 min runoff experiment. In addition, the proposed S-Shaped Richards growth model was a good predictor for estimating the soil detachment of soils experiencing various wetting and drying cycles. Findings from this study suggest that more attention should be given to the influence that soil wetting and drying have on the prediction of soil detachment. Information from this study is expected to be useful for improving soil management strategies for reducing soil erosion.

Solid polymer electrolytes have demonstrated high promise to solve the safety problems caused by conventional liquid electrolytes in lithium ion batteries. However, the inherent flammability of most polymer electrolyte materials remains unresolved, hence hindering their further industrial application. Addressing this challenge, we designed and constructed a thermal-responsive imide-linked covalent organic framework (COF) bearing ortho-positioned hydroxy groups as precursors, which can conduct a thermal rearrangement to transform into a highly crystalline and robust benzoxazole-linked COF upon heating. Benefiting from the release of carbon dioxide through thermal rearrangement reaction, this COF platform exhibited excellent flame retardant properties. By contrast, classic COFs (e.g., boronate ester, imine, olefin, imide linked) were all flammable. Moreover, incorporating polyethylene glycol and Li salt into the COF channels can produce solid polymer electrolytes with outstanding flame retardancy, high ionic conductivity (6.42 ​× ​10−4 ​S ​cm−1) and a high lithium-ion transference number of 0.95. This thermal rearrangement strategy not only opens a new route for the fabrication of ultrastable COFs, but also provides promising perspectives to designing flame-retardant materials for energy-related applications.

Abstract Osteoarthritis (OA) is one of the most common musculoskeletal degenerative diseases and contributes to heavy socioeconomic burden. Current pharmacological and conventional non-pharmacological therapies aim at relieving the symptoms like pain and disability rather than modifying the underlying disease. Surgical treatment and ultimately joint replacement arthroplasty are indicated in advanced stages of OA. Since the underlying mechanisms of OA onset and progression have not been fully elucidated yet, the development of novel therapeutics to prevent, halt, or reverse the disease is laborious. Recently, small molecules of herbal origin have been reported to show potent anti-inflammatory, anti-catabolic, and anabolic effects, implying their potential for treatment of OA. Herein, the molecular mechanisms of these small molecules, their effect on physiological or pathological signaling pathways, the advancement of the extraction methods, and their potential clinical translation based on in vitro and in vivo evidence are comprehensively reviewed.

The osteogenic microenvironment of bone-repairing materials plays a key role in accelerating bone regeneration but remains incompletely defined, which significantly limits the application of such bioactive materials. Here, the transcriptional landscapes of different osteogenic microenvironments, including three-dimensional (3D) hydroxyapatite (HA) scaffolds and osteogenic medium (OM), for mesenchymal stromal cells (MSCs) in vitro were mapped at single-cell resolution. Our findings suggested that an osteogenic process reminiscent of endochondral ossification occurred in HA scaffolds through sequential activation of osteogenic-related signaling pathways, along with inflammation and angiogenesis, but inhibition of adipogenesis and fibrosis. Moreover, we revealed the mechanism during OM-mediated osteogenesis involves the ZBTB16 and WNT signaling pathways. Heterogeneity of MSCs was also demonstrated. In vitro ossification of LRRC75A+ MSCs was shown to have better utilization of WNT-related ossification process, and PCDH10+ MSCs with superiority in hydroxyapatite-related osteogenic process. These findings provided further understanding of the cellular activity modulated by OM conditions and HA scaffolds, providing new insights for the improvement of osteogenic biomaterials. This atlas provides a blueprint for research on MSC heterogeneity and the osteogenic microenvironment of HA scaffolds and a database reference for the application of bioactive materials for bone regeneration.

Abstract Green synthesis of crystalline porous materials for energy-related applications is of great significance but very challenging. Here, we create a green strategy to fabricate a highly crystalline olefin-linked pyrazine-based covalent organic framework (COF) with high robustness and porosity under solvent-free conditions. The abundant nitrogen sites, high hydrophilicity, and well-defined one-dimensional nanochannels make the resulting COF an ideal platform to confine and stabilize the H3PO4 network in the pores through hydrogen-bonding interactions. The resulting material exhibits low activation energy (Ea) of 0.06 eV, and ultrahigh proton conductivity across a wide relative humidity (10–90 %) and temperature range (25–80 °C). A realistic proton exchange membrane fuel cell using the olefin-linked COF as the solid electrolyte achieve a maximum power of 135 mW cm−2 and a current density of 676 mA cm−2, which exceeds all reported COF materials.

A fluorosilicate (FS) nano-crystallized glass ceramic (NGC) is one of the most commonly used gain materials for applications in optical devices due to its excellent thermal stability as well as high-efficiency luminescence. However, FS glass can hardly be used to prepare NGC fibers due to its high preparation temperature. Here, a series of low-temperature fluoro-borosilicate (FBS) glasses were designed for the fabrication of active NGC fibers. By modulating B2O3, the preparation temperature of FBS glass was reduced to 1050 °C, and the crystallization in FBS NGCs was more controllable than in FS NGC. The crystallization of the impure phase was inhibited, and single-phase rare earth (RE)-fluoride nanocrystals were controllably precipitated in the FBS NGCs. The 40Si-20B FBS NGC not only exhibited a higher optical transmittance, but the luminescence efficiency was also much higher than traditional FS NGCs. More importantly, NGC fibers were successfully fabricated by using the designed FBS glass as core glass. Nanocrystals were controllably precipitated and greatly enhanced, and upconversion luminescence was observed in NGC fibers. The designed FBS NGCs provided high-quality optical gain materials and offered opportunities for fabricating a wide range of NGC fibers for multiple future applications, including fiber lasers and sensors.

This study evaluated the effects of dietary soybean lecithin (SBL) on the growth, haematological indices, immunities, antioxidant capabilities, and inflammatory and intestinal barrier functions because little information of dietary SBL could be obtained in juvenile largemouth bass (Micropterus salmoides). The fish were fed identical diets except for SBL added at 0, 2, 4 and 8%. It was found that 4 and 8% SBL significantly increased fish weight gain and daily growth rate (p < 0.05), while 4% SBL was optimal for enhancing RBC, HGB, PLT, MCV, MCH, WBC and MON in blood, and ALB and ALP in serum (p < 0.05). SBL (4%) also significantly elevated the antioxidant enzymes activities of T-SOD, CAT, GR, GPx, GST and T-AOC and GSH contents; increased mRNA transcription levels of Nrf2, Cu/Zn-SOD, CAT, GR, GST3 and GPx3; and decreased MDA contents. Keap1a and Keap1b levels were markedly down-regulated (p < 0.05). SBL (4%) significantly enhanced levels of the immune factors (ACP, LZM and C3) and the mRNA expression levels of innate immune-related genes (C3, C4, CFD, HEPC and MHC-I) compared with the control groups (0%) (p < 0.05). SBL (4%) significantly increased IgM and T-NOS in the intestine (p < 0.05) and significantly decreased levels of TNF-α, IL-8, IL-1β and IFN-γ and increased TGF-β1 at both transcription and protein levels in the liver and intestine (p < 0.05). The mRNA expression levels of MAPK13, MAPK14 and NF-κB P65 were significantly decreased in the intestine in the 4% SBL groups (p < 0.05). Histological sections also demonstrated that 4% SBL protected intestinal morphological structures compared with controls. This included increased intestinal villus height and muscular thickness (p < 0.05). Furthermore, the mRNA expression levels of the intestinal epithelial cell tight junction proteins (TJs) (ZO-1, claudin-3, claudin-4, claudin-5, claudin-23 and claudin-34) and mucin-5AC were significantly up-regulated in the 4% SBL groups compared with the controls (p < 0.05). In conclusion, these results suggested that 4% dietary SBL could not only improve growth, haematological indices, antioxidant capabilities, immune responses and intestinal functions, but also alleviate inflammatory responses, thereby providing reference information for the feed formulations in cultured largemouth bass.

Aspartate family includes five additional amino acids other than aspartate, among which most except aspartate have been reported for their action in pathogenesis by amino acid biosynthesis. However, how aspartate, the initial substrate of this family metabolic pathway, is involved in pathogenesis remains unknown. Here, we focused on aspartate transaminase (AST) that catalyzes transamination reaction between glutamate-aspartate in Magnaporthe oryzae. Three MoAST genes were bioinformatically analyzed, of which MoAST2 was uniquely upregulated when invasive hyphae switched to necrotrophic pathogenesis. MoAST2 deletion (ΔMoast2) caused a drastic reduction in conidiogenesis and appressorium formation. Particularly, ΔMoast2 was observed to be proliferated at the biotrophic phase but inhibited at the necrotrophic stage, and with invisible symptoms detected, suggesting a critical role in necrotrophic phase. Glutamate family restored the ΔMoast2 defects but aspartate family did not, inferring that transamination occurs from aspartate to glutamine. MoAST2 is cytosolic and possessed H2O2 stress tolerance. In parallel, Colletotrichum graminicola AST2, CgAST2 was proven to be a player in necrotrophic anthracnose development. Therefore, conserved AST2 is qualified to be a drug target for disease control.

Abstract Background There is no comparative study with long-term follow-up between posterior-only and combined posterior-anterior approaches in treating lumbar spinal tuberculosis (LSTB) and lumbosacral spinal tuberculosis (LSSTB). This retrospective study aimed to compare and evaluate the long-term outcomes of these two surgical approaches in LSTB and LSSTB. Methods Thirty patients with LSTB and 12 patients with LSSTB underwent posterior-only approach (group A); 26 patients with LSTB and 14 patients with LSSTB were managed with combined posterior-anterior approaches (group B). Analysis and comparison in clinical and radiographic outcomes between the two groups were performed. Results The intra-operative bleeding amount, surgery time, and hospitalization days in group A were less than that in group B (P < 0.05). These patients were followed for a minimum of 7 years. All patients achieved completely healing within 2-year follow-up. Bony fusion was obtained in all patients. The visual analog scale, Japanese Orthopedic Association score, Oswestry Disability index, and Kirkaldy-Willis functional evaluation at the final visit demonstrated that all patients in both groups achieved satisfactory results. There was no significant difference in angle correction or maintaining correction between combined posterior-anterior approaches and posterior-only approach (P > 0.05). Complications occurred in both groups. Conclusions Both combined posterior-anterior approaches and posterior-only approach can achieve satisfactory clinical and radiological outcomes of long-term in treating LSTB and LSSTB, while posterior-only approach can safely and effectively achieve lesion debridement, decompression, and stability reconstruction and maintenance with the advantages of less invasive surgery, less bleeding, shorter surgery time, and hospital stay, and fewer complications. So, posterior-only approach seemed to be superior to the combined posterior-anterior one.

The influence of boiling time (10 –120 min) on nutritional and antioxidant properties of Lentinus edodes pieces and broth was investigated. The results showed that the primary nutrients contents and antioxidant activities of L. edodes pieces decreased with the prolongation of boiling time, while ergosterol content increased. The nutrients contents in L. edodes broth presented an uptrend, and the antioxidant activities increased rapidly in the first 30 min, and then kept stable or decline, which was positively correlated with the change of total phenolic content. Microstructure observation showed that boiling treatment destroyed the cell wall of L. edodes, leading to the dissolution and loss of cell wall components and cytoplasmic contents. From the point of nutrition, the optimal boiling time for L. edodes pieces is 30 min and for broth preparation is 60 min. This study provides a useful reference for rational cooking or processing L. edodes and other mushrooms.

Dihydroxynapthalene-(DHN) and L-dihydroxyphenylalanine (L-DOPA) are two types of dominant melanin in fungi. Fungal melanins with versatile functions are frequently associated with pathogenicity and stress tolerance. In rice blast fungus, Magnaporthe oryzae, DHN melanin is essential to maintain the integrity of the infectious structure, appressoria; but the role of the tyrosinase-derived L-DOPA melanin is still unknown. Here, we have genetically and biologically characterized a tyrosinase gene (MoTyr) in M. oryzae. MoTyr encodes a protein of 719 amino acids that contains the typical CuA and CuB domains of tyrosinase. The deletion mutant of MoTyr (ΔMoTyr) was obtained by using a homologous recombination approach. Phenotypic analysis showed that conidiophore stalks and conidia formation was significantly reduced in ΔMoTyr. Under different concentrations of glycerol and PEG, more appressoria collapsed in the mutant strains than in the wild type, suggesting MoTyr is associated with the integrity of the appressorium wall. Melanin measurement confirmed that MoTyr loss resulted in a significant decrease in melanin synthesis. Accordingly, the loss of MoTyr stunted the conidia germination under stress conditions. Importantly, the MoTyr deletion affected both infection and pathogenesis stages. These results suggest that MoTyr, like DHN pigment synthase, plays a key role in conidiophore stalks formation, appressorium integrity, and pathogenesis of M. oryzae, revealing a potential drug target for blast disease control.

Osteoarthritis (OA) is the most common degenerative joint disease without clear pathophysiological mechanism and effective drugs for treatment. Although various animal models exist, the translation of the outcome into clinics remains difficult due to species differences. In this study, an ex vivo inflammatory OA model was induced using different concentrations of interleukin one beta (IL-1β) and tumor necrosis factor α (TNF-α) on explants from the human femoral head. In the inflammatory OA groups, the gene expression levels of cartilage catabolism (matrix metalloproteinase 1 (MMP1), matrix metalloproteinase 3 (MMP3)), and inflammation (interleukin 6 (IL-6), interleukin 8 (IL-8)) markers were significantly upregulated, while the anabolic genes (collagen 2 (COL2), aggrecan (ACAN), and proteoglycan 4 (PRG4)) were downregulated compared to the control group. The release of cytokines (IL-6, IL-8) and nitric oxide (NO) in the conditioned medium was also upregulated in inflammatory OA groups. The Safranin O/Fast Green staining showed loss of proteoglycan in the superficial zone cartilage after cytokine treatment. The results indicated that an ex vivo inflammation and degeneration model was successfully established using osteochondral explants from the human femoral head. This model can be used to elucidate the in-depth mechanism of inflammatory OA and to screen new drugs for OA treatment.

Human PTEN, a dual-phosphatase tumor suppressor, is frequently dysregulated by alternative splicing. Fungi harbor PTEN homologs, but alternative splicing of fungal PTENs has not been reported as far as we know. Here, we described an alternative splicing case in the PTEN homolog of Magnaporthe oryzae (MoPTEN). Two splice variants of MoPTEN were detected and identified, which are resulted from an intron retention and exclusion (MoPTEN-1/2). Both proteins were different in lipid and protein phosphatase activity and in expression patterns. The MoPTEN deletion mutant (ΔMoPTEN) showed the defects in conidiation, appressorium formation, and pathogenesis. ΔMoPTEN could be completely restored by MoPTEN, but rescued partially by MoPTEN-1 in the defect of conidium and appressorium formation, and by MoPTEN-2 in the defect of invasive development. Assays to assess sensitivity to oxidative stress reveal the involvement of MoPTEN-2 in scavenging exogenous and host-derived H2O2. Taken together, MoPTEN undergoes alternative splicing, and both variants cooperatively contribute to conidium and appressorium development, and invasive hyphae growth in plant cells, revealing a novel disease development pathway in M. oryzae.

Abstract Background The altered concentrations of amino acids were found in the bone marrow or blood of leukemia patients. Metabolomics technology combining mathematical model of biomarkers could be used for assisting the diagnosis of pediatric acute leukemia (AL). Methods The concentrations of 17 amino acids was measured by targeted liquid chromatograph–tandem mass spectrometry in periphery blood collected using dried blood spots. After evaluation, the mathematical models were further evaluated by prospective clinical validation cohort for AL diagnosis. Results The concentrations of 13 in 17 amino acids were statistically different between the periphery blood dried serum dots measured by targeted LC–MS/MS. The receiver operating characteristic analysis for the models of amino acid panel showed that the area under curve for AL diagnosis were 0.848, 0.834 and 0.856 by SVM, RF and XGBoost. The Kappa values in further prospectively evaluated clinical cohort were 0.697, 0.703 and 0.789 (p > 0.05) respectively, and the accuracies for the models were 84.86%, 85.20% and 89.46% respectively with further clinical validation. Conclusions The established mathematical model is a faster, cheaper and more convenient way than conventional methods, and no significant difference on the effect of diagnosis comparing with conventional methods. The mathematical model can be clinically useful for assisting pediatric AL diagnosis.

The room temperature tensile deformation of Ti-22Al-25Nb alloy with an equiaxed α2 phase microstructure and the activated slip system of α2 particles were investigated by a combination of in situ tensile tests and electron backscatter diffraction experiments. The results demonstrate that only a few wide and long slip bands occur in the B2 matrix in the initial stage of yielding. With the tensile displacement increases, a large number of slip bands, including multiple- and cross-slip bands, appear in the B2 matrix and the distance between two adjacent slip bands decreases significantly. Meanwhile, the movement of the slip bands is hindered by the α2 particles and the B2 grain boundaries, and the slip bands appear only in a small number of the α2 particles. From the beginning of the tensile process to the final fracture, there are lots of α2 particles without slip bands. The slip bands penetrate the needle-like lamellar O phase without changing the slip direction. Compared with the α2 particles, the hindering effect of needle-like O phases on the motion of the slip bands is quite small. The microcracks nucleated at the α2/B2 phase boundaries or within the α2 particles, and microcracks propagated along the α2/B2 phase boundaries or across the α2 particles. The fracture surface shows the quasi-cleavage feature, which contains a large number of small and shallow dimples on planar facets. The analysis indicates that the plastic deformation of the alloy is mainly contributed by the B2 phase. For room temperature tensile deformation of α2 phase, there are three types of slip systems that can be activated, including the prism type slip, the basal type slip and the pyramidal type slip. The prism type slip is most likely to be activated, followed by the basal type slip and finally the pyramidal type slip. In addition, the critical resolved shear stress (CRSS) for the pyramidal type slip is the highest among the three types of slip systems. Therefore, the deformation in the α2 phase is mainly contributed by the prism type slip and the basal type slip.

Primary inoculum that survives overwintering is one of the key factors that determine the outbreak of plant disease. Pathogenic resting structures, such as chlamydospores, are an ideal inoculum for plant disease. Puzzlingly, Magnaporthe oryzae, a devastating fungal pathogen responsible for blast disease in rice, hardly form any morphologically changed resting structures, and we hypothesize that M. oryzae mainly relies on its physiological alteration to survive overwintering or other harsh environments. However, little progress on research into regulatory genes that facilitate the overwintering of rice blast pathogens has been made so far. Serine threonine protein kinase AGC/AKT, MoSch9, plays an important role in the spore-mediated pathogenesis of M. oryzae. Building on this finding, we discovered that in genetic and biological terms, MoSch9 plays a critical role in conidiophore stalk formation, hyphal-mediated pathogenesis, cold stress tolerance, and overwintering survival of M. oryzae. We discovered that the formation of conidiophore stalks and disease propagation using spores was severely compromised in the mutant strains, whereas hyphal-mediated pathogenesis and the root infection capability of M. oryzae were completely eradicated due to MoSch9 deleted mutants’ inability to form an appressorium-like structure. Most importantly, the functional and transcriptomic study of wild-type and MoSch9 mutant strains showed that MoSch9 plays a regulatory role in cold stress tolerance of M. oryzae through the transcription regulation of secondary metabolite synthesis, ATP hydrolyzing, and cell wall integrity proteins during osmotic stress and cold temperatures. From these results, we conclude that MoSch9 is essential for fungal infection-related morphogenesis and overwintering of M. oryzae.

The phosphorylation status of proteins, which is determined by protein tyrosine kinases (PTKs) and protein tyrosine phosphatases (PTPs), governs many cellular actions. In fungal pathogens, phosphorylation-mediated signal transduction has been considered to be one of the most important mechanisms in pathogenicity. Colletotrichum graminicola is an economically important corn pathogen. However, whether phosphorylation is involved in its pathogenicity is unknown. A mitochondrial protein tyrosine phosphatase gene, designated CgPTPM1, was deduced in C. graminicola through the use of bioinformatics and confirmed by enzyme activity assays and observation of its subcellular localization. We then created a CgPTPM1 deletion mutant (ΔCgPTPM1) to analyze its biological function. The results indicated that the loss of CgPTPM1 dramatically affected the formation of conidia and the development and differentiation into appressoria. However, the colony growth and conidial morphology of the ΔCgPTPM1 strains were unaffected. Importantly, the ΔCgPTPM1 mutant strains exhibited an obvious reduction of virulence, and the delayed infected hyphae failed to expand in the host cells. In comparison with the wild-type, ΔCgPTPM1 accumulated a larger amount of H2O2 and was sensitive to exogenous H2O2. Interestingly, the host cells infected by the mutant also exhibited an increased accumulation of H2O2 around the infection sites. Since the expression of the CgHYR1, CgGST1, CgGLR1, CgGSH1 and CgPAP1 genes was upregulated with the H2O2 treatment, our results suggest that the mitochondrial protein tyrosine phosphatase PTPM1 plays an essential role in promoting the pathogenicity of C. graminicola by regulating the excessive in vivo and in vitro production of H2O2.

To control the dimension of the supramolecular system was of great significance. We construct a two component self-assembly system, in which the gelator LHC18 and achiral azobenzene carboxylic acid could co-assembly and form gels. By modulating the stoichiometric ratio of the two components, not only the morphology could be transformed from 1D nanaotube to 0D nanospheres but also the supramolecualr chirality could be tuned. This work could provide some insights to the control of dimension and the supramolecular chirality in the two-component systems by simply modulating the stoichiometric ratio.

Bicarbonate (HCO3−) transporter family including the anion exchanger (AE) group is involved in multiple physiological processes through regulating acid-base homeostasis. HCO3− transporters have been extensively studied in mammals, but fungal homologues of AE are poorly understood. Here, we characterized the AE group member (MoAE4) in Magnaporthe oryzae. MoAE4 exhibits more sequence and structure homologies with the reported AE4 and BOR1 proteins. In addition to the common sublocalization on cytomembrane, MoAE4 also localizes on tonoplast. Yeast complementation verified that MoAE4 rescues boron sensitivity and endows NaHCO3 tolerance in the BOR1 deleted yeast. MoAE4 gene is bicarbonate induced in M. oryzae; and loss of MoAE4 (ΔMoAE4) resulted in mycelial growth inhibited by NaHCO3. Lucigenin fluorescence quenching assay confirmed that ΔMoAE4 accumulated less HCO3− in vacuole and more HCO3− in cytosol, revealing a real role of MoAE4 in bicarbonate transport. ΔMoAE4 was defective in conidiation, appressorium formation, and pathogenicity. More H2O2 was detected to be accumulated in ΔMoAE4 mycelia and infected rice cells. Summarily, our data delineate a cytomembrane and tonoplast located HCO3− transporter, which is required for development and pathogenicity in M. oryzae, and revealing a potential drug target for blast disease control.

In the South Yellow Sea Basin, Mesozoic–Paleozoic marine strata are generally well developed with large thickness, and they are characterized by multi-source and multi-stage hydrocarbon accumulation, providing a material basis for the formation of large-scale oil and gas fields. However, no substantial breakthrough has been made in this area. Based on previous research results, the complex tectonic pattern of this superimposed basin was formed by multi-stage tectonic movements and the favorable static conditions for hydrocarbon preservation were reworked or destroyed by later superimposition. Therefore, hydrocarbon preservation conditions are the key factors for restricting the breakthrough of marine oil and gas exploration in this area. In this paper, hydrocarbon preservation conditions of marine strata in the South Yellow Sea Basin were comprehensively analyzed from many aspects, such as tectonic movement, source conditions, caprock characteristics, magmatic activities, and hydrogeological and hydrogeochemical characteristics. It is indicated that the complex tectonic pattern of the South Yellow Sea Basin is resulted from tectonic events in multiple stages, and the development and evolution of regional source rocks are mainly controlled by two stages (i.e., the stable evolution stage of Mesozoic–Paleozoic marine basin and the Mesozoic–Cenozoic tectonic pattern transformation and basin formation stage), so the characteristics of differential oil and gas preservation are presented. Besides, better marine hydrocarbon preservation preconditions in this area are weaker tectonic reworking, development of high-quality thick source rocks, good vertical sealing capacity of caprocks, weaker magmatic activity and confined hydrogeological conditions. It is concluded that the Laoshan Uplift in the central part of the South Yellow Sea Basin is structurally stable with weaker faulting and magmatic activities, so it is better in oil and gas preservation conditions. Besides, several large-scale structural traps with good petroleum geological conditions and complete source–reservoir–caprock assemblages are developed in this area. Therefore, this area is the most promising region for Paleozoic marine oil and gas exploration in this basin.

With the deterioration of the environment and the depletion of fossil fuel energy, renewable energy has attracted worldwide attention because of its continuous availability from nature. Despite this continuous availability, the uncertainty of intermittent power is a problem for grid dispatching. This paper reports on a study of the scheduling and optimization of microgrid systems for photovoltaic (PV) power and electric vehicles (EVs). We propose a mathematical model to address the uncertainty of PV output and EV charging behavior, and model scheduling optimization that minimizes the economic and environmental cost of a microgrid system. A semi-infinite dual optimization model is then used to deal with the uncertain variables, which can be solved with a robust optimization algorithm. A numerical case study shows that the security and stability of the solution obtained by robust optimization outperformed that of stochastic optimization.

BACKGROUND:Sugarcane mosaic virus (SCMV) is responsible for large-scale economic losses in the global production of sugarcane, maize, sorghum, and some other graminaceous species. To understand the evolutionary mechanism of SCMV populations, this virus was studied in Shanxi, China. A total of 86 maize leaf samples (41 samples in 2012 and 45 samples in 2013) were collected from 4 regions of Shanxi. RESULTS:Double-antibody sandwich (DAS)-ELISA and RT-PCR showed 59 samples (30 samples in 2012 and 29 samples in 2013) to be positive for SCMV, from which 10 new isolates of SCMV were isolated and sequenced. The complete genomes of these isolates are 9610 nt long, including the 5' and 3' non-coding regions, and encode a 3063-amino acid polyprotein. Phylogenetic analyses revealed that 24 SCMV isolates could be divided on the basis of the whole genome into 2 divergent evolutionary groups, which were associated with the host species. Among the populations, 15 potential recombination events were identified. The selection pressure on the genes of these SCMV isolates was also calculated. The results confirmed that all the genes were under negative selection. CONCLUSIONS:Negative selection and recombination appear to be important evolutionary factors shaping the genetic structure of these SCMV isolates. SCMV is distributed widely in China and exists as numerous strains with distinct genetic diversity. Our findings will provide a foundation for evaluating the epidemiological characteristics of SCMV in China and will be useful in designing long-term, sustainable management strategies for SCMV.

BACKGROUND:Hepatitis B vaccination is the most important tool available for preventing hepatitis B virus (HBV) infection and reducing the prevalence of infection. However, epidemiological studies have demonstrated that morethan 5% of patients exhibit a non- or hypo-response to the HBV vaccine. Genetic variations associated with T cell immunity contribute to the immune response to HBV vaccination. The deltex 1 (DTX1) gene is involved in T cell anergy, which may also be associated with the immune response to the HBV vaccination. METHODS:We detected 10 single nucleotide polymorphisms (SNPs) in or around the DTX1 gene in 601 infants out of a population from Southwest China, including 299 high responders(HRs; HBsAb > 100 mIU/mL) and 302 non-responders (NRs; HBsAb < 10 mIU/mL). An additional validation study was performed, comprising 230 adult patients(135 HRs and 95 NRs) from Southwest China. RESULTS:This study found that the minor allele 'G' of rs2384077 (adjusted p = 2.63E(-04)), and the minor allele 'C' of rs10744794 (adjusted p = 3.69E(-04)) in the first intron of the DTX1 gene were remarkably associated with the immune response to HBV vaccination in both infant and adult populations. Moreover, a subsequent analysis indicated that haplotypes (A-T, G-C) of the two SNPs were significantly associated with the immune response to HBV vaccination. CONCLUSIONS:Two SNPs (rs2384077 and rs10744794) in an intron of DTX1 and the linkage disequilibrium (LD) block are significantly associated with the immune response to HBV vaccination. The functional element annotation of the LD block between the two SNPs contains four transcriptional regulatory elements. The results suggest that these two SNPs may be involved in the immune response to HBV vaccination.

RNA interference (RNAi) denotes sequence-specific mRNA degradation induced by short interfering double-stranded RNA (siRNA) and has become a revolutionary tool for functional annotation of mammalian genes, as well as for development of novel therapeutics. The practical applications of RNAi are usually achieved by expressing short hairpin RNAs (shRNAs) or siRNAs in cells. However, a major technical challenge is to simultaneously express multiple siRNAs to silence one or more genes. We previously developed pSOS system, in which siRNA duplexes are made from oligo templates driven by opposing U6 and H1 promoters. While effective, it is not equipped to express multiple siRNAs in a single vector. Gibson DNA Assembly (GDA) is an in vitro recombination system that has the capacity to assemble multiple overlapping DNA molecules in a single isothermal step. Here, we developed a GDA-based pSOK assembly system for constructing single vectors that express multiple siRNA sites. The assembly fragments were generated by PCR amplifications from the U6-H1 template vector pB2B. GDA assembly specificity was conferred by the overlapping unique siRNA sequences of insert fragments. To prove the technical feasibility, we constructed pSOK vectors that contain four siRNA sites and three siRNA sites targeting human and mouse β-catenin, respectively. The assembly reactions were efficient, and candidate clones were readily identified by PCR screening. Multiple β-catenin siRNAs effectively silenced endogenous β-catenin expression, inhibited Wnt3A-induced β-catenin/Tcf4 reporter activity and expression of Wnt/β-catenin downstream genes. Silencing β-catenin in mesenchymal stem cells inhibited Wnt3A-induced early osteogenic differentiation and significantly diminished synergistic osteogenic activity between BMP9 and Wnt3A in vitro and in vivo. These findings demonstrate that the GDA-based pSOK system has been proven simplistic, effective and versatile for simultaneous expression of multiple siRNAs. Thus, the reported pSOK system should be a valuable tool for gene function studies and development of novel therapeutics.
