Intestinal protozoan parasitic infections are considered one of the most frequent types of infection caused by these parasites and remain a major health problem for communities. This study aims to detect the frequency of intestinal protozoan infections infection among Zakho general population from October 2018 to June 2022. This cross-sectional study was performed on 2,118 patients referring to private medical diagnostic laboratories in Zakho, Kurdistan Region, Iraq. Samples of fecal matter were collected and subjected to analysis using two different techniques: direct observation under the microscope (wet mount) and formalin-ether concentration methods. Morphological characteristics of trophozoites and cysts were used to identify E. histolytica using microscopical examination. Out of 2,118 recruited samples, 1,155 (54.53%) were male, and 963 (45.47%) were female. The mean age of participants was 20.41 (±19.12), with ages ranging from 1 to 63 years. The overall prevalence of protozoan infections was 395/2118 (18.65%). Out of these, Entamoeba histolytica was the predominant pathogenic protozoa infection 271 (68.61%), followed by Giardia lamblia 100 (25.31%). A significant association was seen between Entamoeba histolytica and age groups (P=0.003) and gender (P=0.004). The highest infection rate was reported among the age group <15 years (55.72%). We also found a higher rate of Giardia infection among age group < 15 years (46%) with significant differences (P=0.002) and a higher rate of Giardia seen in females (55%) with significant association (P=0.014). The frequency of Giardia lamblia and E. histolytica infections in our study was higher than other studies reported in Iraq and other countries, and these infections continue to pose a difficult public health issue and necessitate the implementation of stronger and more effective preventative measures. Viruses are the most abundant biological entities on Earth and play a significant role in the evolution of many organisms and ecosystems. In pathogenic protozoa, the presence of viruses has been linked to an increased risk of treatment failure and severe clinical outcome. Here, we studied the molecular epidemiology of the zoonotic disease cutaneous leishmaniasis in Peru and Bolivia through a joint evolutionary analysis of Leishmania braziliensis and their dsRNA Leishmania virus 1. We show that parasite populations circulate in tropical rainforests and are associated with single viral lineages that appear in low prevalence. In contrast, groups of hybrid parasites are geographically and ecologically more dispersed and associated with an increased prevalence, diversity and spread of viruses. Our results suggest that parasite gene flow and hybridization increased the frequency of parasite-virus symbioses, a process that may change the epidemiology of leishmaniasis in the region. One of the remarkable features of eukaryotes is the nucleus, delimited by the nuclear envelope (NE), a complex structure and home to the nuclear lamina and nuclear pore complex (NPC). For decades, these structures were believed to be mainly architectural elements and, in the case of the NPC, simply facilitating nucleocytoplasmic trafficking. More recently, the critical roles of the lamina, NPC and other NE constituents in genome organisation, maintaining chromosomal domains and regulating gene expression have been recognised. Importantly, mutations in genes encoding lamina and NPC components lead to pathogenesis in humans, while pathogenic protozoa disrupt the progression of normal development and expression of pathogenesis‐related genes. Here, we review features of the lamina and NPC across eukaryotes and discuss how these elements are structured in trypanosomes, protozoa of high medical and veterinary importance, highlighting lineage‐specific and conserved aspects of nuclear organisation. Granular media filtration remains a critical treatment process and regulatory requirement for managing pathogenic protozoa in drinking water. It is a dynamic process in which performance inherently varies. While research has focused on characterizing or maximizing (oo)cyst removal in individual filters, the risk implications of combinations of filters moving through different phases of the filter cycle (leading to temporal variation in plant‐scale performance) have not been described. Increasing threats from climate‐change‐exacerbated landscape disturbances leading to more variable source water quality emphasize the need for such evaluations. Here, a modeling framework was developed to investigate the impacts of individual filter performance variation on plant‐scale performance. It is shown that improving maximal removal during stable operation does not necessarily improve average performance. The effect of other design and operational strategies like increasing the number of filters or implementing proactive operations (e.g., avoiding breakthrough) are analyzed, thereby providing guidance for increasing treatment resilience. Motility of pathogenic protozoa depends on flagella (synonymous with cilia) with axonemes containing nine doublet microtubules (DMTs) and two singlet microtubules. Microtubule inner proteins (MIPs) within DMTs influence axoneme stability and motility and provide lineage-specific adaptations, but individual MIP functions and assembly mechanisms are mostly unknown. Here, we show in the sleeping sickness parasite Trypanosoma brucei, that FAP106, a conserved MIP at the DMT inner junction, is required for trypanosome motility and functions as a critical interaction hub, directing assembly of several conserved and lineage-specific MIPs. We use comparative cryogenic electron tomography (cryoET) and quantitative proteomics to identify MIP candidates. Using RNAi knockdown together with fitting of AlphaFold models into cryoET maps, we demonstrate that one of these candidates, MC8, is a trypanosome-specific MIP required for parasite motility. Our work advances understanding of MIP assembly mechanisms and identifies lineage-specific motility proteins that are attractive targets to consider for therapeutic intervention. Objectives: We aimed to assess the performance of the Novodiag® Stool Parasites (NSP) assay in the diagnosis of the most common intestinal protozoan and microsporidia infections. Methods: A panel of 167 selected stool samples was retrospectively analysed with the NSP assay and compared to routine microscopy and qPCR methods for the detection of pathogenic protozoa and microsporidia. Results: Whereas specificity was high for all protozoa and microsporidia, NSP sensitivity was strongly dependent on the comparative method used as reference. When compared to microscopic methods, NSP sensitivity was high (96.7 to 100%) for Blastocystis hominis, Entamoeba histolytica and Cyclospora cayetanensis but was lower for Giardia intestinalis (85.2%) and ≤50% for Cystoisospora belli and Dientamoeba fragilis. In comparison to conventional qPCR, the NSP assay demonstrated lower sensitivity characteristics dependent on parasite loads, reaching 60 to 70% for G. intestinalis, D. fragilis, Cryptosporidium spp. and E. histolytica. Sensitivity was 100% for Enterocytozoon bieneusi, but none of the five samples containing Encephalitozoon spp. were detected. Conclusions: The overall performance of the NSP assay in the diagnosis of gastrointestinal protozoa and microsporidia seems to be better than or equivalent to that observed with microscopic methods but inferior to that obtainable with classical targeted qPCR. It has been 49 years since the last discovery of a new virus family in the model yeast Saccharomyces cerevisiae. A large-scale screen to determine the diversity of double-stranded RNA (dsRNA) viruses in S. cerevisiae has identified multiple novel viruses from the family Partitiviridae that have been previously shown to infect plants, fungi, protozoans, and insects. Most S. cerevisiae partitiviruses (ScPVs) are associated with strains of yeasts isolated from coffee and cacao beans. The presence of partitiviruses was confirmed by sequencing the viral dsRNAs and purifying and visualizing isometric, non-enveloped viral particles. ScPVs have a typical bipartite genome encoding an RNA-dependent RNA polymerase (RdRP) and a coat protein (CP). Phylogenetic analysis of ScPVs identified three species of ScPV, which are most closely related to viruses of the genus Cryspovirus from the mammalian pathogenic protozoan Cryptosporidium parvum. Molecular modeling of the ScPV RdRP revealed a conserved tertiary structure and catalytic site organization when compared to the RdRPs of the Picornaviridae. The ScPV CP is the smallest so far identified in the Partitiviridae and has structural homology with the CP of other partitiviruses but likely lacks a protrusion domain that is a conspicuous feature of other partitivirus particles. ScPVs were stably maintained during laboratory growth and were successfully transferred to haploid progeny after sporulation, which provides future opportunities to study partitivirus-host interactions using the powerful genetic tools available for the model organism S. cerevisiae. Author summary: This article describes the discovery and characterization of multiple strains and species of viruses from the family Partitiviridae in the brewer's and baker's yeast S. cerevisiae. These novel viruses have bipartite genomes packaged in spherical viral particles with structural homology to members of the family Partitiviridae. Strikingly, yeast partitiviruses are most closely related to viruses from human pathogenic protozoa and not partitiviruses of other fungi. As partitiviruses can positively and negatively contribute to a host's physiology (including important human and plant pathogens), the presence of partitiviruses in S. cerevisiae offers a unique opportunity to study the biology of these viruses in a well-developed model system. The consumption of fresh vegetables is related to healthy lifestyle habits present in culinary preparations in different regions. The presence of pathogenic parasites in these foods can cause gastrointestinal disorders. Thus, the objective of the present study was to carry out a narrative review of the literature on the prevalence of helminths in fresh vegetable samples. The analysis of the studies published from 2016 to 2022 showed that hookworms and Ascaris lumbricoides are the most common pathogenic helminths in fresh vegetable samples, with a prevalence of up to 73.8% and 55.1%, respectively. In addition, studies have shown associations between the presence of helminths and pathogenic protozoa. The results obtained in this review indicate the urgent need to implement actions at all stages of the vegetable production chain, from the water used in planting irrigation to cleaning before sale to the final consumer. © 2022 Society of Chemical Industry. The detection of Giardia duodenalis and Cryptosporidium spp. was performed, along with the identification of the ciliated protozoa biodiversity, to evaluate the correlation between these protozoa in freshwater quality monitoring. Water and sediment samples from two sites in the Atibaia River (Campinas, São Paulo, Brazil) were collected monthly for 2 years (n = 96). Pathogenic protozoa in water and sediment were detected by using immunomagnetic separation, followed by visualization by immunofluorescence assay (IFA). All positive aliquots in IFA were subjected to DNA extraction and subsequently nested PCR. Qualitative (in vivo observation and silver impregnation) and quantitative (in vivo enumeration) analyses were performed for the ciliated protozoa. Giardia cysts were detected in 62.5% of the surface water samples and Cryptosporidium spp. in 25.0%. In the sediment, cysts were detected in 35.4% samples and oocysts in 16.6%. A total of 57 samples positive for Giardia cysts were subjected to sequencing, 40 of which were harboring G. duodenalis (24 were characterized as sub-assemblage AII). For ciliated protozoa, 73 taxa belonging to 53 genera were identified over the period of the study. These results revealed a high degree of contamination by waterborne protozoa in the main water source which supplies drinking water for more than one million people in Campinas (São Paulo), highlighting the need for continuous monitoring of this catchment site. In addition, the present study provides important data regarding the sources of the water body degradation, i.e., fecal contamination of human origin, in addition to the survey of the ciliated protozoa. Transovarial transmission (TOT) is an efficient vertical transmission of pathogens that is observed in many arthropod vectors. This method seems to be an evolutionarily unique development observed only in Babesia sensu stricto (clade VI) and Rickettsia spp., whereas transstadial transmission is the common/default way of transmission. Transovarial transmission does not necessarily contribute to the amplification of tick-borne pathogens but does contribute to the maintenance of disease in the environment. To address the continued rise of multi-drug-resistant microorganisms, the development of novel drugs with new modes of action is urgently required. While humans biosynthesize the essential isoprenoid precursors isopentenyl diphosphate (IPP) and dimethylallyl diphosphate (DMAPP) via the established mevalonate pathway, pathogenic protozoa and certain pathogenic eubacteria use the less well-known methylerythritol phosphate pathway for this purpose. Important pathogens using the MEP pathway are, for example, Plasmodium falciparum, Mycobacterium tuberculosis, Pseudomonas aeruginosa and Escherichia coli. The enzymes of that pathway are targets for antiinfective drugs that are exempt from target-related toxicity. 2C-Methyl-D-erythritol 4-phosphate (MEP), the second enzyme of the non-mevalonate pathway, has been established as the molecular target of fosmidomycin, an antibiotic that has so far failed to be approved as an anti-infective drug. This review describes the development and anti-infective properties of a wide range of fosmidomycin derivatives synthesized over the last four decades. Here we discuss the DXR inhibitor pharmacophore, which comprises a metal-binding group, a phosphate or phosphonate moiety and a connecting linker. Furthermore, non-fosmidomycin-based DXRi, bisubstrate inhibitors and several prodrug concepts are described. A comprehensive structure–activity relationship (SAR) of nearly all inhibitor types is presented and some novel opportunities for further drug development of DXR inhibitors are discussed. Reptiles are frequently kept as pet animals. They are considered as important reservoirs of protozoa with veterinary-medical significance. At a reptile farm in Ireland, fecal samples were collected from 98 captive reptiles, representing 43 species of three orders (Squamata, Testudines, and Crocodylia). After DNA extraction, all samples were screened by conventional PCRs, targeting the ribosomal small subunit (SSU) RNA and alpha-tubulin genes of trichomonads and SSU RNA gene of Acanthamoeba spp. One leopard gecko (Eublepharis macularius) was positive for a not yet reported species/genotype of the genus Monocercomonas, different from M. colubrorum. Various Acanthamoeba genotypes were detected in six reptilian species, i.e., Acanthamoeba genotype T11 in Eunectes notaeus and Heloderma suspectum/horridum; genotype T4 in Varanus exanthematicus, Chlamydosaurus kingii, and Macrochelys temminckii; and the genotype T13 in Iguana iguana. Some of these amoeba species might have clinicopathological significance in both humans and animals. Our findings highlight the importance to monitor pathogenic protozoa in pet as well as wildlife reptiles, as a source of possible infection for animals and humans living nearby. Cryptosporidium spp., Entamoeba histolytica (E. histolytica), and Giardia intestinalis (G. intestinalis) are prevalent in developed and developing countries, including Egypt. These enteric protozoa usually provoke indistinguishable manifestation including diarrhea. There is scarce studies of their true prevalence in Egypt. We aimed to identify the accurate detection of these enteric pathogenic protozoa from human diarrheic samples using a multiplex-PCR (MT-PCR) assay and to evaluate the diagnostic performance of coproscopy and monoplex nested PCR (nPCR). Fecal specimens were collected from 100 diarrheic patients and were examined coproscopically and molecularly using MT-PCR and nPCR assays. Diagnostic yield using MT-PCR, nPCR, and microscopy revealed 25, 24, and 15 cases, respectively, for G. intestinalis and 12, 9, and 2 cases, respectively, for Cryptosporidium spp. Coproscopy showed six positive samples for E. histolytica complex, while E. histolytica copro-DNA was not recovered in any sample. Coproscopy showed lower sensitivity than copro-MT-PCR in identifying Cryptosporidium spp. (17%) and G. intestinalis (60%) with 100% specificity. Coproscopy has limited specificity for detection of E. histolytica complex. Compared to MT-PCR, nPCR showed a sensitivity of (75%) for Cryptosporidium spp. and (96%) for G. intestinalis, with 100% specificity for both parasites. G. intestinalis and Cryptosporidium spp. were the prevailing diarrhea-causing protozoa, while E. histolytica was absent among examined Egyptians. Copro-PCR assays could distinguish between pathogenic E. histolytica and non-pathogenic Entamoeba dispar avoiding misdiagnosis and overmedication. Subsequently, MT-PCR allows simultaneous detection of the three protozoa in a single, affordable, and less time-consuming reaction with high sensitivity and specificity. [ABSTRACT FROM AUTHOR]

Controlling drinking water treatment processes is essential to address water contamination and the adaptability of certain pathogenic protozoa. Sometimes, standard treatment methods and chlorine disinfection may prove insufficient in eliminating pathogenic protozoa. However, ultraviolet (UV) radiation has proved to be more effective than chlorine. This study aims to characterize the eukaryotic community of a drinking water treatment plant that applies a final UV disinfection treatment, focusing on pathogenic protozoa. Fifty water samples (raw water, before and after UV treatment) were evaluated to comply with regulation parameters and identify relevant protozoa. Despite physicochemical and microbiological parameters meeting the regulation, some potentially pathogenic protozoa, such as Blastocystis or Cryptosporidium , were still detected in very low relative abundances in treated water. It was found for the first time in Spain the pathogenic amoebae Naegleria fowleri in one river water, which was not found after the treatment. Moreover, Blastocystis subtypes ST1-ST6 were detected in this study in raw, before and after UV water samples. Blastocystis was only found in 2 two samples after UV treatment, with a very low abundance (≤0.02%). Obtained results demonstrate the effectiveness of water treatment in reducing the prevalence of pathogenic protozoa. • Amplicon sequencing revealed the microbiome of the drinking water treatment process. • UV disinfection treatment is effective in reducing relevant protozoa. • Potentially pathogenic protozoa were found in low relative abundances after treatment. The physical barrier of the intestine and associated mucosal immunity maintains a delicate homeostatic balance between the host and the external environment by regulating immune responses to commensals, as well as functioning as the first line of defense against pathogenic microorganisms. Understanding the orchestration and characteristics of the intestinal mucosal immune response during commensal or pathological conditions may provide novel insights into the mechanisms underlying microbe-induced immunological tolerance, protection, and/or pathogenesis. Over the last decade, our knowledge about the interface between the host intestinal mucosa and the gut microbiome has been dominated by studies focused on bacterial communities, helminth parasites, and intestinal viruses. In contrast, specifically how commensal and pathogenic protozoa regulate intestinal immunity is less well studied. In this review, we provide an overview of mucosal immune responses induced by intestinal protozoa, with a major focus on the role of different cell types and immune mediators triggered by commensal (Blastocystis spp. and Tritrichomonas spp.) and pathogenic (Toxoplasma gondii, Giardia intestinalis, Cryptosporidium parvum) protozoa. Entamoeba histolytica is one of the common pathogenic protozoa encountered in Calabar, Nigeria. Differentiating it from the non-pathogenic species, Entamoeba dispar is not a routine practice in our medical laboratories leading to misdiagnosis, overdiagnosis and drug abuse. Studies suggested that stool antigen assays are more sensitive and specific than microscopy for the diagnosis of Entamoeba histolytica infection. This study was carried out to detect and differentiate E. histolytica from E. dispar, hence determining the actual prevalence of E. histolytica among primary school children in Calabar, Nigeria between February and July 2019. A total of 384 stool samples collected from public and private primary school children, aged 5-15years were examined. Direct stool microscopy and ELISA kit-abnova KA3201 were used for Entamoeba histolytica/dispar complex detection while differentiation between E. histolytica and E. dispar was performed using E. histolytica 11IgG ELISA Kit-abnova KA5130. Prevalence of E. histolytica/E. dispar by microscopy and ELISA were 15(3.91%) and 21(5.47%), respectively. The prevalence of Entamoeba histolytica and that of E. dispar after differentiation were 7(1.82%) and 14(3.65%), respectively. Children in public schools were more significantly infected with E. histolytica 6(3.13%) than their private school counterpart 1(0.52%), (P = 0.0211). Although there was no association between Entamoeba infection and gender, males had an insignificantly higher prevalence of E. histolytica and E. dispar 5(3.0%) and 10(6.2%), respectively than females 2(0.9%) and 4(1.8%), respectively (P = 0.1210). There was no association between Entamoeba infection and age but children between the age of 8-10years were more infected with E. histolytica 5(3.70%) and E. dispar 9(6.67%) than others (P =0.2358). Calabar South recorded an insignificantly higher prevalence for E. histolytica 4(2.08%) and E. dispar 8(4.17%) than Calabar municipal 3(1.56%) and 6(3.13%), respectively (P = 0.7029). Parasitic protozoa are eukaryotic unicellular organisms that depend on a variety of living organisms and can develop intra- and extracellularly inside their hosts. In humans, these parasites cause diseases with a significant impact on public health, such as malaria, toxoplasmosis, Chagas disease, leishmaniasis and amebiasis. The ability of a parasite in establishing a successful infection depends on a series of intricate evolutionarily selected adaptations, which include the development of molecular and cellular strategies to evade the host immune system effector mechanisms. The complement system is one of the main effector mechanisms and the first humoral shield of hosts innate immunity against pathogens. For unicellular pathogens, such as protozoa, bacteria and fungi, the activation of the complement system may culminate in the elimination of the invader mainly via 1- the formation of a pore that depolarizes the plasma membrane of the parasite, causing cell lysis; 2- opsonization and killing by phagocytes; 3- increasing vascular permeability while also recruiting neutrophils to the site of activation. Numerous strategies to avoid complement activation have been reported for parasitic protozoa, such as 1- sequestration of complement system regulatory proteins produced by the host, 2- expression of complement system regulatory proteins, 3- proteolytic cleavage of different complement effector molecules, 4- formation of a physical glycolipid barrier that prevents deposition of complement molecules on the plasma membrane, and 5- removal, by endocytosis, of complement molecules bound to plasma membrane. In this review, we revisit the different strategies of blocking various stages of complement activation described for the main species of parasitic protozoa, present the most recent discoveries in the field and discuss new perspectives on yet neglected strategies and possible new evasion mechanisms. Parasites and other eventually pathogenic organisms require the ability to adapt to different environmental conditions inside the host to assure survival. Some host proteins have evolved as defense constituents, such as lactoferrin (Lf), which is part of the innate immune system. Lf in its iron-free form (apo-Lf) and its peptides obtained by cleavage with pepsin are microbicides. Parasites confront Lf in mucosae and blood. In this work, the activity of Lf against pathogenic and opportunistic parasites such as Cryptosporidium spp., Eimeria spp., Entamoeba histolytica, Giardia duodenalis, Leishmania spp., Trypanosoma spp., Plasmodium spp., Babesia spp., Toxoplasma gondii, Trichomonas spp., and the free-living but opportunistic pathogens Naegleria fowleri and Acanthamoeba castellani were reviewed. The major effects of Lf could be the inhibition produced by sequestering the iron needed for their survival and the production of oxygen-free radicals to more complicated mechanisms, such as the activation of macrophages to phagocytes with the posterior death of those parasites. Due to the great interest in Lf in the fight against pathogens, it is necessary to understand the exact mechanisms used by this protein to affect their virulence factors and to kill them. Background. The study was conducted to observe the prevalence and type of infection caused by intestinal parasites and to compare the techniques that are available for the detection. Method. A total of 112 stool samples were obtained from study participants, and the laboratory examinations were performed at Special Infectious Agents Unit, King Abdulaziz University, Jeddah. One hundred and twelve participants were included in this study. Result. The color of positive specimens was mainly brown (86.4%). Stool consistency in infected cases was soft in (64%) samples. There was no statistically significant difference with the physical characteristics of the stool (P >0:05). In total, 59 of the 112 participants were infected with intestinal parasites, representing 52.7%. Different intestinal protozoa parasites were identified in which Blastocystis hominis (86.4%) was highest. None of the intestinal helminths were detected. Out of the 59 infected cases, single infections were found in (62.7%) samples. The intestinal protozoan parasites in single infections were B. hominis (78.4%), Giardia lamblia (8.1%), and (2.7%) from each Entamoeba histolytica, Cryptosporidium parvum, Entamoeba coli, Endolimax nana, and Chilomastix mesnili. Microscopy, RDTs, and real-time PCR were used for detection and identification of G. lamblia, E. histolytica, and C. parvum. Conclusion. The study reported a high rate of intestinal parasitic infections, mainly with B. hominis. There were no statistical differences in parasite detection between the three techniques in detection of the thirteen cases infected with the pathogenic protozoa. [ABSTRACT FROM AUTHOR]

The flat oyster Ostrea edulis is an oyster species native to Europe. It has declined to functional extinction in many areas of the NE Atlantic for several decades. Factors explaining this decline include over-exploitation of natural populations and diseases like bonamiosis, regulated across both the EU and the wider world and caused by the intracellular protozoan parasite Bonamia ostreae. To date, very limited sequence data are available for this Haplosporidian species. We present here the first transcriptome of B. ostreae. As this protozoan is not yet culturable, it remains extremely challenging to obtain high-quality -omic data. Thanks to a specific parasite isolation protocol and a dedicated bioinformatic pipeline, we were able to obtain a high-quality transcriptome for an intracellular marine micro-eukaryote, which will be very helpful to better understand its biology and to consider the development of new relevant diagnostic tools. Antimicrobial resistance (AMR) is becoming more common in both bacteria and pathogenic protozoa. Therefore, new solutions are being sought as alternatives to currently used agents. There are many new ideas and solutions, especially compounds of natural origin, including essential oils. In the present study, the antiprotozoal activity of a mixture of essential oils (eucalyptus, lavender, cedar and tea tree), organic acids (acetic acid, propionic acid and lactic acid) and metal ions (Cu, Zn, Mn) were tested. As a model, protozoans were selected: Euglena gracilis, Gregarina blattarum, Amoeba proteus, Paramecium caudatum, Pentatrichomonas hominis. The tested concentrations of mixtures were in the range of 0.001–1.5%. The analyses show unexpected, very strong protozoicidal activity of combinations, presenting the synergy of compounds via determination of LD50 and LD100 values. Obtained mixtures showed significantly higher activity against protozoans, compared to chloramphenicol and metronidazole. Most of the analyzed samples show high antiprotozoal activity at very low concentration, in the range of 0.001–0.009%. The most effective combinations for all analyzed protozoans were the cedar essential oil and tea tree essential oil with a mixture of acids and manganese or zinc ions. Innovative combinations of essential oils, organic acids and metal ions are characterized by very high antiprotozoal activity at low doses, which, after further investigation, can be applicable for control of protozoan pathogens. Natural products have been proven to be important starting points for the development of new drugs. Bacteria in the genera Photorhabdus and Xenorhabdus produce antimicrobial compounds as secondary metabolites to compete with other organisms. Our study is the first comprehensive study screening the anti-protozoal activity of supernatants containing secondary metabolites produced by 5 Photorhabdus and 22 Xenorhabdus species against human parasitic protozoa, Acanthamoeba castellanii, Entamoeba histolytica, Trichomonas vaginalis, Leishmania tropica and Trypanosoma cruzi, and the identification of novel bioactive antiprotozoal compounds using the easyPACId approach (easy Promoter Activated Compound Identification) method. Though not in all species, both bacterial genera produce antiprotozoal compounds effective on human pathogenic protozoa. The promoter exchange mutants revealed that antiprotozoal bioactive compounds produced by Xenorhabdus bacteria were fabclavines, xenocoumacins, xenorhabdins and PAX peptides. Among the bacteria assessed, only P. namnaoensis appears to have acquired amoebicidal property which is effective on E. histolytica trophozoites. These discovered antiprotozoal compounds might serve as starting points for the development of alternative and novel pharmaceutical agents against human parasitic protozoa in the future. Filter backwash water (FBW) is a prominent residue from water treatment plants (WTPs) that is often disposed into water bodies or recycled within the WTP without due disinfection. FBW usually contains particles within a size range that includes pathogenic protozoa, as the infective forms of Giardia and Cryptosporidium, parasites responsible for waterborne diseases outbreaks. Quantifying (oo)cysts is essential for addressing this matter, as it might assist research on giardiasis and cryptosporidiosis epidemiology, as well as shed light onto disinfection technologies for FBW. However, (oo)cyst recovery from FBW and other complex matrices still lacks a standard protocol and entails specialized professionals and expensive material. Seeking to provide insight in a reduced-cost recovery method, this study analysed the recovery efficiency (RE) obtained by acid flocculation with ferric sulphate, a common coagulant, on bench-scale simulated FBW. Steps included concentration by flocculation, centrifugation, and quantification by immunofluorescence. Although recovery was sufficient for Cryptosporidium parvum (40.59%), Method 1623.1 recommendations were not reached for Giardia muris (1.76%). Coefficients of variation obtained for both organisms were not satisfactory, highlighting the variability to which environmental matrices are subjected and why defining a methodology for (oo)cyst recovery in WTP residues is important. Protozoan parasites interact with a wide variety of organisms ranging from bacteria to humans, representing one of the most common causes of parasitic diseases and an important public health problem affecting hundreds of millions of people worldwide. The current treatment for these parasitic diseases remains unsatisfactory and, in some cases, very limited. Treatment limitations together with the increased resistance of the pathogens represent a challenge for the improvement of the patient's quality of life. The continuous search for alternative preclinical drugs is mandatory, but the mechanisms of action of several of these compounds have not been described. Electron microscopy is a powerful tool for the identification of drug targets in almost all cellular models. Interestingly, ultrastructural analysis showed that several classes of antiparasitic compounds induced similar autophagic phenotypes in trypanosomatids, trichomonadids, and apicomplexan parasites as well as in Giardia intestinalis and Entamoeba spp. with the presence of an increased number of autophagosomes as well as remarkable endoplasmic reticulum profiles surrounding different organelles. Autophagy is a physiological process of eukaryotes that maintains homeostasis by the self-digestion of nonfunctional organelles and/or macromolecules, limiting redundant and damaged cellular components. Immunity Related GTPases (IRG) are a family of proteins produced during infection that regulate membrane remodeling events in cells, particularly autophagy and mitophagy. The human IRGM gene has been strongly associated with Crohn's disease and other inflammatory diseases through Genome-Wide Association studies. Absence of Irgm1 in mice prompts intestinal inflammation, autoimmunity, and impaired immune control of pathogenic bacteria and protozoa. Although prior work has focused on a prominent role for IRGM/Irgm1 in regulating macrophage function, the work described here addresses a potential role of Irgm1 in regulating the function of mature T cells. Irgm1 was found to be highly expressed in T cells in a manner that varied with the particular T cell subset and increased with activation. Mice with a complete lack of Irgm1, or a conditional lack of Irgm1 specifically in T cells, displayed numerous changes in T cell numbers and function in all subsets examined, including CD4+ (Th1 and Treg) and CD8+ T cells. Related to changes in T cell number, apoptosis was found to be increased in Irgm1-deficient CD4+ and CD8+ T cells. Altered T cell metabolism appeared to be a key driver of the phenotypes: Glucose metabolism and glycolysis were increased in Irgm1-deficient CD4+ and CD8+ T cells, and muting these effects with glycolytic inhibitors partially restored T cell function and viability. Giardia and Cryptosporidium are pathogenic protozoa often present in the environment in their infective form(cysts and oocysts). These parasites are very resistant to disinfection, which makes them important target organisms in environmental quality monitoring and sanitation. Viability assessment provides an interpretation of cell inactivation, and it can be evaluated by membrane integrity as well as enzyme activity, using different staining methods. These are straightforward and adequate to laboratories that lack infrastructure for molecular-based technologies or animal infectivity tests. This study investigated simultaneous staining by a commercial live/dead kit, in order to assess viability of Cryptosporidium parvum oocysts and Giardia muris cysts, comparing it to propidium iodide (PI) incorporation, a common stain applied in viability estimation. Results suggested that, although the central hypothesis of one-panel visualization (α = 0.05) was met, simultaneous staining impaired (oo)cyst detection by immunofluorescence assay (IFA), which was found to be essential to enumeration, as the live/dead test led to poor (oo)cyst labelling or a 10-fold lower recovery when carried out concomitantly to IFA. As for the viability assessment itself, although red dye uptake occurred as expected by dead or weakened organisms, neither live G. muris cysts or C. parvum oocysts present any green fluorescence by esterase metabolism. This may have been caused by low enzyme activity in the infective form and/or wall thickness of these parasites. The results do not exclude the possibility of simultaneous fluorescence staining for protozoa, but it is a starting point for a broader analysis, that may consider, for instance, different incubation conditions. The objectives of this research are to evaluate Giardia and Cryptosporidium contamination in surface water supply in Rio Grande do Sul (RS) State in South Brazil in the years 2016 to 2020, assess seasonality, and to infer the population that may have been exposed to these protozoa through drinking water based on drinking water treatment efficiency. Data were obtained through the drinking water surveillance national information system. From 204 DWT plants in the state, 66 have been analyzed for protozoa. A total of 2304 analyses of protozoa in raw water were evaluated, of which 223 had both Giardia spp. cysts and/or Cryptosporidium spp. oocysts in concentrations that varied from 0.1 to 21.5/L. A total of 2,712,125 people from 48 cities were at risk of having the presence of pathogenic protozoa in their drinking water. The probability of finding these protozoa was higher in winter. Giardia cysts were more likely to be found in a period without rain, suggesting that sewage was the main source of contamination. It is concluded that the springs of Rio Grande do Sul are impacted and the circulation of pathogenic protozoa through the territory is endemic with a probable source of contamination to sewage and livestock activity. The accidental ingestion of treated recreational water is an important transmission route of waterborne protozoa worldwide. The present study aimed to provide the first evaluation of swimming pools in Brazil, analysing the presence of pathogenic protozoa (Toxoplasma gondii, Cryptosporidium spp. and Giardia spp.) by parasitological and molecular methods. A total of 57 samples were collected from 21 public swimming pools, either directly from the pool or filter backwash water and concentrated using the membrane filtration technique. Giardia cysts and Cryptosporidium oocysts were visualized by direct immunofluorescence assay after purification by immunomagnetic separation. Toxoplasma gondii oocysts were detected by autofluorescence visualization using the supernatant discarded during the purification step as a sample. Positive samples were submitted to molecular analysis. The molecular markers were used: SSU‐rRNA, tpi, gdh and bg, for Giardia DNA amplification, and 18S rRNA gene fragment amplification was used for the Cryptosporidium oocysts. The 529‐bp repeat element (REP529) fragment and the 35‐fold repetitive B1 gene were employed as a target for T. gondii. Amplified products were submitted to sequencing and phylogenetic analysis. Giardia cysts were detected in 19.0% and Cryptosporidium oocysts in 9.5% of swimming pools. In one swimming pool (4.7%), both protozoa were detected on at least one occasion. Structures similar to T. gondii oocysts were detected in 33.3% of the samples, ranging from one to 23 per slide. Giardia was confirmed by DNA amplification in three swimming pools; Giardia duodenalis Assemblage A was identified by the phylogenetic positioning of the β‐giardin gene. Toxoplasma gondii DNA was detected in 14.2% of swimming pools. The present study represents the first report of the occurrence of T. gondii oocysts in swimming pools. Purines and pyrimidines are essential nutrients for any cell. Most organisms are able to synthesize their own purines and pyrimidines, but this ability was lost in protozoans that adapted to parasitism, leading to a great diversification in transporter activities in these organisms, especially for the acquisition of amino acids and nucleosides from their hosts throughout their life cycles. Many of these transporters have been shown to have sufficiently different substrate affinities from mammalian transporters, making them good carriers for therapeutic agents. In this review, we summarize the knowledge obtained on purine and pyrimidine activities identified in protozoan parasites to date and discuss their importance for the survival of these parasites and as drug carriers, as well as the perspectives of developments in the field. Extracellular vesicles (EVs) are membranous structures formed during biological processes in living organisms. For protozoan parasites, secretion of EVs can occur directly from the parasite organellar compartments and through parasite-infected or antigen-stimulated host cells in response to in vitro and in vivo physiological stressors. These secreted EVs characteristically reflect the biochemical features of their parasitic origin and activating stimuli. Here, we review the species-specific morphology and integrity of parasitic protozoan EVs in concurrence with the origin, functions, and internalization process by recipient cells. The activating stimuli for the secretion of EVs in pathogenic protozoa are discoursed alongside their biomolecules and specific immune cell responses to protozoan parasite-derived EVs. Ovothiols are histidine-derived thiols that are receiving a great interest for their biological activities in human model systems. Thanks to the position of the thiol group on the imidazole ring of histidine, these compounds exhibit unusual antioxidant properties. They have been revealing a very promising pharmacological potential due to their anti-proliferative and anti-inflammatory properties, as well as anti-fibrotic activities not always related to their antioxidant power. Ovothiols occur in three differentially methylated forms (A, B and C), isolated from ovary, eggs and biological fluids of many marine invertebrates, mollusks, microalgae, and pathogenic protozoa. These molecules are synthesized by two enzymes: the sulfoxide synthase OvoA and the sulfoxide lyase OvoB. OvoA catalyzes the insertion of the sulfur atom of cysteine on the imidazole ring of histidine, leading to the formation of a sulfoxide intermediate. This is then cleaved by OvoB, giving 5-thiohistidine, finally methylated on the imidazole ring thanks to the methyltransferase domain of OvoA. Recent studies have shown that OvoA homologs are encoded in a wide variety of genomes suggesting that ovothiol biosynthesis is much more widespread in nature than initially thought. Here we have investigated the OvoA occurrence in diatoms, one of the most abundant group of microalgae, dominating marine and freshwater environments. They are considered a very good model system for both biology/photophysiology studies and for biotechnological applications. We have performed comparative sequence and phylogenetic analyses of OvoA from diatoms, highlighting a high degree of conservation of the canonical domain architecture in the analyzed species, as well as a clear clustering of OvoA in the two different morphological groups, i.e. centric and pennate diatoms. The in silico analyses have also revealed that OvoA gene expression is modulated by growth conditions. More importantly, we have characterized the thiol fraction from cultures of the coastal centric diatom Skeletonema marinoi , providing the first evidence of ovothiol B biosynthesis in diatoms. Treatments using ozone for filter backwash water (FBW) and calcium oxide for floated residue (FR) were evaluated adopting bench-scale testing for the inactivation of Giardia spp. cysts and Cryptosporidium parvum oocysts. The protocol chosen for protozoa detection involved following the concentration step by direct centrifugation (adding ICN 7X cleaning solution at 1.0%) and purification by immunomagnetic separation (IMS). The FR treatment with calcium oxide (dosage of 23 mg CaO 100 mL−1 and 3-day contact time at 25 °C) proved to be efficient, as no parasites were detected after the treatment. The reduction of calcium oxide dosage (16 mg CaO 100 mL−1 and 3-day contact time at 25 °C) was insufficient to inactivate the protozoa, since potentially viable organisms were identified using propidium iodide (PI). Concerning the disinfection conditions with ozone (5-min and 10-min contact time and dosage of 10 mg O3 L−1 and 7.5 mg O3 L−1, respectively), there was complete removal of the target organisms, as no protozoa were detected after the FBW treatment. From the results obtained, the tested treatments can be considered promising alternatives for water treatment plants (WTPs). However, the costs incurred from these treatments have to be considered. The protozoa Giardia and Cryptosporidium are associated with numerous outbreaks of waterborne diseases worldwide. This study aimed to evaluate the concentration of Giardia spp. cysts, Cryptosporidium spp. oocysts, total coliforms, Escherichia coli and Clostridium perfringens in raw wastewater and their removals at UASB reactor, activated sludge system (operated conventionally and extended aeration) and slow sand filtration. Giardia spp. cysts were present in 100% and Cryptosporidium spp. oocysts in 31.4% of the analysed wastewater samples. The UASB reactor followed by activated sludge system obtained approximately 2.0 log of removal for total coliforms and E. coli, whereas for C. perfringens and Giardia spp. cysts, it obtained 1 log. There was a high percentage of (oo)cysts still viable after secondary treatment, therefore, the risk of contamination of water courses and, consequently, for public health is considerable. However, after tertiary filtration, no (oo)cysts were found in any of the filtered effluent samples, being a good option for future reuse. Seasonal variations did not affect the concentrations and removals of microorganisms observed. Lack of correlations of concentrations of indicator microorganisms and (oo)cysts raise caveats and doubts regarding the true microbiological quality when using only indicator microorganisms. Giardiasis leads to severe diarrhea, and its impacts are especially concerning in low-income sites that lack water and sanitation infrastructure. Monitoring water quality considering the infective form of Giardia is therefore an action that strongly aligns with public health. The standard protocol for cyst quantification in water involves immunofluorescence assays (IFA), which depend on fluorescent probes coupled to antibodies, making it an observer-dependent, and a relatively expensive method. Lens-free holographic microscopy (LHM) offers a possible alternative for imaging, as it relies on portable, lightweight, and low-cost equipment. It is based on phase and amplitude images of holograms recorded with partially coherent lighting. This study, therefore, aimed to test LHM for the detection and counting of Giardia cysts. Purified suspensions of G. duodenalis cysts, with known concentration, were quantified by IFA and LHM. Holograms were acquired by transmission, using three different wavelengths, and were then submitted to automated analysis, using an algorithm developed in Python. Slide preparation and cyst counting by IFA proved to be more laborious compared to LHM. Agglomerations of cysts impaired both methods. LHM quantification showed significant similarity with the reference number of cysts in suspension, which was not obtained by standard immunofluorescence. LHM proved to be an effective technique for detecting and counting Giardia cysts in purified samples and with high concentrations, showing potential for future application in water quality assessment targeting pathogens, but limitations include low sensitivity and a demand for future work considering matrix effect on real environmental samples. J-DNA-binding protein 1 (JBP1) contributes to the biosynthesis and maintenance of base J (β-D-glucosyl-hydroxymethyluracil), an epigenetic modification of thymidine (T) confined to pathogenic protozoa such as Trypanosoma and Leishmania. JBP1 has two known functional domains: an N-terminal T hydroxylase (TH) homologous to the 5-methylcytosine hydroxylase domain in TET proteins and a J-DNA-binding domain (JDBD) that resides in the middle of JBP1. Here, we show that removing JDBD from JBP1 results in a soluble protein (Δ-JDBD) with the N- and C-terminal regions tightly associated together in a well-ordered structure. We found that this Δ-JDBD domain retains TH activity in vitro but displays a 15-fold lower apparent rate of hydroxylation compared with JBP1. Small-angle X-ray scattering (SAXS) experiments on JBP1 and JDBD in the presence or absence of J-DNA and on Δ-JDBD enabled us to generate low-resolution three-dimensional models. We conclude that Δ-JDBD, and not the N-terminal region of JBP1 alone, is a distinct folding unit. Our SAXS-based model supports the notion that binding of JDBD specifically to J-DNA can facilitate T hydroxylation 12-14 bp downstream on the complementary strand of the J-recognition site. We postulate that insertion of the JDBD module into the Δ-JDBD scaffold during evolution provided a mechanism that synergized J recognition and T hydroxylation, ensuring inheritance of base J in specific sequence patterns following DNA replication in kinetoplastid parasites. Cyclospora species are socioeconomically important protistan pathogens. Cyclospora cayetanensis is usually transmitted via food or water to a human host via the faecal-oral route and can cause the gastrointestinal disease cyclosporiasis, which can be complicated by extra-intestinal disorders, particularly in immune-compromised people. Although more than 2 million children die each year from diarrhoeal diseases worldwide, it is not known to what extent cyclosporiasis is involved. Few epidemiological data are available on Cyclospora as a water-borne and food-borne pathogen in both underprivileged communities and developed countries. To gain an improved understanding of human cyclosporiasis, this Review describes the background of Cyclospora, summarises salient aspects of the pathogenesis, epidemiology, diagnosis, treatment, and control of cyclosporiasis, and explores what is known about its prevalence and geographical distribution. The findings show that the effect on human health of cyclosporiasis is likely underestimated, and recommendations are made about areas of future research and the prevention and control of this disease within an international collaborative context. Toxoplasma gondii is an obligate intracellular parasite that causes the zoonoses disease, named toxoplasmosis, with global prevalence. Until now, no cost-effective treatment method has been found to deal with toxoplasma, and vaccination is the best way to deal with the infection. In the case of pathogenic protozoa, mainly live vaccines have had successful results compared to other vaccine platforms. This study evaluated the efficacy of a live experimental vaccine through long-term passages on the Gecko cell line (Z1) in inducing a protective immune response in BALB/c mice. Thirty mice were divided into three equal groups; G1: the immunized/challenged group (injection of attenuated strain), G2: the immunized/unchallenged group (injection of attenuated strain), and G3: the control group (injection of culture medium).One month after immunization, the studied mice were challenged with 1ₓ103 live tachyzoites of Toxoplasma acute RH strain. We performed Serological investigations, including evaluating antibodies, interferon-gamma (IFN-γ), and interleukins 2, 4, 10, and 12 (IL-2,4,10,12). At the study's end, a molecular test was performed on brain and liver tissues in the immunized groups to check the presence of parasites. The results from the serological tests for the evaluation of antibodies, interferon-gamma (IFN-γ), and interleukins 10 and 12 (IL-10, 12) show a significant difference (p < 0.05) between the vaccinated group and the control group, which are essential indicators of protective immunity against toxoplasma infection. Thus, in the vaccinated group, the survival rate of mice against the challenge was 70%. Also, in group two (G2), the attenuated strain of Toxoplasma gondii had no pathogenicity, and all mice survived until the end of the study period. Molecular results also showed the absence of parasites in the brain and liver tissues in this immunized group and the parasite was found in only one case of liver tissue in G1. Therefore, the attenuated strain has caused significant and protective humoral and cellular immune responses in vaccinated groups. This study showed that with the long-term passage of the acute strain on the Gecko cell line, it is possible to quickly obtain a non-diseased attenuated strain with the ability to induce protective immunity. In vitro studies to fourteen previously synthesized chromone-tetrazoles and four novel fluorine-containing analogs were conducted against pathogenic protozoan (Entamoeba histolytica), pathogenic bacteria (Pseudomonas aeruginosa, and Staphylococcus aureus), and human fungal pathogens (Sporothrix schenckii, Candida albicans, and Candida tropicalis), which have become in a serious health problem, mainly in tropical countries.

Bats are considered important reservoirs of pathogens of veterinary and medical relevance worldwide. However, despite the increasing attention paid towards the central role of bats in the spreading of some zoonotic infections, studies on their own parasites remain patchy especially in Neotropical regions. Understanding the relationships occurring between bats and their pathogens is a crucial step to determine the implications undermining the ecology and biology of these animals. The present study aims to assess the endoparasitic fauna of Neotropical bats from Northeastern Brazil, with a particular focus on helminths and protozoa. From October 2016 to March 2017, 54 fresh faecal samples were collected in stable shelters inhabited by three different bat species (i.e., Molossus molossus , Myotis lavali and Noctilio albiventris ). Samples were analysed using four different techniques (i.e., Centrifugal Sedimentation, Flotation Technique, Spontaneous Sedimentation Technique and FLOTAC ® ). A total of 96.29% (52/54) samples were positive for at least one gastrointestinal parasite and 11 different families were identified. In particular, 63.6% (7/11) of the families were of helminths and 36.4% (4/11) of protozoa. Helminths were classified into two different classes (Nematoda and Cestoda) and seven families, whereas the four protozoa classes detected (i.e., Coccidia, Gregarinomorphe, Lobosea and Zoomastigophora) into four families. Eggs of Ancylostomatidae and Hymenolepididae were the most abundan. The findings of this present study demonstrate that Neotropical bats from Northeastern Brazil are parasitized by a wide number of parasites, and some of them may have important implications for Public Health. Polyamines are small polycations that are well conserved in all the living organisms except Archae, Methanobacteriales and Halobacteriales. The most common polyamines are putrescine, spermidine and spermine, which exist in varying concentrations in different organisms. They are involved in a variety of cellular processes such as gene expression, cell growth, survival, stress response and proliferation. Therefore, diverse regulatory pathways are evolved to ensure strict regulation of polyamine concentration in the cells. Polyamine levels are kept under strict control by biosynthetic pathways as well as cellular uptake driven by specific transporters. Reverse genetic studies in microorganisms showed that deletion of the genes in polyamine metabolic pathways or depletion of polyamines have negative effects on cell survival and proliferation. The protein products of these genes are also used as drug targets against pathogenic protozoa. These altogether confirm the significant roles of polyamines in the cells. This mini-review focuses on the differential concentrations of polyamines and their cellular functions in different microorganisms. This will provide an insight about the diverse evolution of polyamine metabolism and function based on the physiology and the ecological context of the microorganisms. [ABSTRACT FROM AUTHOR]
