The acute and chronic toxicity of lead to Anabas testudineus was determined in this study using static replacement bioassay testing. During the chronic toxicity studies, an experiment on the bioremediation of lead toxicity using Ocimum sanctum leaf powder was conducted. The 96 h LC50 values of lead for Anabas testudineus was 1.08 mg/L. Different biomarkers, such as the hepatosomatic index, gonadosomatic index, and fecundity, were significantly lower in fish subjected to 10% and 20% of the 96 h LC50 values of lead, compared to controls. The 45-day chronic exposure of fish to lead concentrations of 0.2 mg/L and above significantly lowered the number of total RBC, hemoglobin content, HCT (%), plasma protein, and cholesterol while decreasing the level of total WBC, plasma glucose, creatinine, serum AST and serum ALT. Fish geographical distribution has pivotal importance in management/conservation policies, especially for community interest species, requiring careful protection measures. However, distribution areas could be difficult to define if the taxonomy has only recently been resolved, and particular attention must be paid where co-generic species share their ranges. Our work reports new insights about the distribution of T. muticellus and T. souffia in Northeastern Italy, where their geographical ranges overlap, and these native species share a cross-border area. Fish samplings and meristic and genetical investigations were performed, and our results showed that T. muticellus seems to expand its range east due to human-induced translocations, while T. souffia occurs only in the cross-border hydrological networks between Italy and Slovenia. Telestes genetic characterization for the Tagliamento River Basin and Northeastern Italy lowland is reported, improving the genus distribution dataset in the Po Plain watershed, where T. muticellus is endemic. The main meristic feature to discriminate the species was the number of lateral line scales, explaining 44.5% of the dissimilarity among the analyzed populations. Findings about the distribution of the investigated populations highlight the critical need for targeted conservation strategies in biodiversity hotspots, where overlapping species distributions, such as T. muticellus and T. souffia, play a vital role in preserving regional biodiversity. Simple Summary: Meagre (Argyrosomus regius) is a fast-growing species used in aquaculture, but it faces significant challenges from Systemic Granulomatosis (SG), a disease marked by granulomas in various internal organs. This study investigated the possible link between Mycobacterium spp. and SG in 34 seemingly healthy meagre from a Sardinian aquaculture facility. Granulomas were identified in 91% of the fish. Several diagnostic methods, including acid-fast staining, microbiology, PCR, and in situ hybridization, were employed. While M. chelonae was detected in a low percentage through PCR (13%), no evidence suggested its role in SG onset. Meagre (Argyrosomus regius) is one of the fast-growing species considered for sustainable aquaculture development along the Mediterranean and Eastern Atlantic coasts. The emergence of Systemic Granulomatosis (SG), a disease marked by multiple granulomas in various tissues, poses a significant challenge in meagre aquaculture. In the current study, we investigate the association of Mycobacterium spp. and SG in offshore aquaculture facilities in Sardinia, Italy. A total of 34 adult seemingly healthy meagre were arbitrarily collected and analyzed, combining histological, microbiological, molecular, metagenomics, and in situ techniques to investigate the presence of pathogens. Ziehl–Neelsen (ZN), periodic acid–schiff (PAS), and Giemsa stains were performed for the detection of acid-fast bacteria, common parasites, and fungi within granulomas, respectively. Granulomas were detected in 91% (31/34) of fish. The affected organs were kidney (88%), liver (47%), heart (41%), intestine (17.6%), and brain (5%). Acid-fast staining, along with Mycobacterium spp. specific quantitative PCR (qPCR), in situ hybridization (ISH) assay, and microbiological analyses showed negative results for the detection of Mycobacterium spp. and other bacteria implicated in granuloma formation. However, PCR amplification and sequencing of the 65-kDa heat shock protein gene revealed the presence of M. chelonae in 13% of both formalin-fixed and frozen liver tissues. Bacterial isolation failed to detect nontuberculous mycobacteria (NTM) and other bacteria typically associated with granulomas. Consistently, the use of an M. chelonae-specific probe in ISH failed to identify this bacterial species in granulomas. Collectively, results do not support the role of M. chelonae in the development of granulomas and suggest rejecting the hypothesis of a potential link between NTM and SG. [ABSTRACT FROM AUTHOR]

Simple Summary: Lactococcus petauri has been described for the first time in 2017 in a sugar glider. A few years later, it has emerged as important pathogen for fish and more recently it has been reported also in terrestrial mammals and humans. A lot of these cases were misdiagnosed as Lactococcus garvieae infection due to the limitation of the available common standard diagnostic techniques used which were unable to discriminate between these two species and relatively high similarity of the two pathogens at genetic and phenotypic levels. Till today, there are only two reported cases of lactococcosis in dogs with no previous cases were reported from Italy. This report is the first description of L. petauri infection in two domestics dog cases from Italy. The results of the current report provide an update on the host tropism, and additional knowledge on virulence factors and antimicrobial resistance of L. petauri. Lactococcus garvieae has been considered for a long time the only causal agent of lactococcosis. In recent years, different papers reported the involvement of other two bacterial species: Lactococcus petauri and Lactococcus formosensis. A different host tropism has been described for these species where L. garvieae and L. petauri are predominant species in fish and humans' infections, while L. formosensis in bovine. L. garvieae has been reported as rare infectious agent in dog. This paper represents the first isolation of L. petauri in two domesticated dog cases from urine and skin samples, respectively. The recovered L. petauri has been identified using PCR and sequencing based on Internal Transcribe Spacer (ITS) and phylogenetic analysis showed that it belongs to the L. petauri cluster with a 100% of identity with sequences previously reported from fish isolates while there were differences with L. petauri isolated from urinary tract infection from humans. L. petauri in human infection has been considered not necessarily deriving from the ingestion of contaminated food but rather as an opportunistic pathogen colonization intestinal tract. Differences among virulotypes have been reported for humans and dogs, and a comparison was also made between the virulotyping of L. petauri and L. garvieae in dogs. The antimicrobial pattern showed susceptibility for the election treatment molecules. These data contribute to our understanding of the host trophism of this species which was misclassified for long time and provide new data on its virulence factors and antimicrobial resistance. Lactococcosis is a common bacterial fish disease caused by Lactococcus garvieae, L. petauri and L. formosensis. Although there are different PCR‐based techniques to identify the etiological agent, none of these can differentiate these two bacteria without sequencing PCR‐amplified fragments. In the present study, we developed a multiplex PCR assay for simultaneous detection and differentiation of L. garvieae and L. petauri. The specificity of the primers was validated against the bacterial DNA of the targeted and non‐targeted bacteria. The sizes of the PCR amplicons were obtained as 204 bp for the DUF1430 domain‐containing protein gene of L. garvieae, 465 bp for the Lichenan permease IIC component gene of L. petauri, and 302 bp for the teichoic acid biosynthesis protein F gene of both L. garvieae and L. petauri. The PCR amplicons were clearly separated by agarose gel electrophoresis. The multiplex PCR assay did not produce any amplification products with the DNA of the non‐targeted bacteria. The multiplex PCR detection limits for L. garvieae and L. petauri were 5 and 4 CFU in pure culture and 50 and 40 CFU/g in spiked tissue samples, respectively. It takes less than 2 h from plate‐cultured bacteria and 3 h from tissue samples to get results. In conclusion, the developed multiplex PCR assay is a rapid, specific, accurate, and cost‐effective method for the detection and differentiation of L. garvieae and L. petauri and is suitable to be used for routine laboratory diagnosis of L. garvieae and L. petauri. Trace element pollution in the Persian Gulf originates from industrial activities, urbanization, shipping, and oil extraction, leading to accumulation in sediments, water, and marine life such as jellyfish. This study investigated trace element bioaccumulation in two jellyfish species, Crambionella orsini and Cassiopea andromeda, across different locations. Jellyfish samples were collected from the Mahshahr and Dilam ports, and their trace element concentrations were analyzed using inductively coupled plasma mass spectrometry (ICP-MS). The study revealed no significant differences in trace element concentrations between C. orsini and C. andromeda. However, levels of copper, iron, manganese, and nickel were significantly higher in specimens from Mahshahr compared to those from Dilam. No significant differences were observed for cadmium, mercury, vanadium, tin, magnesium, and selenium across sites. Lower concentrations of lead, arsenic, zinc, and cobalt were found in C. orsini from Dilam compared to Mahshahr. Additionally, the study found no significant correlation between trace element concentrations in the water and their accumulation in jellyfish bodies. These findings offer valuable insights into the distribution and bioaccumulation of trace elements in jellyfish populations across different marine environments in the Persian Gulf. Sediments of alpine lakes serve as crucial records that reveal the history of lacustrine basins, offering valuable insights into the effects of global changes. One significant effect is the variation in rainfall regimes, which can substantially influence nutrient loads and sedimentation rates in lacustrine ecosystems, thereby playing a pivotal role in shaping biotic communities. In this study, we analyze subfossil chironomid assemblages within a sediment core from an alpine lake (western Italian Alps) to investigate the effects of rainfall and flood regime variations over the past 1200 years. Sediment characterization results highlight changes in sediment textures and C/N ratio values, indicating phases of major material influx from the surrounding landscape into the lake basin. These influxes are likely associated with intense flooding events linked to heavy rainfall periods over time. Flooding events are reflected in changes in chironomid assemblages, which in our samples are primarily related to variations in sediment texture and nutrient loads from the surrounding landscape. Increased abundances of certain taxa (i.e., Brillia, Chaetocladius, Cricotopus, Psectrocladius, Cricotopus/Orthocladius Parorthocladius) may be linked to higher organic matter and vegetation inputs from the surrounding landscape. Biodiversity decreased during certain periods along the core profile due to intense flood regimes and extreme events. These results contribute to our understanding of alpine lake system dynamics, particularly those associated with intense flooding events, which are still understudied. [ABSTRACT FROM AUTHOR]
