Routine sampling of pregnant women at first antenatal care (ANC) visits could make Plasmodium falciparum genomic surveillance more cost-efficient and convenient in sub-Saharan Africa. We compare the genetic structure of parasite populations sampled from 289 first ANC users and 93 children from the community in Mozambique between 2015 and 2019. Samples are amplicon sequenced targeting 165 microhaplotypes and 15 drug resistance genes. Metrics of genetic diversity and relatedness, as well as the prevalence of drug resistance markers, are consistent between the two populations. In an area targeted for elimination, intra-host genetic diversity declines in both populations (p = 0.002-0.007), while for the ANC population, population genetic diversity is also lower (p = 0.0004), and genetic relatedness between infections is higher (p = 0.002) than control areas, indicating a recent reduction in the parasite population size. These results highlight the added value of genomic surveillance at ANC clinics to inform about changes in transmission beyond epidemiological data.

BACKGROUND: The infection of the liver by Plasmodium parasites is an obligatory step leading to malaria disease. Following hepatocyte invasion, parasites differentiate into replicative liver stage schizonts and, in the case of Plasmodium species causing relapsing malaria, into hypnozoites that can lie dormant for extended periods of time before activating. The liver stages of Plasmodium remain elusive because of technical challenges, including low infection rate. This has been hindering experimentations with well-established technologies, such as electron microscopy. A deeper understanding of hypnozoite biology could prove essential in the development of radical cure therapeutics against malaria. RESULTS: The liver stages of the rodent parasite Plasmodium berghei, causing non-relapsing malaria, and the simian parasite Plasmodium cynomolgi, causing relapsing malaria, were characterized in human Huh7 cells or primary non-human primate hepatocytes using Correlative Light-Electron Microscopy (CLEM). Specifically, CLEM approaches that rely on GFP-expressing parasites (GFP-CLEM) or on an immunofluorescence assay (IFA-CLEM) were used for imaging liver stages. The results from P. berghei showed that host and parasite organelles can be identified and imaged at high resolution using both CLEM approaches. While IFA-CLEM was associated with more pronounced extraction of cellular content, samples features were generally well preserved. Using IFA-CLEM, a collection of micrographs was acquired for P. cynomolgi liver stage schizonts and hypnozoites, demonstrating the potential of this approach for characterizing the liver stages of Plasmodium species causing relapsing malaria. CONCLUSIONS: A CLEM approach that does not rely on parasites expressing genetically encoded tags was developed, therefore suitable for imaging the liver stages of Plasmodium species that lack established protocols to perform genetic engineering. This study also provides a dataset that characterizes the ultrastructural features of liver stage schizonts and hypnozoites from the simian parasite species P. cynomolgi.

Skin-penetrating nematodes, including the human threadworm Strongyloides stercoralis and hookworms in the genera Necator and Ancylostoma, are gastrointestinal parasites that are a major cause of neglected tropical disease in low-resource settings worldwide. These parasites infect hosts as soil-dwelling infective larvae that navigate towards hosts using host-emitted sensory cues such as odorants and body heat. Upon host contact, they invade the host by penetrating through the skin. The process of skin penetration is critical for successful parasitism but remains poorly understood and understudied. Here, we review current knowledge of skin-penetration behaviour and its underlying mechanisms in the human parasite S. stercoralis, the closely related rat parasite Strongyloides ratti, and other skin-penetrating nematodes such as hookworms. We also highlight important directions for future investigations into this underexplored process and discuss how recent advances in molecular genetic and genomic tools for Strongyloides species will enable mechanistic investigations of skin penetration and other essential parasitic behaviours in future studies. \nHighlightsThe Bangiales are one of the most common seaweeds in sub-Antarctic and Southeastern Pacific upper intertidal habitats. Here we report records of an ‘olpidioid’ marine obligate parasite infecting Bangiales between 39°20′–41°44′S in the Southeastern Pacific for the first time. The disease resembles the ‘Olpidiopsis blight disease’ reported for farmed Bangiales in Japan and Korea and wild Pyropia from Scotland morphologically and developmentally. The Chilean isolate infects commercial Chilean laver or ‘luche’ (Porphyra sensu lato) and Bangia sensu lato from wild populations in the Southeastern Pacific. Phylogenetic markers (cox2, 18S) identify it as Olpidiopsis porphyrae. We relate the occurrence of this pathogen to both ‘luche’ fisheries in Chile and the potential consequences for its emergent aquaculture in the Southeastern Pacific. The Gyrfalcon (Falco rusticolus) is a top avian predator, an Arctic specialist, and among the bird species most vulnerable to climate change. This vulnerability is driven by their narrow ecological niche, limited or lack of southward migration, and circumpolar distribution where the most rapid climatic changes are occurring. Climatic and habitat changes may alter Gyrfalcon disease ecology due to changes in vector distributions, host ranges, and pathogen life cycles. Warmer Arctic temperatures and accompanying landscape changes may also alter the Gyrfalcon's prey base, and dietary habits can influence transmission of pathogens. To better understand disease ecology in Gyrfalcons, we compared pathogen prevalence across varying time periods at three study sites in Alaska—the Seward Peninsula (2014–2022), the Alaska Peninsula (2021–2022), and the Yukon–Kuskokwim Delta (2008–2013). We collected Gyrfalcon whole blood, thin blood films, cloacal swabs, and fecal samples for serology, haemoparasite assays, microbiological cultures, and fecal tests for parasites. An aliquot of whole blood preserved on filter paper or in Longmire solution was kept for molecular diagnosis of haemoparasites. Serology revealed high exposure to Salmonella (77%), low seroprevalence of avian influenza antibodies (1.5%), exposure to falcon adenovirus type 1 in hatch-year Gyrfalcons (1.3%), and the first report of a Leucocytozoon spp. blood parasite in a Gyrfalcon. We found no antibodies indicative of prior exposure to avian paramyxovirus, West Nile virus, or Chlamydia. One nestling and one hatch-year bird sampled (2 of 12) on the Seward Peninsula exhibited oral plaques from capillarids (Eucoelus spp.) in contrast to those trapped in the Izembek National Wildlife Refuge on the Alaska Peninsula (0 of 6). Parasitism is increasingly seen as an ecological factor contributing to behavioural variation among individuals. Yet, the causal direction of the relationship between animal personality and parasites remains unclear. We measured behavioural traits (i.e. exploration, activity, boldness) in pumpkinseed sunfish, Lepomis gibbosus , before and after an experimental infection using cages in a lake where sunfish were naturally exposed to trematode and cestode infection for 1 month. Despite our initial assumptions (i.e. that all individuals would have the same risk of infection within a cage), we found that initial behavioural traits strongly influenced infection susceptibility: initially bolder and less active fish acquired a higher density of trematode and cestode parasites during the infection period. Following infection, fish body condition decreased with increasing cestode density, suggesting that infection is costly to hosts. Body condition was positively correlated with distance moved, a measure of activity, regardless of individual infection status. The repeatability of exploration and activity behaviour and the strength of the activity–exploration correlation (i.e. behavioural syndrome) were reduced following parasite exposure. Distance moved and trematode density were negatively correlated, suggesting that this infection decreases host activity levels. Since trematodes have a complex life cycle with piscivorous birds as a final host, a decrease in activity following infection may make infected fish more susceptible to bird predation, benefiting the parasite. Our results highlight the close links between behaviour and parasitism. Mason bees (Osmia. spp) can provide an alternative for crop pollination management in addition to the Western honeybees (Apis mellifera). Targeted evidence‐based guidelines can improve cost‐effective management of mason bees. Here, I test the effect of an attractant spray developed in the United States for improving the nesting preference for artificial trap nests of mason bees in Belgium. More specifically, this study investigates the effect of the attractant spray on the nesting preference and nesting performance of the European orchard bee (Osmia cornuta), as well as its effect on the infestation of nest‐associated macroparasites. The number of sealed nesting cavities was twice as high in the trap nests that were treated with the attractant spray. Nesting performance expressed as number of brood cells per cavity, successful cocoon formation, adult emergence, proportion of female bees in the offspring and the number of dead larvae was not different between the treated and the untreated nests. Overall infestation of macroparasites, and infestation of drosophilid flies in particular, were both twice as high in untreated nests. This study provides evidence that the attractant spray improves nesting preference and reduces macroparasite infestation in O. cornuta. The use of this attractant can provide growers and stakeholders with an effective management strategy to prevent infestation by macroparasites and promote efficient bee breeding. Purpose: Studies on monogeneans parasitizing representatives of Siluriformes in Africa remain scarce and fragmentary. In order to contribute to fill this gap, we screened Heterobranchus longifilis from the Lindi River in the Democratic Republic of the Congo. Methods: Heterobranchus longifilis was purchased from fishermen in DR Congo, province of Tshopo, in Kisangani town, the Lindi River. The monogeneans were individually isolated from the gill and transferred directly onto a slide in a drop of Hoyer's medium. Results: Quadriacanthus lindiensis n. sp. is morphologically similar to Quadriacanthus longifilisi. However, it is distinguished from the latter by its non-sclerotized vagina, its dorsal cunei shape and the size of its bars. Conclusion: Only Q. lindiensis n. sp. was found on the gills of the single individual of H. longifilis screened. In the future, a representative sampling should enable us to verify if Q. lindiensis n. sp. co-occurs with other species known to parasitize H. longifilis. [ABSTRACT FROM AUTHOR]

Purpose: This study describes the composition and infection patterns of macro endoparasites associated with the lizard Phyllopezus periosus. Methods: We collected 115 P. periosus over 12 expeditions on the rocky cliffs of the Missão Velha waterfall canyon in northeastern Brazil. Specimens were collected by hand, during active searches conducted between 18:00 h and 23:30 h. The infection patterns were estimated using prevalence, total abundance, and mean intensity of infection for all parasites found. Results: A total of 4256 parasites were collected from 104 hosts, resulting in an exceptionally high overall prevalence of 90.4%. No significant ontogenetic difference in the prevalence of infection was detected. Although juveniles exhibited a high parasite prevalence their parasite loads were significantly lower compared to adults. Six species of endoparasites were found; among these, the nematode Spauligodon oxkutzcabiensis was the most common endoparasite in the digestive tract, while the pentastomid Raillietiella mottae was the only species found in the respiratory tract. Conclusions: Infection rates in Gekkota species appear to correlate with body size, with larger individuals generally exhibiting a higher prevalence. This pattern was also observed across separate populations of P. periosus and other geckos from distinct ecological domains. The dominance of the monoxenous S. oxkutzcabiensis highlights potential transmission mechanisms, suggesting that behaviors such as limb licking after feeding may contribute to its prevalence. The occurrence of Physalopteroides venancioi, Oswaldocruzia sp., Parapharyngodon sp., Physaloptera sp., infecting P. periosus are new records for this host. Purpose: In this study, 143 lutjanid fishes representing four different species from the Northeast coast off Brazil were analyzed for parasites. The aim of the present study is to provide a detailed redescription of R. (I.) vicentei, collected from lutjanid fishes in Brazil, including the first observation of the species using scanning electron microscopy (SEM). Methods: Nematodes found clarified in a crescent glycerin series, were photographed and measured (in mm), and some specimens destined for SEM. The Illustrations were made using an optical microscope with a drawing tube. Results: Nematodes found were identified as Raphidascaris (Ichthyascaris) vicentei, infecting 6.29% of the examined fish. Detailed morphological analysis revealed the following characteristics observed for the first time in this species: labial papillae, narrow lateral alae and the detailed structure of male spicules. These contributions help filling the gaps in the literature and expand the knowledge of this parasite morphology. Conclusions: The presence of R. (I.) vicentei in four lutjanid species may indicates low host specificity, highlighting its adaptability to different hosts. Furthermore, new host records are given. This research provides the most comprehensive morphological description of R. (I.) vicentei to date, generating crucial insights for a better understanding of the parasitic biodiversity off the Brazilian Northeastern coast of Brazil, as well as establishing foundation for future investigations on the ecology and impacts of these parasites in fish populations from this region. Host–parasite interactions are influenced by present and past eco‐evolutionary interactions and the local environment. An ecological community defines the potential host range of each parasite and the potential parasite diversity of each host species. Past and present processes translate potential to realised interaction niches of parasite and host species. Host–parasite interactions are antagonistic, which may slow the saturation of their interaction niches. Intervality, a property of bipartite networks, measures saturation of interaction niches. Intervality of a community increases as the interaction niches of species of one guild (e.g. hosts) become saturated for their interactions with another guild (e.g. parasites). Characteristics driving intervality in host and parasite communities are largely unknown, as well as the effect of environmental change on intervality of these communities. In our study, we assess if the characteristics ‘phylogenetic relatedness' and ‘overlap in ecological interactions' explain intervality of rodent host–helminth parasite communities. In addition, we contrast intervality of these communities from habitats that differ in their history of human‐driven modification. We performed the analyses for the interaction niches of both parasites and hosts, independently. Our results indicated that host and parasite communities were non‐interval or significantly less interval than expected by chance. Phylogenetic relatedness and overlap in ecological interactions did not explain the maximum values of intervality. We speculate that antagonistic coevolution in host–parasite communities may hinder communities to reach saturation, which would explain why it is difficult to find the characteristics that explain intervality of a community. Interestingly, intervality of the interaction niche of parasites (host range) increased with habitat modification (i.e. saturation increased), whereas intervality of the interaction niche of hosts (parasite diversity) decreased as habitat modification increased. Purpose: To report a rare non-endemic case of Leishmania aethiopica in Washington DC. Case report: A 68-year-old female presented for a routine examination with a complaint of right upper eyelid lesions for the past 5 months. On examination, a cluster of elevated and erythematous lesions extending from the medial canthus to the brow area of the right eye were seen. Initial treatment with Valtrex based on a suspected viral etiology failed. Although a biopsy was recommended at this time, the patient declined, and subsequent workup included nasolacrimal duct irrigation, blood work to rule out autoimmune etiology, a course of doxycycline, and an MRI, which yielded no improvement. Upon progression of the lesions into persistent plaques on the eyelids, a punch biopsy was performed, confirming leishmaniasis. The patient was then started on a 28-day course of oral miltefosine which led to complete resolution of her symptoms. Conclusion: This case underlines the importance of a broad differential including non-endemic diseases, particularly in urban areas with frequent patient travel. Furthermore, the delayed punch biopsy in this case highlights the importance of patient counseling to ensure prompt diagnosis and treatment. [ABSTRACT FROM AUTHOR]

Background: Parasitic zoonoses are infections transmitted between vertebrate animals and humans, posing serious public health risks and significant economic consequences. The study aimed at assessing government workers' knowledge, attitudes, and practices (KAP) regarding zoonotic parasites of pet animals in Ethiopia's central Gondar zone. Methods: A cross-sectional study was carried out in selected towns of Central Gondar Zone, Northwest Ethiopia from January 2022 to August 2022. Four hundred randomly selected government employees participated in the study. Data were collected through semi-structured questionnaire administered by face-to-face interview. The chi-squared (χ2) was calculated for association and p-value less than 0.05 was considered as significant. Results: A 57.25% of respondents were males and 42.75% were females. Most of the respondents, 67.5%, had completed college/university education. About 60% of the respondents had 'high knowledge', 57.25% had 'positive attitude', and 73% had 'good practice' regarding pet animals as a source of zoonotic parasites. The chi-square analysis revealed a significant discrepancy (p < 0.05) between knowledge with sex, educational level, and position in the house. The attitude level was significantly associated (p < 0.05) with educational level, religious view, and service year. Significant association was also observed (p < 0.05) between practice level of the respondents and educational level and marital status. The study also showed a significant association between knowledge and attitude (χ2 = 40.4, p ≤ 0.001), knowledge and practice (χ2 = 34.9, p ≤ 0.001), and attitude and practice (χ2 = 12.76, p = 0.013) of the respondents. Conclusion: Although our results revealed that more than half of the participants had good knowledge, attitudes, and practices regarding zoonotic parasites of pet animals, enhanced routine hygienic practices and regular awareness creation programs are recommended to bridge existing gaps. Introduction: Leucocytozoon is an intracellular blood parasite that affects various bird species globally and is transmitted by blackfly vectors. This parasite is responsible for leucocytozoonosis, a disease that results in significant economic losses due to reduced meat and egg production. There is limited knowledge about the epidemiological pattern of leucocytozoonosis and its causative species in Egypt, particularly in pigeons. Methods: The current study involved the collection of 203 blood samples from domestic pigeons from various household breeders and local markets across Qena Province, Upper Egypt. Samples were initially examined for potential Leucocytozoon infection using blood smears, followed by an evaluation of associated risk factors. Molecular identification of the parasite in selected samples (n = 11), which had initially tested positive via blood smears, was further refined through nested PCR and sequence analysis of the mitochondrial cytochrome b gene to ascertain the Leucocytozoon species present. Additionally, histopathological examination of the liver, spleen, and pancreas was conducted on animals that tested positive by blood smears. Results: Interestingly, 26 out of 203 samples (12.08%) had confirmed Leucocytozoon infections based on microscopic analysis. Additionally, all 11 samples that initially tested positive via blood smears were confirmed positive through nested PCR analysis, and their sequencing revealed the presence of Leucocytozoon sabrazesi, marking the first report of this parasite in Egypt. The study into potential risk factors unveiled the prevalence of Leucocytozoon spp. seems host gender-dependent, with males exhibiting a significantly higher infection rate (33.33%). Additionally, adult birds demonstrated a significantly higher infection prevalence than squabs, suggesting an age-dependent trend in prevalence. Seasonality played a significant role, with the highest occurrence observed during summer (37.25%). Histopathological examination revealed the presence of numerous megaloschizonts accompanied by lymphocytic infiltration and multiple focal areas of ischemic necrosis. Conclusion: To our knowledge, this is the first study to shed light on the epidemiological characteristics and molecular characterization of leucocytozoonosis in pigeons in Egypt. Further research endeavors are warranted to curb the resurgence of Leucocytozoon parasites in other avian species across Egypt, thereby refining the epidemiological understanding of the disease for more effective control and prevention measures. The uptake, accumulation, and ontogenetic transfer of microplastics (MPs) in bloodsucking mosquitoes of the genus Anopheles, vectors of helminth and protozoa parasites in humans and animals, were studied under laboratory conditions. In the experiment, 2‐μm‐diameter fluorescent polystyrene spheres were counted in mosquitoes of all stages, from larvae to adults. Larvae from a natural population readily ingested MPs with food, accumulating on average 3.9 × 106 particles per larva within 3 days. The concentration of MPs decreased significantly in mosquitoes during their transition from stage to stage. The average number of detected MPs attained 110 particles per pupa and 3.0 particles per adult. MP uptake by mosquitoes did not affect their survival, whereas the rate of metamorphosis in MP‐consuming mosquitoes was slightly higher. Our data confirmed that MPs can be transferred from feeding larvae to non‐feeding pupae and adult flying Anopheles mosquitoes from natural populations in aquatic ecosystems. [ABSTRACT FROM AUTHOR]

Males and females often differ in ecology, behaviour and lifestyle, and these differences are expected to lead to sex differences in parasite susceptibility. However, neither the sex differences in parasite prevalence, nor their ecological and evolutionary drivers have been investigated across a broad range of taxa using phylogenetically corrected analyses. Using the most extensive dataset yet that includes 755 prevalence estimates from 151 wild bird species in a meta-analytic framework, here we compare sex differences in blood and gastrointestinal parasites. We show that despite sex differences in parasite infection being frequently reported in the literature, only Haemoproteus infections were more prevalent in females than in males. Notably, only seasonality was strongly associated with the sex-specific parasite prevalence of both Leucocytozoon and Haemoproteus, where birds showed greater female bias in prevalence during breeding periods compared to the non-breeding period. No other ecological or sexual selection variables were associated with sex-specific prevalence of parasite prevalence. We suggest that much of the variation in sex-biased prevalence could be idiosyncratic, and driven by local ecology and behavioural differences of the parasite and the host. The spread of parasites and the emergence of disease are currently threatening global biodiversity and human welfare. To address this threat, we need to better understand those factors that determine parasite persistence and prevalence. It is known that dispersal is central to the spatial dynamics of host–parasite systems. Yet past studies have typically assumed that dispersal is a species-level constant, despite a growing body of empirical evidence that dispersal varies with ecological context, including the risk of infection and aspects of host state such as infection status (parasite-dependent dispersal; PDD). Here, we develop a metapopulation model to understand how different forms of PDD shape the prevalence of a directly transmitted parasite. We show that increasing host dispersal rate can increase, decrease or cause a non-monotonic change in regional parasite prevalence, depending on the type of PDD and characteristics of the host–parasite system (transmission rate, virulence, and dispersal mortality). This result contrasts with previous studies with parasite-independent dispersal which concluded that prevalence increases with host dispersal rate. We argue that accounting for host dispersal responses to parasites is necessary for a complete understanding of host–parasite dynamics and for predicting how parasite prevalence will respond to changes such as human alteration of landscape connectivity. Rhoptries are specialized secretory organelles conserved across the Apicomplexa phylum, essential for host cell invasion and critical for subverting of host cellular and immune functions. They contain proteins and membranous materials injected directly into the host cells, participating in parasitophorous vacuole formation. Toxoplasma gondii tachyzoites harbor 8 to 12 rhoptries, 2 of which are docked to an apical vesicle (AV), a central element associated with a rhoptry secretory apparatus prior to injection into the host cell. This parasite is also equipped with 5 to 6 microtubule-associated vesicles, presumably serving as AV replenishment for iterative rhoptry discharge. Here, we characterized a rhoptry protein, rhoptry discharge factor 3 (RDF3), crucial for rhoptry discharge and invasion. RDF3 enters the secretory pathway, localizing near the AV and associated with the rhoptry bulb. Upon invasion, RDF3 dynamically delocalizes, suggesting a critical role at the time of rhoptry discharge. Cryo-electron tomography analysis of RDF3-depleted parasites reveals irregularity in microtubule-associated vesicles morphology, presumably impacting on their preparedness to function as an AV. Our findings suggest that RDF3 is priming the microtubule-associated vesicles for rhoptry discharge by a mechanism distinct from the rhoptry secretory apparatus contribution. Regulated secretion of rhoptry factors is essential for host cell invasion by Apicomplexan parasites. Phoxinus minnows are small freshwater fish found throughout Eurasia. The taxonomy of this genus is still under investigation, and new species are regularly described. They are frequently introduced outside their native range due to their use as live bait for angling, and such fish introductions can have diverse impacts on the recipient areas. Before the rise in interest regarding the diversity of this genus in the past 15 years, studies carried out on their biology and ecology reported their findings for the Eurasian minnow Phoxinus phoxinus Linnaeus, 1758, which was then considered ubiquitous. A parasites checklist for Phoxinus minnows has yet to be published, and the recent progress on the taxonomy of this genus has enabled us to propose a reassignment of parasite records to their proper host. The most thoroughly studied potential species were the North European species Phoxinus sp. 7 sensu Palandačić et al. 2017, Phoxinus isetensis, and P. phoxinus. We were able to highlight a gap in knowledge for numerous species that have received little‐to‐no attention in terms of parasitology. Systematic molecular identification of Phoxinus minnows should be conducted when studying their parasites, as a reliable identification of the host is vital in parasitology research. Future research will certainly reveal a species‐specific composition of the parasitofauna occurring in Phoxinus, especially among widespread parasites such as Gyrodactylus and Diplostomum. Background Refugees to the United States frequently have parasitic infections. If untreated, parasites can cause severe complications. The purpose of this study was to investigate the incidence, management, and outcomes of eosinophilia as a biomarker for parasites. Methods We conducted a retrospective longitudinal chart review of consecutive refugees attending 3 refugee clinics in Rhode Island that manage the health care of all pediatric and adult refugees. Results Among 812 refugees who met inclusion criteria, 147 (18.1%) had eosinophilia upon arrival and almost half had ≥1 symptom. The rates and severity of eosinophilia in those with predeparture presumptive treatment records who did (112/115, 97.4%) or did not (488/498, 98.0%) receive predeparture antiparasitic treatment were similar. All refugees with eosinophilia had ≥1 parasitic test in the United States. The most common attributable parasites were Schistosoma and Strongyloides stercoralis. Overall, parasites were detected in 63 (42.9%) of 147 refugees with eosinophilia by either stool testing, serology, or blood smear, but testing was inconsistent and likely underestimated true incidence. Only some of the identified parasites typically cause eosinophilia. Forty-five (30.6%) refugees with eosinophilia received antiparasitics in the United States. Of 81 (55.1%) individuals who had repeat blood tests, eosinophilia had resolved in 52 (64.2%). Five individuals (3.4%) had alternative diagnoses, including eczema, myelofibrosis, and drug allergy. Conclusions Our findings support Centers for Disease Control and Prevention recommendations to screen for eosinophilia in newly arrived refugees. Follow-up after 3–6 months is critical to confirm resolution of residual eosinophilia, which frequently occurs after effective predeparture treatment or if eosinophilia persists, to diagnose active parasitic infections. Predators negatively affect prey outside of direct attack, and these nonconsumptive effects (NCEs) may cause over half the impacts of predators on prey populations. This "ecology of fear" framework has been extended to host–parasite interactions. The NCEs of parasites are thought to be small relative to those of predators. However, recent research shows ectoparasites exert NCEs on multiple life stages of Drosophila. In this study, we apply recent data to a matrix‐based model of fly populations experiencing infection/consumption and NCEs from an ectoparasitic mite. We found the NCEs of parasites on larvae, which are not actively parasitized, decreased the size of simulated host populations. By contrast, the NCEs on adult flies increased population size through compensatory egg production. The negative NCEs on larvae outweighed the positive effects on adults to reduce population size. Climate change will alter interactions between parasites and their hosts. Warming may affect patterns of local adaptation, shifting the environment to favor the parasite or host and thus changing the prevalence of disease. We assessed local adaptation to hosts and temperature in the facultative ciliate parasite Lambornella clarki , which infects the western tree hole mosquito Aedes sierrensis. We conducted laboratory infection experiments with mosquito larvae and parasites collected from across a climate gradient, pairing sympatric or allopatric populations across three temperatures that were either matched or mismatched to the source environment. Lambornella clarki parasites were locally adapted to their hosts, with 2.6 times higher infection rates on sympatric populations compared with allopatric populations, but they were not locally adapted to temperature. Infection peaked at the intermediate temperature of 12.5°C, notably lower than the optimum temperature for free-living L. clarki growth, suggesting that the host's immune response can play a significant role in mediating the outcome of infection. Serres thought that humans had become the world's parasites and that we must seek a more reciprocal partnership with our host. He put forward a legal justification for writing a new social contract that encompassed the more-than-human. Serres associated the foundation of the "natural contract" with the story of evolution, the biological relation between symbiosis and the parasite. Closely aligned to his proposal, Serres also envisioned the gathering together of a universal history revealed by the knowledge of the diverse sciences of matter and life and stories from the humanities and religion. He thought the "Grand Narrative" would bring people to an intimate understanding of our relation to the living world and the need for a contract of symbiosis. Serres referred to Molière's Tartuffe, the fables of La Fontaine, the adventures of Tintin and Livy's history of Rome to describe an underlying parasitical logic. Crossing social, medical, communicative and historical practices, the parasite is portrayed by Serres as an acquisitive guest, a contagious microbe, a confusing noise and part of ritualistic feasts. Crossing these specific examples, Serres depicts how the general operation of the parasite works in close partnership with the symbiotic. For instance, germs can both invade the body and strengthen immunity. As evolutionary biologists recognise, there is no simple division between parasitic and symbiotic processes. In this paper, I explore the biological–parasitic–symbiotic framework that Serres applies in the context of the ecological crisis and his proposal for a contract of symbiosis. I focus on pioneering research into the history of bacteria carried out by microbiologist Lynn Margulis and associated studies on the evolution of fungi and moss. I describe how this research contributes to Serres's formulation of a universal story and at the same time helps both to supplement and critically engage with Serres's thought on the natural contract. Specifically, I describe how Margulis's contentions about the abilities of micro-organisms differ from Serres's conception of the technology of "nature." I also examine how his notion of the "Biosum," a body that integrates all the perceptions and senses of the living world, informs his understanding of the divergent evolution of humankind. I conclude that Serres's natural contract seeks to reinvent rather than imitate the rest of nature and briefly consider how his vision of inclusive global stories offers a glimpse of a new politics. The Natural Contract (1990) marks the emergence of the environmental question in Serres's work that became central in his later publications. Over the twenty years when the Anthropocene emerged as a distinct narrative entangling geological time with the history of human technology, Serres developed a rather similar "Grand Récit" embracing deep geological time, biological evolution, the process of hominization, and the future of humankind, although he did not enter into conversation with the leading figures of the interdisciplinary field of Anthropocene studies. This paper seeks to characterize Serres's position in the field of environmental philosophy as it has been reconfigured by the introduction of the concept of the Anthropocene. It first briefly surveys the personal pathway that led Serres to his diagnosis of a global climate crisis. It then compares the concepts that guided his analysis with the concepts debated in Anthropocene studies. The first records in the literature compiled on the parasitic fauna of amphibians in Northeastern Brazil date back to the 1990s. Since then, several new studies have been published on parasite-host relationships, parasite communities, and descriptions of new taxa. However, only in the last decade has there been a significant increase in these studies. Given this growth, we aim to provide a complete and updated compilation of helminth records associated with amphibians from the Brazilian Northeastern region and to analyse the dynamics and network structure formed between parasites and their hosts. Therefore, 33 studies were found in the specialized literature that addressed data from eight families, 15 genera, and 34 species of anuran amphibians, distributed mainly in areas of the morphoclimatic domain of the Caatinga and Atlantic Forest remnants in the Brazilian Northeast. These data correspond to 35% of the total known species of the Caatinga, with Leptodactylidae being the most representative taxon. Regarding helminths, 51 species were recorded, belonging to 20 families and 32 genera. To evaluate the structure of the network, we used measures of connectivity, nestedness, modularity, and centrality, that were considered to identify key species. The web presented 247 interactions with a highly connected structure formed by two parasite generalist species, non-nested and non-modular. We concluded that anuran amphibians from the Brazilian Northeast possess a high parasitic diversity, being Bufonidae and Leptodactylidade taxa considered fundamental for the network structure. Background: The burden of zoonotic diseases in developing countries is significantly underestimated, influenced by various factors such as misdiagnosis, underreporting, natural disasters, climate change, resource limitations, rapid unplanned urbanization, poverty, animal migration, travel, ecotourism, and the tropical environmental conditions prevalent in the region. Despite Sri Lanka's provision of a publicly funded free health care system, zoonoses still contribute significantly to the burden of communicable diseases in the country. This study serves as a timely and exhaustive systematic review of zoonoses reported over the past 22 years in Sri Lanka. Materials and Methods: This systematic review adhered to the guidelines provided by the "Preferred Reporting Items for Systematic Reviews and Meta-Analyses" (PRISMA) statement. A systematic literature search was conducted between July and September 2022, utilizing the following databases and sources: Google Scholar, PubMed, Cochrane Library, Weekly Epidemiological Reports, and Rabies Statistical Bulletins published by the Ministry of Health, Sri Lanka. Results: From the initial database search, 1,710 articles were identified. After excluding nonzoonotic diseases, duplicated reports, inaccessible articles, and those not meeting the inclusion criteria, 570 reports were evaluated for eligibility. Of these, 91 reports were selected for data extraction, comprising 58 original research articles, 10 case reports, 16 weekly epidemiological reports, and 7 rabies statistical bulletins. Over the study period (2000–2022), 14 parasitic, 7 bacterial, and 7 viral zoonoses have been reported in Sri Lanka. Notably, leptospirosis emerged as the most reported zoonotic disease in the country. Conclusions: In response to these findings, we strongly recommend the implementation of a tailored, country-specific prevention and control program. To achieve this goal effectively, we emphasize the importance of adopting a country-specific "One Health" approach as a comprehensive framework for managing and controlling zoonotic diseases in Sri Lanka. Although helminth parasites have different life cycles, their hosts share similar immune responses involving Th2 cell-type. Here, we extracted proteins from the larvae of Anisakis simplex complex and Trichinella spiralis to identify common and specific antigens (or allergens) associated with the Th2 immune response. We performed two-dimensional electrophoresis analysis and Matrix-assisted laser desorption ionization–time of flight/time of flight (MALDI-TOF/TOF) experiments. We found 13 potentially immunogenic proteins, which included 5 spots specific to T. spiralis and 8 common to T. spiralis and A. simplex, by tandem mass spectrometry. These molecules were identified structurally as actin, tropomyosin, col cuticle N domain-containing protein, and heat shock proteins. We also identified molecules related to parasite-host immune modulation and interactions. Simple Summary: Free-ranging wild boars are considered exotic animals and cause various ecological, economic and health-related damages in Brazil. With the increasing trend of outdoor pig farming, the chances of domestic animals coming into contact with wildlife have risen, leading to a greater risk of disease transmission. Studies on gastrointestinal parasites in wild boars in Brazil are scarce, so we investigated the occurrence of parasitic helminths in these animals. In our study, a total of 10 animals—5 males and 5 females of different ages—were examined. After these animals were culled by control and hunting teams, their digestive systems were collected and examined. A total of 2750 helminths were collected from the small intestine of all animals, and after evaluating their morphologies, we identified them as Globocephalus urosubulatus. The morphological characteristics observed in the nematodes correspond to those previously described; however, we found that the copulatory bursa was asymmetrical, a characteristic that has not yet been recorded in the literature. Although previous studies have characterized the helminth fauna of wild boars kept in captivity in Brazil, records on these helminths in free-ranging animals are still scarce. In view of this, we aimed in our work to investigate the occurrence and morphological and morphometric characteristics of gastrointestinal helminths in wild Sus scrofa from the northwest region of the State of São Paulo, Brazil. The digestive systems of 10 animals (5 males and 5 females of different ages) were used in this study. Each anatomical segment was washed and sieved under running water, and the helminths were separated and identified using light and scanning electron microscopy, according to their morphological characteristics. A total of 2750 (1152 males and 1598 females) nematode specimens were collected from the small intestine of these wild boars, and all of them presented the morphological characteristics of Globocephalus urosubulatus. However, one characteristic is of particular interest because it has not yet been reported in the literature: a marked asymmetry between the lobes and their respective rays of the copulatory bursa, with the left one being larger than the right one. In this research, we identified the presence of G. urosubulatus in all the examined free-ranging wild boars and reported for the first time in the literature the asymmetry in the copulatory bursa. In the marine environment, fish parasites are present in most seafood species. The most common are nematodes of the genus Anisakis, which can parasitize human tissues, causing anisakiasis and allergies—in some cases with a strong reaction, such as anaphylactic shock. This happens when people ingest live or dead larvae present in the muscles or viscera of a wide range of fish and cephalopods. Consumer education has been positioned as one of the most effective alternatives for its prevention. This study, carried out in Asturias (northwest Spain), sought to identify the seafood products that present the greatest risk of anisakiasis for consumers, taking into account their consumption, the prevalence of Anisakis, and consumer knowledge about this parasitosis. In the results, hake (Merluccius merluccius) and cod (Gadus morhua), frequently consumed in the region and with high parasite prevalence, do not pose a great risk because they are consumed when well cooked. Instead, sardine (Sardina pilchardus), highly consumed and less parasitized, and anchovy (Engraulis encrasicolus), highly parasitized and less consumed, would exhibit a medium risk. Young participants know more about the risks of anisakiasis from raw seafood. The gaps detected in the knowledge about the ability of temperature treatments to eliminate parasites, especially in allergic people, must be addressed for better prevention. Parasitic diseases can affect animal health and welfare, and they may also constitute a danger to public health, particularly in island ecosystems. Fecal samples were collected from 205 dogs and 115 cats on the islands of São Miguel and Terceira, Azores archipelago (Portugal), using the Willis flotation technique and modified Baermann method, for further analysis. The overall prevalence of gastrointestinal parasitism in dogs was 53%, with the following results: Ancylostomatidae (hookworms) (42.44%), Trichuris vulpis (17.56%), Toxocara canis (12.68%) and Cystoisospora spp. (4.39%). In cats, the overall prevalence was also 53%, with the following results: Toxocara cati (31.3%), Ancylostomatidae (30.43%), Cystoisospora spp. (14.78%) and Trichuris sp. (0.87%). The prevalence of lungworms was 0.49% in canines and 20.87% in felines, with Angiostrongylus vasorum and Aelurostrongylus abstrusus species being detected in dogs and cats, respectively. The present survey detected a high prevalence of gastrointestinal infection, in both dogs and cats, probably because the samples came mainly from kennels and catteries and due to the peculiar climatic conditions in this insular territory, with mild temperature and high relative humidity. A considerable prevalence of aelurostrongylosis was also detected (20.87%), so it should be included in the list of differential diagnoses of diseases concerning the respiratory tract in cats of the archipelago. Exosomes are membrane-bound structures released by cells into the external environment that carry a significant amount of important cargo, such as proteins, DNA, RNA, and lipids. They play a crucial role in intercellular communication. Parasites have complex life cycles and can release exosomes at different stages. Exosomes released by parasitic pathogens or infected cells contain parasitic nucleic acids, antigenic molecules, virulence factors, drug-resistant proteins, proteases, lipids, etc. These components can regulate host gene expression across species or modulate signaling pathways, thereby dampening or activating host immune responses, causing pathological damage, and participating in disease progression. This review focuses on the means by which parasitic exosomes modulate host immune responses, elaborates on the pathogenic mechanisms of parasites, clarifies the interactions between parasites and hosts, and provides a theoretical basis and research directions for the prevention and treatment of parasitic diseases. Ladybirds (Coleoptera: Coccinellidae) are a significant predator group that plays a role in agricultural contexts. They serve as important biocontrol agents against several agricultural pests. However, there is limited information available about how ladybirds' populations are regulated by their natural enemies and habitat perturbation. In this study, we evaluated ladybird communities associated with natural and managed areas during the agricultural cycle of 2018 (June–September). We identified seven ladybird species; the most abundant species were Hippodamia convergens (70% individuals) and Cycloneda sanguinea (20% individuals), followed by the exotic species Harmonia axyridis (3.33% individuals), while Paranaemia vittigera, Coleomegilla maculata, Brachiacantha sp., and Olla v-nigrum represented less than 3% each. Ladybirds were more abundant in agricultural fields than in natural habitats. We identified several natural enemies associated with the collected ladybirds, including parasitoids (Dinocampus coccinellae, Hymenoptera: Braconidae), ectoparasitic Acari (Coccipolipus sp., Podapolipidae), and nematodes (Mermithidae). However, none of the natural enemies affected more than 10% of the ladybirds, so natural enemies do not appear to be exerting substantial control over their populations. Our results corroborated that ladybirds can efficiently thrive in anthropogenic habitats in Mexico and therefore may represent good biocontrol agents for agriculture. It is important to consider the ecology of agriculturally important ladybirds in programs for integrated pest management. The role of species interactions in setting species range limits is rarely empirically explored. Here, we quantify host and parasite densities in subtropical eastern Australia (26.65°–30.20°S) to examine whether parasitism might contribute to range limitation of Acropora corals at their cold-range boundary. 79% of Acropora corals had endolithic barnacles (family Pyrgomatidae), with higher parasite load in larger corals and up to 141 barnacles per coral. Parasite load increased poleward and closer to the mainland and was greater in cooler and high nutrient environments. Parasite burden was higher at sites with fewer Acropora corals, broadly consistent with the hypothesis that parasites can fragment host populations where host densities are low, and the parasite is a better disperser than the host. Whilst the mechanism is unclear, our findings suggest that at the high densities recorded here, coral-barnacles could influence range dynamics of Acropora corals at their poleward range limit. Background: Gastrointestinal helminths are a very widespread group of intestinal parasites that can cause major health issues in their hosts, including severe illness or death. Traditional methods of helminth parasite identification using microscopy are time-consuming and poor in terms of taxonomic resolution, and require skilled observers. DNA metabarcoding has emerged as a powerful alternative for assessing community composition in a variety of sample types over the last few decades. While metabarcoding approaches have been reviewed for use in other research areas, the use of metabarcoding for parasites has only recently become widespread. As such, there is a need to synthesize parasite metabarcoding methodology and highlight the considerations to be taken into account when developing a protocol. Methods: We reviewed published literature that utilized DNA metabarcoding to identify gastrointestinal helminth parasites in vertebrate hosts. We extracted information from 62 peer-reviewed papers published between 2014 and 2023 and created a stepwise guide to the metabarcoding process. Results: We found that studies in our review varied in technique and methodology, such as the sample type utilized, genetic marker regions targeted and bioinformatic databases used. The main limitations of metabarcoding are that parasite abundance data may not be reliably attained from sequence read numbers, metabarcoding data may not be representative of the species present in the host and the cost and bioinformatic expertise required to utilize this method may be prohibitive to some groups. Conclusions: Overall, using metabarcoding to assess gastrointestinal parasite communities is preferable to traditional methods, yielding higher taxonomic resolution, higher throughput and increased versatility due to its utility in any geographical location, with a variety of sample types, and with virtually any vertebrate host species. Livestock production in the Greater Mekong Subregion (GMS) reflects the inefficient smallholder 'keeper' system that has been slow to adapt to the rapidly expanding demand for animal protein-sourced foods in the region as urban economies have flourished and food preferences altered. The prolonged surge in demand, with only modest increases in local production, has increased the movement of animals and products into and from the GMS, accompanied by surging risks of transboundary animal disease (TAD) incursions, including the one health (OH) threats of zoonoses and antimicrobial resistance. As a consequence, the region has been subjected to epidemics of highly pathogenic avian influenza (HPAI), new strains of foot-and-mouth disease (FMD) virus, with recent incursions of African swine fever (ASF), lumpy skin disease (LSD), porcine reproductive and respiratory syndrome (PRRS) and risks of peste petits ruminants (OPR) occurring in Southeast Asia (SEA) and beyond. These incidents reflect inadequate biosecurity, a sustainability issue that was clearly of relevance in the covid-19 pandemic, reflecting ecosystem health (EH) deficits, including land-use issues and unregulated trading in wildlife-sourced and poorly processed foods through the predominant 'wet markets'. These challenges are increasingly confounded by slow adaption to the impacts of the climate crisis, including flooding, drought, crop failures and hypothermia episodes. The increase in animal and product movements enhances disease transmission risk, yet coincides with emerging concerns of greenhouse-gas emissions (GHGe) from livestock production, especially from large ruminants, as the world attempts to find pathways in managing the climate crisis. Despite the prolonged collaborative efforts of the SEA China FMD program from 1997 to 2023, a recent review confirmed persistent deficits in biosecurity, vaccine resourcing, disease surveillance, engagement of farmers, and national emergency disease-response capacities. A major project is about to fund major improvements in livestock value chains in Cambodia, including more effective biosecurity, surveillance and emergency disease-response capacities for TADs, antimicrobial resistance (AMR) and zoonoses. Similarly, a private-sector investment in Laos has developed a more climate-resilient livestock-feeding system that decreases GHGe impacts from ruminant production. These developments are likely to extend beyond both countries and be potentially transformational for the livelihoods of many of the poorest citizens in the region. Increased movement of livestock and products in the Greater Mekong Subregion (GMS) has enhanced disease transmission risk, increasing transboundary animal-disease (TAD) incursions and one health (OH) concerns. This has coincided with increasing awareness of greenhouse-gas emissions (GHGe) from livestock, especially contributions to ecosystem health (EH) concerns from large ruminants. Projects addressing biosecurity, vaccines, disease surveillance, feeding systems and national emergency disease-response capacities are required, and private-sector investments developing climate-resilient livestock systems to support the livelihoods of poor rural communities in the GMS should be encouraged. Background: Soybean cyst nematodes (SCN) as animal parasites of plants are not usually interested in killing the host but are rather focused on completing their life cycle to increase population, resulting in substantial yield losses. Remarkably, some agricultural soils after long-term crop monoculture show a significant decline in SCN densities and suppress disease in a sustainable and viable manner. However, relatively little is known about the microbes and mechanisms operating against SCN in such disease-suppressive soils. Results: Greenhouse experiments showed that suppressive soils (S) collected from two provinces of China and transplantation soils (CS, created by mixing 10% S with 90% conducive soils) suppressed SCN. However, SCN suppressiveness was partially lost or completely abolished when S soils were treated with heat (80 °C) and formalin. Bacterial community analysis revealed that the specific suppression in S and CS was mainly associated with the bacterial phylum Bacteroidetes, specifically due to the enrichment of Chitinophaga spp. and Dyadobacter sp., in the cysts. SCN cysts colonized by Chitinophaga spp. showed dramatically reduced egg hatching, with unrecognizable internal body organization of juveniles inside the eggshell due to chitinase activity. Whereas, Dyadobacter sp. cells attached to the surface coat of J2s increased soybean resistance against SCN by triggering the expression of defence-associated genes. The disease-suppressive potential of these bacteria was validated by inoculating them into conducive soil. The Dyadobacter strain alone or in combination with Chitinophaga strains significantly decreased egg densities after one growing cycle of soybeans. In contrast, Chitinophaga strains alone required more than one growing cycle to significantly reduce SCN egg hatching and population density. Conclusion: This study revealed how soybean monoculture for decades induced microbiota homeostasis, leading to the formation of SCN-suppressive soil. The high relative abundance of antagonistic bacteria in the cyst suppressed the SCN population both directly and indirectly. Because uncontrolled proliferation will likely lead to quick demise due to host population collapse, obligate parasites like SCN may have evolved to modulate virulence/proliferation to balance these conflicting needs. Folk plants are the key starting point for antimalarial drug discovery. After crushing and mixing the leaves of Coriandrum sativum with water, one cup of tea is drunk daily for a duration of three to five days as a remedy for malaria by local folks in Ethiopia. Additionally, in vitro experiments conducted on the plant leaf extract elsewhere have also demonstrated the plant's malaria parasite inhibitory effect. There has been no pharmacologic research to assert this endowment in animals, though. This experiment was aimed at evaluating the antimalarial efficacy of C. sativum in Plasmodium berghei infected mice. Methods: The plant's leaf was extracted using maceration with distilled water. The extract was examined for potential acute toxicity. An evaluation of secondary phytoconstituents was done. Standard antimalarial screening models (prophylactic, chemosuppressive, curative tests) were utilized to assess the antiplasmodial effect. In each test, thirty mice were organized into groups of five. To the three categories, the test substance was given at doses of 100, 200 and 400 mg/kg/day before or after the commencement of P. berghei infection. Positive and negative control mice were provided Chloroquine and distilled water, respectively. Rectal temperature, parasitemia, body weight, survival time and packed cell volume were ultimately assessed. Analysis of the data was performed using Statistical Package for Social Sciences. Results: No toxicity was manifested in mice. The extract demonstrated a significant inhibition of parasitemia (p < 0.05) in all the models. The inhibition of parasite load was highest with the upper dose in the suppressive test (82.74%) followed by the curative procedure (78.49%). Likewise, inhibition of hypothermia, weight loss hampering, improved survival and protection against hemolysis were elicited by the extract. Conclusions: The results of our experimental study revealed that the aqueous crude leaf extract of C. sativum exhibits significant antimalarial efficacy in multiple in vivo models involving mice infected with P. berghei. ABSTRACT The Anthropocene is seeing the human footprint rapidly spreading to all of Earth's ecosystems. The fast‐changing biotic and abiotic conditions experienced by all organisms are exerting new and strong selective pressures, and there is a growing list of examples of human‐induced evolution in response to anthropogenic impacts. No organism is exempt from these novel selective pressures. Here, we synthesise current knowledge on human‐induced evolution in eukaryotic parasites of animals, and present a multidisciplinary framework for its study and monitoring. Parasites generally have short generation times and huge fecundity, features that predispose them for rapid evolution. We begin by reviewing evidence that parasites often have substantial standing genetic variation, and examples of their rapid evolution both under conditions of livestock production and in serial passage experiments. We then present a two‐step conceptual overview of the causal chain linking anthropogenic impacts to parasite evolution. First, we review the major anthropogenic factors impacting parasites, and identify the selective pressures they exert on parasites through increased mortality of either infective stages or adult parasites, or through changes in host density, quality or immunity. Second, we discuss what new phenotypic traits are likely to be favoured by the new selective pressures resulting from altered parasite mortality or host changes; we focus mostly on parasite virulence and basic life‐history traits, as these most directly influence the transmission success of parasites and the pathology they induce. To illustrate the kinds of evolutionary changes in parasites anticipated in the Anthropocene, we present a few scenarios, either already documented or hypothetical but plausible, involving parasite taxa in livestock, aquaculture and natural systems. Finally, we offer several approaches for investigations and real‐time monitoring of rapid, human‐induced evolution in parasites, ranging from controlled experiments to the use of state‐of‐the‐art genomic tools. The implications of fast‐evolving parasites in the Anthropocene for disease emergence and the dynamics of infections in domestic animals and wildlife are concerning. Broader recognition that it is not only the conditions for parasite transmission that are changing, but the parasites themselves, is needed to meet better the challenges ahead. Introduction: Parasite-mediated selection is considered one of the potential mechanisms contributing to the coexistence of asexual-sexual complexes. Gibel carp (Carassius gibelio), an invasive fish species in Europe, often forms populations composed of gynogenetic and sexual specimens. Methods: The experimental infection was induced in gynogenetic and sexual gibel carp using eye-fluke Diplostomum pseudospathaceum (Trematoda), and the transcriptome profile of the spleen as a major immune organ in fish was analyzed to reveal the differentially expressed immunity-associated genes related to D. pseudospathaceum infection differing between gynogenetic and sexual gibel carp. Results: High parasite infection was found in gynogenetic fish when compared to genetically diverse sexuals. Although metacercariae of D. pseudospathaceum are situated in an immune-privileged organ, our results show that eye trematodes may induce a host immune response. We found differential gene expression induced by eye-fluke infection, with various impacts on gynogenetic and sexual hosts, documenting for the majority of DEGs upregulation in sexuals, and downregulation in asexuals. Differences in gene regulation between gynogenetic and sexual gibel carp were evidenced in many immunity-associated genes. GO analyses revealed the importance of genes assigned to the GO terms: immune function, the Notch signaling pathway, MAP kinase tyrosine/threonine/phosphatase activity, and chemokine receptor activity. KEGG analyses revealed the importance of the genes involved in 12 immunity-associated pathways - specifically, FoxO signaling, adipocytokine signaling, TGF-beta signaling, apoptosis, Notch signaling, C-type lectin receptor signaling, efferocytosis, intestinal immune network for IgA production, insulin signaling, virion - human immunodeficiency virus, Toll-like receptor signaling, and phosphatidylinositol signaling system. Discussion: Our study indicates the limited potential of asexual fish to cope with higher parasite infection (likely a loss of capacity to induce an effective immune response) and highlights the important role of molecular mechanisms associated with immunity for the coexistence of gynogenetic and sexual gibel carp, potentially contributing to its invasiveness. P21 is a protein secreted by all forms of Trypanosoma cruzi (T. cruzi) with recognized biological activities determined in studies using the recombinant form of the protein. In our recent study, we found that the ablation of P21 gene decreased Y strain axenic epimastigotes multiplication and increased intracellular replication of amastigotes in HeLa cells infected with metacyclic trypomastigotes. In the present study, we investigated the effect of P21 in vitro using C2C12 cell lines infected with tissue culture-derived trypomastigotes (TCT) of wild-type and P21 knockout (TcP21-/-) Y strain, and in vivo using an experimental model of T. cruzi infection in BALB/c mice. Our in-vitro results showed a significant decrease in the host cell invasion rate by TcP21-/- parasites as measured by Giemsa staining and cell count in bright light microscope. Quantitative polymerase chain reaction (qPCR) analysis showed that TcP21-/- parasites multiplied intracellularly to a higher extent than the scrambled parasites at 72h post-infection. In addition, we observed a higher egress of TcP21-/- trypomastigotes from C2C12 cells at 144h and 168h post-infection. Mice infected with Y strain TcP21-/- trypomastigotes displayed higher systemic parasitemia, heart tissue parasite burden, and several histopathological alterations in heart tissues compared to control animals infected with scrambled parasites. Therewith, we propose that P21 is important in the host-pathogen interaction during invasion, cell multiplication, and egress, and may be part of the mechanism that controls parasitism and promotes chronic infection without patent systemic parasitemia. Eustrongylides excisus is a fish-borne zoonotic parasite known to infect various fish species, including Northern pike (Esox Lucius). This nematode, belonging to the family Dioctophymatidae, has a complex life cycle involving multiple hosts. This study aimed to investigate the occurrence of Eustrongylides nematodes in Northern pike (E. Lucius) collected from Mijran Dam (Ramsar, Iran). Between June and October 2023, an investigation was conducted on Northern pike from Mijran Dam in Ramsar, Iran, following reports of reddish parasites in their muscle tissues. Sixty fish were examined at the University of Tehran, revealing live parasites in the muscles, which were then analyzed microscopically and preserved for a multidisciplinary study. The skeletal muscle tissues of 85% (51/60) of fish specimens were infected by grossly visible larvae which were microscopically identified as Eustrongylides spp. In histopathological examination, the lesion was composed of encapsulated parasitic granulomatous myositis. Microscopically, the cystic parasitic granulomas compressed the adjacent muscle fibers, leading to their atrophy and Zenker's necrosis. Moreover, epithelioid macrophages, giant cells and mononuclear inflammatory cells were present around the larvae and between the muscle fibers. Finally, a molecular analysis by examining the ITS gene region, revealed that they belong to the species E. excisus. Eustrongylidiasis in northern Iran necessitates further research into the biology, epidemiology, and control of Eustrongylides nematodes, focusing on various hosts. This study is the first to comprehensively characterize E. excisus in Northern pike in Ramsar, Iran, raising concerns about possible zoonotic transmission. [ABSTRACT FROM AUTHOR]

Introduction: Children with intellectual disability (ID) often face challenges in maintaining proper oral hygiene due to their motor, sensory, and intellectual impairments, which can lead to compromised oral health; therefore, there is a need to enhance the oral health status of these populations and establish an effective system for administering preventive interventions. Here, we aimed to evaluate the prevalence of Entamoeba gingivalis and Trichomonas tenax among children with ID in Lorestan province, in Western Iran through parasitological and molecular methods. Methods: The current descriptive investigation involved 215 in children with ID and 215 healthy children (non-ID) who were referred to health facilities in Lorestan province, Iran between October 2022 and March 2024. The prevalence of protozoa in the oral cavity was found through the utilization of both microscopic analysis and conventional polymerase chain reaction (PCR) techniques. Results: The total prevalence of the E. gingivalis and T. tenax in children with ID was found to be 87 (40.5%) and 92 (42.8%) through microscopic and PCR methods, respectively. Among the positive samples, 57 (61.9%) and 35 (38.1%) children tested positive for E. gingivalis and T. tenax, respectively. In contrast, among the 215 non-ID children in the control group, 39 (18.1%) and 42 (19.5%) tested positive by microscopic and PCR methods, respectively. Among positive samples in non-ID children, 23 (54.7%) and 19 (45.3%) children were positive for E. gingivalis and T. tenax, respectively. Multiple logistic regression analysis indicated that residing in urban areas, parental education, monthly family income, and tooth brushing p<0.001) were identified as independent risk factors for oral cavity parasites. Conclusion: This study identified a notable prevalence of oral cavity parasites in children with ID in Lorestan province, Western Iran. It is imperative to recognize the primary risk factors associated with these parasites, particularly inadequate teeth brushing, in order to enhance public and oral health strategies for children with ID. Therefore, pediatric dental professionals should remain vigilant regarding these risk factors to effectively recognize and address oral health issues in this population, thereby mitigating the occurrence of oral diseases and infections. A prospective study was conducted from September 2022 to March 2023 to examine the occurrence and contamination rates of different parasitic stages on fresh edible fruits and vegetables in local markets of Erbil city. Eight different types of fruits and vegetables were selected from eight markets, and 144 samples were analyzed for parasites using direct sedimentation, indirect floatation (ZnSO4), Kinyoun, and trichrome staining methods. The highest total frequencies of parasites were detected using the direct sedimentation method (18.75%), followed by the indirect floatation method (6.25%), Kinyoun technique (5.55%), and modified trichrome staining (4.16%). A. lumbricoides (Ova) was detected with 4.16% using direct sedimentation, while no positive detections were observed with other techniques. Kinyoun technique identified Cryptosporidium spp. oocysts. For E. histolytica 1.3% were positive using the direct sedimentation method, while the other three methods showed no positive detections. Similar results were observed for G. lamblia where only the modified trichrome staining technique detected 0.69% with trophozoites and 1.3% with cysts. Additionally, the direct sedimentation method revealed the presence of nematodes at 2.77%, but no positive detections were made with other methods. Concerning Taeniid eggs, the indirect floatation identified 0.69% with ova, while other methods showed no positive detections. Lastly, the direct sedimentation method yielded 6.25% with unidentified flagellate protozoa, but no positive detections were found using the indirect floatation method, Kinyoun technique, or modified trichrome staining technique. Several factors influence whether an organism remains in its local habitat. Parasites can, for example, influence host movement by impacting their behavior, physiology, and morphology. In rivers, fish that swim efficiently against the current are able to maintain their position without being displaced downstream, a behavior referred to as positive rheotaxis. We hypothesized that both the presence and number of ectoparasites on a host would affect the ability of fish to avoid downstream displacement and thus prevent them from remaining in their habitat. We used the guppy‐Gyrodactylus host‐ectoparasite model to test whether parasite presence and parasite load had an effect on fish rheotaxis. We quantified rheotaxis of sham‐infected and parasite‐infected fish in a circular flow tank in the laboratory prior to infection and 5–6 days postinfection. Both parasite‐infected and sham‐infected individuals expressed similar levels of positive rheotaxis prior to infection and after infection. However, with increasing parasite numbers, guppies covered less distance in the upstream direction and spent more time in slower flow zones. These results suggest that higher numbers of Gyrodactylus ectoparasites negatively influence rheotactic movements. We searched for common patterns in parasite ecology by investigating species and host contributions to the beta-diversity of infracommunities (=assemblages of parasites harboured by a host individual) in helminths of three species of South African ungulates and fleas of 11 species of South American rodents, assuming that a comparison of patterns in distinctly different parasites and hosts would allow us to judge the generality or, at least, commonness of these patterns. We used data on species' composition and numbers of parasites and asked whether (i) parasite species' attributes (life cycle, transmission mode, and host specificity in helminths; possession of sclerotized combs, microhabitat preference, and host specificity in fleas) or their population structure (mean abundance and/or prevalence) and (ii) host characteristics (sex and age) affect parasite and host species' contributions to parasite beta-diversity (SCBD and HCBD, respectively). We found that parasite species' morphological and ecological attributes were mostly not associated with their SCBD. In contrast, parasite SCBD, in both ungulates and rodents, significantly increased with either parasite mean abundance or prevalence or both. The effect of host characteristics on HCBD was detected in a few hosts only. In general, parasite infracommunities' beta-diversity appeared to be driven by variation in parasite species rather than the uniqueness of the assemblages harboured by individual hosts. We conclude that some ecological patterns (such as the relationships between SCBD and parasite abundance/prevalence) appear to be common and do not differ between different host-parasite associations in different geographic regions, whereas other patterns (the relationships between SCBD and parasite species' attributes) are contingent and depend on parasite and host identities. Migratory movement shapes infectious disease dynamics within migratory populations, but also impacts the disease dynamics of resident (non-migratory) species by transporting pathogens along migratory routes. Migratory animals often dominate local animal biomass and exert strong trophic effects on the resident species that they encounter. Many of these trophic effects could impact infectious disease transmission, yet very little is known about the role of migrant trophic effects in shaping resident disease dynamics. We propose mechanisms via which migrant trophic effects change the local infection risk for resident animals. We then describe a framework that captures the role of migration intensity and duration in modifying migrant transport and trophic effects that can be used to investigate the complex ecological interactions that determine resident parasite dynamics in nature. Migratory animals can bring parasites into resident animal (i.e., non-migratory) home ranges (transport effects) and exert trophic effects that either promote or reduce parasite exposure to resident hosts. Here, we examine the importance of these transport and trophic effects and their interactions for resident parasite dynamics. We propose that migrant transport and trophic effects are impacted by the number of migratory animals entering a resident's home range (migration intensity), the amount of time that migrants spend within a resident's home range (migration duration), and the timing of migrant–resident interactions. We then incorporate migration intensity, duration, and timing into a framework for exploring the net impact of migrant trophic and transport effects on resident animal parasite prevalence. Organisms are exposed to pathogens that cause infections, and consequently, they have developed complex physiological responses that allow them to resist or eliminate such threats; this set of strategies constitutes the immune system. Hosts exhibit great variation in the magnitude and efficiency of their immune responses. Ecoimmunology is an emerging discipline that seeks to understand the intrinsic and extrinsic factors that modulate this variation in immune response and, therefore, susceptibility to diseases across a wide range of species and habitats. On one hand, there is ample evidence that resistance to pathogens and parasites is under genetic control. On the other hand, since immune function involves high costs (e.g., energy, nutritional), it is expected that trade-offs arise between it and other costly physiological processes for the organism. In this review, the main patterns of variation in the major histocompatibility complex (MHC) and its relationship with pathogen resistance in the subterranean rodent Ctenomys talarum (tuco-tucos) are presented, as well as the role of these genes in the context of mate selection. Additionally, evidence is presented for the existence of energy and nutritional costs of different arms of the immune response in this species, as well as the trade-offs established between immune function and other physiological processes, such as reproduction. Finally, the roles of habitat and life history traits in shaping the variation of the immune response in tuco-tucos are discussed, along with the relevance of ecoimmunological studies in wild mammals.
