Abstract

Malnutrition affects growth and development in humans and causes socio-economic losses. Normal maize is deficient in essential amino acids, lysine and tryptophan; and vitamin-A. Crop biofortification is a sustainable and economical approach to alleviate micronutrient malnutrition. We combined favorable alleles of crtRB1 and lcyE genes into opaque2 (o2)-based four inbreds viz. QLM11, QLM12, QLM13, and QLM14 using marker-assisted backcross breeding. These are parents of quality protein maize versions of two elite hybrids viz. Buland and PMH1, grown in India. Gene-based SSRs for o2 and InDel markers for crtRB1 and lcyE were successfully employed for foreground selection in BC1F1, BC2F1, and BC2F2 generations. The recurrent parent genome recovery ranged from 88.9 to 96.0% among introgressed progenies. Kernels of pyramided lines possessed a high concentration of proA (7.14–9.63 ppm), compared to 1.05 to 1.41 ppm in the recurrent parents, while lysine and tryptophan ranged from 0.28–0.44% and 0.07–0.09%, respectively. The reconstituted hybrids (RBuland and RPMH1) showed significant enhancement of endosperm proA (6.97–9.82 ppm), tryptophan (0.07–0.09%), and lysine (0.29–0.43%), while grain yield was at par with their original versions. The dissemination of reconstituted hybrids holds significant promise to alleviate vitamin-A deficiency and protein-energy malnutrition in developing countries.

Abstract

Sugarcane is an economically important crop and is cultivated throughout the world for its sugar and ethanol. Present sugarcane varieties are derived from inter-specific hybridization between Saccharum officinarum and Saccharum spontaneum, resulting in highly polyploid and aneuploid plants with chromosomes ranging from 80 to 120. This complex nature has hindered advances in marker assisted breeding of this crop. Though an array of methodologies have been adopted by different research groups from all over the world who have well established protocols for developing genetically modified (GM) sugarcane. Several genes of economic interest have been transformed in sugarcane, despite variable success in transgene integration and expression. Recently, genome editing approaches (the zinc finger nucleases (ZFNs), transcription activator-like effector nucleases (TALENs) and CRISPR/Cas9 system) have opened new and unique opportunities for researchers to enhance crop production. However, as far as sugarcane is considered, there are still several challenges/constraints (large genome size, polyploidy, low transformation efficiency, transgene silencing and lack of high throughput screening techniques) ahead that deter sugarcane improvement through genetic engineering and need to be resolved to achieve a complete exploitation of the crop through these biotechnological tools. This review is an attempt to unravel the various sugarcane genomic approaches and their applications towards accelerating sugarcane improvement.

Abstract

Sugarcane propagation through conventional means does not provide sufficient planting material of a variety, particularly desirable in case of newly released varieties to achieve large-scale dissemination; this is attributed to slow rate of seed multiplication by conventional sett planting. On the other hand, micropropagation technique of tissue culture ensures production of disease-free and true-to-type planting material of popular (new as well as old) varieties in an abundant quantity in a short period of time. The cultures of meristematic buds or spindle leaves, collected from healthy plants, are established aseptically under controlled nutritional and environmental conditions in vitro, followed by multiplication of shoots and induction of roots; the plantlets are hardened and supplied to growers. Somatic embryogenesis is the process of embryo formation and development from somatic cells of an explant under in vitro conditions. The somatic cells in culture can follow two pathways for somatic embryogenesis, either direct or indirect. The plants regenerated through direct somatic embryogenesis are often uniform; thus, the pathway finds use in clonal propagation and genetic transformation of sugarcane genotypes. In indirect somatic embryogenesis pathway, first callus is induced from cultured explants under the influence of an auxin (mostly 2, 4-D) which is then regenerated into plants; such plants may exhibit somaclonal variation.

4. Direct plant regeneration from cultured young leaf segments of sugarcane

Abstract

Young leaf segments (1.0–1.5 cm) excised from spindle explants of three commercial sugarcane varieties viz. Co J 64, Co J 83 and Co J 86 were cultured on different media compositions based on Murashige and Skoog (1962) salts. Cultured explants exhibited swelling followed by direct shoot regeneration on media containing naphthaleneacetic acid, in all the three varieties. Highest frequency 83.12% shoot regeneration occurred on Murashige and Skoog medium supplemented with naphthaleneacetic acid (5.0 mg l−1) and kinetin (0.5 mg l−1) in variety Co J 83. Medium devoid of naphthaleneacetic acid and supplemented with only kinetin did not induce direct shoot regeneration in any of the varieties thus tried. Subsequently profuse rooting of shoots was observed on the same medium and complete plantlets were recovered within 6 weeks. The plantlets were hardened and transferred to soil. Tissue culture derived field-grown plants were normal and exhibited faster growth and better tillering. This developed single step method of direct plant regeneration can be used for rapid mass cloning and genetic transformation of sugarcane.

Abstract

Decision Support Systems are usually computer applications along with a human component that can sift through large amounts of data and pick between many choices. Among many challenges in front of research managers, checking of duplication of research efforts is a major challenge. Existing systems do provide simple keyword base search wherein one can find similar projects. These systems do not take care of the semantics or sense of the word into consideration. Also, these systems fail to provide results if synonyms of the keywords are used in other research projects. Moreover, if the keywords are matched and are not used in the same sense, it is very much likely that the projects may not be similar. The presented approach in this work uses WordNet Ontology for finding the semantics of the textual information available within the system. Two algorithms viz. Filtered Set and Preferential Coefficient are presented that also take care the structure of the research project proposal. The algorithms are implemented for the PIMS-ICAR database that contains more than 5000 research project documents.

Abstract

The chemical compatibility of wave length shifting fibers with several liquid scintillators has been investigated. Based on systematic characterization of the behavior of the BC-517 family, a time of life of 70–450 years was estimated for the polystyrene based wave length shifting fiber in BC-517P scintillator. Wavelength shifting (WLS) fibers irradiated continuously to a dose of 6.4 Mrads (at .377 Mrad/hr of 60 Co) were observed to decrease from 100% to 5% transmission; however, after 100 hours of annealing, the transmission increased to 90%. GEANT3 simulations of a simplified calorimeter located behind a BaF 2 electromagnetic calorimeter for the GEM detector at SSC showed that the constant term in the energy resolution will change from 1.8 to 2.9 in five years at 10 34 luminosity for pseudorapidity η=3.

Abstract

We have observed B0-B0 mixing in Z0→bb decays using hadronic events containing dileptons. The data sample corresponds to 118 200 hadron events at √s≈Mz. From a fit to the dilepton p and p⊥ spectra, we determine the mixing parameter to be χB=0.178-0.040 +0.049. © 1990.

Abstract

The L3 experiment is one of the six large detectors designed for the new generation of electron-positron accelerators. It is the only detector that concentrates its efforts on limited goals of measuring electrons, muons and photons. By not attempting to identify hadrons, L3 has been able to provide an order of magnitude better resolution for electrons, muons and photons. Vertices and hadron jets are also studied. The construction of L3 has involved much state of the art technology in new principles of vertex detection and in new crystals for large scale electromagnetic shower detection and ultraprecise muon detection. This paper presents a summary of the construction of L3.

Abstract

Within a context of evolution and the rapid integration of digitization in university publishing, the fields of the humanities and social sciences, and in particular those of textbooks, do not appear to have found a satisfactory distribution model. In this article, we shall attempt to explain in what measure this sector is different from other areas of university publishing and what hypotheses can be put forward for its future development. Having suggested a carving up of the university publishing market, we would like to place it briefly in an international context. On the basis of this finding, we will present the hypothesis of aggregation: why this should be required, why it does not yet exist and which needs it will respond to. We shall conclude by summarizing these points which we believe are key to the development of a digital university offer in France for the humanities and social sciences.

Abstract

We present a study of energy-energy correlations based on 83 000 hadronic Z0 decays. From this data we determine the strong coupling constant αs to second order QCD: αs(91.2 GeV)=0.121±0.004(exp.)±0.002(hadr.)-0.006 + 0.009(scale)±0.006(theor.) from the energy-energy correlation and αs(91.2 GeV)=0.115±0.004(exp.)-0.004 +0.007(hadr.) -0.000 +0.002(scale)- 0.005 +0.003 (theor.) from its asymmetry using a renormalization scale μ1=0.1 s. The first error (exp.) is the systematic experimental uncertainly, the statistical error is negligible. The other errors are due to hadronization (hadr.), renormalization scale (scale) uncertainties, and differences between the calculated second order corrections (theor.). © 1991.

Abstract

A search for prompt electron production in p p annihilations from an exposure of 2 GeV/c p 's in a bubble chamber with a track-sensitive target is presented. Subtracting the expected Dalitz decay of η and ω for pair masses me + e − >mπ0 we obtain 95% confidence level upper limits for the ratio R = N e − N π − = N e + e − N π 0 as 7 × 10−5.

Abstract

Cosmic radiation produces, in the atmosphere, various radioisotopes which are efficiently collected during the condensation of moisture and can be detected in rain-water. The production rates of various radioisotopes, mainly those whose half-lives make them suitable for studying meteorological phenomena, have been calculated for all parts of the atmosphere, and are presented in graphical form. For the isotope, 10Be, whose half-life is long compared to the characteristic time of mixing between the stratosphere and the troposphere, the fall-out rate agrees with the calculated production rate. For the isotopes, 7Be, 33P, and 32P, whose half-lives are short compared to the characteristic time of mixing between the stratosphere and the troposphere, the measured fall-out rates agree with the calculations, provided one assumes that, 1. (a) only the activity produced in the troposphere appears in rain-water and contributions from air irradiated in the stratosphere are comparatively rare, 2. (b) the mean period, between successive removals of radioisotopes from air masses, is about 1 month. Tentative measurements on the fall-out of 35S activity indicate an appreciably higher rate than calculated. It is shown that the relative concentrations of any two radioisotopes with appreciably different half-lives, in a particular rain, do not depend on the local meteorological conditions, but only on the latitude and altitude at which the air mass was irradiated and can therefore be used as labels to trace the history of air-masses. The study of such isotope ratios in individual precipitations can, therefore, lead to useful meteorological information.

Abstract

The microbial taxonomy is based on the characteristics of microorganisms that can be objectively observed and measured. There are many scheme of microbial classification, but the latest is the three domain system and is the most accepted. Ontologies are the new form of knowledge representation that acts in synergy with agents and Semantic Web Architecture. Ontologies define domain concepts and the relationships between them, and thus provide a domain language that is meaningful to both humans and machines. The relationships in Ontology are explicitly named and developed with specification of rules and constraints so that they reflect the context of domain for which the knowledge is modelled. Ontologies can be built by using various GUI based software tools, known as Ontology editors. Among all editors Protégé is widely supported by a huge research community. For effective use of Ontology, protégé provides a query interface known as SPARQL query panel. SPARQL is a syntactically-SQL-like language for querying RDF graphs. Microbial Taxonomy Ontology is developed for the three domain system of microbes for the domain Bacteria which will be helpful for the study of Agriculturally Important Microbes (Bacteria). This ontology is built in the Protégé OWL editor from Domain to Genus level. Using this ontology, a query interface can be developed that will help detailed study of microbial taxonomy, classification of microbes as well as exchange knowledge between software agents and systems.
