1. Identification of carboxymethyl (CM)-binding proteins derived from Lolium multiflorum pollen extract and antibody reactivity in Brazilian allergic patients

Abstract: Lolium multiflorum grass is the major pollen allergen source in the southern region of Brazil, but most of its allergens remain poorly characterized. The aim of this study was to investigate antibody reactivity to L. multiflorum crude and carboxymethyl-ligand extracts in allergic patients and healthy individuals. Ion exchange carboxymethyl (CM) chromatography (CM-Sepharose) was used to isolate proteins (S2) from L. multiflorum crude extract (S1), which were assessed by SDS-PAGE. S1- and S2-specific IgE and IgG4 levels were measured by ELISA using sera from 55 atopic and 16 non-atopic subjects. Reactive polypeptide bands in S1 and S2 were detected by immunoblotting, and the most prominent bands in S2 were analyzed by mass spectrometry (MS-MS). Similar IgE and IgG4 levels were observed to both S1 (IgE median absorbance: 1.22; IgG4 median absorbance: 0.68) and S2 (IgE median absorbance: 1.26; IgG4 median absorbance: 0.85) in atopic subjects. 2. Alternative Sample Preparation of Co-28Cr-6Mo to Avoid Strain-induced Phase Transformation

Abstract: The allotropic transformation, characteristic of cobalt-based alloys, occurs at around 970 ºC in Co-28Cr-6Mo alloys. When subjected to fast cooling, such alloys can maintain the high temperature cubic centered phase at room temperature, resulting in a CFC metastable matrix. This metastable condition leads to a nucleation of the hexagonal phase (the stable one), which is induced by deformation or isothermally. In general, Co-based alloys are submitted to solution heat treatment plus aging to control both the precipitation of carbides and the nucleation of the hexagonal phase. When the study of this type of alloy is conducted in the metastable condition, it is extremely important do not induce the hexagonal phase during sample preparation. Traditionally, the metallographic route preparation is carried out using electropolishing to avoid the deformation-induced phase transformation. The need for specialized equipment and hazardous electrolytes to perform electropolishing limits its use. Therefore, the aim of the present work is to propose the use of an adaptation to traditional metallographic techniques in order to prepare Co-28Cr-6Mo alloy samples. To this end, the samples prepared by three different routes were analyzed by X-Ray diffraction, including Rietveld refinement, as well as EBSD in order to identify and quantify the phases present in the structure. A control cold rolled sample was also analyzed. The results showed a significant reduction in the HCP phase fraction, strain-induced during grinding, after the application of the alternative preparation method proposed. Further studies might be useful to validate the present methodology.

3. Financial efficiency in the confinement system for beef cattle

Abstract: ABSTRACT The beef cattle breeding system in Brazil has been growing more and more in the history of animal production, leading the country to fit into the world scenario as the main exporter of beef. Seeking to increase this efficiency, the objective of this paper is to evaluate factors that affect the confinement of beef cattle, influencing the Profitability and Return of the system, to define values that maximize them. To do so, a database was used. It consisted of the observation of 1961 animals, collected in the commercial confinement of Fazenda Bragança, in the city of Lucas do Rio Verde - MT. Data were analyzed using multiple linear regression models, in which the financial gain, Profitability and Return variables were explained by the variables: race, entry weight and days in confinement, selected by the stepwise method. Based on the results, there is greater financial efficiency for animals that enter the feedlot with greater weight and that remain for 163 days. It is concluded that the entry weight and days in confinement significantly influence the financial efficiency, allowing the adjusted equations to help producers in making decisions regarding the formation of lots and, mainly, in the purchase of animals.

Abstract: RESUMO O presente estudo teve como objetivo avaliar a atividade antibacteriana, antioxidante e citotóxica da espécie Opuntia cochenillifera (L.) Mill. Foi realizada a prospecção fitoquímica e espectroscopia de absorção de infravermelho (IV) dos extratos etanólicos brutos e frações dos cladódios grande e pequeno. A atividade antioxidante foi avaliada pelo método da capacidade sequestradora de radicais livres utilizando o radical sintético 2,2-difenil-1-picrilhidrazila (DPPH). A atividade citotóxica foi obtida através do método colorimétrico do Metiltetrazolium (MTT). Já a atividade antibacteriana foi avaliada pelo método de microdiluição em caldo para determinar a concentração inibitória mínima (CIM) frente às estirpes bacterianas Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa e Escherichia coli. A prospecção fitoquímica revelou principalmente a presença de fenóis, esteroides livres, alcaloides, alcanos, além de outras classes químicas. O IV apresentou grupos funcionais como alcanos, carbonilas, grupos de metila, duplas ligações de carbono, grupamentos alquilamina, entre outros. Sobre a citotoxicidade na concentração de 100 μg/mL, os dois extratos brutos, todas as frações do cladódio grande e as frações de clorofórmio e metanol do cladódio pequeno não apresentaram toxicidade. Os extratos brutos e frações do cladódio grande e pequeno, não demonstraram atividade antibacteriana e nem antioxidante. Esses resultados podem fornecer suporte para pesquisas futuras, visando outras atividades biológicas da presente espécie vegetal.

5. Effect of hormone replacement on exercise cardiopulmonary reserve and recovery performance in subclinical hypothyroidism

Abstract: Subclinical hypothyroidism (SH) patients present cardiopulmonary, vascular and muscle dysfunction, but there is no consensus about the benefits of levothyroxine (L-T4) intervention on cardiopulmonary performance during exercise. The aim of the present study was to investigate the effects of L-T4 on cardiopulmonary exercise reserve and recovery in SH patients. Twenty-three SH women, 44 (40-50) years old, were submitted to two ergospirometry tests, with an interval of 6 months of normalization of thyroid-stimulating hormone (TSH) levels (L-T4 replacement group) or simple observation (TSH = 6.90 μIU/mL; L-T4 = 1.02 ng/dL). Patients with TSH >10 μIU/mL were excluded from the study to assure that they would receive treatment in this later stage of SH. Twenty 30- to 57-year-old women with no thyroid dysfunction (TSH = 1.38 μIU/mL; L-T4 = 1.18 ng/dL) were also evaluated. At baseline, lower values of gas exchange ratio reserve (0.24 vs 0.30; P < 0.05) were found for SH patients. The treated group presented greater variation than the untreated group for pulmonary ventilation reserve (20.45 to 21.60 L/min; median variation = 5.2 vs 25.09 to 22.45 L/min; median variation = -4.75, respectively) and for gas exchange ratio reserve (0.19 to 0.27; median variation = 0.06 vs 0.28 to 0.18; median variation = -0.08, respectively). There were no relevant differences in cardiopulmonary recovery for either group at baseline or after follow-up. In the sample studied, L-T4 replacement improved exercise cardiopulmonary reserve, but no modification was found in recovery performance after exercise during this period of analysis.

6. Normal flow-mediated vasodilatation of the brachial artery and carotid artery intima-media thickness in subclinical hypothyroidism

Abstract: Subclinical hypothyroidism (SHT) is a disease for which exact therapeutic approaches have not yet been established. Previous studies have suggested an association between SHT and coronary heart disease. Whether this association is related to SHT-induced changes in serum lipid levels or to endothelial dysfunction is unclear. The aim of this study was to determine endothelial function measured by the flow-mediated vasodilatation of the brachial artery and the carotid artery intima-media thickness (IMT) in a group of women with SHT compared with euthyroid subjects. Triglycerides, total cholesterol, HDL-C, LDL-C, apoprotein A (apo A), apo B, and lipoprotein(a) were also determined. Twenty-one patients with SHT (mean age: 42.4 ± 10.8 years and mean thyroid-stimulating hormone (TSH) levels: 8.2 ± 2.7 µIU/mL) and 21 euthyroid controls matched for body mass index, age and atherosclerotic risk factors (mean age: 44.2 ± 8.5 years and mean TSH levels: 1.4 ± 0.6 µIU/mL) participated in the study. Lipid parameters (except HDL-C and apo A, which were lower) and IMT values were higher in the common carotid and carotid bifurcation of SHT patients with positive serum thyroid peroxidase antibodies (TPO-Ab) (0.62 ± 0.2 and 0.62 ± 0.16 mm for the common carotid and carotid bifurcation, respectively) when compared with the negative TPO-Ab group (0.55 ± 0.24 and 0.58 ± 0.13 mm, for common carotid and carotid bifurcation, respectively). The difference was not statistically significant. We conclude that minimal thyroid dysfunction had no adverse effects on endothelial function in the population studied. Further investigation is warranted to assess whether subclinical hypothyroidism, with and without TPO-Ab-positive serology, has any effect on endothelial function.

Abstract: A mistura formulada dos herbicidas imazethapyr e imazapic é utilizada para controlar arroz-vermelho em cultivos de arroz irrigado. Entretanto, esses herbicidas podem persistir no solo por longos períodos, causando introxicação ao arroz suscetível cultivado em sucessão. Este trabalho teve como objetivo avaliar o efeito de diferentes manejos de solo, durante a entressafra do arroz, sobre a ação residual do imazethapyr e imazapic, em arroz não tolerante. O residual desses herbicidas causou introxicação no arroz suscetível após um ano da última aplicação dos herbicidas. A introxicação atingiu valores máximos até 25 dias após a emergência (DAE), ocorrendo redução da introxicação após esse período, até praticamente desaparecer (60 DAE). O residual do herbicida alterou o estande de plantas, o número de colmos m-2, o número de panículas m-2 e a altura de plantas, porém não afetou a produtividade de grãos do arroz. O revolvimento do solo diminuiu a atividade do herbicida na camada superficial de solo (0-3 m).The mixture of herbicides imazethapyr and imazapic is used to control red rice in irrigated rice crops. However, such herbicides might persist on the soil for a long period causing phytotoxicity on susceptible rice grown in succession. The objective of this work was to determine the effect of different soil tillage systems during the off-season on the residual phytotoxicity of imazethapyr and imazapic on non tolerant rice. Herbicide residues caused phytotoxicity on susceptible rice with the highest values being registered 25 days after emergence and decreasing after this period until almost disappearing 60 days after emergence. Herbicide residues affected plant stand, number of stems per m², number of panicles per m² and plant height, but did not affect grain yield. Soil movement decreased herbicide activity on the superficial soil layer (0-3 cm).

8. Humoral and cellular immune responses to Blomia tropicalis and concanavalin A-binding fractions in atopic patients

Abstract: Blomia tropicalis, Dermatophagoides pteronyssinus and D. farinae are prevalent house dust mites. Concanavalin A-binding components derived from B. tropicalis (Bt-ConA extract) are highly immunogenic in allergic diseases. The aim of the present study was to evaluate the humoral and cellular immune responses to B. tropicalis in mite-sensitized patients. A total of 137 patients with allergic rhinitis with/without asthma and 109 non-atopic subjects were selected and analyzed by the skin prick test, and for total serum IgE and specific IgE levels to both Bt-total and Bt-ConA extracts, their proliferative response and cytokine (IFN-γ and IL-5) production by peripheral blood mononuclear cells (PBMC) stimulated with both extracts. Skin prick test showed that 70% of the patients were sensitized to Bt (Bt+) and similar levels of specific IgE to Bt-total and Bt-ConA extracts were demonstrable in Bt+ patients. Significant PBMC proliferation was observed in response to Bt-total extract in Bt+, but not in Bt- patients and non-atopic subjects (P < 0.001). Bt-ConA extract induced increased proliferative responses in all patient groups compared to medium alone (P < 0.05), but these responses were significantly decreased in the presence of the mannopyranoside ConA inhibitor (P < 0.05). Significant IFN-γ production was observed after Bt-ConA stimulation of Bt+ patients (P < 0.05), while Bt-total extract had no effect. IL-5 production was consistently detected in Bt+ patients after allergen-specific stimulation or with no stimulus, indicating that PBMC from allergic patients are prone to produce Th2 profile cytokines, spontaneously or inductively by allergen restimulation. These data showed that ConA-binding components isolated from B. tropicalis may contain relevant antigens that are involved in both humoral and cellular immune responses. However, without an additional purification procedure to eliminate the residual contamination with ConA, its use in immunotherapeutic procedures cannot be recommended.

9. Controle químico de arroz-vermelho na cultura do arroz irrigado Red rice chemical control in irrigated rice

Abstract: A ocorrência de arroz-vermelho (Oryza spp.) em áreas de arroz irrigado reduz a produtividade de grãos da lavoura e a qualidade do produto colhido. Em vista disso, desenvolveu-se um experimento com o objetivo de comparar duas ferramentas para controle do arroz-vermelho: uma usando o Sistema Clearfield e outra utilizando doses elevadas de clomazone em sementes tratadas com protetor para supressão de arroz-vermelho. Os tratamentos constituíram-se de uma testemunha, sem aplicação de herbicida, três referentes à aplicação da mistura formulada (75 g i.a. ha L-1 de imazethapyr + 25 g i.a. ha L-1 de imazapic) e outros três referentes à aplicação do herbicida clomazone. O tratamento mais eficiente no controle de arroz-vermelho foi a aplicação da mistura formulada de imazethapyr + imazapic em pré-emergência, seguido da mesma dose em pós-emergência. Este tratamento proporcionou 100% de controle de arroz-vermelho, além de não prejudicar o estande inicial de plantas e proporcionar alto rendimento de grãos do arroz irrigado.Red rice (Oryza spp.) reduces rice (Oryza sativa L.) grain yield and quality. A field study was conducted to compare two red rice control tools, the first using the Clearfield™ system and the second using high rates of clomazone and rice seeds treated with safener to suppress red rice emergence. The treatments included: check control without herbicide application, three treatments for the formulated herbicide mixture (imazethapyr 75 g i.a. L-1 + imazapic 25 g i.a. L-1) under the Clearfield system, and three clomazone treatments (clomazone 500 g i.a. L-1). The most efficient treatment for red rice control was the formulated mixture of imazethapyr + imazapic (0.7 L ha-1 PRE followed by 0.7 L ha-1 POST). This treatment promoted 100% red rice control, without affecting plant stand and promoting high grain yield.

Abstract: Após várias décadas de busca de alternativas para controle do arroz-vermelho, desenvolveram-se genótipos de arroz tolerantes a herbicida do grupo químico das imidazolinonas, o qual controla eficientemente esta planta daninha no Sistema Clearfield. O experimento teve como objetivo avaliar: a eficiência do controle de arroz-vermelho com a mistura formulada dos herbicidas imazethapyr (75 g L-1) + imazapic (25 g L-1) (produto comercial Only®); o residual do herbicida no solo através dos danos causados ao azevém e arroz não-tolerante; e a taxa de ocorrência de cruzamento natural entre o arroz-vermelho e o arroz cultivado. O delineamento experimental foi de blocos ao acaso, com três tratamentos e doze repetições. Para determinar o fluxo gênico entre o arroz tolerante a imidazolinonas e o arroz-vermelho, foram coletadas e analisadas as panículas de arroz-vermelho não-controladas. O efeito residual do herbicida em culturas não-tolerantes foi verificado através de coleta de fitomassa de azevém e do estande inicial do cultivar de arroz não-tolerante semeado no ano seguinte. O herbicida testado controlou eficientemente o arroz-vermelho e a fitotoxicidade inicial não reduziu a produtividade do cultivar tolerante. O estande inicial do cultivar IRGA 417 foi afetado pelo residual do herbicida no solo. Os resultados mostraram também que ocorre cruzamento natural entre o arroz-vermelho e o arroz cultivado, e a taxa obtida no experimento foi de 0,065%.After several decades searching for tools to control red rice, imidazolinone tolerant rice was developed to selectively control red rice. To better understand this technology An experiment was conducted in 2004/2005, in Santa Maria, Rio Grande do Sul, Brazil to evaluate: 1) red rice control by imidazolinone herbicides in Clearfield™ rice; 2) evaluate the imidazolinone herbicide carryover effect on rygrass and non-tolerant rice (IRGA 417) and 3) evaluate the outcrossing rates between Clearfield rice and red rice. The experimental design was a randomized block design, with 3 treatments and 12 replications. To determine the outcrossing rates between Clearfield rice and red rice, virtually all the red rice panicle was collected and analyzed in the area. The carryover effect was tested using ryegrass and a non-tolerant rice cultivar (IRGA 417). The herbicides tested controlled red rice. Although injury to Clearfield rice was observed, the herbicide did not affect yield. Herbicide carry-over to non-tolerant rice was observed, reducing plant stand on rice seeded 361 days after herbicide application. Natural out-crossing occurred between Clearfield rice and red rice, at rates of 0.065%.

Abstract: A infestação por arroz-vermelho (Oryza spp.) constitui-se num dos principais fatores limitantes da produtividade de grãos do arroz irrigado. Este trabalho teve como objetivo avaliar o controle de arroz-vermelho e o desempenho de dois genótipos de arroz irrigado, IRGA 422 CL e Tuno CL, tolerantes a herbicidas do grupo das imidazolinonas em resposta a doses e épocas de aplicações da mistura formulada de imazethapyr (75 g L-1) + imazapic (25 g L-1) (produto comercial Only®), em áreas com alta infestação de arroz-vermelho. O experimento foi conduzido em Santa Maria-RS no ano agrícola 2004/05. O delineamento experimental foi de blocos ao acaso em esquema bifatorial (2 x 10), com quatro repetições. O fator A foi composto por dois genótipos de arroz tolerantes às imidazolinonas, um cultivar (IRGA 422 CL) e um híbrido (Tuno CL); e o fator D, pelos tratamentos para controle de arroz-vermelho oriundos de combinações de doses e épocas de aplicação do herbicida. Constatou-se que o híbrido é mais tolerante ao herbicida Only®, quando comparado ao cultivar, sendo possível a utilização de dose total de até 200% no híbrido, em áreas com alta infestação de arroz-vermelho, sem afetar a produtividade. Porém é importante salientar que o incremento da dose do herbicida pode causar problemas de residual a culturas não tolerantes semeadas na seqüência.O controle de arroz-vermelho é total com aplicação fracionada do herbicida em pré e pós-emergência (PRÉ + PÓS), desde que o total aplicado não seja inferior a 125%. Essa condição é atendida pelo tratamento com 75% em PRÉ seguido de 50% em PÓS, o qual propicia a menor dose total entre aqueles com 100% de controle, não afetando a produtividade e apresentando fitotoxicidade semelhante ao tratamento com 100% em PÓS, utilizado como referência.Red rice (Oryza spp.) is one of the main limiting factors to rice (O. sativa) yield. An experiment was carried out to evaluate red rice control and the behavior of two rice genotypes tolerant to the imidazolinone herbicides in response to imazethapyr (75 g L-1 ) + imazapic (25 g L¹ ) application rates and timing. The experiment was conducted in Santa Maria-RS, Brazil in 2004/2005 and was arranged in a factorial scheme, in a randomized block design, with four replications. Factor A included the two rice genotypes tolerant to the imidazolinones, a cultivar (IRGA 422 CL) and a hybrid (Tuno CL); and factor D included the treatments for red rice control, which was a combination of rates and herbicide application timing. The hybrid was found to be more tolerant to the herbicide only than the cultivar. Application rates up to 200% on the hybrid genotype could be done without affecting rice yield. It is important to state that increasing the rate of herbicide application can create carryover problems to non-tolerant crops. Red rice control was total with split application of imazethapyr + imazapic in PRE and POST emergence with the total rate above 125%. The most efficient treatment was application of 75% in PRE followed by 50% in POST, which was the lowest rate promoting 100% control, with relatively low toxicity to the cultivar and without affecting rice yield.

Abstract: The allotropic transformation, characteristic of cobalt-based alloys, occurs at around 970 ºC in Co-28Cr-6Mo alloys. When subjected to fast cooling, such alloys can maintain the high temperature cubic centered phase at room temperature, resulting in a CFC metastable matrix. This metastable condition leads to a nucleation of the hexagonal phase (the stable one), which is induced by deformation or isothermally. In general, Co-based alloys are submitted to solution heat treatment plus aging to control both the precipitation of carbides and the nucleation of the hexagonal phase. When the study of this type of alloy is conducted in the metastable condition, it is extremely important do not induce the hexagonal phase during sample preparation. Traditionally, the metallographic route preparation is carried out using electropolishing to avoid the deformation-induced phase transformation. The need for specialized equipment and hazardous electrolytes to perform electropolishing limits its use. Therefore, the aim of the present work is to propose the use of an adaptation to traditional metallographic techniques in order to prepare Co-28Cr-6Mo alloy samples. To this end, the samples prepared by three different routes were analyzed by X-Ray diffraction, including Rietveld refinement, as well as EBSD in order to identify and quantify the phases present in the structure. A control cold rolled sample was also analyzed. The results showed a significant reduction in the HCP phase fraction, strain-induced during grinding, after the application of the alternative preparation method proposed. Further studies might be useful to validate the present methodology.

Abstract: The shortage of reliable primary taxonomic data limits the description of biological taxa and the understanding of biodiver sity patterns and processes, complicating biogeographical, ecological, and evolutionary studies. This deficit creates a significant taxo nomic impediment to biodiversity research and conservation planning. The taxonomic impediment and the biodiversity crisis are widely recognized, highlighting the urgent need for reliable taxonomic data. Over the past decade, numerous countries worldwide have devoted considerable effort to Target 1 of the Global Strategy for Plant Conservation (GSPC), which called for the preparation of a working list of all known plant species by 2010 and an online world Flora by 2020. Brazil is a megadiverse country, home to more of the world’s known plant species than any other country. Despite that, Flora Brasiliensis, concluded in 1906, was the last comprehensive treatment of the Brazilian flora. The lack of accurate estimates of the number of species of algae, fungi, and plants occurring in Brazil contributes to the prevailing taxonomic impediment and delays progress towards the GSPC targets. Over the past 12 years, a legion of taxonomists motivated to meet Target 1 of the GSPC, worked together to gather and integrate knowledge on the algal, plant, and fungal diversity of Brazil. Overall, a team of about 980 taxonomists joined efforts in a highly collaborative project that used cybertaxonomy to prepare an updated Flora of Brazil, showing the power of scientific collaboration to reach ambitious goals. This paper presents an overview of the Brazilian Flora 2020 and provides taxonomic and spatial updates on the algae, fungi, and plants found in one of the world’s most biodiverse countries. We further identify collection gaps and summarize future goals that extend be yond 2020. Our results show that Brazil is home to 46,975 native species of algae, fungi, and plants, of which 19,669 are endemic to the country. The data compiled to date suggests that the Atlantic Rainforest might be the most diverse Brazilian domain for all plant groups except gymnosperms, which are most diverse in the Amazon. However, scientific knowledge of Brazilian diversity is still un equally distributed, with the Atlantic Rainforest and the Cerrado being the most intensively sampled and studied biomes in the coun try. In times of “scientific reductionism”, with botanical and mycological sciences suffering pervasive depreciation in recent decades, the first online Flora of Brazil 2020 significantly enhanced the quality and quantity of taxonomic data available for algae, fungi, and plants from Brazil. This project also made all the information freely available online, providing a firm foundation for future research and for the management, conservation, and sustainable use of the Brazilian funga and flora. Abstract: The dual active bridge (DAB) is the mostly used DC–DC converter in bidirectional power flow applications. It has several advantages such as high-power density, galvanic isolation, robustness, among others. However, in applications in which direct and reverse power flow have great difference, the converter efficiency can be penalised. A recent proposed isolated converter known as integrated full-bridge-forward DC–DC converter is specifically designed to this application and has low number of active devices compared to DAB converter. In order to evaluate the features of both converters, this Letter proposes a comparative analysis between these two topologies. Besides theoretical details about the converters, this Letter focuses on the comparison of experimental results, including efficiency and zero voltage switching range.

41. Physical properties of Smx B6 single crystals

Abstract: We study the magnetic susceptibility, electrical resistivity, Hall effect, and heat capacity of single crystals of SmxB6 for x=1, 0.94, 0.8, and 0.75. Remarkably, the overall properties of the crystals do not qualitatively depend on the density of vacancies. The topological surface states are seen at low temperatures in the electrical transport properties as the bulk conductivity freezes out. Even a large number of Sm vacancies does not close the hybridization gap. The linear term in the specific heat ¿, of unclear origin, remains large but decreases gradually with x. The shoulder in the density of states, brought by the hybridized 4f states and observed in the susceptibility and the specific heat, does not move but decreases its height with decreasing x. The specific heat also displays a weak Schottky anomaly at about 1.5 K for all x, except for the x = 0.75 sample. This Schottky anomaly is only slightly field dependent and of unknown origin.

Abstract: The Global Strategy for Plant Conservation (GSPC) was established by the Conference of Parties in 2002 to decrease the loss of plant diversity, reduce poverty and contribute to sustainable development. To achieve this overarching goal, the GSPC has established a series of targets, one of which is to ensure that plant diversity is well understood, so that it can be effectively conserved and used in a sustainable manner. Brazil hosts more than 46,000 species of plants, algae and fungi, representing one of the most biodiverse countries on Earth, and playing a key role in the GSPC. To meet the GSPC goals of Target 1 and facilitate access to plant diversity, Brazil committed to preparing the List of Species of the Brazilian Flora (2008-2015) and the Brazilian Flora 2020 (2016-present). Managing all the information associated with such great biodiversity has proven to be an extremely challenging task. Here, we synthesize the history of these projects, focusing on the multidisciplinary and collaborative approach adopted to develop and manage the inclusion of all the knowledge generated though digital information systems. We further describe the methods used, challenges faced, and strategies adopted, as well as summarize advances to date and prospects for completing the Brazilian flora in 2020.

Abstract: The shortage of reliable primary taxonomic data limits the description of biological taxa and the understanding of biodiversity patterns and processes, complicating biogeographical, ecological, and evolutionary studies. This deficit creates a significant taxonomic impediment to biodiversity research and conservation planning. The taxonomic impediment and the biodiversity crisis are widely recognized, highlighting the urgent need for reliable taxonomic data. Over the past decade, numerous countries worldwide have devoted considerable effort to Target 1 of the Global Strategy for Plant Conservation (GSPC), which called for the preparation of a working list of all known plant species by 2010 and an online world Flora by 2020. Brazil is a megadiverse country, home to more of the world's known plant species than any other country. Despite that, Flora Brasiliensis, concluded in 1906, was the last comprehensive treatment of the Brazilian flora. The lack of accurate estimates of the number of species of algae, fungi, and plants occurring in Brazil contributes to the prevailing taxonomic impediment and delays progress towards the GSPC targets. Over the past 12 years, a legion of taxonomists motivated to meet Target 1 of the GSPC, worked together to gather and integrate knowledge on the algal, plant, and fungal diversity of Brazil. Overall, a team of about 980 taxonomists joined efforts in a highly collaborative project that used cybertaxonomy to prepare an updated Flora of Brazil, showing the power of scientific collaboration to reach ambitious goals. This paper presents an overview of the Brazilian Flora 2020 and provides taxonomic and spatial updates on the algae, fungi, and plants found in one of the world's most biodiverse countries. We further identify collection gaps and summarize future goals that extend beyond 2020. Our results show that Brazil is home to 46,975 native species of algae, fungi, and plants, of which 19,669 are endemic to the country. The data compiled to date suggests that the Atlantic Rainforest might be the most diverse Brazilian domain for all plant groups except gymnosperms, which are most diverse in the Amazon. However, scientific knowledge of Brazilian diversity is still unequally distributed, with the Atlantic Rainforest and the Cerrado being the most intensively sampled and studied biomes in the country. In times of “scientific reductionism”, with botanical and mycological sciences suffering pervasive depreciation in recent decades, the first online Flora of Brazil 2020 significantly enhanced the quality and quantity of taxonomic data available for algae, fungi, and plants from Brazil. This project also made all the information freely available online, providing a firm foundation for future research and for the management, conservation, and sustainable use of the Brazilian funga and flora.

Abstract: Several routes have been developed to prepare magnetic core–shell Fe3O4@Au nanoparticles (MNPs). However, only highly monodisperse MNPs are suitable for analytical applications. Herein, we describe the detection of GMO through the use of gold-coated MNPs with fine-tuned properties as platforms. The MNPs were prepared through a procedure that involves the preparation of Fe3O4 cores by thermal decomposition and their coating through reduction of a gold precursor. Different Fe3O4:Au precursor molar ratios (1:1; 1:4; 1:7) were tested on the Fe3O4 encapsulation. Monodisperse quasi-spherical core–shell Fe3O4@Au were obtained for the 1:4 and 1:7 ratios, in contrast, the 1:1 ratio did not lead to complete encapsulation of Fe3O4 cores. Therefore, the Fe3O4@Au obtained from higher Fe3O4:HAuCl4 ratios were tested as platforms for an electrochemical genoassay to detect MON810.
